The anti-allergic active fractionation of hexane extracts of the leaves and stems of Anchietia salutaris,ar. martiana (family Violaceae) nas performed by monitoring their activities with an in vitro bioassay system measuring the inhibitory effects on induced histamine release from guinea pig lung cells. Three known pentacyclic triterpenes (friedelin, alpha-amyrin, beta-amyrin) were isolated, but these compounds were inactive. Aliphatic hydrocarbons and methyl esters of fatty acids (palmitic, oleic, linoleic, linolenic acids) were detected in active fractions. All compounds isolated were detected for the first time in this medicinal plant.

O presente trabalho visou estabelecer uma comparação entre composição de cachaças produzidas por Saccharomyces cerevisiae (Sc) e estirpes de leveduras selvagens [Pichia silvicola (Ps), Pichia anomala 1 (Pa1), Pichia anomala 2 (Pa2) e Dekkera bruxelensis (Db)], isoladas em destilarias da região de Jaboticabal-SP. Os componentes secundários da fração denominada coração foram determinados por cromatografia gasosa. Os níveis dos componentes secundários foram influenciados pelo pH dos respectivos vinhos, os quais dependem da estirpe de levedura empregada no processo fermentativo. A Saccharomyces cerevisiae apresentou valores ligeiramente superiores de componentes secundários, enquanto as estirpes selvagens produziram maiores teores de álcoois superiores. As estirpes selvagens de leveduras mostraram-se adequadas para obtenção de uma cachaça de boa qualidade.

Muitos fungos termofílicos são conhecidos como sendo produtores de lipases e o potencial dessas enzimas nas reações de esterificação (reação inversa à hidrólise), há muito tem sido reconhecida. O aspecto do micélio e diversas observações microscópicas foram efetuadas em Thermomyces lanuginosus cultivado em Placas de Petri com meios de culturas YpSs (extrato de levedura + amido + agar) e mais quatro variações na fonte de carbono neste meio. As matérias-primas usadas e avaliadas foram pentaeritritol, ácido oléico e dioleato de pentaeritritol como possíveis fontes de carbono disponíveis ao microrganismo são comumente encontradas na indústria química como intermediários clássicos na síntese de diversos ésteres. Com o crescimento de Thermomyces lanuginosus em todos os meios propostos e a manutenção da capacidade de reprodução mostramos ser esse fungo altamente promissor em futuros trabalhos de biocatálise com microrganismos vivos.

From the acetone and ethanol extracts of the tubercula of Aristolochia pubescens, two diterpene esters of aristolochic acids were isolated, together with 23 known compounds. 1,3-Diols are frequently involved in biologically important compounds and, therefore, determination of the stereochemistry of these structural elements, in particular those in acyclic systems, has been one of the focuses of attention in natural products chemistry. The modified Mosher's method, commonly used for the determination of the absolute configuration of secondary alcohols, was applied to determine the absolute configuration of 1,3-diols with their di-MTPA esters. Several epimeric pairs of syn- and anti-1,3-diols with known absolute configurations were converted to the corresponding di-MTPA esters and the Delta delta values were then calculated. For the acyclic syn-1,3-diols, the Delta delta values were systematically arranged as predicted from the basic concept of the modified Mosher's method, demonstrating that the method is valid for these compounds. In contrast, the Delta delta values were irregularly arranged for the acyclic anti-1,3-diols and, accordingly, this method is not valid for these cases. These results are complementary to those of the previously reported CD exciton chirality method and, hence, the combined use of the modified Mosher's method and the CD exciton chirality method can determine the absolute configuration of the acyclic 1,3-diols. Also, this method is successfully applicable to cyclic 1,3-diols irrespective of their relative stereochemistry. (C) 2002 Wiley-lass, Inc.

The voltammetric reduction of acetaldehyde was studied in 0.1 M LiOH: LiCl (60: 40 v/v). Welldefined waves can be seen at -1.77 and -1.60 V with the use of hanging mercury and glassy carbon electrodes. Acetaldehyde was shown to react at room temperature with the 2,4-dinitrophenylhydrazine and the product exhibited a differential pulse voltammetric peak at -0.90V, which was well separated from the peaks of the derivative. This allowed the indirect determination of acetaldehyde in the presence of 0.1 M ethanol/tetrabutylammonium perchlorate after 10 min of reaction. Calibration graphs were obtained for 1.00 x 10(-6)-1.00 x 10(-4) M of acetaldehyde. The detection limit is 8.14 x 10(-7) M. The method has been applied satisfactorily to the determination of total aldehyde in fuel ethanol samples without any pretreatment.

Although it is generally accepted that osteoclasts breakdown and resorb bone matrix, the possibility that they may also be able to engulf apoptotic osteoblasts/ lining cells and/or osteocytes remains controversial. Apoptosis of osteoblasts/ lining cells and/or osteocytes and interactions between these cells and osteoclasts are extremely rapid events that are difficult to observe in viva. A suitable in viva model for studying these events is the alveolar bone of young rats because it is continuously. Thus, sections of aldehyde fixed alveolar undergoing intense resorption/remodeling bone of young rats were stained by the combined terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) method and the tartrate-resistant acid phosphatase (TRAP) method for the simultaneous visualization of apoptotic cells and osteoclasts in the same section. The combined TUNEL and TRAP reactions, in the same section, greatly facilitated visualization of relationship between osteoclasts and apoptotic bone cells during alveolar bone remodeling. Our results showed that several TRAP-positive osteoclasts exhibited large vacuoles containing TUNEL positive apoptotic structures, probably derived from osteoblasts/lining cells and/or osteocytes. These results support the idea that alveolar bone osteoclasts are able to internalize dying apoptotic bone cells.
