Ethnopharmacological importance: Many species of plants in the Brazilian cerrado (savanna) are widely used in ethnomedicine. However, the safety and effectiveness of medicinal plants used in communities with little or no access to manufactured drugs should be evaluated. Aim of the study: Evaluate the antimicrobial and cytotoxic activities of extracts from eight plant species, obtained using Brazilian cachaca as the extractor liquid. Materials and methods: The extracts were tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Candida parapsilosis, promastigote forms of Leishmania amazonensis, and poliovirus. In addition, cytotoxic activity was assayed in Vero cells and in human erythrocytes. Results: The plant species Curatella americana, Sclerolobium aureum, and Plathymenia reticulata showed the best activity against yeasts, especially the crude extract of C. americana and its ethyl-acetate fraction. Kielmeyera lathrophyton showed a minimum inhibitory concentration of 250 mu g/ml against S. aureus, and was inactive against Gram-negative bacteria. The extract obtained from Annona coriacea showed the best activity against the promastigote forms of Leishmania amazonensis (IC(50) = 175 mu g/ml). Only C. americana showed potential for antipoliovirus activity. The concentrations of the crude extracts that showed toxicity to VERO cells had CC(50) between 31 and 470 mu g/ml, and the lyophilized Brazilian cachaca showed a CC(50) of 307 mu g/ml. None of the extracts showed toxicity against human erythrocytes. Conclusions: Among the plant species studied. C americana proved to be effective against microorganisms, especially as an antifungal. The results will help in the search for alternative drugs to be used in pharmacotherapy, and will contribute to establish safe and effective use of phytomedicines in the treatment of infectious diseases. Catasetum fimbriatum plants cultivated in the absence of light exhibit continuous shoot growth leading to the formation of nodes and internodes. On the other hand, when these plants are incubated in the presence of light, shoot longitudinal growth is inhibited and pseudobulbs develop just below the shoot apical meristem. These facts provide evidence of a possible influence of light on mitotic cell division in the shoot apex as well as on pseudobulb initiation. The effects of light and dark on the interruption and/or maintenance of shoot apex mitotic activity and the subsequent formation of pseudobulbs in the sub-meristematic regions were investigated by means of histological and hormonal studies. The interruption of shoot apex development occurred around the 150th d of light incubation and seems to have resulted from the establishment of a strong storage sink in the region of the future pseudobulb, in detriment to the continuous activity of the shoot apical meristem. The reduced total cytokinins/IAA ratio in the apex, mainly due to high levels of IAA, could be a key factor in the interruption of cell divisions. Transfer to the dark brings about the resumption of shoot apex development of plants through the re-entrance of cells in the cell cycle which coincides with a significant increase in the total cytokinins/IAA ratio. The anatomy and ultrastructure of root nodules of Anadenanthera peregrina var. falcata (Leguminosae-Mimosoideae) were analysed, as was plant growth. To ensure that nodules developed, seedlings were inoculated with a mixture of six strains of rhizobia. Nodules were produced that differed in appearance-and probably also effectiveness-but their structure was similar and they showed characteristics typical of indeterminate nodules, such as persistent meristematic tissue and a gradient of cells at different stages of development. Many starch grains were present in inner cortex cells and interstitial cells of infected tissue. Infected cells were densely packed with bacteroids, which contained many poly-beta-hydroxybutyrate granules. The high incidence of these granules, together with high levels of starch accumulation in interstitial cells, suggested low N-2-fixation efficiency of the rhizobia isolates used for inoculation. In the symbiosomes of early-senescent infected cells, reticulum-like structures, small vesicles and a fibrillar material were observed; these may be related to bacteroid degradation. In the cytoplasm of late-senescent infected cells, many vesicles and membrane-like structures were observed, probably associated with membrane degradation of bacteroids and peribacteroids. The total biomass of plants inoculated with rhizobia was low and their xylopodia and shoots had low levels of N compared with non-inoculated plants fertilized with ammonium nitrate. However, inoculated plants did not show N-deficiency symptoms and grew better than non-inoculated plants without N fertilization. These growth results, together with ultrastructural observations of nodules, suggest that nitrogen fixation of rhizobia isolates associated with Anadenanthera peregrina var. falcata roots is poor. A toxicidade do alumínio (Al) é um dos fatores mais limitantes para a produtividade. Esta pesquisa foi realizada para avaliar a influência do Al, em solução nutritiva, na altura de plantas, no peso da matéria seca e nas alterações morfoanatômicas de raízes e folhas de milho (Zea mays L.). O experimento foi conduzido em casa de vegetação com tratamentos constituídos de cinco doses de Al (0; 25; 75; 150; e 300 µmol L-1) e seis repetições. As soluções foram constantemente aeradas e o pH foi ajustado a 4,3, inicialmente. A matéria seca da parte aérea e das raízes e a altura das plantas diminuíram significativamente com o aumento da concentração de Al. As raízes de plantas de milho cultivadas em soluções com Al tiveram seu crescimento inibido e apresentaram menos raízes laterais e desenvolvimento do sistema radicular inferior, em comparação com as das plantas-controle. As folhas das plantas crescidas em soluções que continham 75 e 300 µmol L-1 de Al não apresentaram muita diferença anatômica em relação às das plantas-controle. A bainha da folha das plantas exposta ao Al apresentou epiderme uniestratificada revestida por uma fina camada de cutícula e as células da epiderme e do córtex foram as que menos se desenvolveram. No feixe vascular, o metaxilema e protoxilema não tinham paredes secundárias, e o diâmetro de ambos foi muito menor quando comparado com os das plantas-controle.

O tripes Frankliniella schultzei Trybom (Thysanoptera: Thripidae) é um dos mais importantes vetores de tomato spotted wilt tospovirus (TSWV) na cultura do tomate no Brasil. Dois métodos de aplicação do inseticida thiamethoxam foram comparados no controle de tripes em tomate. O experimento foi conduzido em delineamento de blocos ao acaso com sete tratamentos e quatro repetições. Utilizou-se um pulverizador costal manual com lança simples provida de ponta de pulverização com jato cônico vazio JD 14-2 ou modificada, com aplicador tipo esguicho, em substituição à ponta de pulverização. Dosagens de 150 e 200 g i.a. ha-1 foram usadas em única aplicação com esguicho e 50 g i.a. ha-1 em pulverizações semanais, iniciadas aos 48 dias após a semeadura. A eficiência do inseticida em teste foi comparada com diafenthiuron (400 g i.a. ha-1), profenofos + cypermethrin (320+32 g i.a. ha-1, respectivamente) e methamidophos (60 g i.a. 100 L-1 de água), aplicados em pulverização. Não houve diferença estatística entre os métodos de aplicação e doses com thiamethoxam no controle de F. schultzei aos 24 dias após a aplicação, sendo vantajosa a realização de uma única aplicação das maiores dosagens de thiamethoxam com esguicho em comparação às aplicações semanais da menor dosagem em pulverização. A eficiência de controle do tripes com o inseticida thiamethoxam variou de 93 a 95%, nos diferentes métodos e dosagens em teste. Diafenthiuron e profenofos + cypermethrin apresentaram eficiência menor (78 e 88%, respectivamente), porém foram superiores às obtidas com methamidophos (71%). Os produtos e doses utilizados não causaram fitotoxicidade às plantas de tomate.

Introduction. Pitaya (Hylocereus undatus) is an exotic fruit species little known in Brazil and which needs basic studies about plant nutrition, propagation and physiology. Emphasizing the co-existence of juvenile and adult stages in the pitaya canopy, the plant is generally propagated by cuttings. Materials and methods. A completely randomized design with four treatments and five replications was adopted. Each treatment was represented by the part of the canopy from which the cutting was taken ( upper, middle and lower cutting and cuttings from young plants). The following variables were registered: % cuttings with roots, % of live cuttings, root density, root diameter, root area, root length and root dry mass. Results were submitted to variance analyses, Tukey's test at 0.01 probability error and simple correlation analysis. Results and discussion. The results indicated that the position from which the cutting is taken had a quantitative effect on rooting formation of pitaya cuttings. Juvenile cuttings presented 35% more cuttings with roots than adult cuttings. Root density, root area, root length and root dry mass depended on juvenility, the highest results being registered for juvenile cuttings, independently of the variable. Conclusion. Juvenile and adult stages co-exist in the pitaya canopy. Juvenility is an important rooting factor for red pitaya cuttings.

An integrated and interdisciplinary research programme with native medicinal plants from tropical forests has been performed in order to obtain new forest products for sustainable use in regional markets vis-à-vis ecosystem conservation. For the success of this programme ethnopharmacological studies are very important with respect to (i) identification of useful plants including medicinal and aromatic species; (ii) recuperation and preservation of traditional knowledge about native plants; and (iii) identification of potential plants with economic value. The plants are selected with a view to evaluate efficacy and safety (pharmacological and toxicological studies), and phytochemical profile and quality control (phytochemical and chromatographic characterization). These studies are very important to add value to plant products and also to mitigate unscrupulous exploitation of medicinal plants by local communities, since multiple use of plants represents an excellent strategy for sustaining the tropical ecosystem through ex situ and in situ conservation. Thus, conservation of tropical resources is possible in conjunction with improvements in the quality of life of the traditional communities and production of new products with therapeutic, cosmetic and 'cosmeceutic' value. © NIAB 2005.

Isotope screening is a simple test for determining the photosynthetic pathway used by plants. The scope of this work was to classify the photosynthetic type of some herbs and medicinal plants through studies of the carbon isotope composition (δ13C). Also, we propose the use of carbon isotope composition as a tool to control the quality of herbs and medicinal plants. For studies of δ13C, δ 13C‰ = [R (sample)/R (standard) - 1] × 10-3, dry leaves powdered in cryogenic mill were analyzed in a mass spectrometer coupled with an elemental analyzer for determining the ratio R = 13CO2/12CO2. In investigation of δ13C of 55 species, 23 botanical families, and 44 species possessed a C3 photosynthetic type. Six species found among the botanical families Euphorbiaceae and Poaceae were C4 plants, and 5 species found among the botanical families Agavaceae, Euphorbiaceae, and Liliaceae possessed CAM-type photosynthesis. Carbon isotope composition of plants can be used as quality control of herbs and medicinal plants, allowing the identification of frauds or contaminations. Also, the information about the photosynthetic type found for these plants can help in introducing and cultivating exotic and wild herbs and medicinal plants.

Extracts of plants of the Sclerolobium genus are disclosed, as well as methods for preparing same and cosmetic and pharmaceutical formulations containing these extracts. These extracts can be obtained by polar and/or apolar extraction, each fraction containing different anti-oxidants, all of which are useful for topical treatment. Also disclosed is the use of this extract in cosmetic and/or pharmaceutical formulations for topical use.

Rapid tests to assess the nutritional status of plants gerbera can make easier the decision of any adjustments in the fertilization of the substrate. The study was conducted to evaluate nutritionally fertigated gerbera plants with increasing levels of electrical conductivity (EC), using portable meters nitrate and potassium, and chlorophyll. For this, two experiments were conducted. A randomized block design with five levels of EC (0.5, 2.0, 3.5, 5.0 and 6.5 dS m-1) and four replications was used. Cherry cultivar was used in the first experiment. In the second experiment, two cultivars (Cherry and Salmon Rose) were used. In the first experiment, the sap of the leaves was subjected to rapid testing of N-NO3- (equipment Cardy Horiba C -141). N-NO3- and K+ (C-131) were determined in the substrate solution. The intensity of the green leaf, in the second experiment was evaluated with portable Chlorophyll Meter. The N content in the indicator leaf in the first and N and K contents in the plant in the second experiment were determined. Rapid tests are presented as good indicators of the level of N-NO3- and K+ in plant tissue, with the advantages of convenience and speed of determination, facilitating monitoring of fertilization of the substrate by producers and technicians.
