Twelve granulomatous lesions in the thoracic and pelvic limbs of horses from pantaneira breed were evaluated. It was performed suggestive diagnostic of cutaneous pythiosis according to the clinic characteristics of the lesions and histopathological exams using hematoxylin and eosin (HE) and Grocott's methenamine silver (GMS) stains. The confirmation of the diagnoses was performed by the labelled streptavidin biotin (LSAB) method (immunohistochemistry). Extensive focal pyogranulomatous dermatitis associated with pseudo hyphae characteristic of Pythium insidiosum (subcutaneous pythiosis) was the histopathological diagnosis for all 12 horses used in this study. It was confirmed in 100% of the cases by the positive result to Pythium insidiosum in the immunohistochemistry exam, characterized by the visualization of branched and septated structures. It was observed 100% of parity between the techniques. We can conclude that clinical characterization of the granulomatous lesions with pythiosis aspects in horses associated with histopathology suggestive of equine pythiosis constitute a reliable method of diagnostic that can be confirmed by immunohistochemistry technique.

A criptosporidiose, é a enfermidade de veiculação hídrica, possui como agravante a dificuldade de prevenção da contaminação ambiental e ausência de medidas terapêuticas eficazes. Com acentuada importância na bovinocultura, ocasiona inflamação e atrofia das vilosidades intestinais resultando em perda da superfície de absorção. Este estudo teve como objetivo realizar a caracterização molecular da infecção por Cryptosporidium spp. em bezerros do Município de Formiga, Minas Gerais. Um total de 300 amostras de fezes de bezerros holandeses, Nelore e sem raça definida saudáveis foram avaliadas pela técnica de coloração contraste negativo de verde malaquita e por meio da reação de Nested-PCR para amplificação de fragmentos de DNA da subunidade 18S do gene do RNA ribossômico. Ocorrência de 5,33% (16/300) pelo verde malaquita e 4,66% (14/300), pela PCR foi observada, sendo que nenhuma correlação foi verificada entre a positividade e as variáveis estudadas. Por meio da caracterização molecular foram identificadas as espécies Cryptosporidium andersoni e Cryptosporidium ryanae. Como conclusão, observou-se baixa ocorrência da infecção e eliminação de oocistos por Cryptosporidium spp, ausência de sinais clínicos nos animais, houve forte concordância entre os resultados obtidos por meio das duas técnicas utilizadas e pela caracterização molecular (Nested-PCR) foram diagnosticadas as espécies C. andersoni e C. ryanae, presentes em faixas etárias não relatadas na literatura. Estas duas espécies de Cryptosporidium supracitadas são descritas pela primeira vez, parasitando bovinos no estado de Minas Gerais.

The gene encoding TCTP (Translationally Controlled Tumour Protein) is present in all eukaryotes and its product is involved in various cellular processes. Although well characterized in mammals, there are only few works available in the literature related to the analysis of this protein in plants. In this present work, the expression of the gene encoding TCTP was analyzed in different organs/tissues of tomato plants (Solanum lycopersicum cv. Santa Clara). A quantification performed by RT-qPCR revealed the presence of TCTP transcript in all tissues/organs analyzed, with the highest expression level found in leaves. With the exception of fruits in intermediate stage of maturation, for which a small increase on the expression was detected, there was minimal variation in the relative expression of TCTP in other organ/tissues. In parallel, the effects of the constitutive expression of TCTP were investigated using transgenic tobacco lines able to overexpress this protein at different levels (T1, T2 and T3). Seedlings of these lines, and of a non-transgenic control line, were grown in MS culture medium for 21 days. At the end of this period, the length of roots and leaves was taken and the seedlings were photographed. According to Tukey's test, the analysis of the mean root length revealed a significant difference between T1 and T3 lines when compared to the control, although the same was not observed for the T2 line. For leaves, according to Kruskal-Wallis test, there was a statistical difference between the averages of leaf growth obtained for the different lines evaluated. According to these results, we can conclude that TCTP shows an ubiquitous expression in tomato plants, with the highest expression detected in leaves, and also that its overexpression promoted a higher root and leaf development in two of three transgenic tobacco lines tested

The human poliomavirus is the etiologic agente of Progressive multifocal leukoencephalopathy (PML), a disease characterized by focal lesions not expansives of the central nervous system that develops in imunocompromissed patients, specially people with aids. The main aim of the study was to evalute the prevalence of the JCV excretion in urine samples of patients with aids, without PML, to compare two JCV DNA detection techniques through of two diferents genomic regions and to evaluate the genotypic characterization of the positive samples. A total of 75 samples were colected in the Instituto de Infectologia Emílio Ribas, in Sao Paulo, Brazil, between may and november, 2009. To detect the JC virus it was made the DNA extraction and then the polimerase chain reaction (PCR). Firstly a fragment of 215 bp was amplified, which corresponds to the codifying gene of the strutural protein of de JC vírus capsid VP1. All the samples were later submitted to another PCR that uses a pair of primers complementaries to the early region of the JCV (T antigen) amplifying a fragment of 173 bp. Followed by the digestion of the amplified product with the restriction enzime BamH1, resulting in two smaller fragments (120 bp and 53 bp). The JC vírus was detected in 53 samples, for both techniques (70,7% for VP1 PCR, and the restriction enzime BamH1), 34/46 were men (73,9%) and 19/29 were women (65,5%). The JCV excretion was higher in individuals that were over 46 years old. Regarding the seven genotypes described in the literature, the ones that were more prevalent among the JC positive patients were 3B and 3A with 10 samples each (21,0%), the 2B with 9 samples (19,0%) and genotype 6, with six samples (13,0%). As in the brown patients as the white ones, the most prevalent genotype was 3B. In the present study it was observed a high prevalence of JCV DNA (70,7%) and the genotype 3 (43,0%)... Costões rochosos são ecossistemas que propiciam o desenvolvimento de comunidades com grande riqueza de espécies e são ambientes de muita importância para a manutenção dos ecossistemas costeiros. Tais ambientes estão extremamente sujeitos a impactos antrópicos, principalmente no que se refere às contaminações por petróleo. O município da Ilhabela concentra o maior número de costões do Estado de São Paulo, e apresenta um notável histórico desses derrames. A fim de contribuir com o monitoramento dos costões da área do Canal de São Sebastião executado pela CETESB, o presente trabalho tem como objetivo central o estudo da estrutura da comunidade do costão rochoso da Praia da Feiticeira, um dos pontos monitorados pela CETESB entre 1993 e 1996. Além da caracterização das variáveis abióticas, a metodologia, com relação as variáveis biológicas, foi dividida em três partes: estudo quantitativo das espécies dominantes da macrofauna séssil encontradas no campo, baseado na porcentagem de cobertura de Chthamalus bisinuatus (Cirripedia), Brachidontes sp. (Bivalvia) e Tetraclita stalactifera (Cirripedia), gerando gráficos sazonais e demográficos. Para aprimorar as comparações entre os dois trabalhos, foi feito o Teste de hipótese para igualdade de proporções, comparando ano a ano as coletas de inverno, que mostrou que as três espécies dominantes obtiveram mudanças de um trabalho para o outro, rejeitando assim a Ho em todas as situações; estudo qualitativo das espécies de macrofauna e macroalgas encontradas dentro da área amostrada. Este levantamento resultou em uma lista de espécies que foi adicionada ao inventário do programa de monitoramento. O índice de similaridade entre os dois trabalhos foi determinado segundo o coeficiente de Sorrensen, que mostrou uma similaridade média entre as comunidades. The present study is about data characterization and evaluation related to private urban users (legal entity) of the Watersheds of the rivers Piracicaba, Capivari, and Jundiai (PCJ), with the PCJ Collection System, sustained by the Department of Water and Power (DAEE), in order to: provide quantitative numbers about this sector, identifying the cities and economic activities corresponding to the largest water consumers, and determine the sector's share in the total charged contribution. The charge for water use is the most recent and the last instrument which has been implemented for the management of water resources, provided by the institution of the State and National Policy of Water Resources in 1991 and 1997, respectively, regarded as an important step towards the preservation and restoration of water resources. According to the data collected in the PCJ Collection System, the urban sector is the private sector that has the highest number of users in these watersheds (52,95% of users), but with less representation in the financial recovery (only 7%), due to its low water demand in their uses compared to the uses of other sectors. The collected data will also serve as both a parameter for comparing the amount of water used by different economic activities and municipalities in the PCJ watersheds and other locations, and as a tool for water resources management

A alta incidência da cárie no Brasil vem despertando muito interesse para estudos que visem combatê-la. Tem-se observado a procura pela utilização de produtos naturais (plantas medicinais e resinas) que buscam atividade contra a principal bactéria causadora da cárie, que é o Streptococcus mutans, entre estas a geoprópolis (Melipona scutellaris). Todavia, apesar de os estudos mostrarem ação destes produtos de origem animal contra este micro-organismo, muitas vezes a baixa substantividade (bioadesão) deste material ao substrato determina uma baixa efetividade. Este estudo promoveu o desenvolvimento de um sistema líquido cristalino objetivando aumento da substantividade da própolis. Foram desenvolvidos sistemas constituídos por dispersão de quitosana, ácido oleico e álcool cetílico propoxilado e etoxilado (Procetyl® AWS), de acordo com o diagrama de fases ternário. Foram selecionadas 3 formulações, as quais foram submetidas a ensaios de microscopia de luz polarizada, análise de textura, bioadesão e reologia. Os resultados mostraram que os sistemas são viáveis para incorporação do extrato de própolis, aumentando sua bioadesão em ensaios in vitro

The state of São Paulo has four main drainages: Paraná river, Paraíba do Sul river, Ribeira do Iguape river and coastal rivers. The Paraíba do Sul river is born in Sao Paulo and drains an important range of land east of the state. Its ichthyofauna has some similarities and many differences from the continental and coastal drainages which highlights the importance of this study. Surveys conducted in the ichthyofauna of this basin, as in other large river basins in Brazil, is still incomplete. Moreover, there is no consensus about the taxonomic status of many species listed in these surveys. Considering the promising use of DNA barcode as a global system for species identification, the present study is aimed to establishing an inventory of the ichthyofauna of the São Paulo portion of the river Paraíba do Sul and simultaneously build a DNA barcode reference sequence library for fish found. Were obtained and analyzed 354 sequences of the gene cytochrome oxidase c subunit I (COI) belonging to 66 species of São Paulo portion of the Paraíba do Sul river. The average K2P distance between individuals within species of this basin was 0.48%, and 9,87% between species within a genus. Five pairs of species (10 species) showed low levels of interspecific genetic divergence (<2%),but all could be correctly identified. This study showed that the fish species analyzed could be identified efficiently through the use of barcode generating data that can provide information for further studies of this fauna, besides contributing to the global initiative to characterize the species of fish in the world of a molecular point of view. Five pairs of species (10 species) showed low levels of interspecific genetic divergence (<2%), but all could be correctly identified.
