Distribución, concentración y biomasa de los principales recursos pelágicos durante el verano de 1996. Crucero BIC SNP-1 9602-04

El Crucero 9602-04 se realizó a bordo del BIC SNP-1, del 07 de febrero al 05 de abril de 1996 comprendiendo toda el área de litoral peruano desde la frontera sur (18°17 'S) hasta la frontera norte (03°36 'S) y hasta las 120 mn, con el propósito de evaluar los recursos pelágicos. Se presentan los estimados de biomasa y distribución de los principales recursos pelágicos: anchoveta (Engraulis ringens), sardina (Sardinops sagax sagax), jurel (Trachurus picturatus murphyi) y caballa (Scomber japonicus peruanus). Los estimados de biomasa fueron los siguientes: anchoveta 6,69 x 106 t (16,17%),sardina 2,41 x 106 t (22,62%), jurel 3,08 x 106 t (31,23%) y caballa 2,91 x 106 t (21,4%)

Mediciones in situ de TS para el krill (Euphasia superba) a 120 Khz. Verano austral 1998. Perú antar IX

Se realizaron mediciones de la Fuerza de Blanco (TS) del krill (Euphausia superba) durante los 29 lances efectuados en el Crucero de Evaluación Hidroacústica utilizando la ecosonda SIMRAD EK 500 a bordo del BIC Humboldt entre los días 12 y 24 de enero de 1998 a lo largo del Estrecho de Bransfield y alrededores de la Isla Elefante. Se derivaron los valores de b20 a partir de le ecuación de TS de FOOTE (1990), de las longitudes promedios de los individuos capturados durante cada uno de los lances y de las tablas de TS generadas por la ecosonda, determinándose una ecuación de TS para el krill en un rango de longitud comprendido entre 2,1 y 5,3 cm de la siguiente forma: TS = 20 log L - 89,26. Se discute el posible sesgo de la ecuación debido, entre otros aspectos a que no se consideraron los estadíos sexuales del krill.

Con el objetivo de evaluar la biomasa y distribución del krill se llevó a cabo un Crucero de Evaluación Hidroacústica a bordo del BIC Humboldt entre los días 12 y 24 de enero de 1998, a largo del Estrecho de Bransfield y alrededores de la Isla Elefante. Previamente se efectuaron calibraciones de la ecosonda SIMRAD EK 500 utilizando blanco estándar. El trayecto utilizado para el muestreo acústico fue sistemático, paralelo con separaciones de 15 y 12 mn (Estrecho Bransfield e Isla Elefante, respectivamente). Se utilizaron frecuencias de 38 y 120 kHz; la frecuencia de 120 kHz se determinó para la detección entre 2 y 150 mn de profundidad y la de 38 kHz entre 150 - 400 m. Para obtener el área de distribución del krill se utilizó un software de interpolación de datos; y para estimados de biomasa, la metodología de estratificación por cuadrantes de 0,5 - de latitud 1,0 - de longitud. Los resultados obtenidos indican que el krill se encontró en gran parte del área evaluada, con las mayores concentraciones cerca de la Isla D 'Urville, al este de la Isla Rey Jorge, sur de la Isla Robert y en áreas cercanas a la Isla Elefante, distribuidas principalmente entre 50 y 100 m de profundidad.

Effect of early antiretroviral therapy during primary HIV-1 infection on cell-associated HIV-1 DNA and plasma HIV-1 RNA.

Background: Early initiation of combination antiretroviral therapy (ART) during primary HIV-1 infection may prevent the establishment of large viral reservoirs, possibly resulting in improved control of plasma viraemia rebound after ART cessation.Methods: Levels of cell-associated HIV-1 DNA and plasma HIV-1 RNA were measured longitudinally in 32 acutely and recently infected patients, who started ART <= 120 days after the estimated date of infection, and interrupted ART after 18 months (median) of continuous therapy. Averages of HIV-1 DNA and RNA concentrations present in blood 30-365 days after therapy interruption (median duration 300 days, range 195-358) were compared between patients who started ART <= 60 days after the estimated date of infection (early starters), those who started between 61 and 120 days (later starters), and, for HIV-1 RNA only, with 89 untreated participants of the Swiss HIV Cohort Study with documented sero-conversion and longitudinal measurements collected 90-455 days after the first positive HIV test.Results: In early ART starters, average levels of plasma HIV-1 RNA and cell-associated HIV-1 DNA after treatment interruption were 1 log(10) (P=0.008) and 0.4 log(10) (P=0.03) lower compared with later starters. Average post-treatment plasma HIV-1 RNA levels in early starters were significantly lower, respectively, compared with untreated controls (-1.2 log(10); P<0.0004).Conclusions: Early treatment initiation within 2 months after HIV infection compared with later therapy initiation resulted in reduced levels of plasma viraemia and proviral HIV-1 DNA for >= 1 year after subsequent ART cessation. Plasma HIV-1 RNA levels in early starters were also significantly lower than in untreated controls.

Fuel metabolism during exercise in euglycaemia and hyperglycaemia in patients with type 1 diabetes mellitus--a prospective single-blinded randomised crossover trial.

Aims/hypothesis We assessed systemic and local muscle fuel metabolism during aerobic exercise in patients with type I diabetes at euglycaemia and hyperglycaemia with identical insulin levels.Methods This was a single-blinded randomised crossover study at a university diabetes unit in Switzerland. We studied seven physically active men with type I diabetes (mean +/- SEM age 33.5 +/- 2.4 years, diabetes duration 20.1 +/- 3.6 years, HbA(1c) 6.7 +/- 0.2% and peak oxygen uptake [VO2peak] 50.3 +/- 4.5 ml min(-1) kg(-1)). Men were studied twice while cycling for 120 min at 55 to 60% of VO2peak, with a blood glucose level randomly set either at 5 or 11 mmol/l and identical insulinaemia. The participants were blinded to the glycaemic level; allocation concealment was by opaque, sealed envelopes. Magnetic resonance spectroscopy was used to quantify intramyocellular glycogen and lipids before and after exercise. Indirect calorimetry and measurement of stable isotopes and counter-regulatory hormones complemented the assessment of local and systemic fuel metabolism.Results The contribution of lipid oxidation to overall energy metabolism was higher in euglycaemia than in hyperglycaemia (49.4 +/- 4.8 vs 30.6 +/- 4.2%; p<0.05). Carbohydrate oxidation accounted for 48.2 +/- 4.7 and 66.6 +/- 4.2% of total energy expenditure in euglycaemia and hyperglycaemia, respectively (p<0.05). The level of intramyocellular glycogen before exercise was higher in hyperglycaemia than in euglycaemia (3.4 +/- 0.3 vs 2.7 +/- 0.2 arbitrary units [AU]; p<0.05). Absolute glycogen consumption tended to be higher in hyperglycaemia than in euglycaemia (1.3 +/- 0.3 vs 0.9 +/- 0.1 AU). Cortisol and growth hormone increased more strongly in euglycaemia than in hyperglycaemia (levels at the end of exercise 634 52 vs 501 +/- 32 nmol/l and 15.5 +/- 4.5 vs 7.4 +/- 2.0 ng/ml, respectively; p<0.05).Conclusions/interpretation Substrate oxidation in type I diabetic patients performing aerobic exercise in euglycaemia is similar to that in healthy individuals revealing a shift towards lipid oxidation during exercise. In hyperglycaemia fuel metabolism in these patients is dominated by carbohydrate oxidation. Intramyocellular glycogen was not spared in hyperglycaemia.

Mutated regions of nucleophosmin 1 elicit both CD4+ and CD8+ T-cell responses in patients with acute myeloid leukemia.

Mutations in the nucleophosmin gene (NPM1(mut)) are one of the most frequent molecular alterations in acute myeloid leukemia (AML), and immune responses may contribute to the favorable prognosis of AML patients with NPM1(mut). In the present study, we were able to demonstrate both CD4(+) and CD8(+) T-cell responses against NPM1(mut). Ten peptides derived from wild-type NPM1 and NPM1(mut) were subjected to ELISPOT analysis in 33 healthy volunteers and 27 AML patients. Tetramer assays against the most interesting epitopes were performed and Cr(51)-release assays were used to show the cytotoxicity of peptide-specific T cells. Moreover, HLA-DR-binding epitopes were used to test the role of CD4(+) T cells in NPM1 immunogenicity. Two epitopes (epitopes #1 and #3) derived from NPM1(mut) induced CD8(+) T-cell responses. A total of 33% of the NPM1(mut) AML patients showed immune responses against epitope #1 and 44% against epitope #3. Specific lysis of leukemic blasts was detected. To obtain robust immune responses against tumor cells, the activation of CD4(+) T cells is crucial. Therefore, overlapping (OL) peptides were analyzed in ELISPOT assays and OL8 was able to activate both CD8(+) and CD4(+) T cells. The results of the present study show that NPM1(mut) induces specific T-cell responses of CD4(+) and CD8(+) T cells and therefore is a promising target for specific immunotherapies in AML.

Modificações nas formas de fósforo do solo após extrações sucessivas com Mehlich-1, Mehlich-3 e resina trocadora de ânions

O fósforo encontra-se no solo em diversas formas, que variam de acordo com a natureza química dos compostos a que está ligado e à energia de ligação com estes. Assim, a labilidade das formas de P do solo é variável e os métodos de rotina utilizados para avaliação da disponibilidade para as plantas devem ser hábeis em dessorver as formas que têm capacidade de sustentar a absorção das plantas. O objetivo do presente trabalho foi estudar o modo de ação de extratores por meio do acompanhamento das modificações ocorridas nas formas de P do solo após três e treze extrações sucessivas com os métodos Mehlich-1, Mehlich-3 e resina trocadora de ânions (RTA). Foram utilizadas amostras de um Latossolo Vermelho distroférrico típico cultivado sob sistema plantio direto e que recebeu, nos últimos seis anos, doses anuais de 0, 30, 60, 90 e 120 kg ha-1 P2O5, totalizando 0, 180, 360, 540 e 720 kg ha-1 P2O5. Após as extrações sucessivas com os métodos, o solo remanescente foi seco em estufa e realizado o fracionamento químico do P, segundo o fracionamento de Hedley. Os resultados obtidos mostraram que os métodos Mehlich-1 e resina trocadora de ânions atuavam principalmente sobre as frações inorgânicas, sendo parte do P dessorvido por esses extratores readsorvido aos colóides do solo, enquanto o método Mehlich-3 provocava a dessorção de P tanto de formas inorgânicas como de orgânicas. Os métodos Mehlich-1, Mehlich-3 e RTA dessorveram o P de acordo com a labilidade no solo, extraindo, primeiramente, as formas mais lábeis e, posteriormente, as de menor labilidade.

Expression of E-Selectin and Vascular Cell Adhesion Molecule-1 in So-Called 'Negative' Appendices: First Results to Support the Pathological Diagnosis in Borderline Cases.

Background: Since the rate of histologically 'negative' appendices still ranges between 15 and 20%, appendicitis in 'borderline' cases remains a challenging disease. As previously described, cell adhesion molecule expression correlates with different stages of appendicitis. Therefore, it was of interest to determine whether the 'negative' appendix correlated with the absence of E-selectin or vascular cell adhesion molecule-1 (VCAM-1). Methods: Nineteen grossly normal appendices from a series of 120 appendectomy specimens from patients with suspected appendicitis were analysed in frozen sections for the expression of E-selectin and VCAM-1. As control, 5 normal appendices were stained. Results: This study showed a coexpression of E-selectin and VCAM-1 in endothelial cells in early and recurrent appendicitis. In patients with symptoms for less than 6 h, only E-selectin was detected. Cases with fibrosis and luminal obliteration were only positive for VCAM-1. In cases of early appendicitis with symptoms of less than 6 h duration, a discordance between histological and immunohistochemical results was found. Conclusions: This report indicates that E-selectin and VCAM-1 expression could be useful parameters in the diagnosis of appendicitis in borderline cases.

Phosphorus extracted by ion exchange resins and mehlich-1 from oxisols (latosols) treated with different phosphorus rates and sources for varied soil-source contact periods

Despite the large number of studies addressing the quantification of phosphorus (P) availability by different extraction methods, many questions remain unanswered. The aim of this paper was to compare the effectiveness of the extractors Mehlich-1, Anionic Resin (AR) and Mixed Resin (MR), to determine the availability of P under different experimental conditions. The laboratory study was arranged in randomized blocks in a [(3 x 3 x 2) + 3] x 4 factorial design, with four replications, testing the response of three soils with different texture: a very clayey Red Latosol (LV), a sandy clay loam Red Yellow Latosol (LVA), and a sandy loam Yellow Latosol (LA), to three sources (triple superphosphate, reactive phosphate rock from Gafsa-Tunisia; and natural phosphate from Araxá-Minas Gerais) at two P rates (75 and 150 mg dm-3), plus three control treatments (each soil without P application) after four contact periods (15, 30, 60, and 120 days) of the P sources with soil. The soil acidity of LV and LVA was adjusted by raising base saturation to 60 % with the application of CaCO3 and MgCO3 at a 4:1 molar ratio (LA required no correction). These samples were maintained at field moisture capacity for 30 days. After the contact periods, the samples were collected to quantify the available P concentrations by the three extractants. In general, all three indicated that the available P-content in soils was reduced after longer contact periods with the P sources. Of the three sources, this reduction was most pronounced for triple superphosphate, intermediate for reactive phosphate, while Araxá phosphate was least sensitive to the effect of time. It was observed that AR extracted lower P levels from all three soils when the sources were phosphate rocks, while MR extracted values close to Mehlich-1 in LV (clay) and LVA (medium texture) for reactive phosphate. For Araxá phosphate, much higher P values were determined by Mehlich-1 than by the resins, because of the acidity of the extractor. For triple superphosphate, both resins extracted higher P levels than Mehlich-1, due to the consumption of this extractor, particularly when used for LV and LVA.

QUANTIDADE DE FÓSFORO EXTRAÍDO PELAS SOLUÇÕES DE MEHLICH-1 E MEHLICH-3 EM RAZÃO DE DIFERENTES VELOCIDADES DE AGITAÇÃO, TEMPOS DE CONTATO E TEMPERATURAS

A extração de fósforo do solo pode ser comprometida pelas condições em que a análise é realizada e isso pode explicar a variabilidade nos resultados encontrados. O objetivo deste trabalho foi verificar a influência de desvios na velocidade de agitação, no tempo de contato e na temperatura sobre a extração do P pelos extratores Mehlich-1 (M-1) e Mehlich-3 (M-3) em amostras de solo. O estudo foi conduzido por meio de três experimentos executados com 11 amostras de solo provenientes da camada arável de Latossolo, Neossolo, Argissolo, Cambissolo, Planossolo, que são representativas das regiões geomorfológicas do Planalto e da Depressão Central do Estado do Rio Grande do Sul. Para todos os experimentos, adotou-se o delineamento experimental inteiramente casualizado em esquema fatorial 2 × 5 × 11, sendo dois extratores, cinco velocidades, temperaturas ou tempos de contato solo/solução e 11 tipos de solo, com quatro repetições. No primeiro experimento, a mistura solo:solução foi agitada em 80, 100, 120, 140 e 160 oscilações por min (opm). No segundo, após a agitação por 5 min, a mistura solo:solução foi deixada em repouso por períodos de tempo de oito, 12, 16, 20 e 24 h para retirada dos extratos. No terceiro, a temperatura de execução de todo o protocolo de análises foi mantida constante em 15, 20, 25, 30 e 35 °C. O aumento na velocidade de agitação de 120 até 160 opm aumentou a quantidade de P extraído pelos extratores. O aumento no tempo de contato antes da extração da alíquota elevou as quantidades de P extraído pelo M-1 e diminuiu para o M-3. Variações na temperatura não influenciaram significativamente o extrator M-1, mas o M-3 foi mais sensível a variações. Uma padronização na velocidade de agitação, no tempo de repouso e uma climatização dos laboratórios tornam-se necessárias para diminuir a interferência causada sobre as quantidades de P extraído pelos métodos M-1 e M-3 em amostras de solo.

Interleukins (IL)-1 and IL-2 control IL-2 receptor alpha and beta expression in immature thymocytes.

Functional high-affinity interleukin-2 receptors (IL-2R) contain three transmembrane proteins, IL-2R alpha, beta and gamma. We have investigated the expression of IL-2R alpha and beta genes in immature mouse thymocytes. Previous work has shown that during differentiation these cells transiently express IL-2R alpha on their surface. Stimulation of IL-2R alpha+ and IL-2R alpha- immature thymocytes with phorbol 12-myristate 13-acetate and calcium ionophore induces synthesis of IL-2R alpha and IL-2R beta mRNA. Most of this response depends on autocrine stimulation by IL-2. IL-1 synergizes with IL-2 to induce a 120-fold increase in IL-2R alpha mRNA and a 14-fold increase in IL-2R beta mRNA levels. A large proportion of the stimulated cells contains both transcripts. These interleukins do not induce any differentiation to more mature phenotypes. Collectively, these results show that IL-2 plays a major role in the regulation of IL-2R expression in normal immature thymocyte. We suggest that this response to interleukins may be part of a homeostatic mechanism to increase the production of immature thymocytes during stress.
