Objective: This case-control study analyzed mass spectrometry fingerprinting patterns of culture media samples used for embryo culture to predict embryo implantation. Methods: The culture medium harvested after embryo transfer of 22 embryos from 13 patients was used for the experiments. After embryo transfer, the remaining culture media were collected and samples were split in positive (n=8) and negative (n=14) implantation groups according to implantation outcomes (100% or 0% of implantation). Samples were individually diluted and injected directly to the Electrospray ionization (ESI) MS coupled to a Quadrupole Time-of-flight MS (Q-ToF-MS).Ions relative intensities of each spectrum were considered. Data analysis was conducted in MatLab 7.0 version using Partial Least Squares - Discriminant Analysis toolbox. Results: There were 3027 observed ions at 100% and 0% implantation groups by ESI-Q-ToF-MS. The statistical model could categorize the samples in two clusters, based on their positive and negative implantation outcomes. Less intense ions present in the mass spectra with statistical significance have contributed to the major differences to group distinction. Conclusions: Positive and negative implantation embryos showed a specific biochemical pattern present in culture media, which could be detected as a fast, simple and non-invasive way. This biochemical profile could help the selection of the most viable embryo, improving single embryo transfer and thus eliminating the risk and undesirable outcomes of multiple pregnancies. © Todos os direitos reservados a SBRA - Sociedade Brasileira de Reprodução Assistida.

This study evaluated the effect of different fungicide and sodium hypochlorite treatments in reducing the incidence of fungi on seeds of native species of the Atlantic rainforest. The experiment was done using the completely randomized design in a 5×5 factorial with five levels of factor A (forest species) and five levels of factor B (chemical treatment and surface disinfection with sodium hypochlorite) with four replications. The fungal genera Fusarium, Alternaria, Aspergillus and Penicillium were identified. The species 'Jacarandáda- Bahia' and 'Ipê-roxo' had lower fungal incidence, 1.0% and 1.3%, respectively. Except for 'Angico Vermelho', the fungicide pencycurom and sodium hypochlorite were not significantly different from the control in the species evaluated. Seed treatment with fungicides 'Captan' and 'Tiram' yielded satisfactory results in reducing the incidence of fungal contamination in forest species seeds.

Buscando quantificar o efeito do parcelamento da fertirrigação no desenvolvimento da alface, esta pesquisa procurou verificar o número de parcelas de fertirrigação que apresentasse o melhor desempenho no crescimento da cultura. O experimento se deu em blocos casualizados, com três repetições, quatro tratamentos de fertirrigação e três lâminas de irrigação (L1, L2 e L3). Em um dos tratamentos de fertirrigação (F1), toda a adubação necessária se deu no início do plantio; no tratamento F2, a metade da adubação se deu no início do plantio e a outra metade no meio do ciclo da cultura, uma vez planejada a colheita em 40 dias após o plantio; o tratamento F3 foi fracionado: 1/3 da adubação no início do plantio, 1/3 depois de 13 dias e o ultimo terço após 26 dias do plantio; o tratamento F4 seguiu o mesmo raciocínio, fracionando a aplicação em 4 partes, de 10 em 10 dias. Logo, os quatro tratamentos de fertirrigação receberam a mesma dosagem de fertilizantes. Os resultados permitiram concluir que a aplicação F1 resultou no pior desempenho, com massa fresca de 135,06 gramas por planta, enquanto o melhor resultado se deu no tratamento F2, com massa fresca de 161,40 g. Em todas as lâminas, o parcelamento F3 resultou em sistema radicular mais desenvolvido. O tratamento L1 apresentou em média maior desenvolvimento das raízes, seguido do L2 e L3. O tratamento L1 apresentou menor condutividade elétrica no extrato da solução dos vasos, sendo o único tratamento que diferiu estatisticamente dos demais.

The objective of this study was to evaluate different citrus leprosis management tactics during seven seasons, based in pruning and acaricide applications, considering technical and economic aspects of each tactic. The trial was conducted from October 2003 to August 2010 in an orange plantation of Pera cv. located in the municipality of Reginópolis-SP, Brazil. The plants of citrus used were12 years old and grafted onto Cleopatra tangerine. The experimental design used was randomized blocks, in a factorial scheme, made up of factors type of pruning (A), with six levels: (1) drastic pruning, (2) intermediate pruning without leprosis lesions, (3) intermediate pruning with leprosis lesions (4) light pruning, (5) without pruning and (6) replant; acaricide applications factor (B), with three levels: (1) without acaricides applications, (2) with lime sulfur applications and (3) spirodiclofen or cyhexatin applied in rotation; pruning factor to remove leprosis symptomatic branches (C), with two levels: (1) with pruning for removal, (2) without removal pruning. The combination of factors, with respective levels (6 x 3 x 2), resulted in 36 treatments that were repeated four times, with each parcel being made up of three plants in a row. After seven years, it was observed that the types of the pruning and remove of leprosis symptomatic branches used as single management tactic, is not sufficient to leprosis control. Therefore, the results demonstrated that for leprosis management, it is essential the association between tactics, especially the control mite vector. For ensuring the citrus production economically, the use of acaricides highly efficient is essential in B. phoenicis control. The recommendation of the type of pruning should be in function of the leprosis incidence and severity in the orchard. In orchards with low leprosis incidence and severity the light pruning is more appropriate, because it is efficient and ensures greater financial balance. However, in orchards with high leprosis incidence and severity it is indicated the severe pruning for reducing or eliminating inoculum source. Between the severe prunings, the intermediate pruning with leprosis lesions is the most relevant, because the financial return will be faster. Replant is indicated only for young orchards, because does not imply in changes onto cultural practices.
