Nas últimas décadas, muito interesse tem sido focado no potencial biotecnológico das microalgas, principalmente devido à identificação de diversas substâncias bioativas sintetizadas por estas, especialmente ácidos graxos poliinsaturados (PUFAs). PUFAs ômega-3 (PUFAs ω-3) têm sido estudados pela promoção de efeitos benéficos em muitas doenças crônicas, incluindo o câncer, ao contrário de PUFAs ômega-6 (PUFAs ω-6) que, de forma geral, são conhecidos por agravar o desenvolvimento destes quadros. Este estudo objetivou testar a atividade antiproliferativa de extratos lipídicos de duas microalgas marinhas, Isochrysis galbana e Thalassiosira pseudonana, ricas principalmente em PUFAs ω-3, em uma linhagem celular de melanoma, B16F10. Adicionalmente, o efeito dos precursores dos PUFAs ω-3, α-linolênico (ALA), e dos PUFAs ω-6, linoleico (LA) na proliferação celular, foi avaliado nesta linhagem e em uma linhagem melanocítica normal, Melan-A, pelo método MTT. Foi demonstrado que os extratos lipídicos estudados são capazes de induzir inibição de proliferação tempo e concentração dependente na linhagem B16F10 e esta atividade foi também exercida pelo ALA de forma independente das concentrações testadas. Além disso, o extrato lipídico da microalga Thalassiosira pseudonana também induziu citotoxicidade na linhagem B16F10. Desse modo, é possível sugerir que o efeito antiproliferativo dos extratos possa estar relacionado com a alta concentração de ω-3 PUFAs em relação a ω- 6 PUFAs, especialmente para a Thalassiosira pseudonana Adicionalmente, a linhagem Melan-A não foi sensível ao tratamento com o ALA, sugerindo, dessa forma, um efeito antitumoral para este composto. Este estudo ainda indica a possibilidade do uso de microalgas como fontes promissoras de substâncias antitumorais.

Os nanomateriais de carbono como o fulereno (C60) apresenta comportamentos bioquímicos distintos, podendo atuar como antioxidante ou pró-oxidante em diferentes sistemas biológicos. Outra evidência ao C60 refere-se a sua característica lipofilica, na qual oferece ação mais direta a diferentes tipos de membranas celulares. Do mesmo modo ácidos graxos poliinsaturados (AGPs) como o ômega-3 (DHA) e o ômega-6 (LA) são importantes para funções celulares da membrana, sendo considerados antioxidantes clássicos. Dessa forma este estudo avaliou em suspensões celulares de cérebro da carpa (Cyprinus carpio, Cyprinidae), o efeito de C60 após um pré-tratamento com DHA ou LA. Para tal avaliação os ensaios consistiram em um pré-tratamento com AGPs (48h) e após exposição a C60 (2h). Como resultados observamos que a viabilidade celular e a capacidade antioxidante total não apresentaram diferença (p> 0.05) entre todos os grupos. Em relação a valores de espécies ativas de oxigênio e dano lipídico foi observado redução nos seus valores nos grupos expostos ao C60 pré – tratados com AGPs (p<0.05). Em termos de cisteína, ocorre uma redução da sua concentração em todos os grupos expostos ao C60. Porém para glutationa a exposição ao C60 provoca um aumento de sua concentração nos grupo controle (sem AGPs) e no grupo pré – tratado com DHA. Dessa forma consideramos que o pré – tratamento com AGPs é benéfico às células, uma vez que um aumento nos níveis de glutationa e uma diminuição na concentração de espécies ativas de oxigênio e peroxidação lipídica foram observados nos grupos expostos ao C60. Sendo assim um bom estado nutritivo em termos da concentração de AGPs foi considerado benéfico na exposição ao fulereno.

A atividade antiproliferativa in vitro de uma série de amidas graxas sintéticas, em sete linhagens de células tumorais foi investigada. Baseado em GI50, TGI e LC50, os ensaios preliminares mostraram que a maior parte dos compostos mostrou atividade antiproliferativa moderada a boa contra as linhagem de células tumorais testadas, principalmente em células de glioma humano (U251) e câncer de ovário humano com fenótipo de resistentencia a múltiplos fármacos (NCI-ADR/RES). A amida (R,S)-3d, derivada do ácido ricinoleico, mostrou uma elevada seletividade com potência de inibição do crescimento e morte celular para a linhagem de células de glioma. Além disso, as amidas (S)-3c e (S)-3e, derivadas dos ácidos oleico e linoleico respectivamente, foram especificas para glioma e ovário com fenótipo de resistência a múltiplos fármacos com inibição potente do crescimento celular. Estes resultados aliados a um perfil de segurança relativo quando analisado o efeito sobre as linhagens celulares não–tumorais, apontam para que estes compostos sirvam como modelos para o desenvolvimento de candidatos a fármacos para o tratamento de câncer, incluindo cânceres com fenótipo de resistência a múltiplos fármacos.

Neste trabalho desenvolveu-se a síntese de -cetoésteres, a partir da acilação do ácido de Meldrum usando diferentes ácidos graxos, com cadeias saturadas e insaturadas. Inicialmente, foi realizado o experimento utilizando o ácido palmítico (1e) como modelo para obtenção de cloreto de ácido graxo, usando ácido de Meldrum e piridina como catalisador. Porém, após um longo tempo de reação, o -cetoéster palmítico (4e) foi obtido com baixo rendimento, não ultrapassando os 20%. Posteriormente, os experimentos foram realizados utilizando o ácidos graxos 1e, ácido de Meldrum (2), DCC, DMAP, e piridina à temperatura ambiente sob a atmosfera de nitrogênio. No entanto, o protocolo utilizado resultou no β-cetoéster 4e com rendimentos moderados. Em seguida examinou-se o efeito da adição do ácido de Meldrum após a adição dos ácido graxos e DCC. Neste caso, após a adição do DCC aos ácido graxos 1a-j foi observada a formação imediata das O-aciluréias graxas. A seguir a adição de 2,0 equiv de ácido de Meldrum (2) em diclorometano, revelou a formação dos respectivos enol graxos 3a-j. Posteriormente, os β- cetoésteres 4a-j foram obtidos a partir da reação do enol com o metanol. Este protocolo modificado proporcionou o aumento nos rendimentos (74 a 84%) dos β-cetoéster 4a-j. Após, o β-cetoéster 4k foi sintetizado em 75% utilizando o ácido de Meldrum (2) e ácido ricinoléico (1k), obtido a partir do biodiesel de mamona, à temperatura ambiente e sob a atmosfera de nitrogênio. Portanto, foi desenvolvido um método simples para obter β-cetoésteres graxos, a partir do ácido de Meldrum com cadeias graxas diversificadas utilizando DCC e DMAP. Além disso, o presente trabalho relata pela primeira vez a síntese de novos β-cetoésteres graxos derivados dos ácidos oléico, elaídico, ricinoléico, linoléico e linolênico.

Vaccinum myrtillus L. belongs to Ericaceae family, being commonly known for its sweet small fruits: the blueberries. Widely consumed in fresh, these fruits are also used in jams and marmalades due to their digestive and hypoglycemic properties and also due to the presence of several bioactive compounds [!]. Therefore, it has become a very appealing matrix in the development of functional products that, beyond their nutritional properties, will add a long-term beneficial physiological/health effect [2]. In the present work, three novel blueberry based products developed by RBR Foods Company (Portugal), were characterized in terms of their nutritional and chemical properties: carbohydrates, ash, proteins, fat and energetic value (following official methods of food analysis), fatty acids profile (by CG-FID), soluble sugars (by HPLCRI), organic acids (by HPLC-DAD) and tocopherols (by HPLC-fluorescence). The products result from a mixture of the fruits with rose petals (PI), marigold petals (P2) and apple and goji berries (P3). The blueberry fruits were used as control sample. The nutritional profile of the novel products was very similar to the control sample: the carbohydrates were the most abundant macronutrient, followed by proteins and total fat. Regarding sugars, fructose, glucose and sucrose were identified in all the samples. P 1 and P2 didn't show significant differences in comparison to the control, however, P3 revealed a lower concentration of sugars. In terms of fatty acids composition, all the studied samples presented higher contents in polyunsaturated fatty acids, especially due to the contribution of linoleic and alinolenic acids. The results of tocopherols revealed that the control sample only presented two isoforrns of tocopherols, a- and y-tocopherol, being the same observed in P3. However, P 1 revealed the presence of all the isoforrns of tocopherols, while P2 was lacking otocopherol; which is related with the contribution of rose and marigold petals, respectively. The a-tocopherol isoforrn was the most abundant in all the studied samples. Overall, this work contributed to the nutritional characterization of novel blueberry based products and is a part of a wider project that aims the detailed study of these products, namely their potential to be used as functional foods.

Buckler sorrel (Rumex induratus Boiss. & Reut.) is an underutilized leafy vegetable with peculiar sensory properties and potential as a gourmet food. In the food industry, different packaging methods have been used for shelf-life extension, but it is important to know how the quality of minimally processed vegetable is affected by these treatments. Recently, nitrogen and argon have been used for food packaging. Nitrogen is low soluble in water and other food constituents and does not support the growth of aerobic microbes. In turn, argon is biochemically active and appears to interfere with enzymatic oxygen receptor sites. In this study, modified atmospheres enriched with nitrogen and argon were evaluated for shelf-life extension of buckler sorrel leaves. Wild samples were gathered in Bragança, Portugal, considering local consumers’ sites and criteria. Healthy and undamaged leaves were selected, rinsed in tap water, and a portion was immediately analyzed (non-stored control). The remaining fresh material was packaged in polyethylene bags under nitrogen- and argon-enriched atmospheres and a conventional control atmosphere (air). All packaged samples were stored at 4 ºC for 12 days and then analyzed. The headspace gas composition was monitored during storage. Different quality attributes were evaluated, including visual (colour), nutritional (macronutrients, individual sugars and fatty acids) and bioactive (hydrophilic and lipophilic molecules and antioxidant properties) parameters. Different statistical tools were used; the one-way analysis of variance (ANO VA) was applied for analyse the differences among treatments and a linear discriminant analysis (LDA ) was used to evaluate the effects on the overall postharvest quality. The argon-enriched atmosphere better prevent the samples yellowing. The proximate composition did not change significantly during storage. Samples in control atmosphere revealed higher protein and ash contents and lower levels of lipids. The non-stored control samples presented the higher amounts of fructose, glucose and trehalose. The storage time increased the palmitic acid levels and decreased the content in α-linolenic and linoleic acids. The γ- e δ-tocopherols were higher after the 12 days of cold storage. Probably, the synthesis of these lipophilic compounds was a plant strategy to fight against the abiotic stress induced by storage. Higher levels of total phenolics and flavonoids and increased reducing power and β-carotene bleaching inhibition capacity were also found in the stored control samples. Once again, this result may be attributed to the intrinsic plant-protection mechanisms. Overall, the argon atmosphere was more suitable for quality preservation and shelf-life extension of buckler sorrel.

The biochemistry of cheese ripening involves mechanisms such as glycolysis, proteolysis and lipolysis. Fatty acids are released by the action of lipases from different sources, milk, rennet, bacteria, moulds included as secondary starters, and other exogenous lipases, during lipolysis [1]. The composition of the lipid fraction contributes positively to the flavour of cheese, for being precursors of more complex aroma compounds responsible for the characteristic “goaty flavour” of goat cheeses [2]. Goat milk is recognized by its easier digestibility, alkalinity, buffering capacity and certain therapeutic values in medicine and human nutrition [3]. A high total content of fatty acids is strongly linked to a rancid and tart off flavour in goat milk and may be considered undesirable in most cheese varieties [4]. In this sense, the purpose of the present study was to examine the composition and changes in fatty acids and saponification value of goat cheese during curing period (2, 7 and 12 months). Goat cheese was made in industrial unit of Cachão - Mirandela (Trás-os- Montes) with raw milk Serrana goats’ race, salt and rennet from animal origin. During the first two months, the samples were stored in a ripening chamber (9.5-11 °C and RH 75-85%). From the second month to one year, the samples were stored in a preservation chamber (10.5-12 °C and RH 75-85%). The fatty acids profile of the inner part of the cheese was analyzed by gas-chromatography coupled to flame ionization detection (GC-FID). The degree of saponification was determined both in the crust and inside the cheese by HCl titration of ethanol KOH solution of the samples. Twenty-six fatty acids (FA) were identified and quantified in the inner part of the cheese (total fat was 45-46 g/100 g during the curing period). Saturated fatty acids (SFA) did not change up to 7 months of curing, increasing only after 12 months, being palmitic (C16:0), stearic (C18:0), myristic (C14:0) and capric (C10:0) acids the most abundant FA in this class. Monounsaturated fatty acids (MUFA) decreased only after 12 months, and oleic acid (C18:1) was the predominant FA. In polyunsaturated fatty acids (PUFA) class, the most abundant were linoleic (C18:2) and linolenic (C18:3) acids, and followed the same tendency of MUFA. This is corroborated by an increase in the degree of saponification, either in the crust as in the inner part of the cheese, after 12 months of curing, probably related with the saturation of the fatty acids [3]. Extra-long curing can be done in cheeses produced with goat milk up to seven months of storage without changing the total fat and individual FA content.

Deep-fat frying is susceptible to induce the formation of undesirable products as lipid oxidation products and acrylamide in fried foods. Plantain chips produced by small-scale producers are sold to consumers without any control. The objective of this study was to evaluate the quality of plantain chips from local producers in relation to production process parameters and oils, and to identify the limiting factors for the production of acrylamide in plantain chips. Samples of frying oils and plantain chips prepared with either palm olein or soybean oil were collected from 10 producers in Yaoundé. Quality parameters determined in this study were: fatty acid composition of the oils, determined by gas chromatography (GC) of free acid methyl ester; trans fatty acids, determined by Fourier transform infra-red spectroscopy; Tocopherols and tocotrienols as markers of nutritional quality were analyzed by High Performance Liquid Chromatography in isocratic mode. Free fatty acids and acylglycerols as markers of lipid hydrolysis were analyzed by GC of trimethylsilyl derivatives of glycerides. Conjugated dienes, Anisidine value and viscosity as markers of lipid oxidation and thermal decomposition of the oils; acrylamide which is formed through Maillard reaction and identified as a toxic compound in various fried products. Asparagine content of the raw fresh plantain powder was also determined. Fatty acid composition of palm oleins was stable within a day of intermittent frying. In soybean oils, about 57% and 62.5% of linoleic and linolenic acids were lost but trans fatty acids were not detected. Soybean oils were partly hydrolysed leading to the formation of free fatty acids, monoacylglycerols and diacylglycerols. In both oils, tocopherols and tocotrienols contents decreased significantly by about 50%. Anisidine value (AV) and polymers contents increased slightly in fried palm oleins while conjugated hydroperoxides, AV and polymers greatly increased in soybean oils. Acrylamide was not detected in the chips. This is explained by the absence of asparagine in the raw plantains, the other acrylamide precursors being present. This study shows that the plantain chips prepared at the small-scale level in Yaounde with palm olein are of good quality regarding oxidation and hydrolysis parameters and the absence of acrylamide. In contrast, oxidation developed with soybean oil whose usage for frying should be questioned. Considering that asparagine is the limiting factor for the formation of acrylamide in plantain chips, its content depending on several factors such as production parameters and maturity stage should be explored.

Introduction: Preeclampsia is the main complication of pregnancy in developing countries. Calcium starting at 14 weeks of pregnancy is indicated to prevent the disease. Recent advances in prevention of preeclampsia endorse the addition of conjugated linoleic acid. Objective: To estimate the protective effect from calcium alone, compared to calcium plus conjugated linoleic acid in nulliparous women at risk of preeclampsia. Methods: A case-control design nested in the cohort of nulliparous women attending antenatal care from 2010 to 2014. The clinical histories of 387 cases of preeclampsia were compared with 1,054 normotensive controls. The exposure was prescriptions for calcium alone, the first period, or calcium plus conjugated linoleic acid, the second period, from 12 to 16 weeks of gestational age to labor. Confounding variables were controlled, allowing only nulliparous women into the study and stratifying by age, education and ethnic group. Results: The average age was 26.4 yrs old (range= 13-45), 85% from mixed ethnic backgrounds and with high school education. There were no differences between women who received calcium carbonate and those who did not (OR= 0.96; 95% CI= 0.73–1.27). The group of adolescents (13 to 18 yrs old) in the calcium plus conjugated linoleic acid was protected for preeclampsia (OR= 0.00; 95% CI= 0.00–0.44) independent of the confounder variables. Conclusions: 1. Calcium supplementation during pregnancy did not have preventive effects on preeclampsia. 2. Calcium plus Conjugated Linoleic acid provided to adolescents was observed to have preventive effect on Preeclampsia.

Investigou-se o efeito da adição de C18:2n6, por meio da inclusão de óleo de milho em dietas com dois níveis de proteína bruta, sobre o processo de maturação de gametas de pintado, Pseudoplatystoma corruscans, mantidos em tanques-rede. Foram avaliados: taxa de sobrevivência, relação peso x comprimento, fator de condição (K) e índice gonadossomático (IGS). O experimento foi realizado entre março de 2004 e fevereiro de 2006, em 12 tanques-rede, distribuídos em seis viveiros-escavados de 600m² e densidade de estocagem de 20 peixes/tanque-rede. Utilizaram-se três tratamentos (T) com duas repetições/viveiro: T1 com 28% de PB; T2 com 28% de PB + 5% óleo de milho e T3 com 40% de PB. O crescimento foi ligeiramente mais alto nos peixes do T3. As taxas de sobrevivência foram acima de 77%. Pode-se inferir que as rações ofertadas não causaram alterações histomorfológicas durante o processo de maturação gonadal dessa espécie. O IGS e o K foram ligeiramente mais altos nos animais alimentados com a ração enriquecida com óleo de milho.

Freshwater fish are an important source of protein, but they also contain other highly nutritive components such as fats. Polyunsaturated fatty acids (PUFAs) are essential for normal growth, development and reproduction of vertebrates. The antioxidant role of vitamin E in cell membranes prevents fatty acid and cholesterol oxidation, thereby promoting PUFA and subcellular particle stabilization. The effects of vitamin E supplementation on the quality of Nile tilapia (Oreochromis niloticus) carcass were investigated. The experiments were carried out in an experimental laboratory over 106 d. After sex reversal, 400 early juvenile O. niloticus were tested in a completely randomized experiment with 5 treatments (4 repetitions each), consisting of vitamin E monophosphate supplementation at 0, 50, 100, 150 or 200 mg/kg of a base diet. Treatment diets contained equal amounts of protein and energy. Tilapias supplemented with vitamin E contained arachidonic acid (20:4 omega-6; AA) which participates in inflammatory response. Nile tilapia carcasses that received vitamin E at 100 and 150 mg/kg diet had improved carcass quality by increasing the PUFA:SFA ratio and had the highest levels of polyunsaturated fatty acids from the omega-3 (linolenic acid; 18:3 omega-3) and omega-6 (linoleic acid; 18:2 omega-6) series. The present study evaluated the effect of salt reduction on traditional dry-cured sausages' safety, quality and product acceptance, comprising physicochemical and microbiological parameters, biogenic amines, fatty acids, texture profile and sensory analysis. According to our results, salt content had a major effect on microbiological counts, although not compromising the products' safety. Marked differences were identified regarding biogenic amines, in particular for histamine, tyramine and cadaverine, which were detected in larger amounts in products with 3%. Moreover, significant differences in the fatty acids profile have also been found, but only in less abundant components such as linoleic, lauric and heneicosanoic acids. Texture profile analysis of low-salt products, revealed a decrease in hardness and chewiness, along with an increase in adhesiveness values. Sensory evaluations revealed that despite the less intense aroma, products with 3% salt, had a more balanced salt perception. Our results suggest that salt content may be reduced to 50% in dry-cured products, with the obvious health-related advantages.
