Background: Deficiencies in effective flukicide options and growing issues with drug resistance make current strategies for liver fluke control unsustainable, thereby promoting the need to identify and validate new control targets in Fasciola spp. parasites. Calmodulins (CaMs) are small calcium-sensing proteins with ubiquitous expression in all eukaryotic organisms and generally use fluctuations in intracellular calcium levels to modulate cell signalling events. CaMs are essential for fundamental processes including the phosphorylation of protein kinases, gene transcription, calcium transport and smooth muscle contraction. In the blood fluke Schistosoma mansoni, calmodulins have been implicated in egg hatching, miracidial transformation and larval development. Previously, CaMs have been identified amongst liver fluke excretory-secretory products and three CaM-like proteins have been characterised biochemically from adult Fasciola hepatica, although their functions remain unknown.

Methods: In this study, we set out to investigate the biological function and control target potential of F. hepatica CaMs (FhCaMs) using RNAi methodology alongside novel in vitro bioassays.

Results: Our results reveal that: (i) FhCaMs are widely expressed in parenchymal cells throughout the forebody region of juvenile fluke; (ii) significant transcriptional knockdown of FhCaM1-3 was inducible by exposure to either long (~200 nt) double stranded (ds) RNAs or 27 nt short interfering (si) RNAs, although siRNAs were less effective than long dsRNAs; (iii) transient long dsRNA exposure-induced RNA interference (RNAi) of FhCaMs triggered transcript knockdown that persisted for ≥ 21 days, and led to detectable suppression of FhCaM proteins; (iv) FhCaM RNAi significantly reduced the growth of juvenile flukes maintained in vitro; (v) FhCaM RNAi juveniles also displayed hyperactivity encompassing significantly increased migration; (vi) both the reduced growth and increased motility phenotypes were recapitulated in juvenile fluke using the CaM inhibitor trifluoperazine hydrochloride, supporting phenotype specificity.

Conclusions: These data indicate that the Ca(2+)-modulating functions of FhCaMs are important for juvenile fluke growth and movement and provide the first functional genomics-based example of a growth-defect resulting from gene silencing in liver fluke. Whilst the phenotypic impacts of FhCaM silencing on fluke behaviour do not strongly support their candidature as new flukicide targets, the growth impacts encourage further consideration, especially in light of the speed of juvenile fluke growth in vivo.

PURPOSE: Few studies have examined the impact of long-term treatments or exposures on the development of cataract in maturity-onset animal models. We studied the effect of treatment with D-pantethine and exposure to ultraviolet-B (UVB) radiation on the development of lenticular opacity in the Emory mouse. METHODS: A total of 164 Emory mice were randomized by litter at weaning to exposure to UVB light at 12 mJ/cm(2) for 6 hr/day (UV) or usual room light (A), and within litter, were further randomized to bi-weekly intra-peritoneal injections of 0.8 g/kg pantethine (T) or no treatment (C). Retro illumination lens photos were taken at 2, 4, 6, 8, and 10 months after weaning, and graded in masked fashion. The animals were sacrificed at 10 months and the lenses analyzed for total pantethine and total cysteamine. RESULTS: Lens pantethine and cysteamine levels were significantly (P < 0.001) higher for the T as compared to C litters. Mean cataract grade increased monotonically over time for all four groups. Unadjusted mean grade for the AT group at 8 (1.32) and 10 (1.86) months appeared lower than for the other groups (AC: 2.17, 2.39; UVC: 1.77, 2.40; UVT: 1.88, 2.37). However, the mean grade for the pantethine-treated litters did not differ significantly from the untreated litters except at 2 months (when untreated litters had significantly lower grades), when adjusting for UV treatment, gender and litter effect. No significant difference in cataract score existed between UV-exposed and ambient litters. Mortality was higher among pantethine-treated (hazard ratio = 1.8, p = 0.05) and UV-exposed animals (hazard ratio = 1.8, p = 0. 03) than among the untreated and unexposed litters. CONCLUSION: Significantly increased lens levels of pantethine are achieved with long-term intra-peritoneal dosing. The impact of pantethine on the progression of lenticular opacity in the Emory mouse is less than has been reported in other models. This level of chronic UVB exposure appeared to have no effect on the development of cataract in this model.

Chloride-induced corrosion of steel in reinforced concrete structures is one of the main problems affecting their durability, but most previous research projects and case studies have focused on concretes without cracks or not subjected to any structural load. Although it has been recognised that structural cracks do influence the chloride transport and chloride induced corrosion in reinforced concrete structures, there is little published work on the influence of micro-cracks due to service loads on these properties. Therefore the effect of micro-cracks caused by loading on chloride transport into concrete was studied. Four different stress levels (0%, 25%, 50% and 75% of the stress at ultimate load – fu) were applied to 100 mm diameter concrete discs and chloride migration was measured using a bespoke test setup based on the NT BUILD 492 test. The effects of replacing Portland cement CEMI by ground granulated blast-furnace slag (GGBS), pulverised fuel ash (PFA) and silica fume (SF) on chloride transport in concrete under sustained loading were studied. The results have indicated that chloride migration coefficients changed little when the stress level was below 50% of the fu; however, it is desirable to keep concrete stress less than 25% fu if this is practical. The effect of removing the load on the change of chloride migration coefficient was also studied. A recovery of around 50% of the increased chloride migration coefficient was found in the case of concretes subjected to 75% of the fu when the load was removed.

Individuals with autism spectrum disorders (ASD) are reported to allocate less spontaneous attention to voices. Here, we investigated how vocal sounds are processed in ASD adults, when those sounds are attended. Participants were asked to react as fast as possible to target stimuli (either voices or strings) while ignoring distracting stimuli. Response times (RTs) were measured. Results showed that, similar to neurotypical (NT) adults, ASD adults were faster to recognize voices compared to strings. Surprisingly, ASD adults had even shorter RTs for voices than the NT adults, suggesting a faster voice recognition process. To investigate the acoustic underpinnings of this effect, we created auditory chimeras that retained only the temporal or the spectral features of voices. For the NT group, no RT advantage was found for the chimeras compared to strings: both sets of features had to be present to observe an RT advantage. However, for the ASD group, shorter RTs were observed for both chimeras. These observations indicate that the previously observed attentional deficit to voices in ASD individuals could be due to a failure to combine acoustic features, even though such features may be well represented at a sensory level.

Cette étude de cas analyse l'intervention éducative d'un enseignant du primaire qui a mis en oeuvre un dispositif didactique et pédagogique intégrant de façon systématique l'informatique scolaire à une pédagogie du projet dans le but de mieux répondre aux besoins liés à l'apprentissage d'élèves provenant d'un milieu socioéconomiquement faible.Cette étude longitudinale (1998-2001) fait ressortir l'impact de l'agir professionnel sur la réussite scolaire. La problématique de notre recherche s'inscrit dans la foulée de la réforme en cours, dont l'une des visées est de contrer le retard scolaire au primaire et le risque d'échec et d'abandon scolaire précoce au secondaire. Quatre dimensions éclairent l'implémentation du dispositif: l'équité sociale, les changements épistémologiques, une orientation vers la socialisation et le rôle des TIC pour soutenir le développement des compétences transversales. Le cadre conceptuel s'appuie sur une perspective socioconstructiviste cohérente avec la théorie de l'anthropologie culturelle de Vygotsky (1978) pour qui l'apprentissage ne se fait pas seul, mais avec l'aide de pairs plus avancés en contexte de cognition située. La médiation de l'enseignant, par sa réflexion en action , en synchronie avec les besoins immédiats de ses élèves, établit le lien entre enseignant et apprenants, pour qu'ils puissent développer un esprit d'initiative, ainsi que la responsabilité et l'autonomie pour assumer leurs apprentissages. Une méthodologie mixte interreliant des méthodes qualitatives et quantitatives a exigé un recueil de données sur des dimensions descriptives et évaluatives. Des données de deuxième ordre, issues d'une recherche FCAR (98-NT-004), ont permis de dégager des caractéristiques spécifiques à la planification, à la gestion de classe et aux comportements de l'"enseignant-cible" en contexte de pédagogie du projet. Les résultats obtenus par les 23 élèves aux examens sommatifs de 6e année témoignent de l'efficacité de l'intervention. La présente étude pourrait avoir des retombées en tant que modèle pour d'autres chercheurs intéressés à étudier l'impact des TIC comme matériel didactique. Sur le plan de la pratique, ce type d'intervention serait un moyen efficace pour contrer le décrochage scolaire chez les élèves en mobilisant leurs capacités de s'adapter avec succès à leur environnement, malgré les facteurs de risque qui pourraient les inhiber.

As sequelas fisiopatológicas do stress oxidativo são difíceis de quantificar. Apesar dos obstáculos, a relevância médica do stress oxidativo tem vindo a ser cada vez mais reconhecida, sendo hoje em dia encarado como um componente chave de virtualmente todas as doenças. A disfunção erétil (DE) surge neste contexto como uma espécie de barómetro da função endotelial e do dano oxidativo. A quantificação de biomarcadores de stress oxidativo poderá apresentar um enorme impacto na avaliação de pacientes com DE. O rácio glutationa reduzida/oxidada (GSH/GSSG) e a nitrotirosina (3-NT) têm vindo a demonstrar relevância clínica. A consideração de polimorfismos genéticos constitui ainda uma abordagem promissora na avaliação destas relações no futuro. Um método altamente sensível de cromatografia líquida de alta performance (HPLC) foi desenvolvido para a determinação de 3-NT em plasma humano. As concentrações de 3-NT medidos em indivíduos com DE foram 6,6±2,1μM (média±S.D., n = 46). A medição da concentração plasmática de 3-NT poderá revelar-se útil como marcador de dano oxidativo dependente do óxido nítrico (NO). O nível de stress oxidativo pode também ser quantificado através da medição do decréscimo do rácio GSH/GSSG, que tem mostrado alterações numa miríade de patologias, como a DE e a diabetes mellitus. O método proposto para a quantificação do rácio GSH/GSSG em HPLC apresenta a vantagem de avaliação concomitante dos dois parâmetros em apenas uma corrida. O valor do rácio GSH/GSSG obtido a partir de sangue de indivíduos com DE foi 11,9±9,8 (média±S.D., n = 49). Os resultados estatísticos revelaram diferenças significativas (p<0,001) entre ambos a concentração plasmática de 3-NT e o rácio GSH/GSSG de sangue de indivíduos com DE e as respetivas medições em indivíduos saudáveis. Observaram-se ainda diferenças estatisticamente significativas (p≈0,027) entre o rácio GSH/GSSG do sangue de pacientes apenas com diagnóstico de DE e a medição respetiva em indivíduos com DE e comorbilidades cardiovasculares. Estes resultados enfatizam o papel do dano oxidativo na biopatologia da DE, elucidado com o auxílio destas duas metodologias, que poderão ter um amplo campo de aplicação no futuro, dado que se mostraram simples, não dispendiosas e rápidas, podendo eventualmente adequar-se a estudos de rastreio em larga escala.

O desenvolvimento deste trabalho teve como principal objetivo obter uma primeira avaliação da nova proposta de revisão do Regulamento das Características do Comportamento Térmico de Edifícios (RCCTE), relativamente à certificação energética de edifícios existentes. Inicialmente foi feito um estudo da regulamentação térmica de edifícios em Portugal relativamente ao desempenho energético de edifícios. De seguida abordou-se o caso de estudo, onde para uma fração autónoma existente se compara a aplicação do RCCTE com a nova proposta de revisão. Este estudo comparativo foi realizado através de um método detalhado e um outro simplificado, que no caso do RCCTE está relacionado com a Nota Técnica NT-SCE-01 para a certificação térmica de edifícios existentes. Após obtidos os resultados das necessidades energéticas para todas as metodologias, referidas no parágrafo anterior, foi elaborada uma proposta de melhoria e a respetiva análise económica, de acordo com o Fundo de Eficiência Energética, FEE. As medidas de melhoria preconizadas, de acordo com este fundo, foram a substituição das janelas existentes por janelas eficientes e a utilização de coletores solares térmicos para preparação de água quente sanitária, AQS. Por fim, é feita uma análise dos resultados obtidos relativos às necessidades energéticas da fração autónoma, antes e após da proposta de melhoria.

Background Erectile dysfunction (ED) is a prevalent complication of diabetes, and oxidative stress is an important feature of diabetic ED. Oxidative stress-induced damage plays a pivotal role in the development of tissue alterations. However, the deleterious effects of oxidative stress in the corpus cavernosum with the progression of diabetes remain unclear. The aim of this study was to evaluate systemic and penile oxidative stress status in the early and late stages of diabetes. Methods Male Wistar streptozotocin-diabetic rats (and age-matched controls) were examined 2 (early) and 8 weeks (late) after the induction of diabetes. Systemic oxidative stress was evaluated by urinary H2O2 and the ratio of circulating reduced/oxidized glutathione (GSH/GSSG). Penile oxidative status was assessed by H2O2 production and 3-nitrotyrosine (3-NT) formation. Cavernosal endothelial nitric oxide synthase (eNOS) was analyzed by quantitative immunohistochemistry. Dual immunofluorescence was also performed for 3-NT and α-smooth muscle actin (α-SMA) and eNOS–α-SMA. Results There was a significant increase in urinary H2O2 levels in both diabetic groups. The plasma GSH/GSSG ratio was significantly augmented in late diabetes. In cavernosal tissue, H2O2 production was significantly increased in late diabetes. Reactivity for 3-NT was located predominantly in cavernosal smooth muscle (SM) and was significantly reduced in late diabetes. Quantitative immunohistochemistry revealed a significant decrease in eNOS levels in cavernosal SM and endothelium in late diabetes. Conclusions The findings indicate that the noxious effects of oxidative stress are more prominent in late diabetes. Increased penile protein oxidative modifications and decreased eNOS expression may be responsible for structural and/or functional deregulation, contributing to the progression of diabetes-associated ED.

O género Flavivirus (Flaviviridae) inclui mais de setenta vírus com genoma a RNA de cadeia simples, muitos dos quais são importantes agentes patogénicos para o Homem e os outros animais. A maioria dos flavivírus pode ser transmitidos por carraças, mosquitos ou, aparentemente, restringir-se a vertebrados (Cook e Holmes, 2006). No entanto, um grupo de flavivírus designados “não clássicos”, não parece ter hospedeiro vertebrado conhecido. Estes últimos são comumente colocados junto à raiz de árvores filogenéticas do género Flavivirus, sendo frequentemente isolados em mosquitos, justificando a sua designação de vírus específicos de insectos (ISF, do inglês insect-specific flaviviruses) (Farfan-Ale et al., 2009). A classificação dos ISF como flavivírus tem sido suportada por semelhanças ao nível da sua organização genómica, perfil de hidropatia proteica, locais de clivagem conservados da sequência da poliproteína que codificam, e domínios enzimáticos. No entanto, são distintos em termos antigénicos, partilhando o mesmo nível de distância genética quando comparados com outros membros do género que quando comparados com outros dois outros géneros da família Flaviviridae (Cook e Holmes, 2006; Gould et al., 2003). Esta tese apresenta uma caracterização inicial, que inclui a obtenção da sequência genómica quase completa, de um novo ISF. Este vírus, com a designação proposta de OCFVPt, foi isolado de mosquitos adultos classificados como Aedes (Ochlerotatus) caspius (Pallas, 1771), os quais são encontrados em densidades elevadas nas zonas costeiras estuarinas dos distritos de Faro e Setúbal (Almeida et al., 2008). Este vírus replica rapidamente na linha celular C6/36 (derivada de Aedes albopictus), e, como esperado, não replica em células Vero. Contrariamente a outros ISF, o OCFVPt aparentemente causa efeito citopático óbvio em células C6/36, as quais, depois de infectadas, rapidamente se separam do suporte sólido da placa de crescimento, ficando pequenas e redondas. Análises por microscopia electrónica de secções finas de células C6/36 48h após infecção com OCFVPt revelaram uma hiperplasia nuclear acentuada com aumento do espaço entre as cisternas da membrana nuclear, no qual podem ainda ser encontradas vesículas de várias dimensões. O genoma do OCFVPt tem, no mínimo, 9.839 nt e codifica para uma única poliproteína com as caraterísticas normalmente associadas aos membros do género Flavivirus. As árvores filogenéticas geradas após alinhamento de sequências virais mostram que o OCFVPt forma, juntamente com HANKV (Huhtamo et al., 2012) um grupo monofilético distinto dentro da radiação dos ISF.

Cerebral metabolism is compartmentalized between neurons and glia. Although glial glycolysis is thought to largely sustain the energetic requirements of neurotransmission while oxidative metabolism takes place mainly in neurons, this hypothesis is matter of debate. The compartmentalization of cerebral metabolic fluxes can be determined by (13)C nuclear magnetic resonance (NMR) spectroscopy upon infusion of (13)C-enriched compounds, especially glucose. Rats under light α-chloralose anesthesia were infused with [1,6-(13)C]glucose and (13)C enrichment in the brain metabolites was measured by (13)C NMR spectroscopy with high sensitivity and spectral resolution at 14.1 T. This allowed determining (13)C enrichment curves of amino acid carbons with high reproducibility and to reliably estimate cerebral metabolic fluxes (mean error of 8%). We further found that TCA cycle intermediates are not required for flux determination in mathematical models of brain metabolism. Neuronal tricarboxylic acid cycle rate (V(TCA)) and neurotransmission rate (V(NT)) were 0.45 ± 0.01 and 0.11 ± 0.01 μmol/g/min, respectively. Glial V(TCA) was found to be 38 ± 3% of total cerebral oxidative metabolism, accounting for more than half of neuronal oxidative metabolism. Furthermore, glial anaplerotic pyruvate carboxylation rate (V(PC)) was 0.069 ± 0.004 μmol/g/min, i.e., 25 ± 1% of the glial TCA cycle rate. These results support a role of glial cells as active partners of neurons during synaptic transmission beyond glycolytic metabolism.
