While the world is focused on controlling the spread of diseases such as HIV and malaria in the developing world, another approaching epidemic has been largely overlooked. The World Heath Organization predicts that there will be 16 million new cancer cases per year in 2020 and 70% of these will be in the developing world. Many of these cancers are preventable, or treatable when detected early enough. Establishing effective, affordable and workable cancer control plans in African countries is one step in the right direction toward limiting this epidemic.

Organised multilayers were formed from the controlled self-assembly of ferrocene alkyl thiols on Au(111) surfaces. The control was accomplished by increasing the concentration of the thiol solutions used for the assembly. Cyclic voltammetry, ellipsometry, scanning probe microscopy (STM and AFM) and in situ FTIR spectroscopy were used to probe the differences between mono- and multilayers of the same compounds. Electrochemical desorption studies confirmed that the multilayer structure is attached to the surface via one monolayer. The electrochemical behaviour of the multilayers indicated the presence of more than one controlling factor during the oxidation step, whereas the reduction was kinetically controlled which contrasts with the behaviour of monolayers, which exhibit kinetic control for the oxidation and reduction steps. Conventional and imaging ellipsometry confirmed that multilayers with well-defined increments in thickness could be produced. However, STM indicated that at the monolayer stage, the thiols used promote the mobility of Au atoms on the surface. It is very likely that the multilayer structure is held together through hydrogen bonding. To the best of out knowledge, this is the first example of a controlled one-step growth of multilayers of ferrocenyl alkyl thiols using self-assembly techniques.

An in situ method was developed to produce an Ni alloy composite coating reinforced by in situ reacted TiC particles with a gradient distribution, using one-step laser cladding with a pre-placed powder mixture on a 5CrMnMo steel substrate. Dispersed and ultra-fine TIC particles were formed in situ in the coating. Most. of the TiC particles, with a marked gradient distribution, were uniformly distributed within interdendritic regions because of the trapping effect of the advancing solid-liquid interface. In addition, the TiC-gamma-Ni interfaces generated in situ were found to be free from any deleterious surface reaction. Finally, the microhardness also showed a gradient variation, with the highest value of 1250 Hv0.2 and the wear properties of the coating were significantly enhanced.

Cylindrical cellular detonation is numerically investigated by solving two-dimensional reactive Euler equations with a finite volume method on a two-dimensional self-adaptive unstructured mesh. The one-step reversible chemical reaction model is applied to simplify the control parameters of chemical reaction. Numerical results demonstrate the evolution of cellular cell splitting of cylindrical cellular detonation explored in experimentas. Split of cellular structures shows different features in the near-field and far-field from the initiation zone. Variation of the local curvature is a key factor in the behavior of cell split of cylindrical cellular detonation in propagation. Numerical results show that split of cellular structures comes from the self-organization of transverse waves corresponding to the development of small disturbances along the detonation front related to detonation instability.

Arrhenius law implicates that only those molecules which possess the internal energy greater than the activation energy E-a can react. However, the internal energy will not be proportional to the gas temperature if the specific heat ratio gamma and the gas constant R vary during chemical reaction processes. The varying gamma may affect significantly the chemical reaction rate calculated with the Arrhenius law under the constant gamma assumption, which has been widely accepted in detonation and combustion simulations for many years. In this paper, the roles of variable gamma and R in Arrhenius law applications are reconsidered, and their effects on the chemical reaction rate are demonstrated by simulating one-dimensional C-J and two-dimensional cellular detonations. A new overall one-step detonation model with variable gamma and R is proposed to improve the Arrhenius law. Numerical experiments demonstrate that this improved Arrhenius law works well in predicting detonation phenomena with the numerical results being in good agreement with experimental data.

Thermal cracking of China No.3 aviation kerosene was studied experimentally and analytically under supercritical conditions relevant to regenerative cooling system for Mach-6 scramjet applications. A two-stage heated tube system with cracked products collection/analysis was used and it can achieve a fuel temperature range of 700-1100 K, a pressure range of 3.5-4.5 MPa and a residence time of approximately 0.5-1.3 s. Compositions of the cracked gaseous products and mass flow rate of the kerosene flow at varied temperatures and pressures were obtained experimentally. A one-step lumped model was developed with the cracked mixtures grouped into three categories: unreacted kerosene, gaseous products and residuals including liquid products and carbon deposits. Based on the model, fuel conversion on the mass basis, the reaction rate and the residence time were estimated as functions of temperature. Meanwhile, a sonic nozzle was used for the control of the mass flow rate of the cracked kerosene, and correlation of the mass flow rate gives a good agreement with the measurements.

Viruses possess very specific methods of targeting and entering cells. These methods would be extremely useful if they could also be applied to drug delivery, but little is known about the molecular mechanisms of the viral entry process. In order to gain further insight into mechanisms of viral entry, chemical and spectroscopic studies in two systems were conducted, examining hydrophobic protein-lipid interactions during Sendai virus membrane fusion, and the kinetics of bacteriophage λ DNA injection.

Sendai virus glycoprotein interactions with target membranes during the early stages of fusion were examined using time-resolved hydrophobic photoaffinity labeling with the lipid-soluble carbene generator3-(trifluoromethyl)-3-(m-^(125 )I] iodophenyl)diazirine (TID). The probe was incorporated in target membranes prior to virus addition and photolysis. During Sendai virus fusion with liposomes composed of cardiolipin (CL) or phosphatidylserine (PS), the viral fusion (F) protein is preferentially labeled at early time points, supporting the hypothesis that hydrophobic interaction of the fusion peptide at the N-terminus of the F_1 subunit with the target membrane is an initiating event in fusion. Correlation of the hydrophobic interactions with independently monitored fusion kinetics further supports this conclusion. Separation of proteins after labeling shows that the F_1 subunit, containing the putative hydrophobic fusion sequence, is exclusively labeled, and that the F_2 subunit does not participate in fusion. Labeling shows temperature and pH dependence consistent with a need for protein conformational mobility and fusion at neutral pH. Higher amounts of labeling during fusion with CL vesicles than during virus-PS vesicle fusion reflects membrane packing regulation of peptide insertion into target membranes. Labeling of the viral hemagglutinin/neuraminidase (HN) at low pH indicates that HN-mediated fusion is triggered by hydrophobic interactions, after titration of acidic amino acids. HN labeling under nonfusogenic conditions reveals that viral binding may involve hydrophobic as well as electrostatic interactions. Controls for diffusional labeling exclude a major contribution from this source. Labeling during reconstituted Sendai virus envelope-liposome fusion shows that functional reconstitution involves protein retention of the ability to undergo hydrophobic interactions.

Examination of Sendai virus fusion with erythrocyte membranes indicates that hydrophobic interactions also trigger fusion between biological membranes, and that HN binding may involve hydrophobic interactions as well. Labeling of the erythrocyte membranes revealed close membrane association of spectrin, which may play a role in regulating membrane fusion. The data show that hydrophobic fusion protein interaction with both artificial and biological membranes is a triggering event in fusion. Correlation of these results with earlier studies of membrane hydration and fusion kinetics provides a more detailed view of the mechanism of fusion.

The kinetics of DNA injection by bacteriophage λ. into liposomes bearing reconstituted receptors were measured using fluorescence spectroscopy. LamB, the bacteriophage receptor, was extracted from bacteria and reconstituted into liposomes by detergent removal dialysis. The DNA binding fluorophore ethidium bromide was encapsulated in the liposomes during dialysis. Enhanced fluorescence of ethidium bromide upon binding to injected DNA was monitored, and showed that injection is a rapid, one-step process. The bimolecular rate law, determined by the method of initial rates, revealed that injection occurs several times faster than indicated by earlier studies employing indirect assays.

It is hoped that these studies will increase the understanding of the mechanisms of virus entry into cells, and to facilitate the development of virus-mimetic drug delivery strategies.

O cultivo de microalgas é uma matéria prima para produção de biocombustível e de captura de carbono devido a vantagens como alta produção de biomassa e rápido crescimento quando comparado com outras fontes de energia e não necessitar de terra fértil. O presente trabalho teve como objetivo estudar métodos de concentração da biomassa. A microalga utilizada foi a Isochrysis galbana. Os cultivos tiveram duração de 20 dias e concentração inicial de 7.104 cel/mL no meio de cultivo F2/Guillard. e foram realizados em fotobioreatores de 500 mL, 3 L e 12 L. Os experimentos foram conduzidos em foto-período de 12 h claro/escuro, com temperatura de 27 a 29 C. Ao final dos cultivos, as amostras foram levadas para a sequência de processos de separação. Inicialmente, foram realizados ensaios de microfiltração em membrana com porosidade de 0,45 m em procedimento do tipo dead-end e constatou-se a rápida e intensa formação de camada de fouling. Acrescentou-se uma etapa de separação por floculação preliminar à microfiltração, utilizando-se Al2(SO4)3 como agente floculante. O meio coagulado foi então filtrado e microfiltrado. O estudo combinado das 3 etapas de separação possibilitou 99% de remoção de biomassa.O teor de óleo obtido foi de 22,4%. Portanto, o trabalho apresenta uma configuração de concentração da biomassa Isochrysis galbana visando o processo de produção de biocombustíveis

On the basis of signed-digit negabinary representation, parallel two-step addition and one-step subtraction can be performed for arbitrary-length negabinary operands.; The arithmetic is realized by signed logic operations and optically implemented by spatial encoding and decoding techniques. The proposed algorithm and optical system are simple, reliable, and practicable, and they have the property of parallel processing of two-dimensional data. O propósito do presente estudo foi analisar o efeito da aplicação de múltiplas camadas consecutivas de dois sistemas adesivos convencionais de dois passos na difusão resinosa e padrão de distribuição dos componentes monoméricos resinosos. Dezesseis terceiros molares humanos hígidos foram tratados com os sistemas adesivos convencionais de dois passos de acordo com as instruções dos fabricantes ou com aplicações em múltiplas camadas consecutivas. Os espécimes foram seccionados paralelamente aos túbulos dentinários e as superfícies submetidas ao polimento com lixas 600, 1200, 1800, 2000 e 4000. Os espectros Raman foram coletados ao longo de uma linha perpendicular a interface adesivo-resina em intervalos de 1 ou 2 m. As medidas de difusão da resina adesiva e distribuição dos componentes monomériccos foram avaliadas pelos picos Raman de 1113 cm-1, 1609 cm-1 e 1454 cm-1. O gradiente de desmineralização usado na determinação da região de hibridização foi avaliado pelo pico de 960 cm-1 da apatita. De acordo com os resultados obtidos, a aplicação de múltiplas camadas apresentou uma tendência de homogeneização dos componentes poliméricos, dependente da composição química da resina adesiva.

[EN] This paper reports an innovative technique for reagents storage in microfluidic devices by means of a one-step UV-photoprintable ionogel-based microarray on non-modified polymeric substrates. Although the ionogel and the ink-jet printing technology are well published, this is the first study where both are used for long-term reagent storage in lab-on-a-chip devices. This technology for reagent storage is perfectly compatible with mass production fabrication processes since pre-treatment of the device substrate is not necessary and inkjet printing allows for an efficient reagent deposition process. The functionality of this microarray is demonstrated by testing the release of biotin-647 after being stored for 1 month at room temperature. Analysis of the fluorescence of the ionogel-based microarray that contains biotin-647 demonstrated that 90% of the biotin-647 present was released from the ionogel-based microarray after pumping PBS 0.1% Tween at 37 °C. Moreover, the activity of biotin-647 after being released from the ionogel-based microarray was investigated trough the binding capability of this biotin to a microcontact printed chip surface with avidin. These findings pave the way for a novel, one-step, cheap and mass production on-chip reagents storage method applicable to other reagents such as antibodies and proteins and enzymes.

We report on an optical interference method to fabricate array microstructures on the surface of silicon wafers by means of five-beam interference of femtosecond laser pulses. Optical microscope and scanning electron microscope observations revealed microstructures with micrometer-order were fabricated. The diffraction characteristics of the fabricated structures were evaluated. The present technique allows one-step realization of functional optoelectronic devices on silicon surface. We report on an optical interference method to fabricate arrayed holes on metal nickel foil and aluminum film deposited on glass substrate by means of five-beam interference of femtosecond laser pulses. Optical microscope and scanning electron microscope observations revealed that arrayed holes of micrometre-order were fabricated on both metal foil and metal film. The present technique allows one-step, large-area, micrometric processing of metal materials for potential industrial applications.

We report on an optical interference method for transferring periodic microstructures of metal film from a supporting substrate to a receiving substrate by means of five-beam interference of femtosecond laser pulses. Scanning electron microscopy and optical microscopy revealed microstructures with micrometer-order were transferred to the receiving substrate. In the meanwhile, a negative copy of the transferred structures was induced in the metal film on the supporting substrate. The diffraction characteristics of the transferred structures were also evaluated. The present technique allows one-step realization of functional optoelectronic devices. Nos dias atuais, a maioria das operações feitas por empresas e organizações é armazenada em bancos de dados que podem ser explorados por pesquisadores com o objetivo de se obter informações úteis para auxílio da tomada de decisão. Devido ao grande volume envolvido, a extração e análise dos dados não é uma tarefa simples. O processo geral de conversão de dados brutos em informações úteis chama-se Descoberta de Conhecimento em Bancos de Dados (KDD - Knowledge Discovery in Databases). Uma das etapas deste processo é a Mineração de Dados (Data Mining), que consiste na aplicação de algoritmos e técnicas estatísticas para explorar informações contidas implicitamente em grandes bancos de dados. Muitas áreas utilizam o processo KDD para facilitar o reconhecimento de padrões ou modelos em suas bases de informações. Este trabalho apresenta uma aplicação prática do processo KDD utilizando a base de dados de alunos do 9 ano do ensino básico do Estado do Rio de Janeiro, disponibilizada no site do INEP, com o objetivo de descobrir padrões interessantes entre o perfil socioeconômico do aluno e seu desempenho obtido em Matemática na Prova Brasil 2011. Neste trabalho, utilizando-se da ferramenta chamada Weka (Waikato Environment for Knowledge Analysis), foi aplicada a tarefa de mineração de dados conhecida como associação, onde se extraiu regras por intermédio do algoritmo Apriori. Neste estudo foi possível descobrir, por exemplo, que alunos que já foram reprovados uma vez tendem a tirar uma nota inferior na prova de matemática, assim como alunos que nunca foram reprovados tiveram um melhor desempenho. Outros fatores, como a sua pretensão futura, a escolaridade dos pais, a preferência de matemática, o grupo étnico o qual o aluno pertence, se o aluno lê sites frequentemente, também influenciam positivamente ou negativamente no aprendizado do discente. Também foi feita uma análise de acordo com a infraestrutura da escola onde o aluno estuda e com isso, pôde-se afirmar que os padrões descobertos ocorrem independentemente se estes alunos estudam em escolas que possuem infraestrutura boa ou ruim. Os resultados obtidos podem ser utilizados para traçar perfis de estudantes que tem um melhor ou um pior desempenho em matemática e para a elaboração de políticas públicas na área de educação, voltadas ao ensino fundamental.
