The role of wake stiffness on the wake-induced vibration of the downstream cylinder of a tandem pair

When a pair of tandem cylinders is immersed in a flow the downstream cylinder can be excited into wake-induced vibrations (WIV) due to the interaction with vortices coming from the upstream cylinder. Assi, Bearman & Meneghini ( J. Fluid Mech. , vol. 661, 2010, pp. 365–401) concluded that the WIV excitation mechanism has its origin in the unsteady vortex–structure interaction encountered by the cylinder as it oscillates across the wake. In the present paper we investigate how the cylinder responds to that excitation, characterising the amplitude and frequency of response and its dependency on other parameters of the system. We introduce the concept of wake stiffness , a fluid dynamic effect that can be associated, to a first approximation, with a linear spring with stiffness proportional to Re and to the steady lift force occurring for staggered cylinders. By a series of experiments with a cylinder mounted on a base without springs we verify that such wake stiffness is not only strong enough to sustain oscillatory motion, but can also dominate over the structural stiffness of the system. We conclude that while unsteady vortex–structure interactions provide the energy input to sustain the vibrations, it is the wake stiffness phenomenon that defines the character of the WIV response

Zusammenfassung In der vorliegenden Arbeit wurden unterschiedliche Eigenschaften von tandem repetitiver DNA (trDNA) analysiert. Drei der untersuchten trDNA-Familien (Cla-Elemente, Alu-Elemente, 1,688-Satelliten-DNA) stammen aus der genomischen DNA von Insekten, während ein trDNA-Cluster artifiziell aus Hsp70 Promotoren hergestellt wurde. Untersucht wurde die Stabilität dieser trDNA-Sequenzfamilien innerhalb eines Plasmidvektors in E. coli bzw. nach Integration der trDNA auch im Genom von D. melanogaster. Der Schwerpunkt der Arbeit liegt jedoch in der Analyse des Einflusses von trDNA auf die Expression eines benachbarten Reportergens in D. melanogaster.Ziel der Untersuchungen zur Stabilität war es, Eigenschaften von trDNA-Clustern und Mechanismen aufzuzeigen, die die Stabilität derselben in E. coli und im Genom von D. melanogaster beeinflussen. Mit Ausnahme der Alu-Elemente zeigen alle trDNA-Familien eine deutliche Instabilität in E. coli. Am Beispiel der Cla-Elemente wurde gezeigt, daß spezifische Eigenschaften der trDNA-Familie, wie etwa die sequenzbedingte Krümmung der Helixachse, keinen Einfluß auf die Stabilität des trDNA-Clusters haben, sondern die Orientierung des trDNA-Clusters innerhalb des Vektors ausschlaggebend ist. Ein entscheidender Faktor könnte die Orientierung des trDNA-Clusters relativ zur Wanderungsrichtung der Replikationsgabel in E. coli sein. Am Beispiel des trDNA-Clusters aus artifiziellen Hsp70 Promotoren konnte gezeigt werden, daß verschiedene Rekombinationssysteme an der Instabilität in E. coli beteiligt sind. Die meisten beobachteten Deletionen von trDNA sind RecA-abhängig. Zusätzlich findet jedoch in einem kleinen Teil der Plasmide auch eine RecA-unabhängige Rekombination statt. Sowohl in E. coli als auch in D. melanogaster wurde als vorherrschender Mechanismus der trDNA-Instabilität die homologe Rekombination identifiziert. TrDNA-Cluster, die in E. coli deutlich instabil sind, können jedoch im Genom von D. melanogaster weitgehend stabil sein. Auch Faktoren, die in E. coli die Stabilität eines trDNA-Clusters beeinflussen, wie etwa die Orientierung, zeigen in D. melanogaster keinen Einfluß auf die Stabilität der trDNA-Cluster. Ergebnisse aus Stabilitätsuntersuchungen in E. coli können damit nicht ohne Überprüfung auf andere Organismen übertragen werden. Im Hauptteil der Arbeit sollte geklärt werden, ob trDNA generell die Expression benachbarter Gene beeinflußt und welche Eigenschaften der trDNA für diesen inhibitorischen oder stimulierenden Effekt verantwortlich sind. Keine der untersuchten trDNA-Familien zeigt einen inhibitorischen Effekt auf ein benachbartes Reportergen. Entgegen dem Modell von Dorer und Henikoff (1994) führen trDNA-Cluster nicht per se zu der Entstehung von Heterochromatin. Die Ergebnisse der vorliegenden Arbeit zeigen, daß sowohl ein trDNA-Cluster aus Cla-Elementen als auch aus Hsp70 Promotor-Elementen eine deutliche Steigerung der Expression des miniwhite-Reportergens bewirken. Diese Steigerung ist für beide trDNA-Familien unabhängig von der chromosomalen Lage des Transgens im Euchromatin. In beiden Fällen ist der Effekt von der Orientierung des trDNA-Clusters abhängig und verstärkt sich mit wachsender Kopienzahl der trDNA-Einheiten. Während eines der trDNA-Cluster aus trDNA-Einheiten besteht, die bekanntermaßen eine Promotoraktivität aufweisen (Hsp70 Promotoren), war für die Cla-Elemente kein Einfluß auf die Expression eines benachbarten Gens bekannt. Damit wurde für eine trDNA-Familie aus Chironomus nachgewiesen, daß sie auf ein benachbartes Gen ähnlich wirkt wie zusätzliche Promotoren. Die experimentellen Befunde unterstützen ein Modell, demzufolge die Cla-Elemente in gleicher Weise wie tandem repetitive Promotoren auf ein benachbartes Gen expressionssteigernd wirken. Die TATA-Box ist für das Modell ein wichtiges Strukturelement, da diese in beiden expressionssteigernden DNA-Sequenzen der trDNA-Cluster vorkommt und ausschließlich in einer Orientierung wirkt. Das Modell besagt, daß durch die Verbindung einer offenen Chromatinstruktur mit korrekt orientierten Bindungsstellen für TBP der Aufbau von vollständigen Transkriptionskomplexen an den tandem repetitiven Promotoren initiiert wird. Einer Perlenschnur ähnlich wären die Transkriptionskomplexe direkt verfügbar, nachdem ein Transkriptionskomplex den Promotor zur Transkription verlassen hat (Abb. 32). Dies würde zu der beobachteten Steigerung der Expression des Reportergens sowohl durch die tandem repetitiven Hsp70 Promotoren als auch durch die Cla-Elemente führen.

In questo lavoro di tesi sono state sviluppate reazioni domino, tandem e procedure one-pot per ottenere eterocicli enatioarricchiti. Lo sviluppo di queste metodologie sintetiche è molto importante perché permettono di ottenere molecole complesse partendo da prodotti semplici, senza effettuare ripetuti passaggi di purificazione (stop-and-go or step-by-step synthesis). Lo scopo di questo lavoro è di ottenere derivati tetraidrofuranici modificati e derivati ossoazzolinici enantioarrichiti tramite reazioni SN2-Michael o tramite reazioni aldolica-ciclizzazione-Michael usando la catalisi asimmetrica a trasferimento di fase (PTC). Come catalizzatori PTC per imprimere enantioselezione sono stati utilizzati sali di ammonio quaternario derivati dagli alcaloidi della Cinchona. Sono state ottimizzate le condizioni di reazione (base inorganica, temperatura, solvente, tempo di reazione) per i diversi substrati presi in considerazione. I prodotti target sono stati ottenuti con buone rese, ottime diastereoselezioni ma con bassa enantioselezione. I risultati ottenuti richiedono un’ulteriore ottimizzazione e dovranno essere valutate variazioni strutturali dei nucleofili utilizzati. In this thesis were developed domino, tandem reactions and one-pot procedures to obtained enantioenriched heterocycles. The development of these methodologies is very fundamental because they allow to obtain complex molecules starting from raw materials, without carrying out repeated purification steps (stop-and-go or step-by-step synthesis). The purpose of this work is to obtain enantioenriched tetrahydrofuran and oxazoline derivatives through a SN2-Michael reaction or a aldol- cyclization-Michael reaction using the phase-transfer asymmetric catalysis (PTC). For imprint enantioselection we used Cinchona alkaloids quaternary ammonium salts derivatives. The reaction conditions (inorganic base, temperature, solvent, reaction time) were optimised for the different substrates taken into account. The target products were obtained with good yields, excellent diastereoselections but with low enantioselections. The obtained results require further optimization and structural changes in the nucleophiles used must be evaluated.

This thesis is the result of the study of two reactions leading to the formation of important heterocyclic compounds of potential pharmaceutical interest. The first study concerns the reaction of (1,3)-dipolar cycloaddition between nitrones and activated olefins by hydrogen bond catalysis of thioureas derivatives leading to the formation of a five-membered cyclic adducts, an interesting and strategic synthetic intermediate, for the synthesis of benzoazepine. The second project wants to explore the direct oxidative C(sp3)-H α-alkylation of simple amides with subsequent addition of an olefin and cyclization in order to obtain the corresponding oxazine. Both reactions are still under development.

Nowadays obesity can be defined as a global epidemic. The precise identification of circulating biomarkers involved in this pathology could be essential to early diagnose potential co-morbidities and to better address the development of future therapeutic strategies. Published evidences show that circulating steroid hormones and endocannabinoids might have a role in the physiopathology of obesity; however, a precise and reliable quantification of these molecules is still lacking. In the first part of the present thesis, we developed a sensitive, specific and accurate quantification method for nine steroid hormones using a liquid chromatography tandem mass spectrometry (LC-MS/MS) system. This method has been used first for a comparative study with immunoassays, currently used in the clinical practice to quantify these molecules and then to redefine circulating reference intervals in healthy subjects. Furthermore, we measured circulating steroid hormones in three groups of subjects: normo-weight, over-weight and obese, defining different steroid hormones profiles depending on the obesity state. The role of circulating endocannabinoids in humans is still unclear, however there are several evidences concerning their involvement in obesity. In the second part of the thesis, we determined changes of circulating endocannabinoids in obese patients after a weight loss induced by bariatric surgery, currently the most effective long-term treatment for obesity, using LC/MS-MS. We measured basal and dynamic endocannabinoids plasma levels in 12 patients with severe obesity before, one month after and six months after the Roux-en-Y gastric bypass intervention, currently one of the most performed types of bariatric surgery. All together the findings illustrated in this thesis project will help better define the role of steroid hormones and endocannabinoids in the framework of obesity in humans and the role that each type of molecule might have in its pathophysiology.

Il lavoro discusso in questa tesi è lo sviluppo di un metodo sintetico basato sulla catalisi asimmetrica che consente di ottenere precursori di alcuni alcaloidi dell’Ergot in forma enantioarricchita. La reazione sviluppata consiste in un processo domino Friedel-Crafts/nitro-Michael fra indoli sostituiti in posizione 4 con un accettore di Michael e nitroetene. I catalizzatori utilizzati sono in grado di attivare il nitroetene per la Friedel-Crafts mediante legami a idrogeno e di indurre stereoselezione nella successiva ciclizzazione di Michael intramolecolare. Mediante la procedura ottimizzata è possibile ottenere una serie di indoli policiclici, potenziali precursori sintetici di alcaloidi dell’Ergot, con buone rese ed enantioselezioni.

Questa tesi ha come argomento lo studio dei dispositivi fotovoltaici, sistemi che trasformano la radiazione solare in energia elettrica. In particolare è stata approfondita l'analisi di una specifica categoria di celle solari, le tandem cells, dispositivi a giunzione multipla in cui la radiazione solare attraversa due o più giunzioni singole caratterizzate da gap energetici differenti, al fine di massimizzarne l'assorbimento. In primo luogo sono state analizzate le caratteristiche della radiazione solare, assimilabile a quella di un Corpo Nero. Successivamente, dopo una presentazione introduttiva dei parametri principali dei dispositivi fotovoltaici, sono state studiate in dettaglio le celle a giunzione singola, sia da un punto di vista elettrico che termodinamico. Infine è stato esposto il funzionamento delle celle tandem, dando particolare risalto alla risposta elettrica e ai limiti di efficienza di questi dispositivi.

Il fotovoltaico (FV) costituisce il modo più diretto di conversione dell’energia solare in energia elettrica e si basa sull’effetto osservato da Becquerel nel 1839. Si può affermare che tale effetto è rimasto una curiosità di laboratorio dalla metà del XIX secolo fino al 1954, quando la prima cella solare in silicio con un’efficienza di conversione del 6% fu costruita ai Laboratori Bell. Da allora la ricerca in questo settore ha sperimentato una crescita costante ed oggi si può contare su tecnologie mature, in grado di sviluppare alte prestazioni in via di ulteriore miglioramento. Le celle tandem costituiscono ora il migliore esempio di dispositivi fotovoltaici in grado di convertire buona parte della potenza irraggiata dal sole. Aumentare l’efficienza con le celle tandem significa sfruttare le differenti lunghezze d’onda dello spettro solare. Questi dispositivi sono infatti costruiti impilando semiconduttori, disponendoli dal basso in modo crescente secondo i loro valori di energia di gap. A partire dall’analisi delle caratteristiche principali della radiazione solare e del principio di funzionamento delle celle fotovoltaiche, questo lavoro si propone di mettere in evidenza le potenzialità della tecnologia a multigiunzione, che ha già dimostrato rilevanti capacità di ottimizzazione delle prestazioni delle celle solari, facendo ben sperare per il futuro.

The development of procedures for the iridium catalyzed C-H borylation of 1-aryl pyrazolopyrimidines and 1-aryl indazoles is reported. Investigation on the activity of the catalyst revealed the combination of an iridium (I) precursor and tetramethylphenantroline as the best catalytic system. Moreover, the procedures are regioselective resulting in the selective borylation of different C-H bonds within the substrates. The application of C-H borylation to late stage functionalization is demonstrated: a biologically active compound in AstraZeneca's project underwent tandem borylation/oxidation reaction, in order to obtain a functionalized product containing an OH group.

The primary goal of this work is related to the extension of an analytic electro-optical model. It will be used to describe single-junction crystalline silicon solar cells and a silicon/perovskite tandem solar cell in the presence of light-trapping in order to calculate efficiency limits for such a device. In particular, our tandem system is composed by crystalline silicon and a perovskite structure material: metilammoniumleadtriiodide (MALI). Perovskite are among the most convenient materials for photovoltaics thanks to their reduced cost and increasing efficiencies. Solar cell efficiencies of devices using these materials increased from 3.8% in 2009 to a certified 20.1% in 2014 making this the fastest-advancing solar technology to date. Moreover, texturization increases the amount of light which can be absorbed through an active layer. Using Green’s formalism it is possible to calculate the photogeneration rate of a single-layer structure with Lambertian light trapping analytically. In this work we go further: we study the optical coupling between the two cells in our tandem system in order to calculate the photogeneration rate of the whole structure. We also model the electronic part of such a device by considering the perovskite top cell as an ideal diode and solving the drift-diffusion equation with appropriate boundary conditions for the silicon bottom cell. We have a four terminal structure, so our tandem system is totally unconstrained. Then we calculate the efficiency limits of our tandem including several recombination mechanisms such as Auger, SRH and surface recombination. We focus also on the dependence of the results on the band gap of the perovskite and we calculare an optimal band gap to optimize the tandem efficiency. The whole work has been continuously supported by a numerical validation of out analytic model against Silvaco ATLAS which solves drift-diffusion equations using a finite elements method. Our goal is to develop a simpler and cheaper, but accurate model to study such devices.

Questo studio ha condotto allo sviluppo di una nuova e altamente efficiente metodologia per la sintesi di diidropiranoni-spiroossindoli, in alte rese ed eccellenti eccessi enantiomerici. In particolare è stata condotta l’addizione viniloga enantioselettiva tra sistemi 3-alchilidenossindolici e isatine, sfruttando un catalizzatore chirale bifunzionale derivante dalla cincona, in grado di attivare mediante catalisi basica la posizione gamma del suddetto ossindolo e attraverso legame idrogeno l’isatina elettrofilica. Successivamente a questo stadio, in cui si ha la formazione del centro chirale, segue anellazione intramolecolare con formazione di un ciclo lattonico e infine apertura dell’anello ossindolico che dà origine ai diidropiranoni-spiroossindoli enantiomericamente arricchiti.

A liquid chromatographic-mass spectrometric assay with atmospheric pressure chemical ionization for quantification of ondansetron and its main metabolite 8-hydroxyondansetron in human plasma was presented. The enantiomeric separation was achieved on a Chiralcel OD-R column containing cellulose tris-(3,5-dimethylphenylcarbamate). The validation data were within the required limits. The assay was successfully applied to authentic plasma samples. Quantitative results from postoperative patients receiving ondansetron demonstrated a great interindividual variability in postoperative plasma drug concentrations, the metabolites were not detected in their unconjugated form. A wide variation in the S-(+)-/R-(-)-ondansetron concentration ratio between 0.14 and 7.18 is indicative for a stereoselective disposition or metabolism. In further studies CYP2D6 and CYP3A4 genotype dependent metabolism of ondansetron enantiomers as well as of co-administered drugs and clinical efficacy of the medication should be tested.

We describe here a new reversed-phase high-performance liquid chromatography with mass spectrometry detection method for quantifying intact cytokinin nucleotides in human K-562 leukemia cells. Tandem mass spectrometry was used to identify the intracellular metabolites (cytokinin monophosphorylated, diphosphorylated, and triphosphorylated nucleotides) in riboside-treated cells. For the protein precipitation and sample preparation, a trichloroacetic acid extraction method is used. Samples are then back-extracted with diethyl ether, lyophilized, reconstituted, and injected into the LC system. Analytes were quantified in negative selected ion monitoring mode using a single quadrupole mass spectrometer. The method was validated in terms of retention time stabilities, limits of detection, linearity, recovery, and analytical accuracy. The developed method was linear in the range of 1-1,000 pmol for all studied compounds. The limits of detection for the analytes vary from 0.2 to 0.6 pmol.

Protein is an essential component for life, and its synthesis is mediated by codons in any organisms on earth. While some codons encode the same amino acid, their usage is often highly biased. There are many factors that can cause the bias, but a potential effect of mononucleotide repeats, which are known to be highly mutable, on codon usage and codon pair preference is largely unknown. In this study we performed a genomic survey on the relationship between mononucleotide repeats and codon pair bias in 53 bacteria, 68 archaea, and 13 eukaryotes. By distinguishing the codon pair bias from the codon usage bias, four general patterns were revealed: strong avoidance of five or six mononucleotide repeats in codon pairs; lower observed/expected (o/e) ratio for codon pairs with C or G repeats (C/G pairs) than that with A or T repeats (A/T pairs); a negative correlation between genomic GC contents and the o/e ratios, particularly for C/G pairs; and avoidance of C/G pairs in highly conserved genes. These results support natural selection against long mononucleotide repeats, which could induce frameshift mutations in coding sequences. The fact that these patterns are found in all kingdoms of life suggests that this is a general phenomenon in living organisms. Thus, long mononucleotide repeats may play an important role in base composition and genetic stability of a gene and gene functions.
