Forage sorghum can be grown in areas and environmental conditions dry and warm, where the productivity of other forage plants can often be uneconomical. The soil disturbance can be made only on the lines of planting (direct seeding) or entirely from the area for seeding (conventional tillage), as plowing, harrowing, subsoiling and chiseling (minimum tillage). The displacement speed ideal for planting is one in which the groove is opened and closed without removing the over-ground, allowing the distribution of seed spacing and depth constant. The experiment was conducted in a soil classified as Typic Oxisol at Lageado Experimental Farm, Faculty of Agronomic Sciences, UNESP, Botucatu campus. This study aimed to evaluate the response of sorghum in four forward speeds (3, 5, 6 and 9 km h-1) and four systems of soil management: SD (direct seeding), GP (harrow + sowing), LPG (disc harrow and two light disking + sowing) and CR (scarification and seeding). Data was subjected to analysis of variance in a factorial 4 x 4 and a randomized block design with split plots. The following parameters were determined: average speed, average strength of the drawbar, the average power drawbar, theoretical field capacity of the tractor-equipment, fuel consumption per hour. For the conditions under which the experiment was conducted, it was concluded that the hourly fuel consumption was not influenced by tillage systems and was inversely proportional to the increase of speed work, and that the change of speed in the sowing operation did not provide additional the values of average traction force on the bar of the tractor-planter.

The initial growth and mineral nutrition of the physic nut (Jatropha curcas L.) as a function of nitrogen (N) fertilization was investigated. The transplanting of seedlings was carried out in plastic pots filled with 50 dm3 of a Rhodic Hapludox, under a plastic greenhouse. In addition to a control treatment, the dosages of 0, 40, 80, 120, and 160 mg dm−3 N were tested. The results demonstrated that N fertilization for the cultivation of physic nut could be added as top dressing from 60 days after planting with a dosage of 65 mg dm−3. A SPAD index of 46 can be used as a nutritional reference to its initial development. Furthermore, the results suggested that the order of nutrient accumulation by the physic nut plants is as follows: potassium (K) > N > magnesium (Mg) > calcium (Ca) > phosphorus (P) > sulfur (S) > iron (Fe) > manganese (Mn) > boron (B) > zinc (Zn) > copper (Cu).

The objective of this work was to evaluate the utilization by corn plants of P from triple superphosphate fertilizer labeled with P-32 (P-32-TSP), and of P from soil as affected by N rates and by the green manures (GM) sunn hemp (Crotalaria juncea) and millet (Pennisetum glaucum). The experiment was carried out using pots filled with 5 kg Oxisol (Rhodic Hapludox). A completely randomized design was used, in a 4x4x2 factorial arrangement, with four replicates. The treatments were: four P rates as TSP (0, 0.175, 0.350, and 0.700 g P per pot); four N rates as urea (0, 0.75, 1.50, and 2.25 g N per pot); and sunn hemp or millet as green manure. The additions of N and P by the GM were taken into account. After grain physiologic maturation, corn dry matter, P contents, accumulated P, and P recovery in the different treatments were measured. P-32-TSP recovery by corn increased with N increasing rates, and decreased with increasing rates of P-32-TSP. The mineral fertilizer provides most of the accumulated P by corn plants. The recovery of P-32-TSP by corn was 13.12% in average. The green manure species influence the assimilation of P-32-TSP by the plants.

The adoption of no-till system (NTS) combined with crop-livestock integration (CLI) has been a strategy promoted in Brazil, aiming to maximize areas yield and increase agribusiness profitability. This study aimed to evaluate grains yield and phytotechnical attributes from maize and soybean culture by CLI system under NTS after winter annual pure and diversified pastures with the absence or presence of grazing animals. The experiment was installed in Castro (Parana State, Brazil) on in a dystrophic Humic Rhodic Hapludox with a clay texture, using experimental design of randomized complete blocks in 4 x 2 factorial scheme with three replications. Treatments included four pasture combinations (diversified or pure) and animal categories (light and heavy) subjected or not to grazing animals during the winter. During 2008/09 and 2009/10 summers, the area was cultivated with soybeans and maize, respectively, with yield assessment of grains and phytotechnical attributes. Treatments did not alter the yield and weight of a thousand seeds (WTS) of soybeans. In maize culture, the grazing animal during the winter increased the plant population and grains yield, but gave slight decrease in WTS. Pasture combinations (diversified or pure) and animal categories (light and heavy) did not interfere in soybean culture, but benefited the maize crop.

DISTRIBUTION OF NITROGEN AMMONIUM SULFATE (N-15) SOIL-PLANT SYSTEM IN A NO-TILLAGE CROP SUCCESSION The N use by maize (Zea mays, L.) is affected by N-fertilizer levels. This study was conducted using a sandy-clay texture soil (Hapludox) to evaluate the efficiency of N use by maize in a crop succession, based on N-15-labeled ammonium sulfate (5.5 atom %) at different rates, and to assess the residual fertilizer effect in two no-tillage succession crops (signalgrass and corn). Two maize crops were evaluated, the first in the growing season 2006, the second in 2007, and brachiaria in the second growing season. The treatments consisted of N rates of 60, 120 and 180 kg ha(-1) in the form of labeled N-15 ammonium sulfate. This fertilizer was applied in previously defined subplots, only to the first maize crop (growing season 2006). The variables total accumulated N; fertilizer-derived N in corn plants and pasture; fertilizer-derived N in the soil; and recovery of fertilizer-N by plants and soil were evaluated. The highest uptake of fertilizer N by corn was observed after application of 120 kg ha(-1) N and the residual effect of N fertilizer on subsequent corn and Brachiaria was highest after application of 180 kg ha(-1) N. After the crop succession, soil N recovery was 32, 23 and 27 % for the respective applications of 60, 120 and 180 kg ha(-1) N.

O aproveitamento do N pelo milho (Zea mays, L.) é influenciado pelas doses de adubo nitrogenado. O presente trabalho foi desenvolvido em um solo de textura arenoargilosa (Hapludox) e teve por objetivo avaliar a eficiência de utilização do N pela cultura de milho, em uma sucessão de culturas, utilizando-se sulfato de amônio marcado com 15N (5,5 átomos %), em diferentes doses; e o efeito residual desse fertilizante nas duas culturas subsequentes em sucessão (braquiária e milho), sob sistema plantio direto. As avaliações foram feitas em dois cultivos de milho safrinha - o primeiro no ano agrícola 2006 e o segundo em 2007 - e um de braquiária na entressafra. Os tratamentos consistiram de doses de N de 60, 120 e 180 kg ha-1, na forma de sulfato de amônio marcado (15N). Esse adubo foi aplicado em subparcelas, previamente definidas, apenas no primeiro cultivo do milho (safra 2006). Foram avaliados: N-total acumulado; N nas plantas de milho e braquiária proveniente do fertilizante, N no solo proveniente do fertilizante e recuperação de N-fertilizante pelas plantas e pelo solo. O maior aproveitamento do N-fertilizante pelo milho foi obtido no tratamento com 120 kg ha-1 de N, e o maior efeito residual do N-fertilizante pela braquiária e milho subsequente, no tratamento com 180 kg ha-1 de N. Após a sucessão de culturas, a recuperação de N pelo solo foi de 32, 23 e 27 % para os tratamentos com 60, 120 e 180 kg ha-1 de N.

Oggetto di studio del dottorato sono stati i suoli forestali in ambiente litoraneo della Regione Emilia-Romagna. In particolare sono state considerate quattro zone di studio in Provincia di Ravenna: Pineta di San Vitale, aree boscate di Bellocchio, Pineta di Classe e Pineta di Pinarella di Cervia. Lo studio in una prima fase si è articolato nella definizione dello stato del sistema suolo, mediante la caratterizzazione pedologica delle zone di studio. A tale scopo è stata messa a punto un’adeguata metodologia d’indagine costituita da un’indagine ambientale e successivamente da un’indagine pedologica. L’indagine ambientale, mediante fotointerpretazione ed elaborazione di livelli informativi in ambito GIS, ha permesso di individuare ambiti pedogenetici omogenei. L’indagine pedologica in campo ha messo in luce l’elevata variabilità spaziale di alcuni fattori della pedogenesi, in particolar modo l’andamento microtopografico tipico dei sistemi dunali costieri e la profondità della falda freatica del piano campagna. Complessivamente sono stati aperti descritti e campionati 40 profili pedologici. Sugli orizzonti diagnostici di questi sono state eseguite le seguenti analisi: tessitura, pH, calcare totale, carbonio organico, azoto kjeldahl, conduttività elettrica (CE), capacità di scambio cationico (CSC) e calcare attivo. I suoli presentano, ad eccezione della tessitura (generalmente grossolana), un’elevata variabilità delle proprietà chimico fisiche in funzione della morfologia, della profondità e della vicinanza della falda freatica. Sono state riscontrate diverse correlazioni, tra le più significative quelle tra carbonio organico e calcare totale (coeff. di correlazione R = -0.805 per Pineta di Classe) e tra calcare totale e pH (R = 0.736), dalle quali si è compreso in che misura l’effetto della decarbonatazione agisce nei diversi ambiti pedogenetici e tra suoli con diversa età di formazione. Il calcare totale varia da 0 a oltre 400 g.kg-1 e aumenta dalla superficie in profondità, dall’entroterra verso la costa e da nord verso sud. Il carbonio organico, estremamente variabile (0.1 - 107 g.kg-1), è concentrato soprattutto nel primo orizzonte superficiale. Il rapporto C/N (>10 in superficie e molto variabile in profondità) evidenzia una efficienza di umificazione non sempre ottimale specialmente negli orizzonti prossimi alla falda freatica. I tipi di suoli presenti, classificati secondo la Soil Taxonomy, sono risultati essere Mollic/Sodic/Typic Psammaquents nelle zone interdunali, Typic Ustipsamments sulle sommità dunali e Oxiaquic/Aquic Ustipsamments negli ambienti morfologici intermedi. Come sintesi della caratterizzazione pedologica sono state prodotte due carte dei suoli, rispettivamente per Pineta di San Vitale (scala 1:20000) e per le aree boscate di Bellocchio (scala 1:10000), rappresentanti la distribuzione dei pedotipi osservati. In una seconda fase si è focalizzata l’attenzione sugli impatti che le principali pressioni naturali ed antropiche, possono esercitare sul suolo, condizionandone la qualità in virtù delle esigenze del soprasuolo forestale. Si è scelta la zona sud di Pineta San Vitale come area campione per monitorarne mensilmente, su quattro siti rappresentativi, le principali caratteristiche chimico-fisiche dei suoli e delle acque di falda, onde evidenziare possibili correlazioni. Le principali determinazioni svolte sia nel suolo in pasta satura che nelle acque di falda hanno riguardato CE, Ca2+, Mg2+, K+, Na+, Cl-, SO4 2-, HCO3 - e SAR (Sodium Adsorption Ratio). Per ogni sito indagato sono emersi andamenti diversi dei vari parametri lungo i profili, correlabili in diversa misura tra di loro. Si sono osservati forti trend di aumento di CE e degli ioni solubili verso gli orizzonti profondi in profili con acqua di falda più salina (19 – 28 dS.m-1) e profonda (1 – 1.6 m dalla superficie), mentre molto significativi sono apparsi gli accumuli di sali in superficie nei mesi estivi (CE in pasta satura da 17.6 a 28.2 dS.m-1) nei profili con falda a meno di 50 cm dalla superficie. Si è messo successivamente in relazione la CE nel suolo con diversi parametri ambientali più facilmente monitorabili quali profondità e CE di falda, temperatura e precipitazioni, onde trovarne una relazione statistica. Dai dati di tre dei quattro siti monitorati è stato possibile definire tali relazioni con equazioni di regressione lineare a più variabili. Si è cercato poi di estendere l’estrapolabilità della CE del suolo per tutte le altre casistiche possibili di Pineta San Vitale mediante la formulazione di un modello empirico. I dati relativi alla CE nel suolo sia reali che estrapolati dal modello, sono stati messi in relazione con le esigenze di alcune specie forestali presenti nelle zone di studio e con diverso grado di tolleranza alla salinità ed al livello di umidità nel suolo. Da tali confronti è emerso che per alcune specie moderatamente tolleranti la salinità (Pinus pinea, Pinus pinaster e Juniperus communis) le condizioni critiche allo sviluppo e alla sopravvivenza sono da ricondursi, per la maggior parte dei casi, alla falda non abbastanza profonda e non tanto alla salinità che essa trasmette sull’intero profilo del suolo. Per altre specie quali Quercus robur, Populus alba, Fraxinus oxycarpa e Ulmus minor moderatamente sensibili alla salinità, ma abituate a vivere in suoli più umidi, la salinità di una falda troppo prossima alla superficie può ripercuotersi su tutto il profilo e generare condizioni critiche di sviluppo. Nei suoli di Pineta San Vitale sono stati inoltre studiati gli aspetti relativi all’inquinamento da accumulo di alcuni microtossici nei suoli quali Ag, Cd, Ni e Pb. In alcuni punti di rilievo sono stati osservati moderati fattori di arricchimento superficiale per Pb e Cd riconducibili all’attività antropica, mentre le aliquote biodisponibili risultano maggiori in superficie, ma all’interno dei valori medi dei suoli italiani. Lo studio svolto ha permesso di meglio conoscere gli impatti sul suolo, causati dalle principali pressioni esistenti, in un contesto dinamico. In particolare, si è constatato come i suoli delle zone studiate abbiano un effetto tampone piuttosto ridotto sulla mitigazione degli effetti indotti dalle pressioni esterne prese in esame (salinizzazione, sodicizzazione e innalzamento della falda freatica). Questo è dovuto principalmente alla ridotta presenza di scambiatori sulla matrice solida atti a mantenere un equilibrio dinamico con le frazioni solubili. Infine le variabili ambientali considerate sono state inserite in un modello concettuale DPSIR (Driving forces, Pressures, States, Impacts, Responces) dove sono stati prospettati, in via qualitativa, alcuni scenari in funzione di possibili risposte gestionali verosimilmente attuabili, al fine di modificare le pressioni che insistono sul sistema suolo-vegetazione delle pinete ravennati.

Das ADAM10-Gen kodiert für eine membrangebundene Disintegrin-Metalloproteinase, die das Amyloidvorläuferprotein spaltet. Im Mausmodell konnte bewiesen werden, dass die Überexpression von ADAM10 die Plaquebildung vermindern und das Langzeitgedächtnis verbessert. Aus diesem Grund ist es für einen möglichen Therapieansatz für die Alzheimer’sche Erkrankung erforderlich, die Organisation des humanen ADAM10-Gens und seines Promotors aufzuklären. Beim Vergleich der genomischen Sequenzen von humanem und murinem ADAM10 zeigte sich eine hohe Übereinstimmung. Beide Gene umfassen 160 kbp und bestehen aus 16 Exons. Die ersten 500 bp stromaufwärts vom Translationsstartpunkt zwischen dem Menschen, der Maus und der Ratte sind hoch konserviert. Diese Region beinhaltet spezifische regulatorische Elemente, die die ADAM10-Transkription modulieren. In den ersten 2179 bp stromaufwärts vom humanen ADAM10-Translationsstartpunkt fanden sich einige potentiellen Transkriptionsfaktor-bindungsstellen (Brn-2, SREBP, Oct-1, Creb1/cJun, USF, Maz, MZF-1, NFkB und CDPCR3HD). Es wurde eine charakteristische GC-Box und eine CAAT-Box, aber keine TATA-Box identifiziert. Nach Klonierung dieser 2179 bp großen Region wurde eine starke Promotoraktivität, insbesondere in neuronalen Zelllinien, gefunden. Bei der Analyse von Deletionskonstrukten wurde die Region zwischen -508 und -300 als essentiell für die Transkriptionsaktivierung bestimmt. Die Promotoraktivität wird zudem streng herunterreguliert, wenn in die Region 317 bp stromaufwärts vom Startpunkt der Translation eine Punktmutation eingeführt wird. Diese per Computeranalyse als USF-Bindungsstelle deklarierte Region spielt eine zentrale Rolle bei der ADAM10-Transkription. Im EMSA wurde eine Protein-DNA-Interaktion für diese Region gezeigt. Durch transienten Transfektionen in Schneider Drosophila Insektenzellen konnte nachgewiesen werden, dass die Überexpression von Sp1 und USp3 für die ADAM10-Promotoraktivität entscheidend ist. In EMSA-Studien bestätigte sich eine Protein-DNA-Interaktion für die Region -366 bp stromaufwärts vom Translationsstartpunkt. Die Punktmutation in der CAAT-Box veränderte die die Promotoraktivität nicht. Da weiterhin für diese potentielle Bindungsstelle kein Bindungsfaktor vorausgesagt wurde, scheint die CAAT-Box keine Bedeutung bei der Promotorregulation zu spielen. Schließlich fand sich im EMSA eine Protein-DNA-Interaktion für die Bindungsstelle 203 bp stromaufwärts vom Translationsstartpunkt. Diese in Computeranalysen als RXR-Bindungsstelle identifizierte Region ist ebenfalls von Bedeutung in der Promotorregulation. Auf der Suche nach Substanzen, die die ADAM10-Promotoraktivität beeinflussen, wurde ein negativer Effekt durch die apoptoseauslösende Substanz Camptothecin und ein positiver Effekt durch die zelldifferenzierungsauslösende Substanz all-trans Retinsäure festgestellt. Mit dieser Arbeit wurde die genomische Organisation des ADAM10-Gens zusammen mit dem zugehörigen Promotor aufgeklärt und ein neuer Regulationsmechanismus für die Hochregulation der Expression der alpha-Sekretase ADAM10 gefunden. Im Weiteren sollen nun die genauen Mechanismen bei der Hochregulation der alpha-Sekretase ADAM10 durch Retinsäure untersucht und durch Mikroarray-Analysen an RNA-Proben transgener Mäuse, welche ADAM10 überexpremieren, neue therapeutische Ansätze zur Behandlung der Alzheimer´schen Erkrankung identifiziert werden.

Die nahe verwandten T-box Transkriptionsfaktoren TBX2 und TBX3 werden in zahlreichen humanen Krebsarten überexprimiert, insbesondere in Brustkrebs und Melanomen. Die Überexpression von TBX2 und TBX3 hat verschiedene zelluläre Effekte, darunter die Unterdrückung der Seneszenz, die Förderung der Epithelialen-Mesenchymalen Transition sowie invasive Zellmotilität. Im Gegensatz dazu führt ein Funktionsverlust von TBX3 und der meisten anderen humanen T-box-Gene zu haploinsuffizienten Entwicklungsdefekten. Durch Sequenzierung des Exoms von Brustkrebsproben identifizierten Stephens et al. fünf verschiedene Mutationen in TBX3, welche allesamt die DNA-bindende T-box-Domäne betrafen. Die In-Frame-Deletion N212delN wurde zweimal gefunden. Aus der Anhäufung der Mutationen innerhalb der T-box-Domäne wurde geschlossen, dass TBX3 bei Brustkrebs ein Treibergen ist. Da Mutationen innerhalb der T-box-Domäne im Allgemeinen zu einem Funktionsverlust führen, aber die onkogene Aktivität von TBX3 meist auf eine Überexpression zurückzuführen ist, wurden die potentiellen Treibermutationen hinsichtlich einer verminderten oder gesteigerten TBX3-Funktion geprüft. Getestet wurden zwei In-Frame Deletionen, eine Missense- sowie eine Frameshift-Mutante bezüglich der DNA-Bindung in vitro und der Zielgen-Repression in Zellkultur. Zusätzlich wurde eine in silico Analyse der im The Cancer Genome Atlas (TCGA) gelisteten somatischen TBX-Brustkrebsmutationen durchgeführt. Sowohl die experimentelle als auch die in silico Analyse zeigten, dass die untersuchten Mutationen vorwiegend zum Verlust der TBX3-Funktion führen. Um den Mechanismus der Genrepression durch TBX3 besser zu verstehen, wurden weitere TBX3-Mutanten bezüglich ihrer Wirkung auf die p21-Promotoraktivität (p21-Luc-Reporter und endogene p21-Expression) analysiert. Wildtypische p21-Luc-Repression zeigten die zwei Mutationen S674A (Phosphorylierung) und D275K (SUMOylierung), welche posttranslationale Modifikationen verhindern, sowie die Interaktion mit dem Tumorsuppressor Rb1 unterbindende M302A/V304A-Mutation. Erstaunlicherweise war die endogene p21-Repression dieser Mutanten stärker als die des wildtypischen TBX3-Proteins. Alle drei Mutationen führten zu einer Stabilisierung des TBX3-Proteins. Die ursprünglich in Patienten mit Ulna-Mamma Syndrom identifizierte, DNA-bindungsdefekte Y149S-Mutante konnte weder p21-Luc noch endogenes p21 reprimieren. Mutationen in potentiellen Interaktionsdomänen für die Bindung der Co-Repressoren Groucho und C-terminalem Bindeprotein zeigten sowohl auf p21-Luc als auch auf endogenes p21-Gen wildtypische Repressoraktivität, so dass diese Co-Repressoren in COS-7-Zellen wahrscheinlich nicht an der Repression dieses Gens beteiligt sind. Da TBX2 und TBX3 interessante Ziele zur direkten Krebsbekämpfung darstellen, sollte ein zelluläres Reportersystem zur Identifikation TBX2-inhibierender, pharmakologisch aktiver Substanzen etabliert werden. Dazu sollte eine stabile Zelllinie mit vom p21-Promotor reguliertem d2EGFP-Reporter und Doxyzyklin-induzierbarem TBX2-Protein erzeugt werden, da ektopische Expression von TBX2 genetische Instabilität und Toxizität induzieren kann. In dieser Zelllinie sollte die TBX2-Expression zur Reduktion der d2EGFP-Fluoreszenz führen. Zur Erzeugung der Zelllinie wurden die folgenden drei Konstrukte Schritt-für-Schritt stabil in das Genom der Zielzelllinie COS-7 integriert: pEF1alpha-Tet3G, pTRE3G-TBX2 und p21-d2EGFP. Während die Herstellung der doppelt stabilen COS-7-Zelllinie gelang, scheiterte die Herstellung der dreifach stabilen Zelllinie.

The process of adaptive radiation involves multiple events of speciation in short succession, associated with ecological diversification. Understanding this process requires identifying the origins of heritable phenotypic variation that allows adaptive radiation to progress. Hybridization is one source of genetic and morphological variation that may spur adaptive radiation. We experimentally explored the potential role of hybridization in facilitating the onset of adaptive radiation. We generated first- and second-generation hybrids of four species of African cichlid fish, extant relatives of the putative ancestors of the adaptive radiations of Lakes Victoria and Malawi. We com- pared patterns in hybrid morphological variation with the variation in the lake radiations. We show that significant fractions of the interspecific mor- phological variation and the major trajectories in morphospace that charac- terize whole radiations can be generated in second-generation hybrids. Furthermore, we show that covariation between traits is relaxed in second- generation hybrids, which may facilitate adaptive diversification. These results support the idea that hybridization can provide the heritable pheno- typic diversity necessary to initiate adaptive radiation.

1. Predation is a prime force of natural selection. Vulnerability to predation is typically highest early in life, hence effective antipredator defences should work already shortly after birth. Such early defences may be innate, transmitted through non-genetic parental effects or acquired by own early experience. 2. To understand potential joint effects of these sources of antipredator defences on pheno- typic expression, they should be manipulated within the same experiment. We investigated innate, parental and individual experience effects within a single experiment. Females of the African cichlid Simochromis pleurospilus were exposed to the offspring predator Ctenochromis horei or a benign species until spawning. Eggs and larvae were hand-reared, and larvae were then exposed to odour cues signalling the presence or absence of predators in a split-brood design. 3. Shortly after independence of maternal care, S. pleurospilus undergo a habitat shift from a deeper, adult habitat to a shallow juvenile habitat, a phase where young are thought to be par- ticularly exposed to predation risk. Thus, maternal effects induced by offspring predators pres- ent in the adult habitat should take effect mainly shortly after independence, whereas own experience and innate antipredator responses should shape behaviour and life history of S. pleurospilus during the later juvenile period. 4. We found that the manipulated environmental components independently affected different offspring traits. (i) Offspring of predator-exposed mothers grew faster during the first month of life and were thus larger at termination of maternal care, when the young migrate from the adult to the juvenile habitat. (ii) The offspring’s own experience shortly after hatching exerted lasting effects on predator avoidance behaviour. (iii) Finally, our results suggest that S. pleuro- spilus possess a genetically inherited ability to distinguish dangerous from benign species. 5. In S. pleurospilus, maternal effects were limited to a short but critical time window, when young undergo a niche shift. Instead, own environmental sampling of predation risk combined with an innate predisposition to correctly identify predators appears to prepare the young best for the environment, in which they grow up as juveniles.

Anion exchange membranes (AEMs) are a potential method for determining the plant available N status of soils; however, their capacity for use with turfgrass has not been researched extensively. The main objective of this experiment was to determine the relationship between soil nitrate desorbed from AEMs and growth response and quality of turfgrass managed as a residential lawn. Two field experiments were conducted with a bluegrass-ryegrass-fescue mixture receiving four rates of N fertilizer (0, 98, 196, and 392 kg N ha(-1) yr(-1)) with clippings returned or removed. The soils at the two sites were a Paxton fine sandy loam (coarse-loamy, mixed, active, mesic Oxyaquic Dystrudepts) and a variant of a Hinckley gravelly sandy loam (sandy-skeletal, mixed, mesic Typic Udorthents). Anion exchange membranes were inserted into plots and exchanged weekly during the growing seasons of 1998 and 1999. Nitrate-N was desorbed from AEMs and quantified. As N fertilization rates increased, desorbed NO3-N increased. The relationship of desorbed NO3-N from AEMs to clipping yield and turfgrass quality was characterized using quadratic response plateau (QRP) and Cate-Nelson models (C-Ns). Critical levels of desorbed NO3-N ranged from 0.86 to 8.0 microgram cm(-2) d(-1) for relative dry matter yield (DMY) and from 2.3 to 12 microgram cm(-2) d(-1) for turfgrass quality depending upon experimental treatment. Anion exchange membranes show promise of indicating the critical levels of soil NO3-N desorbed from AEMs necessary to achieve maximum turfgrass quality and yield without overapplication of N.
