Background: Peste-des-petits ruminants virus (PPRV) is a non segmented negative strand RNA virus of the genus Morbillivirus within Paramyxoviridae family. Negative strand RNA viruses are known to carry nucleocapsid (N) protein, phospho (P) protein and RNA polymerase (L protein) packaged within the virion which possess all activities required for transcription, post-transcriptional modification of mRNA and replication. In order to understand the mechanism of transcription and replication of the virus, an in vitro transcription reconstitution system is required. In the present work, an in vitro transcription system has been developed with ribonucleoprotein (RNP) complex purified from virus infected cells as well as partially purified recombinant polymerase (L-P) complex from insect cells along with N-RNA (genomic RNA encapsidated by N protein) template isolated from virus infected cells. Results: RNP complex isolated from virus infected cells and recombinant L-P complex purified from insect cells was used to reconstitute transcription on N-RNA template. The requirement for this transcription reconstitution has been defined. Transcription of viral genes in the in vitro system was confirmed by PCR amplification of cDNAs corresponding to individual transcripts using gene specific primers. In order to measure the relative expression level of viral transcripts, real time PCR analysis was carried out. qPCR analysis of the transcription products made in vitro showed a gradient of polarity of transcription from 3' end to 5' end of the genome similar to that exhibited by the virus in infected cells. Conclusion: This report describes for the first time, the development of an in vitro transcription reconstitution system for PPRV with RNP complex purified from infected cells and recombinant L-P complex expressed in insect cells. Both the complexes were able to synthesize all the mRNA species in vitro, exhibiting a gradient of polarity in transcription.

The accuracy of pairing of the anticodon of the initiator tRNA (tRNA(fMet)) and the initiation codon of an mRNA, in the ribosomal P-site, is crucial for determining the translational reading frame. However, a direct role of any ribosomal element(s) in scrutinizing this pairing is unknown. The P-site elements, m(2)G966 (methylated by RsmD), m(5)C967 (methylated by RsmB) and the C-terminal tail of the protein S9 lie in the vicinity of tRNA(fMet). We investigated the role of these elements in initiation from various codons, namely, AUG, GUG, UUG, CUG, AUA, AUU, AUC and ACG with tRNA(CAU)(fmet) (tRNA(fMet) with CAU anticodon); CAC and CAU with tRNA(GUG)(fme); UAG with tRNA(GAU)(fMet) using in vivo and computational methods. Although RsmB deficiency did not impact initiation from most codons, RsmD deficiency increased initiation from AUA, CAC and CAU (2- to 3.6-fold). Deletion of the S9 C-terminal tail resulted in poorer initiation from UUG, GUG and CUG, but in increased initiation from CAC, CAU and UAC codons (up to 4-fold). Also, the S9 tail suppressed initiation with tRNA(CAU)(fMet)lacking the 3GC base pairs in the anticodon stem. These observations suggest distinctive roles of 966/967 methylations and the S9 tail in initiation.

Multiple methods currently exist for rapid construction and screening of single-site saturation mutagenesis (SSM) libraries in which every codon or nucleotide in a DNA fragment is individually randomized. Nucleotide sequences of each library member before and after screening or selection can be obtained through deep sequencing. The relative enrichment of each mutant at each position provides information on its contribution to protein activity or ligand-binding under the conditions of the screen. Such saturation scans have been applied to diverse proteins to delineate hot-spot residues, stability determinants, and for comprehensive fitness estimates. The data have been used to design proteins with enhanced stability, activity and altered specificity relative to wild-type, to test computational predictions of binding affinity, and for protein model discrimination. Future improvements in deep sequencing read lengths and accuracy should allow comprehensive studies of epistatic effects, of combinational variation at multiple sites, and identification of spatially proximate residues.

Interferon-gamma (Ifn gamma), a known immunomodulatory cytokine, regulates cell proliferation and survival. In this study, the mechanisms leading to the selective susceptibility of some tumor cells to Ifn gamma were deciphered. Seven different mouse tumor cell lines tested demonstrated upregulation of MHC class I to variable extents with Ifn gamma; however, only the cell lines, H6 hepatoma and L929 fibrosarcoma, that produce higher amounts of nitric oxide (NO) and reactive oxygen species (ROS) are sensitive to Ifn gamma-induced cell death. NO inhibitors greatly reduce Ifn gamma-induced ROS; however, ROS inhibitors did not affect the levels of Ifn gamma-induced NO, demonstrating that NO regulates ROS. Consequently, NO inhibitors are more effective, compared to ROS inhibitors, in reducing Ifn gamma-induced cell death. Further analysis revealed that Ifn gamma induces peroxynitrite and 3-nitrotyrosine amounts and a peroxynitrite scavenger, FeTPPS, reduces cell death. Ifn gamma treatment induces the phosphorylation of c-jun N-terminal kinase (Jnk) in H6 and L929 but not CT26, a colon carcinoma cell line, which is resistant to Ifn gamma-mediated death. Jnk activation downstream to NO leads to induction of ROS, peroxynitrite and cell death in response to Ifn gamma. Importantly, three cell lines tested, i.e. CT26, EL4 and Neuro2a, that are resistant to cell death with Ifn gamma alone become sensitive to the combination of Ifn gamma and NO donor or ROS inducer in a peroxynitrite-dependent manner. Overall, this study delineates the key roles of NO as the initiator and Jnk, ROS, and peroxynitrite as the effectors during Ifn gamma-mediated cell death. The addition of small amount of boron to Ti and it alloys refines the as-cast microstructure and enhances the mechanical properties. In this paper, we employ nanoindentation on each of the constituent phases in the microstructure and `rule-of-mixture' type analysis to examine their relative contributions to the strength enhancement in a Ti-6Al-4V alloy modified with 0.3 wt% B. Our results indicate to two main contributors to the relatively higher flow strength of B-modified alloy vis-a-vis the base alloy: (a) strengthening of alpha phase due to the reduction in the effective slip length that occurs as a result of the microstructural refinement that occurs upon B addition, and (b) composite strengthening caused by the TiB whiskers present in the alloy. Résumé: Cet article questionne la vision psychodynamique et psychosociale dans le parcours de vie de jeunes immigrés africains, vulnérables, placés en Maison d’Enfants à Caractère Social. Ce placement propose une formation professionnelle ou la poursuite des études, formations indispensables pour rester en France à condition de disposer de papiers en règle. La problématique de la fragmentation psychique et sociale chez ces jeunes dont le processus développemental est marqué par l’exil sera interrogée au regard des concepts tels que Destin et Destinée. Cette dernière sera aussi questionnée à la lumière du rôle et de la valeur du travail chez ces jeunes évoluant dans un nouveau contexte socio-économico-culturel qui tend à les stigmatiser. Par l’intermédiaire des données verbales et non verbales issues d’entretiens psychologiques, d’un groupe de parole et d’une grille d’observation inspirée d’un questionnaire, nous proposons une analyse des trajectoires personnelles au regard d’un processus d’identification. Cette analyse permet déjà d’aller au-delà des caractéristiques individuelles et de mettre en exergue le rôle capital qu’incarne le travail pour asseoir une identité sociale et sortir de l’état intemporel d’exil

Resumen: La prolongación de la edad y el aumento de la expectativa de vida han hecho surgir nuevas necesidades de atención para las personas ancianas y ello ha inducido a la ampliación de las prestaciones de servicios a domicilio. En este sentido, el trabajo de estos profesionales de intervención a domicilio, así como sus misiones, están en plena mutación. Es un sector con dificultades, debido a la dificultad de reclutar un personal especializado y al absentismo inquietante. Después de un análisis sobre el estado de este sector de la profesión en el que se establece la diferencia entre el enfoque físico y el psicológico sobre la salud, nuestra reflexión se centra en los riesgos psicosociales de esta población de profesionales. Se presentan los resultados preliminares de la investigación realizada con una muestra de 38 personas que ejercían laboralmente en este sector. Emerge la expresión de un sufrimiento emocional, en la que el aislamiento de los participantes es uno de los factores que caracterizan los oficios identificados con una fuerte dimensión relacional con personas vulnerables. La dimensión asistencial del trabajo se presenta como fuente de satisfacción del oficio pero, a su vez, puede conducir a una dificultad de gestionar acontecimientos no previstos

Apresenta reflexões sobre a democracia brasileira, abordando a representação, o impacto da imagem do Poder Legislativo sobre a participação popular e os efeitos dessas relações sobre a qualidade do regime democrático vivenciado. Enfoca a correlação entre a confiança da população no poder constituído, o pleno exercício da cidadania e o fortalecimento da democracia, enfatizando o papel da educação cidadã nesse processo.

Fifty-eight species of Decapods are enumerated from the collections examined by the author. Three species described by other authors are inserted in systematic order, thus making the list complete for the Panama region. All available material in the United States National Museum from Panama and Costa Rica is included; it ranges in age from the Oligocene (Culebra formation) to the Pleistocene.
