Inteins or internal proteins are coding sequences that are transcribed and translated with flanking sequences (exteins). After translation, the inteins are excised by an autocatalytic process and the host protein assumes its normal conformation and develops its expected function. These parasitic genetic elements have been found in important, conserved proteins in all three domains of life. Most of the eukaryotic inteins are present in the fungi kingdom and the PRP8 intein is one of the most widespread inteins, occurring in important pathogens such as Cryptococcus neoformans (varieties grubii and neoformans), Cryptococcus gattii, Histoplasma capsulatum and Paracoccidioides brasiliensis. The knowledge of conserved and non-conserved domains in inteins have opened up new opportunities for the study of population variability in pathogenic fungi, including their phylogenetic relationships and recognition or diagnoses of species. Furthermore, inteins in pathogenic fungi should also be considered a promising therapeutic drug target, since once the autocatalytic splicing is inhibited, the host protein, which is typically vital, will not be able to perform its normal function and the fungal cell will not survive or reproduce.

The prevalence of gastrointestinal parasites in stray dogs, and dogs with owners was investigated by fecal examinations from 271 dogs employing sedimentation, simple flotation and centrifugation-flotation methods. The centrifugation-flotation method, when compared to simple flotation or sedimentation methods was generally more accurate in the diagnosis of all intestinal parasites, but statistical differences were detected only in relation to Giardia spp. and Cystoisospora spp. (synonym Isospora spp.). The following parasites, with their respective prevalence, were diagnosed in the fecal samples: Ancylostoma spp. (23.6%); Toxocara canis (5.5%); Trichuris vulpis (4.8%); Spirocerca lupi (1.9%); Dipylidium caninum (0.7%); Giardia spp. (12.2%); Hammondia heydorni (2.6%); Cystoisospora spp. (8.5%); and Sarcocystis spp. (2.2%). The prevalence of most parasites was similar for dogs of mixed-breed and for dogs of a defined-breed, except for Cystoisospora spp. and T canis which showed a significantly higher prevalence in mixed-breed dogs. The prevalence of Ancylostoma spp. (17.1%) was significantly lower in stray dogs than in those with an owner (31.9%) and the prevalence of Giardia spp. and Cystoisospora spp. was higher in stray dogs (P < 0.05). No effect of season on the occurrence of the different parasite genera could be observed, except for Ancylostoma spp., for which an increase in the percentage of dogs shedding eggs was observed at the beginning of Summer with a peak occurrence during April and May (Autumn). The prevalence of Ancylostoma spp., T canis, T vulpis, Giardia spp. and Cystoisospora spp. was higher in adult males than in adult females, but significant differences between the two groups occurred only with Giardia spp. Young animals were found to more frequently shed Nematode eggs in feces than adult animals. Amostras fecais de cabritos machos e fêmeas, de diferentes raças e com até uma ano de idade, foram examinadas para determinação do número de ovos e oocistos por grama de fezes (OPG e OoPG, respectivamente) e coprocultura para identificação genérica dos nematódeos. Ovos de helmintos e oocistos de Eimeria spp. foram observados em 93,06% (188/202) e 77,22% (156/202) das amostras, respectivamente. Pelas coproculturas, foram identificados os gêneros Cooperia em 11,88% (24/202), Haemonchus em 51,98% (105/202), Oesophagostomum em 9,4% (19/202), Strongyloides em 5,94 (12/202) e Trichostrongylus em 20,79% (42/202) das amostras. As espécies de Eimeria encontradas foram E. alijevi em 25,24% (51/202), E. arloingi em 7,42% (15/202), E. caprina em 2,97% (6/202), E. caprovina em 10,39% (21/202), E. christenseni em 4,45% (9/202), E. joklchijevi em 11,38% (23/202), E. hirci em 9,4% (19/202) e E. ninakohlyakimovae em 28,71% (58/202) das amostras. Dentre os parasitas gastrintestinais, houve predominância do gênero Haemonchus e de duas espécies de Eimeria: E. ninakohlyakimovae e E. alijevi.

Este trabalho foi realizado com o objetivo de estimar o grau de infecção dos helmintos gastrintestinais em um rebanho caprino criado no Planalto Norte Catarinense. Foram utilizadas 12 fêmeas jovens e 11 adultas, das quais, a cada 28 dias, foram coletadas amostras de fezes diretamente do reto, totalizando 12 coletas, para quantificação de ovos por grama de fezes (OPG) e cultivo de larvas através de pool das amostras positivas do mesmo grupo. A contagem de OPG variou de zero a 10.400 nos animais jovens e de zero a 7.600 nos adultos. As médias do OPG entre as coletas foram de 583,3 a 4.441,7 no grupo jovem e de 418,2 a 2.181,8 nos adultos, sendo observados ovos da ordem Strongylida, dos gêneros Moniezia e Toxocara, bem como oocistos de coccídeos. Os animais mais jovens foram os mais acometidos, sendo o gênero Haemonchus o mais prevalente.

The aim of present study was to compare the efficiency of a commercial assay and two conventional methods for fecal concentration in detecting canine gastrointestinal parasites. Fecal samples from 254 dogs were processed by centrifugation-sedimentation (CS), centrifugation-flotation (CF) and a commercial assay for fecal concentration (TF-test (R)). The following parasites were detected: Ancylostoma (37.8%), Giardia (16.9%), Toxocara canis (8.7%), Trichuris vulpis (7.1%), Isospora (3.5%), and Sarcocystis (2.7%). The calculated analytical sensitivity indicated that CF was more accurate (P < 0.01) in detecting Ancylostoma, T. canis, T. vulpis and Giardia infections. However, CF showed significantly higher sensitivity only for Ancylostoma, compared to the other two methods. The kappa index value of diagnostic agreement between TF-test and CF was high for T. canis (83%) and moderate for Giardia (72%) and Ancylostoma (63%). Coprological examination was used to estimate the prevalence of gastrointestinal parasites in stray and domiciled dogs from Botucatu, São Paulo State, Brazil. Risk factors for dog infection were assessed in relation to demographic, husbandry and management data. The dog owners completed a questionnaire survey on some aspects of dog parasitism such as parasite species, mechanisms of infection, awareness of zoonotic diseases and history of anthelmintic usage. Parasites were found in the faeces of 138 dogs, with an overall prevalence of 54.3%. Dogs harbouring one parasite were more common (31.4%) than those harbouring two (18.5%), three (3.2%) or four (1.2%). The following parasites and their respective frequencies were detected: Ancylostoma (37.8%), Giardia (16.9%), Toxocara canis (8.7%), Trichuris vulpis (7.1%), Dipylidium caninum (2.4%), Isospora (3.5%), Cryptosporidium (3.1%) and Sarcocystis (2.7%). Stray dogs were found more likely to be poliparasitized (P < 0.01) and presented higher prevalence of Ancylostoma, T. canis and Giardia (P < 0.01) than domiciled ones. Toxocara canis was detected more frequently in dogs with < 6 months of age (P < 0.05) and no effect of sex or breed could be observed (P > 0.05). Except for Ancylostoma, that showed a significantly higher prevalence in dogs living in a multi-dog household (P < 0.01), parasite prevalences were similar in single- and multi-dog household. The answers of dog owners to the questionnaire showed that the majority does not know the species of dog intestinal parasites, the mechanisms of transmission, the risk factors for zoonotic infections, and specific prophylactic measures. The predominance of zoonotic species in dogs in the studied region, associated with the elevated degree of misinformation of the owners, indicates that the risk of zoonotic infection by canine intestinal parasite may be high, even in one of the most developed regions of Brazil.

A study on the nematode parasites of nine species of freshwater fishes from Peixe River São Paulo, State, Brazil. was conducted. Fish were collected between February 2010 and March 2011 and the following species were found: Procamallanus (Spirocamallanus) inopinatus and Contracaecum sp. (larvae) in Astyanax altiparanae; Contracaecum sp. (larvae), Dioctophyma renale (larvae), Philometroides caudata, P. (Spirocamallanus) inopinatus, P. (Spirocamallanus) neocaballeroi (larvae) and P. (Spirocamallanus) saofranciscensis in Acestrorhynchus lacustris; Contracaecum sp. (larvae), Guyanema sp., Hysterothylacium sp. (larvae) and Icthyouris sp. in Cyphocharax modestus; Contracaecum sp. (larvae), Cosmoxynemoides aguirrei and Pharyngodonidae gen. sp. in C. nagelii; Dioctophyma renale (larvae), Hysterothylacium sp. (larvae) and Rhabdochona sp. in Gymnotus sylvius; Capillariidae gen. sp. in Hoplosternum littorale; Cosmoxynema vianai, Guyanema sp., Ichthyouris sp. and Travnema travnema in Steindachnerina insculpta; Contracaecum sp. (larvae), Procamallanus (Spirocamallanus) rebecae (larvae) in Triportheus angulatus and Rhabdochona acuminata in Triportheus nematurus. This is first study of nematode parasites from the Peixe River, therefore all the species found are new geographical records and 19 are new host records.

Fungos são os patógenos mais freqüentemente observados em populações de ácaros, auxiliando o controle destes em diferentes culturas. Vinte e cinco amostras de folhas foram coletadas de cultivos de seringueiras vizinhas a dois fragmentos de vegetação de Cerrado. em cada seringal, 25 plantas foram selecionadas para coletas e sete folhas ao redor da copa foram coletadas até a altura de sete a oito metros do solo. Aproximadamente 250 indivíduos de Calacarus heveae Feres, Phyllocoptruta seringueirae Feres e Tenuipalpus heveae Baker, coletados aleatoriamente, foram montados. Hirsutella thompsonii Fisher foi observado nas três espécies de ácaros e T. heveae foi a espécie mais infectada. Os maiores níveis de infecção ocorreram de novembro a fevereiro (estação chuvosa). Na estação seca, os níveis de infecção ficaram abaixo dos 5%. H. thompsonii tem potencial para ser usado como micoacaricida na estação chuvosa.

O trabalho avaliou a eficiência dos isolados (141/3, 233, 233/1, 245, 245/1, 251, 251/2, 251/5 e 257) de Fusarium oxysporum não patogênico ao tomateiro (Lycopersicon esculentum), no controle da murcha vascular causada por Fusarium oxysporum f. sp. lycopersici, raça 2 em plântulas de tomateiro cv. Viradoro. Para verificar o efeito dos isolados de F. oxysporum não patogênicos, o sistema radicular de plântulas de tomateiro, com 30 dias de idade, foi imerso na suspensão de conídios (10(6) ml-1) e as mudas transplantadas para substrato de cultivo. Após 35 dias do transplante foi verificado que esses isolados não foram patogênicos às plantas de tomateiro, nem afetaram o desenvolvimento das mudas. A eficiência dos isolados de Fusarium oxysporum não patogênicos no controle da murcha foi determinada imergindo-se as raízes de mudas de tomateiro em suspensão de conídios (10(6) conídios ml-1) e transplantando-as em substratos previamente infestados com os isolados de F. oxysporum f.sp. lycopersici, raça 2 (10(5) conídios ml-1 de substrato). Transcorridos 35 dias do transplante, foram realizadas as avaliações da severidade na escala de 1=planta sadia a 6=planta morta ou com vasos coloridos e folhas murchas até o ponteiro e altura das mudas. Os isolados de F. oxysporum não patogênicos foram eficientes em reduzir a severidade da doença e em manter normal o seu desenvolvimento. Esses resultados evidenciam a atividade antagônica dos isolados de F. oxysporum não patogênico no controle da murcha vascular do tomateiro, causada por Fusarium oxysporum f. sp. lycopersici raça 2.

A sample of 471 pre-school children who frequented schools and creches in a poor district of Manaus (Amazonas), Brazil, were randomly submitted to faecal parasitological tests. Two-hundred-and-forty children from both sexes between the ages of 3 and 7 years with Ascaris lumbricoides and/or Giardia lamblia were selected. The objective of the study was to determine the possible influence of these two intestinal parasites and vitamin A and/or zinc supplementation on the serum retinol levels of primary school children. The children were submitted to clinical and anthropometric examinations, dietary interviews and biochemical examinations of retinol and carotene in the serum and of zinc in the hair. The parasitic incidence was 85.0% and about 54% of the children were polyparasitic. During the pretreatment phase, the retinol and carotene serum levels were 36% and 57%, respectively, below the normal levels. Using the Waterlow classification, the anthropometric analyses revealed that 88% of the children showed normal growth. A significant effect was observed of the anti-parasitic medicine on the serum retinol levels.
