Tumor-mobilized bone marrow-derived CD11b(+) myeloid cells promote tumor angiogenesis, but how and when these cells acquire proangiogenic properties is not fully elucidated. Here, we show that CD11b(+) myelomonocytic cells develop proangiogenic properties during their differentiation from CD34(+) hematopoietic progenitors and that placenta growth factor (PlGF) is critical in promoting this education. Cultures of human CD34(+) progenitors supplemented with conditioned medium from breast cancer cell lines or PlGF, but not from nontumorigenic breast epithelial lines, generate CD11b(+) cells capable of inducing endothelial cell sprouting in vitro and angiogenesis in vivo. An anti-Flt-1 mAb or soluble Flt-1 abolished the generation of proangiogenic activity during differentiation from progenitor cells. Moreover, inhibition of metalloproteinase activity, but not VEGF, during the endothelial sprouting assay blocked sprouting induced by these proangiogenic CD11b(+) myelomonocytes. In a mouse model of breast cancer, circulating CD11b(+) cells were proangiogenic in the sprouting assays. Silencing of PlGF in tumor cells prevented the generation of proangiogenic activity in circulating CD11b(+) cells, inhibited tumor blood flow, and slowed tumor growth. Peripheral blood of breast cancer patients at diagnosis, but not of healthy individuals, contained elevated levels of PlGF and circulating proangiogenic CD11b(+) myelomonocytes. Taken together, our results show that cancer cells can program proangiogenic activity in CD11b(+) myelomonocytes during differentiation of their progenitor cells in a PlGF-dependent manner. The organization of lin genes and IS6100 was studied in three strains of Sphingomonas paucimobilis (B90A, Sp+, and UT26) which degraded hexachlorocyclohexane (HCH) isomers but which had been isolated at different geographical locations. DNA-DNA hybridization data revealed that most of the lin genes in these strains were associated with IS6100, an insertion sequence classified in the IS6 family and initially found in Mycobacterium fortuitum. Eleven, six, and five copies of IS6100 were detected in B90A, Sp+, and UT26, respectively. IS6100 elements in B90A were sequenced from five, one, and one regions of the genomes of B90A, Sp+, and UT26, respectively, and were found to be identical. DNA-DNA hybridization and DNA sequencing of cosmid clones also revealed that S. paucimobilis B90A contains three and two copies of linX and linA, respectively, compared to only one copy of these genes in strains Sp+ and UT26. Although the copy number and the sequence of the remaining genes of the HCH degradative pathway (linB, linC, linD, and linE) were nearly the same in all strains, there were striking differences in the organization of the linA genes as a result of replacement of portions of DNA sequences by IS6100, which gave them a strange mosaic configuration. Spontaneous deletion of linD and linE from B90A and of linA from Sp+ occurred and was associated either with deletion of a copy of IS6100 or changes in IS6100 profiles. The evidence gathered in this study, coupled with the observation that the G+C contents of the linA genes are lower than that of the remaining DNA sequence of S. paucimobilis, strongly suggests that all these strains acquired the linA gene through horizontal gene transfer mediated by IS6100. The association of IS6100 with the rest of the lin genes further suggests that IS6100 played a role in shaping the current lin gene organization.

The purpose of this paper is to describe the development and to test the reliability of a new method called INTERMED, for health service needs assessment. The INTERMED integrates the biopsychosocial aspects of disease and the relationship between patient and health care system in a comprehensive scheme and reflects an operationalized conceptual approach to case mix or case complexity. The method is developed to enhance interdisciplinary communication between (para-) medical specialists and to provide a method to describe case complexity for clinical, scientific, and educational purposes. First, a feasibility study (N = 21 patients) was conducted which included double scoring and discussion of the results. This led to a version of the instrument on which two interrater reliability studies were performed. In study 1, the INTERMED was double scored for 14 patients admitted to an internal ward by a psychiatrist and an internist on the basis of a joint interview conducted by both. In study 2, on the basis of medical charts, two clinicians separately double scored the INTERMED in 16 patients referred to the outpatient psychiatric consultation service. Averaged over both studies, in 94.2% of all ratings there was no important difference between the raters (more than 1 point difference). As a research interview, it takes about 20 minutes; as part of the whole process of history taking it takes about 15 minutes. In both studies, improvements were suggested by the results. Analyses of study 1 revealed that on most items there was considerable agreement; some items were improved. Also, the reference point for the prognoses was changed so that it reflected both short- and long-term prognoses. Analyses of study 2 showed that in this setting, less agreement between the raters was obtained due to the fact that the raters were less experienced and the scoring procedure was more susceptible to differences. Some improvements--mainly of the anchor points--were specified which may further enhance interrater reliability. The INTERMED proves to be a reliable method for classifying patients' care needs, especially when used by experienced raters scoring by patient interview. It can be a useful tool in assessing patients' care needs, as well as the level of needed adjustment between general and mental health service delivery. The INTERMED is easily applicable in the clinical setting at low time-costs.

The association between adiposity measures and dyslipidemia has seldom been assessed in a multipopulational setting. 27 populations from Europe, Australia, New Zealand and Canada (WHO MONICA project) using health surveys conducted between 1990 and 1997 in adults aged 35-64 years (n = 40,480). Dyslipidemia was defined as the total/HDL cholesterol ratio &gt;6 (men) and &gt;5 (women). Overall prevalence of dyslipidemia was 25% in men and 23% in women. Logistic regression showed that dyslipidemia was strongly associated with body mass index (BMI) in men and with waist circumference (WC) in women, after adjusting for region, age and smoking. Among normal-weight men and women (BMI<25 kg/m(2)), an increase in the odds for being dyslipidemic was observed between lowest and highest WC quartiles (OR = 3.6, p < 0.001). Among obese men (BMI ≥ 30), the corresponding increase was smaller (OR = 1.2, p = 0.036). A similar weakening was observed among women. Classification tree analysis was performed to assign subjects into classes of risk for dyslipidemia. BMI thresholds (25.4 and 29.2 kg/m(2)) in men and WC thresholds (81.7 and 92.6 cm) in women came out at first stages. High WC (&gt;84.8 cm) in normal-weight men, menopause in women and regular smoking further defined subgroups at increased risk. standard categories of BMI and WC, or their combinations, do not lead to optimal risk stratification for dyslipidemia in middle-age adults. Sex-specific adaptations are necessary, in particular by taking into account abdominal obesity in normal-weight men, post-menopausal age in women and regular smoking in both sexes.

Summary The proinflammatory cytokine macrophage migration inhibitory factor (MIF) has emerged as a central mediator of inflammation and innate immune defense against infections. MIF has been shown to play an important role in the pathogenesis of infectious diseases like sepsis, tuberculosis and autoimmune inflammatory diseases, such as arthritis, inflammatory bowel disease and asthma. Two functional polymorphisms of the MIF gene promoter, a five to eight CATT repeat microsatellite at position -794 and a G/C SNP at position -173, have been associated with increased susceptibility to or severity of autoimmune inflammatory diseases like arthritis, colitis and atopy. The aim of this thesis was to define whether, and if so by which mechanisms, MIF gene polymorphisms influence the susceptibility to or the outcome of one of the most severe and one of the most prevalent infectious diseases: meningococcal sepsis and tuberculosis, respectively. The results of the comparison between 1106 patients suffering from severe meningococcal infections and 434 healthy volunteers showed that carriers of the CATT5-5 genotype were protected from meningococcemia. A transmission disequilibrium test involving 106 families confirmed this association. At baseline and after stimulation with Neisseria meningitidis, the CATT5 MIF promoter drove lower transcriptional activity than the CATT6 or CATT7 alleles in human monocytic cells and whole blood of CATT5-5 healthy individuals tended to produce less MIF than whole blood of CATT6-6 individuals. Beyond, we describe several new MIF gene polymorphisms in Africans. Genotyping the CATT microsatellite and the -173*G/C SNP revealed great genetic diversity in six African ethnic groups. Comparing 471 African tuberculosis cases and 932 matched healthy controls, we observed ethnicity dependent associations of the -173*G/C and the CATT5-8 with susceptibility to or severity of tuberculosis, but confirmation in larger cohorts ìs needed. In conclusion, we report that homozygous carriage of a low expression allele of the MIF gene protects from meningococcal disease. These results support the concept that analyses of MIF genotypes in patients with sepsis may help to classify patients into risk categories and to identify those patients who may benefit from anti-MIF therapeutic strategies.

The repair of wounds is one of the most complex biological processes that occur during human life. After an injury, multiple biological pathways immediately become activated and are synchronized to respond. In human adults, the wound repair process commonly leads to a non-functioning mass of fibrotic tissue known as a scar. By contrast, early in gestation, injured fetal tissues can be completely recreated, without fibrosis, in a process resembling regeneration. Some organisms, however, retain the ability to regenerate tissue throughout adult life. Knowledge gained from studying such organisms might help to unlock latent regenerative pathways in humans, which would change medical practice as much as the introduction of antibiotics did in the twentieth century.

A diabetes mellitus tipo 2 (DM2) é responsável por uma elevada morbilidade e mortalidade em todo o Mundo, essencialmente devido às suas complicações, entre as quais a retinopatia diabética (RD), considerada uma das mais graves, e responsável por 4,8% dos casos de cegueira. Estudos sugerem uma componente genética como um dos principais factores para o desenvolvimento da RD. O gene do VEGF (vascular endothelial growth factor) é um dos mais estudados, por promover a angio©nese e a neovascularização. Outro importante gene candidato é o RAGE (receptor for advanced glycation end products), e, mais recentemente, os genes da paraoxonase, PON1 e PON2. Objectivou-se avaliar a influência dos polimorfismos VEGF -634C/G, RAGE -374T/A, PON1Gln192Arg e PON2Cys310Ser no aparecimento e progressão da RD em indivíduos com DM2 e a sua influência no aparecimento da DM2. Analisaram-se 129 indivíduos, 86 com DM2 e 43 indivíduos saudáveis. Os polimorfismos foram avaliados em todos os indivíduos por PCR-FRLP. A caracterização clínica e a determinação da actividade enzimática da PON1 foram avaliadas em 47 diabéticos. Não se obtiveram diferenças para o polimorfismo do VEGF-634 G/C. O alelo A do polimorfismo RAGE -374A/T mostrou-se mais frequente em indivíduos sem RD ou EMD quando comparados com indivíduos com RD ou EMD. O alelo Q (Gln) e o alelo S (Ser) dos polimorfismos PON1Gln192Arg e PON2Cys310Ser, respectivamente, mostraram-se mais frequentes em indivíduos com DM2 quando comparados com indivíduos saudáveis. Não houve quaisquer diferenças relativamente à actividade enzimática da PON1, nem em relação ao polimorfismo da PON1 nem em relação à presença ou ausência de RD ou EMD. Conclui-se que o alelo A do polimorfismo RAGE -374A/T é factor protector para o aparecimento da RD e do EMD, enquanto os alelos Q e S dos polimorfismos PON1Gln192Arg e PON2Cys310Ser, respectivamente, são factores de risco para o aparecimento da DM2

The pericentric inversion on chromosome 16 [inv(16)(p13q22)] and related t(16;16)(p13;q22) are recurrent aberrations associated with acute myeloid leukemia (AML) M4 Eo. Both abberations result in a fusion of the core binding factor beta (CBFB) and smooth muscle myosin heavy chain gene (MYH11). A selected genomic 6.9-kb BamHl probe detects MYH11 DNA rearrangements in 18 of 19 inv(16)/t(16;16) patients tested using HindIII digested DNA. The rearranged fragments were not detectable after remission in two cases tested, while they were present after relapse in one of these two cases tested.

Trinta amostras de solos foram selecionadas com o objetivo de caracterizar o Fator Capacidade de Fósforo (FCP) e sua relação com propriedades de solos de Entre Ríos, Argentina. Foram estimadas isotermas de adsorção e realizado um teste rápido de adsorção de P, denominado P-remanescente. As equações de Freundlich e Langmuir descreveram adequadamente o fenômeno de adsorção de P. A capacidade máxima de adsorção variou de 46 a 716 mg kg-1 de P no solo, apresentando os solos analisados de muito baixo a médio FCP. A capacidade de adsorção de P aumentou com a quantidade de argila e não foi afetada pelo teor de carbono or¢nico do solo. O P-remanescente permitiu caracterizar os solos em relação ao fator capacidade e estimar sua capacidade máxima de adsorção de P.

Com o objetivo de estudar a aplicabilidade das equações de adsorção de Langmuir e de Freundlich para boro em solos de baixada do Estado do Rio de Janeiro (Glei Pouco Húmico e Cambissolo distrófico), desenvolveram-se ensaios de laboratório, em 1997, em quatro amostras de solo, sendo duas de superfície e duas subsuperficiais. Para isso, as amostras foram equilibradas em soluções de CaCl2 0,01 mol L-1, contendo as seguintes concentrações de boro: 0,0, 1,0, 2,0, 4,0 e 8,0 mg L-1. De posse dos dados, foram construídas isotermas de adsorção e verificados os ajustes das curvas obtidas, em relação aos dados experimentais. Observou-se que as isotermas mediante as equações de Langmuir e de Freundlich se adequaram bem para a adsorção de boro nos solos estudados.

Solos afetados por sais são característicos de zonas áridas/semi-áridas. Este trabalho foi desenvolvido no vale do Açu (RN), no período abril/90 a janeiro/91, objetivando avaliar o efeito do preparo do solo (convencional e convencional + subsolagem) e da aplicação de condicionadores ( gesso, esterco de curral e palha de carnaúba) nas propriedades químicas de um solo aluvial salino sódico. A subsolagem proporcionou efeito eficaz na redução da porcentagem de sódio trocável (PST) e no abaixamento do pH do solo. Nos tratamentos com gesso, foram detectados os maiores valores de cálcio trocável, redução na PST para menos de 15% na camada de 0-0,15 m e aumento na salinidade do solo. O esterco de curral foi benéfico na redução da PST, principalmente quando associado à subsolagem e à gessagem. Existe um desbalanço natural nas relações K:Ca:Mg trocáveis no solo, que se acentua com a gessagem, necessitando de correção potássica para que se atinja o equilíbrio. A proporção entre cátions trocáveis e solúveis no solo ficou entre 13 e 16:1, para cálcio e magnésio, e entre 0,3 e 3,9:1, para o sódio, sendo este o único cátion solúvel que diminuiu com a subsolagem.

A eficácia da cobertura vegetal morta no controle da erosão pode ser avaliada através de dois indicadores principais: a porcentagem de cobertura do solo pelos resíduos culturais e sua persistência sobre a superfície ao longo do tempo. O preparo do solo, por sua vez, pode exercer influência significativa sobre esses indicadores. O trabalho foi realizado no campo, no município de Eldorado do Sul, Depressão Central do Rio Grande do Sul. Avaliou-se a persistência da cobertura vegetal morta durante um período de pousio, que foi de maio de 1989 a abril de 1990, em sucessão à cultura da soja. Os resíduos dessa cultura foram manejados sem preparo, por escarificação e por gradagem. A porcentagem de cobertura do solo pelos resíduos culturais foi quantificada pelo método fotográfico e pelo da transeção linear. A cultura da soja produziu cobertura vegetal morta em pequena quantidade e de baixa durabilidade. A distribuição dos resíduos na superfície, sem preparo do solo, foi o tratamento que possibilitou melhor correlação (R²) entre os índices de cobertura obtidos pelos dois métodos testados. Nas áreas sob gradagem ou escarificação do solo, os índices de cobertura obtidos pelo método fotográfico foram superiores aos da transeção linear, enquanto, na área sem preparo do solo, houve similaridade entre os resultados dos dois métodos.

Integrin adhesion receptors consist of non-covalently linked alpha and beta subunits each of which contains a large extracellular domain, a single transmembrane domain and a short cytoplasmic tail. Engaged integrins recruit to focal structures globally termed adhesion complexes. The cytoplasmic domain of the beta subunit is essential for this clustering. beta1 and beta3 integrins can recruit at distinct cellular locations (i.e. fibrillar adhesions vs focal adhesions, respectively) but it is not clear whether individual beta subunit cytoplasmic and transmembrane domains are by themselves sufficient to drive orthotopic targeting to the cognate adhesion complex. To address this question, we expressed full-length beta3 transmembrane anchored cytoplasmic domains and truncated beta3 cytoplasmic domains as GFP-fusion constructs and monitored their localization in endothelial cells. Membrane-anchored full-length beta3 cytoplasmic domain and a beta3 mutant lacking the NXXY motif recruited to adhesion complexes, while beta3 mutants lacking the NPXY and NXXY motifs or the transmembrane domain did not. Replacing the natural beta subunit transmembrane domain with an unrelated (i.e. HLA-A2 alpha chain) transmembrane domain significantly reduced recruitment to adhesion complexes. Transmembrane anchored beta3 and cytoplasmic domain constructs, however, recruited without discrimination to beta1- and beta3-rich adhesions complexes. These findings demonstrate that membrane anchorage and the NPXY (but not the NXXY) motif are necessary for beta3 cytoplasmic domain recruitment to adhesion complexes and that the natural transmembrane domain actively contributes to this recruitment. The beta3 transmembrane and cytoplasmic domains alone are insufficient for orthotopic recruitment to cognate adhesion complexes.
