A resistência sistêmica adquirida (SAR = systemic acquired resistance) é um importante mecanismo de resistência a doenças em plantas. Neste estudo, a ação do acibenzolar-S-metil (ASM), derivado benzotidiazólico ativador de resistência em plantas foi avaliada sobre a brotação de tubérculos de batata (Solanum tuberosum) e quanto à ação deste na indução de resistência à canela-preta, incitada por Pectobacterium carotovorum subsp. atrosepticum atípica (Pcaa), nas cultivares Asterix, Baronesa e Monalisa. Nas doses, 60, 120, 150, 200 e 250 mg i.a. l -1, o produto não inibiu o número de brotos. Contudo, em concentrações mais elevadas influenciou o comprimento destes. Em casa de vegetação, nas concentrações de 60 e 120 mg i.a. l -1 ASM, tanto no tratamento de tubérculos quanto no de aspersão nas plantas, a cultivar Asterix, respondeu ao tratamento do ASM, conferindo-lhe resistência à canela preta. Na cultivar Baronesa, a resposta ao ASM ocorreu somente no tratamento de tubérculos, e, para a cultivar Monalisa, não houve resposta ao ASM. Verifica-se, neste estudo, que houve ação do ASM sobre a indução de resistência e que este foi específico para determinadas cultivares de batata.

The physico-chemical properties of the new 3d-4f heteronuclear complexes with general formula LnCu3(C11H8N2 O4Br)3·13H2O (where Ln = Pr, Eu, Gd, Tb, Er, Yb and H3(C11H8N2 O4Br) - 5-bromosalicylideneglycylglycine) were studied. The compounds were characterized by elemental, spectral and thermal analyses and magnetic measurements. The formation of Schiff base is evidenced by a strong band at ca. 1646-1650 cm-1 attributable to C=N stretching mode. The presence of water molecules is confirmed by broad absorptions with maximum at 3360 - 3368 cm-1. The Cu(II)-Ln(III) complexes are stable up to ca. 318 K. During dehydration process the water molecules are lost probably in two stages. The magnetic susceptibility data for these complexes change with temperature according to the Curie-Weiss law.
Assessment of hydrochemical quality of ground water under some urban areas within sana'a secreteriat

Groundwater from nine wells of three different districts, located at Sana'a secretariat was analyzed for hydrochemical quality assessment. Measurements of water quality parameters including pH, EC, CO3(2-), HCO3-, Cl-, NO3-, SO4(2-), Ca2+, Mg2+, Fe3+, K+, and Na+ were carried out . Classification of the groundwater samples according to Cl, SO4(2-), CO3(2-) and HCO3-, hardness (H), total dissolved solids (TDS), base-exchange, and meteoric genesis was demonstrated. Suitability of ground water samples for irrigation and industrial uses according to sodium adsorption ration (SAR), ratio of dissolved sodium (RDS), residual sodium carbonate (RSC) and saturation index (SI) was also investigated. The results of this study showed that almost all ground water samples were of good quality that makes them suitable for drinking and domestic uses. Results also indicated that even though some of the ground water samples were suitable for irrigation purposes, almost all of them were found not be good for industrial uses. Despite all drawbacks of the sewerage system built around Sana'a secretariat at the beginning of the first decade of the third millennium, the results of this study indicate that there is scope of significant improvement in Sana'a secretariat ground water quality.

In this dissertation, active galactic nuclei (AGN) are discussed, as they are seen with the high-resolution radio-astronomical technique called Very Long Baseline Interferometry (VLBI). This observational technique provides very high angular resolution (_ 10−300 = 1 milliarcsecond). VLBI observations, performed at different radio frequencies (multi-frequency VLBI), allow to penetrate deep into the core of an AGN to reveal an otherwise obscured inner part of the jet and the vicinity of the AGN’s central engine. Multi-frequency VLBI data are used to scrutinize the structure and evolution of the jet, as well as the distribution of the polarized emission. These data can help to derive the properties of the plasma and the magnetic field, and to provide constraints to the jet composition and the parameters of emission mechanisms. Also VLBI data can be used for testing the possible physical processes in the jet by comparing observational results with results of numerical simulations. The work presented in this thesis contributes to different aspects of AGN physics studies, as well as to the methodology of VLBI data reduction. In particular, Paper I reports evidence of optical and radio emission of AGN coming from the same region in the inner jet. This result was obtained via simultaneous observations of linear polarization in the optical and in radio using VLBI technique of a sample of AGN. Papers II and III describe, in detail, the jet kinematics of the blazar 0716+714, based on multi-frequency data, and reveal a peculiar kinematic pattern: plasma in the inner jet appears to move substantially faster that that in the large-scale jet. This peculiarity is explained by the jet bending, in Paper III. Also, Paper III presents a test of the new imaging technique for VLBI data, the Generalized Maximum Entropy Method (GMEM), with the observed (not simulated) data and compares its results with the conventional imaging. Papers IV and V report the results of observations of the circularly polarized (CP) emission in AGN at small spatial scales. In particular, Paper IV presents values of the core CP for 41 AGN at 15, 22 and 43 GHz, obtained with the help of the standard Gain transfer (GT) method, which was previously developed by D. Homan and J.Wardle for the calibration of multi-source VLBI observations. This method was developed for long multi-source observations, when many AGN are observed in a single VLBI run. In contrast, in Paper V, an attempt is made to apply the GT method to single-source VLBI observations. In such observations, the object list would include only a few sources: a target source and two or three calibrators, and it lasts much shorter than the multi-source experiment. For the CP calibration of a single-source observation, it is necessary to have a source with zero or known CP as one of the calibrators. If the archival observations included such a source to the list of calibrators, the GT could also be used for the archival data, increasing a list of known AGN with the CP at small spatial scale. Paper V contains also calculation of contributions of different sourced of errors to the uncertainty of the final result, and presents the first results for the blazar 0716+714.
Distribución espacial de la rugosidad en parcelas agrícolas en Provincia de Buenos Aires - Argentina

O uso de imagens SAR para estimar e monitorar a umidade superficial do solo requer que se considere outros fatores que influenciam na retrodifusão do sinal-radar, entre os quais a rugosidade da cobertura da superfície à escala de centímetro é muito importante. Há diversos métodos para determinar a rugosidade, mas muitos são caros ou de operação de campo complexa. Neste trabalho, é apresentado um método versátil e econômico que usa máquina fotográfica e tela quadrada. Cada fotografia é processada numericamente obtendo a altura RMS, como parâmetro da rugosidade da cobertura. Por meio de técnicas geoestatísticas de krigagem é estimada a distribuição espacial da rugosidade. São mostradas experiências em áreas com cobertura de trigo, localizadas na área agrícola serrana da Província o Buenos Aires, Argentina. Os valores de RMS encontrados (29 mm < RMS < 48 mm) foram analisados com quatro critérios de rugosidade. É expressa sua utilidade para estimar o estado hídrico superficial de solos em áreas agrícolas mediante sua aplicação como entrada (input) nos modelos de retrodispersão de imagens SAR.

Este trabalho apresenta o Modelo de Regressão Espacial Autorregressivo Misto (SAR) e Modelo do Erro Espacial (CAR) no intuito de investigar a associação entre a produtividade da soja e as variáveis agrometeorológicas relacionadas à precipitação pluvial, temperatura média e radiação solar global. O estudo foi realizado com os dados das safras dos anos agrícolas de 2005/2006 a 2007/2008, da região oeste do estado do Paraná. Como os dados agrometeorológicos estão disponíveis apenas para oito municípios da região em estudo, as estimativas foram obtidas por meio do uso de Polígonos de Thiessen. A estimativa de parâmetros dos modelos ajustados foi obtida utilizando o método de Máxima Verossimilhança. A avaliação do desempenho dos modelos foi realizada com base no coeficiente de determinação (R²), no máximo valor do logaritmo da função verossimilhança e no critério de informação bayesiano de Schwarz (BIC). Este estudo também permitiu verificar a correlação e autocorrelação espacial entre a produtividade da soja e os elementos agrometeorológicos, por meio da análise espacial de área, usando de técnicas como o índice I de Moran Global e Local uni e bivariado, e os testes de significância. O estudo pôde demonstrar que, por meio dos indicadores de desempenho utilizados, os modelos SAR e CAR ofereceram melhores resultados em relação ao modelo de regressão múltipla clássica.

Some models have been developed using agrometeorological and remote sensing data to estimate agriculture production. However, it is expected that the use of SAR images can improve their performance. The main objective of this study was to estimate the sugarcane production using a multiple linear regression model which considers agronomic data and ALOS/PALSAR images obtained from 2007/08, 2008/09 and 2009/10 cropping seasons. The performance of models was evaluated by coefficient of determination, t-test, Willmott agreement index (d), random error and standard error. The model was able to explain 79%, 12% and 74% of the variation in the observed productions of the 2007/08, 2008/09 and 2009/10 cropping seasons, respectively. Performance of the model for the 2008/09 cropping season was poor because of the occurrence of a long period of drought in that season. When the three seasons were considered all together, the model explained 66% of the variation. Results showed that SAR-based yield prediction models can contribute and assist sugar mill technicians to improve such estimates.

Tämän tutkielman tavoitteena oli selvittää meripelastusyksiköiden arviointiprosessin vaiheet sekä, miten katselmoinnit voitaisiin ottaa käyttöön osana arviointiprosessia. Meripelastusyk-siköille ei ole tällä hetkellä määritetty tarkkoja vaatimuksia miehistön koulutuksen tai aluk-sen varustuksen suhteen. Näiden vaatimusten puute, Rajavartiolaitoksen vastuurooli meripe-lastuksen koordinoinnissa ja halu kehittää Suomen meripelastustoimintaa luovat tarpeen ar-viointijärjestelmän luomiseksi ja vaatimusten määrittelemiseksi. Määrittelemällä minimivaa-timukset meripelastusyksiköille parannetaan niiden toimintavalmiutta ja samalla kehitetään koko meripelastustoimintaa. Tutkimus oli luonteeltaan kvalitatiivinen tutkimus. Lähdeaineisto koostui kirjallisista lähteis-tä, joita täydennettiin asiantuntijoiden haastatteluilla. Sisällönanalyysin keinoin luokiteltiin aineiston sisältämät ajatuskokonaisuudet arviointiprosessin osalta. Sen pohjalta luotiin perus-teet laadulliselle käsittelylle, jossa analysoitiin arviointiprosessia meripelastustoimen kannal-ta. Muiden organisaatioiden kokemuksia kerättiin raporttien ja haastatteluiden avulla, ja niitä hyödynnettiin analysoitaessa arviointiprosessin eri vaiheita ja katselmoinnin toteutusta. Mää-rittelemällä tärkeimmät käsitteet luotiin pohja tutkielmassa käytettävälle termistölle. Käsitte-lemällä arvioinnin taustaa ja teoriaa luotiin pohja koko arviointiprosessille ja sille asetetuille vaatimuksille. Meripelastustoimen laadukkuus koostuu asetettujen tavoitteiden saavuttamisesta, toiminnan jatkuvasta kehittämisestä, parhaista käytänteistä ja tiiviistä yhteistyöstä. Meripelastusyksi-köiden vaatimusten määrittelytyö on käynnistetty Rajavartiolaitoksessa. Se on myös edelly-tys katselmointitoiminnan aloittamiselle. Onnistuneen arviointiprosessin kannalta on tärkeää, että katselmoinnin toimivuus ja tuloksellisuus arvioidaan säännöllisesti. Lisäksi katselmoidut yksiköt ja organisaatiot tulee ottaa osaksi arviointiprosessin kehitystyöstä. Arviointiprosessi tulisi käynnistää valitsemalla katselmoijiksi sopivat henkilöt. Henkilöiden valinnassa tulee painottaa heidän kokemuksensa ja ammattitaitonsa lisäksi halukkuutta ja sopivuutta katselmoijiksi. Henkilöiden tulisi osallistua Rajavartiolaitoksen esikunnan koor-dinoimaan arviointiprosessin valmisteluun. Samalla katselmoijat saadaan koulutettua ja si-toutettua paremmin osaksi arviointiprosessia ja sitä varten luotua järjestelmää. Arviointiprosessin tavoitteena on meripelastusyksiköiden suorituskyvyn kehittäminen ja yh-tenäistäminen. Selkeillä ja yhtenevillä vaatimuksilla meripelastusyksiköiden johtaminen hel-pottuu. Meripelastusjohtaja tietää, minkälaisella varustuksella ja miehistöllä meripelastusyk-sikkö on liikkeellä, kun hän määrää sen pelastustehtävään. Katselmoinnin tärkein tavoite on varmistaa, että katselmoitava meripelastusyksikkö täyttää sille asetetut minimikriteerit.

PURPOSE: The aim of this study was to evaluate serum levels of inducible nitric oxide synthase (INOS), myeloperoxidase (MPO), total antioxidant status (TAS), and total oxidative status (TOS) in women with primary ovarian insufficiency (POI) and to compare them with healthy fertile women. We also examined the possible risk factors associated with POI.METHODS: This cross-sectional case control study was conducted in Zekai Tahir Burak Women's Health Education and Research Hospital. The study population consisted of 44 women with POI (study group) and 36 healthy fertile women (control group). In all patients, serum levels of INOS, MPO, TAS, and TOS were determined. INOS and MPO levels were measured by enzyme-linked immunosorbent assay whereas colorimetric method was used for evaluating TAS and TOS levels. Age, body mass index (BMI), obstetric history, smoking status, family history, comorbidities, sonographic findings, complete blood count values, C-reactive protein and baseline hormone levels were also analyzed. Student's t-test or Mann-Whitney U test was used to compare continuous variables between the groups; categorical data were evaluated by using Pearson χ2 or Fisher exact test, when appropriate. Binary logistic regression method was used to identify risk factors for POI.RESULTS: We found significantly elevated levels of INOS (234.1±749.5 versus133.8±143.0; p=0.005), MPO (3,438.7±1,228.6 versus 2,481.9±1,230.1; p=0.001), and TOS (4.3±1.4 versus 3.6±1.4; p=0.02) in the sera of the study group when compared to the BMI-age matched control group. However, difference in serum levels of TAS were not significant between the 2 groups (1.7±0.2 versus 1.6±0.2; p=0.15). Logistic regression method demonstrated that BMI <25 kg/m2, nulliparity, family history of POI, smoking, and elevated serum levels of INOS, MPO, and TOS were independent risk factors for POI.CONCLUSION: We found an increase in INOS, MPO, and TOS in women with POI. These serum markers may be promising in early diagnosis of POI. Further large-scale studies are required to determine whether oxidative stress markers have a role in diagnosing POI.

Spectral sensitivities of visual systems are specified as the reciprocals of the intensities of light (quantum fluxes) needed at each wavelength to elicit the same criterion amplitude of responses. This review primarily considers the methods that have been developed for electrophysiological determinations of criterion amplitudes of slow-wave responses from single retinal cells. Traditional flash methods can require tedious dark adaptations and may yield erroneous spectral sensitivity curves which are not seen in such modifications as ramp methods. Linear response methods involve interferometry, while constant response methods involve manual or automatic adjustments of continuous illumination to keep response amplitudes constant during spectral scans. In DC or AC computerized constant response methods, feedback to determine intensities at each wavelength is derived from the response amplitudes themselves. Although all but traditional flash methods have greater or lesser abilities to provide on-line determinations of spectral sensitivities, computerized constant response methods are the most satisfactory due to flexibility, speed and maintenance of a constant adaptation level

We determined the effects of losartan (40 nmol) and PD 123319 (40 nmol) (both non-peptides and selective antagonists of the AT1 and AT2 angiotensin receptors, respectively), and [Sar¹, Ala8] angiotensin II (ANG II) (40 nmol) (a non-selective peptide antagonist of angiotensin receptors) injected into the paraventricular nucleus (PVN) on the water and salt appetite, diuresis and natriuresis and mean arterial pressure (MAP) induced by administration of 10 nmol of ANG II into the medial septal area (MSA) of male Holtzman rats weighing 250-300 g. The volume of drug solution injected was 0.5 µl over a period of 10-15 s. The responses were measured over a period of 120 min. ANG II alone injected into the MSA induced an increase in all the above parameters (8.1 ± 1.2, 1.8 ± 0.3, and 17.1 ± 1.0 ml, 217 ± 25 µEq/120 min, and 24 ± 4 mmHg, respectively, N = 10-12) compared with vehicle-treated rats (1.4 ± 0.2, 0.6 ± 0.1, and 9.3 ± 0.5 ml, 47 ± 5 µEq/120 min, and 4.1 ± 0.8 mmHg, respectively, N = 10-14). Pretreatment with losartan and [Sar¹, Ala8] ANG II completely abolished the water and sodium intake, and the pressor increase (0.5 ± 0.2, 1.1 ± 0.2, 0.5 ± 0.2, and 0.8 ± 0.2 ml, and 1.2 ± 3.9, 31 ± 4.6 mmHg, respectively, N = 9-12), whereas losartan blunted the urinary and sodium excretion induced by ANG II (13.9 ± 1.0 ml and 187 ± 10 µEq/120 min, respectively, N = 9). Pretreatment with PD 123319 and [Sar¹, Ala8] ANG II blocked the urinary and sodium excretion (10.7 ± 0.8, 9.8 ± 0.7 ml, and 67 ± 13 and 57 ± 17 µEq/120 min, respectively, N = 9), whereas pretreatment with PD 123319 partially blocked the water and sodium intake, and the MAP induced by ANG II administration (2.3 ± 0.3, 1.1 ± 0.1 ml, and 12 ± 3 mmHg, respectively, N = 9-10). These results suggest the angiotensinergic effect of the MSA on the AT1 and AT2 receptors of the PVN in terms of water and sodium homeostasis and MAP modulation.

Lolium multiflorum (Lm) grass pollen is the major cause of pollinosis in Southern Brazil. The objectives of this study were to investigate immunodominant components of Lm pollen allergens and the cross-reactivity of IgE with commercial grass pollen allergen extracts. Thirty-eight serum samples from patients with seasonal allergic rhinitis (SAR), 35 serum samples from patients with perennial allergic rhinitis (PAR) and 30 serum samples from non-atopic subjects were analyzed. Allergen sensitization was evaluated using skin prick test and serum IgE levels against Lm pollen extract were determined by ELISA. Inhibition ELISA and immunoblot were used to evaluate the cross-reactivity of IgE between allergens from Lm and commercial grass pollen extracts, including L. perenne (Lp), grass mix I (GI) and II (GII) extracts. IgE antibodies against Lm were detected in 100% of SAR patients and 8.6% of PAR patients. Inhibition ELISA demonstrated IgE cross-reactivity between homologous (Lm) and heterologous (Lp or GII) grass pollen extracts, but not for the GI extract. Fifteen IgE-binding Lm components were detected and immunoblot bands of 26, 28-30, and 32-35 kDa showed >90% recognition. Lm, Lp and GII extracts significantly inhibited IgE binding to the most immunodominant Lm components, particularly the 55 kDa band. The 26 kDa and 90-114 kDa bands presented the lowest amount of heterologous inhibition. We demonstrated that Lm extract contains both Lm-specific and cross-reactive IgE-binding components and therefore it is suitable for measuring quantitative IgE levels for diagnostic and therapeutic purposes in patients with pollinosis sensitized to Lm grass pollen rather than other phylogenetically related grass pollen extracts.

The a-tocopherol transfer protein (a-TTP) is responsible for the retention of the atocopherol form of vitamin E in living organisms. The detailed ligand transfer mechanism by a-TTP is still yet to be fully elucidated. To date, studies show that a-TTP transfers a-tocopherol from late endosomes in liver cells to the plasma membrane where it is repackaged into very low density lipoprotein (VLDL) and released into the circulation. Late endosomes have been shown to contain a lipid known as lysobisphosphatidic acid (LBP A) that is unique to this cellular compartment. LBPA plays a role in intracellular trafficking and controlling membrane curvature. Taking these observations into account plus the fact that certain proteins are recruited to membranes based on membrane curvature, the specific aim of this project was to examine the effect of LBP A on a-TTP binding to lipid membranes. To achieve this objective, dual polarization interferometry (DPI) and a vesicle binding assay were employed. Whilst DPI allows protein binding affinity to be measured on a flat lipid surface, the vesicle binding assay determines protein binding affinity to lipid vesicles mimicking curved membranes. DPI analysis revealed that the amount of a-TTP bound to lipid membranes is higher when LBPA is present. Using the vesicle binding assay, a similar result was seen where a greater amount of protein is bound to large unilamellar vesicles (LUV s) containing LBP A. However, the effect of LBP A was attenuated when small unilamellar vesicles (SUVs) were replaced with LUVs. The outcome of this project suggests that aTTP binding to membranes is influenced by membrane curvature, which in turn is induced by the presence of LBP A.

Arabidopsis is a model plant used to study disease resistance; Solanum tuberosum or potato is a crop species. Both plants possess inducible defense mechanisms that are deployed upon recognition of pathogen invasion. Transcriptional reprogramming is crucial to the activation of defense responses. The Pathogenesis-Related (PR) genes are activated in these defense programs. Expression of Arabidopsis PR-l and potato PR-10a serve as markers for the deployment of defense responses in these plants. PR-l expression indicates induction of systemic acquired resistance (SAR). Activation of SAR requires accumulation of salicylic acid (SA), in addition to the interaction of the non-expressor of pathogenesis-related genes I (NPRI), with the TGA transcription factors. The PR-10a is activated in response to pathogen invasion, wounding and elicitor treatment. PR-10a induction requires recruitment of the Whirly I (Whyl) activator to the promoter. This locus is also negatively regulated by the silencer element binding factor (SEBF). We established that both the PR-l and PR-10a are occupied by repressors under non-inducing conditions. TGA2 was found to be a constitutive resident and repressor of PR-l, which mediates repression by forming an oligomeric complex on the promoter. The DNA-binding activity of this oligomer required the TGA2 N-terminus (NT). Under resting conditions we determined that the PR-10a is bound by a repressosome containing SEBF and curiously the activator Pto interacting protein 4 (Pti4). In the context of this repressosome, SEBF is responsible for PR-10a binding, yet rWe also showed that PR-l and PR-10a are activated by different means. In PR-l activation the NPRI NT domain alleviates TGA2-mediated repression by interacting with the TGA2 NT. TGA2 remains at the PR-l but adopts a dimeric conformation and forms an enhanceosome with NPRl. In contrast, the PR-10a is activated by evicting the repressosome and recruiting Why! to the promoter. These results advance our understanding of the mechanisms regulating PR-l and PR-10a expression under resting and inducing conditions.
