In the present work, the antitumor effect of fastuosain, a cysteine proteinase from Bromelia fastuosa, was investigated. In the intravenous model of lung colonization in C57Bl/6 mice, fastuosain and bromelain injected intraperitoneally were protective, and very few nodules of B16F10-Nex2 melanoma cells were detected. Tumor cells treated with fastuosain showed reduced expression of CD44 and decreased invasion through Matrigel, lost their cytoplasmic extensions and substrate adherence, and became round and detached, forming strongly bound cell clusters in suspension. Peritoneal cells recruited and activated by fastuosain treatment ( mainly monocytic cells and lymphocytes) migrated to the lung, where pulmonary melanoma metastases grew. Adoptive transference of peritoneal cells recruited by fastuosain had no protective effect against lung metastases in recipient mice. Treatment of green fluorescent protein - chimeric animals with fastuosain did not change the number of cells that migrated to the lung, compared to PBS-injected control mice, but the number of positive major histocompatibility complex class II cells increased with fastuosain treatment. Murine antibodies against fastuosain, bromelain, and cathepsins B and L cross-reacted in ELISA and recognized surface and cytoplasmic components expressed on B16F10-Nex2 cells. Anti-fastuosain antibodies were cytotoxic/lytic to B16F10-Nex2 cells. Antitumor effects of fastuosain involve mainly the direct effect of the enzyme and elicitation of protective antibodies.

Melanose, caused by Diaporthe citri, produces reddish brown lesions on the fruit, leaves, and twigs of citrus trees, and greatly reduces the marketability of fresh fruit. Most of the inoculum is produced in pycnidia on dead twigs in the tree canopy, which exude large numbers of conidia in slimy masses. In this study, detached twigs inoculated with conidia were readily colonized and produced large numbers of pycnidia within 30 to 40 days when they were soaked 3 to 4 h on alternate days. Conidial production was measured by wetting twigs in a rain tower periodically and collecting the conidia in the runoff water. Production began after 80 days and continued for nearly 300 days. In other experiments, production of mature pycnidia on detached twigs was greatest at 94 to 100% relative humidity (RH) and at 28 degrees C. Low RH and temperature, however, favored survival of conidia in exuded masses on twigs. In the field, colonization of detached twigs by D. citri was high in rainy season, moderate in spring and early fall, and minimal in late fall and winter. Twig colonization was positively related to the number of rain days and average temperature, but not to total rainfall. In another experiment, inoculated twigs placed in the tree canopy developed pycnidia and then produced conidial masses for about 200 days. D. citri is a serious pathogen, but a weak parasite, that survives primarily by colonization and reproduction on dead twigs.

The habitat of the mycelial saprobic form of Paracoccidio ides brasiliensis, which produces the infectious propagula, has not been determined and has proven difficult for mycologists to describe. The fungus has been rarely isolated from the environment, the disease has a prolonged latency period and no outbreaks have been reported. These facts have precluded the adoption of preventive measures to avoid infection. The confirmation of natural infections in nine-banded armadillos (Dasypus novemcinctus) with P. brasiliensis, in high frequency and wide geographic distribution, has opened new avenues for the study and understanding of its ecology. Armadillos belong to the order Xenarthra, which has existed in South America ever since the Paleocene Era (65 million years ago), when the South American subcontinent was still a detached land, before the consolidation of what is now known as the American continent. on the other hand, strong molecular evidence suggests that P. brasiliensis and other dimorphic pathogenic fungi - such as Blastomyces dermatitidis, Coccidioides immitis and Histoplasma capsulatum - belong to the family Onygenaceae sensu Into (order Onygenales, Ascomycota), which appeared around 150 million years ago.P. brasiliensis ecology and relation to its human host are probably linked to the fungal evolutionary past, especially its long coexistence with and adaptation to animal hosts other than Homo sapiens, of earlier origin. Instead of being a blind alley, the meaning of parasitism for dimorphic pathogenic fungi should be considered as an open two-way avenue, in which the fungus may return to the environment, therefore contributing to preserve its teleomorphic (sexual) and anamorphic (asexual) forms in a defined and protected natural habitat. This paper discusses the dynamic behaviour of a nonlinear two degree-of-freedom system consisting of a harmonically excited linear oscillator weakly connected to a nonlinear attachment having linear and cubic restoring forces. The effects of the system parameters on the shape of the frequency-response curve are investigated, in particular those yielding the appearance and disappearance of outer and inner detached resonance curves. In contrast to the case when the linear stiffness of the attachment is zero, it is found that multivaluedness occurs at low frequencies as the resonant peak bends to the right. It is also found that as the coefficient of the linear term increases, the range of parameters yielding detached curves reduces. Compared to the case when the attached system has no linear stiffness term, this range of parameters corresponds to smaller values of the damping and nonlinear coefficients. Approximate analytical expressions for the jump-up and jump-down frequencies of the system under investigation are also derived. (C) 2011 Elsevier Ltd. All rights reserved.

The present work aimed to evaluate the endectocide activity of a new injectable long-action formulation, containing ivermectin (IVM) and abamectin (ABA). In each one of the four experiments performed, the following groups were formed: group I: 2.25% IVM (450 mu g/kg) + 1.25% ABA (250 mu g/kg), group II: 3.15% IVM (630 mu g/kg) and group III: control. Eighteen bovine naturally infected by gastrointestinal nematoda were selected for anthelmintic evaluation and necropsied on posttreatment day (PTD) 14 to estimate the total parasitic burden. For the Rhipicephalus (Boophilus) microplus field trial, 30 bovine were selected by means of counts of semi-engorged R. (B.) microplus and the therapeutic and residual efficacy evaluated by tick counts on PTDs 1, 3, 7, 14, 21, 28, 35, 42, 49, 56, 63, 70, 77, 84 and 91. In the stall test, 15 calves were artificially infested with 5000 R. (B.) microplus (Mozzo strain) larvae three times a week and daily collections of all the engorged female ticks detached from each calf were performed until the PTD 80. Forty bovine naturally infected with Dermatobia hominis larvae were selected and the number of larvae was counted by visual and tactile inspection on PTDs 3, 7, 14, 28, 35, 49, 63, 77, 91 and 105. In this trial, a formulation containing 1% doramectin (200 mu g/kg) was also used. IVM + ABA formulation and 3.15% IVM eliminated four of the eight species of nematode identified. The anthelmintic efficacy of the avermectins association against Haemonchus placei, Cooperia spatulata and C. punctata was 89.64%, 98.84% and 97.69%, while 3.15% IVM achieved 30.98%, 84.79% and 75.56%, respectively. The two formulations evaluated showed reduced acaricide action on the PTD I and 3, reaching high efficacy percentages from PTD 14 onward. The IVM + ABA showed efficacy above 95% in the period between PTDs 21 and 49. In the stall test, it observed no difference (P > 0.05) between the two formulations regarding the R. (B.) microplus counts during the entire evaluation period. IVM + ABA reduced the number of ticks from the PTD 1 to 77 (P < 0.05) and 3.15% IVM reduced (P < 0.05) the tick number from PTD 4 up to PTD 80. The three endectocides showed no difference (P > 0.05) regarding the number of D. hominis larvae and prevented this parasite reestablishment until PTD 105. These results indicate that the IVM + ABA association showed higher anthelmintic activity and similar efficacy against arthropods to the formulation containing 3.15% IVM. (C) 2008 Elsevier B.V. Erythrocytes and environmental interferences on sickle cell anaemia Sickle cell anaemia runs na extremely variable clinical course At one end of spectrum, it is characterized by a crippling haemolitic anaemia, interspersed with severe exacerbations, or crises, yet it may be an extremely mild disorder, which is found only by chance on rotine haematological examination. The reasons are only partly understood for these remarkable differences in phenotypic expression of what appears to be the same genetic defect: they include the level of Hb Fetal, coinheritance of the alpha thalassaemia and of other genetic variantsthat has influence as genetic modulation in sickle cell anaemia. However, other genetics abnormalities of erythrocytes: G-6PD deficiency, spherocytosis and deficiencies of anti-oxidant enzymes(SOD, GPx and Catalase) probably interfereon the clinical course of sickle cell anaemia. The haplotypes of the chromosome (Bantu, Benin, Camaroon and Arab-Indian) bearing the sickle gene is associated with assorted haematological and clinical features that are likely, at least in part, to be mediated throgh effects on Hb Fetal concentration. Beyond these factors characterizes as erythrocytes interferents, there are the environmental interferents. Between environmental interferents become detached the socio-economic and cultural situation of each patient. These aspects have influence on the life of affected individuals including social interactions, family relations, peer interaction, intimate relationships, education, enployment, violence, spiritual attitudes and navigating complexities of the health care system, providers and their ancillary functions. As a result of this article it is proposed a protocol of laboratorial management of sickle cell syndrome with detach to sickle cell anaemia.

Rhizoctonia solani causes pre- and post-emergence damping-off, root and hypocotyl rot and foliar blight in soybean. Foliar blight has resulted in yield losses of 31-60% in north and northeast Brazil. The aim of this study was to characterize isolates of R. solani associated with soybean in Brazil. Among 73 Rhizoctonia isolates examined, six were binucleate and 67 were multinucleate. The multinucleate isolates were characterized according to hyphal anastomosis reaction, mycelial growth rate, thiamine requirement, sclerotia production, and RAPD molecular markers. Four isolates that caused hypocotyl rot belonged to AG-4 and using RAPD analysis they grouped together with the HGI subgroup. Another isolate that caused root and hypocotyl rots was thiamine auxotrophic, grew at 35 °C, and belonged to AG-2-2 IIIB. All 62 isolates that caused foliar blight belonged to AG-1 IA. RAPD analysis of R. solani AG-1 IA soybean isolates showed high genetic similarity to a tester strain of AG-1 IA, confirming their classification. The teleomorph of R. solani, Thanatephorus cucumeris was produced in vitro by one AG-1 IA isolate from soybean. The AG-4 and AG-2-2 IIIB isolates caused damping-off and root and hypocotyl rots of soybean seedlings cv. 'FT-Cristalina', under greenhouse conditions. The AG-2-2 IIIB isolate caused large lesions on the cortex tissue, that was distinct from the symptoms caused by AG-4 isolates. The AG-1 IA isolates caused foliar blight in adult soybean plants cv. 'Xingu' under the greenhouse and also in a detached-leaf assay.

It was objectified to quantify the initial growth of varieties of castor bean with boron fertilization. Had been tested to cultivate they AL-Guarani, Mirante 10 and Nordestina to the doses of 0; 0.25; 0.50; 0.75 and 1.0 mg kg -1 of B in controlled environment of greenhouse in the Sector of Cafeicultura of the Department of Agriculture of the UFLA. The characteristics valued were: height of plants, length of roots, leaf number, diameter of stem, mass of dry substance (MS) of leafs, of stem, the root and total. To cultivate Mirante 10 it was the one that got minor growth of the system of root. For leaf number to cultivate AL-Guarani it was superior to Mirante 10 and Nordestina. Also to cultivate better AL-Guarani if it detached how much to the production of total mass of MS, while to cultivate Mirante 10 produced more mass of MS than Nordestina.

This paper discusses the dynamic behaviour of a nonlinear two degree-of-freedom system consisting of a harmonically excited linear oscillator weakly connected to a nonlinear attachment that behaves as a hardening Duffing oscillator. A system which behaves in this way could be a shaker (linear system) driving a nonlinear isolator. The mass of the nonlinear system is taken to be much less than that in the linear system and thus the nonlinear system has little effect on the dynamics of the linear system. Of particular interest is the situation when the linear natural frequency of the nonlinear system is less than the natural frequency of the linear system such that the frequency response curve of the nonlinear system bends to higher frequencies and thus interacts with the resonance frequency of the linear system. It is shown that for some values of the system parameters a complicated frequency response curve for the nonlinear system can occur; closed detached curves can appear as a part of the overall amplitude-frequency response. The reason why these detached curves appear is presented and approximate analytical expressions for the jump-up and jump-down frequencies of the system under investigation are given.
Biomass and yield of peanut grown on tropical soil amended with sewage sludge contaminated with lead

Application of sewage sludge with high lead (Pb) contents may pollute soils and contaminate crops. The objective of this work was to evaluate peanut responses to application of sewage sludge with varying Pb contents in order to supply phosphorus (P) to the plant. A greenhouse experiment was carried out with peanut grown on soil sample from a medium-textured Haplustox. Treatments were arranged in 3 × 2 + 2 factorial scheme, replicated three times, distributed in randomized block design, and consisted of: three Pb rates applied to soil with sewage sludge (3, 21, and 42 mg kg-1) × two times of sewage sludge application (30 days before peanut sowing and at the day of the sowing) + mineral fertilization + control (without sewage sludge and mineral fertilization). Sewage sludge was efficient to supply P to peanut. Sewage sludge containing high rates of Pb, when applied, did not harm biomass and yield of the plant, but increased HCl-extractable Pb in soil and Pb content in shoot, roots, and pod husks. Increase of Pb content in pod husks may represent contamination risk of kernels and their products with fragments from husks detached during manipulation or industrial processing of peanuts. © 2012 Fábio Camilotti et al.

It has been demonstrated that histamine interferes with the recruitment, formation and activity of osteoclasts via H1- and H2-receptors. Cimetidine is a H2-receptor antagonist used for treatment of gastric ulcers that seems to prevent bone resorption. In this study, a possible cimetidine interference was investigated in the number of alveolar bone osteoclasts. The incidence of osteoclast apoptosis and immunoexpression of RANKL (receptor activator of nuclear factor κB ligand) was also evaluated. Adult male rats were treated with 100mg kg-1 of cimetidine for 50days (CimG); the sham group (SG) received saline. Maxillary fragments containing the first molars and alveolar bone were fixed, decalcified and embedded in paraffin. The sections were stained by H&E or submitted to tartrate-resistant acid phosphatase (TRAP) method. TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling) method and immunohistochemical reactions for detecting caspase-3 and RANKL were performed. The number of TRAP-positive osteoclasts, the frequency of apoptotic osteoclasts and the numerical density of RANKL-positive cells were obtained. Osteoclast death by apoptosis was confirmed by transmission electron microscopy (TEM). In CimG, TRAP-positive osteoclasts with TUNEL-positive nuclei and caspase-3-immunolabeled osteoclasts were found. A significant reduction in the number of TRAP-positive osteoclasts and a high frequency of apoptotic osteoclasts were observed in CimG. Under TEM, detached osteoclasts from the bone surface showed typical features of apoptosis. Moreover, a significant reduction in the numerical density of RANKL-positive cells was observed in CimG. The significant reduction in the number of osteoclasts may be due to cimetidine-induced osteoclast apoptosis. However, RANKL immunoexpression reduction also suggests a possible interference of cimetidine treatment in the osteoclastogenesis. © 2012 The Authors Journal of Anatomy © 2012 Anatomical Society.

The objective of this work was to evaluate the agronomic performance and grain quality, especially the grain technological characteristics in the common bean genotypes, cultivated in winter-spring season. The experiment was carried out in Jaboticabal-SP, in a Rhodic Hapludox. The experimental design was a randomized block with 17 treatments (genotypes) with three replications. The plots had been composed for 17 carioca common bean group genotypes (Pérola, BRS Cometa, BRS Pontal, IPR Juriti, CNFC 10703, CNFC 10713, CNFC 10716, CNFC 10721, CNFC 10729, CNFC 10733, CNFC 10742, CNFC 10753, CNFC 10757, CNFC 10758, CNFC 10762, CNFC 10763 and CNFC 10813). BRS Pontal and CNFC 10716 showed simultaneously the higher number of per plant and grain yield. The technological characteristics more expressive was verified in CNFC 10703, CNFC 10713, CNFC 10758, CNFC 10813 and CNFC 10716 genotypes, principally about the protein content, cooking time and grain hydration capacity. The CNFC 10716 genotype detached as promissory, because their elevated agronomic performance and satisfactory technological characteristics.

We explored the interaction between radiation of different wavelength and jasmonic acid (JA) or brassinosteroids (BR) on leaf senescence-induced oxidative stress. Three approaches were used: 1) jasmonic acid insensitive1-1 (jai1-1) and brassinosteroid-deficient [dumpy (dpy)] mutants were treated with red (R) or far-red (FR) radiation; 2) phytochromedeficient aurea (au) and high pigment-1 (hp-1) (radiation exaggerated response) mutants were treated with methyl jasmonate (MeJA) or epibrassinolide (epiBL); and 3) double mutants au jai1-1 and au dpy were produced. Leaf chlorophyll content, lipid peroxidation, and antioxidant enzyme activities were determined. After senescence induction in detached leaves, we verified that the patterns of chlorophyll degradation of hormonal and photomorphogenic mutants were not significantly different in comparison with original cv. Micro-Tom (MT). Moreover, there was no significant change in lipid peroxidation measured as malondialdehyde (MDA) production, as well as catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) activities in the hormonal mutants. Exogenous BR increased CAT and APX activities in MT, au, and hp-1. As concerns the double mutants, severe reduction in H2O2 production which was not accompanied by changes in MDA content, and CAT and APX activities was observed during senescence in au dpy. The results suggest that JA and BR do not participate in light signaling pathway during leaf senescence-induced oxidative stress. © 2013 Springer Science+Business Media Dordrecht.
