Raf-1 activation is a complex process which involves plasma membrane recruitment, phosphorylation, protein-protein and lipid-protein interactions, We now show that PP1 and PP2A serine-threonine phosphatases also have a positive role in Ras dependent Raf-1 activation, General serine-threonine phosphatase inhibitors such sodium fluoride, or beta-glycerophosphate and sodium pyrophosphate, or specific PP1 and PP2A inhibitors including microcystin-LR, protein phosphatase 2A inhibitor I-1 or protein phosphatase inhibitor 2 all abrogate H-Ras and K-Ras dependent Raf-1 activation in vitro. A critical Raf-1 target residue for PP1 and PP2A is S259. Serine phosphatase inhibitors block the dephosphorylation of S259, which accompanies Raf-1 activation, and Ras dependent activation of mutant Raf259A is relatively resistant to serine phosphatase inhibitors. Sucrose gradient analysis demonstrates that serine phosphatase inhibition increases the total amount of 14-3-3 and Raf-1 associated with the plasma membrane and significantly alters the distribution of 14-3-3 and Raf-1 across different plasma membrane microdomains, These observations suggest that dephosphorylation of S259 is a critical early step in Ras dependent Raf-1 activation which facilitates 14-3-3 displacement. Inhibition of PP1 and PP2A therefore causes plasma membrane accumulation of Raf-1/14-3-3 complexes which cannot be activated.

The primary purpose of this experiment was to determine if left hand reaction time advantages in manual aiming result from a right hemisphere attentional advantage or an early right hemisphere role in movement preparation. Right-handed participants were required to either make rapid goal-directed movements to small targets or simply lift their hand upon target illumination. The amount of advance information about the target for a particular trial was manipulated by precuing a subset of potential targets prior to the reaction time interval. When participants were required to make aiming movements to targets in left space, the left hand enjoyed a reaction advantage that was not present for aiming in right space: or simple finger lifts. This advantage was independent of the amount or type of advance information provided by the precue. This finding supports the movement planning hypothesis. With respect to movement execution, participants completed their aiming movements more quickly when aiming with their right hand, particularly in right space. This right hand advantage in right space was due to the time required to decelerate the movement and to make feedback-based adjustments late in the movement trajectory. (C) 2001 Academic Press.

The control of movement is predicated upon a system of constraints of musculoskeletal and neural origin. The focus of the present study was upon the manner in which such constraints are adapted or superseded during the acquisition of motor skill. Individuals participated in five experimental sessions, ill which they attempted to produce abduction-adduction movements of the index finger in time with an auditory metronome. During each trial, the metronome frequency was increased in eight steps from an individually determined base frequency. Electromyographic (EMC) activity was recorded from first dorsal interosseous (FDI), first volar interosseous (FVI), flexor digitorum superficialis (FDS), and extensor digitorum communis (EDC) muscles. The movements produced on the final day of acquisition more accurately matched the required profile, and exhibited greater spatial and temporal stability, than those generated during initial performance. Tn the early stages of skill acquisition, an alternating pattern of activation in FDI and FVI was maintained, even at the highest frequencies. Tn contrast, as the frequency of movement was increased, activity in FDS and EDC was either tonic or intermittent. As learning proceeded, alterations in recruitment patterns were expressed primarily in the extrinsic muscles (EDC and FDS). These changes took the form of increases in the postural role of these muscles, shifts to phasic patterns of activation, or selective disengagement of these muscles. These findings suggest that there is considerable flexibility in the composition of muscle synergies, which is exploited by individuals during the acquisition of coordination.

Two putative promoters from Australian banana streak badnavirus (BSV) isolates were analysed for activity in different plant species. In transient expression systems the My (2105 bp) and Cv (1322 bp) fragments were both shown to have promoter activity in a wide range of plant species including monocots (maize, barley, banana, millet, wheat, sorghum), dicots (tobacco, canola, sunflower, Nicotiana benthamiana, tipu tree), gymnosperm (Pinus radiata) and fern (Nephrolepis cordifolia). Evaluation of the My and Cv promoters in transgenic sugarcane, banana and tobacco plants demonstrated that these promoters could drive high-level expression of either the green fluorescent protein (GFP) or the beta -glucuronidase (GUS) reporter gene (uidA) in vegetative plant cells. In transgenic sugarcane plants harbouring the Cv promoter, GFP expression levels were comparable or higher (up to 1.06% of total soluble leaf protein as GFP) than those of plants containing the maize ubiquitin promoter (up to 0.34% of total soluble leaf protein). GUS activities in transgenic in vitro-grown banana plants containing the My promoter were up to seven-fold stronger in leaf tissue and up to four-fold stronger in root and corm tissue than in plants harbouring the maize ubiquitin promoter. The Cv promoter showed activities that were similar to the maize ubiquitin promoter in in vitro-grown banana plants, but was significantly reduced in larger glasshouse-grown plants. In transgenic in vitro-grown tobacco plants, the My promoter reached activities close to those of the 35S promoter of cauliflower mosaic virus (CaMV), while the Cv promoter was about half as active as the CaMV 35S promoter. The BSV promoters for pregenomic RNA represent useful tools for the high-level expression of foreign genes in transgenic monocots.

The specification of the erythroid lineage from hematopoietic stem cells requires the expression and activity of lineage-specific transcription factors. One transcription factor family that has several members involved in hematopoiesis is the Kruppel-like factor (KLF) family [1]. For example, erythroid KLF (EKLF) regulates beta -globin expression during erythroid differentiation [2-6]. KLFs share a highly conserved zinc finger-based DNA binding domain (DBD) that mediates binding to CACCC-box and GC-rich sites, both of which are frequently found in the promoters of hematopoietic genes. Here, we identified a novel Xenopus KLF gene, neptune, which is highly expressed in the ventral blood island (VBI), cranial ganglia, and hatching and cement glands. neptune expression is induced in response to components of the BMP-4 signaling pathway in injected animal cap explants. Similar to its family member, EKLF, Neptune can bind CACCC-box and GC-rich DNA elements. We show that Neptune cooperates with the hematopoietic transcription factor XGATA-1 to enhance globin induction in animal cap explants. A fusion protein comprised of Neptune's DBD and the Drosophila engrailed repressor domain suppresses the induction of globin in ventral marginal zones and in animal caps. These studies demonstrate that Neptune is a positive regulator of primitive erythropoiesis in Xenopus.

Two hazard risk assessment matrices for the ranking of occupational health risks are described. The qualitative matrix uses qualitative measures of probability and consequence to determine risk assessment codes for hazard-disease combinations. A walk-through survey of an underground metalliferous mine and concentrator is used to demonstrate how the qualitative matrix can be applied to determine priorities for the control of occupational health hazards. The semi-quantitative matrix uses attributable risk as a quantitative measure of probability and uses qualitative measures of consequence. A practical application of this matrix is the determination of occupational health priorities using existing epidemiological studies. Calculated attributable risks from epidemiological studies of hazard-disease combinations in mining and minerals processing are used as examples. These historic response data do not reflect the risks associated with current exposures. A method using current exposure data, known exposure-response relationships and the semi-quantitative matrix is proposed for more accurate and current risk rankings.

The technique of permanently attaching interdigital transducers (IDT) to either flat or curved structural surfaces to excite single Lamb wave mode has demonstrated great potential for quantitative non-destructive evaluation and smart materials design, In this paper, the acoustic wave field in a composite laminated plate excited by an IDT is investigated. On the basis of discrete layer theory and a multiple integral transform method, an analytical-numerical approach is developed to evaluate the surface velocity response of the plate due to the IDTs excitation. In this approach, the frequency spectrum and wave number spectrum of the output of IDT are obtained directly. The corresponding time domain results are calculated by applying a standard inverse fast Fourier transformation technique. Numerical examples are presented to validate the developed method and show the ability of mode selection and isolation. A new effective way of transfer function estimation and interpretation is presented by considering the input wave number spectrum in addition to the commonly used input frequency spectrum. The new approach enables the simple physical evaluation of the influences of IDT geometrical features such as electrode finger widths and overall dimension and excitation signal properties on the input-output characteristics of IDT. Finally, considering the convenience of Mindlin plate wave theory in numerical computations as well as theoretical analysis, the validity is examined of using this approximate theory to design IDT for the excitation of the first and second anti-symmetric Lamb modes. Predicting plant leaf area production is required for modelling carbon balance and tiller dynamics in plant canopies. Plant leaf area production can be studied using a framework based on radiation intercepted, radiation use efficiency (RUE) and leaf area ratio (LAR) (ratio of leaf area to net above-ground biomass). The objective of this study was to test this framework for predicting leaf area production of sorghum during vegetative development by examining the stability of the contributing components over a large range of plant density. Four densities, varying from 2 to 16 plants m(-2), were implemented in a field experiment. Plants were either allowed to tiller or were maintained as uniculm by systematic tiller removal. In all cases, intercepted radiation was recorded daily and leaf area and shoot dry matter partitioning were quantified weekly at individual culm level. Up to anthesis, a unique relationship applied between fraction of intercepted radiation and leaf area index, and between shoot dry weight accumulation and amount of intercepted radiation, regardless of plant density. Partitioning of shoot assimilate between leaf, stem and head was also common across treatments up to anthesis, at both plant and culm levels. The relationship with thermal time (TT) from emergence of specific leaf area (SLA) and LAR of tillering plants did not change with plant density. In contrast, SLA of uniculm plants was appreciably lower under low-density conditions at any given TT from emergence. This was interpreted as a consequence of assimilate surplus arising from the inability of the plant to compensate by increasing the leaf area a culm could produce. It is argued that the stability of the extinction coefficient, RUE and plant LAR of tillering plants observed in these conditions provides a reliable way to predict leaf area production regardless of plant density. Many kinds of transcription factors and regulators play key roles in a variety of developmental processes. In the present survey, genes encoding proteins with conserved HMG-box, bZip domains, and some types of zinc finger motifs were surveyed in the completely sequenced genome of Ciona intestinalis. In the present analysis, 21 HMG-box-containing genes and 26 bZip genes were identified as well as four small groups of zinc finger genes in the Ciona genome. The results also showed that a less redundant set of genes is present in the Ciona genome compared with vertebrate genomes. In addition, cDNA clones for almost all genes identified have been cloned and distributed as a Ciona intestinalis Gene Collection Release I. The present comprehensive analysis therefore provides a means to study the role of these transcription factors in developmental processes of basal chordates.

The Wilms' tumour suppressor gene (WT1) encodes a zinc finger-containing nuclear protein essential for kidney and urogenital development. Initially considered a transcription factor, there is mounting evidence that WT1 has a role in post-transcriptional processing. Using the interspecies heterokaryon assay, we have demonstrated that WT1 can undergo nucleocytoplasmic shuttling. We have also mapped the region responsible for nuclear export to residues 182-324. Our data add further complexity to the role of WT1 in trancriptional and post-transcriptional regulation. Esta pesquisa de mestrado teve como principal objetivo investigar estratégias de cálculo mental, utilizadas por alunos de uma 5ª série/6º ano do ensino fundamental ao resolver cálculos de adição e subtração. Para atingir este objetivo procuramos responder aos questionamentos: Quais estratégias de cálculo mental, alunos da 5ª série/6º ano empregam na resolução de cálculos de adição e subtração? Que relações existem entre o tipo de cálculo envolvido e a estratégia adotada para resolvê-lo? Para respondermos a essas questões, seguimos uma metodologia de natureza qualitativa, configurada como estudo de caso do tipo etnográfico. O trabalho de campo foi desenvolvido em uma turma de 5ª série/6º ano do ensino fundamental de uma escola pública da rede estadual de ensino do município de Serra. A pesquisa aconteceu de maio a dezembro de 2013. Oito alunos resolveram uma atividade diagnóstica composta de quatro sequências de cálculos mentais, a saber, fatos fundamentais do número 5, do número 10, do número 20 e do número 100, dentre adições e subtrações próximas a esses resultados. Todos alunos participaram da etapa de entrevistas. Dos oito alunos, foram escolhidos dados de três que participaram de outras etapas da pesquisa. Os registros realizados pelos alunos na etapa de observação da turma, na etapa diagnóstica e na etapa de intervenção didática, as anotações no caderno de campo e algumas gravações em áudio serviram como fontes de coleta de dados. Utilizamos as estratégias identificadas por Beishuizen (1997), Klein e Beishuizen (1998), Thompson (1999, 2000) e Lucangeli et al. (2003), como categorias de análise. Através da análise de dados, constatamos que as escolhas das estratégias de cálculo mental pelos alunos variaram de acordo com o tipo de sequência de cálculos, a operação aritmética (adição ou subtração) e o estado emocional deles durante a atividade. Foi possível identificar o uso de duas estratégias combinadas, o algoritmo mental e estratégias de contagens nos dedos para grande parte dos cálculos. O uso do algoritmo mental mostrou-se um procedimento de grande sobrecarga mental e, em alguns cálculos de adição sem reserva, serviu apenas como apoio à visualização numérica, sendo executado pelo aluno da esquerda para a direita, semelhantemente à estratégia de decomposição numérica. Os dados deste estudo apontam para: (i) a necessidade de se trabalhar fatos numéricos fundamentais de adição e subtração via cálculo mental de maneira sistemática em sala de aula; (ii) a necessidade de se ensinar estratégias autênticas de cálculo mental para que os alunos não se tornem dependentes de estratégias como contagens e algoritmo mental, que são mais difíceis de serem executadas com êxito; (iii) a importância de entrevistar, individualmente, os alunos a fim de compreender e avaliar o desenvolvimento destes em tarefas de cálculo mental.

A fonação reversa é a produção de voz durante a inspiração, realizada espontaneamente em situações como o suspiro. OBJETIVO: Realizar uma revisão da literatura, descrevendo achados relacionados à utilização da fonação reversa na prática clínica, à anatomofisiologia de sua produção e seus efeitos no trato vocal, e às indicações e contra-indicações da técnica para os distúrbios e para o aperfeiçoamento da voz. RESULTADOS: Foram encontrados relatos de mudanças significativas no trato vocal durante a produção da fonação reversa, como o relaxamento dos ventrículos, o afastamento das pregas ventriculares, o aumento da freqüência fundamental, o movimento inverso da onda mucosa, além da facilitação do estudo dinâmico da laringe, quando associada à endoscopia, o que possibilita melhor definição da localização das lesões nas camadas da lâmina própria da mucosa das pregas vocais. CONCLUSÃO: Existem poucos estudos que descrevem o comportamento laríngeo durante a fonação reversa e, para que essa técnica utilizada de forma mais precisa e objetiva, acredita-se que ainda devem ser realizados estudos que visem a comprovar sua eficácia na prática clínica.

O objetivo deste trabalho foi avaliar a qualidade dos bulbos de duas variedades de cebola colhidos em três estádios de maturação da planta. Bulbos das variedades Texas Grano e Baia Periforme foram colhidos em 2006 no campo experimental da Universidade Federal de Viçosa, de plantas com 10% de estalo natural, 10% de estalo induzido e 50% de estalo natural. Na colheita foram avaliados a massa da matéria fresca total, o diâmetro, a cor (L*, a* e b*), o teor de massa da matéria seca, os sólidos solúveis totais e o teor relativo de água da polpa e das películas externas do bulbo. No cultivar Texas Grano, a massa da matéria fresca total, o diâmetro e os teores de sólidos solúveis totais foram maiores nos bulbos colhidos com 10 e 50% de estalo natural. O teor relativo de água das películas externas de bulbos variedade Texas Grano foi maior somente quando colhidos com 10% de estalo natural, o que representa bulbos com películas mais úmidas. Bulbos colhidos com 10% de estalo natural e 10% de estalo induzido tiveram valores de L* maiores do que aqueles colhidos com 50% de estalo natural, indicando que a antecipação da colheita reduz a intensidade da cor das películas. O menor valor de b* para a película do bulbo da Texas Grano, colhido com 10% estalo natural, decorre do amarelo menos intenso da casca. No cultivar Baia Periforme, os índices de maturação não influenciaram a massa da matéria fresca total, o diâmetro, teor de massa da matéria seca, o teor de sólidos solúveis totais e os índices de cor a* e b*. O estádio de maturação da planta, na colheita, não influencia o tamanho e o teor de sólidos solúveis dos bulbos na variedade Baia Periforme. Para a Texas Grano, bulbos maiores, com películas mais amareladas e menor teor de umidade são obtidos quando a colheita é realizada com 50% de estalo natural.

The search for higher profitability in wheat crop with cost reduction technologies that may promote sustainability is an important matter in Brazilian agriculture. This study evaluated the profitability of no-tilled wheat, reducing nitrogen topdressing doses with the cultivation of green manure before the wheat crop. The experiment was carried out in Selvíria (MS), Brazil, in 2009/10. The experiment was arranged in a randomized block design with 36 treatments in splitplots and four replicates. The plots were formed by six types of green manure: Cajanus cajan L. BRS Mandarin, Crotalaria juncea L., Pennisetum americanum L. BRS 1501, fallow area and mixed cropping of Pennisetum americanum L. + Cajanus cajan L. and Pennisetum americanum L. + crotalaria which provided straw for no-tilled wheat in the winter, following the rice crop in the summer. The subplots were formed by six levels of topdressing nitrogen (0, 25, 50, 75, 100 and 125 kg N ha-1) using urea as a nitrogen source. The wheat grown after green manure in the previous winter crop, with no nitrogen topdressing and a rate of 25 kg ha-1 N, had more frequently production costs above the gross income. Wheat production cost after the mixed cropping Pennisetum americanum L. + Cajanus cajan L. and Pennisetum americanum L. + Crotalaria juncea L. from the previous winter crop, combined with nitrogen rates of 50 and 75 kg N ha-1, provided better profitability compared with the other green manures evaluated.
