A heparina foi isolada no início do século XX e permanece, até os dias atuais, como um dos mais importantes e eficientes agentes terapêuticos de ação antitrombótica. Sua atividade anticoagulante deve-se à ativação da antitrombina (AT), uma serpina responsável pela inibição fisiológica de serino-proteinases plasmáticas, tais como fIIa e fXa. Os esforços no sentido da elucidação dos aspectos estruturais e dinâmicos associados ao reconhecimento molecular da heparina pela AT, em nível atômico, vêm encontrando diversas dificuldades, principalmente associadas aos compostos sacarídicos. Em decorrência de sua elevada polaridade e flexibilidade, glicosaminoglicanos como a heparina são difíceis de estudar e modelar. Soma-se a isto o fato de que os resíduos de iduronato presentes na heparina (IdoA) apresentam um incomum equilíbrio conformacional entre estados de cadeira (1C4) e bote-torcido (2SO), sendo esta última estrutura postulada como a possível conformação bioativa. Sendo assim, este trabalho apresenta um estudo de modelagem molecular do perfil conformacional da heparina, tanto em solução quanto complexada à AT, utilizando cálculos ab initio e simulações de dinâmica molecular (DM) Em decorrência da ausência de parâmetros capazes de descrever polissacarídeos nos campos de força atualmente disponíveis, cargas atômicas foram geradas, utilizando-se os esquemas de Mulliken, Löwdin e cargas ajustadas ao Potencial Eletrostático, através de cálculos quantum-mecânicos na base 6-31G** e testadas na simulação de DM da heparina. Diversas condições de simulação, tais como modelos de água, concentrações de sais e as conformações 1C4 e 2SO do IdoA, foram avaliadas de forma a identificar as melhores condições para a descrição conformacional da heparina em solução. O protocolo de DM obtido foi então utilizado no estudo do complexo AT-heparina. Os resultados obtidos estão de acordo com os dados experimentais atualmente disponíveis acerca da conformação da heparina e da contribuição energética de cada resíduo de aminoácido da AT para a formação do complexo com a heparina. A partir dos cálculos ab initio realizados, foi proposto um refinamento na estrutura da heparina determinada por RMN, enquanto que as simulações de DM permitiram reinterpretar aspectos da estrutura tridimensional da AT determinada por cristalografia de raios-X. Adicionalmente, os dados obtidos sugerem que não há requerimento conformacional para a interação do IdoA com a AT Globalmente, os dados indicam que simulações de DM podem ser utilizadas para representar adequadamente a conformação da heparina, assim como para caracterizar e quantificar suas interações com a AT. Assim sendo, propomos o uso de simulações de DM do complexo entre a AT e a heparina, ou entre a AT e compostos derivados da heparina, como uma ferramenta útil no processo de desenvolvimento de novos agentes antitrombóticos e anticoagulantes.

O ramo de lavagem de roupas é um importante setor de serviços na sociedade moderna e responsável por uma parcela significativa no consumo de água no meio urbano. No entanto, são poucas as empresas que se preocupam em lançar seus efluentes dentro de um padrão de qualidade estabelecido pelos órgãos ambientais ou, ainda, reciclar a água no processo de lavagem. Portanto, o objetivo do presente trabalho foi estudar o tratamento e a reciclagem do efluente gerado por uma lavanderia industrial. Assim, realizou-se inicialmente um estudo de caracterização do efluente e uma investigação, em laboratório, das diferentes alternativas de tratamento, entre as quais a coagulação/floculação, adsorção/coagulação/ floculação e a Reação de Fenton, os quais foram avaliados em relação a padrões físicoquímicos e ecotoxicológicos de qualidade. Nas condições otimizadas em laboratório, realizou-se o tratamento de efluentes em escala industrial. Após, avaliou-se a possibilidade de reciclagem do efluente no próprio processo de lavagem de roupas e fez-se uma avaliação de custos considerando as diferentes alternativas de processos. Por fim, realizou-se um estudo de caracterização do lodo gerado. Os resultados obtidos demonstram que o efluente bruto da lavanderia não atinge os padrões físico-químicos exigidos pela legislação e apresenta um alto grau de toxicidade no meio aquático, devendo ser tratado para lançamento no corpo hídrico receptor. O efluente tratado por coagulação/floculação melhorou significativamente a qualidade da água, porém ainda apresentou resultados insatisfatórios quanto ao parâmetro surfactantes. Os índices toxicológicos melhoraram, porém ainda mostraram um nível de toxidade elevado. O efluente tratado por adsorção/coagulação/floculação apresentou bons resultados tanto nos parâmetros físicoquímicos quanto nos ensaios de toxicidade, mostrando ser a melhor opção de processo. A aplicação deste processo em escala industrial demonstrou que o processo apresenta uma grande confiabilidade em relação a todos os parâmetros de qualidade exigidos pelo órgão ambiental e a reciclagem do efluente pode ser realizada para fins de lavagem de roupas. O tratamento através da Reação de Fenton apresentou bons resultados em relação aos parâmetros físico-químicos, porém a presença de agentes oxidantes de forma residual foi tóxica sobre os organismos avaliados. A Reação de Fenton, apesar de gerar um efluente com boas características para reciclagem, apresenta um custo de insumos no processo bastante elevado. O lodo é um fator importante o processo de tratamento. Apresenta-se rico nos elementos alumínio e cálcio e é classificado, de acordo com a NBR 10.004, como um resíduo não perigoso (Classe II - Não Inerte). Esse lodo deve ser devidamente secado e envido para um aterro de resíduos industriais. Porém, apresenta potencial para reúso como matéria-prima na construção civil.

A lavagem de roupas é um importante serviço para a sociedade moderna. Os efluentes de lavanderias industriais, de maneira geral, contêm sujeiras removidas das roupas e substâncias adicionadas na lavagem que normalmente são tratados por processo físicoquímico de coagulação/floculação/sedimentação. Os sais de alumínio e os produtos convencionalmente empregados para a correção do pH são agentes inorgânicos não biodegradáveis que acrescentam elementos químicos à água ou ao lodo. Como principal dificuldade do processo destaca-se o lodo inorgânico gerado, de difícil manuseio por parte das empresas em função do volume gerado e do elevado teor de umidade. O objetivo geral da presente da pesquisa foi avaliar a aplicabilidade e a eficiência do uso de um coagulante vegetal, basicamente tanino modificado pela reação de Mannich, no tratamento do efluente de uma lavanderia industrial. Realizaram-se estudos de coagulação/floculação com reagentes tradicionais (sulfato de alumínio e poliacrilamida catiônica), coagulação/floculação com um coagulante alternativo (tanino catiônico e poliacrilamida aniônica) e adsorção/coagulação/floculação (carvão ativado em pó, tanino catiônico e poliacrilamida aniônica). A dosagem de reagentes foi otimizada através de “Teste de Jarros” e a eficiência de cada uma das alternativas foi avaliada em termos de remoção de sólidos sedimentáveis, sólidos suspensos, turbidez, DQO e surfactantes. Analisou-se a contribuição de cada um destes processos em termos de ânions cloretos e sulfatos residual bem como a toxicidade aguda para populações do microcrustáceo Daphnia similis e do peixe Pimephales promelas. A massa e o volume de lodo gerado em cada processo foi quantificado e o lodo caracterizado em termos de sua periculosidade conforme a NBR 10.004 e comportamento termogravimétrico Os resultados demonstram que, em termos da legislação vigente, o efluente bruto não atende aos padrões de carga orgânica (DQO) e surfactantes. Os ensaios ecotoxicológicos indicam que o efluente pode ser considerado extremamente tóxico para o microcrustáceo Daphnia similis, como para o peixe Pimephales promelas. O tratamento do efluente realizado por coagulação/floculação usando o tanino catiônico como agente coagulante remove os sólidos suspensos e uma fração considerável da carga orgânica e de surfactantes. Em termos da legislação vigente, o efluente somente não atende ao padrão referente ao lançamento de surfactantes. Os ensaios ecotoxicológicos indicam que efluente passa a ser considerado pouco tóxico para a Daphnia similis e ainda tóxico para o peixe Pimephales promelas. O tratamento do efluente por heteroagregação entre carvão ativado, tanino catiônico e polímero floculante permite remover os sólidos suspensos, e níveis mais significativos de carga orgânica e de surfactantes, atendendo a todos padrões de lançamento exigidos para este tipo de efluente. Os ensaios ecotoxicológicos indicam que efluente passa a ser considerado não tóxico para a Daphnia similis e para a Pimephales promelas. O lodo gerado com o uso do tanino catiônico como agente coagulante é classificado como um resíduo Não Inerte - Classe II conforme a NBR 10.004, unicamente devido ao fato de exceder a concentração de surfactantes no ensaio de solubilização conforme a NBR 10.006. O lodo apresenta ainda um alto teor de matéria orgânica, o que facilita a sua eliminação por tratamento térmico ou biológico Comparado ao agente coagulante tradicionalmente empregado para tal fim, o sulfato de alumínio, o tanino catiônico apresentou resultados em relação a qualidade do efluente tratado muito parecidos. Entretanto, pode-se citar algumas evidentes vantagens: menor custo, uso de uma matéria prima renovável, menor contribuição de ânions sulfatos ao efluente final, menor geração de massa de lodo, e obtenção de um lodo orgânico com maior facilidade de eliminação. Esses fatores todos permitem concluir que a substituição do sulfato de alumínio pelo tanino catiônico contribui para um processo de tratamento de efluentes mais limpo.

O presente trabalho, desenvolvido na estação de tratamento de água de Novo Hamburgo, avalia uma alternativa aos tradicionais coagulantes metálicos. Esta alternativa é um coagulante orgânico de origem vegetal produzido na região metropolitana de Porto Alegre. Este coagulante a base de tanino extraído da acácia negra tem o nome comercial de Veta organic. Foi verificada, além da qualidade da água tratada, do custo do tratamento e do impacto ambiental gerado durante o processo de potabilização da água, a operacionalidade da ETA com o coagulante. Foram pesquisados os fenômenos físico-químicos que ocorrem na coagulação. Foram feitas análises dos principais resíduos do tratamento (água de lavagem dos filtros e lodo dos decantadores), definindo alguns parâmetros para sua disposição no solo. A caracterização dos efluentes do tratamento confirmou alguns preceitos da literatura, como a relação DBO/DQO e o alto teor de umidade. Outros parâmetros apresentaram valores divergentes dos publicados em outros trabalhos, sendo, provavelmente, o principal motivo a substituição do coagulante. Foi traçado um comparativo com o sulfato de alumínio, determinando vantagens e desvantagens da substituição do sulfato de alumínio pelo Veta organic. Este comparativo considerou aspectos relacionados à operação e manutenção da ETA, segurança e salubridade dos envolvidos com o processo de tratamento, qualidade e quantidade da água tratada, volume e contaminação dos resíduos do tratamento, custo para tratar um metro cúbico de água e adequação às leis, normas e portarias referentes a abastecimento público de água e poluição por despejos industriais. Os resultados deste trabalho permitiram verificar a redução na concentração de alumínio na água tratada e nos resíduos oriundos do tratamento com Veta organic em relação ao tratamento com sulfato de alumínio. Também, constatou-se a redução no volume de lodo gerado nos decantadores com o uso do Veta organic se comparado com os valores para quantidade de produção de lodo com sulfato de alumínio definidos em literatura. Foi determinado o aumento no custo do tratamento na ETA-NH com a introdução do novo coagulante. Além disso, foram feitas sugestões referentes à operação da estação que podem aumentar a eficiência do tratamento com o Veta organic, tendo sido abordadas questões relativas ao processo de tratamento, verificadas durante o trabalho de campo, com uso deste produto. A alteração mais importante na operação da ETA e que trouxe mais benefícios no tratamento com Veta organic foi a utilização de ar na retrolavagem dos filtros, tendo sido verificada a redução da cor e turbidez da água de lavagem dos filtros, com o auxílio do ar na lavagem, até valores estáveis.

The occurrence of bioactive compounds in marine organisms comes awaking the interest of the pharmaceutical industry. Heparin, a sulfated polysaccharide which presence was already identified in several marine invertebrates, is very attractive due its remarkable functional versatility. Besides to intervene in blood coagulation, this molecule has a great anti-inflammatory potential. However, its strong anticoagulant activity difficult the clinical exploitation of its anti-inflammatory properties. Thus, the aims of this work were to evaluate the effect of a heparin-like compound (heparinoid), isolated from the cephalotorax of the Litopenaeus vannamei shrimp, on the inflammatory response, hemostasia and synthesis of antithrombotic heparan sulfate by endothelial cells, besides studying some aspects concerning its structure. The purified heparinoid was structurally characterized following an analytical boarding, involving electrophoresis and chromatography. The structural analysis have shown that this compound possess a high content of glucuronic acid residues and disulfated disaccharide units. In contrast to mammalian heparin, the heparinoid was incapable to stimulate the synthesis of heparan sulfate by endothelial cells in the tested concentrations, beyond to show reduced anticoagulant activity and hemorrhagic effect. In a model of acute inflammation, the compound isolated from the shrimp reduced more than 50% of the cellular infiltration. Besides reduce the activity of MMP-9 and proMMP-2 of the peritoneal lavage of inflamed animals, the heparinoid also reduced the activity of MMP-9 secreted by activated human leukocytes. These results demonstrate the potential of heparinoid from L. vannamei to intervene in the inflammatory response. For possessing reduced anticoagulant activity and hemorrhagic effect, this compound can serve as a structural model to direct the development of more specific therapeutical agents to the treatment of inflammatory diseases

Since the first description of sulfated polysaccharides from seaweeds, the biological activities of these compounds have been evaluated under different aspects and experimental procedures. Among the broad biological activities presented by seaweed polysaccharides, anticoagulant action appears as a promising function. In this present study we have obtained sulfated polysaccharides from the green seaweed Codium isthmocladium by proteolytic digestion, followed by separation into five fractions (0.3, 0.5, 0.7, 0.9 and 1.2) by sequential acetone precipitation. The chemical analyses have demonstrated that all fractions are composed mainly by sulfated polysaccharides. The anticoagulant activity of these fractions was determined by activated partial thromboplastin time (aPTT) and prothrombin time test (PT) using citrate normal human plasma. None fraction has shown anticoagulant activity by PT test. Furthermore, all of them have shown anticoagulant activity by aPTT test. These results indicated that the molecular targets of these sulfated polysaccharides are mainly in the intrinsic via of the coagulation cascade. Agarose gel electrophoresis in 1,3-diaminopropane acetate buffer, pH 9.0, stained with 0.1% toluidine blue showed the presence of two or three bands in several fractions while the fraction 0.9 showed a single spot. By anion exchange chromatography, the acid polysaccharides from the 0.9 acetone fraction were separated into two new fractions eluted respectively with 2.0 and 3.0 M NaCl. These compounds showed a molecular weight of 6.4 and 7.4 kDa respectively. Chemical analyses and infrared spectroscopy showed that Gal 1 and Gal 2 are sulfated homogalactans and differ one from the other in degree and localization of sulfate groups. aPPT test demonstrated that fractions 2,0 and 3,0M (Gal1 and Gal 2, respectively) have anticoagulant activity. This is the first time that anticoagulant sulfated homogalatans have been isolated from green algae. To prolong the coagulation time to double the baseline value in the aPTT, the required amount of sulfated galactan 1 (6,3mg) was similar to low molecular heparin Clexane®, whereas only 0,7mg of sulfated galactan 2 was needed to obtain the same effect. Sulfated galactan 2 in high doses (250mg) induces platelet aggregation. These results suggest that these galactans from C. isthmocladum have a potential application as an anticoagulant drug

In addition, pharmacological activities of PS from plants have not been carrying out. The aim of this work was extract PS from the angiosperm Halodule wrightii and study their anticoagulant and antioxidant activities. Histological analysis showed the presence of the PS manly in the roots. A polysaccharide-rich extract was obtained from H. wrightii by proteolysis followed by methanol and TCA precipitation. Chemical, infra-red analysis and agarose gel electrophoresis in 1.3 diaminopropane acetate buffer confirmed the presence of sulfated polysaccharides made by glucose, galactose, xylose and sulfate residues in the proportion 1: 0,9: 1: 1. In addition polyacrilamide electrophoresis have shown that extract is mainly compose by 11kDa sulfated polysaccharides. Pharmacological analysis have shown total antioxidant capacity (CAT) that resulted in 15,21 μg for equivalent of ascorbic acid, scavenging activity of the DPPH radical with 41,36 % of scavenging, activity of reducing power with the maximum of 0,290 nm (50 % of vitamin C activity) and scavenging activity superoxide radical (O2-) with a maximum of 32,23 %. Chelating activity of metal less than 4% and scavenging activity of the radical hydroxyl (OH-) less than 2%. Time of activated partial tromboplastin (aPTT) doubling the time of coagulation from 20μg of and protrombin time (PT) was not present. The data indicate that PS from Halodule wrightii could be considered for future applications in medicine, food production or cosmetic industry

Heparan sulfate (HS) and Heparin (Hep) glycosaminoglycans (GAGs) are heterogeneous and highly charged polysaccharides. HS is structurally related to Hep but is much less substituted with sulfo groups than heparin and has a more varied structure (or sequence). Because of structural similiarities between these two polymers, they have been described together as heparinoids . Both chains bind a variety of proteins and mediate various physiologically important processes including, blood coagulation, cell adhesion and growth factor regulation. Heparinoids with structural characteristics similar to these described from HS and/or Hep from mammalian tissues have been isolated from different species of invertebrates, although only a few heparinoids from unusual sources have been characterized. The present study describes the presence of unusual heparinoids population from Artemia franciscana, isolated after proteolysis and fractionation by ion exchange resin and named, F-3.0M. The study model in vivo were hemostasis (rat tail scarification) and inflamatoty activity. The tests in vitro were used for coagulations assays (PT and APTT). The analyse of the heparinoids eluted with 3,0M NaCl showed electrophoretic migration in different buffer systems a single band with a behaviour intermediate between those of mammalian HEP and HS. The main products obtained from Artemia heparinoids after enzymatic degradation with heparitinases I and II from F. heparinum were N-sulphated disaccharides (∆U-GlcNS,6S/ ∆U,2S-GlcNS and ∆U-GlcNS) and N-acetylated disaccharides (∆U, GlcNAc). This heparinoid had a lower hemorrhagic effect (400μg/ml) when compared to unfractiionated heparins(25μg/ml).The results also suggest a negligible APTT activity of this heparinoid (62.2s). No action was observed on PT indicating that F-3.0M haven t action on the extrinsic pathway. The results showed that the fraction F- 3.0M have inhibitory effect on migration of leukocytes, 64.5% in the concentration of 10 μg/ml (P<0.001). The search for new heparin and/or heparan sulphates analogs devoid of anticoagulant activity is an atractive alternative and may open up a wide variety of new therapeutic applications

The exopolysaccharides are extracellular compounds produced by some species of fungi and bacteria. It is suggested that these molecules, even when in the form of complex polysaccharide-peptide, are the main bioactive molecules of many fungus. Some of the biological activities displayed by these compounds can be accentuated and others may arise when you add chemically polar or nonpolar groups to polysaccharides. The fruiting body of Pleurotus sajor-caju produces a heteropolysaccharide with antineoplastic and antimicrobial activity, but other biological activities of this polymer have not been evaluated. In this work the exopolysaccharide of Pleurotus sajor-caju was sulfated chemically and structurally characterized. We also evaluated the antiproliferative, antioxidant and anticoagulant activities from native exopolysaccharide (PN) and its sulfated derivated (PS). Polyacrylamide gel electrophoresis, infrared spectroscopy and nuclear magnetic resonance (¹³C) proved successful in sulfation of PN to obtain PS. Analysis by gas chromatography-mass spectroscopy showed that PN and PS are composed of mannose, galactose, 3-O-methyl-galactose and glucose in proportion percentage of 44,9:16,3:19,8:19 and 49, 7:14,4:17,7:18,2, respectively. The percentage of sulfate found in PS was 22.5%. Antioxidants assays revealed that the sulfation procedure affects differently the activities of exopolysaccharides, while the total antioxidant capacity, the scavenging activity of superoxide radical and ferric chelating were not affected by sulfation, on the other hand the chemical modification of PN enhanced the scavenging activity of hydroxyl radical and reducing power. PS also showed anticoagulant activity in a dose-dependent manner and clotting time was 3.0 times higher than the baseline value in APTT at 2 mg/mL. The exopolysaccharide not presented antiproliferative activity against HeLa tumor cells, but PS affects the cellular proliferation in a time-dependent manner. After 72 h, the inhibition rate of PS (2.0 mg/mL) on HeLa cells was about 60%. The results showed that PN sulfation increase some of their activities.

The corn cob is an agricultural by-product still little used, this in part due to the low knowledge of the biotechnological potential of their molecules. Xylan from corn cobs (XSM) is a polysaccharide present in greater quantity in the structure of plant and its biotechnology potential is little known. This study aimed to the extraction, chemical characterization and evaluation of biological activities of xylan from corn cobs. To this end, corncobs were cleaned, cut, dried and crushed, resulting in flour. This was subjected to a methodology that combines the use of alkaline conditions with waves of ultrasound. After methanol precipitation, centrifugation and drying was obtained a yield of 40% (g/g flour). Chemical analysis indicated a high percentage of polysaccharides in the sample (60%) and low contamination by protein (0.4%) and phenolic compounds (> 0.01%). Analysis of monosaccharide composition indicated the presence of xylose:glucose:arabinose:galactose:mannose:glucuronic acid in a molar ratio 50:20:15:10:2.5:2.5. The presence of xylan in the sample was confirmed by nuclear magnetic resonance (¹H and ¹³C) and infrared spectroscopy (IR). Tests were conducted to evaluate the antioxidant potential of XSM. This showed a total antioxidant capacity of 48.45 EAA/g sample. However, did not show scavenging activity of superoxide and hydroxyl radical and also reducing power. But, showing a high capacity chelating iron ions with 70% with about 2 mg/mL. The ability to XSM to influence cell proliferation in culture was also evaluated. This polymer did not influence the proliferation of normal fibroblast cells (3T3), however, decreased the rate of proliferation of tumor cells (HeLa) in a dose-dependent, reaching an inhibition of about 50% with a concentration around 2 mg/mL. Analyzing proteins related to cell death, by immunoblotting, XSM increases the amount of Bax, Bcl-2 decrease, increase cytochrome c and AIF, and reduce pro-caspase-3, indicating the induction of cell death induced apoptosis dependent and independent of caspase. XSM did not show anticoagulant activity in the PT test. However, the test of activated partial thromboplastin time (aPTT), XSM increased clotting time at about 5 times with 600 μg of sample compared with the negative control. The presence of sulfate on the XSM was discarded by agarose gel electrophoresis and IR. After carboxyl-reduction of XSM the anticoagulant activity decreased dramatically. The data of this study demonstrate that XSM has potential as antioxidant, antiproliferative and anticoagulant compound. Future studies to characterize these activities of XSM will help to increase knowledge about this molecule extracted from corn and allow their use in functional foods, pharmaceuticals and chemical industries.

The present study examines the chemical composition and their effects on free radicals, inflammation, angiogenesis, coagulation, VEGF effects and cellular proliferation of a polysaccharides from alga Sargassum vulgare. The sulfated polysaccharide was extracted from brown seaweed by proteolysis with enzymes maxataze. The presence of proteins and sugars were observed in crude polysaccharides. Fractionation of this crude extract was made with growing concentration of acetone (0.3-1.5 v) and produced four groups of polysaccharides. Anionic polysaccharides from brown seaweed Sargassum vulgare, SV1and PSV1 were fractionated (SV1) and purified (PSV1), and displayed with high total sugars and sulfate content and very low level of protein. This fucan SV1 contains low levels of protein and high carbohydrate and sulfate content. This polysaccharides prolonged activated partial thromboplastin time (aPTT) at 50 μg (>240 s). SV1 was found to have no effect on prothrombin time (PT), corresponding to the extrinsic pathway of coagulation. SV1 exhibits high antithrombotic action in vivo, with a concentration ten times higher than heparin. Polysaccharides from S. vulgare promoted direct inhibition enzymatic activity of thrombin and stimulated enzymatic activity of FXa. SV1 showed optimal inhibitory activity of thrombin (50.2±0.28%) at a concentration of 25 μg/mL. Its antioxidant action on scavenging radicals by DPPH was (22%), indicating the polymer has no cytotoxic action (hemolytic) on ABO and Rh blood types in different erythrocyte groups and displays strong anti-inflammatory action on all concentrations tested in the carrageenan-induced paw edema model, demonstrated by reduced edema and cellular infiltration. Angiogenesis is a dynamic process of proliferation and differentiation. It requires endothelial proliferation, migration, and tube formation. In this context, endothelial cells are a preferred target for several studies and therapies. The antiangiogenic efficacy of polysaccharides was examined in vivo in the chick chorioallantoic membrane (CAM) model by using fertilized eggs. Decreases in the density of the capillaries were assessed and scored. The results showed that SV1 and PSV1 have an inhibitory effect on angiogenesis. These results were also confirmed by inhibition tubulogenesis in rabbit aorta endothelial cell (RAEC) in matrigel. These compounds were assessed in Apoptosis assay (Annexin V - FITC / PI) and cell viability by MTT assay of RAEC. These polysaccharides do not affect the viability and do not have apoptotic or necrotic action. RAEC cell when incubated with SV1 and PSV1showed inhibition of VEGF secretion, observed when compounds were incubated at 25, 50 and 100 μg/μL. The VEGF secretion with the RAEC cell line for 24 h, was more effective for PSV1 at 50 μg/μL(71.4%) than SV1 100 μg/μL (75.9%). SV1 and PSV1 had an antiproliferative action (47%) against tumor cell line HeLa. Our results indicate that these sulfated polysaccharides have antiangiogenic and antitumoral actions

In the present study, extracts rich-sulfated polysaccharides were obtained from three different species of Dictyotales (a class of brown macroalgae): Canistrocarpus cervicornis, Dictyota mertensii and Dictyopteris delicatula and their anticoagulant and antioxidant activities were evaluated. All extracts showed anticoagulant activity on aPTT assay, but not on PT assay. Extracts also exhibited total antioxidant activity, superoxide radical scavenging capacity and ferric chelating property. The extract from C. cervicornis showed the best results and was choose to have their sulfated polysaccharides fractioned and subsequently analysed. Thus, six fractions (CC-0.3, CC-0.5, CC-0.7, CC-1.0, CC-1.2 and CC-2.0) were obtained by proteolysis followed by sequential acetone precipitation. Agarose gel eletrophoresis stained with blue toluidine, confirmed the presence of sulfated polysaccharides in all fractions. Chemical analyses showed that all fractions presented heterofucans mainly constitued by fucose, galactose, glucuronic acid and sulfate. Any fraction changed the PT. However, all fractions were able to double the aPTT on a dose-dependent manner. CC- 0.3, CC-0.5, CC-0.7 and CC-1.0 needed only 0.100 mg/mL to double the aPTT, result only 1.25 times higher than the Clexane® (0.080 mg/mL), a commercial low molecular heparin. The heterofucans presented appreciable total antioxidant capacity, low capacity on scavenging hydroxyl radical and good efficiency on scavenging superoxide radicals (except CC-1.0). CC-1.2 showed 43.1 % on superoxide radical scavenging. This result was higher than that showed by the same concentration of gallic acid (41.8 %), a known antioxidant. Furthermore, the heterofucans showed excelent activity on ferrous chelating activity (except CC-0.3). CC-0.5, CC-0.7 and CC-1.0 showed the highest activities with 47.0 % of ferrous chelating activity, a result 2.0 times lesser than that exhibited by the same concentration of EDTA. These results clearly indicated the beneficial effects of heterofucans extracted from C. cervicornis as potential anticoagulant and antioxidant agents. However additional steps of purification, structural studies, besides in vivo experiments are needed for these fucans may be used as therapeutic agents

Proteinases are enzymes distributed widely founded in several organisms and perform many different functions, from maintaining homeostasis to the worsening of some diseases such as cancer, autoimmune diseases and infections. The proteins responsible of controlling the action of these enzymes are the inhibitors, that are classified based on their target proteases and are founded since simple organisms, such as bacteria, to higher organisms, such as larger plants and mammals. Plant proteinase inhibitors act by reducing or inactivating the activity of target proteases, thus, these proteins have been studied as potential tools in the treatment of diseases related to protease activities. In this context, an inhibitor of chymotrypsin from Erythrina velutina, called EvCI was previously purified and it was observed that this protein plays in vitro anticoagulant activity and anti-inflammatory activity in in vivo model. Aiming to reduce the environmental impact caused by the purification EvCI in high amounts and to facilitate the process of obtaining this protein, the recombinant chymotrypsin inhibitor from Eryhrina velutina was produced after cloning and expression in Escherichia coli. The bacteria were grown in LB medium and after induction of the expression this material was subjected to procedures for cell lysis and the product was applied on Nickel-affinity column. The proteins adsorbed were digested by thrombin and applied on Chymotrypsin-Sepharose affinity column, obtaining the purified inhibitor, named recEvCI. After electrophoresis, the recombinant inhibitor showed an approximately molecular mass of 17 kDa, and reduced the chymotrypsin and elastase activities in vitro. The recombinant inhibitor was sequenced and was found similar amino acids residues when compared to other inhibitors deposited in the database, with some modifications. recEvCI showed high stability under pH variations and reducing conditions, maintaining its activity around 80%. This protein increased the blood coagulation time in vitro by acting on the intrinsic pathway and did not show cytotoxicity against strains of mouse 3T3 fibroblasts and RAW 264.7 macrophages. recEvCI showed microbicide activity related to release of nitric oxide and consequently the activation of macrophages, futhermore having proinflammatory effects assessed by increased release of TNF-α. These results indicate that recEvCI can be biotechnologically used as a new tool in the control of coagulation-related diseases as well as can be an activating agent of the immune system in immunosuppressed individuals
