Chromosomal aberration (CA) assays have been widely used, not only to assess the genotoxic effects of chemical agents, but also to evaluate their action mechanisms on the genetic material of exposed organisms. This is of particular interest, since such analyses provide a better knowledge related to the action of these agents on DNA. Among test organisms, Allium cepa is an outstanding species due to its sensitivity and suitable chromosomal features, which are essential for studies on chromosomal damage or disturbances in cell cycle. The goal of the present study was to analyze the action mechanisms of chemical agents present in petroleum polluted waters. Therefore, CA assay was carried out in A. cepa meristematic cells exposed to the Guaeca river waters, located in the city of Sao Sebastiao, SP, Brazil, which had its waters impacted by an oil pipeline leak. Analyses of the aberration types showed clastogenic and aneugenic effects for the roots exposed to the polluted waters from Guaeca river, besides the induction of cell death. Probably all the observed effects were induced by the petroleum hydrocarbons derived from the oil leakage. (C) 2008 Elsevier B.V. All rights reserved.

A análise das alterações cromossômicas em leucemias tem uma aplicação direta no diagnóstico, prognóstico e tratamento dos pacientes. Além disso, permite o entendimento dos processos biológicos envolvidos na carcinogênese. Este trabalho apresenta os resultados do estudo cariotípico de 51 casos de diferentes tipos de leucemias. Os cromossomos foram obtidos através de cultura de células de sangue periférico, realizadas por 24 ou 48 horas, sem estimulação mitogênica. em 74% dos pacientes foram observadas anomalias cromossômicas clonais como translocações, deleções, monossomias e trissomias. Muitas alterações foram compatíveis com outras previamente descritas e outras não, como a translocação envolvendo os cromossomos 9 e 22, que origina o cromossomo Philadelphia e uma translocação complexa envolvendo os cromossomos 4, 7 e 11. Os resultados reforçam a importância da análise cromossômica em leucemia e seus benefícios para o paciente.

OBJETIVO: em decorrência dos questionamentos sobre o efeito deletério das radiações emitidas pelo campo eletromagnético (CEM) dos tipos ELF (extremely low frequency) e VLF (very low frequency) transmitidas pelos monitores de vídeo dos computadores (CRT), foi avaliada a freqüência de anomalias cromossômicas estruturais e a cinética do ciclo celular em indivíduos expostos por seu trabalho à radiação dos CRT. MÉTODOS: A pesquisa de aberrações cromossômicas foi realizada em 2.000 metáfases de primeira divisão celular obtidas de culturas de 48h de linfócitos de sangue venoso periférico de dez indivíduos expostos ao CRT (grupo E) e de dez controles (grupo C). A cinética do ciclo celular foi pesquisada pelos índices mitótico (IM) e de proliferação celular (IPC). RESULTADOS: A análise estatística evidenciou freqüências significativamente maiores de metáfases com anomalias cromossômicas (E=5,9%; C=3,7%) e anomalias/célula (E=0,066±0,026; C=0,040±0,026) nos indivíduos expostos aos CRTs. As alterações citogenéticas mais comuns foram as quebras cromatídicas, com freqüência de 0,034±0,016 no grupo E e de 0,016±0,015 no grupo C. As freqüências de IM e IPC não apresentaram diferenças significantes entre os grupos avaliados. CONCLUSÕES: Os resultados sugerem um efeito genotóxico do CEM emitido pelos CRTs devido à freqüência mais elevada de quebras cromatídicas, enfatizando a necessidade de haver um número maior de estudos com diferentes técnicas que vise a investigar a ação do CEM sobre o material genético.

JUSTIFICATIVA E OBJETIVOS: Neste estudo compararam-se médicos expostos aos gases anestésicos com indivíduos não expostos, para a investigação de alterações cromossômicas, verificação da possível interferência dos anestésicos inalatórios na cinética celular e avaliação do risco genotóxico associado à exposição ocupacional. MÉTODO: Foram realizadas culturas temporárias de linfócitos do sangue periférico para a obtenção de metáfases, necessárias para a análise de aberrações cromossômicas e trocas entre cromátides irmãs. RESULTADOS: A análise citogenética mostrou um aumento nas freqüências de aberrações cromossômicas por célula no grupo exposto, quando comparado ao grupo controle, denotando o efeito clastogênico desses compostos. Com relação ao parâmetro de trocas entre cromátides irmãs, os gases anestésicos não demonstraram efeito indutor. A comparação entre os índices mitótico e de proliferação celular também mostrou que não há diferenças significativas entre os grupos exposto e controle. CONCLUSÕES: Os resultados obtidos sugerem que os anestesiologistas podem representar um grupo de risco entre pessoas ocupacionalmente expostas e as condições de trabalho devem ser melhoradas.

To determine whether the venom of Apis mellifera can exert a radioprotective effect, by reducing the frequency of chromosome aberrations induced by radiation, five different experiments were performed on bone marrow cells of Wistar rats.Animals weighing about 100 g were injected intraperitoneally with different venom concentrations (1.0 or 0.5 mul) 1 or 24 h before, or 30 min after being submitted to 3 or 4 Gy of gamma radiation, and sacrificed 24 h after the last treatment. For each experiment in addition to the group of animals submitted to combined treatment (venom + radiation) and to their control, there was also one group treated with radiation only and another treated with venom only. A decrease in the frequency of chromosome aberrations, and fragments in particular, as well as in the number of cells with aberrations was observed in the experiments in which venom was administered 24 h before irradiation, and the effect was more marked at the higher venom concentration (1 mul/100 g weight).

Significant interindividual variations in health outcome may be caused by the inheritance of variant polymorphic genes, such as CYP2D6 and CYP2E1 for activation, and GSTM1 and GSTT1 for detoxification of chemicals. However. mechanistic studies linking the inheritance of predisposing genes with genotoxic effects towards cancer have yet to be systematically conducted. We have studied 54 lung cancer patients and 50 matched normal controls, who have been cigarette smokers, to elucidate the role of polymorphic genes in cancer. Our data indicates that the inheritance of unfavorable CYP2D6, CYP2E1, and GSTT1 genes is strongly correlated with the smoking-related lung cancer. For heavy cigarette smokers (> 30 pack-years), the smoking habit is the strongest predictor of lung cancer risk irrespective of the inheritance of unfavorable metabolizing genes. For moderate to light smokers (< 30 pack-years), the genetic predisposition plays on important role For the risk (odds ratio = 3.46; 95% CL = 0.46-40.2). Using a subgroup of the study population, we observed that cigarette smokers having the defective GST genes have significantly more chromosome aberrations as determined by the fluorescence-in-situ-hybridization (FISH) technique than smokers with the normal GST genes (P < 0.001). In conclusion, our study provides data to indicate that individuals who have inherited unfavorable metabolizing genes have increased body burden of toxicants to cause increased genetic damage and to have increased risk for cancer. Studies like ours can be used to understand the basis for interindividual variations in cancer outcome, to identify high risk individuals and to assess health risk. (C) 1997 Wiley Liss, Inc.

Several studies have suggested that dietary supplementation with antioxidants can influence the response to chemotherapy as well as the development of adverse side effects that result from treatment with antineoplastic agents. The emphasis of the present study was to investigate whether the administration of a single dose of oral glutamine had any protective effect against cisplatin-induced clastogenicity. Cisplatin was administered to Wistar rats either alone or after treatment with glutamine. The rats were treated with glutamine (300 mg/kg b.w.) by gavage 24 h before the administration of cisplatin (5 mg/kg b.w., i.p.) and then sacrificed 24h after treatment with cisplatin. Glutamine significantly reduced (by about 48%) the clastogenicity of cisplatin in rat bone marrow cells. The antioxidant action of glutamine presumably modulates the clastogenic action of cisplatin. A simple and sensitive method for determining atmospheric ammonia (NH3), using a hanging drop, is described. A colorimetric sensor is composed of two optical fibers and the source of monochromatic light implemented was a red light emitting diode (LED) (635 nm). Preliminary experiments were carried out in order to optimize the geometry of the sensor. These tests showed that the best signal absorbance was obtained using a 22 muL deionized water drop for sampling the gas and as addition of 4 muL of each of the reactants to form the blue dye (indophenol). Some important analytical parameters were also studied, including sampling time and flow rate. The analytical curve was constructed with a concentration range of 3-20 ppbv of gaseous NH3 standard. The detection limit reached was of ca 0.5 ppbv. It was observed that formaldehyde and diethylamine did not interfere. However, studies showed that hydrogen sulfide caused a negative interference of 20%, when present in the atmosphere in a concentration equal to that of NE3. The method considered here was shown to be easy to apply, making it possible to make a determination every 17 min.

The rate of chromatid breaks was studied in cows with a history of sub-fertility by means of a test based on measurement of the average of breaks induced in lymphocytes of peripheral blood cultures. Fourteen female specimens were divided into two groups: fertile and sub-fertile. Peripheral blood lymphocytes were cultured and prepared for cytogenetic analysis. Two types of culture were established for each animal to evaluate the response of peripheral blood lymphocyte cultures to the genotoxic effects of bleomycin. The first culture did not receive bleomycin treatment (spontaneous chromosome aberrations). Our results showed that median breaks per cell (b/c) (+/-semirange) for spontaneous culture of the fertile and sub-fertile animals and bleomycin sensitivity assay for fertile and sub-fertile animals were 0.00 +/- 0.06, 0.02 +/- 0.03, 0.08 +/- 0.05 and 0.22 +/- 0.09, respectively. There was no significant difference (P > 0.05) in the chromosomal breakage in lymphocytes not exposed to bleomycin; however, in comparing the number of chromatid breaks per cell in cultures treated with bleomycin, the sub-fertile group showed a significantly higher (P < 0.05) level than the fertile group. These findings have implications both for identifying cattle with less than optimum fertility as well as for providing potential avenues to study the origins of sub-fertility. (C) 2004 Elsevier B.V. The measurement of sulfur dioxide in air at the parts-per-billion level is described. The experimental arrangement consists of two optical fibers placed on opposite sides of a liquid droplet of malachite green solution. After light has been passed through the droplet, the transmitted light is measured by a referenced photodetection arrangement. The light used in this absorption process is from a monochromatic source (lambda(max) 625 nm). This arrangement permits the variation of color in the droplet to be measured. The sulfur dioxide in the sample is collected by the droplet; it reacts with malachite green resulting in a colorless dye. The decoloration of the solution is proportional to the concentration of sulfur dioxide sampled. The signal depends on the sample flow rate. The present technique is simple, inexpensive, and permits a fast and near real time measurement while consuming very little reagent, (C) 1999 Academic Press.
