sigma(S) is responsible for the transcriptional regulation of genes related to protection against stresses and bacterial survival and it accumulates in the cell under conditions of stress, such as nutrient limitation. An increase in the levels of sigma(S) causes a reduction in the expression of genes that are transcribed by RNA polymerase associated with the principal sigma factor, sigma(70). phoA, that encodes alkaline phosphatase (AP) is expressed under phosphate shortage conditions, and is also repressed by sigma(S). Here we show that in a Pi-limited chemostat, accumulation of rpoS mutations is proportional to the intrinsic level of sigma(S) in the cells. Acquisition of mutations in rpoS relieves repression of the PHO genes. We also devised a non-destructive method based on the rpoS effect on AP that differentiates between rpo(S+) and rpoS mutants, as well as between high and low-sigma(S) producers. Using this method, we provide evidence that sigma(S) contributes to the repression of AP under conditions of Pi excess and that AP variation among different strains is at least partly due to intrinsic variation in sigma(S) levels. Consequently, a simple and non-destructive AP assay can be employed to differentiate between strains expressing different levels of sigma(S) on agar plates.

A non-destructive sorting method to separate single-walled carbon nanotubes (SWNTs) by diameter was recently proposed. By this method, SWNTs are suspended in water by surfactant encapsulation and the separation is carried out by ultracentrifugation in a density gradient. SWNTs of different diameters are distributed according to their densities along the centrifuge tube. A mixture of two anionic surfactants, namely sodium dodecylsulfate (SDS) and sodium cholate (SC), presented the best performance in discriminating nanotubes by diameter. Unexpectedly, small diameter nanotubes are found at the low density part of the centrifuge tube. We present molecular dynamics studies of the water-surfactant-SWNT system to investigate the role of surfactants in the sorting process. We found that surfactants can actually be attracted towards the interior of the nanotube cage, depending on the relationship between the surfactant radius of gyration and the nanotube diameter. The dynamics at room temperature showed that, as the amphiphile moves to the hollow cage, water molecules are dragged together, thereby promoting the nanotube filling. The resulting densities of filled SWNT are in agreement with measured densities.

Steatosis is diagnosed on the basis of the macroscopic aspect of the liver evaluated by the surgeon at the time of organ extraction or by means of a frozen biopsy. In the present study, the applicability of laser-induced fluorescence (LIF) spectroscopy was investigated as a method for the diagnosis of different degrees of steatosis experimentally induced in rats. Rats received a high-lipid diet for different periods of time. The animals were divided into groups according to the degree of induced steatosis diagnosis by histology. The concentration of fat in the liver was correlated with LIF by means of the steatosis fluorescence factor (SFF). The histology classification, according to liver fat concentration was, Severe Steatosis, Moderate Steatosis, Mild Steatosis and Control (no liver steatosis). Fluorescence intensity could be directly correlated with fat content. It was possible to estimate an average of fluorescence intensity variable by means of different confidence intervals (P=95%) for each steatosis group. SFF was significantly higher in the Severe Steatosis group (P < 0.001) compared with the Moderate Steatosis, Mild Steatosis and Control groups. The various degrees of steatosis could be directly correlated with SFF. LIF spectroscopy proved to be a method capable of identifying the degree of hepatic steatosis in this animal model, and has the potential of clinical application for non-invasive evaluation of the degree of steatosis.

A robust, direct, rapid and non-destructive X-ray diffraction crystallography method to detect the polyprenylated benzophenones 7-epi-clusianone (1) and guttiferone A (2) in extracts from Garcinia brasiliensis is presented. Powder samples of benzophenones 1 and 2, dried hexane extracts from G. brasiliensis seeds and fruit`s pericarp, and the dried ethanolic extract from G. brasiliensis seeds were unambiguously characterized by powder X-ray diffractometry. The calculated X-ray diffraction peaks from crystal structures of analytes 1 and 2, previously determined by single-crystal X-ray diffraction technique, were overlaid to those of the experimental powder diffractograms, providing a practical identification of these compounds in the analyzed material and confirming the pure contents of the powder samples. Using the X-ray diffraction crystallography method, the studied polyprenylated benzophenones were selectively and simultaneously detected in the extracts which were mounted directly on sample holder. In addition, reference materials of the analytes were not required for analyses since the crystal structures of the compounds are known. High performance liquid chromatography analyses also were comparatively carried out to quantify the analytes in the same plant extracts showing to be in agreement with X-ray diffraction crystallography method. The motivation for this thesis work is the need for improving reliability of equipment and quality of service to railway passengers as well as a requirement for cost-effective and efficient condition maintenance management for rail transportation. This thesis work develops a fusion of various machine vision analysis methods to achieve high performance in automation of wooden rail track inspection.The condition monitoring in rail transport is done manually by a human operator where people rely on inference systems and assumptions to develop conclusions. The use of conditional monitoring allows maintenance to be scheduled, or other actions to be taken to avoid the consequences of failure, before the failure occurs. Manual or automated condition monitoring of materials in fields of public transportation like railway, aerial navigation, traffic safety, etc, where safety is of prior importance needs non-destructive testing (NDT).In general, wooden railway sleeper inspection is done manually by a human operator, by moving along the rail sleeper and gathering information by visual and sound analysis for examining the presence of cracks. Human inspectors working on lines visually inspect wooden rails to judge the quality of rail sleeper. In this project work the machine vision system is developed based on the manual visual analysis system, which uses digital cameras and image processing software to perform similar manual inspections. As the manual inspection requires much effort and is expected to be error prone sometimes and also appears difficult to discriminate even for a human operator by the frequent changes in inspected material. The machine vision system developed classifies the condition of material by examining individual pixels of images, processing them and attempting to develop conclusions with the assistance of knowledge bases and features.A pattern recognition approach is developed based on the methodological knowledge from manual procedure. The pattern recognition approach for this thesis work was developed and achieved by a non destructive testing method to identify the flaws in manually done condition monitoring of sleepers.In this method, a test vehicle is designed to capture sleeper images similar to visual inspection by human operator and the raw data for pattern recognition approach is provided from the captured images of the wooden sleepers. The data from the NDT method were further processed and appropriate features were extracted.The collection of data by the NDT method is to achieve high accuracy in reliable classification results. A key idea is to use the non supervised classifier based on the features extracted from the method to discriminate the condition of wooden sleepers in to either good or bad. Self organising map is used as classifier for the wooden sleeper classification.In order to achieve greater integration, the data collected by the machine vision system was made to interface with one another by a strategy called fusion. Data fusion was looked in at two different levels namely sensor-level fusion, feature- level fusion. As the goal was to reduce the accuracy of the human error on the rail sleeper classification as good or bad the results obtained by the feature-level fusion compared to that of the results of actual classification were satisfactory.

Since the last decade the problem of surface inspection has been receiving great attention from the scientific community, the quality control and the maintenance of products are key points in several industrial applications.The railway associations spent much money to check the railway infrastructure. The railway infrastructure is a particular field in which the periodical surface inspection can help the operator to prevent critical situations. The maintenance and monitoring of this infrastructure is an important aspect for railway association.That is why the surface inspection of railway also makes importance to the railroad authority to investigate track components, identify problems and finding out the way that how to solve these problems. In railway industry, usually the problems find in railway sleepers, overhead, fastener, rail head, switching and crossing and in ballast section as well. In this thesis work, I have reviewed some research papers based on AI techniques together with NDT techniques which are able to collect data from the test object without making any damage. The research works which I have reviewed and demonstrated that by adopting the AI based system, it is almost possible to solve all the problems and this system is very much reliable and efficient for diagnose problems of this transportation domain. I have reviewed solutions provided by different companies based on AI techniques, their products and reviewed some white papers provided by some of those companies. AI based techniques likemachine vision, stereo vision, laser based techniques and neural network are used in most cases to solve the problems which are performed by the railway engineers.The problems in railway handled by the AI based techniques performed by NDT approach which is a very broad, interdisciplinary field that plays a critical role in assuring that structural components and systems perform their function in a reliable and cost effective fashion. The NDT approach ensures the uniformity, quality and serviceability of materials without causing any damage of that materials is being tested. This testing methods use some way to test product like, Visual and Optical testing, Radiography, Magnetic particle testing, Ultrasonic testing, Penetrate testing, electro mechanic testing and acoustic emission testing etc. The inspection procedure has done periodically because of better maintenance. This inspection procedure done by the railway engineers manually with the aid of AI based techniques.The main idea of thesis work is to demonstrate how the problems can be reduced of thistransportation area based on the works done by different researchers and companies. And I have also provided some ideas and comments according to those works and trying to provide some proposal to use better inspection method where it is needed.The scope of this thesis work is automatic interpretation of data from NDT, with the goal of detecting flaws accurately and efficiently. AI techniques such as neural networks, machine vision, knowledge-based systems and fuzzy logic were applied to a wide spectrum of problems in this area. Another scope is to provide an insight into possible research methods concerning railway sleeper, fastener, ballast and overhead inspection by automatic interpretation of data.In this thesis work, I have discussed about problems which are arise in railway sleepers,fastener, and overhead and ballasted track. For this reason I have reviewed some research papers related with these areas and demonstrated how their systems works and the results of those systems. After all the demonstrations were taking place of the advantages of using AI techniques in contrast with those manual systems exist previously.This work aims to summarize the findings of a large number of research papers deploying artificial intelligence (AI) techniques for the automatic interpretation of data from nondestructive testing (NDT). Problems in rail transport domain are mainly discussed in this work. The overall work of this paper goes to the inspection of railway sleepers, fastener, ballast and overhead.

The report examines the factors which may be a contributing cause to the problems that are present when ferritic stainless steel are eddy current tested in a warm condition. The work is carried out at Fagersta Stainless AB in Fagersta which manufactures stainless steel wire. In the rolling mill there is an eddy current equipment for detection of surface defects on the wire. The ferritic stainless steels cause a noise when testing and this noise complicates the detection of defects.Because of this, a study was made of how the noise related to factors such as steel grade, temperature, size and velocity. By observing the signal and with the possibilities to change the equipment settings the capability to let a signal filter reduce the noise level were evaluated. Theories about the material's physical properties have also been included, mainly the magnetic properties, electrical conductivity and the material's tendency to oxidize.Results from the tests show that a number of factors do not affect the inductive test significantly and to use a filter to reduce the noise level does not seem to be a viable option. The level of noise does not relate to the presence of superficial particles in form of oxides.The ferritic stainless steels showed some difference in noise level. Which noise level there was did match well with the steels probability for a precipitation of a second phase, and precipitation of austenite may in this case contribute to noise when using an eddy current instrument.The noise is probably due to some physical material property that varies within the thread.

Tourism as an example of alternative for sustainable development in the Lençóis Maranhenses. The objective of this work is to analyze the prospects for sustainable development in tourism in the aforementioned region. An attempt is made to evaluate the five dimensions of sustainable - ecological, economic, social, space and cultural. The base for discussion is the information gathered from the people who actually live in the region. A type of the case study was built up. The research involved speaking with the people about the viability of tourism projects, not only with an eye on economic success, job creation and income for the region. There was also an attempt to tackle the issue of social and environment impact. Data was collected through verbal contact. A variety of people were interviewed, locals, businessmen, authorities, civil representatives and they spoke about the advantages and disadvantages of tourism as the saw it and as it affected them. Their answers and comments constitute the knowledge gleaned. The development of possible new strategies is the result of this work. Strategies that should be investigated by the local government/public administration. One such strategy would involve strengthening ties whit society and hoping that the improved dialogue would encourage sustainability. Another would mean improving interdepartmental contact to streamline the performance of public bodies so that development can be better planned and non-destructive.

This work deals with the development of a prototype of a helicopter quadrotor for monitoring applications in oil facilities. Anomaly detection problems can be resolved through monitoringmissions performed by a suitably instrumented quadrotor, i.e. infrared thermosensors should be embedded. The proposed monitoring system aims to reduce accidents as well as to make possible the use of non-destructive techniques for detection and location of leaks caused by corrosion. To this end, the implementation of a prototype, its stabilization and a navigation strategy have been proposed. The control strategy is based on dividing the problem into two control hierarchical levels: the lower level stabilizes the angles and the altitude of the vehicle at the desired values, while the higher one provide appropriate references signals to the lower level in order the quadrotor performs the desired movements. The navigation strategy for helicopter quadrotor is made using information provided by a acquisition image system (monocular camera) embedded onto the helicopter. Considering that the low-level control has been solved, the proposed vision-based navigation technique treats the problem as high level control strategies, such as, relative position control, trajectory generation and trajectory tracking. For the position control we use a control technique for visual servoing based on image features. The trajectory generation is done in a offline step, which is a visual trajectory composed of a sequence of images. For the trajectory tracking problem is proposed a control strategy by continuous servovision, thus enabling a navigation strategy without metric maps. Simulation and experimental results are presented to validate the proposal

Numerosas pesquisas têm estudado os métodos não-destrutivos de avaliação de materiais e sua aplicação àqueles de matrizes complexas, como é o caso da madeira. Um dos primeiros métodos não-destrutivos investigados para aplicação nesses casos foi o da vibração transversal. Apesar de sua concepção simples, e a despeito dos grandes avanços obtidos nessa área com outros métodos, como, por exemplo, o ultra-som, o método de vibração transversal para a determinação do módulo de elasticidade da madeira revela-se como de grande potencial de aplicação, sobretudo pela precisão do modelo matemático a ele associado e pela possibilidade de sua aplicação a peças de dimensões estruturais (in-grade testing). Neste trabalho, apresenta-se o uso desse método na determinação do módulo de elasticidade de três espécies de eucalipto. Foram ensaiados não-destrutivamente e por ensaios mecânicos convencionais de flexão corpos-de-prova de 2 cm x 2 cm x 46 cm de E. grandis, E. saligna e E. citriodora. Os ensaios não-destrutivos foram conduzidos com uso do sistema BING - Beam Identification by Non-destructive Grading, que permite a análise das vibrações do material nos domínios do tempo e da freqüência. Os resultados obtidos revelaram boa correlação entre os dois tipos de ensaios empregados, justificando o início dos ensaios com peças de dimensões estruturais, para a viabilização da técnica nas práticas de classificação estrutural.

A madeira de Pinus sp. tem utilização crescente na indústria madeireira brasileira. O decréscimo constante do suprimento de árvores adultas com grandes diâmetros, provenientes de florestas naturais, tornou comum a produção de madeira em ciclos curtos, com grande proporção de madeira juvenil. Resultados de diversas pesquisas têm reportado que o módulo de elasticidade e a resistência a diferentes solicitações mecânicas são seriamente afetados pela presença de madeira juvenil. Este trabalho teve por objetivo determinar o módulo de elasticidade da madeira juvenil e adulta de Pinus taeda L. a partir da constante dinâmica C LL, obtida em ensaios não-destrutivos de ultra-som. A madeira de P. taeda era originária de plantios da Estação Experimental de Itapeva - SP, sendo amostrados seis indivíduos arbóreos com 34 anos de idade. Os corpos-de-prova (4 cm x 4 cm x 45 cm) foram obtidos separadamente das regiões de madeira juvenil e adulta da prancha central, previamente submetida à secagem industrial (umidade final de 12%), para a determinação da constante dinâmica por meio de ensaios de ultra-som. Para avaliar a sensibilidade do método do ultra-som, os corpos-de-prova foram ensaiados destrutivamente à compressão paralela. Os resultados mostraram boa sensibilidade do método do ultra-som (R² » 0,90) na avaliação desse parâmetro mecânico da madeira juvenil e adulta.

Currently there is still a high demand for quality control in manufacturing processes of mechanical parts. This keeps alive the need for the inspection activity of final products ranging from dimensional analysis to chemical composition of products. Usually this task may be done through various nondestructive and destructive methods that ensure the integrity of the parts. The result generated by these modern inspection tools ends up not being able to geometrically define the real damage and, therefore, cannot be properly displayed on a computing environment screen. Virtual 3D visualization may help identify damage that would hardly be detected by any other methods. One may find some commercial softwares that seek to address the stages of a design and simulation of mechanical parts in order to predict possible damages trying to diminish potential undesirable events. However, the challenge of developing softwares capable of integrating the various design activities, product inspection, results of non-destructive testing as well as the simulation of damage still needs the attention of researchers. This was the motivation to conduct a methodological study for implementation of a versatile CAD/CAE computer kernel capable of helping programmers in developing softwares applied to the activities of design and simulation of mechanics parts under stress. In this research it is presented interesting results obtained from the use of the developed kernel showing that it was successfully applied to case studies of design including parts presenting specific geometries, namely: mechanical prostheses, heat exchangers and piping of oil and gas. Finally, the conclusions regarding the experience of merging CAD and CAE theories to develop the kernel, so as to result in a tool adaptable to various applications of the metalworking industry are presented

O uso de cápsulas porosas para extração de solução do solo é interessante por ser um ensaio não-destrutivo. Entretanto, persistem dúvidas sobre a liberação de íons da própria cápsula, que podem contaminar a solução extraída. Foram realizados testes na Faculdade de Ciências Agronômicas/UNESP, Botucatu (SP), com o objetivo de verificar a liberação de Ca e de Mg por cápsulas porosas de porcelana. No primeiro, foram empregados quatro tratamentos: T1 - lavagem das cápsulas com água destilada, forçando sua passagem pelas cápsulas, utilizando uma bomba a vácuo; T2 - lavagem das cápsulas com HCl 0,1 mol L-1, forçando sua passagem pelas cápsulas, utilizando uma bomba a vácuo; T3 - lavagem das cápsulas com água destilada, sem vácuo, e T4 - lavagem das cápsulas com HCl 0,1 mol L-1, sem vácuo, em um tempo de imersão de 24 h (para T3 e T4). No segundo teste, as cápsulas tratadas com HCl 0,1 mol L-1 no primeiro teste foram lavadas com água destilada novamente e deixadas de molho em água destilada e deionizada durante 45 min. Após estes procedimentos, as cápsulas foram imersas em água destilada, tomando-se alícotas desta solução para determinação de pH e dos teores de Ca e Mg após contato com as cápsulas por 0; 0,5; 1,0; 2,0; 4,0 e 24 h. Cada tratamento teve quatro repetições. Não se observou liberação significativa de Ca e Mg das cápsulas porosas para a solução, quando foram preparadas utilizando-se da passagem de HCl 0,1 mol L-1 a vácuo e água destilada, e deixadas, a seguir, em água destilada e deionizada durante 4 h.

The continuous inputs from agriculture areas and human or industrial sewer have been provided a widely demographic explosion of tropical tanner-grass and para grasss plants in the rivers and water reservoirs of Brazil. Considering the importance of these noxious weeds there is a vast necessity of basic studies on aspects related to reproduction, growing, development, nutritional requirements, responses to control systems and others. In most studies, leaf area knowledge is crucial and it is one of the most difficult characteristics to be measured because it usually requires expensive equipments or it uses a destructive technique. The objective of this work was to develop equations for leaf blade dimensional parameters to allow determination of the leaf area of African tanner-grass (Brachiaria subquadripara (Trin.) Hitchc) and para grass (Brachiaria mutica (Forsk.) Stapf.). Correlations between real leaf area and dimensional parameters of leaf blade such as length along the main vein (L) and maximum width (W) perpendicular to the main axis were studied. Only the linear equations allowed a good leaf area estimate. It is suggested to opt for a simple linear equation involving respective products of length times maximum width considering linear coefficient equal to zero. Thus, B. subquadripara (SS) and B. mutica (SM) non destructive leaf blade area estimates may be performed by the following formulas: SS = 0.7719 (L x W) and SM = 0.7479 (L x W), with coefficients of determination (R(2)) of 0.9594 and 0.9408, respectively.

O clorofilômetro (SPAD-502, Minolta) é um aparelho portátil que permite obtenção do índice relativo de clorofila (IRC) de modo simples, rápido e não destrutivo no próprio campo, o qual se correlaciona com a concentração de N em várias culturas, e que pode tornar mais rápida a correção de deficiência, otimizando a utilização da fertirrigação. Objetivou-se com este trabalho verificar se o IRC pode ser indicativo do estado nutricional em N, podendo auxiliar no ajuste da fertirrigação nitrogenada durante o ciclo do cafeeiro. O experimento foi desenvolvido em Botucatu, SP, em área com cafeeiros do cultivar Catuaí Vermelho, com dois anos, no espaçamento de 2,5 x 0,8 m e irrigado por gotejamento. O delineamento experimental utilizado foi em blocos casualizados, com quatro repetições, aplicando-se um tratamento sem adubação, mas com irrigação, e mais quatro tratamentos utilizando fertirrigação com 33, 66, 100 e 133 % da dose de N recomendada, parcelada semanalmente (52,8; 105,6; 160,0; e 212,7 kg ha-1, respectivamente). O IRC aumentou linearmente com as doses de N, e a produtividade correlacionou-se significativamente com o IRC, do florescimento até a colheita, porém não com a concentração de N foliar. Os IRCs nos cafeeiros com maior produtividade foram de 81,5 a 83,2 (florescimento e início de expansão dos frutos), de 76,2 a 78,3 (expansão dos frutos), de 68,3 a 69,8 (início da granação), de 64,0 a 65,9 (na granação) e de 61,7 a 62,7 unidades SPAD (na maturação). O clorofilômetro pode ser utilizado para definir a probabilidade de resposta ao N no decorrer do ciclo do cafeeiro.
