The Emissions around the M25 motorway (EM25) campaign took place over the megacity of London in the United Kingdom in June 2009 with the aim of characterising trace gas and aerosol composition and properties entering and emitted from the urban region. It featured two mobile platforms, the UK BAe-146 Facility for Airborne Atmospheric Measurements (FAAM) research aircraft and a ground-based mobile lidar van, both travelling in circuits around London, roughly following the path of the M25 motorway circling the city. We present an overview of findings from the project, which took place during typical UK summertime pollution conditions. Emission ratios of volatile organic compounds (VOCs) to acetylene and carbon monoxide emitted from the London region were consistent with measurements in and downwind of other large urban areas and indicated traffic and associated fuel evaporation were major sources. Sub-micron aerosol composition was dominated by secondary species including sulphate (24% of sub-micron mass in the London plume and 29% in the non-plume regional aerosol), nitrate (24% plume; 20% regional) and organic aerosol (29% plume; 31% regional). The primary sub-micron aerosol emissions from London were minor compared to the larger regional background, with only limited increases in aerosol mass in the urban plume compared to the background (~12% mass increase on average). Black carbon mass was the major exception and more than doubled in the urban plume, leading to a decrease in the single scattering albedo from 0.91 in the regional aerosol to 0.86 in the London plume, on average. Our observations indicated that regional aerosol plays a major role on aerosol concentrations around London, at least during typical summertime conditions, meaning future efforts to reduce PM levels in London must account for regional as well as local aerosol sources.

We investigated the impact of managed retreat on mercury (Hg) biogeochemistry at a site subject to diffuse contamination with Hg. We collected sediment cores from an area of land behind a dyke one year before and one year after it was intentionally breached. These sediments were compared to those of an adjacent mudflat and a salt marsh. The concentration of total mercury (THg) in the sediment doubled after the dyke was breached due to the deposition of fresh sediment that had a smaller particle size, and higher pH. The concentration of methylmercury (MeHg) was 27% lower in the sediments after the dyke was breached. We conclude that coastal flooding during managed retreat of coastal flood defences at this site has not increased the risk of Hg methylation or bioavailability during the first year. As the sediment becomes vegetated, increased activity of Hg-methylating bacteria may accelerate Hg-methylation rate.

Two decades ago, Canada, Mexico, and the United States created a continental economy. The road to integration from the signing of the North American Free Trade Agreement has not been a smooth one. Along the way, Mexico lived through a currency crisis, a democratic transition, and the rising challenge of Asian manufacturing. Canada stayed united despite surging Quebecois nationalism during the 1990s; since then, it has seen dramatic economic changes with the explosion of hydrocarbon production and a much stronger currency. The United States saw a stock-market bust, the shock of 9/11, and the near-collapse of its financial system. All of these events have transformed the relationships that emerged after NAFTA entered into force in 1994. Given the tremendous changes, one might be skeptical that the circumstances and details of the negotiation and ratification of NAFTA hold lessons for the future of North America. However, the road to NAFTA had its own difficulties, and many of the issues involved in the negotiations underpin today's challenges. NAFTA was conceived at a time of profound change in the international system. When Mexican leaders surveyed the world two decades ago, they saw emerging regional groupings in Europe, Asia, and South America. Faced with a lack of interest or compatibility, they instead doubled down on North America. How did Mexican leaders reconsider their national interests and redefine Mexico's role in the world in light of those transformations? Unpublished Mexican documents from SECOFI, the secretariate most involved in negotiating NAFTA, help illustrate Mexican thinking about its interests and role at that time. Combining those insights with analysis of newly available evidence from U.S. presidential archives, this paper sheds light on the negotiations that concluded two decades ago.

Aircraft do not fly through a vacuum, but through an atmosphere whose meteorological characteristics are changing because of global warming. The impacts of aviation on climate change have long been recognised, but the impacts of climate change on aviation have only recently begun to emerge. These impacts include intensified turbulence and increased take-off weight restrictions. Here we investigate the influence of climate change on flight routes and journey times. We feed synthetic atmospheric wind fields generated from climate model simulations into a routing algorithm of the type used operationally by flight planners. We focus on transatlantic flights between London and New York, and how they change when the atmospheric concentration of carbon dioxide is doubled. We find that a strengthening of the prevailing jet-stream winds causes eastbound flights to significantly shorten and westbound flights to significantly lengthen in all seasons. Eastbound and westbound crossings in winter become approximately twice as likely to take under 5 h 20 min and over 7 h 00 min, respectively. For reasons that are explained using a conceptual model, the eastbound shortening and westbound lengthening do not cancel out, causing round-trip journey times to increase. Even assuming no future growth in aviation, the extrapolation of our results to all transatlantic traffic suggests that aircraft will collectively be airborne for an extra 2000 h each year, burning an extra 7.2 million gallons of jet fuel at a cost of US$ 22 million, and emitting an extra 70 million kg of carbon dioxide, which is equivalent to the annual emissions of 7100 average British homes. Our results provide further evidence of the two-way interaction between aviation and climate change.

We combined measurements of tree growth and carbon dioxide exchange to investigate the effects of selective logging on the Aboveground Live Biomass (AGLB) of a tropical rain forest in the Amazon. Most of the measurements began at least 10 months before logging and continued at least 36 months after logging. The logging removed similar to 15% of the trees with Diameter at Breast Height (DBH) greater than 35 cm, which resulted in an instantaneous 10% reduction in AGLB. Both wood production and mortality increased following logging, while Gross Primary Production (GPP) was unchanged. The ratio of wood production to GPP (the wood Carbon Use Efficiency or wood CUE) more than doubled following logging. Small trees (10 cm < DBH < 35 cm) accounted for most of the enhanced wood production. Medium trees (35 cm < DBH < 55 cm) that were within 30 m of canopy gaps created by the logging also showed increased growth. The patterns of enhanced growth are most consistent with logging-induced increases in light availability. The AGLB continued to decline over the study, as mortality outpaced wood production. Wood CUE and mortality remained elevated throughout the 3 years of postlogging measurements. The future trajectory of AGLB and the forest`s carbon balance are uncertain, and will depend on how long it takes for heterotrophic respiration, mortality, and CUE to return to prelogging levels.

Gills are the first site of impact by metal ions in contaminated waters. Work on whole gill cells and metal uptake has not been reported before in crustaceans. In this study, gill filaments of the American lobster, Homarus americanus, were dissociated in physiological saline and separated into several cell types on a 30, 40, 50, and 80% sucrose gradient. Cells from each sucrose solution were separately resuspended in physiological saline and incubated in (65)Zn(2+) in order to assess the nature of metal uptake by each cell type. Characteristics of zinc accumulation by each kind of cell were investigated in the presence and absence of 10 mM calcium, variable NaCl concentrations and pH values, and 100 mu M verapamil, nifedipine, and the calcium ionophore A23187. (65)Zn(2+) influxes were hyperbolic functions of zinc concentration (1-1,000 mu M) and followed Michaelis-Menten kinetics. Calcium reduced both apparent zinc binding affinity (K (m)) and maximal transport velocity (J (max)) for 30% sucrose cells, but doubled the apparent maximal transport velocity for 80% sucrose cells. Results suggest that calcium, sodium, and protons enter gill epithelial cells by an endogenous broad-specificity cation channel and trans-stimulate metal uptake by a plasma membrane carrier system. Differences in zinc transport observed between gill epithelial cell types appear related to apparent affinity differences of the transporters in each kind of cell. Low affinity cells from 30% sucrose were inhibited by calcium, while high affinity cells from 80% sucrose were stimulated. (65)Zn(2+) transport was also studied by isolated, intact, gill filament tips. These intact gill fragments generally displayed the same transport properties as did cells from 80% sucrose and provided support for metal uptake processes being an apical phenomenon. A working model for zinc transport by lobster gill cells is presented.

Propolis, a natural product of plant resins, is used by the bees to seal holes in their honeycombs and protect the hive entrance. However, propolis has also been used in folk medicine for centuries. Here, we apply the power of Saccharomyces cerevisiae as a model organism for studies of genetics, cell biology, and genomics to determine how propolis affects fungi at the cellular level. Propolis is able to induce an apoptosis cell death response. However, increased exposure to propolis provides a corresponding increase in the necrosis response. We showed that cytochrome c but not endonuclease G (Nuc1p) is involved in propolis-mediated cell death in S. cerevisiae. We also observed that the metacaspase YCA1 gene is important for propolis-mediated cell death. To elucidate the gene functions that may be required for propolis sensitivity in eukaryotes, the full collection of about 4,800 haploid S. cerevisiae deletion strains was screened for propolis sensitivity. We were able to identify 138 deletion strains that have different degrees of propolis sensitivity compared to the corresponding wild-type strains. Systems biology revealed enrichment for genes involved in the mitochondrial electron transport chain, vacuolar acidification, negative regulation of transcription from RNA polymerase II promoter, regulation of macroautophagy associated with protein targeting to vacuoles, and cellular response to starvation. Validation studies indicated that propolis sensitivity is dependent on the mitochondrial function and that vacuolar acidification and autophagy are important for yeast cell death caused by propolis.

Energy intensity of the total primary energy supply (TPES), total final energy consumption (TFC) and LOSSES in the conversion from TPES to TFC were analyzed for the World, OECD and Rest of the World (ROW) countries. LOSSES increased significantly for all groups of countries due to the increase of electricity production from coal in the period studied (1971-2008). Electricity share final consumption almost doubled, increasing from 8.8% to 17.2% in the period studied. However the energy intensity of LOSSES remained practically constant, which reflects the fact that the efficiency of electricity generation from coal (the main source of electricity) remained practically constant in that period. Despite the attractiveness of end-use devices running on electricity such as computers, which is typical of modern societies, the CO(2) emissions are bound to increase unless coal is replaced by less carbon emitting sources such as natural gas, renewables and nuclear energy. This study develops a simplified model describing the evolutionary dynamics of a population composed of obligate sexually and asexually reproducing, unicellular organisms. The model assumes that the organisms have diploid genomes consisting of two chromosomes, and that the sexual organisms replicate by first dividing into haploid intermediates, which then combine with other haploids, followed by the normal mitotic division of the resulting diploid into two new daughter cells. We assume that the fitness landscape of the diploids is analogous to the single-fitness-peak approach often used in single-chromosome studies. That is, we assume a master chromosome that becomes defective with just one point mutation. The diploid fitness then depends on whether the genome has zero, one, or two copies of the master chromosome. We also assume that only pairs of haploids with a master chromosome are capable of combining so as to produce sexual diploid cells, and that this process is described by second-order kinetics. We find that, in a range of intermediate values of the replication fidelity, sexually reproducing cells can outcompete asexual ones, provided the initial abundance of sexual cells is above some threshold value. The range of values where sexual reproduction outcompetes asexual reproduction increases with decreasing replication rate and increasing population density. We critically evaluate a common approach, based on a group selection perspective, used to study the competition between populations and show its flaws in addressing the evolution of sex problem.

Calcium (Ca2+) is a critical regulator of many aspects of the Plasmodium reproductive cycle. In particular, intra-erythrocyte Plasmodium parasites respond to circulating levels of the melatonin in a process mediated partly by intracellular Ca2+. Melatonin promotes the development and synchronicity of parasites, thereby enhancing their spread and worsening the clinical implications. The signalling mechanisms underlying the effects of melatonin are not fully established, although both Ca2+ and cyclic AMP (cAMP) have been implicated. Furthermore, it is not clear whether different strains of Plasmodium use the same, or divergent, signals to control their development. The aim of this study was to explore the signalling mechanisms engaged by melatonin in P. chabaudi, a virulent rodent parasite. Using parasites at the throphozoite stage acutely isolated from mice erythrocytes, we demonstrate that melatonin triggers cAMP production and protein kinase A (PKA) activation. Interestingly, the stimulation of cAMP/PKA signalling by melatonin was dependent on elevation of Ca2+ within the parasite, because buffering Ca2+ changes using the chelator BAPTA prevented cAMP production in response to melatonin. Incubation with melatonin evoked robust Ca2+ signals within the parasite, as did the application of a membrane-permeant analogue of cAMP. Our data suggest that P. chabaudi engages both Ca2+ and cAMP signalling systems when stimulated by melatonin. Furthermore, there is positive feedback between these messengers, because Ca2+ evokes cAMP elevation and vice versa. Melatonin more than doubled the observed extent of parasitemia, and the increase in cAMP concentration and PKA activation was essential for this effect. These data support the possibility to use melatonin antagonists or derivates in therapeutic approach.

SammanfattningHögskolan Dalarna har i samarbete med Skogsägarna Mellanskog, Naturbränsle i Mellan¬sverige AB och GDE-Net genomfört studier på en ny metod för uttag av skogsbränsle från slutavverkningar. Metoden går ut på att timmer tas ut som enda rundvirkessortiment. Resten av trädet, samt klenare träd som inte håller timmerdimension, tas ut som ett bränslesortiment. Metoden har jämförts med en konventionell slutavverkning med uttag av timmer, massaved och GROT-flis.Enligt genomförda försök skulle en avverkning enligt den nya metoden (långa toppar) ge ett högre drivningsnetto och drygt dubbelt så mycket bränsleflis som en konventionell avverk¬ning. En anledning till det högre drivningsnettot är att kostnaden för flisning blir lägre än vid flisning av GROT och att flisen betalas bättre än GROT-flis. Resultaten är beroende av de faktiska beståndsförutsättningarna och gällande prisrelationer mellan massaved och bränsle¬flis.Faktorer som har en positiv inverkan på drivningsnettot vid uttag av ”långa toppar” är t.ex. stora uttagsvolymer och korta terrängtransportavstånd samt bestånd med en hög andel virke av låg kvalitet eller udda sortiment som betalas dåligt på rundvirkesmarknaden.SummaryIn Sweden forest energy from final felling is traditionally harvested as logging residues after harvesting of timber (saw logs) and pulpwood, but depending on the market situation other methods with higher yield of forest energy might be of interest. Dalarna University has study a new method called “Undelimbed long tops” where only saw timber was taken out as an industrial assortment. The rest of the trees and smaller trees that don’t hold timber dimensions was left intact on the clear-felled area and been chipped later on. The study was done in different stands with some different conditions. The results have been compared with the traditional method for final felling. The surplus (forest owners net income) was higher in almost all stands when the method with “undelimbed long tops” was used, compared to the traditional method for taking out forest energy, and the volume of chips was more than doubled. A reason for the higher income from long tops is that the costs for chipping is lower and the prize of chips is higher compared to chips from logging residues. Other reason is that forest owners will not be paid for wasted pulpwood, but will be fully paid for the chips from such pulpwood. Factors that will have a positive influence on the ULT-method are for example large logging volumes and short distance between the logging area and the landing, different kinds of price reductions on pulpwood and large volumes of rotten wood or low paid industrial assortments.
Chloroquine is grossly under dosed in young children with malaria : implications for drug resistance

Background: Plasmodium falciparum malaria is treated with 25 mg/kg of chloroquine (CQ) irrespective of age. Theoretically, CQ should be dosed according to body surface area (BSA). The effect of dosing CQ according to BSA has not been determined but doubling the dose per kg doubled the efficacy of CQ in children aged <15 years infected with P. falciparum carrying CQ resistance causing genes typical for Africa. The study aim was to determine the effect of age on CQ concentrations. Methods and Findings: Day 7 whole blood CQ concentrations were determined in 150 and 302 children treated with 25 and 50 mg/kg, respectively, in previously conducted clinical trials. CQ concentrations normalised for the dose taken in mg/kg of CQ decreased with decreasing age (p<0.001). CQ concentrations normalised for dose taken in mg/m(2) were unaffected by age. The median CQ concentration in children aged <2 years taking 50 mg/kg and in children aged 10-14 years taking 25 mg/kg were 825 (95% confidence interval [CI] 662-988) and 758 (95% CI 640-876) nmol/l, respectively (p = 0.67). The median CQ concentration in children aged 10-14 taking 50 mg/kg and children aged 0-2 taking 25 mg/kg were 1521 and 549 nmol/l. Adverse events were not age/concentration dependent. Conclusions: CQ is under-dosed in children and should ideally be dosed according to BSA. Children aged <2 years need approximately double the dose per kg to attain CQ concentrations found in children aged 10-14 years. Clinical trials assessing the efficacy of CQ in Africa are typically performed in children aged <5 years. Thus the efficacy of CQ is typically assessed in children in whom CQ is under dosed. Approximately 3 fold higher drug concentrations can probably be safely given to the youngest children. As CQ resistance is concentration dependent an alternative dosing of CQ may overcome resistance in Africa.

Background: Although associated adverse pregnancy outcomes, no international or Swedish consensus exists that identifies a cut-off value or what screening method to use for definition of gestational diabetes mellitus. This study investigates the following: i) guidelines for screening of GDM; ii) background and risk factors for GDM and selection to OGTT; and iii) pregnancy outcomes in relation to GDM, screening regimes and levels of OGTT 2 hour glucose values. Methods: This cross-sectional and population-based study uses data from the Swedish Maternal Health Care Register (MHCR) (2011 and 2012) combined with guidelines for GDM screening (2011-2012) from each Maternal Health Care Area (MHCA) in Sweden. The sample consisted of 184, 183 women: 88, 140 in 2011 and 96,043 in 2012. Chi-square and two independent samples t-tests were used. Univariate and multivariate logistic regression analyses were performed. Results: Four screening regimes of oral glucose tolerance test (OGTT) (75 g of glucose) were used: A) universal screening with a 2-hour cut-off value of 10.0 mmol/L; B) selective screening with a 2-hour cut-off value of 8.9 mmol/L; C) selective screening with a 2-hour cut-off value of 10.0 mmol/L; and D) selective screening with a 2-hour cut-off value of 12.2 mmol/L. The highest prevalence of GDM (2.9%) was found with a 2-hour cut-off value of 8.9 mmol/L when selective screening was applied. Unemployment and low educational level were associated with an increased risk of GDM. The OR was 4.14 (CI 95%: 3.81-4.50) for GDM in obese women compared to women with BMI <30 kg/m(2). Women with non-Nordic origin presented a more than doubled risk for GDM compared to women with Nordic origin (OR = 2.24; CI 95%: 2.06-2.43). Increasing OGTT values were associated with increasing risks of adverse pregnancy outcomes. Conclusions: There was no consensus regarding screening regimes for GDM from 2011 through 2012 when four different regimes were applied in Sweden. Increasing levels of OGTT 2-hour glucose values were strongly associated with adverse pregnancy outcomes. Based on these findings, we suggest that Sweden adopts the recent recommendations of the International Association of Diabetes and Pregnancy Study Group (IADPSG) concerning the performance of OGTT and the diagnostic criteria for GDM.

O transporte de massa por migração relativo à redução do proton em um ultramicroeletrodo de platina foi investigado. O efeito da migração sobre as correntes limite foi primeiramente estudado para uma só espécie eletroativa em solução através da comparação dos voltampérogramas obtidos na ausência de eletrólito suporte assim como na presença de um grande excesso do mesmo (efeito do eletrólito). O comportamento do proton na água e dos ácidos do tipo BH+ e HA- em acetonitrila foi estudado e uma expressão para quantificar o efeito do eletrólito é proposta. Ela considera as condutâncias equivalentes e as cargas das espécies iônicas em solução, o número de elétrons envolvido na reação eletroquímica e o tipo de eletrodo utilisado. Os fenômenos de exaltação da corrente de migração que podem se manifestar quando a redução de uma espécie eletroativa é precedida pela redução de uma segunda espécie presente simultâneamente na solução foram igualmente estudados. Observa-se que a exaltação da corrente de migração de uma espécie iônica ocorre mesmo quando sua transformação eletroquímica precede aquela da espécie molecular. Nêste caso, se as mobilidades iônicas são próximas, a altura da onda da espécie molecular é duas vezes maior que na ausência da espécie iônica.

A eficiência da técnica de cultura de anteras, em escala comercial, ainda pode ser considerada baixa quando medida em número de plantas duplo-haplóides férteis obtidas para cada antera estabelecida in vitro. Dessa forma, o presente trabalho é pioneiro no estudo detalhado da embriogênese in vitro do micrósporo e do grão de pólen de cevada (Hordeum vulgare L. ssp. vulgare). Com o objetivo de contribuir para o aperfeiçoamento da técnica de cultura de anteras foi analisada a embriogênese, com especial ênfase na etapa da indução, através de análises citológicas e histológicas de anteras cultivadas in vitro. Foram analisadas uma cultivar brasileira de cevada, em comparação com linhagens de duas outras cultivares brasileiras, que foram selecionadas, por seleção divergente para maior ou para menor resposta na indução da rota embriogênica e, respectivamente, para menor ou para maior capacidade de regenerar plântulas verdes. Somente foram estabelecidas em cultivo in vitro as anteras que apresentaram micrósporos e pólens jovens, das linhagens selecionadas da cultivar A-05 (S3A22 e S3A23), e da cultivar BR-2(S3B63 e, apenas na cultura de anteras, S3B61), bem como da cultivar MN-599 (nãoselecionada). Para as análises histológicas, foram fixadas, a cada dois dias, duas anteras, correspondentes a cada fileira da mesma espiga, após o início do cultivo in vitro. As anteras em cultivo e respectivas estruturas multicelulares foram fixadas em FAA 50%, desidratadas em série etílica e incluídas em hidroxietilmetacrilato. Os blocos de resina polimerizada foram secionados longitudinalmente com 3 mm de espessura. Para as análises citológicas foram fixadas, de cada espiga recém-coletada, três espiguetas sendo uma da base, outra do meio e outra do ápice. Após o pré-tratamento à baixa temperatura (5 °C), porém antes do cultivo in vitro, foram fixadas três anteras (amostras utilizadas como controles). A cada três dias, durante o cultivo, três anteras foram fixadas (até 18 dias). As anteras em cultivo e estruturas multicelulares foram fixadas em Farmer e FAA 50%, transferidas após 24 horas para etanol 70%. Na cultura in vitro das anteras houve diferenças entre uma das linhagens da cultivar A-05 em relação a cultivar MN- 599, na produção inicial de estruturas embriogênicas, diferença que desapareceu na produção total. Entretanto, houve diferenças na formação dos xiii embriões: a cv.MN-599 formou embriões bem diferenciados ao passo que a linhagem S3A22 produziu um número aparentemente menor, sendo que os embriões não eram bem diferenciados. A linhagem S3B63 não apresentou embriões até o final da análise histológica. Considerando que a amostra dessa linhagem, mantida em cultura, formou plantas verdes, pode-se propor que a formação de embriões deve ocorrer posteriormente ao desenvolvimento da cv.MN-599. Cabe destacar que houve diferenças significativas entre as cultivares A-05 e BR-2 quanto à regeneração de plântulas verdes. Esses resultados indicam ter havido maior eficiência da seleção em relação à etapa da regeneração. Com relação às categorias classificatórias dos micrósporos e grãos de pólen, constatou-se que desde o início da análise histológica (2o dia de cultivo in vitro) até o final (34o dia), foram observados micrósporos, o mesmo tendo sido observado na análise citológica. Os grãos de pólen multinucleados ocorreram praticamente em todo o período de cultivo in vitro, em ambas análises; não ocorrendo nos controles da citologia (antes do cultivo); os multinucleados foram observados a partir do 3o dia, enquanto que os multicelulares a partir do 4o dia de cultivo. As estruturas multicelulares foram observadas a partir do 8o dia. A quantidade e o tamanho das estruturas multicelulares foram variáveis ao longo da análise histológica, sendo que do 14o ao 20o dia foram encontradas as de maiores dimensões, resultantes da proliferação celular por mitoses sucessivas. A partir do 22o dia (cultivar MN- 599), a ocorrência de estruturas multicelulares no interior dos lóculos da antera diminuiu, predominando o processo de proliferação externo às anteras. Para as linhagens, a partir do 18o dia foram observadas estruturas multicelulares liberadas das anteras. A análise das estruturas multicelulares permitiu classificá-las em quatro categorias: 1. SFD: Sem forma definida; 2. MAC: meristema apical caulinar; 3. MAR: meristema apical radical embrionário adventício; e 4. Embriões. As estruturas amorfas apareceram em maior número, quando comparadas com as outras categorias. Em síntese: as linhagens selecionadas e a cultivar diferiram não apenas no tempo necessário para a formação dos embriões, mas também no desenvolvimento dos mesmos, que foi mais diferenciado na cultivar MN-599, porém sendo observados mais cedo na linhagem S3A22 e S3A23, do que na cultivar MN-599.
