“Advanced Watershed Science and Policy (ESSP 660)” is a graduate class taught in the Master of Science in Coastal and Watershed Science & Policy program at California State University Monterey Bay (CSUMB). In 2007, the class was taught in four 4-week modules, each focusing on a local watershed issue. This report is one outcome of one of those 4-week modules taught in the fall 2007 session. Humans infected with Bordetella pertussis, the whooping cough bacterium, show evidences of impaired host defenses. This pathogenic bacterium produces a unique adenylate cyclase toxin (ACT) which enters human phagocytes and catalyzes the unregulated formation of cAMP, hampering important bactericidal functions of these immune cells that eventually cause cell death by apoptosis and/or necrosis. Additionally, ACT permeabilizes cells through pore formation in the target cell membrane. Recently, we demonstrated that ACT is internalised into macrophages together with other membrane components, such as the integrin CD11b/CD18 (CR3), its receptor in these immune cells, and GM1. The goal of this study was to determine whether ACT uptake is restricted to receptor-bearing macrophages or on the contrary may also take place into cells devoid of receptor and gain more insights on the signalling involved. Here, we show that ACT is rapidly eliminated from the cell membrane of either CR3-positive as negative cells, though through different entry routes, which depends in part, on the target cell physiology and characteristics. ACT-induced Ca2+ influx and activation of non-receptor Tyr kinases into the target cell appear to be common master denominators in the different endocytic strategies activated by this toxin. Very importantly, we show that, upon incubation with ACT, target cells are capable of repairing the cell membrane, which suggests the mounting of an anti-toxin cell repair-response, very likely involving the toxin elimination from the cell surface.

ENGLISH: The anchoveta is the major constituent of the important bait and reduction fisheries of the Gulf of Panama. It is a short-lived species, the great majority of the catch consisting of fish in their first year of life. Fish for reduction are caught chiefly in the Isla Verde area, between Punta Chame and the entrance of the Panama Canal. In 1960 and 1961 anchovetas were tagged with metal internal tags and released in the major areas of occurrence of this species. The tags were recovered from the meal in the reduction plants with magnets. From the 53,380 fish tagged in 1960, 745 tags were returned during the 1960 season, 246 during the 1961 season, and 8 during the 1962 season. From the 113,202 tagged in 1961, 373 tags were returned during the 1961 season and 48 during the 1962 season. Complete catch statistics are available, and length-frequency and length-weight data were used to convert these from pounds to numbers of fish of each year class. The annual survival rate for the fish of the 1959 year class in the Isla Verde area was estimated to be 0.086 by the Chapman-Robson method, 0.102 by the year-class method, and 0.088 by the Jackson positive method. The first method is considered to give the best estimate. Six estimates of the population of fish of the 1959 year class in the Isla Verde area were obtained from the sample tag ratios of six experiments conducted in that area in 1960. The estimates differed due to the temporal decrease in the population, but the downward trend corresponded fairly well to what was expected from the total annual mortality rate. It was estimated that the population of 1959-year class fish was about 818 million on March 8, 1960, and about 70 million on March 8, 1961. As the population of anchovetas decreases during the season the effort increases sufficiently that the catch remains roughly constant. This is described as the "constant absolute catch" type fishery. Of the original population of fish in the Isla Verde area at the beginning of the 1960 season, about 11 per cent were caught and 81 per cent died of natural causes. Evaluation of growth and mortality data demonstrated that beginning the fishery for the youngest age group later than March 8 (the date it began in 1960) would reduce the yield per recruit, while increasing the fishing effort would greatly increase it. Further, it is believed unlikely that increases in the catch in the Isla Verde area alone would noticeably decrease the number of recruits to that area. Therefore there is no foreseeable need for regulation of the fishery. SPANISH: El principal constituyente de la importante pesquería para carnada y para reducción en el Golfo de Panamá es la anchoveta. Es una especie de vida corta cuya pesca, en su mayor parte, está constituida por peces que se encuentran en su primer año de vida. Para la industria de reducción los peces son capturados principalmente en el área de Isla Verde, entre Punta Chame y la entrada del Canal de Panamá. En 1960 y 1961 las anchovetas fueron marcadas con marcas metálicas internas y liberadas en las áreas más importantes en que se encuentra esta especie. Las marcas fueron recobradas de la harina en las plantas de reducción por medio de magnetos. De los 53,380 peces marcados en 1960, fueron devueltas 745 marcas durante la temporada pesquera de 1960, 246 durante la de 1961, y 8 durante la de 1962. De los 113,202 marcados en 1961, 373 marcas fueron devueltas durante la temporada pesquera de 1961 y 48 durante la de 1962. Se dispone de estadísticas completas de captura, y los datos de frecuencia-longitud y de longitud-peso fueron usados para convertir éstos de libras a números de peces de cada clase anual. La tasa anual de supervivencia correspondiente a la clase anual de 1959 en el área de Isla Verde estimó en 0.086 por medio del método Chapman-Robson; en 0.102 por método de la clase anual; y en 0.088 por el método positivo de Jackson. Se considera que el primer método dé la mejor estimación. Seis estimaciones de la población de peces de la clase anual 1959 en el área de Isla Verde fueron obtenidas según la proporción de marcas halladas en las muestras correspondientes a seis experimentos efectuados en aquella área en 1960. Las estimaciones variaron debido a la disminución temporal de la población, pero esta tendencia descendente correspondió bastante bien a lo que se esperaba según la tasa total de mortalidad anual. Se estimó que la población de peces de la clase anual de 1959 era de unos 818 millones el 8 de marzo de 1960, y aproximadamente de unos 70 millones el 8 de marzo de 1961. Conforme a que la población de anchovetas disminuye durante la temporada pesquera, el esfuerzo aumenta lo suficientemente como para que la pesca se mantenga más o menos constante. Este es el tipo de pesquería descrito como de "captura absoluta constante". De la población original de peces en el área de Isla Verde al comienzo de la temporada pesquera de 1960, cerca del 11 por ciento fue capturada y el 81 por ciento murió por causas naturales. La evaluación de los datos del crecimiento mortalidad demostraron que al comenzar la pesquería a explotar grupo de edad más joven en una fecha posterior al 8 de marzo (la fecha en que comenzó en 1960) se reduciría el rendimiento por recluta, mientras que al aumentar el esfuerzo de pesca lo aumentaría considerablemente. Más aún, se cree improbable que el aumento en la pesca en el área de Isla Verde de por sí disminuyera perceptiblemente el número de reclutas en esa área. Bi-weekly phytoplankton samples were collected at 0, 10, and 20 m and enumerated by the Utermöhl sedimentation technique; 14C productivity measurements at 10 m, oblique zooplankton tows, and routine hydrographic observations were also made. Northerly winds induce upwelling during December-April, followed by a rainy season; a slight resurgence in upwelling may occur during July and/or August. Annual variations in upwelling intensity and rainfall occur. During upwelling, the upper 50 m, about 30 per cent of the total volume of the Gulf of Panama, is replaced with water 5 to 10 C colder than the more stratified, turbid and nutrient impoverished watermass present during the rainy season. The mean annual runoff accompanying an average annual precipitation of 2731 mm is estimated to equal a layer of fresh water 3.2 m thick. About 10 per cent of the phytoplankton phosphate and inorganic nitrogen requirements during the rainy season are accreted. (PDF contains 260 pages.)

To improve the cod stocks in the Baltic Sea, a number of regulations have recently been established by the International Baltic Sea Fisheries Commission (IBSFC) and the European Commission. According to these, fishermen are obliged to use nets with escape windows (BACOMA nets) with a mesh size of the escape window of 120 mm until end of September 2003. These nets however, retain only fish much larger than the legal minimum landing size would al-low. Due to the present stock structure only few of such large fish are however existent. As a consequence fishermen use a legal alternative net. This is a conventional trawl with a cod-end of 130 mm diamond-shaped meshes (IBSFC-rules of 1st April 2002), to be increased to 140 mm on 1st September 2003, according to the mentioned IBSFC-rule. Due legal alterations of the net by the fishermen (e.g. use of extra stiff net material) these nets have acquired extremely low selective properties, i. e. they catch very small fish and produce great amounts of discards. Due to the increase of the minimum landing size from 35 to 38 cm for cod in the Baltic, the amount of discards has even increased since the beginning of 2003. Experiments have now been carried out with the BACOMAnet on German and Swedish commercial and research vessels since arguments were brought forward that the BACOMA net was not yet sufficiently tested on commercial vessels. The results of all experiments conducted so far, are compiled and evaluated here. As a result of the Swedish, Danish and German initiative and research the European Commission reacted upon this in June 2003 and rejected the increase of the diamond-meshed non-BACOMA net from 130 mm to 140mm in September 2003. To protect the cod stocks in the Baltic Sea more effectively the use of traditional diamond meshed cod-ends with-out escape window are prohibited in community waters without derogation, becoming effective 1st of September 2003. To enable more effective and simplified control of the bottom trawl fishery in the Baltic Sea the principle of a ”One-Net-Rule“ is enforced. This is going to be the BACOMA net, with the meshes of the escape window being 110 mm for the time being. The description of the BACOMA net as given in the IBSFC-rules no.10 (revision of the 28th session, Berlin 2002) concentrates on the cod-end and the escape window but only to a less extent on the design and mesh-composition of the remaining parts of the net, such as belly and funnel and many details. Thus, the present description is not complete and leaves, according to fishermen, ample opportunity for manipulation. An initiative has been started in Germany with joint effort from scientists and the fishery to better describe the entire net and to produce a proposal for a more comprehensive description, leaving less space for manipulation. A proposal in this direction is given here and shall be seen as a starting point for a discussion and development towards an internationally uniform net, which is agreed amongst the fishery, scientists and politicians. The Baltic Sea fishery is invited to comment on this proposal, and recommendations for further improvement and specifications are welcomed. Once the design is agreed by the Baltic Fishermen Association, it shall be proposed to the IBSFC and European Commission via the Baltic Fishermen Association.

ENGLISH: One phase of the duties of the Inter-American Tropical Tuna Commission is the gathering and interpretation of data concerning the life history of the commercially important bait species throughout the Eastern Pacific Ocean. During 1958 the Commission established a laboratory in Manta, Ecuador to study tuna. It was subsequently found that this fishery was dependent upon one species of anchovy, Anchoa naso, which was locally referred to as "colorado." During the calendar year 1959 approximately 380,000 scoops of bait were taken by the Manta tuna fleet (Schaefer, 1960), which at that time numbered about 23 vessels. Since then the fleet has increased by about 25 per cent and it is probable that the bait catch has increased also. Virtually nothing has been reported concerning the life history of this species. Hildebrand (1943) reviewed its taxonomy and reported standard lengths ranging from 32 to 135 mm. Peterson (1956) examined specimens from Central America and found them to range from 27 to 66 mm. He also indicated that the species spawned over a long period of time. The present report describes some aspects of the life history of Anchoa naso in Ecuadorian waters. The findings are based on 121 collections taken during the period March 1959 through June 1961. SPANISH: Una fase de las obligaciones de La Comisión Interamericana del Atún Tropical es la obtensión e interpretación de los datos concernientes a la historia natural de las especies de carnada comercialmente importantes en todo el Océano Pacifico Oriental. En el año de 1958 la Comisión estableció un laboratorio en Manta, Ecuador, para estudiar el atún. Se encontró subsecuentemente que esta pesquería dependía de una especie de anchoa, Anchoa naso, conocida localmente con el nombre de colorado. Durante el año calendario de 1959, la flota atunera de Manta, que en ese tiempo alcanzaba a unos 23 barcos, obtuvo aproximadamente 380,000 copas (scoops) de carnada (Schaefer, 1960). Desde entonces la flota ha aumentado en un 25 por ciento, y es probable que la captura de peces-cebo haya aumentado también. Nada se ha informado virtualmente sobre la historia natural de esta especie. Hildebrand (1943) revisó su taxonomia e informó sobre su longitud estándar, que varia entre los 32 y 135 mm. Peterson (1956) examinó especímenes de la América Central, y encontró que variaban entre los 27 y 66 mm. También indicó que la especie desova durante un largo periodo de tiempo. El presente informe describe algunos aspectos de la historia natural de la Anchoa naso en aguas ecuatorianas. The ritterazine and cephalostatin natural products have biological activities and structures that are interesting to synthetic organic chemists. These products have been found to exhibit significant cytotoxicity against P388 murine leukemia cells, and therefore have the potential to be used as anticancer drugs. The ritterazines and cephalostatins are steroidal dimers joined by a central pyrazine ring. Given that the steroid halves are unsymmetrical and highly oxygenated, there are several challenges in synthesizing these compounds in an organic laboratory.

Ritterazine B is the most potent derivative in the ritterazine family. Its biological activity is comparable to drugs that are being used to treat cancer today. For this reason, and the fact that there are no reported syntheses of ritterazine B to date, our lab set out to synthesize this natural product.

Herein, efforts toward the synthesis of the western fragment of ritterazine B are described. Two different routes are explored to access a common intermediate. An alkyne conjugate addition reaction was initially investigated due to the success of this key reaction in the synthesis of the eastern fragment. However, it has been found that a propargylation reaction has greater reactivity and yields, and has the potential to reduce the step count of the synthesis of the western fragment of ritterazine B.

Politically the Colorado river is an interstate as well as an international stream. Physically the basin divides itself distinctly into three sections. The upper section from head waters to the mouth of San Juan comprises about 40 percent of the total of the basin and affords about 87 percent of the total runoff, or an average of about 15 000 000 acre feet per annum. High mountains and cold weather are found in this section. The middle section from the mouth of San Juan to the mouth of the Williams comprises about 35 percent of the total area of the basin and supplies about 7 percent of the annual runoff. Narrow canyons and mild weather prevail in this section. The lower third of the basin is composed of mainly hot arid plains of low altitude. It comprises some 25 percent of the total area of the basin and furnishes about 6 percent of the average annual runoff.

The proposed Diamond Creek reservoir is located in the middle section and is wholly within the boundary of Arizona. The site is at the mouth of Diamond Creek and is only 16 m. from Beach Spring, a station on the Santa Fe railroad. It is solely a power project with a limited storage capacity. The dam which creats the reservoir is of the gravity type to be constructed across the river. The walls and foundation are of granite. For a dam of 290 feet in height, the back water will be about 25 m. up the river.

The power house will be placed right below the dam perpendicular to the axis of the river. It is entirely a concrete structure. The power installation would consist of eighteen 37 500 H.P. vertical, variable head turbines, directly connected to 28 000 kwa. 110 000 v. 3 phase, 60 cycle generators with necessary switching and auxiliary apparatus. Each unit is to be fed by a separate penstock wholly embedded into the masonry.

Concerning the power market, the main electric transmission lines would extend to Prescott, Phoenix, Mesa, Florence etc. The mining regions of the mountains of Arizona would be the most adequate market. The demand of power in the above named places might not be large at present. It will, from the observation of the writer, rapidly increase with the wonderful advancement of all kinds of industrial development.

All these things being comparatively feasible, there is one difficult problem: that is the silt. At the Diamond Creek dam site the average annual silt discharge is about 82 650 acre feet. The geographical conditions, however, will not permit silt deposites right in the reservoir. So this design will be made under the assumption given in Section 4.

The silt condition and the change of lower course of the Colorado are much like those of the Yellow River in China. But one thing is different. On the Colorado most of the canyon walls are of granite, while those on the Yellow are of alluvial loess: so it is very hard, if not impossible, to get a favorable dam site on the lower part. As a visitor to this country, I should like to see the full development of the Colorado: but how about THE YELLOW!
