The taxonomy of the Hanuman langur (Semnopithecus spp.), a widely distributed Asian colobine monkey, has been in a flux for a long time due to much disagreement between various classification schemes. However, results from a recent field-based morphological study were consistent with Hill's (Ceylon J Sci 21:277-305, 1939) species level classification scheme. Here we tested the validity of S. hypoleucos and S. priam, the two South Indian species recognized by Hill. To this end, one mitochondrial and four nuclear markers were sequenced from over 72 non-invasive samples of Hanuman langurs and S. johnii collected from across India. The molecular data were subjected to various tree building methods. The nuclear data was also used in a Bayesian structure analysis and to determine the genealogical sorting index of each hypothesized species. Results from nuclear data suggest that the South Indian population of Hanuman langur consists of two units that correspond to the species recognized by Hill. However in the mitochondrial tree S. johnii and S. priam were polyphyletic probably due to retention of ancestral polymorphism and/or low levels of hybridization. Implications of these results on conservation of Hanuman langurs are also discussed.

Callithrix jacchus (common marmoset) is a New World primate monkey, used as an animal model in biomedical research. Marmoset-specific follicle-stimulating hormone (FSH) preparation is required to improve superovulation protocols and to develop homologous FSH monitoring assays in these monkeys. In this study, we document the large-scale expression of recombinant marmoset FSH in methylotropic yeast, Pichia pastoris. The recombinant preparation was found to be immunologically active in Western blotting and radioimmunoassay. The preparation displayed receptor binding ability in radioreceptor assay. Based on the receptor binding ability, the yield of fermentation was estimated to be 7.2 mg/L. FSH-induced cAMP assay and estradiol assay revealed that the recombinant hormone is able to induce signal transduction. Both immunological and in vitro biological activity of marmoset FSH was found to be comparable to purified human pituitary FSH, which served as reference hormone for these assays. Thus, the study suggests that a Pichia expression system can be used for large-scale expression of bioactive recombinant marmoset FSH.

Resumen: Son numerosas las fuentes literarias que dan cuenta de los raptos de doncellas llevados a cabo por animales, de cuya unión se desprenden a su vez los nacimientos monstruosos de niños que reciben cualidades de su padre-animal, tanto físicamente como en su carácter, aunque en ocasiones lleguen a conservar en gran parte su apariencia antropomórfica. Historias de este tipo se propagan durante los siglos xv y xvi en libros de misceláneas y casos extraños, pero continúan aún vivos en los siglos posteriores, cuando adquieren nuevas formas y variantes a través del folklore, la narrativa y la lírica popular. Analizaremos particularmente los raptos ejecutados por el oso y el simio, dos animales en los que el hombre del Medioevo vio especialmente la lujuria.

Sensory-motor circuits course through the parietal cortex of the human and monkey brain. How parietal cortex manipulates these signals has been an important question in behavioral neuroscience. This thesis presents experiments that explore the contributions of monkey parietal cortex to sensory-motor processing, with an emphasis on the area's contributions to reaching. First, it is shown that parietal cortex is organized into subregions devoted to specific movements. Area LIP encodes plans to make saccadic eye movements. A nearby area, the parietal reach region (PRR), plans reaches. A series of experiments are then described which explore the contributions of PRR to reach planning. Reach plans are represented in an eye-centered reference frame in PRR. This representation is shown to be stable across eye movements. When a sequence of reaches is planned, only the impending movement is represented in PRR, showing that the area is more related to movement planning than to storing the memory of reach targets. PRR resembles area LIP in each of these properties: the two areas may provide a substrate for hand-eye coordination. These findings yield new perspectives on the functions of the parietal cortex and on the organization of sensory-motor processing in primate brains.

Waking up from a dreamless sleep, I open my eyes, recognize my wife’s face and am filled with joy. In this thesis, I used functional Magnetic Resonance Imaging (fMRI) to gain insights into the mechanisms involved in this seemingly simple daily occurrence, which poses at least three great challenges to neuroscience: how does conscious experience arise from the activity of the brain? How does the brain process visual input to the point of recognizing individual faces? How does the brain store semantic knowledge about people that we know? To start tackling the first question, I studied the neural correlates of unconscious processing of invisible faces. I was unable to image significant activations related to the processing of completely invisible faces, despite existing reports in the literature. I thus moved on to the next question and studied how recognition of a familiar person was achieved in the brain; I focused on finding invariant representations of person identity – representations that would be activated any time we think of a familiar person, read their name, see their picture, hear them talk, etc. There again, I could not find significant evidence for such representations with fMRI, even in regions where they had previously been found with single unit recordings in human patients (the Jennifer Aniston neurons). Faced with these null outcomes, the scope of my investigations eventually turned back towards the technique that I had been using, fMRI, and the recently praised analytical tools that I had been trusting, Multivariate Pattern Analysis. After a mostly disappointing attempt at replicating a strong single unit finding of a categorical response to animals in the right human amygdala with fMRI, I put fMRI decoding to an ultimate test with a unique dataset acquired in the macaque monkey. There I showed a dissociation between the ability of fMRI to pick up face viewpoint information and its inability to pick up face identity information, which I mostly traced back to the poor clustering of identity selective units. Though fMRI decoding is a powerful new analytical tool, it does not rid fMRI of its inherent limitations as a hemodynamics-based measure.

O presente estudo teve como objetivo descrever o conteúdo das representações sociais acerca da Aids para os usuários soropositivos em acompanhamento ambulatorial da rede pública de saúde e analisar a interface das representações sociais da Aids com o cotidiano dos indivíduos que vivem com o HIV, especialmente no que concerne à sua organização e ao processo de adesão ao tratamento. Trata-se de um estudo exploratório-descritivo, pautado na abordagem qualitativa e orientado pela Teoria das Representações Sociais. Os sujeitos consistiram em 30 usuários em acompanhamento ambulatorial de um Hospital Público Municipal localizado na cidade do Rio de Janeiro referenciado para clientes soropositivos ao HIV/Aids. Os dados foram coletados por meio de entrevista e analisados através da análise de conteúdo. Como resultados, emergiram 6 categorias, quais sejam: Elementos de memória da Ancoragem da Aids na sociedade e o seu processo de transformação, onde foi explicitada a ancoragem da Aids no outro, na África, no macaco, no homossexual e uma nova ancoragem apresentada consiste na cronicidade do diabetes, deixando a síndrome de ser sinônimo de morte; Transmissão e Prevenção da Aids segundo as pessoas que convivem com a síndrome, na qual os sujeitos apresentaram quase todas as formas cientificamente comprovadas quanto aos meios de transmissão do vírus HIV; O cotidiano dos indivíduos soropositivos permeado pelo processo de vulnerabilidade ao HIV, no âmbito do qual entende-se que o reconhecimento do risco individual frente à epidemia irá influenciar, sobretudo, as práticas e os comportamentos das pessoas; Discriminação e ocultamento no conviver com o HIV, onde se apresenta como estratégias de sobrevivência social o ocultamento do estado de soropositividade ao HIV. Assim, podem continuar a vida como pessoas consideradas normais, sem serem acusadas e discriminadas, sejam no âmbito familiar, social ou no trabalho; além disso, os sujeitos do estudo declararam que eram preconceituosos antes do diagnóstico; o processo de adesão ao tratamento na cotidianidade de indivíduos soropositivos, observando-se, nesta categoria, que um dos grandes motivadores da adesão ao tratamento consiste no fato dos usuários acreditarem no resultado positivo da terapêutica; o enfrentamento cotidiano experiênciado pelos sujeitos que convivem com o HIV, onde a forma como os sujeitos organizam o seu cotidiano para enfrentar e conviver com o HIV reflete diretamente em suas atitudes e em suas práticas, tanto no processo da adesão, como nas relações sociais (o outro) e, principalmente, na relação individual (o eu). Conclui-se que a representação social da Aids apresenta-se multifacetada e dependente do contexto histórico e social no qual o indivíduo está inserido, seus valores, cultura, nível de informação e conhecimento.

A study was conducted on the adsorption of Escherichia coli bacteriophage T4 to activated carbon. Preliminary adsorption experiments were also made with poliovirus Type III. The effectiveness of such adsorbents as diatomaceous earth, Ottawa sand, and coconut charcoal was also tested for virus adsorption.
The kinetics of adsorption were studied in an agitated solution containing virus and carbon. The mechanism of attachment and site characteristics were investigated by varying pH and ionic strength and using site-blocking reagents.
Plaque assay procedures were developed for bacteriophage T4 on Escherichia coli cells and poliovirus Type III on monkey kidney cells. Factors influencing the efficiency of plaque formation were investigated.
The kinetics of bacteriophage T4 adsorption to activated carbon can be described by a reversible second-order equation. The reaction order was first order with respect to both virus and carbon concentration. This kinetic representation, however, is probably incorrect at optimum adsorption conditions, which occurred at a pH of 7.0 and ionic strength of 0.08. At optimum conditions the adsorption rate was satisfactorily described by a diffusion-limited process. Interpretation of adsorption data by a development of the diffusion equation for Langmuir adsorption yielded a diffusion coefficient of 12 X 10-8 cm2/sec for bacteriophage T4. This diffusion coefficient is in excellent agreement with the accepted value of 8 X 10-8 cm2/sec. A diffusion-limited theory may also represent adsorption at conditions other than the maximal. A clear conclusion on the limiting process cannot be made.
Adsorption of bacteriophage T4 to activated carbon obeys the Langmuir isotherm and is thermodynamically reversible. Thus virus is not inactivated by adsorption. Adsorption is unimolecular with very inefficient use of the available carbon surface area. The virus is probably completely excluded from pores due to its size.
Adsorption is of a physical nature and independent of temperature. Attraction is due to electrostatic forces between the virus and carbon. Effects of pH and ionic strength indicated that carboxyl groups, amino groups, and the virus's tail fibers are involved in the attachment of virus to carbon. The active sites on activated carbon for adsorption of bacteriophage T4 are carboxyl groups. Adsorption can be completely blocked by esterifying these groups.

Na década de 80 dramáticas ocorrências atingiram o país e o mundo na área da saúde com a descoberta da Síndrome da Imunodeficiência Adquirida (Aids) síndrome, causada pelo Vírus da Imunodeficiência Humana (HIV). O objetivo deste estudo foi descrever as representações sociais do HIV/Aids e as memórias sociais do cuidado de enfermagem construídas na década de 80 pela equipe de enfermagem. Optou-se por uma abordagem qualitativa, embasada na teoria de representações sociais e nos conceitos do campo da memória social. Os sujeitos do estudo foram 20 profissionais de enfermagem de serviços ambulatoriais e/ou da atenção básica, atuantes em 11 instituições públicas de saúde da cidade do Rio de Janeiro que possuem o Programa Nacional de DST/Aids. A coleta de dados foi realizada através de entrevista semiestruturada e questionário de caracterização sócio profissional. A análise dos dados deu-se em duas etapas, a primeira atraves da técnica de análise de conteúdo temática destinada a identificar nos depoimentos dos entrevistados os conteúdos discursivos relativos a década de 80. Posteriormente os trechos selecionados foram submetidos à análise lexical pelo software Alceste 4.10. Obteve-se três classes temáticas que abordaram: As percepções e as ações do cuidado de enfermagem na década de 80; Os primeiros contatos profissionais e pessoais com HIV/Aids e A mídia e a construção das representações sociais do HIV/Aids. Na primeira classe os profissionais de enfermagem relatam as memórias referentes aos cuidados prestados na década de 80, descrevendo como esse cuidado era prestado, o medo da contaminação e os profissionais que atuavam na prestação de serviços. Na classe 2 os sujeitos resgatam as primeiras vivências com as pessoas com HIV/Aids e os sentimentos experimentados neste primeiro contato. As características físicas, os aspectos emocionais, a introdução do AZT e o abandono familiar são elementos destacados. Na classe 3 são relatas as memórias referentes ao início da epidemia de HIV/Aids, com destaque para as ancoragens representacionais do surgimento do vírus, tendo especialmente o macaco como hospedeiro. Os meios de comunicação surgiram como formadores das memórias do início da epidemia, veiculando imagens, como a do cantor Cazuza, fortemente citado pelos sujeitos. Conclui-se que este estudo permitiu compreender, através das memórias e das representações, como se constituiu a atuação dos profissionais no início da epidemia, assim como a permanência de elementos simbólicos até hoje nas representações sociais do HIV/Aids.

O sagui-da-serra-escuro (Callithrix aurita) (É.Geoffroy, 1812) é uma espécie endêmica da Mata Atlântica e ameaçada de extinção, considerada em perigo. Callithrix jacchus e C. penicillata são espécies invasoras no Estado do Rio de Janeiro, que competem com C. aurita pelos mesmos recursos, além de formarem híbridos. Nesse contexto, avaliamos a distribuição espacial das espécies de saguis (nativa e invasoras) no interior e entorno do Parque Nacional da Serra dos Órgãos (PARNASO), RJ, Brasil, através do método de playback, entrevistas, e a modelagem de ocupação e detecção. Além disso, comparamos os dados de distribuição atual com os registros de ocorrência anteriores a esse estudo. Os resultados mostraram que a população de saguis nativos e invasores presentes no interior no parque possuem probabilidade de ocupação (0,20 e 0,22), detecção (0,22 e 0,26) e abundância (12,65 e 13,68 grupos) semelhantes. A ocupação de C. aurita está relacionada às áreas do parque mais afastadas da estrada e com menor interferência humana, provavelmente pelo efeito da elevada altitude nessas regiões. Enquanto que a ocupação dos saguis invasores está relacionada à proximidade dos limites do parque e a maior interferência humana, tanto em maiores altitudes quanto em menores. Registramos o processo de hibridação entre saguis nativos e invasores, e a formação de grupos mistos entre eles, em diversos pontos da região de Petrópolis, inclusive no interior do parque, evidenciando as consequências do processo de invasão. Concluímos que a população de Callithrix aurita no PARNASO é pequena, restrita a uma única região do parque, e parte dela possui contato com grupos de saguis invasores, ou está bem próxima deles. As espécies de saguis invasores ocorrem em alguns locais no interior do parque e por todo o seu entorno. Portanto, há uma pressão dos saguis invasores em direção ao interior do PARNASO. Diante deste processo de extinção local de uma espécie endêmica, ameaçada de extinção, e com uma restrita distribuição geográfica, alertamos para a urgente necessidade de iniciar um eficiente programa de manejo das espécies de saguis invasores, juntamente com a reintrodução de grupos de Callithrix aurita.

In 1989-1991, the U.S. Fish and Wildlife Service surveyed breeding populations of seabirds on the entire California coast. This study was sponsored by the Minerals Management Service in relation to outer continental shelf oil and gas leasing. At 483 nesting sites (excluding terns and skimmers in southern California), we estimated 643,307 breeding birds of 21 seabird species including: 410 Fork-tailed Storm-petrel (Oceanodroma furcata); 12,551 Leach's Storm-petrel (O. leucorhoa); 7,209 Ashy Storm-petrel (O. homochroa); 274 Black Storm-petrel (O. melania); 11,916 Brown Pelican (Pelecanus occidentalis); 10,037 Double-crested Cormorant (Phalacrocorax auritus); 83,394 Brandt's Cormorant (P. penicillatus); 14,345 Pelagic Cormorant (P. pelagicus); 888 Black Oystercatcher (Haemotopus bachmani); 4,764 California Gull (Larus californicus); 61,760 Western Gull (L. occidentalis); 2,838 Caspian Tern (Sterna caspia) (excluding southern California); 3,550 Forster's Tern (S. forsteri) (excluding southern California); 272 Least Tern (S. albifrons) (excluding southern California); 351,336 Common Murre (Uria aalge); 15,470 Pigeon Guillemot (Cepphus columba); 1,821 Marbled Murrelet (Brachyramphus marmoratus); 1,760 Xantus' Murrelet (Endomychura hypoleuca); 56,562 Cassin's Auklet (Ptychoramphus aleuticus); 1,769 Rhinoceros Auklet (Cerorhinca monocerata); and 276 Tufted Puffin (Fratercula cirrhata). The inland, historical or hybrid breeding status of American White Pelican (P. erythrorynchus), American Oystercatcher (H. palliatus), Heermann's Gull (L. heermanni), Ring-billed Gull (L. delawarensis), Glaucous-winged Gull (L. glaucescens) and Black Tern (Chlidonias niger) are discussed. Estimates for Gull-billed Tern (S. nilotica), Royal Tern (S. maxima), Elegant Tern (S. elegans) and Black Skimmer (Rhynchops niger) will be included in the final draft of this report. Overall numbers were slightly lower than reported in 1975-1980 surveys (summarized in Sowls et al. 1980. Catalog of California seabird colonies. U.S. Dept. Int., Fish Wildl. Serv., Biol. Serv. Prog., FWS/OBS 37/80). Recent declines were found or suspected for Fork-tailed Storm-petrel, Leach's Storm-petrel, White Pelican, Black Tern, Caspian Tern, Least Tern, Common Murre and Marbled Murrelet. Recent increases were found or suspected for Brown Pelican, Double-crested cormorant, California Gull, Western Gull, Forster's Tern and Rhinoceros Auklet. Similar numbers were found for other species or trends could not be determined without additional surveys, studies and/or more in-depth comparisons with previous surveys. The status of terns and skimmers in southern California has not yet been finalized.

为筛选出与认知、记忆相关的脑部表达基因及基因家族,利用TRIzol试剂从恒河猴大脑前额叶组织抽提总RNA,再用纯化试剂盒从总RNA中成功纯化出mRNA。按照Stratagene公司的cDNASynthesisKit(200401)、ZAP-cDNASynthesisKit(200400)和ZAP-cDNAGigapackⅢGoldCloningKit(200450)三个试剂盒的操作说明,构建了恒河猴大脑前额叶组织的cDNA文库。文库总库容为2·0×106克隆;绝大多数的cDNA插入片段≥0·5kb,平均长度≈1·0kb;cDNA片段与噬菌体载体重组率为97·3%。文库各项指标均达要求,为克隆大脑PFC区表达基因、测定基因编码区序列、揭示具有多种剪切组合模式等提供了可靠资源,也为相关基因表达调控的研究提供了方便。

目的 从体外培养成熟囊胚中分离并鉴定猕猴胚胎干细胞(embryonic stem cell , ES cell) 。方法 猕猴卵母细 胞经体外成熟培养、体外受精和早期胚胎体外成熟培养后,获得猕猴囊胚。当囊胚由透明带自然孵出后,用细玻璃针剥离 囊胚中的内细胞团(inner cell mass , ICM) 并与饲养细胞进行共培养。由ICM分离,培养并鉴定胚胎干细胞集落。结果 由 4 只FSH 超排猕猴中共取得92 个处于GV 期的猕猴卵母细胞,选取其中的22 个用HECM210 培养基培养后,获得6 个高质 量的囊胚,由此6 个囊胚中分离得到3 个内细胞团,并由此最终获得1 株猕猴ES 细胞,即RS5 细胞。RS5 细胞具高比例核P 质比,核仁多,其细胞集落边缘平整,其内各单个细胞清晰。经约5 个月的连续传代后,仍保持了正常二倍体的核型,其染 色体数目为42 条。碱性磷酸酶细胞组织化学染色为阳性,说明RS5 细胞为未分化态的胚胎干细胞。经高密度和长时间 培养后,RS5 细胞可进一步分化为多种类型细胞。结论 RS5 细胞株具有自我更新能力和多分化潜能,属于胚胎干细胞。

BACKGROUND: Effects of 17beta-estradiol and progesterone on rhesus monkey oocyte maturation in vitro were evaluated by embryo development subsequent to IVF. METHODS AND RESULTS: In experiment 1, immature cumulus-oocyte complexes collected from unstimulated adult females during the non-breeding season were matured in modified medium CMRL-1066 containing various combinations of gonadotrophins (FSH + LH), estradiol and/or progesterone. Formation of morulae and blastocysts was greatest in oocytes matured in medium containing estradiol and/or progesterone, with or without gonadotrophins (morula 38-46%, blastocyst 14-20%) than in control oocytes matured without estradiol or progesterone (morula 14%, blastocyst 0%). In experiment 2, cumulus-oocyte complexes from unstimulated prepubertal female monkeys were matured in medium with gonadotrophins, estradiol or progesterone. The best development to the morula stage was obtained with oocytes matured with gonadotrophins and estradiol or gonadotrophins and progesterone (43 and 25 morulae, respectively), while control oocytes matured with gonadotrophins but without steroid hormones gave the poorest morula developmental response (12%). However, there was no difference in blastocyst development across all groups (0-3%). CONCLUSIONS: These results demonstrate that during rhesus monkey oocyte maturation in vitro: (i) estradiol or progesterone can improve oocyte developmental competence; (ii) immature oocytes from prepubertal versus adult females have differential responses to challenge with estradiol or progesterone.
