Context: Genetic polymorphisms at the perilipin (PLIN) locus have been investigated for their potential utility as markers for obesity and metabolic syndrome (MS). We examined in obese children and adolescents (OCA) aged 7-14 yr the association of single-nucleotide polymorphisms (SNP) at the PLIN locus with anthropometric, metabolic traits, and weight loss after 20-wk multi-disciplinary behavioral and nutritional treatment without medication. Design: A total of 234 OCA [body mass index (BMI = 30.4 +/- 4.4 kg/m(2); BMI Z-score = 2.31 +/- 0.4) were evaluated at baseline and after intervention. We genotyped four SNPs (PLIN1 6209T -> C, PLIN4 11482G -> A, PLIN5 13041A -> G, and PLIN6 14995A -> T). Results: Allele frequencies were similar to other populations, PLIN1 and PLIN4 were in linkage disequilibrium (D` = 0.999; P < 0.001). At baseline, no anthropometric differences were observed, but minor allele A at PLIN4 was associated with higher triglycerides (111 +/- 49 vs. 94 +/- 42 mg/dl; P = 0.003), lower high-density lipoprotein cholesterol (40 +/- 9 vs. 44 +/- 10 mg/dl; P = 0.003) and higher homeostasis model assessment for insulin resistance (4.0 +/- 2.3 vs. 3.5 +/- 2.1; P +/- 0.015). Minor allele A at PLIN4 was associated with MS risk (age and sex adjusted) hazard ratio 2.4 (95% confidence interval = 1.1-4.9) for genotype GA and 3.5 (95% confidence interval = 1.2-9.9) for AA. After intervention, subjects carrying minor allele T at PLIN6 had increased weight loss (3.3 +/- 3.7 vs. 1.9 +/- 3.4 kg; P = 0.002) and increased loss of the BMI Z-score (0.23 +/- 0.18 vs. 0.18 +/- 0.15; P +/- 0.003). Due to group size, risk of by-chance findings cannot be excluded. Conclusion: The minor A allele at PLIN4 was associated with higher risk of MS at baseline, whereas the PLIN6 SNP was associated with better weight loss, suggesting that these polymorphisms may predict outcome strategies based on multidisciplinary treatment for OCA. Objectives: E-cadherin and beta-catenin are adhesion molecules responsible for the maintenance of normal epithelial cell phenotype. A disturbance in epithelial cell adhesion, which leads to a more invasive and metastatic phenotype, is a hallmark of tumor progression. Several immunohistochemical studies have reported a strong correlation between loss of their expression to higher stage and grade in prostate carcinoma, but their influence in metastatic process is not yet known. The aim of this study is to verify the role of adhesion molecules in the progression of prostate cancer (PC), assessing the expression of E-cadherin and beta-catenin in bone metastasis. Materials and Methods: Twenty-eight bone metastases of prostate carcinoma were submitted to immunohistochemistry analysis for E-cadherin and beta-catenin expression. In 6 patients, we were able to assess the expression of the adhesion molecules in the primary tumors and their respective metastases. The definition of normal expression for both antibodies was strong and diffuse expression in more than 70% of tumor cells. Results: In bone metastases, there was loss of expression of E-cadherin and beta-catenin in 86% and 82%, respectively. Among the primary tumors, E-cadherin and beta-catenin expression was normal in 83% and 50% cases, respectively. Considering the 6 patients with paired primary and bone metastasis, we found loss of expression for both E-cadherin and beta-catenin in most of the cases. Conclusions: Comparing primary PC and its metastasis, we showed persistent loss of E-cadherin and beta-catenin expression. This phenomenon may be related to metastatic potential in PC, because we have shown underexpression for E-cadherin and beta-catenin in 86% and 82% of bone metastases.

The tissue microarray (TMA) technique allows multiple tissue samples in a single block. Commercial adhesive tape is used to avoid the loss of tissue samples during the immunostaining process. Few reports exist in the literature comparing the use of these adhesive tapes to other adhesive techniques. The objective of this study was to compare loss of sections adhered to slides using commercial adhesive tapes versus using silanized only slides. TMA was constructed with varying tissues using a fixed-base device (Beecher Instruments), placing 108 cylinders of 1 mm diameter in duplicate, spaced 1.2 mm apart. Section of 4 mu m were cut from the TMA block and adhered to 30 silanized slides and 30 commercial glass slides using adhesive tape, according to manufacturer`s recommendations. Vimentin immunoexpression was evaluated by immunohistochemistry. Antigenic recovery was realized in citrate buffer using a microwave oven. Cylinder loss in the immunohistochemical process was quantified and expressed as: total (>80%), almost complete (75-79%), or partial (50-74%). The commercial adhesive tape group presented lesser total loss (1.1 versus 6.4%), almost complete loss (2.2 versus 3.5%), and partial loss (2.1 versus 3.8%) than the silanized slide group (ANOVA, P < 0.05). The sum of total and almost complete losses in the silanized slide group was 9.9%, greater than the losses in slides using commercial adhesive tapes (3.3%) and less than reported and considered acceptable in the literature (10-30%). In conclusion, the use of silanized only slides presents very satisfactory results, requires less training, and reduces costs significantly, thus justifying their use in research.

Objective To evaluate the extent sensory and motor blocks produced by the epidural injection of different volumes of 0.25% bupivacaine (Bu) with methylene blue (MB), in dogs. Study design Prospective experimental trial. Animals Twenty healthy adult mongrel dogs, weighing 9.9 +/- 1.9 kg. Methods Dogs were randomly allocated into one of four groups that received 0.2, 0.4, 0.6 or 0.8 mL kg-1 of an epidural solution containing 0.25% Bu and MB. Sensory block was evaluated against time by pinching the tail, hind limb interdigital web, toenail bases and the skin over the vertebral dermatomes. Motor block was assessed by ataxia, hind limb weight-bearing ability and by loss of muscle tone of the tail and pelvic limbs. Data were collected at 2, 5, 10, 15 and 30 minutes after the end of epidural injection. After the final time point, dogs were euthanatized and laminectomies were conducted to expose the extent of the dural dye staining. Results The volumes 0.2, 0.4, 0.6 and 0.8 mL kg-1 of 0.25% Bu and MB blocked a mean of 5, 14.2, 20.2 and 21 dermatomes, respectively. The extent of the senory block increased up to a volume of 0.6 mL kg-1. Motor block was longer-lasting and more intense than sensory block. Complete dyeing of the spinal cord with MB was achieved in some dogs at 0.4 mL kg-1 and all dogs at 0.6 mL kg-1. Conclusions The volume of anesthetic injected into the epidural space plays an important role in the quality of the epidural anesthesia. At 0.25%, bupivacaine provided an efficient sensory block at 0.6 mL kg-1. Clinical relevance Relatively high volumes (0.6 mL kg-1) of 0.25%, BU and MB were needed to produce an effective sensory and motor block caudal to the umbilicus, but all spinal cord segments were reached by MB at this dose.

Esta tese apresenta os resultados da pesquisa que teve como objetivo analisar como os trabalhadores vivenciam os efeitos subjetivos produzidos pelo processo de trabalho de um Centro de Atenção Psicossocial Álcool e Drogas (CAPSad). A pesquisa foi realizada sob uma abordagem qualitativa, em um CAPSad do município de Vila Velha, Espírito Santo. A coleta de dados se deu por meio de cinco etapas: 1. Análise documental das políticas vigentes sobre uso de drogas; 2. Análise de prontuários; 3. Entrevista coletiva com dez trabalhadores; 4. Oitenta horas de observação do cotidiano de trabalho; 5. Entrevista em profundidade com treze trabalhadores. Para análise de dados foi utilizada a técnica da Análise Temática. Constatamos que no plano das políticas sobre o assunto,há prevalência de ideias relacionadas à repressão dos usuários, apesar da tentativa do Ministério da Saúde (MS) em abordar a redução de danos como uma estratégia que valoriza o sujeito e sua singularidade. A análise ainda apontou as dificuldades que os profissionais enfrentam neste município para atuar segundo as diretrizes do MS, uma vez que as ações municipais dão ênfase à repressão, à religiosidade e ao amedrontamento como estratégia de prevenção, com apoio da justiça e da polícia. Enfatizamos que tais ambiguidades repercutem no trabalho e para o trabalhador. Apontamos ainda outros aspectos que geram efeitos para os trabalhadores: condições de trabalho precárias (devido à estrutura do serviço, baixos salários e rede de atenção inexistente), falta de reconhecimento (devido à omissão da gerência e à ausência de normas) e sobrecarga (devido à falta de profissionais e aos conflitos nas divisões de tarefas). Essas situações levam a efeitos subjetivos como: desgaste, adoecimento, medo, incapacidade de agir, apatia, desvalorização, desmotivação e no aprisionamento do trabalhador. Notamos que estes efeitos são todos negativos e que os profissionais os vivenciam por meio do distanciamento afetivo no processo de trabalho, o que repercute negativamente na possibilidade de produção de um cuidado efetivo. Sugerimos que haja investimentos na formação de todos os trabalhadores que atuam nesse local, com foco na educação permanente, uma vez que por meio desta há o incentivo da aprendizagem e o enfrentamento criativo dos efeitos vivenciados no cotidiano.É preciso que haja diálogo, seja entre os trabalhadores e a gestão, entre os próprios trabalhadores e entre trabalhadores e usuários.

This article recommends a new way to improve Refugee Status Determination (RSD) procedures by proposing a network society communicative model based on active involvement and dialogue among all implementing partners. This model, named after proposals from Castells, Habermas, Apel, Chimni, and Betts, would be mediated by the United Nations High Commissioner for Refugees (UNHCR), whose role would be modeled after that of the International Committee of the Red Cross (ICRC) practice.

Motion compensated frame interpolation (MCFI) is one of the most efficient solutions to generate side information (SI) in the context of distributed video coding. However, it creates SI with rather significant motion compensated errors for some frame regions while rather small for some other regions depending on the video content. In this paper, a low complexity Infra mode selection algorithm is proposed to select the most 'critical' blocks in the WZ frame and help the decoder with some reliable data for those blocks. For each block, the novel coding mode selection algorithm estimates the encoding rate for the Intra based and WZ coding modes and determines the best coding mode while maintaining a low encoder complexity. The proposed solution is evaluated in terms of rate-distortion performance with improvements up to 1.2 dB regarding a WZ coding mode only solution.

Num mundo em que as redes de telecomunicações estão em constante evolução e crescimento, o consumo energético destas também aumenta. Com a evolução tanto por parte das redes como dos seus equipamentos, o custo de implementação de uma rede tem-se reduzido até ao ponto em que o maior obstáculo para o crescimento das redes é já o seu custo de manutenção e funcionamento. Nas últimas décadas têm sido criados esforços para tornar as redes cada fez mais eficientes ao nível energético, reduzindo-se assim os seus custos operacionais, como também a redução dos problemas relacionados com as fontes de energia que alimentam estas redes. Neste sentido, este trabalho tem como objectivo principal o estudo do consumo energético de redes IP sobre WDM, designadamente o estudo de métodos de encaminhamento que sejam eficientes do ponto de vista energético. Neste trabalho formalizámos um modelo de optimização que foi avaliado usando diferentes topologias de rede. O resultado da análise mostrou que na maioria dos casos é possível obter uma redução do consumo na ordem dos 25%.

This paper presents a variable speed autonomous squirrel cage generator excited by a current-controlled voltage source inverter to be used in stand-alone micro-hydro power plants. The paper proposes a system control strategy aiming to properly excite the machine as well as to achieve the load voltage control. A feed-forward control sets the appropriate generator flux by taking into account the actual speed and the desired load voltage. A load voltage control loop is used to adjust the generated active power in order to sustain the load voltage at a reference value. The control system is based on a rotor flux oriented vector control technique which takes into account the machine saturation effect. The proposed control strategy and the adopted system models were validated both by numerical simulation and by experimental results obtained from a laboratory prototype. Results covering the prototype start-up, as well as its steady-state and dynamical behavior are presented. (C) 2011 Elsevier Ltd. All rights reserved.

This paper proposes a computationally efficient methodology for the optimal location and sizing of static and switched shunt capacitors in large distribution systems. The problem is formulated as the maximization of the savings produced by the reduction in energy losses and the avoided costs due to investment deferral in the expansion of the network. The proposed method selects the nodes to be compensated, as well as the optimal capacitor ratings and their operational characteristics, i.e. fixed or switched. After an appropriate linearization, the optimization problem was formulated as a large-scale mixed-integer linear problem, suitable for being solved by means of a widespread commercial package. Results of the proposed optimizing method are compared with another recent methodology reported in the literature using two test cases: a 15-bus and a 33-bus distribution network. For the both cases tested, the proposed methodology delivers better solutions indicated by higher loss savings, which are achieved with lower amounts of capacitive compensation. The proposed method has also been applied for compensating to an actual large distribution network served by AES-Venezuela in the metropolitan area of Caracas. A convergence time of about 4 seconds after 22298 iterations demonstrates the ability of the proposed methodology for efficiently handling large-scale compensation problems.

This paper presents a new and efficient methodology for distribution network reconfiguration integrated with optimal power flow (OPF) based on a Benders decomposition approach. The objective minimizes power losses, balancing load among feeders and subject to constraints: capacity limit of branches, minimum and maximum power limits of substations or distributed generators, minimum deviation of bus voltages and radial optimal operation of networks. The Generalized Benders decomposition algorithm is applied to solve the problem. The formulation can be embedded under two stages; the first one is the Master problem and is formulated as a mixed integer non-linear programming problem. This stage determines the radial topology of the distribution network. The second stage is the Slave problem and is formulated as a non-linear programming problem. This stage is used to determine the feasibility of the Master problem solution by means of an OPF and provides information to formulate the linear Benders cuts that connect both problems. The model is programmed in GAMS. The effectiveness of the proposal is demonstrated through two examples extracted from the literature.

RESUMO: Introdução – A insuficiência de convergência (IC) pode desencadear alterações da atenção visual. Pretende-se investigar se existem alterações na atenção visual em estudantes do ensino superior com IC. Metodologia – Estudo quantitativo, comparativo e correlacional. Participaram 44 estudantes com idades compreendidas entre os 18 e os 24 anos. Formaram-se dois grupos, um com Visão Binocular Normal (VBN) e outro com IC. O grupo com IC incluiu os indivíduos que apresentaram alterações no ponto próximo de convergência (PPC) e/ou na convergência para perto (C’). Para avaliar a atenção visual utilizou-se o teste de cancelamento de sinos. Resultados – O grupo com VBN foi composto por 32 indivíduos (23 do género feminino) e o grupo com IC por 12 indivíduos (11 do género feminino). No teste de atenção visual verificou-se que o número médio de sinos identificados foi de 34,6 para o grupo com VBN e de 34,3 no grupo com IC. O tempo médio de realização do teste foi de 167,9s e de 198,3s para os grupos de VBN e IC, respetivamente. Observou-se uma correlação moderada positiva entre o PPC e o tempo médio de realização do teste (r≈0,63) e uma correlação fraca positiva entre o número médio de sinos identificados e a C’ (r≈0,16). Por outro lado, a correlação entre o PPC e o número médio de sinos identificados (r≈-0,48) foi fraca negativa e entre a C’ e o tempo médio (r≈-0,05) foi ínfima negativa. Discussão/Conclusões – O grupo com VBN apresenta um número médio de sinos identificados superior ao grupo com IC. Verifica-se ainda que o grupo com IC demorou mais tempo na realização do teste, comparativamente com o grupo com VBN. Estes resultados apontam para uma possível relação entre a IC e a diminuição da atenção visual.
