Improvements have been made on the currently available hydride generator system manufactured by SpectraMetrics Incorporated, because the system was found to be unsatisfactory with respect to the following: 1. the drying agent, anhydrous calcium chloride, 2. the special sample tube, 3. the direction of argon flow through the Buchner funnel when it came to dealing with real sample, that is, with reference only to aqueous extracts of soil samples. Changes that were made on the system included the replacement of anhydrous calcium chloride with anhydrous calcium sulphate and the replacement of the special sample tube with a modified one made from silica. Re-directing the flow of argon through the top of the Buchner funnel appeared to make the system compatible with aqueous extracts of soil samples. The interferences from 1000 ~g/mL of nickel(II) , cobalt(II), iron(III), copper(II) have been eliminated with the aid of 1.4 M hydrochloric acid and 1% (weight/volume) L-cystine. Greater than 90% recovery of 0.3 ~g/mL arsenic signal was achieved in each case. Furthermore, 103% of arsenic signal was accomplished in the presence of 1000 ~g/mL cadmium with 5 M Hel. tVhen each of the interferents was present in solution at 1000 ppm, a recovery of 85% was achieved by using 5 M hydrochloric acid and 3% (weight/volume) L-cystine. Without L-cystine and when 1.4 M hydrochloric acid was used, the recoveries were 0% (Ni), 0% (Co), 88% (Fe), 15% (Cu), 18% (Cd). Similarly, a solution containing 1000 ppm of each interferent gave a zero percent recovery of arsenic. The reduction of trivalent and pentavalent arsenic at a pH less than one has also been investigated and shown to be quantitative if peak areas are measured. The reproducibility determination of a 0.3 Vg/mL standard arsenic solution by hydride generation shows a relative standard deviation of 3.4%. The detection limits with and without Porapak Q have been found to be 0.6 ng/mL and 1.0 ng/mL, respectively.

Trilobites ¥tere collected from Ordovician and Devonian formations of Ontario} New York} Ohio} Oklahoma} and Indiana. Diversity was generally low} but 19..?telllS and Ph..~tY>ps ¥tere the most abundant species from the Ordovician and Devonian} respectively. Recent marine arthropods ¥tere collected from the Atlantic shore of the middle Florida Keys} and from the Pacific and lagoonal waters at Cape Beale} B. C. Fresh-water arthropods were collected along the shore of the Severn River in northcentral Ontario. Cuticles ¥tere analyzed for major} minor and trace elements, 180 and 13C isotopes, as ¥tell as examined by scanning electron micr?scope to identify original and diagenetic fabrics. Examination of trilobite cuticles by scanning electron microscope revealed several microstructures consistent with those observed in Recent arthropods. Microstructures} such as setae and tegumental gland duct openings} in like sized Lim/IllS and Isoteline trilobites may indicate common ancestral origins for these organisms, or simply parallel cuticle evolutions. The dendritic microstructure, originally' thought to be a diagenetic indicator, was found in Recent specimens and therefore its presence in trilobites may be suggestive of the delicate nature of diagenesis in trilobites. The absence of other primary microstructures in trilobites may indicate alteration, taxonomic control} or that there is some inherent feature of S EM examination which may' not allow detection of some features} while others are apparently visit·le onl~1 under SH.·1. The region of the cuticle sampled for examination is also a major influence in detecting pristine microstructures, as not all areas of trilobite and Recent arthropod cuticles will have microstructures identifiable in a SEM study. Subtleties in the process of alteration, however} ma~·· leave pristine microstructures in cuticles that are partial~/ silicified or do 10m itized, and degree and type of alteration may vary stratigraphically and longitudinally within a unit. The presence of fused matrices, angular calcite rhombs, and pyrite in the cuticle are thought to be indicative of altered cuticles, although pyritization may not affect the entire cuticle. t-~atural processes in Recent arthropods, such as molting, lead to variations in cuticle chemistries, and are thought to reflect the area of concentration of the elements during calcification. The level of sodium in Recent arthropods was found to be higher than that in trilobites, but highly mobile when sUbjected to the actions of VY'€'athering. Less saline water produced lovy'€'r magnesium and higher calcium values in Recent specimens .. and metal variations in pristine Ordovician trilobite cuticle appears to follow the constraints outlined for Recent arthropods, of regulation due to the chemislry of the surrounding medium. In diagenetic analysis, sodium, strontium and magnesium proved most beneficial in separating altered from least altered trilobites. Using this criterion, specimens from shale show the least amount of geochemical alteration, and have an original mineralogy of 1.7 - 2.4 mole % MgC03 (8000 t(> 9500 ppm magnesium) for both /s>..?/e/11S lJA'i.riff!11S and PseIAit'11J17ites I..itmirpin..itl/~ and 2.8 - 3.3 mole % MgC03 (5000 to 7000 ppm magnesium) for Ph.i{).?PS This is Slightly lower than the mineralogy of Recent marine arthropods (4.43 - 12.1 mole % MgC03), and slightly higher than that of fresh-water crayfish (0.96 - 1.82 mole % MgC03). Geochemically pristine trilobites were also found to possess primary microstructures. Stable isotope values and trends support the assertion that marine-meteoriclburial fluids were responsible for the alteration observed in a number of the trilobite specimens. The results of this stUdy suggest that fossil material has to be evaluated separately along taxonomic and lithological lines to arrive at sensible diagenetic and e nvironmenta I interpretations.

A survey of predominantly industrial silicon carbide has been carried out using Magic Angle Spinning nuclear magnetic resonance (MAS nmr); a solid state technique. Three silicon carbide polytypes were studied; 3C, 6H, and 15R. The 13C and 29 Si MAS nmr spectra of the bulk SiC sample was identified on the basis of silicon (carbon) site type in the d iff ere n t pol Y t Y pes • Out to 5.00 A fro mac en t r a lsi 1 i con (0 r carbon) atom four types of sites were characterized using symmetry based calculations. This method of polytype analysis was also considered, in the prelminary stages, for applications with other polytypic material; CdBr 2 , CdI 2 , and PbI 2 " In an attempt to understand the minor components of silicon carbide, such as its surface, some samples were hydrofluoric acid washed and heated to extreme temperatures. Basically, an HF removable species which absorbs at -110 ppm (Si0 2 ) in the 29 Si MAS nmr spectrum is found in silicon carbide after heating. Other unidentified peaks observed at short recycle delays in some 29 Si MAS nmr spectra are considered to be impurities that may be within the lattice. These components comprise less than 5% of the observable silicon. A Tl study was carried out for 29 Si nuclei in a 3C ii polytype sample, using the Driven Equilibrium Single-Pulse Observation of T1 (DESPOT) technique. It appears as though there are a number of nuclei that have the same chemical shift but different T1 relaxation times. The T1 values range from 30 seconds to 11 minutes. Caution has to be kept when interpreting these results because this is the first time that DESPOT has been used for solid samples and it is not likely in full working order. MAS nmr indicates that the 13C and 29 Si ~sotropic chemical shifts of silicon carbide appear to have a reciprocal type of relationship_ Single crystal nmr analysis of a 6H sample is accordance with this finding when only the resultant isotropic shift is considered. However, single crystal nmr also shows that the actual response of the silicon and carbon nuclear environment to the applied magnetic field at various angles is not at all reciprocal. Such results show that much more single crystal nmr work is required to determine the actual behavior of the local magnetic environment of the SiC nuclei.

A high performance liquid chromatographic method employing two columns connected in series and separated~y·a.switching valve has been developed for the analysis of the insecticide/ nematicide oxamyl (methyl-N' ,N'-dimethyl-N-[(methylcarbamoyl) oxy]-l-thiooxarnimidate) and two of its metabolites. A variation of this method involving two reverse phase columns was employed to monitor the persistence and translocation of oxamyl in treated peach seedlings. It was possible to simultaneously analyse for oxamyl and its corresponding oxime (methyl-N',N'-dimethyl-N-hydroxy-l-thiooxamimidate}, a major metabolite of oxamyl in plants, without prior cleanup of the samples. The method allowed detection of 0.058 pg oxamyl and 0.035 p.g oxime. On treated peach leaves oxamyl was found to dissipate rapidly during the first two-week period, followed by a period of slow decomposition. Movement of oxamyl or its oxime did not occur in detectable quantities to untreated leaves or to the root or soil. A second variation of the method which employed a size exclusion column as·the first column and a reverse phase column as the second was used to monitor the degradation of oxamyl in treated, planted corn seeds and was suitable for simultaneous analysis of oxamyl, its oxime and dimethylcyanoformamide (DMCF), a metabolite of oxamyl. The method allowed detection of 0.02 pg oxamyl, 0.02 p.g oxime and 0.005 pg DMCF. Oxamyl was found to persist for a period of 5 - 6 weeks, which is long enough to permit oxamyl seedtreatment to be considered as a potential means of protecting young corn plants from nematode attack. Decomposition was found to be more rapid in unsterilized soil than in sterililized soil. DMCF was found to have a nematostatic effect at high concentrations ( 2,OOOpprn), but at lower concentrations no effect on nematode mobility was observed. Oxamyl, on the other hand, was found to reduce the mobility of nematodes at concentrations down to 4 ppm.

Niagara Peninsula of Ontario is the largest viticultural area in Canada. Although it is considered to be a cool and wet region, in the last decade many water stress events occurred during the growing seasons with negative effects on grape and wine quality. This study was initiated to understand and develop the best strategies for water management in vineyards and those that might contribute to grape maturity advancement. The irrigation trials investigated the impact of time of initiation (fruit set, lag phase and veraison), water replacement level based on theoretical loss through crop evapotranspiration (ETc; 100,50 and 25%) and different irrigation strategies [partial root zone drying (PRD) versus regulated deficit irrigation (RD!)] on grape composition and wine sensory profiles. The irrigation experiments were conducted in a commercial vineyard (Lambert Vineyards Inc.) located in Niagara-on-the-Lake, Ontario, from 2005 through 2009. The two experiments that tested the combination of different water regimes and irrigation time initiation were set up in a randomized block design as follows: Baco noir - three replicates x 10 treatments [(25%, 50% and 100% of ETc) x (initiation at fruit set, lag phase and veraison) + control]; Chardonnay - three replicates x seven treatments [(25%, 50% and 100% of ETc) x (initiation at fruit set and veraison) + control]. The experiments that tested different irrigation strategies were set up on two cultivars as follows: Sauvignon blanc - four replicates x four treatments [control, fully irrigated (100% ETc), PRD (100% ETc) and RDI (25% ETc)]; Cabemet Sauvignon - four replicates x five treatments [control, fully irrigated (100% ETc), PRD (100% ETc), RDI (50% ETc) and RDI (25% ETc)]. The controls in each experiment were nonirrigated. The irrigation treatments were compared for many variables related to soil water status, vine physiology, berry composition, wine sensory profile, and hormone composition [(abscisic acid (ABA) and its catabolites]. Soil moisture profile was mostly affected by irrigation treatments between 20 and 60 em depth depending on the grapevine cultivar and the regime of water applied. Overall soil moisture was consistently higher throughout the season in 100 and 50% ETc compare to the control. Transpiration rates and leaf temperature as well as shoot growth rate were the most sensitive variables to soil water status. Drip irrigation associated with RDI treatments (50% ETc and 25% ETc) had the most beneficial effects on vine physiology, fruit composition and wine varietal typicity, mainly by maintaining a balance between vegetative and reproductive parts of the vine. Neither the control nor the 100 ETc had overall a positive effect on grape composition and wine sensory typicity. The time of irrigation initiation affected the vine physiology and grape quality, the most positive effect was found in treatments initiated at lag phase and veraison. RDI treatments were overall more consistent in their positive effect on grape composition and wine varietal typicity comparing to PRD treatment. The greatest difference between non-irrigated and irrigated vines in most of the variables studied was found in 2007, the driest and hottest season of the experimental period. Soil water status had a greater and more consistent effect on red grapevine cultivars rather than on white winegrape cultivars. To understand the relationships among soil and plant water status, plant physiology and the hormonal profiles associated with it, abscisic acid (ABA) and its catabolites [phaseic acid (PA), dihydrophaseic acid (DPA), 7-hydroxy-ABA (TOH-ABA), 8' -hydroxy-ABA, neophaseic acid and abscisic acid glucose ester (ABA-GE)] were analyzed in leaves and berries from the Baco noir and Chardonnay irrigation trials over two growing seasons. ABA and some of its catabolites accurately described the water status in the vines. Endogenous ABA and some of its catabolites were strongly affected in Baco noir and Chardonnay by both the water regime (i.e. ET level) and timing of irrigation initiation. Chardonnay grapevines produced less ABA in both leaves and berries compared to Baco noir, which indicated that ABA synthesis is also cultivar dependant. ABA-GE was the main catabolite in treatments with high water deficits, while PA and DPA were higher in treatments with high water status, suggesting that the vine produced more ABA-GE under water deficits to maintain rapid control of the stomata. These differences between irrigation treatments with respect to ABA and catabolites were particularly noticeable in the dry 2007 season. Two trials using exogenous ABA investigated the effect of different concentrations of ABA and organs targeted for spraying, on grape maturation and berry composition of Cabemet Sauvignon grapevines, in two cool and wet seasons (2008-2009). The fIrst experiment consisted of three replicates x three treatments [(150 and 300 mg/L, both applications only on clusters) + untreated control] while the second experiment consisted in three replicates x four treatments [(full canopy, only clusters, and only leaves sprayed with 300 ppm ABA) + untreated control]. Exogenous ABA was effective in hastening veraison, and improving the composition of Cabemet Sauvignon. Ability of ABA to control the timing of grape berry maturation was dependant on both solution concentration and the target organ. ABA affected not only fruit composition but also yield components. Berries treated with ABA had lower weight and higher skin dry mass, which constitutes qualitative aspects desired in the wine grapes. Temporal advancement of ripening through hormonal control can lead to earlier fruit maturation, which is a distinct advantage in cooler areas or areas with a high risk of early frost occurrence. Exogenous ABA could provide considerable benefits to wine industry in terms of grape composition, wine style and schedule activities in the winery, particularly in wet and cool years. These trials provide the ftrst comprehensive data in eastern North America on the response of important hybrid and Vitis vinifera winegrape cultivars to irrigation management. Results from this study additionally might be a forward step in understanding the ABA metabolism, and its relationship with water status. Future research should be focused on ftnding the ABA threshold required to trigger the ripening process, and how this process could be controlled in cool climates.

L’effort physique a été reconnu comme l’un des déterminants majeurs qui affecte l’absorption pulmonaire et la cinétique des solvants industriels, composés volatils très répandus dans le milieu de travail. L’objectif global de ce projet était de caractériser la relation entre divers niveaux de charge de travail et les concentrations biologiques de l’acétone et du styrène ou de ses métabolites utilisés comme des indicateurs biologiques de l’exposition (IBEs) à ces solvants. Des modèles pharmacocinétiques à base physiologique ont été développés et validés afin de reproduire une exposition professionnelle à l’acétone et au styrène, individuellement et en combinaison, durant une semaine complète de travail (8h/jour, 5 jours). Les simulations ont été effectuées suivant une exposition aux valeurs limite d’exposition (500 ppm et 20 ppm, respectivement) et à des charges de travail de 12,5 W (repos), 25 W et 50 W. Les valeurs prédites par les modèles ont été comparées aux valeurs de référence des IBEs actuels. Le niveau d’acétone dans l’urine obtenu à la fin du dernier quart de travail était 3,5 fois supérieur à la valeur au repos (28 mg/L) pour un effort de 50 W, tandis que les niveaux de styrène dans le sang veineux et de ses métabolites dans l’urine ont augmenté d’un facteur d’environ 3,0 en comparaison avec les valeurs au repos, respectivement de 0,17 mg/L et 144 mg/g créatinine. Pour une co-exposition à des concentrations de 20 ppm de styrène et 200 ppm d’acétone et à une activité physique de 50 W, les simulations ont montré une augmentation de 10% du styrène sanguin et une faible diminution de ses métabolites dans l’urine. Les valeurs simulées par les modèles pour l’acétone ou le styrène montrent que des travailleurs dont la charge de travail équivaut à plus de 25 W sont susceptibles d’avoir des concentrations internes pouvant dépasser les valeurs de référence des IBEs respectifs de ces solvants et peuvent être à risque. Les résultats soulignent ainsi l’importance de tenir compte de la charge de travail dans la détermination d’une valeur de référence pour la surveillance biologique de l’acétone et du styrène.

L'évaluation des risques de l'exposition aux mélanges de produits chimiques par voies multiples peut être améliorée par une compréhension de la variation de la dose interne due à l’interaction entre les produits. Les modèles pharmacocinétiques à base physiologique (PBPK) sont des outils éprouvés pour prédire l'ampleur de ces variations dans différents scénarios. Dans cette étude, quatre composés organiques volatils (COV) (toluène, nhexane, cyclohexane et isooctane) ont été choisis pour représenter des produits pétroliers (essence) pouvant contaminer l'eau potable. Premièrement, les modèles PBPK ont simulé l'exposition à un seul COV par une voie (inhalation ou gavage). Ensuite, ces modèles ont été interconnectés pour simuler l'exposition à un mélange par voies multiples. Les modèles ont été validés avec des données in vivo chez des rats Sprague-Dawley (n=5) exposés par inhalation (50 ppm ; toluène, hexane, et 300 ppm ; cyclohexane, isooctane; 2-h) ou par gavage (8,3; 5,5; 27,9 et 41,27 mg/kg pour le toluène, l’hexane, le cyclohexane et l’isooctane, respectivement). Des doses similaires ont été utilisées pour l'exposition au mélange par voies multiples. Les AUC (mg/L x min) pour le toluène, l'hexane, le cyclohexane et l'isooctane étaient respectivement de 157,25; 18,77; 159,58 et 176,54 pour les données expérimentales, et 121,73; 21,91; 19,55 et 170,54 pour les modèles PBPK. Les résultats des modèles PBPK et les données in vivo (simple COV par voies multiples vs. mélange par voies multiples) ont montré des interactions entre les COVs dans le cas de l'exposition au mélange par voies multiples. Cette étude démontre l'efficacité des modèles PBPK pour simuler l'exposition aux mélanges de COV par voies multiples.

Un modèle pharmacocinétique à base physiologique (PBPK) d’exposition par inhalation à l’éthanol a antérieurement été développé en se basant sur des données provenant d’une étude chez des volontaires exposés par inhalation à plus de 5000 ppm. Cependant, une incertitude persiste sur la capacité du modèle PBPK à prédire les niveaux d’éthanolémie pour des expositions à de faibles concentrations. Ces niveaux sont fréquemment rencontrés par une large partie de la population et des travailleurs suite à l’utilisation de produits tels que les vernis et les solutions hydroalcooliques (SHA). Il est ainsi nécessaire de vérifier la validité du modèle existant et de déterminer l’exposition interne à l’éthanol dans de telles conditions. Les objectifs du mémoire sont donc 1) de documenter les niveaux d’éthanolémie résultant de l’exposition par inhalation à de faibles concentrations d’éthanol (i.e., ≤ 1000 ppm) et de valider/raffiner le modèle PBPK existant pour ces concentrations ; et 2) de déterminer les concentrations d’éthanol atmosphérique provenant d’utilisation de SHA et de vernis et de prédire les niveaux d’éthanolémie découlant de leur utilisation. Les données toxicocinétiques récoltées chez des volontaires nous suggèrent qu’il est insuffisant de limiter au foie la clairance métabolique de l’éthanol lors d’exposition à de faibles niveaux d’éthanol, contrairement aux expositions à de plus forts niveaux. De plus, il a clairement été démontré qu’un effort physique léger (50 W) influençait à la hausse (2-3 fois) l’éthanolémie des volontaires exposés à 750 ppm. L’ajout au modèle PBPK d’une clairance métabolique de haute affinité et de faible capacité associée aux tissus richement perfusés a permis de simuler plus adéquatement la cinétique de l’éthanolémie pour des expositions à des concentrations inférieures à 1000 ppm. Des mesures de concentrations d’éthanol dans l’air inhalé générées lors d’utilisation de SHA et de vernis ont permis de simuler des expositions lors de l’utilisation de ces produits. Pour l’utilisation de 1,5 g et 3 g de SHA dans un local peu ventilé, des concentrations sanguines maximales (Cmax) de 0.383 et 0.366 mg.L-1 ont été respectivement simulées. Dans un local bien ventilé, les Cmax simulées étaient de 0.264 et 0.414 mg.L-1. Selon les simulations, une application de vernis résulterait en une Cmax respectivement de 0.719 mg.L-1 et de 0.729 mg.L-1, chez les hommes et femmes. Les Cmax sanguines d’éthanol estimées suites aux différentes simulations sont inférieures à la concentration toxique pour les humains (100 mg.L-1). Ainsi, de telles expositions ne semblent pas être un danger pour la santé. Les résultats de cette étude ont permis de mieux décrire et comprendre les processus d’élimination de l’éthanol à faibles doses et permettront de raffiner l’évaluation du risque associé à l’inhalation chronique de faibles niveaux d’éthanol pour la population, particulièrement chez les travailleurs.

Triple quadrupole mass spectrometers coupled with high performance liquid chromatography are workhorses in quantitative bioanalyses. It provides substantial benefits including reproducibility, sensitivity and selectivity for trace analysis. Selected Reaction Monitoring allows targeted assay development but data sets generated contain very limited information. Data mining and analysis of non-targeted high-resolution mass spectrometry profiles of biological samples offer the opportunity to perform more exhaustive assessments, including quantitative and qualitative analysis. The objectives of this study was to test method precision and accuracy, statistically compare bupivacaine drug concentration in real study samples and verify if high resolution and accurate mass data collected in scan mode can actually permit retrospective data analysis, more specifically, extract metabolite related information. The precision and accuracy data presented using both instruments provided equivalent results. Overall, the accuracy was ranging from 106.2 to 113.2% and the precision observed was from 1.0 to 3.7%. Statistical comparisons using a linear regression between both methods reveal a coefficient of determination (R2) of 0.9996 and a slope of 1.02 demonstrating a very strong correlation between both methods. Individual sample comparison showed differences from -4.5% to 1.6% well within the accepted analytical error. Moreover, post acquisition extracted ion chromatograms at m/z 233.1648 ± 5 ppm (M-56) and m/z 305.2224 ± 5 ppm (M+16) revealed the presence of desbutyl-bupivacaine and three distinct hydroxylated bupivacaine metabolites. Post acquisition analysis allowed us to produce semiquantitative evaluations of the concentration-time profiles for bupicavaine metabolites.

In recent years, we observed a significant increase of food fraud ranging from false label claims to the use of additives and fillers to increase profitability. Recently in 2013, horse and pig DNA were detected in beef products sold from several retailers. Mass spectrometry has become the workhorse in protein research and the detection of marker proteins could serve for both animal species and tissue authentication. Meat species authenticity will be performed using a well defined proteogenomic annotation, carefully chosen surrogate tryptic peptides and analysis using a hybrid quadrupole-Orbitrap mass spectrometer. Selected mammalian meat samples were homogenized, proteins were extracted and digested with trypsin. The samples were analyzed using a high-resolution mass spectrometer. The chromatography was achieved using a 30 minutes linear gradient along with a BioBasic C8 100 × 1 mm column at a flow rate of 75 µL/min. The mass spectrometer was operated in full-scan high resolution and accurate mass. MS/MS spectra were collected for selected proteotypic peptides. Muscular proteins were methodically analyzed in silico in order to generate tryptic peptide mass lists and theoretical MS/MS spectra. Following a comprehensive bottom-up proteomic analysis, we were able to detect and identify a proteotypic myoglobin tryptic peptide [120-134] for each species with observed m/z below 1.3 ppm compared to theoretical values. Moreover, proteotypic peptides from myosin-1, myosin-2 and -hemoglobin were also identified. This targeted method allowed a comprehensive meat speciation down to 1% (w/w) of undesired product.

La sclérose latérale amyotrophique est une maladie neurodégénérative fatale caractérisée par la dégénérescence progressive des neurones moteurs centraux et périphériques. L’un des premiers signes de la maladie est la dénervation de la jonction neuromusculaire (JNM). Les diverses unités motrices (UM) ne présentent toutefois pas la même vulnérabilité à la dénervation dans la SLA: les UM rapide fatigables sont en fait les plus vulnérables et les UM lentes sont les plus résistantes. Alors que des études précédentes ont démontré dans plusieurs modèles animaux de la SLA de nombreuses variations synaptiques, les découvertes ont été contradictoires. Par ailleurs, le type d’UM n’a pas été tenu en compte dans ces divers travaux. Nous avons donc émis l’hypothèse que la présence de la mutation SOD1 pourrait affecter différemment la transmission synaptique des UM, en accord avec leur vulnérabilité sélective. En effectuant des enregistrements électrophysiologiques et de l’immunohistochimie, nous avons étudié la transmission synaptique des différents types d’UM du muscle à contraction rapide Extensor Digitorum Longus (EDL; rapide fatigable (FF) MU) et du muscle à contraction lente Soleus (SOL; lente (S) and rapide fatigue-résistante (FR) MU) de la souris SOD1G37R et leur congénères WT. Pour identifier le type d’UM, un marquage par immunohistochimie des chaînes de myosine a été effectué. Un triple marquage de la JNM a également été effectué pour vérifier son intégrité aux différents stades de la maladie. À P160, dans la période asymptomatique de la maladie, alors qu’aucune altération morphologique n’était présente, l’activité évoquée était déjà altérée différemment en fonction des UM. Les JNMs FF mutantes ont démontré une diminution de l’amplitude des potentiels de plaque motrice (PPM) et du contenu quantique, alors que les JNMs lentes démontraient pratiquement le contraire. Les JNMs FR montraient quant à elles une force synaptique semblable au WT. À P380, dans la période présymtomatique, de nombreuses altérations morphologiques ont été observées dans le muscle EDL, incluant la dénervation complète, l’innervation partielle et les extensions du nerf. La transmission synaptique évoquée des UM FF étaient toujours réduites, de même que la fréquence des potentiels de plaque motrice miniatures. À P425, à l’apparition des premiers symptômes, l’activité synaptique des JNMs S était redevenue normale alors que les JNMs FR ont montré à ce moment une diminution du contenu quantique par rapport au contrôle. De manière surprenante, aucun changement du ratio de facilitation n’a été observé malgré les changements flagrants de la force synaptique. Ces résultats révèlent que la fonction de la JNM est modifiée différemment en fonction de la susceptibilité des UM dans l’ALS. Cette étude fournit des pistes pour une meilleure compréhension de la physiologie de la JNM durant la pathologie qui est cruciale au développement d’une thérapie adéquate ciblant la JNM dans la SLA.

Salicylaldehyde Schiff base of amino-methylated polystyrene has been developed as a novel reagent for the removal of Fe(III) from solutions. The selectivity of the metal ion uptake over a wide range of different concentrations of metal ion, effect of pH, ligand concentration and the influence of other foreign ions were studied. A very good selectivity was achieved for the removal of the ion. It was found that 0.01 g of the ligand was sufficient to achieve about 96% removal of the metal ion in terms of concentration (ppm) from a 30 ppm solution in acidic pH.

A set of six new polystyrene anchored metal complexes have been synthesized by the reaction of the metal salt with the polystyrene anchored Schiff base of vanillin. These complexes were characterized by elemental analyses, Fourier transform infrared spectroscopy, diffuse reflectance studies, thermal studies, and magnetic susceptibility measurements. The elemental analyses suggest a metal : ligand ratio of 1 : 2. The ligand is unidentate and coordinates through the azomethine nitrogen. The Mn(II), Fe(III), Co(II), Ni(II), and Cu(II) complexes are all paramagnetic while Zn(II) is diamagnetic. The Cu(II) complex is assigned a square planar structure, while Zn(II) is assigned a tetrahedral structure and Mn(II), Fe(III), Co(II), and Ni(II) are all assigned octahedral geometry. The thermal analyses were done on the ligand and its complexes to reveal their stability. Further, the application of the Schiff base as a chelating resin in ion removal studies was investigated. The polystyrene anchored Schiff base gave 96% efficiency in the removal of Ni(II) from a 20-ppm solution in 15 min, without any interference from ions such as Mn(II), Co(II), Fe(III), Cu(II), Zn(II), U(VI), Na , K , NH4 , Ca2 , Cl , Br , NO3 , NO2 ,and CH3CO2 . The major advantage is that the removal is achieved without altering the pH.

This paper reports the synthesis of a series of six new polystyrene anchored metal complexes of Co(II), Fe(III), Ni(II), Cu(II), Zn(II), and dioxouanium(VI) using the polystyrene anchored Schiff base of 2-nitrobenzaldehyde and the corresponding metal salts. The metal salts used were anhydrous FeCl3, CoCl2 Æ 6H2O, Ni(CH3COO)2 Æ 4H2O, Cu(CH3- COO)2 Æ H2O, Zn(CH3COO)2 Æ 2H2O, and UO2(CH3COO) Æ 2H2O. Physico chemical characterizations have been made from diffuse reflectance and vibrational spectra, elemental analysis, magnetic measurements, and TG studies. The elemental analysis suggest a 1:2 metal:ligand ratio when the complexation has carried out at 70 C for about 12 h reflux. The ligand is monodentate and coordinates through the azomethine nitrogen. The Fe(III), Co(II), Ni(II), and Cu(II) complexes are all paramagnetic whereas Zn(II) and U(VI) are diamagnetic. Zn(II) is assigned a tetrahedral structure, Cu(II) and Co(II) are assigned a square planar structure and Fe(III), Ni(II), and U(VI) are all assigned an octahedral structure. The polystyrene anchored ligand has been developed as an excellent reagent for the removal of Cu(II). Optimum conditions have been developed for the removal of metal ion from solutions by studying the effect of change of concentration of metal ion, ligand, effect of pH, time of reflux, and interference effect of other ions. It was found that within a span of 20 min it is possible to remove 90% of the metal ion from a 30 ppm metal ion solution in the pH range 4–5.5.

The MgAl2O4 ceramics were prepared by the conventional solid-state ceramic route and the dielectric properties studied in the microwave frequency region (3–13 GHz). The phase purity and crystal structure were identified using the X-ray diffraction technique. The MgAl2O4 spinel ceramics show interesting microwave dielectric properties (εr = 8.75, Qux f = 68 900 GHz (loss tangent = 0.00017 at 12.3 GHz), τf =−75 ppm/◦C). The MgAl2O4 has high negative τf, which precludes its immediate use in practical applications. Hence the microwave dielectric properties of MgAl2O4 spinels were tailored by adding different mole fractions of TiO2. The εr and Q factor of the mixed phases were increased with the molar addition of TiO2 into the spinel to form mixtures based on (1−x)MgAl2O4-xTiO2 (x = 0.0−1.0). For x = 0.25 in (1−x)MgAl2O4-xTiO2, the microwave quality factor reaches a maximum value of Qux f = 105 400 GHz (loss tangent = 0.00007 at 7.5 GHz) where εr and τf are 11.035 and −12 ppm/◦C, respectively. The microwave dielectric properties of the newly developed 0.75MgAl2O4-0.25TiO2 dielectric is superior to several commercially available low loss dielectric substrates.
