The aim of the present study was to test the hypothesis that oxidative stress and alteration of oxidative metabolism and apoptosis of neutrophils in dogs vary with the stage of leishmaniasis and to determine the contribution of uremia to such alterations. Dogs with leishmaniasis were classified into two stages: moderate (Leish II, n = 20) or very severe (i.e. with concurrent uremia; Leish IV, n = 20) according to the LeishVet Consensus. The two leishmaniasis groups were compared with uremic dogs without leishmaniasis (Uremic, n = 10) and to healthy dogs (Control, n = 30). To determine oxidative stress, total antioxidant/oxidant capacity, lipid peroxidation, total glutathione and the plasma antioxidants albumin, uric acid and bilirubin were quantified. Superoxide production was determined using the hydroethidine probe and viability and apoptosis were measured using annexin V-PE by capillary flow cytometry. Oxidative stress was present in both uremia and leishmaniasis with reduced total antioxidant capacity and was associated with increased induced production of superoxide and apoptosis. The greatest amount of oxidants was observed in animals with moderate disease only. Neutrophils from uremic dogs with and without leishmaniasis had decreased viability and an increased apoptosis rate in addition to increased lipid peroxidation. In conclusion, oxidative stress occurs in both stages of leishmaniasis with differences in intensity and levels of plasma markers; however, uremia does contribute to the decreased spontaneous viability of neutrophils in dogs in the final stage of the disease. © 2013 Elsevier Ltd. Bacterial fruit blotch of cucurbits (BFB), caused by the seed borne Gramnegative bacterium Acidovorax citrulli is a serious threat to cucurbit industry worldwide. Since late 1980`s after devastating outbreaks in watermelon fields in southern United States, BFB has spread worldwide and has been reported in other cucurbit crops such as melon, pumpkin, cucumber and squash. To date, there is evidence for the existence of at least two genetically and pathogenically distinct populations of A. citrulli. In Brazil, the first report of BFB was in 1991, in a watermelon field in São Paulo. Although widespread in the country, BFB has been a major problem to melon production. More precisely, BFB has caused significant yield losses to melon production in northeastern Brazil, which concentrates > 90% of the country`s melon production. Despite the management efforts and the recent advances in A. citrulli research, BFB is still a continuous threat to the cucurbit industry, including seed producers, growers and transplant nurseries. To better understand the population structure of A. citrulli strains in Brazil, and to provide a basis for the integrated management of BFB, we used pulsed-field gel electrophoresis (PFGE), multilocus sequence analysis (MLSA) of housekeeping and virulence-associated genes and pathogenicity tests on different cucurbit seedlings to characterize a Brazilian population of A. citrulli strains from different hosts and regions. Additionally, we conducted for the first time a comparative analysis of the A. citrulli group I and II population at genomic level and showed that these two groups differ on their genome sizes due to the presence of eight DNA segments, which are present in group II and absent in group I genomes. We also provide the first evidence to suggest that temperature might be a driver in the ecological adaptation of A. citrulli populations under nutrient-rich or -depleted conditions. Finally, in order to improve the routine detection of A. citrulli on melon seedlots, we designed a new primer set that is able to detect the different Brazilian haplotypes, thus minimizing the risk of false-negatives on PCR-based seed health testing.

The aim of this experiment was evaluate the effect of the bioestimulant administered in grafted and non-grafted japanese cucumber (Cucumis sativus L.) plants, under greenhouse conditions, in gas exchanges during the development of the plant and in the increase of yield. The experiment was carried out in the experimental area of Agronomic Sciences University of UNESP, Campus of Botucatu, São Paulo State, Brazil. The experiment design was completely randomized, in a factorial arrangement of 2x5, grafted and non-grafted plants and 5 treatments with bioestimulant: control, indolbutyric acid 0,0005% + citocinine 0,0009% + gibberelic acid 0,005% 250 mL ha-1; indolbutyric acid 0,0005% + citocinine 0,0009% + gibberelic acid 0,005% 375 mL ha-1; indolbutyric acid 0,0005% + citocinine 0,0009% + gibberelic acid 0,005% 500 mL ha-1; Yuca extract (Yucca elephantipes) + manganese + iron + copper + sulfur 375 mL ha-1 applied 15 days after the transplant, in intervals of 7 days between the applications, via leaf. The effect of the treatments were evaluated through the observations of the following characteristics: production of fruits (number and mass), average mass of the fruit and measures of gas exchanges. It can be concluded that indolbutyric acid 0,0005% + citocinine 0,0009% + gibberelic acid 0,005% 250 mL ha-1 increased CO2 assimilation rate and the water efficiency, influencing in addition to increase the quantity of the fruits

Cucumber (Cucumis sativus L.) is a plant of the Cucurbitaceae family of great commercial importance and their culture is widespread throughout the country. The production of vegetables, high-quality care begins with training ranging from seedlings and shading techniques have an important role in creating environments more conducive to the production of seedlings and many researchers and producers of seedlings have opted for the use of color shading screens, making this way, the wavelength and the amount of incident energy are changed in favor of the plants. Work will be performed at the Department of Forest Sciences, Faculty of Agricultural Sciences, Universidade Estadual Paulista (UNESP), Botucatu campus. The statistical design will be in three blocks, each containing 100 plots of each treatment (seedlings covered with blue screen, red and black). Evaluations of dry mass and height will be made at 7, 14, 21 and 28 days after emergence, and 5 will be selected seedlings of each treatment. At 14 and 28 DAE was measured, and SPAD chlorophyll content at 30 DAE, was evaluated by measuring gas exchange in the second pair of leaves, fully expanded for each plant, which was put in a chamber controlled with infrared gas analyzer (IRGA LI 6400, Licor) to perform measurements of photosynthesis. Treatment with black shading showed statistically better results for total dry matter, plant height, SPAD chlorophyll content and net assimilation of CO2, showing a better quality seedlings and its use is recommended in the cultivation of cucumber Taiko
