A permeabilidade ao ar pode ser utilizada como indicador da qualidade física do solo. O objetivo deste trabalho foi determinar a permeabilidade ao ar e os índices de continuidade de poros para a cama de semeadura em um Latossolo Vermelho distrófico cultivado no sistema semeadura direta e submetido à escarificação mecânica e escarificação biológica, utilizando a cultura do nabo forrageiro. O estudo foi conduzido em área experimental da Universidade Estadual de Ponta Grossa, Estado do Paraná. Os tratamentos implantados foram: sistema semeadura direta por 18 anos consecutivos (SD); semeadura direta submetido à escarificação mecânica (SDE); e semeadura direta submetido à escarificação biológica por meio da cultura do nabo forrageiro (SDNF). O delineamento experimental utilizado foi em blocos casualizados com quatro repetições. As amostragens de solo foram feitas aos seis e 18 meses após a implantação dos tratamentos, correspondentes às semeaduras das culturas do milho (outubro de 2009) e da soja (novembro de 2010), respectivamente, nas camadas de 0,00-0,05 e 0,05-0,10 m de profundidade. A permeabilidade ao ar foi determinada pelo permeâmetro de carga constante nos potenciais mátricos -6, -10, -30 e -100 kPa. Foram definidos os seguintes índices de continuidade de poros: índice N, índice K1 e volume de poros bloqueados; e a porosidade de aeração. Os resultados foram submetidos à análise de variância e, quando significativos, as médias dos tratamentos foram comparadas pelo teste Tukey (p<0,05). Para a camada de 0,00-0,05 m, a permeabilidade ao ar e o índice K1 no SDNF no potencial mátrico de -6 kPa foram significativamente maiores que em SD e SDE. O índice N, o volume de poros bloqueados e a porosidade de aeração não apresentaram diferenças significativas entre os tratamentos em ambas as profundidades. Os efeitos benéficos da escarificação biológica na permeabilidade do solo ao ar e no índice de continuidade de poros K1 persistiram aos 18 meses após sua aplicação. A escarificação mecânica resultou em maior continuidade do sistema poroso do solo, avaliado pelo índice K1, com persistência desse efeito aos 18 meses após sua aplicação.

By an analysis of the exchange of carriers through a semiconductor junction, a general relationship for the nonequilibrium population of the interface states in Schottky barrier diodes has been derived. Based on this relationship, an analytical expression for the ideality factor valid in the whole range of applied bias has been given. This quantity exhibits two different behaviours depending on the value of the applied bias with respect to a critical voltage. This voltage, which depends on the properties of the interfacial layer, constitutes a new parameter to complete the characterization of these junctions. A simple interpretation of the different behaviours of the ideality factor has been given in terms of the nonequilibrium charging properties of interface states, which in turn explains why apparently different approaches have given rise to similar results. Finally, the relevance of our results has been considered on the determination of the density of interface states from nonideal current-voltage characteristics and in the evaluation of the effects of the interfacial layer thickness in metal-insulator-semiconductor tunnelling diodes.

Uma topossequência apresenta solos com características estruturais e morfológicas distintas que podem alterar a susceptibilidade à deformação estrutural em curtas distâncias espaciais. Os objetivos deste estudo foram avaliar a capacidade de suporte de carga e a suscetibilidade à compactação e mensurar o efeito que cargas acima e abaixo da pressão de preconsolidação causam na permeabilidade ao ar de solos numa topossequência da Depressão Central do Rio Grande do Sul, Brasil. Amostras indeformadas foram coletadas nos horizontes de três perfis de Argissolos (Ap, A1, AB, BA, Bt1 e Bt2; Ap, A1, A2, AB, BA, Bt1 e Bt2; e Ap, A1, BA, Bt1 e Bt2) e dois de Gleissolos (A1, A2, Bg; A, Bg e Cg), no longo de uma topossequência. Essas amostras foram submetidas ao teste de compressão uniaxial, sendo avaliada a permeabilidade ao ar antes e após compressão. Na condição de capacidade de campo, os Gleissolos apresentaram menor suscetibilidade à compactação e maior capacidade de suporte de carga que os Argissolos. O teor de argila esteve diretamente correlacionado com a capacidade de suporte de carga. Portanto, foi inversamente proporcional à suscetibilidade à compactação, exceto em condições de elevada umidade do solo quando a água lubrificou as superfícies aumentando a susceptibilidade à compactação de quaisquer solos. Até 25 kPa de carga ao solo não resultou degradação física do solo, mas quando foram aplicadas cargas de 200 kPa, mesmo na condição de capacidade de campo, houve redução significativa do volume de macroporos e porosidade total em níveis inferiores ao limite crítico (0,10 m3 m-3).

Approaching or looming sounds (L-sounds) have been shown to selectively increase visual cortex excitability [Romei, V., Murray, M. M., Cappe, C., & Thut, G. Preperceptual and stimulus-selective enhancement of low-level human visual cortex excitability by sounds. Current Biology, 19, 1799-1805, 2009]. These cross-modal effects start at an early, preperceptual stage of sound processing and persist with increasing sound duration. Here, we identified individual factors contributing to cross-modal effects on visual cortex excitability and studied the persistence of effects after sound offset. To this end, we probed the impact of different L-sound velocities on phosphene perception postsound as a function of individual auditory versus visual preference/dominance using single-pulse TMS over the occipital pole. We found that the boosting of phosphene perception by L-sounds continued for several tens of milliseconds after the end of the L-sound and was temporally sensitive to different L-sound profiles (velocities). In addition, we found that this depended on an individual's preferred sensory modality (auditory vs. visual) as determined through a divided attention task (attentional preference), but not on their simple threshold detection level per sensory modality. Whereas individuals with "visual preference" showed enhanced phosphene perception irrespective of L-sound velocity, those with "auditory preference" showed differential peaks in phosphene perception whose delays after sound-offset followed the different L-sound velocity profiles. These novel findings suggest that looming signals modulate visual cortex excitability beyond sound duration possibly to support prompt identification and reaction to potentially dangerous approaching objects. The observed interindividual differences favor the idea that unlike early effects this late L-sound impact on visual cortex excitability is influenced by cross-modal attentional mechanisms rather than low-level sensory processes.

Hepatitis A virus (HAV), the prototype of genus Hepatovirus, has several unique biological characteristics that distinguish it from other members of the Picornaviridae family. Among these, the need for an intact eIF4G factor for the initiation of translation results in an inability to shut down host protein synthesis by a mechanism similar to that of other picornaviruses. Consequently, HAV must inefficiently compete for the cellular translational machinery and this may explain its poor growth in cell culture. In this context of virus/cell competition, HAV has strategically adopted a naturally highly deoptimized codon usage with respect to that of its cellular host. With the aim to optimize its codon usage the virus was adapted to propagate in cells with impaired protein synthesis, in order to make tRNA pools more available for the virus. A significant loss of fitness was the immediate response to the adaptation process that was, however, later on recovered and more associated to a re-deoptimization rather than to an optimization of the codon usage specifically in the capsid coding region. These results exclude translation selection and instead suggest fine-tuning translation kinetics selection as the underlying mechanism of the codon usage bias in this specific genome region. Additionally, the results provide clear evidence of the Red Queen dynamics of evolution since the virus has very much evolved to re-adapt its codon usage to the environmental cellular changing conditions in order to recover the original fitness.

The pursuit of high response rates to minimise the threat of nonresponse bias continues to dominate decisions about resource allocation in survey research. Yet a growing body of research has begun to question this practice. In this study, we use previously unavailable data from a new sampling frame based on population registers to assess the value of different methods designed to increase response rates on the European Social Survey in Switzerland. Using sampling data provides information about both respondents and nonrespondents, making it possible to examine how changes in response rates resulting from the use of different fieldwork methods relate to changes in the composition and representativeness of the responding sample. We compute an R-indicator to assess representativity with respect to the sampling register variables, and find little improvement in the sample composition as response rates increase. We then examine the impact of response rate increases on the risk of nonresponse bias based on Maximal Absolute Bias (MAB), and coefficients of variation between subgroup response rates, alongside the associated costs of different types of fieldwork effort. The results show that increases in response rate help to reduce MAB, while only small but important improvements to sample representativity are gained by varying the type of effort. These findings lend further support to research that has called into question the value of extensive investment in procedures aimed at reaching response rate targets and the need for more tailored fieldwork strategies aimed both at reducing survey costs and minimising the risk of bias.

As the list of states adopting the HWTD continues to grow, there is a need to evaluate how results are utilized. AASHTO T 324 does not standardize the analysis and reporting of test results. Furthermore, processing and reporting of the results among manufacturers is not uniform. This is partly due to the variation among agency reporting requirements. Some include only the midpoint rut depth, while others include the average across the entire length of the wheel track. To eliminate bias in reporting, statistical analysis was performed on over 150 test runs on gyratory specimens. Measurement location was found to be a source of significant variation in the HWTD. This is likely due to the nonuniform wheel speed across the specimen, geometry of the specimen, and air void profile. Eliminating this source of bias when reporting results is feasible though is dependent upon the average rut depth at the final pass. When reporting rut depth at the final pass, it is suggested for poor performing samples to average measurement locations near the interface of the adjoining gyratory specimens. This is necessary due to the wheel lipping on the mold. For all other samples it is reasonable to only eliminate the 3 locations furthest from the gear house. For multi‐wheel units, wheel side was also found to be significant for poor and good performing samples. After eliminating the suggested measurements from the analysis, the wheel was no longer a significant source of variation.

In the vast majority of bottom-up proteomics studies, protein digestion is performed using only mammalian trypsin. Although it is clearly the best enzyme available, the sole use of trypsin rarely leads to complete sequence coverage, even for abundant proteins. It is commonly assumed that this is because many tryptic peptides are either too short or too long to be identified by RPLC-MS/MS. We show through in silico analysis that 20-30% of the total sequence of three proteomes (Schizosaccharomyces pombe, Saccharomyces cerevisiae, and Homo sapiens) is expected to be covered by Large post-Trypsin Peptides (LpTPs) with M(r) above 3000 Da. We then established size exclusion chromatography to fractionate complex yeast tryptic digests into pools of peptides based on size. We found that secondary digestion of LpTPs followed by LC-MS/MS analysis leads to a significant increase in identified proteins and a 32-50% relative increase in average sequence coverage compared to trypsin digestion alone. Application of the developed strategy to analyze the phosphoproteomes of S. pombe and of a human cell line identified a significant fraction of novel phosphosites. Overall our data indicate that specific targeting of LpTPs can complement standard bottom-up workflows to reveal a largely neglected portion of the proteome.

Neurally adjusted ventilatory assist or NAVA is a new assisted ventilatory mode which, in comparison with pressure support, leads to improved patient-ventilator synchrony and a more variable ventilatory pattern. It also improves arterial oxygenation. With NAVA, the electrical activity of the diaphragm is recorded through a nasogastric tube equipped with electrodes. This electrical activity is then used to pilot the ventilator. With NAVA, the patient's respiratory pattern controls the ventilator's timing of triggering and cycling as well as the magnitude of pressurization, which is proportional to inspiratory demand. The effect of NAVA on patient outcome remains to be determined through well-designed prospective studies.

Previously we determined that S81 is the highest stoichiometric phosphorylation on the androgen receptor (AR) in response to hormone. To explore the role of this phosphorylation on growth, we stably expressed wild-type and S81A mutant AR in LHS and LAPC4 cells. The cells with increased wild-type AR expression grow faster compared with parental cells and S81A mutant-expressing cells, indicating that loss of S81 phosphorylation limits cell growth. To explore how S81 regulates cell growth, we tested whether S81 phosphorylation regulates AR transcriptional activity. LHS cells stably expressing wild-type and S81A mutant AR showed differences in the regulation of endogenous AR target genes, suggesting that S81 phosphorylation regulates promoter selectivity. We next sought to identify the S81 kinase using ion trap mass spectrometry to analyze AR-associated proteins in immunoprecipitates from cells. We observed cyclin-dependent kinase (CDK)9 association with the AR. CDK9 phosphorylates the AR on S81 in vitro. Phosphorylation is specific to S81 because CDK9 did not phosphorylate the AR on other serine phosphorylation sites. Overexpression of CDK9 with its cognate cyclin, Cyclin T, increased S81 phosphorylation levels in cells. Small interfering RNA knockdown of CDK9 protein levels decreased hormone-induced S81 phosphorylation. Additionally, treatment of LNCaP cells with the CDK9 inhibitors, 5,6-dichloro-1-β-D-ribofuranosylbenzimidazole and Flavopiridol, reduced S81 phosphorylation further, suggesting that CDK9 regulates S81 phosphorylation. Pharmacological inhibition of CDK9 also resulted in decreased AR transcription in LNCaP cells. Collectively these results suggest that CDK9 phosphorylation of AR S81 is an important step in regulating AR transcriptional activity and prostate cancer cell growth.

This paper focused on four alternatives of analysis of experiments in square lattice as far as the estimation of variance components and some genetic parameters are concerned: 1) intra-block analysis with adjusted treatment and blocks within unadjusted repetitions; 2) lattice analysis as complete randomized blocks; 3) intrablock analysis with unadjusted treatment and blocks within adjusted repetitions; 4) lattice analysis as complete randomized blocks, by utilizing the adjusted means of treatments, obtained from the analysis with recovery of interblock information, having as mean square of the error the mean effective variance of this same analysis with recovery of inter-block information. For the four alternatives of analysis, the estimators and estimates were obtained for the variance components and heritability coefficients. The classification of material was also studied. The present study suggests that for each experiment and depending of the objectives of the analysis, one should observe which alternative of analysis is preferable, mainly in cases where a negative estimate is obtained for the variance component due to effects of blocks within adjusted repetitions.

O trabalho foi desenvolvido com o objetivo de avaliar a evolução física do processo de remoção de água das sementes em secador estacionário, com cilindro central perfurado e distribuição radical de ar. A pesquisa foi conduzida com sementes de soja, variando o fluxo (26,9, 28,4 e 33,2 m³/minuto/t) e a temperatura do ar insuflado (42, 46 e 50ºC), considerando a posição das sementes (17, 34 e 51 cm em relação ao cilindro de insuflação) e o tempo de secagem (zero a doze horas, com intervalos de duas horas). Foram caracterizados o ar ambiente, o ar insuflado, as temperaturas e os teores de água da massa, as velocidades e curvas de secagem. As avaliações realizadas destacaram vantagens físicas operacionais da combinação de 28,4 m³/minuto/t com 46ºC e o contrário, com a combinação de 26,9 m³/minuto/t com 42ºC.
