Serine/threonine protein kinase AMP-activated protein kinase (AMPK) is a key metabolic stress-responsive factor that promotes the adaptation of cells to their microenvironment. Elevated concentrations of intracellular AMP, caused by metabolic stress, are known to activate AMPK by phosphorylation of the catalytic subunit. Recently, the tumor suppressor serine/threonine protein kinase LKB1 was identified as an upstream kinases, AMPKKs. In the current study, we found that stimulation with growth factors also caused AMPK-alpha subunit phosphorylation. Interestingly, even an LKB1-nonexpressing cancer cell line, HeLa, exhibited growth factor-stimulated AMPK-alpha subunit phosphorylation, suggesting the presence of an LKB1-independent pathway for AMPK-alpha subunit phosphorylation. In the human pancreatic cancer cell line PANC-1, AMPK-alpha subunit phosphorylation promoted by IGF-I was suppressed by antisense ataxia telangiectasia mutated (ATM) expression. We found that IGF-1 also induced AMPK-alpha subunit phosphorylation in the human normal fibroblast TIG103 cell line, but failed to do so in a human fibroblast AT2-KY cell line lacking ATM. Immunoprecipitates of ATM collected from IGF-1-stimulated cells also caused the phosphorylation of the AMPK-alpha subunit in vitro. IGF-1-stimulated ATM phosphorylation at both threonine and tyrosine residues, and our results demonstrated that the phosphorylation of tyrosine in the ATM molecule is important for AMPK-alpha subunit phosphorylation during IGF-1 signaling. These results suggest that IGF-1 induces AMPK-alpha subunit phosphorylation via an ATM-dependent and LKB1-independent pathway. (C) 2004 Elsevier Inc. All rights reserved.

Aim To evaluate whether the T1D susceptibility locus on chromosome 16q contributes to the genetic susceptibility to T1D in Russian patients. Method Thirteen microsatellite markers, spanning a 47-centimorgan genomic region on 16q22-q24 were evaluated for linkage to T1D in 98 Russian multiplex families. Multipoint logarithm of odds (LOD) ratio (MLS) and nonparametric LOD (NPL) values were computed for each marker, using GENEHUNTER 2.1 software. Four microsatellites (D16S422, D16S504, D16S3037, and D16S3098) and 6 biallelic markers in 2 positional candidate genes, ICSBP1 and NQO1, were additionally tested for association with T1D in 114 simplex families, using transmission disequilibrium test (TDT). Results A peak of linkage (MLS = 1.35, NPL = 0.91) was shown for marker D16S750, but this was not significant (P = 0.18). The subsequent linkage analysis in the subset of 46 multiplex families carrying a common risk HLA-DR4 haplotype increased peak MLS and NPL values to 1.77 and 1.22, respectively, but showed no significant linkage (P = 0.11) to T1D in the 16q22-q24 genomic region. TDT analysis failed to find significant association between these markers and disease, even after the conditioning for the predisposing HLA-DR4 haplotype. Conclusion Our results did not support the evidence for the susceptibility locus to T1D on chromosome 16q22-24 in the Russian family data set. The lack of association could reflect genetic heterogeneity of type 1 diabetes in diverse ethnic groups.

Efficient insulin action requires spatial and temporal coordination of signaling cascades. The prototypical insulin receptor substrate, IRS-1 plays a central role in insulin signaling. By subcellular fractionation IRS-1 is enriched in a particulate fraction, termed the high speed pellet (HSP), and its redistribution from this fraction is associated with signal attenuation and insulin resistance. Anecdotal evidence suggests the cytoskeleton may underpin the localization of IRS-1 to the HSP. In the present study we have taken a systematic approach to examine whether the cytoskeleton contributes to the subcellular fractionation properties and function of IRS-1. By standard microscopy or immunoprecipitation we were unable to detect evidence to support a specific interaction between IRS-1 and the major cytoskeletal components actin (microfilaments), vimentin (intermediate filaments), and tubulin (microtubules) in 3T3-L1 adipocytes or in CHO.IR.IRS-1 cells. Pharmacological disruption of microfilaments and microtubules, individually or in combination, was without effect on the subcellular distribution of IRS-1 or insulin-stimulated tyrosine phosphorylation in either cell type. Phosphorylation of Akt was modestly reduced (20-35%) in 3T3-L1 adipocytes but not in CHO.IR.IRS-1 cells. In cells lacking intermediate filaments (Vim(-/-)) IRS-1 expression, distribution and insulin-stimulated phosphorylation appeared normal. Even after depolymerisation of microfilaments and microtubules, insulin-stimulated phosphorylation of IRS-1 and Akt were maintained in Vim-/- cells. Taken together these data indicate that the characteristic subcellular fractionation properties and function of IRS-1 are unlikely to be mediated by cytoskeletal networks and that proximal insulin signaling does not require an intact cytoskeleton. Esta pesquisa se dedica ao debate em torno da redação e composição do livro de Amós, e pretende, primeiramente, apresentar e criticar algumas hipóteses sobre as camadas literárias, que poderiam pertencer ao próprio profeta, e os acréscimos tardios. Como primeiro passo para uma abordagem do problema redacional, este estudo examina algumas contribuições de especialistas estrangeiros e de estudiosos brasileiros, e tenta fazer um julgamento crítico de suas hipóteses sobre a redação do livro de Amós. A questão do processo redacional do livro de Amós tem sido, na verdade, objeto de uma animada discussão entre muitos estudiosos. Em segundo lugar, a leitura diacrônica é complementada com o estudo sincrônico do livro, isto é, com o debate em torno da estrutura não só da composição em Amós 6,1-14, mas também do livro todo. Em terceiro lugar, a partir da avaliação do resultado da pesquisa e com a utilização do método histórico-crítico, a composição em Amós 6,1-14 é analisada como uma unidade literária de sentido. Para tal análise são, indubitavelmente, relevantes algumas considerações prévias sobre o gênero literário da lamentação, a denúncia profética da injustiça social e a instituição do banquete do marzeah. Em seqüência, esta tese analisa o fenômeno literário dos panfletos proféticos e da organização social, que poderia estar por detrás de sua autoria. Por isso, e levando em conta o desafio de novas propostas metodológicas, esta pesquisa visa dar uma contribuição específica ao estudo de Amós. Ela se empenha em demonstrar que, em relação a Amós 6,1-14, o leitor está diante de uma criação coletiva. De fato, a mencionada composição é o resultado não só da pregação oral do próprio profeta, mas também, em razão de sua forma compósita, das vozes proféticas de pessoas oprimidas, que, impulsionadas pelo Espírito, confirmaram a denúncia severa de Amós e até a completaram com suas próprias experiências e sofrimentos. Finalmente, um despretensioso excursus examina o livro de Amós em um contexto literário mais amplo, para dar uma visão geral do debate em torno do processo redacional do Livro dos Doze Profetas, particularmente a partir de sua perspectiva escatológica mediante a categoria teológica do Dia do Senhor.(AU)

O cântico de Judite 16,1-12, síntese da parte em prosa do livro, faz memória da ação do Deus de Israel em favor dos oprimidos, libertando-os do poder dos impérios opressores. No centro do poema (v. 5), situa-se a ação do Todo-poderoso por mão de fêmea. A vitória de Judite é uma ironia não só à guerra, mas também às mulheres. Por um lado, as armas utilizadas pela mulher, para matar o comandante-em-chefe beleza do rosto, perfumes, veste festiva, sandália, diadema nos cabelos , são aparentemente insignificantes diante do poderio do exército inimigo, o que representa a vitória dos fracos sobre os fortes. Por outro, numa sociedade patriarcal e androcêntrica, beleza e sedução são consideradas como armas essencialmente femininas. Assim, enquanto a narrativa diverte a audiência, ela adverte aos homens que a mulher bela é perigosa, e, por sua causa, até o general mais poderoso pode perder a cabeça. Entre os séculos 4 a.C. e 2 d.C., há muitas narrativas judaicas que ressaltam o perigo de se olhar para uma mulher bela. No contexto dos movimentos sociais de resistência do período helenista, a literatura historiográfica acentua o protagonismo dos homens, enquanto a ação da mulher como protagonista só aparece no campo da ficção, e ainda para reforçar a atuação masculina. Ler Judite 16,1-12 a partir da ótica de gênero nos desafia a compreender os mecanismos que continuam expropriando o corpo e o desejo de mulheres e homens. É um convite para entoarmos novos cânticos, pautados por relações entre iguais, numa vivência solidária e de reciprocidade.(AU)

O presente estudo consiste em uma pesquisa sobre o ingresso das crianças de seis anos no Ensino Fundamental com duração de nove anos, em quatro escolas públicas do Estado de São Paulo. Como fundamento para as análises desta política educacional, o referencial teórico adotado pauta-se em estudos sobre a legislação, a concepção de infância, os conhecimentos sobre os processos de aprendizagem da leitura e da escrita, a formação e a prática dos professores. Trata-se de uma pesquisa de cunho qualitativo, estruturada na análise das proposições dos documentos oficiais, na aplicação de um questionário (para 25 professoras) e realização de entrevistas de aprofundamento com cinco professoras dentre aquelas que responderam o questionário. O propósito da aplicação do questionário e da realização de entrevistas apresentou como objetivos: traçar o perfil e conhecer aspectos da vida profissional das professoras (sujeitos da pesquisa); analisar as ideias que norteiam suas práticas de alfabetização; analisar como foram preparadas para esta nova realidade; discutir sobre os avanços e/ou as dificuldades encontradas para desenvolverem o trabalho pedagógico com as crianças do 1º ano e analisar sobre o que acreditam que deve ser garantido para o sucesso da escolaridade destas crianças. A pesquisa revela que incluir as crianças de seis anos no ensino fundamental foi, para as entrevistadas, uma medida positiva, no entanto, a implantação ocorreu sem o preparo das escolas e dos professores. Aos docentes que assumiram as classes de 1º ano coube o desafio de, mesmo sem as condições estruturais e a devida formação, organizarem o tempo e os espaços escolares. Ficou evidente nas análises, acerca do relato das práticas das professoras entrevistadas, a preocupação dada à alfabetização e o letramento, entretanto, todas reconhecem a importância da dimensão lúdica nas atividades pedagógicas da sala de aula. Esta preocupação decorre das interpretações que fazem das determinações da Secretaria Estadual de Educação e das pressões indiretas percebidas por elas quanto ao compromisso com a alfabetização neste 1º ano da escolaridade obrigatória, entre elas, a expectativa da comunidade escolar pais e professoras dos anos seguintes do Ensino Fundamental. Por fim, os dados revelam, ainda, a necessidade de se garantir a formação dos educadores, bem como de discussões sobre o currículo das turmas de seis anos.

Objective: To investigate the relationship between maternal psychopathological symptomatology during pregnancy and at 6 and 12 months postnatally and maternal use of controlling and restrictive feeding practices at 1 year. Research Methods and Procedures: Eighty-seven women completed a measure of psychological distress during pregnancy and at 6 and 12 months postpartum, and at 12 months postnatally these women reported their usage of controlling and restrictive feeding practices and were observed feeding their infants. Results: General psychological distress, particularly anxious psychopathology, during pregnancy and at 6 and 12 months postnatally was significantly associated with maternal use of restrictive feeding practices at 1 year, even when controlling for length of breast-feeding and the infants' weights at 1 year. Contrary to expectations, depression and eating psychopathology as measured by the SCOFF eating disorder measure during pregnancy or at 6 or 12 months postnatally were not associated with the use of controlling or restrictive feeding practices at 1 year. Discussion: These findings indicate that anxious maternal psychopathology may partially explain the development of maternal use of restriction when feeding. Copyright © 2005 NAASO.

The Digital Observatory for Protected Areas (DOPA) has been developed to support the European Union’s efforts in strengthening our capacity to mobilize and use biodiversity data, information and forecasts so that they are readily accessible to policymakers, managers, experts and other users. Conceived as a set of web based services, DOPA provides a broad set of free and open source tools to assess, monitor and even forecast the state of and pressure on protected areas at local, regional and global scale. DOPA Explorer 1.0 is a web based interface available in four languages (EN, FR, ES, PT) providing simple means to explore the nearly 16,000 protected areas that are at least as large as 100 km2. Distinguishing between terrestrial, marine and mixed protected areas, DOPA Explorer 1.0 can help end users to identify those with most unique ecosystems and species, and assess the pressures they are exposed to because of human development. Recognized by the UN Convention on Biological Diversity (CBD) as a reference information system, DOPA Explorer is based on the best global data sets available and provides means to rank protected areas at the country and ecoregion levels. Inversely, DOPA Explorer indirectly highlights the protected areas for which information is incomplete. We finally invite the end-users of DOPA to engage with us through the proposed communication platforms to help improve our work to support the safeguarding of biodiversity.

Full text: The title of the book gives us a major clue on the innovative approach developed by Anne Freadman in her analysis of a particular Colette corpus, the one devoted to auto-biographical writing: Les Vrilles de la vigne, Mes apprentissages, La Maison de Claudine, Sido ,L’E ́toile Vesper and Le Fanal bleu. Freadman follows the powerful lure of Rimbaldianvieilles vieilleries and its echoes with Colette’s fondness for collecting objects, people and memories. To this must be added a technical aspect, that of the study of the genre of Colette’s writing. Freadman argues that, by largely avoiding the autobiographical form, the writer achieves a new way of ‘telling time’, collecting anecdotes and detail taken from the quotidian and setting them within an all-encompassing preoccupation with time. This provides the second part of the title.The sonata form directs the sequence of the book, orchestrated into five parts,from ‘exposition’ to ‘first subject’ to‘bridge’ to ‘second subject’ to ‘recapitulation’. This has the advantage of enabling Freadman to move and progress between distinct themes—autobiography first,then alternative forms—with grace,whilst preserving within her own writing what she sees as the essence of Colette’s relationship to time in her ‘Livres-Souvenirs’, the telling of time. This‘telling of time’ is itself therefore cleverly subjected to the time constraints and freedoms of musical composition. Freadman’s ‘Exposition’ takes us through a discussion of the autobiographical genre, analysing the texts against anumber of theorists, from Lejeune to Benjamin and Ricoeur, before launching into ‘Colette and Autobiography’. It argues pertinently that Colette did not write a ‘sustained’ autobiography, even inthe most autobiographical of her writings, Mes apprentissages. Measured against Goodwin’s three sources for autobiography, confession, apologia and memoirs, Colette’s autobiographical writings appear to be at odds with all of them. Freadman then goes on in Part II of her argument, to persuasively uncover a project that rejects self-scrutiny and with no autobiographical strategy. In ‘Collecting Time’, despite claims of continuity, narrative logic and causality areabandoned in favour of a collection offragments, family stories that are built up generation after generation into familylegends. A close and fruitful analysis of Sidoleads us to a study of ‘The Art of Ending’, concentrating on L’E ́toile Vesperandle Fanal Bleu. The closing chapter gives a fascinating reading of La Naissance du jouras an exemplar of the way in which the two subjects developed in Freadman’s volume are cast together:Colette’s own working through the autobiographical genre, and her refusal to write memoirs, in favour of collecting memories, and the strategies she uses for her purpose. In ‘Recapitulation’, her concluding chapter, Freadman adroitlyen capsulates her analysis in a fetching title: ‘Fables of Time’. Indeed, the wholepremise of her book is to move away from autobiographical genre, having acknowledged the links and debt the corpus owes to it, and into a study of the multiple and fruitful ways in which Colette tells time.The rich and varied readings of thematerial, competently informed by theoretical input, together with acute sensitivity to the corpus, mark out this study as incontournable for Colette scholars.

Presently monoethanolamine (MEA) remains the industrial standard solvent for CO2 capture processes. Operating issues relating to corrosion and degradation of MEA at high temperatures and concentrations, and in the presence of oxygen, in a traditional PCC process, have introduced the requisite for higher quality and costly stainless steels in the construction of capture equipment and the use of oxygen scavengers and corrosion inhibitors. While capture processes employing MEA have improved significantly in recent times there is a continued attraction towards alternative solvents systems which offer even more improvements. This movement includes aqueous amine blends which are gaining momentum as new generation solvents for CO2 capture processes. Given the exhaustive array of amines available to date endless opportunities exist to tune and tailor a solvent to deliver specific performance and physical properties in line with a desired capture process. The current work is focussed on the rationalisation of CO2 absorption behaviour in a series of aqueous amine blends incorporating monoethanolamine, N,N-dimethylethanolamine (DMEA), N,N-diethylethanolamine (DEEA) and 2-amino-2-methyl-1-propanol (AMP) as solvent components. Mass transfer/kinetic measurements have been performed using a wetted wall column (WWC) contactor at 40°C for a series of blends in which the blend properties including amine concentration, blend ratio, and CO2 loadings from 0.0-0.4 (moles CO2/total moles amine) were systematically varied and assessed. Equilibrium CO2 solubility in each of the blends has been estimated using a software tool developed in Matlab for the prediction of vapour liquid equilibrium using a combination of the known chemical equilibrium reactions and constants for the individual amine components which have been combined into a blend.From the CO2 mass transfer data the largest absorption rates were observed in blends containing 3M MEA/3M Am2 while the selection of the Am2 component had only a marginal impact on mass transfer rates. Overall, CO2 mass transfer in the fastest blends containing 3M MEA/3M Am2 was found to be only slightly lower than a 5M MEA solution at similar temperatures and CO2 loadings. In terms of equilibrium behaviour a slight decrease in the absorption capacity (moles CO2/mole amine) with increasing Am2 concentration in the blends with MEA was observed while cyclic capacity followed the opposite trend. Significant increases in cyclic capacity (26-111%) were observed in all blends when compared to MEA solutions at similar temperatures and total amine concentrations. In view of the reasonable compromise between CO2 absorption rate and capacity a blend containing 3M MEA and 3M AMP as blend components would represent a reasonable alternative in replacement of 5M MEA as a standalone solvent.

A két részből álló cikk a magyarországi reformfolyamat és posztszocialista átalakulás négy olyan jellegzetességét emeli ki, amelyek megszakítás nélkül, a politikai szférában bekövetkezett rendszerváltást követően is, folyamatosan jellemezték az elmúlt harminc év fejlődését: a lakosság anyagi jólétének prioritása, erős paternalista jóléti állam, a reformfolyamat és átmenet fokozatossága és a politikai nyugalom. Az 1995. március 12én bejelentett stabilizációs program - amennyiben megvalósul - jelenthet elmozdulást ezektől a jellegzetességektől. A cikk politikai gazdaságtani megközelítésben vizsgálja, hogy miért alakult ki ez a négy jellegzetesség, miképpen hatottak azok egymásra, milyen kedvező és kedvezőtlen hatásokat fejtettek ki. Az első rész az 1956os forradalom hatásából kiindulva politikatörténeti áttekintést ad, majd a mai jólét, biztonság és nyugalom prioritását és a társadalmi adósság felhalmozódását, végül a stabilizációs program gazdasági és politikai hatásait elemzi. / === / The article, consisting of two parts, emphasizes four characteristic features of the Hungarian reform process and the postsocialist transformation, which, uninterruptedly, characterized the development over the last thirty years, even after the systemic change in the political sphere. These were: priority of the material welfare of the population, a strong, paternalistic welfare state, the gradual progress of the reform process and the transition, as well as political calmness. The stabilization programme, announced on March 12,1995, may imply - if it materializes - a shift away from these characteristics. The article investigates, from the aspect of political economy, why the four characteristic features had developed, how they affected each other and what were their advantageous and disadvantageous impacts. Setting out from the impacts of the 1956 revolution, the first part gives an overview of political history and then analyses the priority of today's welfare, security and calm as well as the accumulation of societal debt and, finally, the economic and political impacts of the stabilization programme.

In the years 2002, 2003 and 2004 we collected samples of macroinvertebrates on a total of 36 occasions in Badacsony bay, in areas of open water (in the years 2003 and 2004 reed-grassy) as well as populated by reed (Phragmites australis) and cattail (Typha angustifolia). Samples were taken using a stiff hand net. The sampling site includes three microhabitats differentiated only by the aquatic plants inhabiting these areas. Our data was gathered from processing 208 individual samples. The quantity of macroinvertebrates is represented by biovolume value based on volume estimates. We can identify taxa in abundant numbers found in all water types and ooze; as well as groups associated with individual microhabitats with various aquatic plants. We can observe a notable difference between the years in the volume of invertebrate macrofauna caused by the drop of water level, and the multiplication of submerged macrophytes. There are smaller differences between the samples taken in reeds and cattail stands. In the second half of 2003 – which was a year of drought – the Najas marina appeared in open waters and allowed to support larger quantities of macroinvertebrates. In 2004 with higher water levels, the Potamogeton perfoliatus occurring in the same area has had an even more significant effect. This type of reed-grass may support the most macroinvertebrates during the summer. From the aspect of diversity relations we may suspect different characteristics. The reeds sampling site proved to be the richest, while the cattail microhabitat is close behind, open water (with submerged macrophytes) is the least diverse microhabitat.
