O trabalho foi realizado a campo para avaliar a eficiência de diversos tipos de leite de vaca sobre o oídio da abóbora cultivar Piramoita, causado por Sphaerotheca fuliginea. Utilizou-se o delineamento experimental de blocos ao acaso com seis tratamentos, cinco repetições e cinco plantas por parcela. Os tratamentos foram: leite cru, leite pasteurizado do tipo C, leite integral do tipo longa vida, leite pasteurizado tipo C + Yakult®, leite integral do tipo longa vida + Yakult® e água como controle. Os melhores resultados no controle da doença foram obtidos com leite de vaca cru, com leite do tipo C e leite do tipo C + Yakult®. Concluiu-se que leite é uma alternativa viável no controle do oídio, mesmo após o início da infecção, no campo. Sua utilização na forma de leite cru mais eficiente e na forma de longa vida o menos eficiente.

O botulismo alimentar ocorre pela ingestão de toxinas pré-formadas pelo Clostridium botulinum, consideradas as mais potentes dentre as toxinas conhecidas. Caracteriza-se como uma doença de extrema gravidade, de evolução aguda, provoca distúrbios digestivos e neurológicos, em conseqüência à ingestão de diversos tipos de alimentos. As conservas caseiras estão entre os alimentos que oferecem maior risco à população consumidora. Os produtos de origem animal são frequentemente associados aos surtos da doença, destacando-se os embutidos, tais como salsichas, salames, presuntos e patês. Derivados do leite e enlatados, bem como produtos fermentados, são passíveis de provocar a intoxicação. As outras formas naturais da doença são botulismo por feridas e botulismo infantil, normalmente associado ao consumo de mel contendo esporos do Clostridium botulinum.

Na produção de álcool por fermentação com leveduras, a floculação manifesta-se como um mecanismo natural de agregação de células. Essa condição pode ser induzida por vários fatores, entre eles, a interação entre bactérias floculentas como Lactobacillus fermentum e as leveduras. Esse fato torna-se prejudicial para o processo, pois tanto nas unidades que se utilizam do sistema de recuperação de células por centrifugação, como nas que não o utilizam, ocorrem perdas excessivas de fermento, em conseqüência dos problemas operacionais decorrentes da floculação, comprometendo seriamente o desempenho industrial. No presente trabalho, avaliou-se a floculação em vinho proveniente de fermentação experimental, sob duas condições de pH de fermento tratado utilizadas como inóculo e temperaturas de fermentação, não se observando diferenças significativas entre elas. Com este estudo, pôde-se ainda avaliar a capacidade de dispersão dos flocos em três condições de pH no tratamento do fermento em fase industrial, mostrando diferenças altamente significativas entre elas. A maior capacidade de dispersão no tratamento do fermento é desejável para o controle da floculação industrial, permitindo a sua centrifugação, com conseqüente separação das bactérias contaminantes do fermento. Essas constatações tornaram-se possíveis pelo emprego da técnica usual de determinação da floculação por espectrofotometria, modificada pela desfloculação prévia das amostras de vinho e fermento, o que conferiu maior estabilidade nas leituras.

Objective: To determine the effects produced by the ingestion of a fermented soy product (soy yoghurt), supplemented with isoflavones and associated with a resistive exercise program, on the bone metabolism of mature ovariectomized (Ovx) and sham-ovariectomized (sham-Ovx) rats.Methods: A total of 56 rats were used. They were divided into 2 sedentary control groups, the Ovx control group (C-Ovx) and the sham-Ovx control group (C-Sovx), each with 7 sedentary animals, and 2 treated groups, Ovx and sham-Ovx, with 21 animals each. These two treated groups were subdivided into three subgroups of seven animals each, which received the following treatments: consuming the soy yoghurt + sedentary, only subjected to resistive exercise, and consuming the soy yoghurt + resistive exercise. Both the program of resistive exercise and the consumption of soy yoghurt (at 3 mL/(kg body weight day)) continued for 12 weeks. The soy yoghurt was supplemented with isoflavones at 50 mg/100 g of product. The animals were sacrificed and their right-side femurs and tibias removed and assessed for bone mineral density (BMD). The alkaline phosphatase activity (AP) was determined in the blood serum.Results: There was a significant increase in both femur and tibia BMD values and in serum alkaline phosphatase activity in all the treated subgroups, compared with the control groups (p < 0.05).Conclusion: the ingestion of the soy yoghurt supplemented with isoflavones was capable of preventing a loss of bone mass in Ovx rats and of increasing bone mass in sham rats, whilst the resistive exercise program was effective in augmenting the bone mass in sham and Ovx rats. The comprehension of the structure of starch granules is important for the understanding of its physicochemical properties. Native and sour cassava starches after being analyzed with respect to their pasting properties and baking expansion capacity, were treated with 2.2 N HCl at 38 degreesC for a maximum of nine days. The starch granules remaining after lintnerization were analyzed for amylose content and intrinsic viscosity, by X-ray diffraction, scanning electron microscopy and chromatographic analysis. The results indicated that the acid hydrolysis on all starches occurred in two steps. The first one, with high hydrolysis rate, was characterized by a quick degradation of the amorphous part of the granules whereas the second step, with lower hydrolysis rate, was characterized by a higher resistance of the organized areas of the granules to acid treatment. Most of the amylose chains were found in the amorphous areas of starch granules only a small percentage was involved in the crystalline regions. The microscopic and chromatographic analysis demonstrated that the acid hydrolysis was not able to disrupt the entire granular crystalline structure. Fermented starch showed amylose and/or amylopectin chain fractions resistant to pullulanase, probably due to structural alterations during fermentation.

Enterococcus faecium CRL 183, a strain isolated from NSLAB cheese starter, has been the focus of much research on its potential probiotic capacity, although its survival through the gastrointestinal tract has not been demonstrated so far. In order to determine the capacity of E. faecium CRL 183 to survive such conditions, this strain was administered daily to rats for 30 weeks. The experimental animals were divided into Group I: those that did not receive E. faecium, Group II: those that received a pure culture of E. faecium CRL 183 and Group III: animals that received E. faecium CRL 183 in the form of a fermented soy-based product. Faecal samples were collected at the beginning and at the 50%, 75% and 100% stages of the experimental period. Isolation and counts of Enterococcus were carried out on KF selective media. To distinguish the various Enterococcus species in the faeces, biochemical (API Strep 20) and molecular (PCR) tests were performed. Initially, E. faecium was absent from the intestinal flora of the rats; however, after 15 weeks of administration, E. faecium could be recovered from the faeces of Groups II and III, demonstrating that E. faecium CRL 183 was able to survive gastrointestinal transit under the study conditions. This is further evidence of the probiotic qualities of this strain. The safety of the strain was also investigated with regard to body weight and serum biochemical analysis.

Yields and starch pasting characteristics obtained from wet milling of maize samples with low and high levels of defect grains were compared to those from sound samples. Defect grain groups ere established taking into account the defect degree. Thus the first group consisted of fermented, molded, heated and sprouted grains and the second of insect damaged. hollow, fermented (up to 1/4) grains and those injured by other causes. The grain groups, if present at low levels in the samples, 10% for first group and 17% for second group did not affect the chemical composition of starch and its pasting properties. obtained by the rapid visco analyser. Samples with high levels of grain groups (up to 100%). affected wet milling yields and starch viscosity. Samples with 100% of grains in the first group decreased starch, germ yield and peak viscosity and increased gluten yield. The effect of milk processing on the microstructure of probiotic low-fat yogurt was studied. Skim milk fortified with skim milk powder was subjected to three treatments prior to innoculation: thermal treatment at 85 degrees C for 30 min, high hydrostatic pressure at 676 MPa for 5 min, and combined treatments of high hydrostatic pressure (HHP) and heat. The processed milk was then fermented by using two different starter cultures containing Streptococcus thermophilus, Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus acidophilus, and Bifidobacterium longum. The microstructure of heat-treated milk yogurt had fewer interconnected chains of irregularly shaped casein micelles, forming a network that enclosed the void spaces. on the other hand, microstructure of HHP yogurt had more interconnected clusters of densely aggregated protein of reduced particle size, with an appearance more spherical in shape, exhibiting a smoother more regular surface and presenting more uniform size distribution. The combined HHP and heat milk treatments led to compact yogurt gels with increasingly larger casein micelle clusters interspaced by void spaces, and exhibited a high degree of cross-linking. The rounded micelles tended to fuse and form small irregular aggregates in association with clumps of dense amorphous material, which resulted in improved gel texture and viscosity. The in vitro gas production of four single roughages and their paired combinations (1:1 on dry matter basis) were evaluated. Two roughage samples (100 mg) per treatment were fermented with ruminal fluid during a 48 h incubation period. Total 48 h gas volumes of fermentation dry matter (DM), neutral detergent fiber (NDF) and soluble compounds in neutral detergent (NDS) were for sugarcane = 16.8, 11.2, 6.9 mL; sugarcane + corn silage = 20.1, 12.6, 9.1 mL; sugarcane + 60-day elephantgrass = 16.5, 17.6 mL; sugarcane + 180-day elephantgrass = 13.8, 8.2, 5.9 mL; corn silage = 18.8, 16.8, 4.7 mL; corn silage + 60-day elephantgrass = 16.3, 15.4, 2.4 mL; corn silage + 180-day elephantgrass = 16.1, 11.8, 4.2 mL; 60-day elephantgrass = 16.9, 19.0 mL and 180-day elephantgrass = fermented 10.7, 12.2 mL, respectively. The NDS gas production was not possible to estimate for sugarcane + 60-day elephantgrass, 60-day elephantgrass and 180-day elephantgrass. The present data shows that the curves subtraction method can be an option to evaluate the contribution of the soluble fractions in roughages to digestion kinetics. However, this method underestimates the NDS gas contribution when roughages are low in crude protein and soluble carbohydrates. It is advisable to directly apply the two-compartmental mathematical model to the digestion curves for roughage DM, when determining the NDS gas volume and the digestion rate. This method is more straightforward and accurate when compared to the curve subtraction method. Non-structural carbohydrates combined with fiber and protein promoted a positive associative effect in sugarcane + corn silage (50:50) mixture. Therefore, it can be concluded that the soluble fraction of roughages greatly contributes to gas production. (C) 2004 Elsevier B.V. All rights reserved.

The present study had the aim of testing the effect of different fermentation periods on the germination and vigour of pitomba seeds. The experiment was carried out in the greenhouse of the Seed Section of the Agrarian Sciences Center of the Federal University of Paraiba, Areia, PB. The fruits were picked directly from maternal trees located in the same municipal district, peeled manually and fermented for 24, 48, 72, 96 and 120 hours, in addition to seeds with pulp (without fermentation). The experiment was entirely randomized with four replications of 25 seeds per treatment. After each fermentation period, the seeds were washed in tap water and left in the laboratory environment for 24 hours on paper towels. Water content, germination and vigor (germination velocity index, seedling length and dry mass., relative frequency and medium time of germination) were measured. Less water content was shown in the seeds fermented for 96 hours (38.5%), while the largest germination percentages were observed after 76 hours of fermentation (93%). In relation to vigour, the best values occurred with 86 and 105 hours of fermentation. Fermentation is recommended for up to 105 hours as appropriate to removal of the pitomba seed aril.

This work was carried out at the Fazenda Experimental Lageado, at Botucatu, SP, to evaluate the physiological quality of cucumber (Cucumis sativus L.), seeds harvested at different fruit maturity stages and post-harvest resting periods. Fruits of cultivar Pérola were harvested at 6 ages (20, 25, 30, 35, 40 and 45 days after anthesis) and they were stored at room temperature for 0, 5, 10 or 15 days before seed extraction. After this, seeds were fermented for 2 days, washed and dried. External visual characteristics of fruits and seeds were observed at harvest. Seed quality was evaluated as for moisture, dry matter content, dimensions, germination, and vigour. Results showed that seed germination and vigour increased in fruits aging from 20 to 40 days. Post-harvest periods improved seed quality of 20-day old fruits not reaching the seed quality levels observed in 40 or 45-day old fruits, without post-harvest resting period. Seeds from fruits at 25, 30 and 35 days of age, with 10 or 15 days of post-harvest resting period reached the same quality levels of seeds from fruits with 40-45 days old.

Charqui meat is a tropical intermediate meat product formulated using hurdle technology, a concept described by Leistner (1,2). Salt, sodium nitrite, dehydration, and packaging are hurdles sequentially applied to inhibit deteriorating microorganisms with the possibility of selecting desirable microbiota. This would characterize charqui and its derivatives as fermented meat products. This paper reviews biochemical, physico-chemical, and ultrastructural changes that occur during processing of charquis.
