Com o passar do tempo, a população mundial vem se conscientizando mais sobre problemas ambientais. Isso fez surgir uma demanda por tecnologias novas que possam se encaixar no cenário de sustentabilidade. Instabilidades frequentes no cenário político-econômico global acabam por elevar o preço do barril do petróleo. Assim a indústria química começa a buscar por alternativas que tenham a mesma versatilidade do petróleo. Dentre as opções de combustíveis renováveis, destaca-se o bio-óleo de pirólise. Seu interesse em estudos científicos vem do fato de poder-se utilizar do rejeito de processos como matéria prima, não necessitando competir por espaço com a plantação de alimentos. Sua composição pode ser representada por ácidos e fenóis. Em especial destacamos o ácido acético e fenóis oxigenados como m-cresol, o-cresol, p-cresol e guaiacol por estarem presentes em grandes quantidades. Sua separação das frações menos polares do bio-óleo pode ser realizada por meio de extração com água que é um reagente abundante e de baixo custo. O conhecimento das propriedades desses componentes puros é amplamente disponível na literatura, porém dados de composições das fases corrosivas, como misturas ternárias de água-ácido acético-m-cresol/o-cresol/p-cresol/guaiacol nas temperaturas de (298,15, 310,65 e 323,15) K são escassos. Devido a isso, o uso de modelos termodinâmicos para simulação do comportamento desses sistemas torna-se interessante. Todavia, quando são testadas as capacidades dos modelos clássicos, como o UNIFAC e NRTL, percebe-se que os mesmos não conseguem reproduzir o comportamento da binodal dos componentes corrosivos. Sendo assim, essa dissertação investigou soluções para melhorar a descrição desses sistemas, assim como obteve dados experimentais para tais sistemas de misturas ternárias de água-ácido acético-m-cresol/o-cresol/p-cresol/guaiacol nas temperaturas de (298,15, 310,65 e 323,15) K; desenvolveu-se uma metodologia para estimar parâmetros do modelo NRTL a partir de dados de composição da binodal e verificou-se a possibilidade de utilizar o modelo UNIFAC para prever o comportamento de equilíbrio de fases. Como resultado foram obtidos 314 novos dados experimentais, obtiveram-se parâmetros para o modelo NRTL que conseguem reproduzir com acurácia a forma da binodal com a metodologia proposta e verificou-se a necessidade de aperfeiçoamento no estudo do modelo UNIFAC para otimizar sua utilização na previsão do comportamento dos sistemas estudados

Os peixes são vertebrados que vivem em vários habitats. Adaptações de sua fisiologia e de sua bioquímica são estudadas para entender como conseguem sobreviver aos desafios presentes em cada ambiente. A diminuição da concentração de oxigênio dissolvido na água é um fenômeno natural cíclico em águas do Pantanal e da Amazônia. A crescente poluição antrópica dos rios da Amazônia e do Pantanal pode, somada à hipoxia, oferecer ameaça à permanência de muitas espécies de peixes. Ao estudarmos a carboxilesterase (CarbE), enzima importante para a biotransformação de xenobióticos, observamos que sua atividade diminuía em plasma de pacu, um peixe típico do Pantanal, mantido em hipoxia durante 42 horas. As carboxilesterases participam de inúmeras reações químicas no organismo, incluindo uma transesterificação capaz de produzir ésteres etílicos tóxicos de ácido graxo (FAEE, fatty acid ethyl esters) a partir de etanol e ésteres de ácidos graxos. Já que a diminuição da atividade de CarbE poderia ser uma vantagem, pois que o etanol (um produto da glicólise em peixes sob hipoxia) seria menos esterificado, resolvemos saber mais sobre a bioquímica da CarbE do plasma e do fígado de pacus. Os pacus foram submetidos à hipoxia por diminuição da concentração até 0,5 mg O2/L por meio de borbulhamento de nitrogênio na água. Os animais ficaram nessas condições por 42 horas, quando então coletamos sangue e retiramos seus fígados. A atividade de CarbE ensaiada foi 50% menor no soro e 25% menor nos microssomos de fígado se comparada com a de peixes sob 6 mg O2/L. A CarbE isolada do soro dos pacus em normoxia possui massa molecular relativa de 56.000. A eletroforese em gel desnaturante com a fração purificada rendeu três bandas, mas o gel nativo apresentou só duas bandas com atividades sobre α-naftil acetato. Inferimos que mais do que uma isoforma da enzima está no plasma dos pacus. A CarbE isolada do soro não possui atividade de lipase sobre o Tween 20, mas os microssomos dos fígados dos animais em normoxia e hipoxia possuem. No entanto, a CarbE possui atividade sobre acil-CoA assim como o microssomo de fígado. A enzima pura apresenta Vmáx três vezes maior e uma KM quatro vezes maior do que a atividade presente nos microssomos. Além disso, os microssomos do fígado dos pacus em normoxia podem hidrolisar acil-CoA com o dobro da velocidade daqueles em hipoxia. A atividade clássica de CarbE do soro foi inibida por 4-HNE a 4 mM. A atividade de acil-CoA dos microssomos de fígado também foi sensível a 4-HNE a 4 mM, enquanto 2 mM de 4-HNE inibiu metade desta atividade do soro.

预计到本世纪末,大气CO2浓度将会增加到540~970ppm,大气CO2浓度升高所引起的全球气候变化已经受到广泛的关注。植物生长依赖CO2,并且对大气CO2浓度升高在结构和生理上产生响应。目前已有大量报道,从生态系统、群落、种群、个体、器官、组织、生理以及生化等水平上研究高浓度CO2所对植物产生的影响。但是有关高浓度CO2对植物有性生殖影响的报道却很少,同时多数实验均建立在短期的生殖响应,忽视了植物在长期高CO2浓度下具有的反馈作用和CO2浓度变化对植物的驯化作用。植物有性生殖与其生态适应性和农作物籽粒产量的关系极为密切;同时,植物有性生殖特性的变化,也可作为预测植物对全球气候变化响应的重要指标之一。为此,利用高浓度CO2对植物进行长期选择实验将很有必要。研究结果将为预测未来大气CO2浓度增加的条件下陆地生态系统的演变趋势、全球变化对植物有性生殖响应的方式和机制提供新的思路和有效方法。 在本研究中,我们以模式植物拟南芥(Arabidopsis thaliana)作为实验材料,利用370和700ppm CO2对其进行连续8个世代处理,首先研究高浓度CO2对每一个世代的拟南芥有性生殖特性的影响,然后比较各个世代中各种生殖特性指标变化的规律,从细胞、组织和个体尺度上揭示拟南芥有性生殖对全球变化的响应模式。此外,在700ppm CO2处理下,我们对拟南芥叶片生理、生化以及结构的变化进行了相关研究。两部分研究结果及主要结论如下: 首先,在每一个世代中,与370ppm CO2相比较,700ppm CO2处理显著促进了拟南芥开花,缩短生长周期,增加花、角果及种子等生殖的产量,降低种子N含量,提高种子C/N比、种子千粒重以及生殖生物量所占总生物量的比例等,而对种子萌发率、角果所含种子数目以及角果长度则无显著影响。但是, 通过对相同CO2浓度处理条件下,不同世代之间的研究结果比较发现,不同世代之间相关的生殖生物学指标并无显著差异。 其次,高浓度CO2显著降低叶片气孔密度、气孔指数、气孔导度以及蒸腾速率。在高浓度CO2处理下,叶肉细胞中叶绿体数目、叶绿体宽度和表观面积、淀粉粒大小和数量、叶片和细胞壁厚度等都显著增加,但是基粒内囊体膜的数量却显著下降。叶片中碳水化合物如可溶性总糖、淀粉以及纤维素含量在高浓度CO2下分别显著增加71.9%、78.7% 和 22.3%。此外,在高浓度CO2处理下,叶片中多数激素如如吲哚乙酸(indole-3-acetic acid, IAA)、赤霉素(gibberellin, GA)、玉米素核苷(zeatin riboside, ZR)、二氢玉米素核苷(dihydrozeatin riboside, DHZR)和异戊烯基腺苷(isopentenyl adenosine, iPA)均都显著地增加,而脱落酸(abscisic acid, ABA)含量却有所下降。最后,叶片中各种矿物质元素含量如N、P、K、Ca和Mg等含量在高浓度CO2处理下也都显著下降,而C/N比增加24.8%。 以上结果表明: (1) 在每一个世代中,700ppm CO2处理对拟南芥各种有性生殖特性具有显著的影响,但是高浓度CO2处理对植物所引起的效应在多个世代以内并不能够传递给后代,所以在多个有性生殖世代内,高浓度CO2处理对植物生长、生殖没有驯化作用。 (2) 在高浓度CO2处理下,拟南芥叶片中叶绿体超微结构的变化,可能主要是由于叶绿体中淀粉粒数量和体积大小显著增加而引起。 (3) 在高浓度CO2处理下,由于拟南芥叶片内与促进细胞分裂与伸长的激素含量显著增加,从而对拟南芥植株生长发育速率的提高起了重要的作用。 (4) 拟南芥生长在高浓度CO2条件下,其叶片中各种矿质元素含量(如N、P、K、Ca和Mg)均显著降低,究其原因可能是,第一由于叶片中碳水化合物含量的显著增加而对矿物质元素具有稀释作用;第二由于蒸腾速率下降,引起矿质元素从根部随着蒸腾流运输到地上部分的含量相应减少。

During the low temperature setting of fish paste, myosin heavy chain (MHC) is polymerized to cross-linked myosin heavy chain (CMHC), which is considered to occur by the action of endogenous transglutaminase (TGase). In this study the contribution of TGase on the setting of Alaska pollack surimi at different temperatures was studied. Alaska pollack surimi was ground with 3% NaCl, 30% h2o and with or without ethylene glycol bis (β-aminoethylether) N, N, N¹,N¹- tetra acetic acid (EGTA), an inhibitor of TGase. Among the pastes without EGTA, highest TGase activity was observed at 25°C but breaking force of the gel set at 25°C was lower than that set at 30°, 35°, and 40°C. Addition of EGTA (5m mol/kg) to the paste suppressed TGase activity at all setting temperatures from 20° to 40°C. Gelation of the pastes and cross-linking of MHC on addition of EGTA were suppressed completely at 20° and 25°C, partially at 30° and 35°C, and not at all at 40°C. The findings suggested that during the setting of Alaska pollack surimi TGase mediated cross-linking of MHC was strong at around 25°C but the thermal aggregation of MHC by non-covalent bonds was strong at above 35°C. Setting of surimi at 40°C and cross-linking of its MHC did not involve TGase.

An attempt was made to prepare an intermediate moisture (around 44% moisture) marinated (pH around 4) fish product. Fillets from Sciaenid fish (each fish weighing 70-80 gm) were dipped in a solution containing 7% acetic acid, 20% common salt and 1% propionic acid for 2 hours. After soaking, the soaked fillets were partially dried to about 44% moisture. Three effective hurdles like low pH (by using 7% acetic add and 1% propionic acid), low water activity (by using 20% salt and partially drying the fillets) and preservative (1% propionic add), were used to prepare a shelf-stable product at room temperature. The dried product was sprayed with 0.0 5% BHA in 50% alcohol and further dried for 10 minutes to remove added water and alcohol, thereby another hurdle (preservative) against fat oxidation. The product was packed in 300 gauge polythene bags and stored in transparent screw cap plastic jars. Fortnightly samples were drawn and subjected to biochemical, bacteriological and organoleptic evaluation to study its storage characteristics. The product was in good acceptable form up to 4 months at ambient temperature. The product needed one hour soaking in water with two changes of water in between to make it free from excess salt and acid smell.

A method for the preparation of fish pickles from a lean variety of fish namely pink perch (Nemipterus japonicus) is described. Dipping the fish in 10% sodium chloride solution containing 6% acetic acid before pickling, was found desirable for retaining the meaty texture of the product. The product has no fish smell or flavour and has a shelf life of more than six months at ambient temperatures and scored very well in organoleptic tests.

A simple process is described for extraction of rays from shark fins. The process consists in treating the rays with acetic acid to soften the tissue, separation of the rays by hand and drying. White fins yield almost double the quantity of rays compared to black fins.

Chitosan may be used to reduce the bacterial load of water. Material prepared according to the method of Radhakrishnan & Prahu described in Res. & Ind., 16(4), pp. 265, used in 1% solution in 1% acetic acid was added at 10 ppm level to contaminated water and allowed to stand for 45 min. Cultures of E. coli, Staphylococci and a mixture of the 2 were inoculated into ordinary and muddy water. Bacterial load was determined, and it is shown that chitosan has excellent qualities as a coagulant/water clarifying agent, especially for muddy waters or those contaminated with suspended matter or bacteria.

Results of experiments carried out on canning edible oyster (Crassostrea madrasensis) meat from farmed as well as wild specimens are discussed. The canning yield of meat was 15% higher from farmed oysters compared to wild specimens. The meat from wild oysters was highly slimy and therefore required an additional pre-treatment of washing in brine containing acetic acid to prevent formation of lumps of meat in the can causing problems for proper heat penetration while processing.

The juice extracted from a locally abundant cheap variety of citrus fruit namely, Citrus reticulata was utilized for pickling. The paper highlights the trials made to select the optimum concentrations of acetic acid and sodium chloride to be used along with the juice of C. reticulata so as to obtain the best, product. The product can be stored well at room temperature for six months.

A simple and cheap process for the preservation of mussel meat by marinading is described. The method involves blanching the mussel meat shucked from depurated live mussels in 3% boiling sodium chloride solution for 5 min followed by preserving it in a solution containing 3% acetic acid and 3% sodium chloride. The product stored in closed glass jars has a storage life of approximately 16 weeks at room temperature (23-30°C), after which the quality began to deteriorate. Texture of the meat is least affected and closely resembles that of the fresh meat.

A sensitive and selective liquid chromatography-tandem mass spectrometry method was developed and validated for the simultaneous quantitative determination of microcystin-LR (MC-LR) and its glutathione conjugate (MC-LR-GSH) in fish tissues. The analytes were extracted from fish liver and kidney using 0.01 M EDTA-Na-2-5% acetic acid, followed by a solid-phase extraction (SPE) on Oasis HLB and silica cartridges. High-performance liquid chromatography (HPLC) with electrospray ionization mass spectrometry, operating in selected reaction monitoring (SRM) mode, was used to quantify MC-LR and its glutathione conjugate in fish liver and kidney. Recoveries of analytes were assessed at three concentrations (0.2, 1.0, and 5 mu g g(-1) dry weight [DW]) and ranged from 91 to 103% for MC-LR, and from 65.0 to 75.7% for MC-LR-GSH. The assay was linear within the range from 0.02 to 5.0 mu g g(-1) DW, with a limit of quantification (LOQ) of 0.02 mu g g(-1) DW. The limit of detection (LOD) of the method was 0.007 mu g g(-1) DW in both fish liver and kidney. The overall precision was determined on three different days. The values for within- and between-day precision in liver and kidney were within 15%. This method was applied to the identification and quantification of MC-LR and its glutathione conjugate in liver and kidney of fish with acute exposure of MC-LR. (c) 2007 Elsevier B.V. The performance of polymer-fullerene bulk heterojunction (BHJ) solar cells is strongly dependent on the vertical distribution of the donor and acceptor regions within the BHJ layer. In this work, we investigate in detail the effect of the hole transport layer (HTL) physical properties and the thermal annealing on the BHJ morphology and the solar cell performance. For this purpose, we have prepared solar cells with four distinct formulations of poly(3,4- ethylenedioxythiophene) poly(styrenesulfonate) (PEDOT:PSS) buffer layers. The samples were subjected to thermal annealing, applied either before (pre-annealing) or after (post-annealing) the cathode metal deposition. The effect of the HTL and the annealing process on the BHJ ingredient distribution - namely, poly(3-hexylthiophene) (P3HT) and [6,6]-phenyl C61 butyric acid methyl ester (PCBM) - has been studied by spectroscopic ellipsometry and atomic force microscopy. The results revealed P3HT segregation at the top region of the films, which had a detrimental effect on all pre-annealed devices, whereas PCBM was found to accumulate at the bottom interface. This demixing process depends on the PEDOT:PSS surface energy; the more hydrophilic the surface the more profound is the vertical phase separation within the BHJ. At the same time those samples suffer from high recombination losses as evident from the analysis of the J-V measurements obtained in the dark. Our results underline the significant effect of the HTL-active and active-ETL (electron transport layer) interfacial composition that should be taken into account during the optimization of all polymer-fullerene solar cells. © 2012 The Royal Society of Chemistry.

The dithiophene donor-acceptor copolymers that are bridged either with carbon (C-PCPDTBT) or silicon atoms (Si-PCPDTBT) belong to a promising family of materials for use in photoactive layers for organic photovoltaic cells (OPVs). In this work, we implement the non-destructive Spectroscopic Ellipsometry technique in the near infrared to the far ultraviolet spectral region in combination with advanced theoretical modeling to investigate the vertical distribution of the C-PCPDTBT and Si-PCPDTBT polymer and fullerene ([6,6]-phenyl C71-butyric acid methyl ester - PC70BM) phases in the blend, as well as the effect of the polymer-to-fullerene ratio on the distribution mechanism. It was found that the C-PCPDTBT:PC70BM blends have donor-enriched top regions and acceptor-enriched bottom regions, whereas the donor and acceptor phases are more homogeneously intermixed in the Si-PCPDTBT:PC70BM blends. We suggest that the chemical incompatibility of the two phases as expressed by the difference in their surface energy, may be a key element in promoting the segregation of the lower surface phase to the top region of the photoactive layer. We found that the increase of the photoactive layer thickness reduces the polymer enrichment at the cathode, producing a more homogeneous phase distribution of donor and acceptor in the bulk that leads to the increase of the OPV efficiency. © 2014 Elsevier B.V.

The oligohaline cyanobacterium Aphanizomenon flos-aquae (L.) Ralfs (A. flos-aquae) has been reported in several countries to produce paralytic shellfish poisons (PSPs) or protracted toxic effects. In the past years, A. flos-aquae blooms have occurred annually in the eutrophic Lake Dianchi (300 km(2) in area, located in southwestern China). Material from natural blooms dominated by A. flosaquae was collected and lyophilized. Acute toxicity testing was performed by mouse bioassay using extracts from the lyophilized material. Clear symptoms of PSPs, intoxications were observed. To confirm the production of PSPs, a strain of A. flos-aquae (DC-1) was isolated and maintained in culture. Histopathological effects were studied by examining the organ damages using transmission electron microscopy (TEM). Slight hepatocytic damage with swollen mitochondria was found. The ultrastructural pulmonary lesions were characterized by distortied nuclei and indenting of karyotheca, together with degeneration and tumefaction of mitochondria and endoplasmic reticulum. Control animals injected with acetic acid did not exhibit histopathological damage in any organ. Toxic effects of cultured algal cells on enzymatic systems in the mouse were studied using sublethal doses of extracts. Significant glutathione-S-transferase (GST) and lactate dehydrogenase (LDH) increases, together with decrease of the glutathione (GSH) level, were measured. These results indicated a potential role of PSPs intoxicating and metabolizing in the test animals. HPLC-FLD and LC/MS analysis of extracts from cultured material demonstrated the PSP toxins produced by A. flos-aquae bloom.
