Pro gradu -tutkielmani tavoitteena on analysoida ympäristön merkityksiä yhdysvaltalaiskirjailija Don DeLillon (1936-) romaanissa White Noise (1985). Lähestyn romaania ekokriittisen kirjallisuudentutkimuksen näkökulmasta ja kytken sen ekokriitikko Lawrence Buellin ajatukseen nk. ympäristöalitajunnasta. Analysoin DeLillon romaania myös yhteydessä filosofi Jean Baudrillardin ajatukseen postmodernin ajan länsimaisessa ja erityisesti amerikkalaisessa yhteiskunnassa vallalla olevasta simulaatioiden järjestelmästä. White Noise -romaanin todellisuus vastaa Baudrillardin ajatusta yhteiskunnasta, jossa representaatiot ja simulaatiot ovat korvanneet todellisuuden. Media, erityisesti televisio, tuottaa jatkuvasti kuvia ja simulaatioita, joiden kyllästämässä todellisuudessa aineellinen maailma ja luonto jäävät tavoittamattomiin. White Noise -romaanin henkilöiden yhteys aineelliseen ympäristöönsä ja luonnonilmiöihin on katkennut, sillä heidän arkensa pyörii pitkälti kuluttamisen ja televisionkatselun ympärillä. Romaanin todellisuudessa myös identiteetistä on tullut eräänlainen tuote, jonka jokainen voi rakentaa mieleisekseen kulutusvalinnoillaan. Identiteettiproblematiikan ohella myös kuolemalla on keskeinen asema tutkielmassani. White Noise -romaanin päähenkilö Jack Gladney kärsii paniikinomaisesta kuolemanpelosta, jota pyrkii torjumaan erilaisin keinoin siinä kuitenkaan onnistumatta. Tavoitteenani on osoittaa, että tämä piinaava pelko kuolemaa kohtaan on syntynyt simulaatioyhteiskunnan tuloksena. Vieraantuminen aineellisesta maailmasta ja luonnon prosesseista on johtanut vieraantumiseen ruumiista ja kuolemasta. Analysoin kuolemaa romaanissa eräänlaisena simulaatioiden maailman äärirajana, viimeisenä luonnollisena tapahtumana. White Noise -romaanin päähenkilö Jack Gladney ahdistuu kulutuskeskeisessä, simulaatioiden kyllästämässä elinympäristössään. Tulkitsen tämän ahdistuksen tarpeena tunnistaa tärkeä vuorovaikutussuhde yksilön ja hänen aineellisen ympäristönsä välillä. Jack ei ole vielä täysin sulautunut osaksi simulaatioiden maailmaa, vaan hän tiedostaa kytköksen itsensä ja aineellisen maailman välillä. Tämä romaanista implisiittisesti esiin nouseva tiedostamisen tunne korostaa ihmisen ja ympäristön sekä laajemmin kulttuurin ja luonnon välttämätöntä yhteyttä. DeLillon romaanista on löydettävissä ajatus ympäristöalitajunnasta, joka alleviivaa ympäristön ja luonnon merkitystä ihmiselle.

RESUMO - (Tibouchina sect. Pleroma (D. Don) Cogn. (Melastomataceae) no estado de São Paulo). Tibouchina Aubl. (Melastomeae), com ca. de 308 espécies, ocupa posição central entre as Melastomataceae neotropicais de fruto capsular. Muitas destas espécies estão especialmente concentradas nas regiões sudeste e centro-oeste do Brasil. O gênero Tibouchina encontra-se dividido em 11 seções. Este trabalho teve como objetivo estudar as espécie de Tibouchina sect. Pleroma no estado de São Paulo. São relacionadas as seguintes espécies: T. chamissoana Cogn., T. clavata (Pers.) Wurdack, T. estrellensis (Raddi) Cogn., T. grandifolia Cogn., T. granulosa (Desr.) Cogn., T. langsdorffiana (Bonpl.) Baill., T. martialis (Cham.) Cogn., T. riedeliana Cogn., T. serrana P. Guimarães & A. B. Martins, T. stenocarpa (Schrank et Mart. ex DC.) Cogn., T. ursina (Cham.) Cogn. e T. urvilleana (DC.) Cogn. Estas espécies estão distribuidas principalmente na região litorânea do estado. Foi elaborada chave de identificação e são apresentadas descrições, comentários, relação do material examinado e ilustrações.

The acid mining drainage is considered the most significant environmental pollution problem around the world for the extensive formation acidic leachates containing heavy metals. Adsorption is widely used methods in water treatment due to it easy operation and the availability of a wide variety of commercial adsorbent (low cost). The primary goal of this thesis was to investigate the efficiency of neutralizing agents, CaCO3 and CaSiO3, and metal adsorption materials with unmodified limestone from Company Nordkalk Oy. In addition to this, the side materials of limestone mining were tested for iron adsorption from acidic model solution. This study was executed at Lappeenranta University of Technology, Finland. The work utilised fixed-bed adsorption column as the main equipment and large fluidized column. Atomic absorption spectroscopy (AAS) and x-ray diffraction (XRD) was used to determine ferric removal and the composition of material respectively. The results suggest a high potential for the studied materials to be used a low cost adsorbents in acid mine drainage treatment. From the two studied adsorbents, the FS material was more suitable than the Gotland material. Based on the findings, it is recommended that further studies might include detailed analysis of Gotland materials.

Jacaranda copaia (Aubl.) D. Don is a pioneer tree widespread in the Brazilian Amazon, usually found colonizing forest gaps and altered areas, and the forest fragment edges. This study investigated aspects of the floral biology, breeding system and pollinators of J. copaia trees. Flowering lasts from August to November, during the low rainfall period extending up to four weeks per tree and 3-4 months for the population as a whole, characterizing a cornucopia flowering pattern. The fruit set ends in the beginning of the rainy season, with wind dispersed winged seeds. Fruit set from open pollination was 1.06% (n = 6,932). Hand pollination using self-pollen (n = 2,099) did not set fruits. Cross-pollination resulted in 6.54% fruit set (n = 2,524), representing six times more than the natural pollination rate (1.06%, n = 6,932). Flowers excluded from insect visitation (automatic self-pollination) did not set fruits (n = 5,372). Pollen tube growth down to ovary was detected under fluorescence microcoscopy in cross-pollinated and selfed pistils. The species is an obligate allogamous plant, with late-acting self-incompatibility system. Approximately 40 species of native bees visited the flowers, but the main pollinators were medium-sized solitary bees as Euglossa and Centris species due to the compatibility between their body sizes with the corolla tube, direct contact with the reproductive structures and high frequency of visits.

Dietary calcium lowers the risk of nephrolithiasis due to a decreased absorption of dietary oxalate that is bound by intestinal calcium. The aim of the present study was to evaluate oxaluria in normocalciuric and hypercalciuric lithiasic patients under different calcium intake. Fifty patients (26 females and 24 males, 41 ± 10 years old), whose 4-day dietary records revealed a regular low calcium intake (<=500 mg/day), received an oral calcium load (1 g/day) for 7 days. A 24-h urine was obtained before and after load and according to the calciuria under both diets, patients were considered as normocalciuric (NC, N = 15), diet-dependent hypercalciuric (DDHC, N = 9) or diet-independent hypercalciuric (DIHC, N = 26). On regular diet, mean oxaluria was 30 ± 14 mg/24 h for all patients. The 7-day calcium load induced a significant decrease in mean oxaluria compared to the regular diet in NC and DIHC (20 ± 12 vs 26 ± 7 and 27 ± 18 vs 32 ± 15 mg/24 h, respectively, P<0.05) but not in DDHC patients (22 ± 10 vs 23 ± 5 mg/24 h). The lack of an oxalate decrease among DDHC patients after the calcium load might have been due to higher calcium absorption under higher calcium supply, with a consequent lower amount of calcium left in the intestine to bind with oxalate. These data suggest that a long-lasting regular calcium consumption <500 mg was not associated with high oxaluria and that a subpopulation of hypercalciuric patients who presented a higher intestinal calcium absorption (DDHC) tended to hyperabsorb oxalate as well, so that oxaluria did not change under different calcium intake.

This work aims to evaluate deoxynivalenol degradation by Aspergillus oryzae and Rhizopus oryzae in a submerged fermentation system and to correlate it to the activity of oxydo-reductase enzymes. The submerged medium consisted of sterile distilled water contaminated with 50 μg of DON and 4 × 10(6) spore.mL-1 inoculum of Aspergillus oryzae and Rhizopus oryzae species, respectively in each experiment. Sampling was performed every 24 hours for monitoring the peroxidase specific activity, and every 48 hours for determining mycotoxin levels. Results showed that the fungi species were able to decrease DON levels as the peroxidase activity increased. The 48 hours fermentation interval presented the highest peroxidase specific activity (ΔABS/minute.μg.protein-1), 800 and 198, while the highest DON degradation velocity was 10.8 and 12.4 ppb/hour, respectively in both cases for Rhizopus oryzae and Aspergillus oryzae.

A técnica "Random Amplified Polymorphic DNA" (RAPD) surgiu como uma ferramenta útil para testar a pureza genética e a discriminação de cultivares em muitas espécies. É uma técnica simples, rápida, relativamente de baixo custo e permite o uso de DNA extraído de sementes secas, o que é muito importante em um programa de análise de sementes. O uso desta tecnologia no teste da pureza genética pode ser muito interessante para algumas espécies, como a vinca (Catharanthus roseus (L.) G.Don), pois pouco se conhece a respeito da seqüência de seu DNA. É interessante, também, pelo fato de existir grande número de "primers" comercialmente disponíveis, que podem ser prontamente utilizados para gerar dados. Essa técnica pode ser mais facilmente utilizada para gerar padrões de bandas polimórficas suficientes para discriminar genótipos diferentes. Todavia, no presente estudo, os padrões RAPD de bandas obtidas de amostras de DNA extraído de sementes de vinca em "bulk" foram não-consistentes, o mesmo ocorrendo com o uso de DNA extraído de sementes individuais de um mesmo cultivar, o que evidencia que a técnica não é aplicável para testar a pureza genética e a discriminação de cultivares de vinca. Entretanto, os padrões de bandas RAPD gerados a partir de DNA extraído de tecido foliar de plântulas foram mais reproduzíveis e poderiam ser considerados na caracterização de cultivares.

Sequence-Characterized Amplified Region (SCAR) appears as a useful technique for genetic purity testing and variety discrimination, applicable to species in which some other techniques have failed. In particular, this technique is very attractive with species in which RAPD results were not consistent. The RAPD polymorphic bands were cloned, sequenced and from the sequence information, primers pairs for normal PCR were developed. Since the probability of obtaining successful SCAR primers from RAPD polymorphic bands was about 50%, a larger number of RAPD polymorphic bands are needed to develop sufficient SCAR primers for varietal discrimination in vinca. In addition, the efficiency of the SCAR technique is strongly affected by the quality of DNA extracted from seeds. The SCAR banding patterns obtained from vinca seed were consistent and repeatable making the results reliable for genetic purity testing and variety discrimination. The SCAR technique is simple, fast, relatively inexpensive and allows the use of DNA extracted from dry seeds, which is very important in a seed-quality evaluating program
