Overactivation of ionotropic glutamate receptors in oligodendrocytes induces cytosolic Ca2+ overload and excitotoxic death, a process that contributes to demyelination and multiple sclerosis. Excitotoxic insults cause well-characterized mitochondrial alterations and endoplasmic reticulum (ER) dysfunction, which is not fully understood. In this study, we analyzed the contribution of ER-Ca2+ release through ryanodine receptors (RyRs) and inositol triphosphate receptors (IP(3)Rs) to excitotoxicity in oligodendrocytes in vitro. First, we observed that oligodendrocytes express all previously characterized RyRs and IP(3)Rs. Blockade of Ca2+-induced Ca2+ release by TMB-8 following alpha-amino-3-hydroxyl-5-methyl-4-isoxazole-propionate (AMPA) receptor-mediated insults attenuated both oligodendrocyte death and cytosolic Ca2+ overload. In turn, RyR inhibition by ryanodine reduced as well the Ca2+ overload whereas IP3R inhibition was ineffective. Furthermore, AMPA-triggered mitochondrial membrane depolarization, oxidative stress and activation of caspase-3, which in all instances was diminished by RyR inhibition. In addition, we observed that AMPA induced an ER stress response as revealed by alpha subunit of the eukaryotic initiation factor 2 alpha phosphorylation, overexpression of GRP chaperones and RyR-dependent cleavage of caspase-12. Finally, attenuating ER stress with salubrinal protected oligodendrocytes from AMPA excitotoxicity. Together, these results show that Ca2+ release through RyRs contributes to cytosolic Ca2+ overload, mitochondrial dysfunction, ER stress and cell death following AMPA receptor-mediated excitotoxicity in oligodendrocytes. Abstract. The atomic motion is coupled by the fast and slow components due to the high frequency vibration of atoms and the low frequency deformation of atomic lattice, respectively. A two-step approximate method was presented to determine the atomic slow motion. The first step is based on the change of the location of the cold potential well bottom and the second step is based on the average of the appropriate slow velocities of the surrounding atoms. The simple tensions of one-dimensional atoms and two-dimensional atoms were performed with the full molecular dynamics simulations. The conjugate gradient method was employed to determine the corresponding location of cold potential well bottom. Results show that our two-step approximate method is appropriate to determine the atomic slow motion under the low strain rate loading. This splitting method may be helpful to develop more efficient molecular modeling methods and simulations pertinent to realistic loading conditions of materials.

Many particles proposed by theories, such as GUT monopoles, nuclearites and 1/5 charge superstring particles, can be categorized as Slow-moving, Ionizing, Massive Particles (SIMPs).

Detailed calculations of the signal-to-noise ratios in vanous acoustic and mechanical methods for detecting such SIMPs are presented. It is shown that the previous belief that such methods are intrinsically prohibited by the thermal noise is incorrect, and that ways to solve the thermal noise problem are already within the reach of today's technology. In fact, many running and finished gravitational wave detection ( GWD) experiments are already sensitive to certain SIMPs. As an example, a published GWD result is used to obtain a flux limit for nuclearites.

The result of a search using a scintillator array on Earth's surface is reported. A flux limit of 4.7 x 10^(-12) cm^(-2)sr^(-1)s^(-1) (90% c.l.) is set for any SIMP with 2.7 x 10^(-4) less than β less than 5 x 10^(-3) and ionization greater than 1/3 of minimum ionizing muons. Although this limit is above the limits from underground experiments for typical supermassive particles (10^(16)GeV), it is a new limit in certain β and ionization regions for less massive ones (~10^9 GeV) not able to penetrate deep underground, and implies a stringent limit on the fraction of the dark matter that can be composed of massive electrically and/ or magnetically charged particles.

The prospect of the future SIMP search in the MACRO detector is discussed. The special problem of SIMP trigger is examined and a circuit proposed, which may solve most of the problems of the previous ones proposed or used by others and may even enable MACRO to detect certain SIMP species with β as low as the orbital velocity around the earth.

An internally circulating fluidized bed (ICFB) was applied to investigate the behavior of chlorine and sulfur during cofiring RDF and coal. The pollutant emissions in the flue gas were measured by Fourier transform infrared (FTIR) spectrometry (Gasmet DX-3000). In the tests, the concentrations of the species CO, CO2, HCl, and SO2 were measured online. Results indicated when cofiring RDF and char, due to the higher content of chlorine in RDF, the formation of HCl significantly increases. The concentration Of SO2 is relatively low because alkaline metal in the fuel ash can absorb SO2. The concentration of CO emission during firing pure RDF is relatively higher and fluctuates sharply. With the CaO addition, the sulfur absorption by calcium quickly increases, and the desulfuration ratio is bigger than the dechlorination ratio. The chemical equilibrium method is applied to predict the behavior of chlorine. Results show that gaseous HCl emission increases with increasing RDF fraction, and gaseous KCl and NaCl formation might occur.

Gold Coast Water is responsible for the management of the water and wastewater assets of the City of the Gold Coast on Australia’s east coast. Treated wastewater is released at the Gold Coast Seaway on an outgoing tide in order for the plume to be dispersed before the tide changes and renters the Broadwater estuary. Rapid population growth over the past decade has placed increasing demands on the receiving waters for the release of the City’s effluent. The Seaway SmartRelease Project is designed to optimise the release of the effluent from the City’s main wastewater treatment plant in order to minimise the impact of the estuarine water quality and maximise the cost efficiency of pumping. In order to do this an optimisation study that involves water quality monitoring, numerical modelling and a web based decision support system was conducted. An intensive monitoring campaign provided information on water levels, currents, winds, waves, nutrients and bacterial levels within the Broadwater. These data were then used to calibrate and verify numerical models using the MIKE by DHI suite of software. The decision support system then collects continually measured data such as water levels, interacts with the WWTP SCADA system, runs the models in forecast mode and provides the optimal time window to release the required amount of effluent from the WWTP. The City’s increasing population means that the length of time available for releasing the water with minimal impact may be exceeded within 5 years. Optimising the release of the treated water through monitoring, modelling and a decision support system has been an effective way of demonstrating the limited environmental impact of the expected short term increase in effluent disposal procedures. Part I

Studies of vibrational relaxation in excited electronic states of simple diatomic molecules trapped in solid rare-gas matrices at low temperatures are reported. The relaxation is investigated by monitoring the emission intensity from vibrational levels of the excited electronic state to vibrational levels of the ground electronic state. The emission was in all cases excited by bombardment of the doped rare-gas solid with X-rays.

The diatomics studied and the band systems seen are: N2, Vegard-Kaplan and Second Positive systems; O2, Herzberg system; OH and OD, A 2Σ+ - X2IIi system. The latter has been investigated only in solid Ne, where both emission and absorption spectra were recorded; observed fine structure has been partly interpreted in terms of slightly perturbed rotational motion in the solid. For N2, OH, and OD emission occurred from v' > 0, establishing a vibrational relaxation time in the excited electronic state of the order, of longer than, the electronic radiative lifetime. The relative emission intensity and decay times for different v' progressions in the Vegard-Kaplan system are found to depend on the rare-gas host and the N2 concentration, but are independent of temperature in the range 1.7°K to 30°K.

Part II

Static crystal field effects on the absorption, fluorescence, and phosphorescence spectra of isotopically mixed benzene crystals were investigated. Evidence is presented which demonstrate that in the crystal the ground, lowest excited singlet, and lowest triplet states of the guest deviate from hexagonal symmetry. The deviation appears largest in the lowest triplet state and may be due to an intrinsic instability of the 3B1u state. High resolution absorption and phospho- rescence spectra are reported and analyzed in terms of site-splitting of degenerate vibrations and orientational effects. The guest phosphorescence lifetime for various benzene isotopes in C6D6 and sym-C6H3D3 hosts is presented and discussed.

We describe a method to generate an ultra-slow atomic beam by velocity selective resonance (VSR). A VSR experiment on a metastable helium beam in a magnetic field is presented and the results show that the transverse velocity of the defected beam can be cooled and precisely controlled to less than the recoil velocity, depending on the magnitude of the magnetic field. We extend this idea to a cold atomic cloud to produce an ultra-slow Rb-87 beam that can be used as a source of an atomic fountain clock or a space clock.

While synoptic surveys in the optical and at high energies have revealed a rich discovery phase space of slow transients, a similar yield is still awaited in the radio. Majority of the past blind surveys, carried out with radio interferometers, have suffered from a low yield of slow transients, ambiguous transient classifications, and contamination by false positives. The newly-refurbished Karl G. Jansky Array (Jansky VLA) offers wider bandwidths for accurate RFI excision as well as substantially-improved sensitivity and survey speed compared with the old VLA. The Jansky VLA thus eliminates the pitfalls of interferometric transient search by facilitating sensitive, wide-field, and near-real-time radio surveys and enabling a systematic exploration of the dynamic radio sky. This thesis aims at carrying out blind Jansky VLA surveys for characterizing the radio variable and transient sources at frequencies of a few GHz and on timescales between days and years. Through joint radio and optical surveys, the thesis addresses outstanding questions pertaining to the rates of slow radio transients (e.g. radio supernovae, tidal disruption events, binary neutron star mergers, stellar flares, etc.), the false-positive foreground relevant for the radio and optical counterpart searches of gravitational wave sources, and the beaming factor of gamma-ray bursts. The need for rapid processing of the Jansky VLA data and near-real-time radio transient search has enabled the development of state-of-the-art software infrastructure. This thesis has successfully demonstrated the Jansky VLA as a powerful transient search instrument, and it serves as a pathfinder for the transient surveys planned for the SKA-mid pathfinder facilities, viz. ASKAP, MeerKAT, and WSRT/Apertif.

Hairpin pyrrole-imdazole polyamides are cell-permeable, sequence-programmable oligomers that bind in the minor groove of DNA. This thesis describes studies of Py-Im polyamides targeted to biologically important DNA repeat sequences for the purpose of modulating disease states. Design of a hairpin polyamide that binds the CG dyad, a site of DNA methylation that can become dysregulated in cancer, is described. We report the synthesis of a DNA methylation antagonist, its sequence specificity and affinity informed by Bind-n-Seq and iteratively designed, which improves inhibitory activity in a cell-free assay by 1000-fold to low nanomolar IC50. Additionally, a hairpin polyamide targeted to the telomeric sequence is found to trigger a slow necrotic-type cell death with the release of inflammatory molecules in a model of B cell lymphoma. The effects of the polyamide are unique in this class of oligomers; its effects are characterized and a functional assay of phagocytosis by macrophages is described. Additionally, hairpin polyamides targeted to pathologically expanded CTG•CAG triplet repeat DNA sequences, the molecular cause of myotonic dystrophy type 1, are synthesized and assessed for toxicity. Lastly, ChIP-seq of Hypoxia-Inducible Factor is performed under hypoxia-induced conditions. The study results show that ChIP-seq can be employed to understand the genome-wide perturbation of Hypoxia-Inducible Factor occupancy by a Py-Im polyamide.

This report covers the period April to September, 1989. During this period sampling of invertebrates has concentrated on planktonic animals and those associated with one of the dominant macrophytes in the system, Nuphar lutea, the yellow water lily, since these are particularly important in the diets of larval and juvenile cyprinid fish. A proportion of samples has been partly analysed and some preliminary data are presented here.

The River Great Ouse is a highly managed large lowland river in eastern England. It drains rich arable land in the Midlands and Eastern England and over the years nutrient concentrations have increased and there is a general perception that the clarity of the water has decreased. The main river channels have been dredged a number of times partly for flood control reasons but also for recreational boating and navigation activities. The period covered by this first report has been used to develop specific methodology and instrumentation for measuring turbidity, suspended solids and underwater irradiance for conditions found in the middle abd lower reaches of the River Great Ouse. Sampling strategies have been developed and an extensive sampling programme is now underway covering phytoplankton, suspended solids and turbidity in relation to algal epiphyte growth on underwater macrophytes. Preliminary data are presented relating light levels on the river bed to the river bed profile, turbidity levels and phytoplankton chlorophyll a concentrations. Studies are underway concerning the extent of macrophyte cover and periphyton densities.

Genetic engineering now makes possible the insertion of DNA from many organisms into other prokaryotic, eukaryotic and viral hosts. This technology has been used to construct a variety of such genetically engineered microorganisms (GEMs). The possibility of accidental or deliberate release of GEMs into the natural environment has recently raised much public concern. The prospect of deliberate release of these microorganisms has prompted an increased need to understand the processes of survival, expression, transfer and rearrangement of recombinant DNA molecules in microbial communities. The methodology which is being developed to investigate these processes will greatly enhance our ability to study microbial population ecology.
