Whole-body coverage using MRI was developed almost 2 decades ago. The first applications focused on the investigation of the skeleton to detect neoplastic disease, mainly metastases from solid cancers, and involvement by multiple myeloma and lymphoma. But the extensive coverage of the whole musculoskeletal system, combined with the exquisite sensitivity of MRI to tissue alteration in relation to different pathologic conditions, mainly inflammation, has led to the identification of a growing number of indications outside oncology. Seronegative rheumatisms, systemic sclerosis, inflammatory diseases involving muscles or fascias, and multifocal osseous, vascular, or neurologic diseases represent currently validated or emerging indications of whole-body MRI (WB-MRI). We first illustrate the most valuable indications of WB-MRI in seronegative rheumatisms that include providing significant diagnostic information in patients with negative or ambiguous MRI of the sacroiliac joints and the lumbar spine, assessing disease activity in advanced (ankylosed) central disease, and evaluating the peripherally dominant forms of spondyloarthropathy. Then we review the increasing indications of WB-MRI in other rheumatologic and nonneoplastic disorders, underline the clinical needs, and illustrate the role of WB-MRI in the positive diagnosis and evaluation of disease burden, therapeutic decisions, and treatment monitoring.

BACKGROUND: Exercise prevents the adverse effects of a high-fructose diet through mechanisms that remain unknown. OBJECTIVE: We assessed the hypothesis that exercise prevents fructose-induced increases in very-low-density lipoprotein (VLDL) triglycerides by decreasing the fructose conversion into glucose and VLDL-triglyceride and fructose carbon storage into hepatic glycogen and lipids. DESIGN: Eight healthy men were studied on 3 occasions after 4 d consuming a weight-maintenance, high-fructose diet. On the fifth day, the men ingested an oral (13)C-labeled fructose load (0.75 g/kg), and their total fructose oxidation ((13)CO2 production), fructose storage (fructose ingestion minus (13)C-fructose oxidation), fructose conversion into blood (13)C glucose (gluconeogenesis from fructose), blood VLDL-(13)C palmitate (a marker of hepatic de novo lipogenesis), and lactate concentrations were monitored over 7 postprandial h. On one occasion, participants remained lying down throughout the experiment [fructose treatment alone with no exercise condition (NoEx)], and on the other 2 occasions, they performed a 60-min exercise either 75 min before fructose ingestion [exercise, then fructose condition (ExFru)] or 90 min after fructose ingestion [fructose, then exercise condition (FruEx)]. RESULTS: Fructose oxidation was significantly (P < 0.001) higher in the FruEx (80% ± 3% of ingested fructose) than in the ExFru (46% ± 1%) and NoEx (49% ± 1%). Consequently, fructose storage was lower in the FruEx than in the other 2 conditions (P < 0.001). Fructose conversion into blood (13)C glucose, VLDL-(13)C palmitate, and postprandial plasma lactate concentrations was not significantly different between conditions. CONCLUSIONS: Compared with sedentary conditions, exercise performed immediately after fructose ingestion increases fructose oxidation and decreases fructose storage. In contrast, exercise performed before fructose ingestion does not significantly alter fructose oxidation and storage. In both conditions, exercise did not abolish fructose conversion into glucose or its incorporation into VLDL triglycerides. This trial was registered at clinicaltrials.gov as NCT01866215.

Insulin resistance is a fundamental pathogenic factor that characterizes various metabolic disorders, including obesity and type 2 diabetes. Adipose tissue contributes to the development of obesity-related insulin resistance through increased release of fatty acids, altered adipokine secretion, and/or macrophage infiltration and cytokine release. Here, we aimed to analyze the participation of the cyclin-dependent kinase 4 (CDK4) in adipose tissue biology. We determined that white adipose tissue (WAT) from CDK4-deficient mice exhibits impaired lipogenesis and increased lipolysis. Conversely, lipolysis was decreased and lipogenesis was increased in mice expressing a mutant hyperactive form of CDK4 (CDK4R24C). A global kinome analysis of CDK4-deficient mice following insulin stimulation revealed that insulin signaling is impaired in these animals. We determined that insulin activates the CCND3-CDK4 complex, which in turn phosphorylates insulin receptor substrate 2 (IRS2) at serine 388, thereby creating a positive feedback loop that maintains adipocyte insulin signaling. Furthermore, we found that CCND3 expression and IRS2 serine 388 phosphorylation are increased in human obese subjects. Together, our results demonstrate that CDK4 is a major regulator of insulin signaling in WAT.

The signalling function of melanin-based colouration is debated. Sexual selection theory states that ornaments should be costly to produce, maintain, wear or display to signal quality honestly to potential mates or competitors. An increasing number of studies supports the hypothesis that the degree of melanism covaries with aspects of body condition (e.g. body mass or immunity), which has contributed to change the initial perception that melanin-based colour ornaments entail no costs. Indeed, the expression of many (but not all) melanin-based colour traits is weakly sensitive to the environment but strongly heritable suggesting that these colour traits are relatively cheap to produce and maintain, thus raising the question of how such colour traits could signal quality honestly. Here I review the production, maintenance and wearing/displaying costs that can generate a correlation between melanin-based colouration and body condition, and consider other evolutionary mechanisms that can also lead to covariation between colour and body condition. Because genes controlling melanic traits can affect numerous phenotypic traits, pleiotropy could also explain a linkage between body condition and colouration. Pleiotropy may result in differently coloured individuals signalling different aspects of quality that are maintained by frequency-dependent selection or local adaptation. Colouration may therefore not signal absolute quality to potential mates or competitors (e.g. dark males may not achieve a higher fitness than pale males); otherwise genetic variation would be rapidly depleted by directional selection. As a consequence, selection on heritable melanin-based colouration may not always be directional, but mate choice may be conditional to environmental conditions (i.e. context-dependent sexual selection). Despite the interest of evolutionary biologists in the adaptive value of melanin-based colouration, its actual role in sexual selection is still poorly understood.

The nutrient load to the Gulf of Finland has started to increase as a result of the strong economic recovery in agriculture and livestock farming in the Leningrad region. Also sludge produced from municipal wastewater treatment plant of the Leningrad region causes the great impact on the environment, but still the main options for its treatment is disposal on the sludge beds or Landfills. The aim of this study was to evaluate the implementation of possible joint treatment methods of manure form livestock and poultry enterprises and sewage sludge produced from municipal wastewater treatment plants in the Leningrad region. The study is based on published data. The most attention was put on the anaerobic digestion and incineration methods. The manure and sewage sludge generation for the whole Leningrad region and energy potential produced from their treatment were estimated. The calculations showed that total amount of sewage sludge generation is 1 348 000 t/a calculated on wet matter and manure generation is 3 445 000 t/a calculated on wet matter. The potential heat release from anaerobic digestion process and incineration process is 4 880 000 GJ/a and 5 950 000 GJ/a, respectively. Furthermore, the work gives the overview of the general Russian and Finnish legislation concerning manure and sewage sludge treatment. In the Gatchina district it was chosen the WWTP and livestock and poultry enterprises for evaluation of the centralized treatment plant implementation based on anaerobic digestion and incineration methods. The electricity and heat power of plant based on biogas combustion process is 4.3 MW and 7.8 MW, respectively. The electricity and heat power of plant based on manure and sewage sludge incineration process is 3.0 MW and 6.1 MW, respectively.

ABSTRACT In experiments conducted in a growth chamber, the chronological time and the accumulated degree-days were determined for the duration of incubation, latent and infectious periods of Phakopsora pachyrhizi cultivars BRSGO 7560 and BRS 246 RR. Detached soybean leaflets were placed in gerbox-type acrylic boxes and inoculated with 20 x 103 uredospores/mL. The study was conducted at 12-h photoperiod and temperatures of 10ºC, 15ºC, 22ºC, 25ºC and 30°C for 30 days. Lesions and uredia/cm2were evaluated and the number of uredia per lesion was quantified after the beginning of sporulation. The sporulation potential was also quantified for cultivars BRSGO 7560 and BRS 246 RR. The steps of the infection process can be quantified based on both the chronological time and the accumulated heat. The cultivar BRSGO 7560 produced 4,012.8 spores/cm2 and BRS 246 RR, 7,348.4 uredospores/cm2. The largest number of uredia was produced at 25ºC in both cultivars; however, BRS 246 RR presented 372.7 uredia/cm2 and BRSGO 7560, 231.6 uredia/cm2. At 10ºC and 30°C, leaf infection did not occur in both cultivars.

Este trabalho apresenta os resultados da avaliação qualitativa de impactos ambientais na etapa de implantação florestal, considerando-se as atividades de limpeza e preparo do terreno, por meio da aplicação dos métodos de matriz de identificação e interação de impactos, da lista descritiva (checklist) e da combinação/ponderação de atributos. Tal avaliação foi feita com base na identificação e caracterização qualitativa dos impactos ambientais, cujas metodologias possibilitaram a visualização rápida e hierarquizada dos impactos. Através dessa matriz, evidenciaram-se 348 possíveis relações de impactos (capacidade total da matriz), das quais 304 (87,6%) foram identificadas, em que 209 impactos (60,05%) recaíram sobre o meio biofísico e 95 (27,31%) sobre o meio socioeconômico. Dos 209 impactos no meio biofísico, todos apresentaram valor negativo, evidenciando-se ser esse o compartimento ambiental mais impactado negativamente quando da realização das atividades de limpeza e preparo do terreno. Já, dos 95 impactos no meio socioeconômico, 72 (75,78%) foram de valor positivo e 23 (24,21%) de valor negativo, demonstrando que o meio socioeconômico é o compartimento ambiental mais afetado positivamente por ocasião da realização das atividades. Foram identificados os impactos ambientais positivos e negativos mais significantes que, com a combinação/ponderação de atributos, mostraram ser, em sua maioria, de significância média local e regional. Todos os impactos evidenciados apresentaram temporalidade no curto prazo, evidenciando estarem sobre o alcance quanto ao diagnóstico e gestão ambiental adequados. Os resultados contribuem e reafirmam a necessidade de se instituir o processo de avaliação de impactos ambientais na rotina técnico-administrativa da empresa como instrumento de gestão ambiental.

O presente trabalho apresenta o desenvolvimento e calibração de tensiômetro de leitura automática, sendo o tensiômetro de mercúrio utilizado como padrão de comparação. Os ensaios foram conduzidos no Laboratório de Mecânica e Eletrônica do Departamento de Engenharia Agrícola da UFC. Esse equipamento difere do tensiômetro tradicional por substituir o manômetro de mercúrio por sensor de pressão. Tal dispositivo gera uma saída com valor de até no máximo 4,5 volts. O tensiômetro eletrônico é conectado a uma placa de aquisição de dados (DAQ) contendo um microprocessador, conversor analógico/digital (ADC) e saída serial, sendo tal placa ligada a um microcomputador. A bancada de ensaio constituiu-se de três caixas plásticas preenchidas com solo de textura franco- arenosa. Os dados foram coletados durante um mês e as equações geradas foram do tipo Count = offset + apsim, em que psim representa o potencial matricial fornecido pelo tensiômetro de mercúrio (kPa) e count a saída digial do ADC, com valores adimensionais de 0 a 4.095. Foram obtidos os valores máximo e mínimo de offset de 348,572 e 261,026, e de coeficiente angular de 36,675 kPa-1 e 34,421 kPa-1. Os resultados da regressão linear simples indicaram a existência de regressão a menos de 0,1% de significância e valores de coeficientes de correlação nunca inferiores a 0,9994.

Neste trabalho, avaliou-se o desempenho de dois reatores anaeróbios de fluxo ascendente com manta de lodo (UASB), em série, seguidos de um reator operado em batelada sequencial (RBS) com etapa aeróbia, no tratamento de águas residuárias de suinocultura. O sistema de tratamento anaeróbio em dois estágios foi alimentado com águas residuárias de suinocultura com concentrações médias de sólidos suspensos totais (SST), de 4.427 a 16.425 mg L-1 . As cargas orgânicas volumétricas (COV) aplicadas no reator UASB do primeiro estágio variaram de 14,8 a 24,4 g DQO (L d)-1. Os tempos de detenção hidráulica (TDH) foram de 28 e 11 h e de 14 e 6 h no primeiro e segundo reatores UASB, respectivamente. O RBS foi operado com 1 e 2 ciclos diários de alimentação e com concentrações de SST do afluente, de 1.348 a 2.036 mg L-1 . As maiores eficiências de remoção de DQOtotal ocorreram com os maiores TDH, com valores médios de 78 a 88% nos reatores UASB, em dois estágios. Com o tratamento do efluente dos reatores UASB no RBS, as eficiências médias de remoção aumentaram para 93 a 97%, 92 a 98%, 57 a 78%, 71 a 88% e 68 a 85% para a DQO total, SST, P-total, nitrogênio total Kjeldahl (NTK) e nitrogênio total (NT), respectivamente. Para os coliformes termotolerantes, as remoções foram de 93,80 a 99,99%.
