The optimum conditions for the production of low methoxyl pectin using pectinmethylesterase (PME) from acerola (Malpighia glabra L.), immobilized in gelatin, have been established by factorial design and response surface methodology. In the case of the free enzymes the optimum conditions for activity, within ranges adequate for food processing, are low NaCl concentrations (0.10 M), relatively high temperatures (55 degreesC) and slightly basic pH values (pH = 9). The temperature and pH seem to have strong influence on the observed activity. In the immobilized enzyme, optimum NaCl concentration was 0.15 M, while the optimum pH remained at 9.0. When the (X) over bar chart is in use, samples are regularly taken from the process, and their means are plotted on the chart. In some cases, it is too expensive to obtain the X values, but not the values of a correlated variable Y. This paper presents a model for the economic design of a two-stage control chart, that is. a control chart based on both performance (X) and surrogate (Y) variables. The process is monitored by the surrogate variable until it signals an out-of-control behavior, and then a switch is made to the (X) over bar chart. The (X) over bar chart is built with central, warning. and action regions. If an X sample mean falls in the central region, the process surveillance returns to the (Y) over bar chart. Otherwise. The process remains under the (X) over bar chart's surveillance until an (X) over bar sample mean falls outside the control limits. The search for an assignable cause is undertaken when the performance variable signals an out-of-control behavior. In this way, the two variables, are used in an alternating fashion. The assumption of an exponential distribution to describe the length of time the process remains in control allows the application of the Markov chain approach for developing the cost function. This paper presents an economic design of (X) over bar control charts with variable sample sizes, variable sampling intervals, and variable control limits. The sample size n, the sampling interval h, and the control limit coefficient k vary between minimum and maximum values, tightening or relaxing the control. The control is relaxed when an (X) over bar value falls close to the target and is tightened when an (X) over bar value falls far from the target. A cost model is constructed that involves the cost of false alarms, the cost of finding and eliminating the assignable cause, the cost associated with production in an out-of-control state, and the cost of sampling and testing. The assumption of an exponential distribution to describe the length of time the process remains in control allows the application of the Markov chain approach for developing the cost function. A comprehensive study is performed to examine the economic advantages of varying the (X) over bar chart parameters.

An experiment was carried out at Sâo Manuel Experimental Farm, FCA-UNESP, Botucatu-SP, in 1994 and 1995, aiming to determine the biomass production and essential oil yield of mentrasto (Ageratum conyzoides L. - Asteraceae) in different stages of development. The statistical design used was a complete randomized block with 8 replications and 3 treatments (pre-flowering, flowering and post-flowering). The spacing was 70 cm between rows and 50 cm between plants, with 70 plants/parcel. The seedlings were transplanted 40 days after sowing. After harvesting, the fragments (roots, stems, leaves, flowers) and whole plants were washed, separated and weighted. The extractions of essential oil were done by hydrodistillation, using Clevenger apparatus, for two hours. There were statistic differences (Tukey 5%) in fresh weight of all vegetal parts w'hen harvested in different development stages. About essential oil yield, statistic differences were not verified among the treatments.

This paper deals with the joint economic design of x̄ and R charts when the occurrence times of assignable causes follow Weibull distributions with increasing failure rates. The variable quality characteristic is assumed to be normally distributed and the process is subject to two independent assignable causes (such as tool wear-out, overheating, or vibration). One cause changes the process mean and the other changes the process variance. However, the occurrence of one kind of assignable cause does not preclude the occurrence of the other. A cost model is developed and a non-uniform sampling interval scheme is adopted. A two-step search procedure is employed to determine the optimum design parameters. Finally, a sensitivity analysis of the model is conducted, and the cost savings associated with the use of non-uniform sampling intervals instead of constant sampling intervals are evaluated.

We develop an economic model for X̄ control charts having all design parameters varying in an adaptive way, that is, in real time considering current sample information. In the proposed model, each of the design parameters can assume two values as a function of the most recent process information. The cost function is derived and it provides a device for optimal selection of the design parameters. Through a numerical example one can foresee the savings that the developed model possibly provides. © 2001 Elsevier Science B.V. All rights reserved.

The investigation was conducted under greenhouse conditions at UNESP, Brazil, to evaluate the effects of the commercial substrates Plantmax and Sunshine and its mixtures with charred rice hulls, and the effects of floating and conventional seedling production systems in lettuce cultivation. The experiment was a randomized block experimental design, in 4 x 4 factorial treatment arrangement with two replications. Seedlings produced in the floating system were more precocious in relation to the conventional system. From transplant to harvest, the seedling production systems had similar mean time of production.

This investigation was conducted at UNESP, Jaboticabal campus, São Paulo, Brazil, from March 8 to June 21, 1999, to evaluate net melon yield and fruit quality, using soilless culture. 'Bônus 2' and 'Mission' cultivars were grown in a substrate of ground quartz or thick sand, in a randomized block experimental design. Leaf and stem dry matter production, leaf size and soluble solids content of the fruit, of 'Bônus 2' were superior to 'Mission'. There was a significant interaction for the evaluated characteristics of fruit dry mass production and yield. The largest accumulation of fruit dry mass and yield of the 'Bônus 2' cultivar (61,325 kg/ha) was observed when a substrate of ground quartz was used, while the yield for the 'Mission' cultivar was higher in a substrate of thick sand (42,125 kg/ha).

The aim of the work was to evaluate the performance of four cultivars of iceberg lettuce, in a greenhouse, with and without ground cover. The experimental design adopted was that of subdivided parcels, where parcels were provided with and without ground cover consisting of black polyethylene and the subparcels consisted of the cultivars. There were three repetitions per treatment and six central plants of each parcel were evaluated. Drip irrigation was utilized. The cultivars studied were: 'Laurel', 'AF-389', 'Mayara' and 'Tainá'. The seeds were planted in Styrofoam trays with 288 cells containing the substrate Plantmax Hortaliças® A. The trays were kept in a greenhouse until transplanting, 25 days after seeding. The parameters examined were: fresh weight of the aerial part per plant, longitudinal (DL) and transverse (DT) diameters of the head, height of the plant and mean number of leaves per plant. Only among the cultivars there were significant differences in transverse and longitudinal diameters of the head and height of the plant. No significant interactions were detected between ground cover and cultivars. With regard to longitudinal and transverse diameters, 'AF-839' (DL = 27.11 cm; DT = 24.53 cm) showed smaller diameters than those of 'Laurel' (DL = 31.13 cm; DT = 30.55 cm), but did not differ from the other cultivars. In regard to height of the plants, 'Laurel' (20.66 cm) had the greatest value and 'AF-839' (16.02 cm) the lowest. The cultivars did not differ with respect to mean number of leaves and fresh weight of the aerial part of the plant. It is concluded that there were no significant differences in the cultivation of the iceberg lettuce with and without ground cover. The cultivar 'Laurel' stood out with respect to longitudinal and transverse diameters and height of the plant, only differing from the 'AF-839' with regard to the diameters.

The optimal combination of the mechanical characteristics of austempered spheroidal graphitic cast steel together with modern casting techniques yielded an economically promising product. The maximum potential of the usage of these steels is related to fabrication and characterization techniques, among which, one of the most important is the cooling diagram (TTT curve). In this work, 3 heats of graphitic steels with the following nominal compositions were cast: 1.0 % C, 2.3 % Si, 0.4 % Mn, and with niobium contents of. 0.0 %, 0.5 % and 1.0 %. TTT curves were determined by dilatometric testing and test specimens of these steels were austempered. The samples were then characterized by hardness testing and optical and SEM microscopy. Tensile, impact (no notch) and wear testing were also performed. The addition of niobium produced significant alterations in the TTT diagrams. Increasing niobium content moves the pearlite transformation nose to the right and the bainitic transformation nose to the left. Tensile strength of these alloys was high, in the range of 1700 MPa and impact values were around of 45 Joules for alloy with 1 % Nb, 49 Joules for alloy with 0.5 % Nb and fracture did not occur for the alloy without the addition of Nb.

The objective of this work is to introduce and demonstrate the technical feasibility of the continuous flash fermentation for the production of butanol. The evaluation was carried out through mathematical modeling and computer simulation which is a good approach in such a process development stage. The process consists of three interconnected units, as follows: the fermentor, the cell retention system (tangential microfiltration) and the vacuum flash vessel (responsible for the continuous recovery of butanol from the broth). The efficiency of this process was experimentally validated for the ethanol fermentation, whose main results are also shown. With the proposed design the concentration of butanol in the fermentor was lowered from 11.3 to 7.8 g/l, which represented a significant reduction in the inhibitory effect. As a result, the final concentration of butanol was 28.2 g/l for a broth with 140 g/l of glucose. Solvents productivity and yield were, respectively, 11.7 g/l.h and 33.5 % for a sugar conversion of 95.6 %. Positive aspects about the flash fermentation process are the solvents productivity, the use of concentrated sugar solution and the final butanol concentration. The last two features can be responsible for a meaningful reduction in the distillation costs and result in environmental benefits due to lower quantities of wastewater generated by the process. © 2008 Berkeley Electronic Press. The study was performed in a greenhouse with the aim of evaluating the production of butterhead lettuce cultivars with and without plastic film mulch in the summer. The cultivars studied were 'Elisa', 'Lídia', 'Herman' and 'Luísa'. The experimental design was a split plot, in which the main plot was plastic film mulch (with or without black polyethylene film) and the subplots were the cultivars, with three repetitions. The seedlings were grown in Styrofoam trays of 288 cells and transplanted 25 days after seeding in a space of 25 × 25 cm. Harvests were carried out 45 days after transplanting and the following variables were determined: fresh and dry weights of the aerial part, number of leaves per plant, plant height and the longitudinal and transverse diameters of the plant. Significant differences were found only among the cultivars for the characteristics studied, with no cover × cultivar interaction. Fresh weight of the aerial part of 'Lídia' (270.9 g) was greater than that of 'Herman' (142.5 g), but did not differ from those of 'Luísa' (229.8 g) or 'Elisa' (204.8). The dry weight of the aerial part of 'Lídia' (12.4 g) differed statistically only with respect to that of 'Herman' (7.9 g). The cultivars 'Luísa' and 'Elisa' had an aerial part dry weight of 11.4 and 11.2 g, respectively. The number of leaves per plant of the cultivars 'Luísa' (45.1) and 'Elisa' (43.0) was higher than for 'Herman' (24.05), but did not differ from that of the cultivar 'Lídia' (39.5). The mean transverse and longitudinal diameters, as well as the mean height of the plants, showed that 'Lídia', 'Elisa' and 'Luísa' had the greatest values. The cultivars 'Lídia', 'Elisa' and 'Luísa' were the most productive and plastic film mulch had no effect on lettuce production under protected cultivation.

The present study sought biotensoactive production from soybean oil fry waste using Pseudomonas aeruginosa ATCC 10145 and Pseudomonas aeruginosa isolated from the soil of a petroleum station having undergone gasoline and diesel oil spills. The results of the experiments were analyzed using a complete factorial experimental design, investigating the concentration of soybean oil waste, ammonia sulfate and residual brewery yeast. Assays were performed in 250-mL Erlenmeyer beakers containing 50 mL of production medium, maintained on a rotary shaker at 200 rpm and a temperature of 30±1 °C for a 48-hour fermentation period. Biosurfactant production was monitored through the determination of rhamnose, surface tension and emulsification activity. The Pseudomonas aeruginosa ATCC 10145 strain and isolated Pseudomonas aeruginosa were able to reduce the surface tension of the initial mexlium from 61 mN/m to 32.5 mN/m and 30.0 mN/m as well as produce rhamnose at concentrations of 1.96 and 2.89 g/L with emulsification indices of 96% and 100%, respectively.
