Histone deacetylase inhibitors (HDACIs) interfere with the epigenetic process of histone acetylation and are known to have analgesic properties in models of chronic inflammatory pain. The aim of this study was to determine whether these compounds could also affect neuropathic pain. Different class I HDACIs were delivered intrathecally into rat spinal cord in models of traumatic nerve injury and antiretroviral drug-induced peripheral neuropathy (stavudine, d4T). Mechanical and thermal hypersensitivity was attenuated by 40% to 50% as a result of HDACI treatment, but only if started before any insult. The drugs globally increased histone acetylation in the spinal cord, but appeared to have no measurable effects in relevant dorsal root ganglia in this treatment paradigm, suggesting that any potential mechanism should be sought in the central nervous system. Microarray analysis of dorsal cord RNA revealed the signature of the specific compound used (MS-275) and suggested that its main effect was mediated through HDAC1. Taken together, these data support a role for histone acetylation in the emergence of neuropathic pain.

The objective of the present work was to induce somatic embryogenesis from zygotic embryos of Passiflora cincinnata Masters. Zygotic embryos formed calli on media with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 4.5 mu M benzyladenine (BA) after 30 days of in vitro culture. A concentration of 18.1 mu M 2,4-D resulted in the largest number of somatic embryos. Embryogenic calli were yellowish and friable, forming whitish proembryogenic masses. Morphologically, embryogenic cells were small and had large nuclei and dense cytoplasm, whereas non-embryogenic cells were elongated, with small nuclei and less dense cytoplasm. Calli cultured under white light on basal Murashige and Skoog`s medium with activated charcoal produced embryos in all developmental stages. There were differences among the treatments, with some leading to the production of calli with embryos and some only to callus formation. Some abnormalities were associated with somatic embryos, including fused axes, fused cotyledons and polycotyledonary embryos. Production of secondary somatic embryos occurred in the first cycle of primary embryo development. Secondary embryos differentiated from the surface of the protodermal layer of primary embryos with intense cell proliferation, successive mitotic divisions in the initial phase of embryoid development, and a vascular system formed with no connection to the parental tissue. This secondary embryogenic system of P. cincinnata is characterized by intense proliferation and maintenance of embryogenic competence after successive subcultures. This reproducible protocol opens new prospects for massive propagation and is an alternative to the current organogenesis-based transformation protocol.

The present volume is the fruit of a research initiative on Access to Knowledge begun in 2004 by Yochai Benkler, Eddan Katz, and myself. Access to Knowledge is both a social movement and an approach to international and domestic policy. In the present era of globalization, intellectual property and information and communications technology are major determinants of wealth and power. The principle of access to knowledge argues that we best serve both human rights and economic development through policies that make knowledge, knowledge-creating tools, and nowledgeembedded goods as widely available as possible for decentralized innovation and use. Open technological standards, a balanced approach to intellectual property rights, and expansion of an open telecommunications infrastructure enable ordinary people around the world to benefit from the technological advances of the information age and allow them to generate a vibrant, participatory and democratic culture. Law plays a crucial role in securing access to knowledge, determining whether knowledge and knowledge goods are shared widely for the benefit of all, or controlled and monopolized for the benefit of a few.

Dietary modifications may significantly reduce cardiovascular disease (CVD) risk factors, including cholesterol and atherosclerosis. The present study addressed the effects of the crude extract from the pulp fruit of Tamarindus indica L. on lipid serum levels and early atherosclerotic lesions in hypercholesterolemic hamsters in vivo, and the extract's antioxidant action, in vitro. Animals were fed on either chow or atherogenic diet during 10 weeks and concomitantly received either water or T indica L. extract for drinking. Treatment of hypercholesterolemic hamsters with the T. indica pulp fruit extract (5%) led to a decrease in the levels of serum total cholesterol (50%), non-HDL cholesterol (73%) and triglyceride (60%), and to an increase of high-density lipoprotein (HDL) cholesterol levels (61%). In vitro, the extract presented radical scavenging ability, as assessed by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and superoxide radicals assays, and led to decreased lipid peroxidation in serum, as assessed by the thiobarbituric acid reactive substances (TBARS) assay. In vivo, the extract improved the efficiency of the antioxidant defense system, as assessed by the superoxide dismutase, catalase and glutathione peroxidase activities. Together these results indicate the potential of tamarind extracts in diminishing the risk of atherosclerosis development in humans. (c) 2005 Elsevier Ltd. All rights reserved.

Guazuma ulmifolia is as popular reforestation tree all over Latin America. It is characteristic of the initial stages of the secondary sucession and presents potential utility in the restoring of degraded areas. There is no information about fruit, seed and seedling morphology, which is of fundamental importance for identification, extraction, management and seed germination as well as for the characterization of post-seminal development and normal seedling pattern. To obtain such information, external fruit, and external and internal seed structures were studied considering shape, size, micropile and embryo localization; and tegumentar structures. All stages of this work were conduced in the Universidade Estadual Paulista (UNESP), Campus of Jaboticabal city. The fruits were collected in a mixed plantation in Jaboticabal city, State of São Paulo, Brazil. For the bio metric study eight repetitions of ten fruits and eight repetitions of 100 seeds were utilized. For seed internal traits study, 50 seeds were drenched in a distiled water, cut, and observed with a scanning electron microscope and a stereomicroscope, For post-seminal study ten repetitions of seven seeds were scarificated chemically with sulphuric acid during 50 min, and placed to germinate ina culture medium, at 30 degrees C, and eight hours of photoperiod. We found elipsoid, woody, indehiscent, pentacarpelar fruits, with a mean lenght of 22.61 mm (diameter 24.88 mm) and 64.0 seeds per fruit. Seed shape varies, mean length is 3.07 mm (width of 2.36 mm). The seed is bitegumented, tegmic, with a continuous, axial and curved embryo. The germination is epigeal and the seedlings are fanerocotiledoneus. Drawings of all stages are included.

The effect of boron (B) on cotton growth and fruit shedding may be due not only to physiological or biochemical effects, but also to vascular tissue malformation. This experiment investigated petiole and floral peduncle anatomical alterations and growth of cotton supplied with deficient and sufficient B in nutrient solution. Cotton (Gossypium hirsutum cv. 'Delta Opal') plants were grown in solutions containing 0, 1.5, 3.0, 4.5, and 6.0 mu mol L-1 of B from 22 to 36 d after plant emergence (DAPE). From 36 to 51 DAPE, B was omitted from the nutrient solution. Petioles from young leaves and floral bud peduncles (first position of the first sympodial) were sampled and the cross-section anatomy observed under an optical microscope. The number of vascular bundles of the petiole was decreased in B-deficient plants and the xylem was disorganized. Phloem elements in the peduncle vascular cylinder of B-deficient plants did not show clear differentiation. The few xylem elements that were formed were also disorganized. Modifications caused by B deficiency may have impaired B and photosynthate translocation into new cotton growth. Boron accumulation in the shoot of B-deficient plants suggested that there was some B translocation within the plant. It could be inferred that cotton growth would be impaired by the decrease in carbohydrate translocation rather than by B deficiency in the tissue alone.

Eutherian mammals share a common ancestor that evolved into two main placental types, i.e., hemotrophic (e.g., human and mouse) and histiotrophic (e.g., farm animals), which differ in invasiveness. Pregnancies initiated with assisted reproductive techniques (ART) in farm animals are at increased risk of failure; these losses were associated with placental defects, perhaps due to altered gene expression. Developmentally regulated genes in the placenta seem highly phylogenetically conserved, whereas those expressed later in pregnancy are more species-specific. To elucidate differences between hemotrophic and epitheliochorial placentae, gene expression data were compiled from microarray studies of bovine placental tissues at various stages of pregnancy. Moreover, an in silico subtractive library was constructed based on homology of bovine genes to the database of zebrafish - a nonplacental vertebrate. In addition, the list of placental preferentially expressed genes for the human and mouse were collected using bioinformatics tools (Tissue-specific Gene Expression and Regulation [TiGER] - for humans, and tissue-specific genes database (TiSGeD) - for mice and humans). Humans, mice, and cattle shared 93 genes expressed in their placentae. Most of these were related to immune function (based on analysis of gene ontology). Cattle and women shared expression of 23 genes, mostly related to hormonal activity, whereas mice and women shared 16 genes (primarily sexual differentiation and glycoprotein biology). Because the number of genes expressed by the placentae of both cattle and mice were similar (based on cluster analysis), we concluded that both cattle and mice were suitable models to study the biology of the human placenta. Postbloom fruit drop (PFD) of citrus caused by Colletotrichum acutatum produces orange-brown lesions on petals and induces the abscission of young fruitlets and the retention of the calyces. Despite the fact that C. acutatum is not highly sensitive to benomyl in culture, this fungicide provides good control of the disease under field conditions. This study was undertaken to determine the effect of benomyl on various stages of disease development to understand the basis for its effectiveness in the field. We found that benomyl at 1.0 μg/ml reduced colony area of C. acutatum by about 75% and completely inhibited growth of C. gloeosporioides. Benomyl did not prevent conidial germination even at 100 μg/ml, but reduced germ tube elongation at 10 and 100 μg/ml. When benomyl was applied to flower clusters on screen-house-grown plants before inoculation, disease severity was greatly reduced. Applications at 24 and 48 h, but not at 72 h, after inoculation reduced PFD severity. Application of benomyl to symptomatic petals not bearing conidia did not prevent or reduce production of inoculum. Application to petals bearing conidia reduced viability of these fungal propagules by only about 50%. The viability of appressoria on mature leaves was not affected by benomyl application. Even when appressoria on mature leaves were stimulated to germinate by treatment with flower extracts, subsequent application of benomyl did not reduce propagule numbers below original levels. Benomyl appears to act by preventing infection and early development of the fungus in petals. However, once symptoms have developed, this fungicide has only minimal effects on further disease development and spread.

Brazilian Myrtaceae comprises several genera of trees and shrubs used for ornamental and fruit production. In addition to the well known Guava, Pitanga and Jaboticaba, other species could be used for fruitculture, due the value and quality of their fruits and adaptation to some climate conditions mainly the subtropical one. Nine species of Eugenia were evaluated at Jaboticabal, located at 48° W and 21° S in São Paulo state in a germplasm bank. The average rain by year is 1431 mm and the temperature 22,2° C at an altitude of 575 m. The species are Eugenia klozschiana Berg. (Pero-do-campo), E. stipitata Mc Vaugh (Araça-boi), E. tomentosa Camb. (Cabeludinha), E. dysentherica DC. (Cagaita), E. brasiliensis Berg. (Grumixama), E.pitanga (Pitanga-anã), E. luchsnathiana Berg. (Pitomba), E. uvalha Camb. (Uvaia) and E. involucrata DC. (Cereja-do-rio-grande). The evaluations comprised tree development, fruit quality and leaf and flower morphological studies. The main results are: the trees of Pera-do-campo and Pitanga-anã are small shrubs of 1 to 2 m height, Araça-boi and Cabeludinha are small trees, 3 to 5 m high, and the other species are tall trees, with 5 to 10 m height. The species adapted well to the subtropical conditions, except for Araça-boi, which is native to the Amazonian region and exhibited a severe fungus disease infection. In relation to fruit quality, all the species had edible fruit, some were sweet and juicy, Cabeludinha, Grumixama, Pitomba, Cereja-do-rio grande and Pitanga-anã, while others had high acidity (Araça-boi, Pera-do-campo, Cagaita and Uvaia and were more suitable for processing. Simple, single leaves were characteristic of all species, but with different sizes and shapes., With the addition of color, smell and other characteristics, leaf size and shape were useful for comparative classification. Flower components and structure are described also.

Flavonoids have been widely incorporated into cosmetic and dermatological formulations, affording benefits such as antioxidant action, improved skin tone and fewer lines and wrinkles. Brazil has a huge biodiversity, with one of the richest flora in the world, and existing studies justify the quest for greater research efforts in this area. The cajazeira (Spondias lutea L.), a plant of the Anacardiaceae family from tropical America, is widely disseminated in Brazil. This plant was chosen because of the presence of flavonoids that exhibit antioxidant activity. The purpose of this research was to develop a stable topical formulation containing Spondias lutea extract with the aim of preventing skin diseases caused by UV radiation. Hydro ethanolic extract of Spondias lutea fruit was prepared and assayed for its the flavonoids content. The antioxidant activity was estimated by DPPH and superoxide assay. The physicochemical stability and skin permeation of the cream containing 8% (w/w) of extract were assessed. The results showed that the S. lutea extract possessed antioxidant activity, and that it is possible to obtain a stable cosmetic containing the extract, which is able to penetrate the skin. Thus, it is possible to use this extract to produce an anti-aging cosmetic.

Objectives were to evaluate the role of canonical WNT signaling in development of the preimplantation embryo. Signaling was activated with 2-Amino-4-(3,4-(methylenedioxy)benzylamino)-6-(3-methoxyphenyl)pyrimidine (AMBMP) and inhibited with Dickkopf-related protein 1 (DKK1). Treatment of bovine embryos with AMBMP at day 5 after insemination decreased development to the blastocyst stage at day 7 and reduced numbers of trophectoderm and inner cell mass cells. At high concentrations, AMBMP caused disorganization of the inner cell mass. DKK1 blocked actions of AMBMP but did not affect development in the absence of AMBMP. Examination of gene expression in day 6 morulae by microarray revealed expression of 16 WNT genes and other genes involved in WNT signaling; differences in relative expression were confirmed by PCR for 7 genes. In conclusion, the preimplantation embryo possesses a functional WNT signaling system and activation of the canonical pathway can inhibit embryonic development.
