Abstract: INTRODUCTION: Leprosy is mainly transmitted among family members who share genetic and ambient factors. The clinical form of leprosy in the index case and kinship could be risk factors for leprosy transmission. High antibody levels in household contacts (HC) in the absence of neural or skin lesions may characterize latent infection. This study aimed to evaluate the association between seropositivity for anti-phenolic glycolipid-I immunoglobulin M antibodies (APGL-I) in HC and the clinical classification of the index case and to analyze the association between APGL-I positivity with other factors such as age, kinship, and gender. METHODS: We performed a survey among 320 HC of 120 leprosy patients who were evaluated and followed-up in a leprosy outpatient clinic of a university hospital. All HC underwent complete skin examination, peripheral nerve palpation, skin sensory tests, and serologic tests for the detection and quantification of APGL-I. RESULTS: The overall seropositivity rate was 20%, and was greatly affected by kinship. APGL-I seropositivity was higher in siblings (41%), followed by parents (28%), spouses (26%), other (19%), and offspring (14%). Independent risk factors for seropositivity were being siblings (OR 3.3) and being a HC of an index case with indeterminate leprosy (OR 5.3). APGL-I seropositivity was associated with index cases with a bacillary index of 4 (88%; p<.001). Seropositivity among HC was not significantly associated with their gender and age. There was no statistical difference in the seropositivity rates of HC of index patients with paucibacillary and multibacillary leprosy. CONCLUSIONS: Strict evaluation and follow-up of HC with positive results for APGL-I is recommended. Special attention should be paid during the screening of siblings of the index cases, HC of patients with a high bacillary index, and HC of patients with indeterminate leprosy.

This paper addresses the growing difficulties automobile manufacturers face within their after sales business: an increasing number of trade obstacles set up by import countries discriminates against the foreign suppliers and impedes the international sales of genuine parts. The purpose of the study is to explore the emergence of trade restrictive product certification systems, which affect spare parts exports of automobile manufacturers. The methodology used includes review of the literature and an empirical study based on qualitative interviews with representatives of major stakeholders of the automotive after sales business. Relevant key drivers, which initiate the introduction of technical regulations in importing countries, are identified and analysed to evaluate their effect on the emerging trade policy. The analysis of the key drivers outlines that several interacting components, such as the global competitiveness of the country, macroeconomic and microeconomic factors, and certain country-specific variables induce trade restrictive product certification systems. The findings allow for an early detection of the emergence of product certification systems and provide a means to early recognise the risks and opportunities for the sales of automotive spare parts in the automakers’ target markets. This allows the manufacturers to react immediately and adapt in time to the upcoming changes.

Poor water quality condition has been pointed out as one of the major causes for the high mortality of ornamental fishes exported from the state of Amazonas, Brazil. The purpose of the current study was to define water quality standards for cardinal tetra (Paracheirodon axelrodi), by establishing the lower and higher for lethal temperature (LT50), lethal concentration (LC50) for total ammonia and nitrite and LC50 for acid and alkaline pH. According to the findings, cardinal tetra is rather tolerant to high temperature (33.3 ºC), to a wide pH range (acid pH=2.9 and alkaline pH=8.8) and to high total ammonia concentration (23.7 mg/L). However, temperatures below 19.6 ºC and nitrite concentrations above 1.1 mg/L NO2- may compromise fish survival especially during long shipment abroad.

The aim of this study was not only to determine the red blood cells parameters, thrombocyte and leukocyte counts in farmed Brycon amazonicus (matrinxã), to compare these parameters among Bryconinae species from literature, and also to investigate the presence of special granulocytic cells in these fish. The results of the blood cells parameters here established for farmed B. amazonicus, a species of great economic importance in Brazilian aquaculture, could help a better understanding of the blood features in natural populations of this Amazon species. Blood parameters varied between Bryconinae species investigated, mainly the red blood cell counts, hemoglobin, hematocrit and mean corpuscular volume (MCV). The presence of the blood granulocytes, neutrophils and heterophils in matrinxã suggest that both leukocytes can be a characteristic for Bryconinae family. Furthermore, it indicates that the existence of special granulocytic cells in the blood of Bryconinae species from literature is an artifact, and this was herein discussed.

In order to determine the lethal dose (96-h LD50) of the bacteria Aeromonas hydrophila to matrinxã, Brycon amazonicus, to be applied in challenge tests, 90 fish (63.23 ± 6.39 g) were divided into five treatments, with different bacterial solutions: T1 - Control (0.9% NaCl saline solution); T2 (4 x 10(11) cells/ mL); T3 (5 x 10(11) cells/ mL); T4 (1.36 x 10(12) cells/ mL) and T5 (3.06 x 10(12) cells/ mL). Fish were previously anesthetized with benzocaine (60 mg L-1), inoculated in the peritoneal cavity with the bacterial suspensions and then distributed into fifteen 80-L test chambers, where the water variables were monitored and fish mortality was observed. The experiment was randomly designed in three replicates and the 96-h LD50 was estimated according to the trimmed Spearman-Karber method. Water quality variables remained within adequate ranges for fish health and performance. Fish mortality rate increased with the bacterial concentrations of A. hydrophila (T1 = 0%; T2 = 16.66%; T3 = 44.44%; T4 = 72.22% and T5 = 100%), and the first mortalities were observed after 57 h, although the signs of the bacterial infection were already observed 24 h after the inoculation. The results indicate that the 96-h LD50 value of A. hydrophila to matrinxã is 6.66 x 10(11) cells/ mL.

Metabolic adaptation is considered an emerging hallmark of cancer, whereby cancer cells exhibit high rates of glucose consumption with consequent lactate production. To ensure rapid efflux of lactate, most cancer cells express high levels of monocarboxylate transporters (MCTs), which therefore may constitute suitable therapeutic targets. The impact of MCT inhibition, along with the clinical impact of altered cellular metabolism during prostate cancer (PCa) initiation and progression, has not been described. Using a large cohort of human prostate tissues of different grades, in silico data, in vitro and ex vivo studies, we demonstrate the metabolic heterogeneity of PCa and its clinical relevance. We show an increased glycolytic phenotype in advanced stages of PCa and its correlation with poor prognosis. Finally, we present evidence supporting MCTs as suitable targets in PCa, affecting not only cancer cell proliferation and survival but also the expression of a number of hypoxia-inducible factor target genes associated with poor prognosis. Herein, we suggest that patients with highly glycolytic tumours have poorer outcome, supporting the notion of targeting glycolytic tumour cells in prostate cancer through the use of MCT inhibitors.

OBJETIVO: O presente estudo teve como objetivo conhecer evidências de validade e precisão do Alcohol Use Disorder Identification Test (AUDIT). MÉTODOS: Contou-se com uma amostra de conveniência (não probabilística) de 547 estudantes universitários de Fortaleza (CE), com idade média de 21,6 anos (dp = 4,86; amplitude de 18 a 53), a maioria do sexo masculino (51,5%), solteira (91,4%) e católica (62,5%). Os participantes responderam ao AUDIT e a perguntas demográficas. Procurando conhecer a estrutura fatorial, além de estatísticas descritivas, realizou-se uma Análise de Componentes Principais. Adicionalmente, a fim de avaliar a precisão do instrumento, efetuaram-se cálculos de alfa de Cronbach (consistência interna), correlações de r de Pearson e coeficiente de correlação intraclasse - ICC (precisão teste-reteste). RESULTADOS: De acordo com a análise de componentes principais com rotação oblimin, a estrutura bifatorial do AUDIT mostrou-se coerente, com todos os itens apresentando saturações satisfatórias, superior a |0,40|, tendo o Fator 1 explicado 47,5% da variância total com alfa de 0,84 e o Fator 2 explicado 11,6% da variância total com alfa de 0,69. Os resultados do teste-reteste indicaram correlação forte entre os dados obtidos na primeira (t1) e segunda (t2) aplicação (r tt = 0,94, p < 0,01), sem diferença significativa de médias nos dois tempos (m t1 = 0,37, dp = 0,49; m t2 = 0,34, dp2= 0,47; p > 0,05), com ICC satisfatório (0,96). CONCLUSÕES: Os achados apoiaram a adequação psicométrica do AUDIT, com as análises fatoriais exploratórias apontando como mais satisfatória a estrutura com dois fatores, bem como atestaram sua boa estabilidade temporal.

Mutations or amplification of the MET proto-oncogene are involved in the pathogenesis of several tumours, which rely on the constitutive engagement of this pathway for their growth and survival. However, MET is expressed not only by cancer cells but also by tumour-associated stromal cells, although its precise role in this compartment is not well characterized. Here we show that MET is required for neutrophil chemoattraction and cytotoxicity in response to its ligand hepatocyte growth factor (HGF). Met deletion in mouse neutrophils enhances tumour growth and metastasis. This phenotype correlates with reduced neutrophil infiltration to both the primary tumour and metastatic sites. Similarly, Met is necessary for neutrophil transudation during colitis, skin rash or peritonitis. Mechanistically, Met is induced by tumour-derived tumour necrosis factor (TNF)-a or other inflammatory stimuli in both mouse and human neutrophils. This induction is instrumental for neutrophil transmigration across an activated endothelium and for inducible nitric oxide synthase production upon HGF stimulation. Consequently, HGF/MET-dependent nitric oxide release by neutrophils promotes cancer cell killing, which abates tumour growth and metastasis. After systemic administration of a MET kinase inhibitor, we prove that the therapeutic benefit of MET targeting in cancer cells is partly countered by the pro-tumoural effect arising from MET blockade in neutrophils. Our work identifies an unprecedented role of MET in neutrophils, suggests a potential 'Achilles' heel' of MET-targeted therapies in cancer, and supports the rationale for evaluating anti-MET drugs in certain inflammatory diseases.

This research uses the textile/text axis concept as a conceptual tool to investigate the role of textile and text in contemporary women’s art practice and theorizing, investigating textile as a largely hitherto unacknowledged element in women’s art practice of the late 20th and early 21st centuries. Textile and text share a common etymological root, from the Latin textere to weave, textus a fabric. The thesis illuminates the pathways whereby textile and text played an important role in women reclaiming a speaking voice as creators of culture and signification during a revolutionary period of renewal in women’s cultural contribution and positioning. The methodological approach used in the research consisted of a comprehensive literature review, the compilation of an inventory of relevant women artists, developing a classificatory system differentiating types of approaches, concerns and concepts underpinning women’s art practice vis a vis the textile/text axis and a series of three in-depth case studies of artists Tracey Emin, Louise Bourgeois and Faith Ringgold. The thesis points to the fact that contemporary women artists and theorists have rounded their art practice and aesthetic discourse in textile as prime visual metaphor and signifier, turning towards the ancient language of textile not merely to reclaim a speaking voice but to occupy a ground breaking locus of signification and representation in contemporary culture. The textile/text axis facilitated women artists in powerfully countering a culturally inscribed status of Lacanian ‘no-woman’ (a position of abjection, absence and lack in the phallocentric symbolic). Turning towards a language of aeons, textile as fertile wellspring, the thesis identifies the methodologies and strategies whereby women artists have inserted their webs of subjectivities and deepest concerns into the records and discourses of contemporary culture. Presenting an anatomy of the textile/text axis, the thesis identifies nine component elements manifesting in contemporary women’s aesthetic practice and discourse. In this cultural renaissance, the textile/text axis, the thesis suggests, served as a complex lexicon, a system of labyrinthine references and signification, a site of layered meanings and ambiguities, a body proxy and a corporeal cartography, facilitating a revolution in women’s aesthetic praxis.

FUNDAMENTO: Medir Qualidade de Vida (QV) relacionada à saúde auxilia na avaliação da eficiência de um tratamento e identifica problemas de maior impacto na QV do paciente. No entanto, essas medidas são mais seguras se avaliadas por instrumentos genéricos e específicos conjuntamente, fazendo-se necessário verificar se há compatibilidade entre esses e evitar repetições e contradições entre os domínios. OBJETIVO: Descrever o perfil de qualidade de vida de pacientes hipertensos e avaliar a compatibilidade de um instrumento específico (MINICHAL) e outro genérico (SF-36). MÉTODOS: Cem pacientes hipertensos adultos em tratamento ambulatorial entrevistados. A média da QVRS medida pelo MINICHAL foi de 6,64 (DP 6,04) no estado mental e média de 5,03 (DP 4,11) no estado manifestações somáticas. As médias para o instrumento SF-36 foram por ordem de classificação: limitação por aspectos físicos 47,3 (DP 42,9), vitalidade 57,4 (DP 19,7), limitação por aspectos emocionais 58 (DP 44,7), capacidade funcional 58,7 (DP 27,8), dor 60,4 (DP 26,3), estado geral de saúde 60,7 (DP 22,7), saúde mental 66,8 (DP 22,1) e aspectos sociais 78 (DP 26,1). RESULTADOS: O MINICHAL apresentou correlação significativa (p < 0,001) com o SF-36 em todos os domínios. CONCLUSÃO: O MINICHAL provou ser um instrumento útil na avaliação da QVRS em pacientes hipertensos. (Arq Bras Cardiol. 2012; [online].ahead print, PP.0-0)

Der Begriff der Aufarbeitung beschreibt die verfahrenstechnischen Grundoperationen zur Isolierung des im Fermenter enthaltenen Wertproduktes. Bei der mikrobiellen Herstellung verschiedenster Produkte ist die Aufarbeitung aber der kosten- und arbeitsintensivste Faktor, so dass Optimierungen hinsichtlich der Effizienz wichtig sind. Die vorliegende Arbeit wurde im Helmholtz-Zentrum für Infektionsforschung angefertigt. Die Arbeitsgruppe Mikrobielle Wirkstoffe beschäftigt sich mit der Suche nach neuen antibiotisch wirkenden Sekundärmetaboliten aus Bodenbakterien und filamentösen Pilzen. Die Feststoff- Flüssig- Extraktion wird dabei zur Isolierung der Sekundärmetabolite genutzt. Das Myzel wird standardmäßig im Labormaßstab mit einer 30-minütigen Extraktion im Ultraschallbad behandelt. Die durchgeführten Versuche zeigen jedoch, dass andere Methoden des Leistungseintrages weniger Zeit beanspruchen, höhere Substanzausbeuten hervorbringen können und geringere Lösungsmittelvolumen zur Extraktion der Sekundärmetabolite genutzt werden können. Doch neben den genannten Vorteilen zeigte sich auch, dass der Verschmutzungsgrad mit intrazellulären Substanzen durch den mechanischen Energieeintrag zunehmen kann. (...) Neben der Feststoff- Flüssig- Extraktion des Myzels spielt auch die Extraktion eines standardmäßig eingesetzten Adsorberharzes XAD16 in der Arbeitsgruppe MWIS eine große Rolle. Dies wird zur Adsorption der Sekundärmetabolite aus der Fermentationsbrühe oder Überstand genutzt, nach der Fermentation mittels Filtration abgeerntet und anschließend extrahiert. Für diesen Schritt wurden verschiedene Downstreammethoden mit unterschiedlichen Lösungsmittelvolumen und Extraktionszeiten hinsichtlich der Substanzausbeute untersucht. (...)

In der vorliegenden Arbeit werden Untersuchungen zum Einfluss von Licht auf humane in vitro-Zellkulturen beschrieben. Zunächst wird das Licht technisch erläutert und das Farbspektrum des Lichts betrachtet. Das natürliche Farbspektrum der Sonne ändert sich über den Tag, des Weiteren werden die Farben des Lichts in der Wissenschaft in verschiedenen Bereichen verwendet. Der Mensch hat einen geregelten Tagesablauf, der mit dem Tagesverlauf der Sonne parallel abläuft. Durch verschiedene Einflüsse kann diese Parallelität gestört werden. Für die Untersuchungen zum Einfluss des Lichts werden in vitro Modellsysteme verwendet, insbesondere Keratinozyten, die genauer beschrieben werden. Außerdem werden für die Untersuchungen organische Leuchtdioden (kurz: OLED) verwendet. Es wird eine Auswahl an Versuchs-OLEDs getroffen, welche für erste Versuche verwendet werden. In dieser Arbeit sollen erste Erkenntnisse über die Auswirkungen von Licht auf humane Zellen ermittelt werden, um aus der großen Auswahl von OLEDs Farbspektren zur genaueren Betrachtung herauszuziehen und später eine Anwendung in der Medizin zu ermöglichen. Hierzu werden das Wachstum und der Stoffwechselumsatz der Zellen betrachtet. Die Zellen werden mit Hilfe eines Versuchsaufbaus aus einer vorangegangenen Praktikumsarbeit beleuchtet. Als Lichtspektrum wurden 5 Farben ausgewählt: blau, grün, orange, warmweiß und kaltweiß. Als weitere Parameter werden eine feste Beleuchtungszeit, sowie eine identische Leuchtdichte bei allen Farben festgelegt. Aus den Versuchen können erste Erkenntnisse gezogen werden, jedoch müssen diese durch weitere Versuche bestätigt bzw. genauer betrachtet werden.
