This thesis entitled Physicochemical and molecular characterization of bacteriophages ΦSP-1and ΦSP-3, specific for pathogenic Salmonella and evaluation of their potential as biocontrol agent . Salmonella were screened using standard methodologies from various environmental samples including chicken caecum. Salmonella strains, which were previously isolated and stocked in the lab, were also included in this study as host, for screening Salmonella specific lytic phages. The Salmonella strain in this study designated as S49 which helped in phage propagation by acting as host bacteria was identified as Salmonella enterica subsp. enterica by 16S rRNA gene analysis and serotyping . A total of three Salmonella specific phage named as ΦSP-1, ΦSP-2 and ΦSP-3 were isolated from chicken intestine samples via an enrichment protocol employing the double agar overlay method. ΦSP-1 and ΦSP-3 showing consistent lytic nature were selected for further study and were purified by repeated plating after picking of single isolated plaques from the lawns of Salmonella S49 plates. Both the phages produced small, clear plaques indicating their lytic nature. ΦSP-1 and ΦSP-3 were concentrated employing PEG-NaCl precipitation method before further characterization. The focus of present study was to isolate, characterize and verify the efficacy of lytic bacteriophages against the robust pathogen Salmonella, capable of surviving under various hostile conditions. Two phages, ΦSP-1 and ΦSP-3, belonging to two families, Podovoridae and Siphoviridae were isolated.

Organizar, adecuar, estructurar y desarrollar los contenidos del Diseño Curricular de Escuelas de Música especializada, en interrelación con el Diseño Curricular de Música en la educación infantil y primaria. Definir los objetivos didácticos de cada nivel y diseñar actividades de enseñanza-aprendizaje. Desarrollar y potenciar capacidades de comprensión y expresión del hecho sonoro. Desarrollar criterios de evaluación. El trabajo de campo se realizó en cada uno de los ciclos de educación infantil y primaria en los ámbitos de conocimiento : expresión rítmica, melódica, armónica (instrumentos) y percepción o escucha activa con un total de 254 alumnos. Mediante la creación de plantillas de análisis de actitudes, procedimientos y conceptos que permitiesen adecuar y potenciar las distintas posibilidades de acoplamiento de los niños con el lenguaje musical, se observaron y analizaron las actitudes diferenciadas -valoración del propio cuerpo, sensibilidad y respeto por las manifestaciones musicales, curiosidad e interés por descubrir y utilizar las posibilidades de los materiales sonoros, disfrute con las expresiones rítmicas, vocales e instrumentales- que surgen de la relación de las actividades y conceptos musicales y por otra se intentó desvelar cuándo y cómo dichas actitudes se convierten en una comunicación estético-musical-social que desarrollan las aptitudes del niño. El niño/a desde una perspectiva cognitiva y metacognitiva, mantiene y desarrolla una serie de acoplamientos con el entorno y la música que parecen producir un diálogo entre los aspectos sociológicos del conocimiento y los aspectos cognitivos de la vida en una constante búsqueda de equilibrio. El modelo de análisis creado y los resultados obtenidos en función de la adecuación de contenidos propuestos en el diseño curricular y en las unidades didáctivas validadas, en las que se han buscado las dimensiones pertinentes para analizar las relaciones intermusicales en la percepción auditiva, vivenciación, interiorización y reproducción rítmica, melódica, instrumental, vocal y corporal constatan que la evolución no es lineal y el proceso es autoconstructivo.

Antología de textos para lectura compartida. Contiene 50 actividades fotocopiables de ficción, no ficción y poesía. Cada texto tiene una mini versión anotada que muestra cómo puede trabajarse en la lección. Las notas para el profesor muestran cómo pueden utilizarse para enseñar múltiples objetivos y se dan ideas para la lecto-escritura. Incluyen historias con configuración familiar, relatos significativos de cuentos infantiles, historias y poemas de otras culturas, poemas para jugar con el lenguaje, informes no cronológicos, instrucciones, extractos de diccionarios.

O seguinte trabalho de investigação foi realizado no âmbito da dissertação de Mestrado em Ciências da Educação: Educação Especial - Domínio Cognitivo-Motor. Este projeto de investigação intitula-se «Sexualidade nas NEE – Trissomia 21: Perspetivas dos docentes do Ensino Regular do 1º, 2º e 3º Ciclo» e tem como objetivo o estudo das perspetivas dos professores perante manifestações sexuais de alunos portadores de Trissomia 21 e divide-se em duas partes fundamentais: o enquadramento teórico e o enquadramento prático. No que diz respeito ao enquadramento teórico, primeiramente procedeu-se à definição da patologia; resumo da sua história; etiologia, ou seja, causas; tipos de trissomia; características, físicas e cognitivas; diagnóstico e intervenção. Seguidamente aprofundaram-se questões ligadas à parte da sexualidade: ideias gerais sobre a sexualidade dos deficientes, o desenvolvimento sexo-afetivo na trissomia 21, contraceção e deficiência mental e princípios e prática da Educação Sexual. Quanto ao enquadramento prático, o estudo do problema levantado, assim como das suas variáveis e hipóteses, foi feito através de um inquérito por questionário, respondido por 36 professores do Ensino Regular do 1º, 2º e 3º ciclo. O questionário em questão continha 15 perguntas de resposta fechada e duas de resposta aberta, onde os professores inquiridos teriam de responder a questões importantes relacionadas com a interação professor – aluno com Trissomia 21, nomeadamente quanto às suas perspetivas face a manifestações sexuais de alunos portadores desta patologia.

The reaction of 1,2-diacyloxycyclohex-3-enes with iodinating agents in the presence of water has been investigated. The process is inherently diastereoselective, with many reactions giving only two of the four possible diastereoisomers which could be obtained. However, the regiocontrol is variable: highest selectivities are observed when pivalates are present on the periphery of the cycloalkene, when single regio- and diastereoisomers are obtained from the reactions. The transforming growth factorβ(TGFβ) superfamily plays an important role in the myocardial response to hypertrophy. We have investigated the protein expression of TGFβ1,β2andβ3in left ventricular tissue, and determined their subcellular distribution in myocytes by immunoblotting and immunocytochemistry during the development of left ventricular hypertrophy (LVH), using isoform specific antibodies to TGFβ1,β2andβ3. LVH was produced in rats by aortic constriction (AC) and LV tissue was obtained at days (d)0, 1, 3, 7, 14, 21 and 42 following operation. Compared with age matched sham-operated controls (SH), TGFβ1levels in LV tissue of AC rats increased significantly from d1–d14 (P<0.03) concomitant with the adaptive growth of LV tissue. In contrast, TGFβ3levels decreased in LV tissue of AC rats from d3 post-operation (significant from d14–d42,P<0.03). No significant difference in TGFβ2levels were observed from SH and AC rats after operation. Antibodies to TGFβ1stained intercalated disks, sarcolemmal membranes and cytoplasm, but not nuclei, of cardiomyocytes on LV sections from untreated and SH rats. However, a trans-localisation of TGFβ1to the nuclei of cardiomyocytes was observed in AC hearts. Antibodies to TGFβ3stained T tubules, cytoplasm and the nuclei of cardiomyocytes from untreated and SH rats. However, by d7 post-AC operation, TGFβ3expression was lost rapidly from nuclei of cardiomyocytes followed by a reduction in total TGFβ3immunofluorescence in myocytes. Antibodies to TGFβ2stained sarcolemmal membranes of cardiomyocytes from both SH and AC rats without significant difference between groups. Thus, the differential pattern of protein expression and subcellular distribution of TGFβ1,β2andβ3in myocytes during the development of LVH suggests that these molecules play different roles in the response of cardiomyocytes to LVH.

PSNCBAM-1 has recently been described as a cannabinoid CB1 receptor allosteric antagonist associated with hypophagic effects in vivo; however, PSNCBAM-1 effects on CB1 ligand-mediated modulation of neuronal excitability remain unknown. Here, we investigate PSNCBAM-1 actions on CB1 receptor-stimulated [35S]GTPγS binding in cerebellar membranes and on CB1 ligand modulation of presynaptic CB1 receptors at inhibitory interneurone-Purkinje cell (IN-PC) synapses in the cerebellum using whole-cell electrophysiology. PSNCBAM-1 caused non-competitive antagonism in [35S]GTPγS binding studies, with higher potency against the CB receptor agonist CP55940 than for WIN55,212-2 (WIN55). In electrophysiological studies, WIN55 and CP55940 reduced miniature inhibitory postsynaptic currents (mIPSCs) frequency, but not amplitude. PSNCBAM-1 application alone had no effect on mIPSCs; however, PSNCBAM-1 pre-treatment revealed agonist-dependent functional antagonism, abolishing CP55940-induced reductions in mIPSC frequency, but having no clear effect on WIN55 actions. The CB1 antagonist/inverse agonist AM251 increased mIPSC frequency beyond control, this effect was reversed by PSNCBAM-1. PSNCBAM-1 pre-treatment also attenuated AM251 effects. Thus, PSNCBAM-1 reduced CB1 receptor ligand functional efficacy in the cerebellum. The differential effect of PSNCBAM-1 on CP55940 versus WIN55 actions in [35S]GTPγS binding and electrophysiological studies and the attenuation of AM251 effects are consistent with the ligand-dependency associated with allosteric modulation. These data provide the first description of functional PSNCBAM-1 allosteric antagonist effects on neuronal excitability in the mammalian CNS. PSNCBAM-1 allosteric antagonism may provide viable therapeutic alternatives to orthosteric CB1 antagonists/inverse agonists in the treatment of CNS disease.

The copper(II) complex [Cu(bdoa)(H2O)2] (bdoaH2 = benzene-1,2-dioxyacetic acid) reacts with triphenylphosphine (1:4 mol ratio) to give the colourless copper(I) complex [Cu(η1-bdoaH)(PPh3)3] (1) in good yield. The X-ray crystal structure of the complex shows the copper atom at the centre of a distorted tetrahedron, and is ligated by the phosphorus atoms of the three triphenylphosphines and one carboxylate oxygen atom of the bdoaH− ligand. Significant intermolecular hydrogen-bonding exists between the pendant carboxylate OH function of one molecule and the uncoordinated “ketonic” oxygen of a neighbouring molecule. Complex 1 is non-conducting in chloroform but ionizes readily in acetonitrile. The cyclic voltammogram of an acetonitrile solution of 1 shows a single irreversible anodic peak for the oxidation of the PPh3 ligands and the copper(I) centre, and a single irreversible cathodic peak for the reduction of the bdoaH− ion. IR and mass spectral data for 1 are given.

Protein sequences from characterized type III secretion (TTS) systems were used as probes in silico to identify several TTS gene homologs in the genome sequence of Brucella suis biovar 1 strain 1330. Four of the genes, named flhB, fliP, fliR, and fliF on the basis of greatest homologies to known flagellar apparatus proteins, were targeted in PCR and hybridization assays to determine their distribution among other Brucella nomen species and biovars. The results indicated that flhB, fliP, fliR and fliF are present in Brucella melitensis, Brucella ovis, and Brucella suis biovars 1, 2 and 3. Similar homologos have been reported previously in Brucella abortus. Using RT-PCR assays, we were unable to detect any expression of these genes. It is not yet known whether the genes are the cryptic remnants of a flagellar system or are actively involved in a process contributing to pathogenicity or previously undetected motility, but they are distributed widely in Brucella and merit further study to determine their role.

The physical properties of the La(0.6)Y(0.1)Ca(0.3)MnO(3) compound have been investigated, focusing on the magnetoresistance phenomenon studied by both dc and ac electrical transport measurements. X-ray diffraction and scanning electron microscopy analysis of ceramic samples prepared by the sol-gel method revealed that specimens are single phase and have average grain size of similar to 0.5 mu m. Magnetization and 4-probe dc electrical resistivity rho(T,H) experiments showed that a ferromagnetic transition at T(C) similar to 170 K is closely related to a metal-insulator (MI) transition occurring at essentially the same temperature T(MI). The magnetoresistance effect was found to be more pronounced at low applied fields (H <= 2.5 T) and temperatures close to the MI transition. The ac electrical transport was investigated by impedance spectroscopy Z(f,T,H) under applied magnetic field H up to 1 T. The Z(f,T,H) data exhibited two well-defined relaxation processes that exhibit different behaviors depending on the temperature and applied magnetic field. Pronounced effects were observed close to T (C) and were associated with the coexistence of clusters with different electronic and magnetic properties. In addition, the appreciable decrease of the electrical permittivity epsilon`(T,H) is consistent with changes in the concentration of e(g) mobile holes, a feature much more pronounced close to T (C).

The synthesis and characterization of lanthanide(III) citrates with stoichiometries 1:1 and 2:3; [LnL center dot xH(2)O] and [Ln(2)(LH)(3)center dot 2H(2)O], Ln=La, Ce, Pr, Nd, Sm and Eu are reported. L stands for (C6O7H5)(3-) and LH for (C6O7H6)(2-). Infrared absorption spectra of both series evidence coordination of carboxylate groups through symmetric bridges or chelation. X-ray powder patterns show the amorphous character of [LnL center dot xH(2)O]. The compounds [Ln(2)LH(3)center dot 2H(2)O] are crystalline and isomorphous. Emission spectra of Eu compounds suggest C-2v symmetry for the coordination polyhedron of [LnL center dot xH(2)O] and C-4v for [Ln(2)(LH)(3)center dot 2H(2)O]. Thermal analyses (TG-DTG-DTA) were carried out for both series. The thermal analysis patterns of the two series are quite different and both fit in a 4-step model of thermal decomposition, with lanthanide oxides as final products.
