O método construtivo com painéis portantes de concreto é economicamente viável, porém relativamente novo no cenário nacional, sobretudo no caso dos pré-moldados. As incertezas referentes às peculiaridades desse método, bem como a nova norma brasileira de painéis pré-moldados, ainda em elaboração, vem a motivar uma análise probabilística dos critérios de projeto disponíveis. Utilizando-se a técnica da confiabilidade estrutural, é possível propagar as incertezas referentes às variáveis a uma resposta final no índice de confiabilidade, sendo um cálculo totalmente probabilístico. Neste trabalho, emprega-se tal técnica com informações estatísticas referentes a lajes de concreto moldadas in loco para verificar, de maneira mais verossímil, a segurança dos critérios de projeto impostos pelo Precast Concrete Institute Design Handbook - Precast and Prestressed Concrete - 7th Edition (2010) às fases transitórias (desforma, transporte e içamento) e pela Norma Brasileira ABNT NBR 6118: 2014 - Projeto de estruturas de concreto, à fase em uso. Prossegue-se a uma análise crítica dos resultados bem como sugestões para diminuir a variação dos resultados, sobretudo pela calibração de novos coeficientes parciais de segurança, processo para o qual este trabalho pode servir de base.

O emprego da flotação por ar dissolvido (FAD) para o pós-tratamento de efluentes de reatores anaeróbios aparenta ser atraente considerando algumas características desse processo físico-químico. A FAD é reconhecidamente um processo de alta taxa, particularmente eficiente na remoção de material particulado em suspensão e de flocos produzidos pela coagulação química de águas residuárias. Além disso, há produção de lodo espesso e provavelmente arraste de parcela de gases e de compostos voláteis, presentes nos efluentes anaeróbios. Entretanto, a concepção de sistemas de FAD deve ser precedida por ensaios em unidades de flotação em escala de laboratório, permitindo a determinação dos principais parâmetros do processo. Neste trabalho, são apresentados e discutidos os resultados obtidos em laboratório e em instalação piloto de flotação com escoamento contínuo recebendo efluente de reator anaeróbio de manta de lodo (UASB), com 18 m3 de volume, tratando esgoto sanitário. Os ensaios em unidade em escala de laboratório foram realizados utilizando diferentes dosagens de cloreto férrico (entre 30 e 110 mg/L) ou de polímero catiônico (entre 1,0 e 16,0 mg/L), atuando como coagulantes. Além disso, foram estudadas as condições de floculação (tempo de 15 e de 25 min, e gradiente médio de velocidade de floculação entre 30 e 100 s-1) e diferentes valores de quantidade de ar fornecido ao processo (S*, entre 4,7 e 28,5 g de ar por m3 de efluente). Com a instalação piloto de FAD foram realizados apenas ensaios preliminares variando-se a taxa de aplicação superficial (140 e 210 m3/m2/d) para diferentes valores de S* (14,8 a 29,5 g de ar por m3 de efluente). Com o emprego de dosagem de 65 mg/L de cloreto férrico, de tempo de 15 min e gradiente médio de velocidade de floculação de 80 s-1 e de 19 g de ar por m3 de efluente, foram observados excelentes resultados em laboratório, com elevadas remoções de DQO (89%), de fosfato total (96%), de sólidos suspensos totais (96%), de turbidez (98%), de cor aparente (91%), de sulfetos (não detectado) e NTK (47%). Considerando o sistema UASB e FAD, nos testes em laboratório, foram observadas remoções globais de 97,7% de DQO, de 98,0% de fosfato total, de 98,9% de SST, de 99,5% de turbidez, de 97,8% de cor aparente e de 59,0% de NTK. Nos ensaios com a instalação piloto de FAD, o sistema apresentou remoções de 93,6% de DQO, de 87,1% de SST, de 90% de sulfetos e de 30% de NTK.

The electroreduction of nitrate on Pt(1 0 0) electrodes in phosphate buffer neutral solution, pH 7.2, is reported. The sensitivity of the reaction to the crystallographic order of the surface is studied through the controlled introduction of defects by using stepped surfaces with (1 0 0) terraces of different length separated by monoatomic steps, either with (1 1 1) or (1 1 0) symmetry. The results of this study show that nitrate reduction occurs mainly on the well defined (1 0 0) terraces in the potential region where H adsorption starts to decrease, allowing the nitrate anion to access the surface. Adsorbed NO has been detected as a stable intermediate in this media. An oxidation process observed at 0.8 V has been identified as leading to the formation of adsorbed NO and being responsible for a secondary reduction process observed in the subsequent negative scan. Using in situ FTIRS, ammonium was found to be the main product of nitrate reduction. This species can be oxidized at high potentials resulting in adsorbed NO and nitrate (probably with nitrite as intermediate).

En Argentina, en consonancia con el resto del mundo, la Nanotecnología es considerada un área estratégica. Sin embargo, las investigaciones en Nanobiotecnología todavía constituyen un área de vacancia. El uso de nanomateriales para desarrollar plataformas bioanalíticas que permitan la construcción de biosensores ofrece múltiples ventajas y una promisoria perspectiva de aplicación en diversas áreas. En la actualidad, los laboratorios de análisis clínicos, la industria farmacéutica y alimentaria, y los laboratorios de control bromatológico y ambiental requieren de metodologías analíticas que proporcionen resultados exactos, reproducibles, rápidos, sensibles y selectivos empleando pequeños volúmenes de muestra, con un mínimo consumo de reactivos y una producción de deshechos limpia y escasa. Las investigaciones en nanobiosensores se encuentran dirigidas hacia el logro de estas metas. Uno de los grandes desafíos es lograr biosensores miniaturizados con potencialidad para el desarrollo de dispositivos de medición descentralizada (“point of care”) y la detección simultánea de multianalitos. Aún cuando se han hecho innumerables desarrollos en los casi 50 años de vida de los biosensores, todavía hay numerosos interrogantes por dilucidar. La modificación con nanomateriales juega un rol preponderante en los transductores tanto en los electroquímicos como en los plasmónicos. El uso de películas delgadas de Au para SPR modificadas con grafeno u óxido de grafeno, es un campo de una enorme potencialidad y sin embargo es muy poco explotado, por lo que reviste gran importancia. En lo referido a la capa de biorreconocimiento, se trabajará con moléculas capaces de establecer interacciones de bioafinidad, como los anticuerpos y también moléculas que son muy poco usadas en nuestro país y en Latinoamérica como ADN, aptámeros, PNA y lectinas. RESUMEN: El Objetivo general de este proyecto es desarrollar nuevas plataformas bioanalíticas para la detección de diferentes eventos de bioafinidad a partir de la integración de transductores electroquímicos (EQ) y plasmónicos con materiales nanoestructurados (nanotubos de carbono, nanoláminas de grafeno, nanoalambres metálicos); biomoléculas (ADN, “peptide nucleic acid” (PNA), aptámeros, anticuerpos, lectinas) y polímeros funcionalizados con moléculas bioactivas. Las arquitecturas supramoleculares resultantes estarán dirigidas al desarrollo de biosensores EQ y plasmónicos para la cuantificación de biomarcadores de relevancia clínica y medioambiental. Se funcionalizarán CNT, grafeno, óxido de grafeno, nanoalambres metálicos empleando homopéptidos y proteínas con alta afinidad por cationes metálicos, los que se integrarán a transductores de carbono y oro y biomoléculas de reconocimiento capaces de formar complejos de afinidad (antígeno-anticuerpo, aptámero-molécula blanco, ADN-ADN, PNA-ADN, lectinas-hidratos de carbono, ligandos-cationes metálicos y avidina-biotina). Se sintetizarán y caracterizarán nuevos monómeros y polímeros funcionalizados con moléculas bioactivas y/o grupos rédox empleando diferentes rutas sintéticas. Se desarrollarán genosensores para la detección del evento de hibridación de secuencias de interés médico (cáncer de colon y de mama, tuberculosis); aptasensores para la detección de marcadores proteicos de T. cruzi, enfermedades cardiovasculares y contaminantes catiónicos; inmunosensores para la detección de biomarcadores proteicos relacionados con enfermedades cardiovasculares y cáncer; y biosensores de afinidad con lectinas para la detección de hidratos de carbono. La caracterización de las plataformas y las señales analíticas se obtendrán empleando las siguientes técnicas: voltamperometrías cíclica, de pulso diferencial y de onda cuadrada; stripping; resonancia de plasmón superficial; espectroscopía de impedancia electroquímica; microscopías de barrido electroquímico, SEM, TEM, AFM,SNOM, espectroscopías: UV-vis, FTIR,Raman;RMN, TGA y DSC.

Il principale componente non idrocarburico contenuto nel gas naturale è l’acqua che viene rimossa mediante assorbimento con glicole trietilenico, il processo di trattamento più comune nella produzione di gas naturale. La presenza di acqua libera nel gas deve essere rimossa totalmente, per evitare la formazione di condensa nelle condizioni di trasporto e di distribuzione più critiche cioè a pressione elevata e a bassa temperatura. Obiettivo di questa tesi è l’analisi delle cause e dei fenomeni che portano a rilevanti perdite di glicole (TEG) in impianti di disidratazione del gas naturale operanti con elevate concentrazioni di CO2 e H2S nel gas di processo. Le perdite, in relazione alle diverse condizioni operative e concentrazioni dei gas menzionati, possono arrivare a raggiungere valori pari a 3-4 volte l'entità attesa, con punte che raggiungono 10 volte tali valori. Il lavoro di tesi è stato focalizzato su un impianto attualmente in esercizio, situato in Arabia Saudita. L’attività è stata condotta presso la Comart di Ravenna, azienda specializzata nella progettazione di impianti nel settore Oil&Gas. Inizialmente sono state studiate le caratteristiche di un impianto di disidratazione di gas naturale mediante assorbimento con glicole trietilenico. Dopo l’analisi delle possibili fonti di perdita, la colonna di alimentazione del ribollitore (Still Column) e il relativo condensatore, sulla base dei dati operativi degli impianti, sembra essere il punto su cui si concentrano le perdite. Vengono presentati i dettagli costruttivi di tale colonna al fine di determinarne i possibili malfunzionamenti. Scopo della tesi sarà l'identificazione, partendo dall'attuale configurazione dell’impianto, delle cause che portano a tali valori di perdite e l'individuazione di una configurazione colonna/condensatore (con eventuali altri equipment necessari) in grado di minimizzare le perdite contenendo al minimo i costi aggiuntivi della soluzione.

Oceanographic changes in the western equatorial Pacific during the past 6 m.y. are inferred from carbon isotopic analyses of planktonic and benthic foraminifers from Ontong Java Plateau (DSDP Site 586). Sample spacing is 1.5 m (ca. 35,000-75,000 yr). An overall trend of d13C toward lighter values is evident for the last 5 m.y. in all four foraminiferal taxa analyzed (G. sacculifer, Pulleniatina, P. wuellerstorfi, and O. umbonatus). This trend is interpreted as an enrichment of the global ocean with 12C, because of the addition of carbon from organic carbon reservoirs (or lack of removal of carbon to such reservoirs), as a consequence of an overall drop in sea level. Differences between shallow- and deep-water d13C decrease slightly during this time interval, suggesting a moderate drop in productivity. This drop is not sufficient to explain the drop in sedimentation rate, however, much of which apparently must be ascribed to winnowing effects. A marked convergence in the d13C values of planktonic taxa exists within the last 2 m.y. We propose that this convergence indicates nutrient depletion in thermocline waters, caused by the vigorous removal of phosphate in marginal upwelling regions, or by the stripping of intermediate waters in their source regions. No large shifts are seen in the carbon isotope record of the last 6 m.y., in contrast to the oxygen isotope record. Some indication of cyclicity is present, with a period between 0.5 and 1.0 m.y. (especially in the earlier portion of the record).

We have discovered a new type of galaxy in the Fornax Cluster: 'ultra-compact' dwarfs (UCDs). The UCDs are unresolved in ground-based imaging and have spectra typical of old stellar systems. Although the UCDs resemble overgrown globular clusters, based on VLT UVES echelle spectroscopy, they appear to be dynamically distinct systems with higher internal velocity dispersions and M/L ratios for a given luminosity than Milky Way or M31 globulars. Our preferred explanation for their origin is that they are the remnant nuclei of dwarf elliptical galaxies which have been tidally stripped, or 'threshed' by repeated encounters with the central cluster galaxy, NGC1399. If correct, then tidal stripping of nucleated dwarfs to form UCDs may, over a Hubble time, be an important source of the plentiful globular cluster population in the halo of NGC1399, and, by implication, other cD galaxies. In this picture, the dwarf elliptical halo contents, up to 99% of the original dwarf luminosity, contribute a significant fraction of the populations of intergalactic stars, globulars, and gas in galaxy clusters.

Stratum corneum (SC) desorption experiments have yielded higher calculated steady-state fluxes than those obtained by epidermal penetration studies. A possible explanation of this result is a variable diffusion or partition coefficient across the SC. We therefore developed the diffusion model for percutaneous penetration and desorption to study the effects of either a variable diffusion coefficient or variable partition coefficient in the SC over the diffusion path length. Steady-state flux, lag time, and mean desorption time were obtained from Laplace domain solutions. Numerical inversion of the Laplace domain solutions was used for simulations of solute concentration-distance and amount penetrated (desorbed)-time profiles. Diffusion and partition coefficients heterogeneity were examined using six different models. The effect of heterogeneity on predicted flux from desorption studies was compared with that obtained in permeation studies. Partition coefficient heterogeneity had a more profound effect on predicted fluxes than diffusion coefficient heterogeneity. Concentration-distance profiles show even larger dependence on heterogeneity, which is consistent with experimental tape-stripping data reported for clobetasol propionate and other solutes. The clobetasol propionate tape-stripping data were most consistent with the partition coefficient decreasing exponentially for half the SC and then becoming a constant for the remaining SC. Skin penetration of the tetrapeptide Ac-Ala-Ala-Pro-Val-NH2 was assessed. This peptide sequence fits the P-P-1 subsites of elastase and inhibits human neutrophil elastase competitively. Consequently this peptide may be therapeutically useful in a variety of inflammatory disorders, including psoriasis. in which elevated levels of human neutrophil elastase have been reported. Peptide penetration was assessed across whole human skin, whole skin with the stratum corneum removed by tape stripping and epidermis, which had been removed from the dermis by heat separation. The influence of 75% aqueous ethanol as a potential penetration enhancer of the tetrapeptide across epidermis was also assessed. The tetrapeptide did not penetrate whole human skin or epidermis, even under the influence of 75% aqueous ethanol. However, when the stratum corneum was removed tetrapeptide flux of 73.39 mug cm(-2) h(-1) was achieved. The study demonstrates that the stratum corneum is the main barrier to tetrapeptide skin penetration and must be overcome if therapeutically relevant amounts of tetrapeptide are to be delivered to the skin.

This thesis describes the study of various grating based optical fibre sensors for applications in refractive index sensing. The sensitivity of these sensors has been studied and in some cases enhanced using novel techniques. The major areas of development are as follows. The sensitivity of long period gratings (LPGs) to surrounding medium refractive index (SRI) for various periods was investigated. The most sensitive period of LPG was found to be around 160 µm and this was due to the core mode coupling to a single cladding mode but phase matching at two wavelength locations, creating two attenuation peaks, close to the waveguide dispersion turning point. Large angle tilted fibre gratings (TFGs) have similar behaviour to LPGs, in that they couple to the co-propagating cladding modes. The tilted structure of the index modulation within the core of the fibre gives rise to a polarisation dependency, differing the large angle TFG from a LPG. Since the large angle TFG couple to the cladding mode they are SRI sensitive, the sensitivity to SRI can be further increased through cladding etching using HF acid. The thinning of the cladding layer caused a reordering of the cladding modes and shifted to more SRI sensitive cladding modes as the investigation discovered. In a SRI range of 1.36 to 1.40 a sensitivity of 506.9 nm/URI was achieved for the etched large angle TFG, which is greater than the dual resonance LPG. UV inscribed LPGs were coated with sol-gel materials with high RIs. The high RI of the coating caused an increase in cladding mode effective index which in turn caused an increase in the LPG sensitivity to SRI. LPGs of various periods of LPG were coated with sol-gel TiO2 and the optimal thickness was found to vary for each period. By coating of the already highly SRI sensitive 160µm period LPG (which is a dual resonance) with a sol-gel TiO2, the SRI sensitivity was further increased with a peak value of 1458 nm/URI, which was an almost 3 fold increase compared to the uncoated LPG. LPGs were also inscribed using a femtosecond laser which produced a highly focused index change which was no uniform throughout the core of the optical fibre. The inscription technique gave rise to a large polarisation sensitivity and the ability to couple to multiple azimuthal cladding mode sets, not seen with uniform UV inscribed gratings. Through coupling of the core mode to multiple sets of cladding modes, attenuation peaks with opposite wavelength shifts for increasing SRI was observed. Through combining this opposite wavelength shifts, a SRI sensitivity was achieved greater than any single observed attenuations peak. The maximum SRI achieved was 1680 nm/URI for a femtosecond inscribed LPG of period 400 µm. Three different types of surface plasmon resonance (SPR) sensors with a multilayer metal top coating were investigated in D shape optical fibre. The sensors could be separated into two types, utilized a pre UV inscribed tilted Bragg grating and the other employed a post UV exposure to generate surface relief grating structure. This surface perturbation aided the out coupling of light from the core but also changed the sensing mechanism from SPR to localised surface plasmon resonance (LSPR). This greatly increased the SRI sensitivity, compared to the SPR sensors; with the gold coated top layer surface relief sensor producing the largest SRI sensitivity of 2111.5nm/URI was achieved. While, the platinum and silver coated top layer surface relief sensors also gave high SRI sensitivities but also the ability to produce resonances in air (not previously seen with the SPR sensors). These properties were employed in two applications. The silver and platinum surface relief devices were used as gas sensors and were shown to be capable of detecting the minute RI change of different gases. The calculated maximum sensitivities produced were 1882.1dB/URI and 1493.5nm/URI for silver and platinum, respectively. Using a DFB laser and power meter a cheap alternative approach was investigated which showed the ability of the sensors to distinguish between different gases and flow rates of those gases. The gold surface relief sensor was coated in a with a bio compound called an aptamer and it was able to detect various concentrations of a biological compound called Thrombin, ranging from 1mM to as low as 10fM. A solution of 2M NaCl was found to give the best stripping results for Thrombin from the aptamer and showed the reusability of the sensor. The association and disassociation constants were calculated to be 1.0638×106Ms-1 and 0.2482s-1, respectively, showing the high affinity of the Aptamer to thrombin. This supports existing working stating that aptamers could be alternative to enzymes for chemical detection and also helps to explain the low detection limit of the gold surface relief sensor.
