This thesis aims to present a color segmentation approach for traffic sign recognition based on LVQ neural networks. The RGB images were converted into HSV color space, and segmented using LVQ depending on the hue and saturation values of each pixel in the HSV color space. LVQ neural network was used to segment red, blue and yellow colors on the road and traffic signs to detect and recognize them. LVQ was effectively applied to 536 sampled images taken from different countries in different conditions with 89% accuracy and the execution time of each image among 31 images was calculated in between 0.726sec to 0.844sec. The method was tested in different environmental conditions and LVQ showed its capacity to reasonably segment color despite remarkable illumination differences. The results showed high robustness.

This paper aims to present three new methods for color detection and segmentation of road signs. The images are taken by a digital camera mounted in a car. The RGB images are converted into IHLS color space, and new methods are applied to extract the colors of the road signs under consideration. The methods are tested on hundreds of outdoor images in different light conditions, and they show high robustness. This project is part of the research taking place in Dalarna University / Sweden in the field of the ITS.

O carrapato Boophilus microplus é um ectoparasita hematófago que infesta os rebanhos bovinos de regiões tropicais e subtropicais, causando grande prejuízo à pecuária. O principal método de controle deste parasita baseia-se no uso de acaricidas, entretanto, o uso de vacinas tem sido estudado como um método de controle promissor. A Boophilus Yolk pro-Cathepsin (BYC) é uma aspártico proteinase presente no ovo do carrapato e envolvida na embriogênese que foi anteriormente testada como imunógeno vacinal. Neste estudo, o cDNA da BYC foi amplificado por PCR e clonado em dois vetores de expressão para produção de duas formas da proteína recombinante com cauda de histidina, rBYC e rBYC-Trx (fusionada com tioredoxina). As duas formas foram expressas em Escherichia coli na forma de corpúsculos de inclusão (CI) e comparadas quanto ao nível de expressão, solubilidade e rendimento na purificacão. Três agentes desnaturantes (N-lauroil sarcosina, hidrocloreto de guanidina e uréia) foram testados para solubilização dos CIs. Sarcosina foi o agente mais eficiente, solubilizando mais de 90 % de rBYC-Trx e rBYC. As duas proteínas recombinantes foram purificadas em cromatografia de afinidade por metal (Ni2+), sob condições desnaturantes. O rendimento na purificação da proteína solúvel foi de 84 % para r-BYC-Trx e 6 % para rBYC. As duas formas foram reconhecidas por soro de coelhos, camundongos e bovinos previamente imunizados com BYC nativa, demonstrando a existência de epítopos comuns entre a BYC nativa e as formas recombinantes expressas em E. coli. Para verificar o potencial vacinal da proteína recombinante, um grupo de bovinos Hereford foi imunizado com rBYC e desafiado com 20.000 larvas de B. microplus por animal. Os soros dos bovinos imunizados reconheceram a BYC nativa em ELISA e “Western blot”, com títulos entre 500 e 4.000. Os resultados do desafio mostraram uma proteção parcial contra a infestação, com 25 % de proteção global. O perfil de expressão de citocinas (IL-2, IL-4, IL-10, IFN-γ) foi verificado por RT-PCR, porém os resultados não permitiram identificar a polarização da resposta imune em Th1 ou Th2. Os resultados de imunoproteção obtidos com a BYC recombinante foram similares aos obtidos na imunização de bovinos com BYC nativa, indicando a possibilidade de uso da forma recombinante como imunógeno vacinal.

Computer vision is a field that uses techniques to acquire, process, analyze and understand images from the real world in order to produce numeric or symbolic information in the form of decisions [1]. This project aims to use computer vision to prepare an app to analyze a Madeira Wine and characterize it (identify its variety) by its color. Dry or sweet wines, young or old wines have a specific color. It uses techniques to compare histograms in order to analyze the images taken from a test sample inside a special container designed for this purpose. The color analysis from a wine sample using an image captured by a smartphone can be difficult. Many factors affect the captured image such as, light conditions, the background of the sample container due to the many positions the photo can be taken (different to capture facing a white wall or facing the floor for example). Using new technologies such as 3D printing it was possible to create a prototype that aims to control the effect of those external factors on the captured image. The results for this experiment are good indicators for future works. Although it’s necessary to do more tests, the first tests had a success rate of 80% to 90% of correct results. This report documents the development of this project and all the techniques and steps required to execute the tests.

Aim. Extrinsic compression of the popliteal artery and absence of surrounding anatomical abnormalities characterize the functional popliteal artery entrapment syndrome (PAES). The diagnosis is confirmed to individuals who have typical symptoms of popliteal entrapment and occlusion or important stenosis of the popliteal artery with color duplex sonography (CDS), magnetic resonance imaging (MRI) or arteriography during active plantar flexion-extension maneuvers. However, variable result findings in normal asymptomatic subjects have raised doubts as to the validity of these tests. The purpose of this study was to compare the frequency of popliteal artery compression in 2 groups of asymptomatic subjects, athletes and non-athletes.Methods. Forty-two individuals were studied. Twenty-one subjects were indoor soccer players, and 21 were sedentary individuals. Physical activity was evaluated through questionnaires, anthropometric measurements, and cardiopulmonary exercise test. Evaluation of popliteal artery compression was performed in lower limbs with CDS, ankle-brachial index (ABI) measurements and continuous wave Doppler of the posterior tibial artery.Results. The athletes studied fulfilled the criteria of high level of physical activity whereas sedentary subjects met the criteria of low level of activity. Popliteal artery compression was observed with CDS in 6 (14.2%) studied subjects; 2 of whom (4.7%) were athletes and 4 (9.5%) were non-athletes. This difference was not statistically significant (p=0.21). Doppler of the tibial arteries and ABI measurements gave good specificity and sensibility in the identification of popliteal artery compression.Conclusion. The frequency of popliteal artery compression during maneuvers in normal subjects was 14.2% irrespective of whether or not they performed regular physical activities. Both Doppler and ABI showed good agreement with CDS and should be considered in screening popliteal arteries in individuals suspected of PAES.

The yolk syncytial layer (YSL) has been regarded as one of the main obstacles for a successful cryopreservation of fish embryos. The purpose of this study was to identify and characterize the YSL in Prochilodus lineatus, a fish species found in southeastern Brazil and considered a very important fishery resource. Embryos were obtained through artificial breeding by hormonal induction. After fertilization, the eggs were incubated in vertical incubators with a controlled temperature (28 degrees C). Embryos were collected in several periods of development up to hatching and then fixed with 2% glutaralclehyde and 4% paraformaldehyde in 0.1 M sodium phosphate buffer (pH 7.3). Morphological analyses were carried out under either light, transmission or scanning electron microscopy. The formation of the YSL in P. lineatus embryos starts at the end of the cleavage stage (morula), mainly at the margin of the blastoderm, and develops along the embryo finally covering the entire yolk mass (late gastrula) and producing a distinct intermediate zone between the yolk and the endodermal cells. The YSL was characterized by the presence of microvilli on the contact region with the yolk endoderm. A cytoplasmic mass, full of mitochondria, vacuoles, ribosomes, endomembrane nets and euchromatic nuclei, indicated a high metabolic activity. This layer is shown as an interface between the yolk and the embryo cells that, besides sustaining and separating the yolk, acts as a structure that makes it available for the embryo. The structural analyses identified no possible barriers to cryoprotectant penetration.

The objective of this study was to determine whether replacing the egg yolk with soybean lecithin in the Botu-Crio (R) cryodiluent would maintain the fertility of cryopreserved stallion sperm. Two experiments were performed to evaluate cell freezability. In experiment 1, sperm from 15 stallions were frozen in Botu-Crio (R) (BC) or Botu-Crio (R) which contained 45 g/L soybean lecithin (BCLS45) in place of the egg yolk. In experiment 2, we compared different concentrations of soybean lecithin: 0, 10.0, 12.5, 15.0, 17.5 and 20.0 g/L (BC, BCLS10, BCLS12.5, BCLS17.5 and BCLS20, respectively). In experiment 1, sperm frozen in BC and BCLS45 exhibited similar (P > 0.05) percentages of total motile sperm (61% and 61%, respectively); progressively motile sperm (27% and 27%, respectively) and sperm with intact plasma membranes (IMP; 53% and 57%, respectively). Similarly, sperm frozen in BC or BC containing any concentration of soybean lecithin maintained similar (P > 0.05) percentages of total motile sperm (61-68%) and progressively motile sperm (27-31%). In the first fertility trial, we used cryopreserved semen from a single stallion was inseminated into mares. The semen from the sperm that were frozen in BC diluent resulted in a higher fertility rate (66%, 16/24) compared to the sperm that were frozen in BCLS45 diluent (17%, 5/29; P < 0.01). Similarly, in a second fertility trial, the mares that were inseminated with the sperm that were frozen in BC diluent exhibited a higher fertility rate (66%, 16/24) compared to the mares that were inseminated with the sperm that were frozen in BCLS20 (40%, 10/25; Pc 0.05). Finally, in a third trial, the sperm that were frozen in BC resulted in a higher fertility rate in mares (75%, 18/24) compared to the sperm that were frozen in BCLS10 (41%, 10/24; P < 0.05). Although replacing the egg yolk in the BC cryodiluent with soybean lecithin provided similar laboratory results for stallion sperm, after cryopreservation, the sperm that was frozen with soybean lecithin in the diluent correlated with lower fertility rates. Based on these results, we concluded that the use of BCLS can be used as an alternative diluent for cryopreserving stallion sperm. However, the resulting reduced fertility rate is a matter of concern. Further studies are necessary to clarify the reasons for this decrease in fertility and to determine the optimal lecithin concentration for diluents to freeze stallion sperm. (C) 2011 Elsevier B.V. The objective of this study was to evaluate the effect of different supplementation levels of Rhodocyclus gelatinosus biomass on the performance and pigmentation of broilers. Cobb broilers were raised in pens and were fed a sorghum-based diet until 35 days of age. At the 36th day of age, ninety-six birds were randomly distributed to four treatments consisting of experimental diets with different supplementation levels of Rhodocyclus gelatinosus biomass (0, 75, 150 and 300 ppm) added to the finisher basal diet, which were given for seven days. A completely randomized experimental design was used, with four treatments, four replicates and six broilers per parcel. Birds and diets were weighed at the 36th and at the 42nd day of age for performance analysis. There was no significant effect on broiler feed intake. Significant differences in weight gain and feed conversion were seen between the control group and the groups supplemented with 75 and 150 ppm. The treatment with the highest supplementation level and the control group had best weight gain and feed conversion. Color analysis showed that R. gelatinosus biomass supplementation resulted in more yellow breast skin and increased darkening and color purity in breast and thigh meat of broilers, but had no effect on lightness or color purity of breast and thigh skins.
