Due to the increasing demand from clients and the search for better performances in the heavy vehicles industry, a progressive evolution in technology in a general way was needed. This paper uses a scientific method to validate, prior to its manufacture, the project of an agricultural wheel for sugar cane harvesters. Monte Carlo Simulation is used in conjunction with Finite Elements Method, in order to simulate the wheel's behavior in a cornering test, identify possible failure regions and get an estimate for its life under fatigue. To this end, test conditions according to EUWA Standards were simulated and obeyed, relevant to fatigue. Simulation results were interesting, according to industry experts involved in the project and manufacture of the product in question, and have provided important elements for the decision making regarding improvements that could be made on the product project before its execution

Photoprotection of the agarophyte red alga Gracilaria tenuistipitata against ultraviolet radiation (UVR) was investigated in algae submitted for 1 week to photosynthetically active radiation (PAR, 260 mu mol photons m(-2) s(-1)) or PAR + UVR (UV-A, 8.13 W m(-2) and UV-B, 0.42 W m(-2)) under different nitrogen concentrations: 0, 0.1, and 0.5 mM of NO3-. Photosynthetic pigments decreased during the time of the experiment mainly under low nitrogen supply and UVR. Incubation under high nitrogen supply (0.5 mM) sustained the photosynthetic levels over time. In contrast, mycosporine-like amino acids (MAAs) increased up to eightfold in the presence of UVR and 0.5 mM NO3-. Under PAR + UVR, maximal quantum yield was positively correlated to MAA abundance, whereas under PAR no correlation was found. The photosynthetic yield of algae cultivated during seven days under PAR + UVR was less affected by a 30-min exposure of high UVR (16 W m(-2)) and fully recovered after transferring to low PAR irradiances, whereas algae kept under PAR were more affected by UV exposure and no full recovery was observed. Growth rates decreased after three days in the presence of UVR and under low nitrate supply. However, these rates were similar when compared with treatments of PAR and PAR + UVR after seven days, with the exception of samples in 0 mM NO3-, indicating that the acclimation after one week's exposure is related to nitrate supply. In conclusion, the lowest negative effect of UVR on photosynthesis and growth rate in high N-supply-grown algae could be explained by the stimulation of photoprotection mechanisms, such as accumulation of MAAs. Photostimulation of MAA accumulation by UVR under high N supply was observed in G. tenuistipitata even after 20 years in culture without the induction of this photomorphogenic light signal.

O vigor da semente tem sido avaliado por meio de testes fisiológicos, bioquímicos e de tolerância a estresse. Mais recentemente, com o uso da análise computadorizada de imagens de plântulas, podem ser obtidas informações objetivas, em período relativamente curto, com menor interferência humana. O objetivo nesta pesquisa foi identificar testes eficientes para avaliação do vigor de sementes de alface, com ênfase para a análise de imagens digitais de plântulas. Sementes de 12 lotes (seis da cultivar Vanda e seis da Vera) foram avaliadas por meio de testes de germinação, germinação a baixa temperatura (15ºC), emergência de plântulas e envelhecimento acelerado tradicional e com solução salina (NaCl 40%) saturada, a 41ºC/48 h e 41ºC/72 h. Utilizou-se, também, o sistema de análise computadorizada de imagens de plântulas (SVIS®), desenvolvido pela Ohio State University (EUA), que forneceu dados referentes ao índice de vigor, uniformidade e crescimento de plântulas. Os testes foram realizados a cada três meses, durante 12 meses de armazenamento, sob condições ambiente. Com base nos resultados obtidos, concluiu-se que os testes de envelhecimento acelerado com uso de solução saturada de NaCl, a 41ºC por 48 e 72 horas e de velocidade de germinação, e os parâmetros obtidos com o software SVIS, índice de crescimento de plântulas e comprimento da raiz primária, são eficientes na avaliação do vigor de lotes de sementes de alface.

O planejamento e a evolução terapêutica de crianças com transtorno fonológico estão diretamente relacionados à avaliação inicial e aos testes complementares aplicados. Acompanhar a evolução do caso por meio de verificações regulares acrescenta informações importantes à avaliação diagnóstica, o que permite fortalecer achados iniciais a respeito da dificuldade subjacente identificada na avaliação inicial. Assim, no presente estudo de caso verificou-se a efetividade e a eficiência da aplicação do índice de porcentagem de consoantes corretas revisado (PCC-R) bem como dos testes complementares de inconsistência de fala, de estimulabilidade e de habilidades metafonológicas no acompanhamento da intervenção terapêutica em crianças com transtorno fonológico. Participaram deste estudo três crianças do gênero masculino. Na data da avaliação inicial o Caso 1 tinha 6 anos e 9 meses de idade, o Caso 2, 8 anos e 10 meses, e o Caso 3, 9 anos e 7 meses. Além da avaliação específica da fonologia, foram aplicados testes complementares que auxiliaram na verificação da dificuldade subjacente específica em cada um dos casos. Desta forma, os sujeitos foram submetidos à avaliação de habilidades metafonológicas, à prova de inconsistência de fala e de estimulabilidade. A análise conjunta dos dados permitiu constatar que os testes selecionados foram efetivos e eficientes tanto para complementar o diagnóstico como para indicar mudanças nos três casos de crianças com transtorno fonológico.

Animal coloration often serves as a signal to others that may communicate traits about the individual such as toxicity, status, or quality. Colorful ornaments in many animals are often honest signals of quality assessed by mates, and different colors may beproduced by different biochemical pigments. Investigations of the mechanisms responsible for variation in color expression among birds are best when including a geographically and temporally broad sample. In order to obtain such a sample, studies such as this often use museum specimens; however, in order for museum specimens toserve as an accurate replacement, they must accurately represent living birds, or we must understand the ways in which they differ. In this thesis, I investigated the link between feather corticosterone, a hormone secreted in response to stress, and carotenoid-basedcoloration in the Red-winged Blackbird (Agelaius phoeniceus) in order to explore a mechanistic link between physiological state and color expression. Male Red-winged Blackbirds with lower feather corticosterone had significantly brighter red epaulets than birds with higher feather corticosterone, while I found no significant changes in red chroma. I also performed a methodological comparison of color change in museum specimens among different pigment types (carotenoid and psittacofulvin) and pigments in different locations in the body (feather and bill carotenoids) in order to quantify colorchange over time. Carotenoids and psittacofulvins showed significant reductions in red brightness and chroma over time in the collection, and carotenoid color changed significantly faster than psittacofulvin color. Both bill and feather carotenoids showed significant reductions in red brightness and red chroma over time, but change of both red chroma and red brightness occurred at a similar rate in feathers and bills. In order to use museum specimens of ecological research on bird coloration specimen age must be accounted for before the data can be used; however, once this is accomplished, museum- based color data may be used to draw conclusions about wild populations.

The gene for agouti signaling protein (ASIP) is centrally involved in the expression of coat color traits in animals. The Mangalitza pig breed is characterized by a black-and-tan phenotype with black dorsal pigmentation and yellow or white ventral pigmentation. We investigated a Mangalitza x Piétrain cross and observed a coat color segregation pattern in the F2 generation that can be explained by virtue of two alleles at the MC1R locus and two alleles at the ASIP locus. Complete linkage of the black-and-tan phenotype to microsatellite alleles at the ASIP locus on SSC 17q21 was observed. Corroborated by the knowledge of similar mouse coat color mutants, it seems therefore conceivable that the black-and-tan pigmentation of Mangalitza pigs is caused by an ASIP allele a(t), which is recessive to the wild-type allele A. Toward positional cloning of the a(t) mutation, a 200-kb genomic BAC/PAC contig of this chromosomal region has been constructed and subsequently sequenced. Full-length ASIP cDNAs obtained by RACE differed in their 5' untranslated regions, whereas they shared a common open reading frame. Comparative sequencing of all ASIP exons and ASIP cDNAs between Mangalitza and Piétrain pigs did not reveal any differences associated with the coat color phenotype. Relative qRT-PCR analyses showed different dorsoventral skin expression intensities of the five ASIP transcripts in black-and-tan Mangalitza. The a(t) mutation is therefore probably a regulatory ASIP mutation that alters its dorsoventral expression pattern.

Most published genomewide association studies (GWAS) in sheep have investigated recessively inherited monogenic traits. The objective here was to assess the feasibility of performing GWAS for a dominant trait for which the genetic basis was already known. A total of 42 Manchega and Rasa Aragonesa sheep that segregate solid black or white coat pigmentation were genotyped using the SNP50 BeadChip. Previous analysis in Manchegas demonstrated a complete association between the pigmentation trait and alleles of the MC1R gene, setting an a priori expectation for GWAS. Multiple methods were used to identify and quantify the strength of population substructure between black and white animals, before allelic association testing was performed for 49 034 SNPs. Following correction for substructure, GWAS identified the most strongly associated SNP (s26449) was also the closest to the MC1R gene. The finding was strongly supported by the permutation tree-based random forest (RF) analysis. Importantly, GWAS identified unlinked SNP with only slightly lower p-values than for s26449. Random forest analysis indicated these were false positives, suggesting interpretation based on both approaches was beneficial. The results indicate that a combined analytical approach can be successful in studies where a modest number of animals are available and substantial population stratification exists.
