This study reports the implementation of GMPs in a mozzarella cheese processing plant. The mozzarella cheese manufacturing unit is located in the Southwestern region of the state of Parana, Brazil, and processes 20,000 L of milk daily. The implementation of GMP took place with the creation of a multi-disciplinary team and it was carried out in four steps: diagnosis, report of the diagnosis and road map, corrective measures and follow-up of GMP implementation. The effectiveness of actions taken and GMP implementation was compared by the total percentages of non-conformities and conformities before and after implementation of GMR Microbiological indicators were also used to assess the implementation of GMP in the mozzarella cheese processing facility. Results showed that the average percentage of conformity after the implementation of GMP was significant increased to 66%, while before it was 32% (p < 0.05). The populations of aerobic microorganisms and total coliforms in equipment were significantly reduced (p < 0.05) after the implementation of GMP, as well as the populations of total coliforms in the hands of food handlers (p < 0.05). In conclusion, GMP implementation changed the overall organization of the cheese processing unity, as well as managers and food handlers' behavior and knowledge on the quality and safety of products manufactured. OBJETIVOS: Analisar a reincidência de violência infantil no Município de Curitiba - Paraná e compreender o fenômeno com base na perspectiva de gênero. MÉTODOS: Estudo de abordagem quantitativa do tipo descritivo exploratório. Dentre as 338 notificações de violência contra crianças de zero a 9 anos de idade junto à Rede de Proteção à Criança e ao Adolescente dessa cidade com última notificação em 2009 foram analisados 300 casos por serem reincidentes. RESULTADOS: A totalidade dos casos foi de violência intrafamiliar e a reincidência mais frequente foi a negligência, tendo com principal agressora a mãe. A violência sexual apareceu mais entre as meninas, com casos reincidentes no mesmo tipo ou com a negligência antecedendo. A manifestação da violência reincidente contra crianças apresentou-se sobreposta, recorrente, com tendência a agravar-se com a evolução dos casos. CONCLUSÃO: A violência contra as crianças é determinada por relações de poder determinadas pelas categorias gênero e geração, que produzem iniquidades potencializadoras da vulnerabilidade familiar em relação à violência. O olhar crítico sobre essa questão pode indicar medidas de superação no tocante à assistência e à prevenção de sua ocorrência e reincidência tanto do setor saúde como de outros setores.

Oltre un miliardo di persone non ha oggi accesso all’acqua potabile; più di due miliardi è il numero di coloro che vivono in condizioni igienico-sanitarie realmente proibitive. Sono 80 i paesi nel mondo (con il 40% della popolazione totale) in cui si riscontra difficoltà di approvvigionamento e presenza di risorse idriche che mancano dei requisiti che dovrebbero essere assicurati per la tutela della salute: quotidianamente e sistematicamente il diritto di accesso all’acqua, che nessun individuo dovrebbe vedersi negato, viene violato. Scarsità di acqua e non omogenea distribuzione sulla superficie terrestre sono fattori che concorrono alla crisi della risorsa, cui contribuiscono processsi di natura ambientale (cambiamenti climatici, desertificazione), di natura economica (le sorti dell’industria agroalimentare, la globalizzazione degli scambi, il bisogno crescente di energia), di natura sociale (migrazioni, urbanizzazione, crescita demografica, epidemie), di natura culturale (passaggio dal rurale all’urbano, dall’agricoltura di sussistenza a quella di profitto). Nell’ottica di uno sviluppo sostenibile un aumento indiscriminato dell’offerta non può costituire soluzione al continuo incremento della domanda di acqua. Si rende pertanto necessaria la definizione di politiche e strumenti di cambiamento nei modelli di consumo e nella pianificazione che consentano una riduzione degli squilibri nella distribuzione e nella gestione della risorsa a livello domestico e civile, industriale, agricolo. L’uso efficiente, e quindi sostenibile, dell’acqua è da perseguirsi secondo le modalità: • Risparmio, inteso come minore consumo di acqua all’inizio del ciclo. • Riciclo dell’acqua in circuito chiuso, inteso come riuso dell’acqua di scarico, o uso multiplo dell’acqua. Una idonea utilizzazione dipende da una idonea progettazione, che abbia come finalità: • La destinazione in via prioritaria delle fonti e delle risorse di più elevata qualità agli usi idropotabili, con una graduale sostituzione del consumo per altri usi con risorse di minore pregio. • La regolamentazione dell’uso delle acque sotterranee, mediante la limitazione del ricorso all’impiego di pozzi solo in mancanza di forniture alternative per uso civile, industriale, agricolo. • L’incentivazione ad un uso razionale della risorsa, anche mediante l’attuazione di idonee politiche tariffarie. • L’aumento dell’efficienza delle reti di adduzione e distribuzione, sia civili che irrigue. • La promozione di uso efficiente, riciclo e recupero di acqua nell’industria. • Il miglioramento dell’efficienza ed efficacia delle tecniche di irrigazione. • La promozione del riutilizzo delle acque nei vari settori. • La diffusione nella pratica domestica di apparati e tecnologie progettati per la riduzione degli sprechi e dei consumi di acqua. In ambito agricolo la necessità di un uso parsimonioso della risorsa impone il miglioramento dell’efficienza irrigua, pari appena al 40%. La regione Emilia Romagna a livello locale, Israele a livello internazionale, forniscono ottimi esempi in termini di efficacia dei sistemi di trasporto e di distribuzione, di buona manutenzione delle strutture. Possibili soluzioni verso le quali orientare la ricerca a livello mondiale per arginare la progressiva riduzione delle riserve idriche sono: • Revisione dei costi idrici. • Recupero delle riserve idriche. • Raccolta dell’acqua piovana. • Miglioramento degli impianti di distribuzione idrica. • Scelta di metodi di coltivazione idonei alle caratteristiche locali. • Scelta di colture a basso fabbisogno idrico. • Conservazione della risorsa attraverso un sistema di irrigazione efficiente. • Opere di desalinizzazione. • Trasferimento idrico su vasta scala da un’area all’altra. Si tratta di tecniche la cui attuazione può incrementare la disponibilità media pro capite di acqua, in particolare di coloro i quali non ne posseggono in quantità sufficiente per bere o sono privi di sistemi igienico-sanitari sufficienti.

Self-incompatibility (SI) systems have evolved in many flowering plants to prevent self-fertilization and thus promote outbreeding. Pear and apple, as many of the species belonging to the Rosaceae, exhibit RNase-mediated gametophytic self-incompatibility, a widespread system carried also by the Solanaceae and Plantaginaceae. Pear orchards must for this reason contain at least two different cultivars that pollenize each other; to guarantee an efficient cross-pollination, they should have overlapping flowering periods and must be genetically compatible. This compatibility is determined by the S-locus, containing at least two genes encoding for a female (pistil) and a male (pollen) determinant. The female determinant in the Rosaceae, Solanaceae and Plantaginaceae system is a stylar glycoprotein with ribonuclease activity (S-RNase), that acts as a specific cytotoxin in incompatible pollen tubes degrading cellular RNAs. Since its identification, the S-RNase gene has been intensively studied and the sequences of a large number of alleles are available in online databases. On the contrary, the male determinant has been only recently identified as a pollen-expressed protein containing a F-box motif, called S-Locus F-box (abbreviated SLF or SFB). Since F-box proteins are best known for their participation to the SCF (Skp1 - Cullin - F-box) E3 ubiquitine ligase enzymatic complex, that is involved in protein degradation through the 26S proteasome pathway, the male determinant is supposed to act mediating the ubiquitination of the S-RNases, targeting them for the degradation in compatible pollen tubes. Attempts to clone SLF/SFB genes in the Pyrinae produced no results until very recently; in apple, the use of genomic libraries allowed the detection of two F-box genes linked to each S haplotype, called SFBB (S-locus F-Box Brothers). In Japanese pear, three SFBB genes linked to each haplotype were cloned from pollen cDNA. The SFBB genes exhibit S haplotype-specific sequence divergence and pollen-specific expression; their multiplicity is a feature whose interpretation is unclear: it has been hypothesized that all of them participate in the S-specific interaction with the RNase, but it is also possible that only one of them is involved in this function. Moreover, even if the S locus male and female determinants are the only responsible for the specificity of the pollen-pistil recognition, many other factors are supposed to play a role in GSI; these are not linked to the S locus and act in a S-haplotype independent manner. They can have a function in regulating the expression of S determinants (group 1 factors), modulating their activity (group 2) or acting downstream, in the accomplishment of the reaction of acceptance or rejection of the pollen tube (group 3). This study was aimed to the elucidation of the molecular mechanism of GSI in European pear (Pyrus communis) as well as in the other Pyrinae; it was divided in two parts, the first focusing on the characterization of male determinants, and the second on factors external to the S locus. The research of S locus F-box genes was primarily aimed to the identification of such genes in European pear, for which sequence data are still not available; moreover, it allowed also to investigate about the S locus structure in the Pyrinae. The analysis was carried out on a pool of varieties of the three species Pyrus communis (European pear), Pyrus pyrifolia (Japanese pear), and Malus × domestica (apple); varieties carrying S haplotypes whose RNases are highly similar were chosen, in order to check whether or not the same level of similarity is maintained also between the male determinants. A total of 82 sequences was obtained, 47 of which represent the first S-locus F-box genes sequenced from European pear. The sequence data strongly support the hypothesis that the S locus structure is conserved among the three species, and presumably among all the Pyrinae; at least five genes have homologs in the analysed S haplotypes, but the number of F-box genes surrounding the S-RNase could be even greater. The high level of sequence divergence and the similarity between alleles linked to highly conserved RNases, suggest a shared ancestral polymorphism also for the F-box genes. The F-box genes identified in European pear were mapped on a segregating population of 91 individuals from the cross 'Abbé Fétel' × 'Max Red Bartlett'. All the genes were placed on the linkage group 17, where the S locus has been placed both in pear and apple maps, and resulted strongly associated to the S-RNase gene. The linkage with the RNase was perfect for some of the F-box genes, while for others very rare single recombination events were identified. The second part of this study was focused on the research of other genes involved in the SI response in pear; it was aimed on one side to the identification of genes differentially expressed in compatible and incompatible crosses, and on the other to the cloning and characterization of the transglutaminase (TGase) gene, whose role may be crucial in pollen rejection. For the identification of differentially expressed genes, controlled pollinations were carried out in four combinations (self pollination, incompatible, half-compatible and fully compatible cross-pollination); expression profiles were compared through cDNA-AFLP. 28 fragments displaying an expression pattern related to compatibility or incompatibility were identified, cloned and sequenced; the sequence analysis allowed to assign a putative annotation to a part of them. The identified genes are involved in very different cellular processes or in defense mechanisms, suggesting a very complex change in gene expression following the pollen/pistil recognition. The pool of genes identified with this technique offers a good basis for further study toward a better understanding of how the SI response is carried out. Among the factors involved in SI response, moreover, an important role may be played by transglutaminase (TGase), an enzyme involved both in post-translational protein modification and in protein cross-linking. The TGase activity detected in pear styles was significantly higher when pollinated in incompatible combinations than in compatible ones, suggesting a role of this enzyme in the abnormal cytoskeletal reorganization observed during pollen rejection reaction. The aim of this part of the work was thus to identify and clone the pear TGase gene; the PCR amplification of fragments of this gene was achieved using primers realized on the alignment between the Arabidopsis TGase gene sequence and several apple EST fragments; the full-length coding sequence of the pear TGase gene was then cloned from cDNA, and provided a precious tool for further study of the in vitro and in vivo action of this enzyme.

Allergies are a complex of symptoms derived from altered IgE-mediated reactions of the immune system towards substances known as allergens. Allergic sensibilization can be of food or respiratory origin and, in particular, apple and hazelnut allergens have been identified in pollens or fruits. Allergic cross-reactivity can occur in a patient reacting to similar allergens from different origins, justifying the research in both systems as in Europe a greater number of people suffers from apple fruit allergy, but little evidence exists about pollen. Apple fruit allergies are due to four different classes of allergens (Mal d 1, 2, 3, 4), whose allergenicity is related both to genotype and tissue specificity; therefore I have investigated their presence also in pollen at different time of germination to clarify the apple pollen allergenic potential. I have observed that the same four classes of allergens found in fruit are expressed at different levels also in pollen, and their presence might support that the apple pollen can be considered allergenic as the fruit, deducing that apple allergy could also be indirectly caused by sensitization to pollen. Climate changes resulting from increases in temperature and air pollution influence pollen allergenicity, responsible for the dramatic raise in respiratory allergies (hay fever, bronchial asthma, conjunctivitis). Although the link between climate change and pollen allergenicity is proven, the underlying mechanism is little understood. Transglutaminases (TGases), a class of enzymes able to post-translationally modify proteins, are activated under stress and involved in some inflammatory responses, enhancing the activity of pro-inflammatory phospholipase A2, suggesting a role in allergies. Recently, a calcium-dependent TGase activity has been identified in the pollen cell wall, raising the possibility that pollen TGase may have a role in the modification of pollen allergens reported above, thus stabilizing them against proteases. This enzyme can be involved also in the transamidation of proteins present in the human mucosa interacting with surface pollen or, finally, the enzyme itself can represent an allergen, as suggested by studies on celiac desease. I have hypothesized that this pollen enzyme can be affected by climate changes and be involved in exhacerbating allergy response. The data presented in this thesis represent a scientific basis for future development of studies devoted to verify the hypothesis set out here. First, I have demonstrated the presence of an extracellular TGase on the surface of the grain observed either at the apical or the proximal parts of the pollen-tube by laser confocal microscopy (Iorio et al., 2008), that plays an essential role in apple pollen-tube growth, as suggested by the arrest of tube elongation by TGase inhibitors, such as EGTA or R281. Its involvement in pollen tube growth is mainly confirmed by the data of activity and gene expression, because TGase showed a peak between 15 min and 30 min of germination, when this process is well established, and an optimal pH around 6.5, which is close to that recorded for the germination medium. Moreover, data show that pollen TGase can be a glycoprotein as the glycosylation profile is linked both with the activation of the enzyme and with its localization at the pollen cell wall during germination, because from the data presented seems that the active form of TGase involved in pollen tube growth and pollen-stylar interaction is more exposed and more weakly bound to the cell wall. Interestingly, TGase interacts with fibronectin (FN), a putative SAMs or psECM component, inducing possibly intracellular signal transduction during the interaction between pollen-stylar occuring in the germination process, since a protein immunorecognised by anti-FN antibody is also present in pollen, in particular at the level of pollen grain cell wall in a punctuate pattern, but also along the shank of the pollen tube wall, in a similar pattern that recalls the signal obtained with the antibody anti TGase. FN represents a good substrate for the enzyme activity, better than DMC usually used as standard substrate for animal TGase. Thus, this pollen enzyme, necessary for its germination, is exposed on the pollen surface and consequently can easily interact with mucosal proteins, as it has been found germinated pollen in studies conducted on human mucus (Forlani, personal communication). I have obtained data that TGase activity increases in a very remarkable way when pollen is exposed to stressful conditions, such as climate changes and environmental pollution. I have used two different species of pollen, an aero allergenic (hazelnut, Corylus avellana) pollen, whose allergenicity is well documented, and an enthomophylus (apple, Malus domestica) pollen, which is not yet well characterized, to compare data on their mechanism of action in response to stressors. The two pollens have been exposed to climate changes (different temperatures, relative humidity (rH), acid rain at pH 5.6 and copper pollution (3.10 µg/l)) and showed an increase in pollen surface TGase activity that is not accompanied to an induced expression of TGase immunoreactive protein with AtPNG1p. Probably, climate change induce an alteration or damage to pollen cell wall that carries the pollen grains to release their content in the medium including TGase enzyme, that can be free to carry out its function as confirmed by the immunolocalisation and by the in situ TGase activity assay data; morphological examination indicated pollen damage, viability significantly reduced and in acid rain conditions an early germination of apple pollen, thus possibly enhancing the TGase exposure on pollen surface. Several pollen proteins were post-translationally modified, as well as mammalian sPLA2 especially with Corylus pollen, which results in its activation, potentially altering pollen allergenicity and inflammation. Pollen TGase activity mimicked the behaviour of gpl TGase and AtPNG1p in the stimulation of sPLA2, even if the regulatory mechanism seems different to gpl TGase, because pollen TGase favours an intermolecular cross-linking between various molecules of sPLA2, giving rise to high-molecular protein networks normally more stable. In general, pollens exhibited a significant endogenous phospholipase activity and it has been observed differences according to the allergenic (Corylus) or not-well characterized allergenic (Malus) attitude of the pollen. However, even if with a different intensity level in activation, pollen enzyme share the ability to activate the sPLA2, thus suggesting an important regulatory role for the activation of a key enzyme of the inflammatory response, among which my interest was addressed to pollen allergy. In conclusion, from all the data presented, mainly presence of allergens, presence of an extracellular TGase, increasing in its activity following exposure to environmental pollution and PLA2 activation, I can conclude that also Malus pollen can behave as potentially allergenic. The mechanisms described here that could affect the allergenicity of pollen, maybe could be the same occurring in fruit, paving the way for future studies in the identification of hyper- and hypo- allergenic cultivars, in preventing environmental stressor effects and, possibly, in the production of transgenic plants.

The horizontal and vertical system neurons (HS and VS cells) are part of a conserved set of lobula plate giant neurons (LPGNs) in the optic lobes of the adult brain. Structure and physiology of these cells are well known, predominantly from studies in larger Dipteran flies. Our knowledge about the ontogeny of these cells is limited and stems predominantly from laser ablation studies in larvae of the house fly Musca domestica. These studies suggested that the HS and VS cells stem from a single precursor, which, at least in Musca, has not yet divided in the second larval instar. A regulatory mutation (In(1)omb[H31]) in the Drosophila gene optomotor-blind (omb) leads to the selective loss of the adult HS and VS cells. This mutation causes a transient reduction in omb expression in what appears to be the entire optic lobe anlage (OLA) late in embryogenesis. Here, I have reinitiated the laser approach with the goal of identifying the presumptive embryonic HS/VS precursor cell in Drosophila. The usefulness of the laser ablation approach which has not been applied, so far, to cells lying deep within the Drosophila embryo, was first tested on two well defined embryonic sensory structures, the olfactory antenno-maxillary complex (AMC) and the light-sensitive Bolwing´s organ (BO). In the case of the AMC, the efficiency of the ablation procedure was demonstrated with a behavioral assay. When both AMCs were ablated, the response to an attractive odour (n-butanol) was clearly reduced. Interestingly, the larvae were not completely unresponsive but had a delayed response kinetics, indicating the existence of a second odour system. BO will be a useful test system for the selectivity of laser ablation when used at higher spatial resolution. An omb-Gal4 enhancer trap line was used to visualize the embryonic OLA by GFP fluorescence. This fluorescence allowed to guide the laser beam to the relevant structure within the embryo. The success of the ablations was monitored in the adult brain via the enhancer trap insertion A122 which selectively visualizes the HS and VS cell bodies. Due to their tight clustering, individual cells could not be identified in the embryonic OLA by conventional fluorescence microscopy. Nonetheless, systematic ablation of subdomains of the OLA allowed to localize the presumptive HS/VS precursor to a small area within the OLA, encompassing around 10 cells. Future studies at higher resolution should be able to identify the precursor as (an) individual cell(s). Most known lethal omb alleles do not complement the HS/VS phenotype of the In(1)omb[H31] allele. This is the expected behaviour of null alleles. Two lethal omb alleles that had been isolated previously by non-complementation of the omb hypomorphic allele bifid, have been reported, however, to complement In(1)omb[H31]. This report was based on low resolution paraffin histology of adult heads. Four mutations from this mutagenesis were characterized here in more detail (l(1)omb[11], l(1)omb[12], l(1)omb[13], and l(1)omb[15]). Using A122 as marker for the adult HS and VS cells, I could show, that only l(1)omb[11] can partly complement the HS/VS cell phenotype of In(1)omb[H31]. In order to identify the molecular lesions in these mutants, the exons and exon/intron junctions were sequenced in PCR-amplified material from heterozygous flies. Only in two mutants could the molecular cause for loss of omb function be identified: in l(1)omb[13]), a missense mutation causes the exchange of a highly conserved residue within the DNA-binding T-domain; in l(1)omb[15]), a nonsense mutation causes a C-terminal truncation. In the other two mutants apparently regulatory regions or not yet identified alternative exons are affected. To see whether mutant OMB protein in the missense mutant l(1)omb[13] is affected in DNA binding, electrophoretic shift assays on wildtype and mutant T-domains were performed. They revealed that the mutant no longer is able to bind the consensus palindromic T-box element.

Scopo del lavoro della presente tesi è stato quello di valutare la prevalenza di Chlamydiaceae, ed in particolare Chlamydia psittaci, in una popolazione aviare con caratteristiche sinantropiche quale il colombo di città (Columba livia var. domestica) dell’areale veneziano. Per evidenziare il patogeno sono state utilizzate metodiche sierologiche, d’isolamento e biomolecolari tradizionali. Contestualmente, mediante tecnologie molecolari innovative, quali microarray ed MLVA (Multilocus VNTR Assay) sono stati valutati i genotipi di C. psittaci e le eventuali altre specie di Chlamydia presenti in tale popolazione. Inoltre, si è proceduto ad una classificazione delle lesioni anatomo-patologiche ed ad una loro correlazione con la presenza del patogeno. I risultati dimostrano che la prevalenza di C. psittaci nella popolazione oggetto dello studio è del 10%. Durante tale studio è stata dimostrata la presenza di un ceppo di C. psittaci atipico, in quanto non classificabile con le attuali tecniche a disposizione. La genotipizzazione dei ceppi di C. psittaci conferma la presenza nel colombo di città del genotipo B, E ed E/B, che solitamente risultano essere coinvolti con minore frequenza in episodi di infezione umana. Inoltre sono stati dimostrati alcuni ceppi classificati come Chlamydia spp., in quanto le metodologie applicate e le conoscenze attuali non permettono ulteriori distinzioni, prospettando la possibilità di un nuovo ceppo. Infine, attraverso l’analisi dei dati raccolti durante la prima fase di campionamenti e successivamente confermati durante la seconda fase, siamo riusciti a strutturare un sistema di selezione, basato su caratteristiche funzionali ed anatomopatologiche, che permette di selezionare in sede necroscopica i colombi molto probabilmente infetti, permettendo conseguentemente una migliore organizzazione e gestione dei campioni di interesse contenendo nel contempo i costi ma mantenendo elevati gli standards diagnostici.
Gene expression analysis in ‘Candidatus Phytoplasma mali’-resistant and -susceptible Malus genotypes

Apple proliferation (AP) disease is the most important graft-transmissible and vector-borne disease of apple in Europe. ‘Candidatus Phytoplasma mali’ (Ca. P. mali) is the causal agent of AP. Apple (Malus x domestica) and other Malus species are the only known woody hosts. In European apple orchards, the cultivars are mainly grafted on one rootstock, M. x domestica cv. M9. M9 like all other M. x domestica cultivars is susceptible to ‘Ca. P. mali’. Resistance to AP was found in the wild genotype Malus sieboldii (MS) and in MS-derived hybrids but they were characterised by poor agronomic value. The breeding of a new rootstock carrying the resistant and the agronomic traits was the major aim of a project of which this work is a part. The objective was to shed light into the unknown resistance mechanism. The plant-phytoplasma interaction was studied by analysing differences between the ‘Ca. P. mali’-resistant and -susceptible genotypes related to constitutively expressed genes or to induced genes during infection. The cDNA-Amplified Fragment Length Polymorphism (cDNA-AFLP) technique was employed in both approaches. Differences related to constitutively expressed genes were identified between two ‘Ca. P. mali’-resistant hybrid genotypes (4551 and H0909) and the ‘Ca. P. mali’-susceptible M9. 232 cDNA-AFLP bands present in the two resistant genotypes but absent in the susceptible one were isolated but several different products associated to each band were found. Therefore, two different macroarray hybridisation experiments were performed with the cDNA-AFLP fragments yielding 40 sequences encoding for genes of unknown function or a wide array of functions including plant defence. In the second approach, individuation and analysis of the induced genes was carried out exploiting an in vitro system in which healthy and ‘Ca. P. mali’-infected micropropagated plants were maintained under controlled conditions. Infection trials using in vitro grafting of ‘Ca. P. mali’ showed that the resistance phenotype could be reproduced in this system. In addition, ex vitro plants were generated as an independent control of the genes differentially expressed in the in vitro plants. The cDNA-AFLP analysis in in vitro plants yielded 63 bands characterised by over-expression in the infected state of both the H0909 and MS genotypes. The major part (37 %) of the associated sequences showed homology with products of unknown function. The other genes were involved in plant defence, energy transport/oxidative stress response, protein metabolism and cellular growth. Real-time qPCR analysis was employed to validate the differential expression of the genes individuated in the cDNA-AFLP analysis. Since no internal controls were available for the study of the gene expression in Malus, an analysis on housekeeping genes was performed. The most stably expressed genes were the elongation factor-1 α (EF1) and the eukaryotic translation initiation factor 4-A (eIF4A). Twelve out of 20 genes investigated through qPCR were significantly differentially expressed in at least one genotype either in in vitro plants or in ex vitro plants. Overall, about 20% of the genes confirmed their cDNA-AFLP expression pattern in M. sieboldii or H0909. On the contrary, 30 % of the genes showed down-regulation or were not differentially expressed. For the remaining 50 % of the genes a contrasting behaviour was observed. The qPCR data could be interpreted as follows: the phytoplasma infection unbalance photosynthetic activity and photorespiration down-regulating genes involved in photosynthesis and in the electron transfer chain. As result, and in contrast to M. x domestica genotypes, an up-regulation of genes of the general response against pathogens was found in MS. These genes involved the pathway of H2O2 and the production of secondary metabolites leading to the hypothesis that a response based on the accumulation of H2O2 in MS would be at the base of its resistance. This resembles a phenomenon known as “recovery” where the spontaneous remission of the symptoms is observed in old susceptible plants but occurring in a stochastic way while the resistance in MS is an inducible but stable feature. As additional product of this work three cDNA-AFLP-derived markers were developed which showed independent distribution among the seedlings of two breeding progenies and were associated to a genomic region characteristic of MS. These markers will contribute to the development of molecular markers for the resistance as well as to map the resistance on the Malus genome.

Die durch eine männchenspezifisch auftretende heterochromatische Bande und ein hemizygotes Cla-Element-Cluster gekennzeichnete geschlechtsbestimmende Region („sex determining region“: SDR) auf Chromosom III von C. riparius stellt ein frühes Stadium in der Evolution von Geschlechtschromosomen dar. Diese eindeutig lokalisierte chromosomale Region, die den molekular noch unbekannten männchen¬bestimmenden Faktor M enthalten muss, ist im Vergleich zu den Y-Chromosomen anderer Dipterenarten wie unter anderem M. domestica, die ebenfalls einen dominanten Männchenbestimmer besitzen, relativ klein. Aus diesem Grund bietet die SDR von C. riparius eine Möglichkeit, den männchenbestimmenden Faktor einzugrenzen und zu identifizieren. In der vorliegenden Arbeit konnte ein Bereich einer Größe von ca. 200 kb aus der SDR von C. riparius charakterisiert und analysiert werden. Durch bioinformatische Sequenzanalysen konnten an 20 Stellen der SDR mögliche Genstrukturen nachgewiesen werden. Von den gefundenen möglichen Genen ist bisher die Funktion in C. riparius unbekannt. Bei den Genen mit vermuteter Funktion deutet nichts eindeutig auf eine Beteiligung an der Geschlechtsbestimmung von C. riparius hin. Da allerdings davon auszugehen ist, dass für die Funktion des Männchenbestimmers M ein Gen rekrutiert wurde, welches zur Interaktion mit dem nachgeschalteten Gen der Geschlechts¬bestimmungskaskade fähig ist, muss die geschlechtsbestimmende Funktion des Gens M nicht unbedingt offensichtlich sein. Aus geschlechtsbestimmenden Genkaskaden anderer Dipteren bekannte Gene wie transformer und doublesex konnten im analysierten Bereich nicht nachgewiesen werden, obwohl zumindest zu doublesex homologe Gene im Genom von C. riparius vorkommen. Um möglicherweise proto-X- und proto-Y-Chromosom miteinander vergleichen zu können und einen Hinweis auf die chromosomale Herkunft der analysierten Sequenzen aus der SDR zu erlangen, wurden Sequenzen von 31 teilweise parallel liegenden BAC-Klonen aus der untersuchten Region verglichen. Dabei zeigte sich, dass die Klone zwei Gruppen bilden, deren Sequenzen sich durch 500 SNPs und 110 Indels unterschiedlicher Größe (1-800 Bp) unterscheiden, was für eine Herkunft von zwei sich erst seit kurzer Zeit unterscheidenden Geschlechtschromosomen spricht. Die zwölf größten dieser Indels wurden auf geschlechtsspezifische Unterschiede hin untersucht. Dabei zeigte sich, dass die Unterschiede zwischen den beiden Klongruppen zwar im 30 Jahre alten Laborstamm, der auch für die Konstruktion der durchsuchten BAC-Bibliotheken verwendet wurde, tatsächlich geschlechtsspezifisch sind, in zwei Wildfangpopulationen jedoch keine derartige Geschlechtsspezifität aufweisen. Somit kann keine Aussage zur Herkunft der untersuchten Klone aus der SDR von C. riparius getroffen werden, und es bleibt unklar, ob die analysierten Sequenzen vom proto-X oder vom proto-Y-Chromosom stammen.

Il ritrovamento della Casa dei due peristili a Phoinike ha aperto la strada a un’intensa opera di revisione di tutti i dati relativi all’edilizia domestica nella regione, con studi comparativi verso nord (Illiria meridionale) e verso sud (il resto dell’Epiro, ovvero Tesprozia e Molossia). Tutta quest’area della Grecia nord-occidentale è stata caratterizzata nell’antichità da un’urbanizzazione scarsa numericamente e tardiva cronologicamente (non prima del IV sec. a.C.), a parte ovviamente le colonie corinzio-corciresi di area Adriatico- Ionica d’età arcaica (come Ambracia, Apollonia, Epidamnos). A un’urbanistica di tipo razionale e programmato (ad es. Cassope, Orraon, Gitani in Tesprozia, Antigonea in Caonia) si associano numerosi casi di abitati cresciuti soprattutto in rapporto alla natura del suolo, spesso diseguale e montagnoso (ad es. Dymokastro/Elina in Tesprozia, Çuka e Aitoit nella Kestrine), talora semplici villaggi fortificati, privi di una vera fisionomia urbana. D’altro canto il concetto classico di polis così come lo impieghiamo normalmente per la Grecia centro-meridionale non ha valore qui, in uno stato di tipo federale e dominato dall’economia del pascolo e della selva. In questo contesto l’edilizia domestica assume caratteri differenti fra IV e I sec. a.C.: da un lato le città ortogonali ripetono schemi egualitari con poche eccezioni, soprattutto alle origini (IV sec. a.C., come a Cassope e forse a Gitani), dall’altro si delinea una spiccata differenziazione a partire dal III sec., quando si adottano modelli architettonici differenti, come i peristili, indizio di una più forte differenziazione sociale (esemplari i casi di Antigonea e anche di Byllis in Illiria meridionale). I centri minori e fortificati d’altura impiegano formule abitative più semplici, che sfruttano l’articolazione del terreno roccioso per realizzare abitazioni a quote differenti, utilizzando la roccia naturale anche come pareti o pavimenti dei vani.

Kolumnare Apfelbäume (Malus x domestica) stellen aufgrund ihres auffälligen Phänotyps eine ökonomisch interessante Wuchsform dar. Diese extreme Form des Kurztriebwuchses zeichnet sich durch einen insgesamt sehr schlanken, säulenförmigen Habitus aus, welcher eine dichte Pflanzung und damit einhergehend Ertragssteigerungen im Vergleich zu normalwüchsigen Bäumen ermöglicht. Verursacht wird der Phänotyp durch die Anwesenheit eines einzelnen, dominanten Allels des Columnar (Co)-Gens. Bis auf die approximative Lokalisation des Gens auf Chromosom 10 ist über mögliche Identität und Funktion bislang nichts bekannt.rnIn der vorliegenden Arbeit wurde ein erster Versuch unternommen, mit Hilfe von Next Generation Sequencing (NGS) Technologien und RNA-Seq Einblicke in das Transkriptom des Sprossapikalmeristems (SAM) kolumnarer Apfelbäume zu gewinnen. So konnte gezeigt werden, dass unabhängig vom Zeitpunkt der Entnahme des Materials mehrere hundert Gene differentiell reguliert werden. Diese lassen sich funktional in mehrere überrepräsentierte Kategorien gruppieren, von denen sich einige wiederum mit dem kolumnaren Phänotyp assoziieren lassen. Durch den Einsatz weiterer Expressionsstudien (Microarrays, qRT-PCR) konnten frühere Ergebnisse bezüglich des Hormonhaushalts auf Genebene bestätigt und neue Erkenntnisse gewonnen werden, die eine mögliche Erklärung für den Phänotyp darstellen. Weiterhin ergab der Vergleich aller durchgeführten Expressionsstudien eine Anreicherung signifikant differentiell regulierter Gene auf Chromosom 10, was auf einen „selective sweep“ hindeutet. Eine potentielle epigenetische Regulation dieser Gene durch das Genprodukt von Co könnte daher möglich sein. Mehr als die Hälfte dieser Gene lassen sich darüber hinaus aufgrund ihrer Funktion direkt mit dem kolumnaren Phänotyp assoziieren.rnDiese Ergebnisse zeigen, dass die Anwesenheit des Co-Allels massive Veränderungen in der Genregulation des SAMs mit sich bringt, wobei einige dieser differentiell regulierten Gene mit großer Wahrscheinlichkeit an der Etablierung des kolumnaren Phänotyps beteiligt sind. Auch wenn die Funktion des Co-Genproduktes nicht abschließend geklärt werden konnte, sind doch anhand der Resultate schlüssige Hypothesen diesbezüglich möglich.rn

La ricerca si propone di analizzare una di quelle stagioni architettoniche controverse e lontane dalle internazionali strade maestre del nascente Neues Bauen: il romanticismo-nazionale svedese riletto attraverso l’esperienza del suo massimo esponente, Ragnar Östberg (1866-1945). L’obiettivo della tesi non è solamente quello di una revisione della critica storiografica, facendo così luce su una di quelle personalità considerate marginali, quanto quello di ricavare dalla lettura comparata di due tra i suoi progetti, fino ad ora mai indagati, quegli elementi che fanno dell’architettura un “fatto urbano” in cui la collettività può riconoscersi e parallelamente un fatto di rappresentazione della stessa. L’arcipelago di Stoccolma e quel processo di “renovatio urbis” a cui fu sottoposta proprio agli albori del XX secolo furono gli scenari in cui presero vita i due progetti: il complesso formato dallo Stockholms Stadshuset e la vicina parte mai realizzata del Nämndhuset, e villa Geber. Condensano due dimensioni che la città immersa nel paesaggio contiene: la natura urbana dell’edificio municipale e quella domestica della villa urbana isolata. La ricerca intesse un itinerario di disvelamento attraverso una matrice duale di lettura: “genius loci” e memorie urbane. I capitoli cercano di dimostrare come i due casi-studio siano espressione di quella pendolarità di ricerca tra lo spirito del luogo e le rimembranze delle forme urbane della tradizione. Questa analisi ci conduce in un viaggio alla ricerca dell’atlante delle “memorie urbane”, raccolte nei viaggi e nella formazione, comprendendo così il mondo analogico di riferimenti culturali con altre architetture europee della tradizione. I due progetti sorgono in opposte aree di espansione di Stoccolma e, pur nella loro diversità di scala, sono chiara espressione di appropriatezza al luogo e di strutture formali analoghe. Stockholm Stadshuset-Nämndhuset e villa Geber esprimono il metodo di Östberg, dove i riferimenti raccolti dall’imagination passive sono tramutati ed assemblati grazie alla imagination active.

L’obiettivo del lavoro è stato quello di definire la filiera di smaltimento dei rifiuti di plastica di origine domestica. La modellazione della stessa ha preso come riferimento un insieme di informazioni rese disponibili dal CONAI, Consorzio Nazionale degli Imballaggi. Noti gli attori e le attività svolte al fine dell’avvio a recupero della frazione raccolta, è stato sviluppato un modello economico adeguato al fine di ripartire i costi delle attività in funzione della gestione del materiale. Tale modellazione non prende in esame le spese e gli introiti allocati al Consorzio, ma si focalizza sulle attività di raccolta, selezione e avvio a recupero. Ci si è poi concentrati sulla modellazione economica dell’avvio a recupero della frazione di plastica raccolta dal Comune di Bologna. Ripartendo la responsabilità di gestione tra il gestore del servizio (Hera), il Centro Comprensoriale (CC Akron Granarolo), i Centri di Selezione (CSS Argeco e Idealservice) e i riciclatori (tra quelli convenzionati al consorzio) si è definito il modello economico per il caso in esame. In particolare, si è adottato un approccio simulativo di allocazione del materiale selezionato in vista della modalità di vendita di alcuni prodotti tramite asta online. La quantificazione economica in sé risente di assunzioni definite per la determinazione di una componente di costo che possa essere il può possibile esaustiva delle operazioni svolte. Si è poi proceduto definendo una stima della quantità di diossido di carbonio prodotta in fase di combustione di carburante per la movimentazione dei flussi del caso di studio. I valori in esame sono stati desunti come prodotto tra la quantità di CO2 emessa per i chilometri percorsi a seconda della tipologia di mezzo adottata per la movimentazione dei flussi. I dati relativi alle quantità di anidrite carbonica prodotte sono stime in grammi di CO2 al chilometro realizzate da Arpa e distinte in funzione delle soluzioni tecnologiche da implementare secondo le disposizioni redatte dalla Comunità Europea. La nota metodologica adottata prevede di analizzare i risultati ottenuti in funzione della distanza percorsa, dei costi di trasporto e della quantità di CO2 generata per le attività.

Extracts of 644 plant materials were screened for tannins and 204 extracts gave a positive test. Quantitative evaluation of tannins in positive extracts showed that 16 contained appreciable quantities. Insecticidal activity was tested against Musco domestica and Tribolium casteneum in 162 extracts and was confirmed in 17 and 4 extracts respectively.
