We used a network of 20 carbon dioxide- and octenol-supplemented light traps to sample adult mosquitoes throughout Russell Island in southern Moreton Bay, south-east Queensland. Between February and April 2001, an estimated 1365 564 adult female mosquitoes were collected. In contrast to an average catch of 9754 female mosquitoes per trap night on Russell Island, reference traps set on Macleay Island and on the mainland returned average catches of 3172 and 222, respectively. On Russell Island, Ochlerotatus vigilax (Skuse), Coquillettidia linealis (Skuse), Culex annulirostris Skuse and Verrallina funerea (Theobald), known or suspected vectors of Ross River (RR) and/or Barmah Forest (BF) viruses, comprised 89.6% of the 25 taxa collected. When the spatial distributions of the above species were mapped and analysed using local spatial statistics, all were found to be present in highest numbers towards the southern end of the island during most of the 7 weeks. This indicated the presence of more suitable adult harbourage sites and/or suboptimal larval control efficacy. As immature stages and the breeding habitat of Cq. linealis are as yet undescribed, this species in particular presents a considerable impediment to proposed development scenarios. The method presented here of mapping the numbers of mosquitoes throughout a local government area allows specific areas that have high vector numbers to be defined.

Coquillettidia linealis is a severe pest on some of the Moreton Bay islands in Queensland, Australia, but little is known of its breeding habitats and biology. Because of its high abundance and its association with Ross River (RR) and Barmah Forest (BF) viruses by field isolation, its vector competence was evaluated in the laboratory by feeding dilutions of both viruses in blood. For RR, Cq. linealis was of comparable efficiency to Ochlerotatus vigilax (Skuse), recognised as being a major vector. The extracellular loop 3 (ECL3) of the mammalian gonadotropin-releasing hormone receptor (GnRH-R) contains an acidic amino acid (Glu(301) in the mouse GnRH-R,) that confers agonist selectivity for Are in mammalian GnRH. It is proposed that a specific conformation of ECL3 is necessary to orientate the carboxyl side chain of the acidic residue for interaction with Arg(8) of GnRH, which is supported by decreased affinity for Arg(8) GnRH but not Gln(8) GnRH when an adjacent Pro is mutated to Ala. To probe the structural contribution of the loop domain to the proposed presentation of the carboxyl side chain, we synthesized a model peptide (CGPEMLNRVSEPGC) representing residues 293-302 of mouse ECL3, where Cys and Gly residues are added symmetrically at the N and C termini, respectively, allowing the introduction of a disulfide bridge to simulate the distances at which the ECL3 is tethered to the transmembrane domains 6 and 7 of the receptor. The ability of the ECL3 peptide to bind GnRH with low affinity was demonstrated by its inhibition of GnRH stimulation of inositol phosphate production in cells expressing the GnRH-R. The CD bands of the ECL3 peptides exhibited a superposition of predominantly unordered structure and partial contributions from beta-sheet structure. Likewise, the analysis of the amide I and amide III bands from micro-Raman and FT Raman experiments revealed mainly unordered conformations of the cyclic and of the linear peptide. NMR data demonstrated the presence of a beta-hairpin among an ensemble of largely disordered structures in the cyclic peptide. The location of the turn linking the two strands of the hairpin was assigned to the three central residues L-296, N-297, and R-298. A small population of structured species among an ensemble of predominantly random coil conformation suggests that the unliganded receptor represents a variety of structural conformers, some of which have the potential to make contacts with the ligand. We propose a mechanism of receptor activation whereby binding of the agonist to the inactive receptor state induces and stabilizes a particular structural state of the loop domain, leading to further conformational rearrangements across the transmembrane domain and signal propagating interaction with G proteins. Interaction of the Glu(301) of the receptor with Arg(8) of GnRH induces a folded configuration of the ligand. Our proposal thus suggests that conformational changes of both ligand and receptor result from this interaction.

A large number of herbaceous and woody plants from tropical woodland, savanna, and monsoon forest were analysed to determine the impact of environmental factors (nutrient and water availability, fire) and biological factors (microbial associations, systematics) on plant delta(15)N values. Foliar delta(15)N values of herbaceous and woody species were not related to growth form or phenology, but a strong relationship existed between mycorrhizal status and plant delta(15)N. In woodland and savanna, woody species with ectomycorrhizal (ECM) associations and putative N-2-fixing species with ECM/arbuscular (AM) associations had lowest foliar delta(15)N values (1.0-0.6parts per thousand), AM species had mostly intermediate delta(15)N values (average +0.6parts per thousand), while non-mycorrhizal Proteaceae had highest delta(15)N values (+2.9 to +4.1parts per thousand). Similar differences in foliar delta(15)N were observed between AM (average 0.1 and 0.2parts per thousand) and non-mycorrhizal (average +0.8 and +0.3parts per thousand) herbaceous species in woodland and savanna. Leguminous savanna species had significantly higher leaf N contents (1.8-2.5% N) than non-fixing species (0.9-1.2% N) indicating substantial N acquisition via N-2 fixation. Monsoon forest species had similar leaf N contents (average 2.4% N) and positive delta(15)N values (+0.9 to +2.4parts per thousand). Soil nitrification and plant NO3- use was substantially higher in monsoon forest than in woodland or savanna. In the studied communities, higher soil N content and nitrification rates were associated with more positive soil delta(15)N and plant delta(15)N. In support of this notion, Ficus, a high NO3- using taxa associated with NO3- rich sites in the savanna, had the highest delta(15)N values of all AM species in the savanna. delta(15)N of xylem sap was examined as a tool for studying plant delta(15)N relations. delta(15)N of xylem sap varied seasonally and between differently aged Acacia and other savanna species. Plants from annually burnt savanna had significantly higher delta(15)N values compared to plants from less frequently burnt savanna, suggesting that foliar N-15 natural abundance could be used as marker for assessing historic fire regimes. Australian woodland and savanna species had low leaf delta(15)N and N content compared to species from equivalent African communities indicating that Australian biota are the more N depauperate. The largest differences in leaf delta(15)N occurred between the dominant ECM Australian and African savanna (miombo) species, which were depleted and enriched in N-15, respectively. While the depleted delta(15)N of Australian ECM species are similar to those of previous reports on ECM species in natural plant communities, the N-15-enriched delta(15)N of African ECM species represent an anomaly.

The volume of the primary (PCS) and secondary (SCS) circulatory system in the Atlantic cod Gadus morhua was determined using a modified dye dilution technique. Cod (N=10) were chronically cannulated in the second afferent branchial artery with PE-50 tubing. Evans Blue dye was bound to harvested fish plasma at a concentration of 1 mg dye ml(-1) plasma, and injected at a concentration of 1 mg kg(-1) body mass. Serial sampling from the cannula produced a dye dilution curve, which could be described by a double exponential decay equation. Curve analysis enabled the calculation of the primary circulatory and total distribution volume. The difference between these volumes is assumed to be the volume of the SCS. From the dilution curve, it was also possible to calculate flow rates between and within the systems. The results of these experiments suggest a plasma volume in the PCS of 3.42+/-0.89 ml 100 g(-1) body mass, and in the SCS of 1.68+/-0.35 ml 100 g(-1) body mass (mean +/- S.D.) or approximately 50% that of the PCS. Flow rates to the SCS were calculated as 2.7% of the resting cardiac output. There was an allometric relationship between body mass and blood volumes. Increasing condition factor showed a tendency towards smaller blood volumes of the PCS, expressed as percentage body mass, but this was not evident for the volume of the SCS.

Forest fires are suggested as a potential and significant source of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/Fs), even though no studies to date provide sufficient evidence to confirm forest fires as a source of PCDD/Fs. Recent investigations in Gueensland, Australia have identified a widespread contamination of PCDDs (in particular OND) in soils and sediments in the coastal region from an unknown source of PCDD/Fs. Queensland is predominately rural; it has few known anthropogenic sources of PCDD/Fs, whereas forest fires are a frequent occurrence. This study was conducted to assess forest fires as a potential source of the unknown PCDD/F contamination in Queensland. A combustion experiment was designed to assess the overall mass of PCDD/Fs before and after a simulated forest fire. The results from this study did not identify an increase in Sigma-PCDD/Fs or OCDD after the combustion process. However, specific non-2,3,7,8 substituted lower chlorinated PCDD/Fs were elevated after the combustion process, suggesting formation from a precursor. The results from this study indicate that forest fires are unlikely to be the source of the unknown PCDD contamination in Gueensland, rather they are a key mechanism for the redistribution of PCDD/Fs from existing sources and precursors.

Cariniana estrellensis (Raddi.) Kuntze e C. legalis (Mart.) Kuntze são arbóreas nativas do Brasil que, além de possuírem alto poder econômico, são objeto de interesse em programas de recuperação de áreas degradadas e em plantios comerciais. A escassez de informações relacionadas ao desempenho ecofisiológico dessas espécies em condições ambientais estressantes dificultam o manejo e conservação das mesmas. Dessa forma, o presente estudo objetivou avaliar a ecofisiologia das espécies em um gradiente de irradiância, por meio de dois experimentos. No experimento 1, plantas de C. estrellensis com 12 meses de idade foram submetidas a quatro tratamentos: 40%, 50%, 70% e 100% de irradiância, durante 104 dias. Ao final desse período foram feitas análises de crescimento, do conteúdo de pigmentos fotossintéticos, de trocas gasosas, da fluorescência da clorofila a, do conteúdo foliar de carboidratos solúveis, das características anatômicas foliares e caulinares e da plasticidade fenotípica da espécie. No experimento 2, plantas de C. estrellensis e C. legalis com 14 meses de idade foram submetidas a dois tratamentos: 30% e 100% de irradiância (sombra e sol, respectivamente), durante 30 dias. Ao final desse período foram feitas análises do estresse oxidativo das espécies, por meio da quantificação da atividade das enzimas catalase e peroxidase do ascorbato e por meio da quantificação do conteúdo foliar de pigmentos fotossintéticos. No experimento 1, em 70% de irradiância, as plantas apresentaram melhor crescimento em altura e diâmetro, maior massa seca de folhas (MSF), de caule (MSC) e de raiz (MSR). Em 70% e 100% de irradiância, as plantas apresentaram folhas menores (AFU) e mais espessas (AFE e MFE) resultando em menor área foliar total (AFT). Nesses tratamentos as plantas também apresentaram menor conteúdo foliar de clorofila a (Chl a) e b (Chl b), porém, maior razão Chl a/b e maior conteúdo de carotenóides, o que implicou em menor razão Chl a/Carot. Taxas fotossintéticas maiores foram encontradas nas plantas em 70% e inibidas em 40% e 50%, em função da baixa irradiância solar, e em 100%, possivelmente pela ocorrência de fotoinibição, como mostraram os parâmetros do fluxo de energia do fotossistema II. De acordo com a análise da fluorescência da clorofila a, em pleno sol, as plantas apresentaram menor densidade de centros de reação ativos (RC/ABS) e maior dissipação de energia (DI0/ABS), culminando com menor desempenho do fotossistema II (PIabs) e desempenho total (PITotal). O conteúdo foliar de carboidratos solúveis foi maior nas plantas em 70%, seguido das plantas em 100% de irradiância, com exceção da glicose, que não variou entre os tratamentos. A maior espessura encontrada nas folhas sob 100% de irradiância foi em função da maior espessura das epidermes adaxial e abaxial e dos parênquimas paliçádico e esponjoso. E o maior diâmetro do caule em 70% de irradiância se deu pela maior espessura do xilema e floema secundários. No experimento 2, as plantas em pleno sol de ambas as espécies também apresentaram menor conteúdo foliar de clorofila a (Chl a) e b (Chl b) e maior razão Chl a/b. No entanto, o conteúdo de carotenóides foi maior, o que implicou em menores razões Chl a/Carot. A atividade da catalase (CAT) variou em função do tempo e da espécie, apresentando uma queda em C. estrellensis aos 16 dias, possivelmente em função de fotoinativação, e um aumento em C. legalis aos 30 dias. Já a atividade da peroxidase do ascorbato (APX) não variou em função do tempo, da espécie ou dos tratamentos. O estudo da plasticidade fenotípica mostrou que C. estrellensis é uma espécie plástica, principalmente em função das variáveis de fotossíntese e trocas gasosas, sendo capaz de sobreviver no gradiente de irradiância testado, o que viabiliza o seu uso em projetos de recuperação de áreas degradadas. E, uma vez que as análises ecofisiológicas mostraram que C. estrellensis e C. legalis apresentaram melhor desempenho em luminosidade moderada, sugere-se que ambas comportaram-se como espécies intermediárias no processo de sucessão florestal. No entanto, uma vez que a concentração de pigmentos foliares e a produção de enzimas antioxidantes inferiram maior susceptibilidade de C. estrellensis à fotoinibição em alta irradiância, sugere-se maior viabilidade do uso de C. legalis em projetos de recuperação de áreas degradas.

As ações de reflorestamento com o pau-brasil (Caesalpinia echinata Lam.) depende de informações de suas características ecofisiológicas sujeitas às variações ontogenéticas e ambientais. O objetivo desse trabalho foi caracterizar alguns aspectos morfológicos, anatômicos, fisiológicos e estruturais de parede celular de C. echinata nas fases juvenil, jovem e adulto em condições naturais em um fragmento da Floresta Atlântica. Foi analisada a biometria, concentração de nutrientes e dos pigmentos cloroplastídicos dos foliólulos, anatomia foliar e do xilema secundário do caule e a constituição dos polímeros estruturais de parede celular. Os indivíduos juvenis localizados no estrato inferior da floresta se destacaram pela maior área foliar específica e maiores teores de pigmentos cloroplastídicos bem como pelas maiores dimensões de suas células guardas associado às maiores concentrações de K e Ca foliar. Estruturalmente, os indivíduos juvenis apresentaram menores elementos de vasos e teores de lignina. Os indivíduos jovens apresentaram valores intermediários das variáveis analisadas. Já os indivíduos adultos, cujas copas alcançavam o dossel, se destacaram pelo maior espessamento do limbo, da cutícula e do parênquima lacunoso, teor de água foliar, densidade estomática e teor de lignina foliar e caulinar cuja capacidade de síntese foi associada ao maior teor de P foliar. O conteúdo de celulose foliar e caulinar não variou entre as diferentes fases ontogenéticas. As hemiceluloses são do tipo xilanos com possibilidade de presença de xiloglucano dada a maior fração de xilose (±12% MS) e galactose (±1% MS). A glucose foi o monossacarídeo mais representativo (±40% MS) sem diferenças ontogenéticas. As diferenças morfológicas, anatômicas, fisiológicas e estruturais parecem, também, estarem sob controle da irradiância mais intensa na copa dos indivíduos adultos. Os resultados denotam que o plantio consorciado com espécies de crescimento rápido seja a melhor ação para o reflorestamento de C. echinata.

The objectives of this study were to identify anthophilous butterflies on psychophilous flowers of four Asteraceae species in an Atlantic Forest fragment in Viçosa, Minas Gerais State, Southeastern Brazil, and to determine whether there are species in common with other lepidopteran inventories of the Southeastern and Midwestern regions of Brazil. It is the first inventory of anthophilous butterflies of a semideciduous forest fragment in Zona da Mata, State of Minas Gerais. A total of 108 species were recorded, representing the fourth largest lepidopteran survey in this State. The results demonstrated that Asteraceae species may be important tools for monitoring anthophilous butterflies. The similarity with other inventories ranged from 1 to 92.55%. Fifteen species were reported for the first time in the State of Minas Gerais, and among them, Melanis alena and Thisbe irenea were observed in this study only.

The mycelium and young fruiting bodies of Agaricus blazei were submitted to supercritical CO2 extraction, in a modified commercial flow apparatus, at temperatures from 40 to 80 ºC, pressures up to 600 bar and CO2 flow-rates from 2.0 to 9.0 g.min-1. The best extraction conditions of secondary metabolites, whereby the degree of solubilization (g extract/100 g of fungi) is the highest, was obtained with pure CO2 at 400 bar, 70 ºC and a CO2 flow rate of 5.7g.min-1. The extract in that conditions were analysed by GC-Ms. In order to increase the extraction yield of secondary metabolites, which are mostly present in glycolipid fractions, a polar compound (ethanol) was used as co-solvent in the proportions of 5 and 10 % (mol/mol). The presence of ethanol increased the yield when compared with the extraction with pure CO2. Moreover, a simple model was applied to the supercritical CO2 extraction of secondary metabolites from Agaricus blazei.
