The purpose of this research was to study the genetic diversity and genetic relatedness of 60 genotypes of grapevines derived from the Germplasm Bank of Embrapa Semiárido, Juazeiro, BA, Brazil. Seven previously characterized microsatellite markers were used: VVS2, VVMD5, VVMD7, VVMD27, VVMD3, ssrVrZAG79 and ssrVrZAG62. The expected heterozygosity (He) and polymorphic information content (PIC) were calculated, and the cluster analysis were processed to generate a dendrogram using the algorithm UPGMA. The He ranged from 81.8% to 88.1%, with a mean of 84.8%. The loci VrZAG79 and VVMD7 were the most informative, with a PIC of 87 and 86%, respectively, while VrZAG62 was the least informative, with a PIC value of 80%. Cluster analysis by UPGMA method allowed separation of the genotypes according to their genealogy and identification of possible parentage for the cultivars 'Dominga', 'Isaura', 'CG 26916', 'CG28467' and 'Roni Redi'.

The use of Passiflora species for ornamental purposes has been recently developed, but little is known about pollen viability and the potential for crossing different species. The objective of this study was to evaluate the pollen viability of six Passiflora species collected from different physiological stages of development through in vitro germination and histochemical analysis using dyes. The pollen was collected in three stages (pre-anthesis, anthesis and post-anthesis). Three compositions of culture medium were used to evaluate the in vitro germination, and two dyes (2,3,5-triphenyltetrazolium chloride, or TTC, and Lugol's solution) were used for the histochemical analysis. The culture medium containing 0.03% Ca(NO3) 4H2O, 0.02% of Mg(SO4 ).7H2O, 0.01% of KNO3, 0,01% of H3BO3, 15% sucrose, and 0.8% agar, pH 7.0, showed a higher percentage of pollen grains germinated. Anthesis is the best time to collect pollen because it promotes high viability and germination. The Lugol's solution and TTC dye overestimated the viability of pollen, as all accessions showed high viability indices when compared with the results obtained in vitro.

Frutos das cultivares Arapaho, Brazos, Caingangue, Cherokee, Choctaw, Comanche, Ébano, Guarani, Tupy e Xavante, e uma espécie de amoreira-vermelha (Rubus rosifolius Smith) foram avaliados quanto à composição química. Avaliou-se também a variação genética entre as cultivares de amoreira-preta e a espécie de amoreira-vermelha. Os resultados demonstraram variações na composição química dos frutos estudados. A amora- vermelha apresentou menor teor de umidade e maiores valores para os componentes cinzas, açúcares totais, açúcares redutores, açúcares näo redutores, pectina total, pectina solúvel, fenóis, flavonoides, licopeno, β-caroteno e vitamina A. Os teores de umidade e antocianinas, a porcentagem de solubilização e a atividade antioxidante foram maiores nos frutos da cultivar Ébano. Verificou-se que os frutos da amoreira-vermelha e da cultivar Ébano apresentaram o maior grau de divergência genética para as variáveis analisadas. Isto indica a possibilidade de uso das mesmas em programas de melhoramento que visem à melhoria da composição química.

Estudou-se o efeito de quatro condições de manejo sobre a aceitação de banana 'Pacovan'. Os frutos foram colhidos no ponto de maturação fisiológica ideal para o armazenamento, em quatro propriedades rurais do município de Bananeiras - PB, seguindo-se delineamento inteiramente casualizado. Os tratamentos foram identificados como: P1: irrigação+adubação química e orgânica +controle químico de plantas invasoras; P2: controle manual de plantas invasoras; P3: irrigação+adubação química+controle manual de plantas invasoras; P4: controle manual e químico de plantas invasoras. Em cada propriedade, foram colhidos cinco cachos de banana que foram submetidos à climatização com carbureto de cálcio por 12 horas para uniformização da maturação. Após esse período, foram mantidos à temperatura ambiente até sua completa maturação. O painel sensorial foi composto por 62 julgadores não treinados, com idade média de 18 anos. Os atributos aroma, sabor, textura e cor da polpa foram avaliados por meio de teste de aceitação, utilizando-se de escala hedônica estruturada de 9 pontos. Para o teste de intenção de compra, foi utilizada escala estruturada de 5 pontos. As amostras foram apresentadas aos julgadores na forma de rodelas, com espessura aproximada de 1,5 cm. Os resultados foram submetidos à análise de variância (ANOVA), e as médias, comparadas pelo teste de Tukey, ao nível de 5% de probabilidade. Para todos os atributos avaliados,os valores situaram-se entre 7,34 e 7,68, posicionando-se na escala hedônica entre os termos 'gostei moderadamente' e 'gostei muito'. Para intenção de compra, o percentual de aceitação foi de 74,19; 72,58; 72,58 e 70,97%, respectivamente, para as amostras P1, P2, P3 e P4. O tipo de manejo não influenciou na aceitação da banana 'Pacovan', sendo os frutos considerados pelos julgadores como produto de boa aceitação.

A amora-preta é uma fruta que vem despertando a atenção de produtores e consumidores devido ao seu sabor agradável, cor atrativa e por apresentar em sua composição elevado teor de compostos bioativos. O grande entrave para consumo e a comercialização dos frutos da amoreira é sua elevada taxa respiratória, o que reduz sua vida útil. Uma alternativa viável para o aproveitamento econômico dessas frutas consiste em sua industrialização, podendo ser congeladas, enlatadas, processadas na forma de polpa, ou na forma de sucos e geleias. O objetivo deste trabalho foi avaliar os efeitos da radiação gama na conservação da polpa de amora. A irradiação foi realizada no laboratório de irradiação gama do Centro de Desenvolvimento da Tecnologia Nuclear (CDTN), em uma fonte de cobalto 60, com as seguintes doses de radiação: 0,75 kGy; 1,5 kGy e 3 kGy, sob uma taxa de dose de 3,24 kGy.h-1 . A polpa não irradiada foi utilizada como controle. As polpas de amora irradiadas foram armazenadas à temperatura de 4ºC, sendo avaliadas nos tempos de 0; 7; 15; 30 e 60 dias. Para verificar os efeitos da radiação gama no processamento na polpa, foi realizada a caracterização físico-química e química através das análises de acidez, pH, sólidos solúveis, sólidos totais, teor de antocianinas, atividade antioxidante e cor. Também foi feita análise microbiológica de acordo com a legislação brasileira vigente. O processo de irradiação aumentou a vida útil de prateleira da polpa em até 60 dias, sendo que o tratamento com uma dose de 1,5 kGy foi a que proporcionou a melhor qualidade microbiológica.

Fish eggs are associated with microbes, whose roles range from mutualism to parasitism. Recent laboratory experiments have shown that the taxonomic composition of associated microbial communities on the egg influences embryonic development. Host genetics also plays an important role in determining the consequences for embryonic growth and survival in this interaction. Moreover, it has been found that the importance of host genetics increases during embryogenesis. These findings suggest that during embryogenesis, the host increasingly influences the composition of its associated microbial community. However, little is known about the composition of microbial communities associated with naturally spawned eggs in the wild. We sampled fertilized whitefish eggs (Coregonus spp.) of different developmental stages from six sub-Alpine lakes and used a universal primer pair and 454 pyrosequencing in order to describe the taxonomic composition of egg-associated bacterial communities. We found bacterial communities on early embryos to be very diverse and to resemble the bacterial composition of the surrounding water environment. The bacterial communities on late embryos were significantly less diverse than on early embryos and displayed a clear shift in taxonomic composition that corresponded poorly with the bacterial composition of the surrounding water environment. The main bacterial components on whitefish eggs in this study were Proteobacteria, Actinobacteria, and Firmicutes, while the five most common families were Leuconostocaceae, Streptococcaceae, Comamonadaceae, Oxalobacteraceae and Moraxellaceae. Their putative relationships with the host are discussed. We conclude that natural symbiotic bacterial communities become more specialized during embryogenesis because of specific interactions with their embryo host.

RESUMO A podridão floral dos citros (PFC) é uma importante doença dessa cultura, responsável por elevadas perdas de produção. Normalmente, essa doença mostra-se limitante quando ocorrem prolongados períodos chuvosos durante o florescimento das plantas ou quando existe intenso molhamento foliar. Duas espécies de Colletotrichum estão associadas à doença: C. acutatum e C. gloeosporioides. Entretanto, recentemente, tem-se verificado que, mesmo sob condições não tão propícias, a doença tem ocorrido com relativa frequência, suspeitando-se do envolvimento de outras espécies de Colletotrichumou de novas condições de adaptação das espécies descritas. Este trabalho teve como objetivo determinar se há ou não outra espécie de Colletotrichum associada a PFC e avaliar a viabilidade do emprego de marcadores moleculares ISSR na caracterização taxonômica de isolados de Colletotrichum spp. associados a sintomas de PFC em flores, assim como de tecidos foliares e frutos cítricos assintomáticos. Para tanto, foi empregada uma combinação de iniciadores específicos, levando em conta a região ITS e marcadores moleculares ISSR. Os marcadores ISSR mostraram-se eficientes na caracterização taxonômica dos isolados de Colletotrichum analisados. A população avaliada foi constituída apenas por C. acutatum e C. gloeosporioides, descartando o envolvimento de uma espécie adicional. Foi constatada alta diversidade genética entre os isolados analisados, o que também se mostra convergente quanto às diferenças fenotípicas observadas sob condições de campo. Entretanto, não foi encontrada relação quanto à origem e as espécies de Colletotrichum spp. associadas. De modo inédito, ainda que assintomaticamente, foi detectada a presença de um isolado de C. acutatum associado a frutos cítricos.

At present, despite extensive laboratory investigations, most cases of porcine abortion remain without an etiological diagnosis. Due to a lack of recent data on the abortigenic effect of order Chlamydiales, 286 fetuses and their placentae of 113 abortion cases (1-5 fetuses per abortion case) were investigated by polymerase chain reaction (PCR) methods for family Chlamydiaceae and selected Chlamydia-like organisms such as Parachlamydia acanthamoebae and Waddlia chondrophila. In 0.35% of the cases (1/286 fetuses), the Chlamydiaceae real-time PCR was positive. In the Chlamydiaceae-positive fetus, Chlamydia abortus was detected by a commercial microarray and 16S ribosomal RNA PCR followed by sequencing. The positive fetus had a Porcine circovirus-2 coinfection. By the Parachlamydia real-time PCR, 3.5% (10/286 fetuses of 9 abortion cases) were questionable positive (threshold cycle values: 35.0-45.0). In 2 of these 10 cases, a confirmation by Chlamydiales-specific real-time PCR was possible. All samples tested negative by the Waddlia real-time PCR. It seems unlikely that Chlamydiaceae, Parachlamydia, and Waddlia play an important role as abortigenic agents in Swiss sows.

Chlamydophila abortus and Waddlia chondrophila cause abortion in ruminants. We investigated the role of Parachlamydia acanthamoebae in bovine abortion. Results of immunohistochemical analyses were positive in 30 (70%) of 43 placentas from which Chlamydia-like DNA was amplified, which supports the role of Parachlamydia spp. in bovine abortion.

Given the dual role of many plant traits to tolerate both herbivore attack and abiotic stress, the climatic niche of a species should be integrated into the study of plant defense strategies. Here we investigate the impact of plant reproductive strategy and components of species' climatic niche on the rate of chemical defense evolution in the milkweeds using a common garden experiment of 49 species. We found that across Asclepias species, clonal reproduction repeatedly evolved in lower temperature conditions, in species generally producing low concentrations of a toxic defense (cardenolides). Additionally, we found that rates of cardenolide evolution were lower for clonal than for nonclonal species. We thus conclude that because the clonal strategy is based on survival, long generation times, and is associated with tolerance of herbivory, it may be an alternative to toxicity in colder ecosystems. Taken together, these results indicate that the rate of chemical defense evolution is influenced by the intersection of life-history strategy and climatic niches into which plants radiate.

Marine microorganisms, including Aeromonas, are a source of compounds for drug development that have generated great expectations in the last decades. Aeromonas infections produce septicaemia, and ulcerative and haemorrhagic diseases in fish. Among the pathogenic factors associated with Aeromonas, the lipopolysaccharides (LPS), a surface glyconconjugate unique to Gram-negative bacteria consisting of lipid A (lipid anchor of the molecule), core oligosaccharide and O-specific polysaccharide (O antigen), are key elicitors of innate immune responses. The chemical structure of these three parts has been characterized in Aeromonas. Based on the high variability of repeated units of O-polysaccharides, a total of 97 O-serogroups have been described in Aeromonas species, of which four of them (O:11; O:16; O:18 and O:34) account for more than 60% of the septicemia cases. The core of LPS is subdivided into two regions, the inner (highly conserved) and the outer core. The inner core of Aeromonas LPS is characterized by the presence of 3-deoxy-D-manno-oct-2-ulosonic (ketodeoxyoctonic) acid (Kdo) and L-glycero-D-manno-Heptoses (L,D-Hep), which are linked to the outer core, characterized by the presence of Glc, GlcN, Gal, and GalNAc (in Aeromonas salmonicida), D,D-Hep (in Aeromonas salmonicida), and L,D-Hep (in Aeromonas hydrophila). The biological relevance of these differences in the distal part of the outer core among these species has not been fully assessed to date. The inner core is attached to the lipid A, a highly conserved structure that confers endotoxic properties to the LPS when the molecule is released in blood from lysed bacteria, thus inducing a major systemic inflammatory response known as septic or endotoxic shock. In Aeromonas salmonicida subsp. salmonicida the Lipid A components contain three major lipid A molecules, differing in acylation patterns corresponding to tetra-, penta- and hexaacylated lipid A species and comprising of 4′-monophosphorylated β-2-amino-2-deoxy-D-glucopyranose-(1→6)-2-amino-2-deoxy-D-glucopyranose disaccharide. In the present review, we discuss the structure-activity relationships of Aeromonas LPS, focusing on its role in bacterial pathogenesis and its possible applications.

We analysed concentrations of cadmium, lead, mercury and selenium in blood from males and females of the 2 sibling species of giant petrels, the northern Macronectes halli and the southern M. giganteus, breeding sympatrically at Bird Island (South Georgia, Antarctica). Blood samples were collected in 1998 during the incubation period, from 5 November to 10 December. Between species, cadmium and lead concentrations were significantly higher for northern than for southern giant petrels, which probably resulted from northern giant petrels wintering in more polluted areas (mainly on the Patagonian Shelf and Falkland Islands) compared to southern giant petrels (wintering mainly around South Georgia and the South Sandwich Islands). Between sexes, cadmium concentrations were significantly higher for females than for males in both species, corresponding to the more pelagic habits of females compared to the more scavenging habits of males. Lead and cadmium concentrations in circulating blood decreased significantly over the incubation period, suggesting that when breeding at Bird Island, exposure to the source of pollution had ended, and these metals had been cleared from the blood and excreted, or rapidly transferred to other tissues. Association of lead and cadmium with a common source of pollution was further corroborated by a significant positive correlation between the levels of the 2 elements found. Mercury levels were similar between the species, but showed an opposite trend between sexes, with males showing higher levels than females in northern giant petrels, and the opposite was true in southern giant petrels, with no changes throughout incubation. Selenium levels were similar between sexes, but significantly greater for northern than for southern giant petrels. Moreover, there was a significant increase in the selenium levels over the incubation period in northern giant petrels. Age of adult birds did not affect metal concentrations. Coefficients of variation of metal levels were consistently lower for northern than for southern giant petrels, particularly for mercury, suggesting that the former species is more dietary specialised than the latter. Contaminant analyses, when combined with accurate information on seabird movements, obtained through geolocation or satellite tracking, help us to understand geographic variation of pollution in the marine environment.
