To determine which species and populations of Anopheles transmit malaria in any given situation, immunological assays for malaria sporozoite antigen can replace traditional microscopical examination of freshly dissected Anopheles. We developed a wicking assay for use with mosquitoes that identifies the presence or absence of specific peptide epitopes of circumsporozoite (CS) protein of Plasmodium falciparum and two strains of Plasmodium vivax (variants 210 and 247). The resulting assay (VecTest(TM) Malaria) is a rapid, one-step procedure using a 'dipstick' test strip capable of detecting and distinguishing between P. falciparum and P. vivax infections in mosquitoes. The objective of the present study was to test the efficacy, sensitivity, stability and field-user acceptability of this wicking dipstick assay. In collaboration with 16 test centres world-wide, we evaluated more than 40 000 units of this assay, comparing it to the standard CS ELISA. The 'VecTest(TM) Malaria' was found to show 92% sensitivity and 98.1% specificity, with 97.8% accuracy overall. In accelerated storage tests, the dipsticks remained stable for >15 weeks in dry conditions up to 45degreesC and in humid conditions up to 37degreesC. Evidently, this quick and easy dipstick test performs at an acceptable level of reliability and offers practical advantages for field workers needing to make rapid surveys of malaria vectors.

The Load-Unload Response Ratio (LURR) method is an intermediate-term earthquake prediction approach that has shown considerable promise. It involves calculating the ratio of a specified energy release measure during loading and unloading where loading and unloading periods are determined from the earth tide induced perturbations in the Coulomb Failure Stress on optimally oriented faults. In the lead-up to large earthquakes, high LURR values are frequently observed a few months or years prior to the event. These signals may have a similar origin to the observed accelerating seismic moment release (AMR) prior to many large earthquakes or may be due to critical sensitivity of the crust when a large earthquake is imminent. As a first step towards studying the underlying physical mechanism for the LURR observations, numerical studies are conducted using the particle based lattice solid model (LSM) to determine whether LURR observations can be reproduced. The model is initialized as a heterogeneous 2-D block made up of random-sized particles bonded by elastic-brittle links. The system is subjected to uniaxial compression from rigid driving plates on the upper and lower edges of the model. Experiments are conducted using both strain and stress control to load the plates. A sinusoidal stress perturbation is added to the gradual compressional loading to simulate loading and unloading cycles and LURR is calculated. The results reproduce signals similar to those observed in earthquake prediction practice with a high LURR value followed by a sudden drop prior to macroscopic failure of the sample. The results suggest that LURR provides a good predictor for catastrophic failure in elastic-brittle systems and motivate further research to study the underlying physical mechanisms and statistical properties of high LURR values. The results provide encouragement for earthquake prediction research and the use of advanced simulation models to probe the physics of earthquakes.

The ability of introduced organisms to invade undisturbed native habitats is a major concern in conservation biology and has resulted in a re-evaluation of the introduction of exotic biocontrol agents, especially of generalist predators. One such agent is Stratiolaelaps miles (Berlese), a predatory mite described from Italy, known from throughout the Holarctic, and apparently accidentally introduced to other areas of the world, including Australia. Initial investigations revealed that putative S. miles could be found in both disturbed and relatively pristine habitats in Queensland, Australia. However, analysis of the mitochondrial DNA of five populations showed most to be highly divergent genetically. Subsequent morphological analysis established two species groups: the lamington-group from cool-temperate to subtropical rainforests in Eastern Australia and the more eurytopic miles-group with a cosmopolitan distribution. We describe two new species from each of these complexes (Stratiolaelaps womersleyi, Stratiolaelaps lamington; Stratiolaelaps marilyn, Stratiolaelaps lorna, respectively), and resurrect Stratiolaelaps scimitus (Womersley), a species which often appears to have been confused with S. miles. Additionally, the large genetic distances among morphologically homogenous species in the miles-group suggest that the apparently cosmopolitan S. miles may be composed of a suite of cryptic species of potentially varying utility in biological control. Direct numerical simulation has been carried out for turbulent flow set up by a rotating cylinder with two backward-facing steps axisymmetrically mounted in the circumferential direction. This flow geometry creates a qualitatively similar flow pattern as observed near, a sudden, pipe expansion or a plane backward-facing step, characterized by flow separation and reattachment. A region of intense turbulence intensity and high wall-shear-stress fluctuations is formed in,the recirculating I region downstream of the step, where high mass-transfer capacity was also experimentally observed. Since, corrosion is frequently mass-transfer., controlled, our findings, put forward this apparatus as useful tool for future corrosion research.

Heat transfer levels have been investigated behind a rearward-facing step in a superorbital expansion tube. The heat transfer was measured along a flat plate and behind 2 and 3mm steps with the same length to step height ratio. Results were obtained with air as the test gas at speeds of 6.76kms(-1) and 9-60kms(-1) corresponding to stagnation enthalpies of 26MJ/kg and 48MJ/kg respectively. A laminar boundary layer was established on the flat plate and measured heat transfer levels were consistent with classical empirical correlations. In the case of flow behind a step, the measurements showed a gradual rise in heat transfer from the rear of the step to a plateau several step heights downstream for both flow conditions. Reattachment distance was estimated to be approximately 1.6 step heights downstream of the 2mm step at the low enthalpy condition through the use of flow visualisation.

Activation of macrophages with lipopolysaccharide (LPS) induces the rapid synthesis and secretion of proinflammatory cytokines, such as tumor necrosis factor (TNFalpha), for priming the immune response [1, 2]. TNFalpha plays a key role in inflammatory disease [3]; yet, little is known of the intracellular trafficking events leading to its secretion. In order to identify molecules involved in this secretory pathway, we asked whether any of the known trafficking proteins are regulated by LPS. We found that the levels of SNARE proteins were rapidly and significantly up- or downregulated during macrophage activation. A subset of t-SNAREs (Syntaxin 4/SNAP23/Munc18c) known to control regulated exocytosis in other cell types [4, 5] was substantially increased by LPS in a temporal pattern coinciding with peak TNFalpha secretion. Syntaxin 4 formed a complex with Munc18c at the cell surface of macrophages. Functional studies involving the introduction of Syntaxin 4 cDNA or peptides into macrophages implicate this t-SNARE in a rate-limiting step of TNFalpha secretion and in membrane ruffling during macrophage activation. We conclude that in macrophages, SNAREs are regulated in order to accommodate the rapid onset of cytokine secretion and for membrane traffic associated with the phenotypic changes of immune activation. This represents a novel regulatory role for SNAREs in regulated secretion and in macrophage-mediated host defense.

For dynamic closed loop control of a multilevel converter with a low pulse number (ratio of switching frequency to synthesized fundamental), natural sampled pulse-width modulation (PWM) is the best form of modulation. Natural sampling does not introduce distortion or a delayed response to the modulating signal. However previous natural sampled PWM implementations have generally been analog. For a modular multilevel converter, a digital implementation has advantages of accuracy and flexibility. Re-sampled uniform PWM is a novel digital modulation technique which approaches the performance of natural PWM. Both hardware and software implementations for a five level multilevel converter phase are presented, demonstrating the improvement over uniform PWM.

Religião e técnica, ainda que em momentos e pesos distintos na arquitetônica do pensamento de Jürgen Habermas, acabam por se constituir como temas dialogais referenciais na obra de tal autor, a partir dos quais estrutura seu pensamento em um crescendo teórico que se estende desde seus primeiros ensaios até seus mais recentes escritos. O objetivo desta dissertação é, a partir de uma revisão bibliográfica, analisar as aproximações encetadas por Habermas acerca destes pólos dialogais explicitando movimentos e aportes teóricos elaborados em cada etapa. Propomo-nos a trabalhar em um primeiro momento o conceito de técnica explicitando a influência herdada da Escola de Frankfurt, bem como suas reconstruções e propostas aos impactos de uma razão instrumentalizada na sociedade. O ponto culminante da análise habermasiana acerca do conhecimento tecnocientífico será o delineamento dos limites ético-normativos de tais conhecimentos e ao mesmo tempo a exposição das patologias e insuficiências de uma razão estratégica, ou unicamente relacionada a fins, o que culmina com sua proposta de um “naturalismo mitigado”. A partir deste horizonte teórico trabalharemos o conceito de religião e sua importância no marco teórico hodierno de Habermas. Por fim, analisamos sua aproximação ao conceito de religião e a importância deste movimento na guinada interpretativa acerca desta, culminando com sua reposição no interior do pensamento de tal autor.

Esta dissertação teve como objeto o Ensino Médio Integrado no Espírito Santo, sua trajetória e implantação a partir do Decreto 5.154/2004. O objetivo principal foi analisar a política do ensino médio integrado à educação profissional técnica de nível médio implementada no Espírito Santo. Investigar a educação pública pressupõe contextualizar a sociedade e as construções sociais que se projetam. O ensino médio no Brasil, enquanto etapa intermediária entre o ensino fundamental e a educação superior constitui-se de grande complexidade na estruturação de políticas públicas, no que tange à sua expansão qualitativa e quantitativa. Foram estabelecidos os seguintes objetivos específicos: identificar os pressupostos teóricos e pedagógicos contidos na Proposta Pedagógica da unidade escolar EEEFM Arnulpho Mattos; analisar a matriz curricular e a proposta pedagógica; relacionar o projeto pedagógico do curso com a prática educacional relatada nos encontros com os alunos; analisar a organização escolar e sua gestão política e pedagógica em face da proposta do EMI; relacionar a proposta do EMI, o princípio da politecnia e os relatos dos alunos; mapear a oferta do EMI no estado e o seu investimento financeiro. Nesse cenário, identificamos a relação entre trabalho e educação que contextualiza a relação da escola com a sociedade no sistema capitalista, em especial no ensino médio. Assim, nossa hipótese foi que esta política tem dificuldades de alcançar seus objetivos porque sua estrutura material segue a lógica da escola capitalista, assentada na perspectiva da Teoria do Capital Humano. A formação profissional no ensino médio é uma demanda social e histórica da classe trabalhadora, que necessita ingressar mais rápido no mercado de trabalho do que os filhos das elites brasileiras. A metodologia empregada foi uma pesquisa do tipo qualitativa, sendo um estudo de caso da EEEF Arnulpho Mattos. Os procedimentos metodológicos adotados para a investigação foram: observação, entrevistas semi-estruturadas, a análise documental, a revisão bibliográfica e os princípios metodológicos inscritos na sociologia da experiência (DUBET, 1994). Na articulação desses procedimentos, verificamos que a política do ensino médio integrado no ES teve, em seu início, um planejamento de implantação bem estruturado mas interrompido e, posteriormente, caracterizou-se pela descontinuidade das ações. Ademais, constatamos que os alunos ingressam e permanecem no curso sem compreenderem de fato o que significa a formação integrada entre a área geral e a técnica/profissional. Nos grupos de discussão percebemos que bastaram alguns encontros para que se pudesse estabelecer uma visão e relação diferenciadas com a experiência social do ensino médio integrado, principalmente por serem alunos trabalhadores. Acrescentamos às nossas conclusões que há iniciativas da escola acerca da integração entre as disciplinas de formação geral e técnicas, mas sem apoio ou orientação da unidade gestora, ou seja, a Sedu. A ampliação da oferta do EMI ocorreu no estado sem investimento na formação e valorização dos professores, sem concurso público e sem uma infra-estrutura necessária para a formação de jovens e adultos com capacidade de entender e de interferir no mundo do trabalho.

One of the current frontiers in the clinical management of Pectus Excavatum (PE) patients is the prediction of the surgical outcome prior to the intervention. This can be done through computerized simulation of the Nuss procedure, which requires an anatomically correct representation of the costal cartilage. To this end, we take advantage of the costal cartilage tubular structure to detect it through multi-scale vesselness filtering. This information is then used in an interactive 2D initialization procedure which uses anatomical maximum intensity projections of 3D vesselness feature images to efficiently initialize the 3D segmentation process. We identify the cartilage tissue centerlines in these projected 2D images using a livewire approach. We finally refine the 3D cartilage surface through region-based sparse field level-sets. We have tested the proposed algorithm in 6 noncontrast CT datasets from PE patients. A good segmentation performance was found against reference manual contouring, with an average Dice coefficient of 0.75±0.04 and an average mean surface distance of 1.69±0.30mm. The proposed method requires roughly 1 minute for the interactive initialization step, which can positively contribute to an extended use of this tool in clinical practice, since current manual delineation of the costal cartilage can take up to an hour.

“A memória não é um instrumento para explorar o passado, mas o seu teatro” (Walter Benjamin), cujos actores são pessoas comuns que recordam e recriam eventos passados através de diferentes formas de representação. Assim sendo, a memória é dinâmica e reconstrutiva e permite perpetuar a experiência, bem como criar e legitimar a identidade de cada um (Eber and Neal, 2001:37). O objectivo deste artigo é analisar os desenhos de Jack Sullivan sobre a comunidade das docas de Cardiff, conhecida como Butetown ou “Tiger Bay”, e a sua importância enquanto prática cultural na (re)construção de uma memória colectiva e imaginada. De que forma é que esta comunidade das docas é recordada e de que modo são as suas práticas quotidianas representadas através de desenhos? Quem e o que é que Sullivan representa? O que deixa na obscuridade? Como é que a memória individual se torna em memória colectiva? A este artigo subjaz a ideia de que os desenhos de Jack Sullivan, que estiveram na origem dos seus reconhecidos quadros compilados em Tramp Steamers, Seamen & Sailor Town, se apresentam como uma “forma de lembrança” de eventos passados, tanto para aqueles que viveram nesta área de Cardiff, como para aqueles que lhe eram espacial e temporalmente alheios. Estes desenhos constroem as memórias individuais e colectiva, tornando esta comunidade das docas acessível a todos.
