(Reports, 27 February 2015, p. 998) identify wheat DNA from an 8000-calendar-years-before-the-present archaeological site in southern England and conclude that wheat was traded to Britain 2000 years before the arrival of agriculture. The DNA samples are not dated, either directly or from circumstantial evidence, so there is no chronological evidence to support the claim

CONTEXT: Fetal ovarian development and primordial follicle formation underpin future female fertility. Prokineticin (PROK) ligands regulate cell survival, proliferation and angiogenesis in adult reproductive tissues including the ovary. However, their expression and function during fetal ovarian development remains unclear.

OBJECTIVE: To investigate expression and localization of the PROK ligands, receptors and their downstream transcriptional targets in the human fetal ovary.

SETTING: This study was conducted at the University of Edinburgh.

PARTICIPANTS: Ovaries were collected from 37 morphologically normal human fetuses.

DESIGN AND MAIN OUTCOME MEASURES: mRNA and protein expression of PROK ligands and receptors was determined in human fetal ovaries using qRT-PCR, immunoblotting and immunohistochemistry. Functional studies were performed using a human germ tumour cell line (TCam-2) stably transfected with PROKR1.

RESULTS: Expression of PROK1 and PROKR1 was significantly higher in mid-gestation ovaries (17-20 weeks) than at earlier gestations (8-11 and 14-16 weeks). PROK2 significantly increased across the gestations examined. PROKR2 expression remained unchanged. PROK ligand and receptor proteins were predominantly localised to germ cells (including oocytes within primordial follicles) and endothelial cells, indicating these cell types to be the targets of PROK signalling in the human fetal ovary. PROK1 treatment of a germ cell line stably-expressing PROKR1 resulted in ERK phosphorylation, and elevated COX2 expression.

CONCLUSIONS: Developmental changes in expression and regulation of COX2 and pERK by PROK1 suggest that PROK ligands may be novel regulators of germ cell development in the human fetal ovary, interacting within a network of growth and survival factors prior to primordial follicle formation.

Fetal ovarian development and primordial follicle formation are imperative for adult fertility in the female. Data suggest the interleukin (IL)6-type cytokines, leukaemia inhibitory factor (LIF), IL6, oncostatin M (OSM) and ciliary neurotrophic factor (CNTF), are able to regulate the survival, proliferation and differentiation of fetal murine germ cells (GCs) in vivo and in vitro. We postulated that these factors may play a similar role during early human GC development and primordial follicle formation. To test this hypothesis, we have investigated the expression and regulation of IL6-type cytokines, using quantitative reverse transcription polymerase chain reaction and immunohistochemistry. Expression of transcripts encoding OSM increased significantly across the gestational range examined (8-20 weeks), while expression of IL6 increased specifically between the first (8-11 weeks) and early second (12-16 weeks) trimesters, co-incident with the initiation of meiosis. LIF and CNTF expression remained unchanged. Expression of the genes encoding the LIF and IL6 receptors, and their common signalling subunit gp130, was also found to be developmentally regulated, with expression increasing significantly with increasing gestation. LIF receptor and gp130 proteins localized exclusively to GCs, including oocytes in primordial follicles, indicating this cell type to be the sole target of IL6-type cytokine signalling in the human fetal ovary. These data establish that IL6-type cytokines and their receptors are expressed in the human fetal ovary and may directly influence GC development at multiple stages of maturation.

CONTEXT: The formation of primordial follicles occurs during fetal life yet is critical to the determination of adult female fertility. Prior to this stage, germ cells proliferate, enter meiosis, and associate with somatic cells. Growth and survival factors implicated in these processes include activin A (INHBA), the neurotrophins BDNF and NT4 (NTF5), and MCL1. The prostaglandins have pleiotrophic roles in reproduction, notably in ovulation and implantation, but there are no data regarding roles for prostaglandins in human fetal ovarian development.

OBJECTIVE: The aim of the study was to investigate a possible role for prostaglandin (PG) E(2) in human fetal ovary development.

DESIGN: In vitro analysis of ovarian development between 8 and 20 wk gestation was performed.

MAIN OUTCOME MEASURE(S): The expression patterns of PG synthesis enzymes and the PGE(2) receptors EP2 and EP4 in the ovary were assessed, and downstream effects of PGE(2) on gene expression were analyzed.

RESULTS: Ovarian germ cells express the PG synthetic enzymes COX2 and PTGES as well as the EP2 and EP4 receptors, whereas COX1 is expressed by ovarian somatic cells. Treatment in vitro with PGE(2) increased the expression of BDNF mRNA 1.7 +/- 0.16-fold (P = 0.004); INHBA mRNA, 2.1 +/- 0.51-fold (P = 0.04); and MCL1 mRNA, 1.15 +/- 0.06-fold (P = 0.04), but not that of OCT4, DAZL, VASA, NTF5, or SMAD3.

CONCLUSIONS: These data indicate novel roles for PGE(2) in the regulation of germ cell development in the human ovary and show that these effects may be mediated by the regulation of factors including BDNF, activin A, and MCL1.

Five to ten percent of individuals with melanoma have another affected family member, suggesting familial predisposition. Germ-line mutations in the cyclin-dependent kinase (CDK) inhibitor p16 have been reported in a subset of melanoma pedigrees, but their prevalence is unknown in more common cases of familial melanoma that do not involve large families with multiple affected members. We screened for germ-line mutations in p16 and in two other candidate melanoma genes, p19ARF and CDK4, in 33 consecutive patients treated for melanoma; these patients had at least one affected first or second degree relative (28 independent families). Five independent, definitive p16 mutations were detected (18%, 95% confidence interval: 6%, 37%), including one nonsense, one disease-associated missense, and three small deletions. No mutations were detected in CDK4. Disease-associated mutations in p19ARF, whose transcript is derived in part from an alternative codon reading frame of p16, were only detected in patients who also had mutations inactivating p16. We conclude that germ-line p16 mutations are present in a significant fraction of individuals who have melanoma and a positive family history.

Arsenic (As) uptake and distribution in the roots, shoots, and grain of wheat (Triticum durum) grown in 2 As polluted soils (192 and 304 mg kg -1 respectively), and an uncontaminated soil (14 mg kg-1 ), collected from Scarlino plain (Tuscany, Italy), was investigated with respect with phosphorus fertilization. Three different level of phosphorus (P) fertilization: PO [0 kg ha-1], Pl [75 kg ha-1], and P2 [150 kg ha-1], as KH2PO4 of P, were applied. The presence of high concentrations of As in soils reduced plants growth, decreased grain yield and increased root, shoot and grain As concentrations, especially in the absence of P fertilization. The P fertilization decreased the As concentration in all the tissues as well as the translocation of As to the shoot and grain. This observation may be useful in certain areas of the world with high levels of As in soils, to reduce the potential risk posed to human health by As entering the food-chain. © by PSP.

In this study we have investigated the uptake and distribution of arsenic (As) and phosphate (Pi) in roots, shoots, and grain of wheat grown in an uncontaminated soil irrigated with solutions containing As at three different concentrations (0.5, 1 and 2 mg l-1) and in the presence or in the absence of P fertilization. Arsenic in irrigation water reduced plants growth and decreased grain yield. When Pi was not added (P-), plants were more greatly impacted compared to the plus Pi (P+) treatments. The differences in mean biomass between P- and P+ treatments at the higher As concentrations demonstrated the role of Pi in preventing As toxicity and growth inhibition. Arsenic concentrations in root, shoot and grain increased with increasing As concentration in irrigation water. It appears that P fertilization minimizes the translocation of As to the shoots and grain whilst enhancing P status of plant. The observation that P fertilization minimises the translocation of arsenic to the shoots and grain is interesting and may be useful for certain regions of the world that has high levels of As in groundwater or soils. © 2008 Springer Science+Business Media B.V.

Analgesics which affect prostaglandin (PG) pathways are used by most pregnant women. As germ cells (GC) undergo developmental and epigenetic changes in fetal life and are PG targets, we investigated if exposure of pregnant rats to analgesics (indomethacin or acetaminophen) affected GC development and reproductive function in resulting offspring (F1) or in the F2 generation. Exposure to either analgesic reduced F1 fetal GC number in both sexes and altered the tempo of fetal GC development sex-dependently, with delayed meiotic entry in oogonia but accelerated GC differentiation in males. These effects persisted in adult F1 females as reduced ovarian and litter size, whereas F1 males recovered normal GC numbers and fertility by adulthood. F2 offspring deriving from an analgesic-exposed F1 parent also exhibited sex-specific changes. F2 males exhibited normal reproductive development whereas F2 females had smaller ovaries and reduced follicle numbers during puberty/adulthood; as similar changes were found for F2 offspring of analgesic-exposed F1 fathers or mothers, we interpret this as potentially indicating an analgesic-induced change to GC in F1. Assuming our results are translatable to humans, they raise concerns that analgesic use in pregnancy could potentially affect fertility of resulting daughters and grand-daughters.

Chromosome structure and behaviour in both meiosis of the germ cells and mitosis of the embryo from fertilisation to the two-cell stage in Bursaphelenchus xylophilus were examined by DAPI staining and three-dimensional reconstruction of serial-section images from confocal laser-scanning microscopy. By this method, each chromosome’s shape and behaviour were clearly visible in early embryogenesis from fertilisation through the formation and fusion of the male and female pronuclei to the first mitotic division. The male pronucleus was bigger than that of the female, although the oocyte is larger and richer in nutrients than the sperm. From the shape of the separating chromosomes at anaphase, the mitotic chromosomes appeared to be polycentric or holocentric rather than monocentric. Each chromosome was clearly distinguishable in the male and female germ cells, pronuclei of the one-cell stage embryo, and the early embryonic nuclei. The haploid number of chromosomes (N) was six (2n = 12), and all chromosomes appeared similar. The chromosome pair containing the ribosomal RNA-coding site was visualised by fluorescence in situ hybridisation. Unlike the sex determination system in Caenorhabditis elegans (XX in hermaphrodite and XO in male), the system for B. xylophilus may consist of an XX female and an XY male.

Com o presente trabalho pretendeu-se determinar e compreender melhor quais os alvos do Alumínio (Al) nas plantas, e contribuir para um melhor entendimento dos mecanismos de tolerância presentes em genótipos com elevado grau de tolerância ao Al. O Al é um dos maiores constituintes do solo e torna-se biodisponível em solos com baixo pH. Nesses casos, a exposição ao Al afecta negativamente o crescimento das plantas conduzindo a uma diminuição da produção. Estes factos são especialmente visíveis nos cereais, sendo a exposição ao Al uma das principais causas das quebras de produção nestas espécies. O Capítulo I consiste numa revisão geral sobre a toxicidade do Al nas plantas, apontando os seus principais alvos. Apresenta também os mecanismos de resistência, que inclui Al-destoxificação externa e interna, em diferentes espécies. O Capítulo II aborda os estudos sobre a exposição de curto prazo ao Al em duas espécies de cereais: Triticum aestivum L. e Secale cereale L., tendo-se sempre utilizado um genótipo Al-tolerante e um Al-sensível para cada espécie. Este capítulo está dividido em três estudos: no Capítulo II.1 realça-se o efeito da exposição a 185 μM de Al no equilíbrio nutricional em trigo. Verificou-se que em ambos os genótipos (sensível e tolerante) o perfil de macro e micro nutrientes se alterou, tendo uma interferência negativa, sobretudo no nível de P, Mg e K. Além disso, registaram-se diferenças na diferenciação da endoderme consoante o grau de tolerância/sensibilidade do genótipo. No Capítulo II.2 apresenta-se uma visão mais abrangente dos efeitos da exposição a 185 μM de Al em trigo, incluindo parâmetros fisiológicos, estruturais, citológicos e genotóxicos. Demonstra-se, pela primeira vez, que a progressão do ciclo celular é diferentemente regulada, dependendo da tolerância/sensibilidade do genótipo e que, mesmo em zonas já diferenciadas da raiz a exposição ao Al leva à deposição de calose. O Capítulo II.3 aborda os efeitos da exposição de 1.1 mM de Al em centeio, numa perspectiva bastante alargada. Apresenta-se o desequilíbrio nutricional, sobretudo no genótipo sensível, assim como a translocação de Al para a parte aérea nesse mesmo genótipo. Analisa-se também o comportamento de ambos os genótipos no que se refere ao ciclo celular, diferenciação da endoderme, crescimento radicular, reservas de hidratos de carbono, entre outros. Os resultados apontam para estratégias bem definidas adoptadas pelo genótipo tolerante de forma a minimizar a acção do Al no sistema radicular. O Capítulo III compreende a exposição longa ao Al. Dois genótipos de centeio com diferentes graus de tolerância ao Al foram expostos a 1.11 mM e 1.85 mM de Al durante 21 dias, tendo sido usados dois pontos de amostragem (15 e 21 dias). Este capítulo está dividido em dois estudos: No Capítulo III. 1 analisamse os mecanismos antioxidantes (folhas e raízes) como resposta à exposição ao Al, dando-se especial atenção ao ciclo do ascorbato-glutationas. A exposição ao Al levou a stress oxidativo e a alterações na actividade de enzimas antioxidantes e no conteúdo de antioxidantes não-enzimáticos. Demonstra-se que os dois órgãos apresentam respostas diferentes à exposição ao Al e que a capacidade de sobreviver em ambientes ricos em Al depende da eficácia da resposta antioxidante. Para além disso, a resposta do ciclo ascorbato-glutationas parece estar dependente do tipo de órgão, grau de tolerância e do tempo de exposição ao Al. No Capítulo III. 2 analisam-se os efeitos da exposição ao Al na fotossíntese. Verificou-se que o Al afecta negativamente a taxa fotossintética em ambos os genótipos, embora as alterações que o Al provoca nas trocas gasosas e no Ciclo de Calvin sejam dependentes do genótipo. Verificou-se também que os danos no genótipo sensível surgem mais cedo do que no genótipo tolerante, mas que ambos apresentam susceptibilidade ao Al após exposição de longo termo. Por fim, no Capítulo IV são apresentadas as conclusões da Tese de Doutoramento.

Following the 1978 rural reform, a series of agricultural reforms were introduced in China with an aim to create incentives for the farmers to produce more. However, the nineties’ reforms towards liberalization eventually resulted in a huge drop in agricultural production, which apparently motivated the grain self-sufficiency program in 1998. For a dataset that covers wheat production during these reforms, we examine how and to what extent these reforms affected the Total Factor Productivity (TFP) and the welfare of wheat farmers in China, both at the national and at the regional level. We find that although the nineties' price reforms led to a relatively faster growth of the incentivized TFP of wheat production, they failed to improve profits vis a vis welfare for the farmers. A series of weather shocks in the early nineties resulted in a scarcity of cultivable land and a shortage of agricultural labour, which eventually led to a sharp increase in their relative prices. The introduction of grain self-sufficiency program stabilized these agricultural prices but destroyed the growth in TFP for most regions. However, this reform resulted in some improvement in farmers’ welfare. Wheat farmers in China therefore experienced a trade off between productivity and welfare; competition boosted their productivity and regulation improved their welfare. Not only these findings add a completely new set of results to the existing literature, they can also form a strong basis for future agricultural reforms in China.

In 1893 Attorney Henry Drushel Perky and William H. Ford invented a machine for the preparation of cereals for food. Perky soon realized that the actual cereal biscuits were more popular than the machines and he opened Shredded Wheat plants in Massachusetts and Niagara Falls, New York. In 1904, the Niagara Falls, Canada plant was opened. These factories also served as tourist attractions. In 1907, one hundred thousand people visited the plants on both sides of the border. In 1928, the company was sold to The National Biscuit Company and the product name changed to Nabisco Shredded Wheat. The name of the plant was changed to Nabisco Foods in 1956 to reflect the variety of foods that were being produced at that time. In 1985, Nabisco was purchased by J.R. Reynolds thus forming RJR Nabisco. In 1994, RJR sold its breakfast cereal business to Kraft Foods and the international licenses to General Mills. The present study is an attempt to find out the ralation between RNA/DNA ratio, protein,percentage growth rate and specific growth rate of prawn,Penaeus indicus with respect to Nervous system, Eyestalk and Muscle tissues during ontogenesis. We have isolated and purified a natural agglutinin in the hemolymph of P.indicus with antigenecity, agglutinating, hemolytic and antibacterial properties. The influence of growth and environmental parameters on the level of agglutinin in the hemolymph was studied. Agglutinin concentration during normal growth process was compared. The agglutinin concentration in the hemolymph was quantified through developing ELISA, which is useful in health monitoring studies of individual species. Complete amino acid composition of both the subunits of P.indicus agglutinin were analysed. P.indicus agglutinin showed similarity to those proteins having antigenecity,hemolytic and agglutinating properties.Hence, agglutinin was considered as a natural defence protein in the hemolymph of P.indicus responsible for immune surveillance. The humoral defence mechanism of agglutinin was a co-operative effort with hemocytes and complement system. The composition of isolated agglutinin of P.indicus amino acids will be helpful in the synthesis of new antibacterial analogues which can be used against disease causing organisms.
