We report several classes of human interspersed repeats that resemble fossils of DNA transposons, elements that move by excision and reintegration in the genome, whereas previously characterized mammalian repeats all appear to have accumulated by retrotransposition, which involves an RNA intermediate. The human genome contains at least 14 families and > 100,000 degenerate copies of short (180-1200 bp) elements that have 14- to 25-bp terminal inverted repeats and are flanked by either 8 bp or TA target site duplications. We describe two ancient 2.5-kb elements with coding capacity, Tigger1 and -2, that closely resemble pogo, a DNA transposon in Drosophila, and probably were responsible for the distribution of some of the short elements. The deduced pogo and Tigger proteins are related to products of five DNA transposons found in fungi and nematodes, and more distantly, to the Tc1 and mariner transposases. They also are very similar to the major mammalian centromere protein CENP-B, suggesting that this may have a transposase origin. We further identified relatively low-copy-number mariner elements in both human and sheep DNA. These belong to two subfamilies previously identified in insect genomes, suggesting lateral transfer between diverse species.

As queimaduras na infância compreendem importante causa de atendimento hospitalar e internação e podem desencadear sequelas físicas e emocionais. Dependendo da gravidade e complexidade das lesões, a hospitalização da criança é prolongada para realização de tratamento adequado e ela é afastada tanto do convívio familiar como do social. O número de crianças em idade escolar que sofrem queimaduras, e consequentemente interrompem as atividades escolares, por períodos curtos ou até as abandonam, é significativo, tornando digno de preocupação o processo de reinserção escolar. Este estudo teve como objetivo interpretar os significados do processo de reinserção escolar de crianças sobreviventes de queimaduras. Realizou-se uma pesquisa com abordagem metodológica qualitativa, fundamentada no referencial teórico da Antropologia Interpretativa e no método etnográfico. Após a aprovação do estudo pelos Comitês de Ética em Pesquisa com Seres Humanos das instituições envolvidas, convidaram-se crianças sobreviventes de queimaduras em seguimento em um hospital-escola do interior paulista. Coletaram-se dados no hospital, no domicílio e na escola das crianças, de janeiro de 2012 a dezembro de 2013, por meio de entrevistas em profundidade audiogravadas e observação participante, complementadas pelo diário de campo. Participaram da pesquisa 14 crianças e os atores sociais envolvidos neste processo, como familiares, profissionais de saúde, professores, vizinha e amigo, totalizando 57 participantes. A coleta e análise dos dados ocorreram concomitantemente, e esta última seguiu os pressupostos da análise temática indutiva. Identificaram-se códigos, os quais, posteriormente, embasaram a construção das duas unidades de sentidos: \"Fatos e fatores prévios ao retorno escolar\" e \"A volta à escola\". A partir destas, identificou-se o núcleo temático, \"O olho puxa essas coisas. A gente olha mesmo\": enxergando o outro como diferente, o qual apresenta a explicação compreensiva do processo de reinserção escolar, fornecendo os significados da experiência. Os significados foram explicados por meio dos conceitos antropológicos de estigma, identidade e corporeidade. O culto ao corpo presente no contexto cultural brasileiro foi fator intensificador do estigma sofrido pelas crianças. Identificaram-se fatores facilitadores do processo, como: dar continuidade às atividades escolares e manter contato com professores e colegas durante a hospitalização; preparar a escola para receber a criança; abordar sobre queimadura na escola para evitar curiosidade; encarar a presença da discriminação e trabalhar as diferenças. Estas estratégias fazem com que todos os envolvidos se sintam ao menos mais confortáveis durante esse difícil processo que merece atenção e envolvimento dos familiares, profissionais de saúde e equipe escolar. A partir dos resultados desta pesquisa, esperamos empoderar as crianças e seus familiares acerca da melhor maneira para lidar com as queimaduras durante a reinserção escolar e contribuir para o desenvolvimento de ações e estratégias baseadas na cultura para que este processo ocorra da forma menos traumática possível

Introdução: A violência doméstica contra as mulheres apresenta-se, na atualidade, como relevante questão social e problema de saúde pública que afeta a maioria dos países. Portanto, não está mais restrita a países considerados do terceiro mundo e tende a ampliarse e a se generalizar. Em Moçambique pouco se investigou sobre a importância e papel das Delegacias de Mulheres no processo de implementação da Lei 29/2009 sobre Violência Doméstica contra a Mulher. Objetivo: Conhecer como profissionais de Delegacias de Mulheres acolhem e encaminham aos Tribunais os casos de violência doméstica; como interpretam o texto da lei, seu alcance, limitação e desafios na defesa dos direitos das mulheres vivendo em situação de violência na cidade de Maputo. Metodologia: Trata-se de pesquisa de natureza qualitativa, na qual foram realizadas entrevistas com 21 profissionais, operadores de Delegacias da cidade de Maputo Moçambique, que autorizaram a entrevista, segundo Termo de Consentimento Livre e Esclarecido. Resultados e Discussão: Os entrevistados apresentaram, em seus relatos, grande identificação com o tipo de trabalho exercido, considerando as Delegacias como espaços significativos para o reconhecimento da cidadania das mulheres, assim como um campo de atuação políticojurídica em defesa dos direitos das mesmas via atuação na implementação da Lei 29/2009. Reconhecem a relevância da Lei, tendo em vista a significativa expressão da violência como prática naturalizada, na sociedade moçambicana, pela cristalização de valores tradicionais do poder masculino sobre as mulheres, secundados por diferentes práticas culturais. Tais entrevistados Identificam alcances, mas igualmente limites no processo de implementação da mesma, destacando a importância de sua maior divulgação em todas as regiões do país, quer urbanas, sobretudo nas periferias, assim como zonas rurais. Destacam, igualmente, a necessidade de maior capacitação dos próprios agentes do setor jurídico, considerando a multiplicidade de aspectos envolvidos na prática cotidiana dos serviços, notadamente nas delegacias, em relação à aplicação e ampliação do alcance da Lei. Considerações Finais: As tradicionais práticas culturais vigentes na sociedade moçambicana apresentaram-se como aspecto limitante para a implementação da Lei, assim como a remissão das penas, prevista na Lei, que, impossibilita a punição do agressor, em termos de detenção, uma vez que substitui a mesma por prestação de serviços à comunidade, pagamento de cestabásica e/ou multas, tendo em vista, sobretudo, sua estreita relação com o artigo 37 sobre a salvaguarda da família. Diante disso, foram sugeridas, para maior alcance e efetividade na aplicação da Lei, modalidades como o trabalho reflexivo com grupos de homens, com o objetivo de desconstrução dos tradicionais valores sobre masculinidade vigentes nas relações entre homens e mulheres nessa sociedade, que mantém tanto a desigualdade quanto a iniquidade de gênero, pela permanência de tradicionais valores culturais, a exemplo do lobolo e da poligamia

Estima-se que 52% da população mundial faz uso de álcool, sendo a droga mais consumida no mundo. Ao usuário, o álcool torna-se prejudicial devido às consequências nos níveis biológicos, sociais e funcionais. Assim, a redução do uso abusivo da substância é um dos objetivos da Organização Mundial de Saúde (OMS) e uma das prioridades na agenda de saúde pública mundial. No Brasil, a Política do Ministério da Saúde para a Atenção Integral aos Usuários de Álcool e Outras Drogas teve como objetivo a criação de uma rede de atenção integral a eles - a RAPS (Rede de Atenção Psicossocial). A RAPS é considerada um grande avanço da Reforma Psiquiátrica, já que integra os diversos pontos de atenção disponíveis no Sistema Único de Saúde (SUS). Um dos pontos da RAPS é a Atenção Básica (AB), que através da atuação das equipes da Estratégia Saúde da Família (ESF) tem a possibilidade de monitoração, prevenção do uso e colaboração na reinserção social dos usuários de álcool e outras drogas devido à proximidade e criação de vínculo entre o serviço e usuário. Para que o vínculo seja estabelecido o Agente Comunitário de Saúde (ACS) é a peça fundamental, visto que conhece a comunidade e reconhece suas necessidades, além de ser a figura que medeia as relações entre a equipe de saúde e os usuários. Assim sendo, o objetivo deste estudo foi descrever e analisar o discurso de ACS sobre o uso de álcool e a assistência prestada na AB. Trata-se de um estudo qualitativo de teor descritivo, cuja pesquisa ocorreu em cinco municípios da região central do Estado de Santa Catarina. Foram realizadas entrevistas semiestruturadas, analisadas através do método da Análise de Conteúdo. A análise das entrevistas resultou na formulação de duas categorias e quatro subcategorias empíricas. Os resultados evidenciaram que os ACS percebem o consumo de álcool como inerente a população em virtude da cultura caracterizada pelo consumo habitual e festivo da droga. Eles percebem que o uso do álcool torna-se um problema quanto à definição social atribuída pela comunidade, ressaltando as consequências para a família e outras perdas vivenciadas pelos usuários com base nas repercussões sociais. Quanto à assistência prestada por eles aos usuários de álcool, os resultados indicaram uma prática desprovida de instrumentos ou habilidades para a abordagem adequada do uso, evidenciando uma prática infundada pelos ACS. A prática está pautada também nas crenças em relação aos usuários de álcool, que estão muito ligadas aos estigmas relacionados a estes usuários em geral e não em evidências científicas. Conclui-se que a partir do conhecimento das percepções e práticas deste profissional, é possível direcionar ações que potencialize a prática dos ACS, já que são profissionais com grandes possibilidades de atuação diante da prevenção e tratamento do abuso de álcool e reabilitação social do usuário

For more than half of a century, Colombia has been living in a state of violence, a nationwide political violence. As the time goes by, this situation gets even worse. Now the violence is implanted for different interests, such as personal, political, social and economical interests. For this reason, the information for this thesis was gathered from the time of the independence, through the era of violence until today; considering a reflection that begins with the perspective of Marx, passing by a theoretical compilation of the conflict, with an anthropological, psychological, biological and sociological perspective. In addition, different theories about violence have been studied to recognize the ideological approach of the different armed movements that have emerged in Colombia. Statistical, economical and social data including class struggle, social stratification, exclusion and gender perspective among others, have been considered from an anthropological and interdisciplinary approach for studying the violence. A strong interest in the social reintegration process, through an ethnographic study, based on the actors backgrounds, their lives, experiences and their geographical location have provided information that will allow to know the reality of those who are living in the process, those who survive it and those who go on with their lives in society, and those who after doing it return to the armed groups. Population displaced by violence, refugees, has contributed to this study about their possibilities to return to the society or to be excluded by it. With this purpose, a theoretical and a documentary analysis as well as fieldwork have been done, trying to bring forward tools and guidelines to make some progress in developing effective solutions for social integration of Colombian armed conflict victims.

Using qualitative methods, this study explored potential risk factors for suicide, as defined by Joiner's Interpersonal-Psychological Theory of Suicide (IPTS), in a population of Soldiers returning from deployment in Operation Enduring Freedom/Operation Iraqi Freedom (OEF/OIF). Sixty-eight Soldiers participated in semi-structured interviews during the period of transition from deployment to the garrison environment. These Soldiers were asked about changes in perception of pain, experiences of perceived burdensomeness, and lack of belonging. Interviews were transcribed and analyzed. A phenomenological methodology was employed (Creswell, 2006). In response to questions about perception of pain, Soldiers discussed both positive and negative changes in their experience of physical and emotional pain. When asked about experiences of perceived burdensomeness, Soldiers described changes related to deployment, such as injuries and combat related guilt, as well as changes related to transition from combat, including care seeking, reintegration into family and society, and emotional distancing. Regarding the experience of lack of belonging, Soldiers described difficulties related to the deployment, such as combat injuries, leadership roles, and individual differences, as well as difficulties related to reintegration such as symptoms of emotional numbing and distancing. Findings highlight the potential utility of IPTS in exploring both acute and chronic suicide risk factors associated with deployment and transition, as well as potential treatment strategies that may reduce suicide risk in the population of Soldiers during reintegration.

Abstract This paper investigates themes and parallels related to the traumatic experiences women face within the correctional setting and how these experiences influence women's behavior choices that increase their risk of recidivism. Intersubjective Systems Theory is used to conceptualize the distinct dynamics and impact of trauma with this particular population. Intersubjectivity also informs the changes needed to create an environment that would help women in correctional settings to heal, avoid recidivism, and foster successful community reintegration. Principles from Intersubjective Systems Theory are reviewed in this paper to demonstrate: (a) how developmental trauma impacts the lives of incarcerated women, (b) how these women's attempts at self-healing are often maladaptive and lead to arrests, (c) how the current climate in corrections leads to retraumatization and promotes later recidivism, and (d) what changes in the corrections system would promote optimal healing and better outcomes for incarcerated women. Improved outcomes are defined as healthy boundaries, empowerment in choice of relationships, improvement of social support and occupational skills, and reduction of relapse and recidivism.

Guanylate cyclase activating proteins are EF-hand containing proteins that confer calcium sensitivity to retinal guanylate cyclase at the outer segment discs of photoreceptor cells. By making the rate of cGMP synthesis dependent on the free intracellular calcium levels set by illumination, GCAPs play a fundamental role in the recovery of the light response and light adaptation. The main isoforms GCAP1 and GCAP2 also localize to the synaptic terminal, where their function is not known. Based on the reported interaction of GCAP2 with Ribeye, the major component of synaptic ribbons, it was proposed that GCAP2 could mediate the synaptic ribbon dynamic changes that happen in response to light. We here present a thorough ultrastructural analysis of rod synaptic terminals in loss-of-function (GCAP1/GCAP2 double knockout) and gain-of-function (transgenic overexpression) mouse models of GCAP2. Rod synaptic ribbons in GCAPs−/− mice did not differ from wildtype ribbons when mice were raised in constant darkness, indicating that GCAPs are not required for ribbon early assembly or maturation. Transgenic overexpression of GCAP2 in rods led to a shortening of synaptic ribbons, and to a higher than normal percentage of club-shaped and spherical ribbon morphologies. Restoration of GCAP2 expression in the GCAPs−/− background (GCAP2 expression in the absence of endogenous GCAP1) had the striking result of shortening ribbon length to a much higher degree than overexpression of GCAP2 in the wildtype background, as well as reducing the thickness of the outer plexiform layer without affecting the number of rod photoreceptor cells. These results indicate that preservation of the GCAP1 to GCAP2 relative levels is relevant for maintaining the integrity of the synaptic terminal. Our demonstration of GCAP2 immunolocalization at synaptic ribbons at the ultrastructural level would support a role of GCAPs at mediating the effect of light on morphological remodeling changes of synaptic ribbons.

We find that the formation of MWC 656 (the first Be binary containing a black hole) involves a common envelope phase and a supernova explosion. This result supports the idea that a rapidly rotating Be star can emerge out of a common envelope phase, which is very intriguing because this evolutionary stage is thought to be too fast to lead to significant accretion and spin up of the B star. We predict ∼10–100 of B-BH binaries to currently reside in the Galactic disc, among which around 1/3 contain a Be star, but there is only a small chance to observe a system with parameters resembling MWC 656. If MWC 656 is representative of intrinsic Galactic Be-BH binary population, it may indicate that standard evolutionary theory needs to be revised. This would pose another evolutionary problem in understanding black hole (BH) binaries, with BH X-ray novae formation issue being the prime example. Future evolution of MWC 656 with an ∼5 M⊙ BH and with an ∼13 M⊙ main-sequence companion on an ∼60 d orbit may lead to the formation of a coalescing BH–NS (neutron star) system. The estimated Advanced LIGO/Virgo detection rate of such systems is up to ∼0.2 yr−1. This empirical estimate is a lower limit as it is obtained with only one particular evolutionary scenario, the MWC 656 binary. This is only a third such estimate available (after Cyg X-1 and Cyg X-3), and it lends additional support to the existence of so far undetected BH–NS binaries.

Despite the ultimatum delivered in October 2010 to the French government by Viviane Reding, Vice-President of the European Commission, to adapt its national immigration law ‘to the letter’ of the Citizens Directive 2004/38, the country has continued to evict and expel Romanian and Bulgarian nationals of Roma origin. This paper examines the state of affairs with respect to France’s policy on eviction and expulsion of Roma and assesses the way in which the controversy has developed and can be understood from the perspective of citizenship of the EU. On the basis of an examination of the subsequent responses by the European Commission and the EU member states involved, as well as of a recent bilateral agreement concluded between France and Romania on the reintegration of families of Romanian citizens belonging to the Roma minority who have exercised their freedom to move, the paper suggests that there has been a paradigm shift in the priorities driving EU policy responses and politics. This shift has led to an ethnicisation of citizenship of the Union, where ethnicity increasingly plays a decisive role in the allocation and attribution of responsibility to secure and safeguard the union freedoms.

La probation est aujourd’hui profondément ancrée dans notre système de justice pénale. Parmi les programmes de surveillance communautaire (probation, condamnation avec sursis ou libération conditionnelle), elle constitue de loin le programme le plus commun, avec près de 98 000 contrevenants adultes soit 61 % de la population adulte placée sous surveillance correctionnelle au Canada. Pour autant, les chercheurs ne manifestent depuis plusieurs décennies que peu d’intérêt pour la question. La mesure se banalisant, la recherche s’étiole. L’orientation de la recherche a amené le sujet à la marge des études sur la punition. En réponse à une demande existante (Phelps, 2015), ce mémoire est guidé par une approche sociologique centrée sur l’échelle individuelle, au niveau macroscopique. Il vise à la compréhension de l’expérience des contrevenants placés en probation.

The collapse of the USSR brought about conditions conducive to the dynamic development of relations between Central Asia and China. These relations have evolved from deep mistrust to the continually growing Chinese presence primarily in the region’s economy but also increasingly in its politics. Central Asia is playing a growing role in those areas of the economy which China sees as strategic (in particular in energy and communications). China’s ambitions and capabilities with regard to new areas and its geopolitical competitors are also being tested here. For the time being, it can be said that China has achieved vast success: its offer is appealing for the region, and Beijing has outpaced its Western rivals and has seriously challenged Russia’s position in the region. It remains, however, an open question whether China will be able to maintain and secure its present achievements and trends, which will be a serious challenge given the eternal instability in the region and Russia’s reintegration ambitions.

Over the past few decades substantial progress has been achieved in the democratisation and modernisation of the countries of Southeastern Europe. The formal democratic institutions were introduced, a liberalisation of trade resulted in the reintegration of the region into the global economy and some of them successfully applied to join the EU and NATO. But the transformation process is far from over. In the past years the achievements of the political and economic transformation have been rolled back and the quality of governance has declined in many countries. To avoid a further deterioration of the situation in the region the new impulse for institutional transformation and a change of the development model are needed. This report is the outcome of a discussion held during the conference “The Freedom Challenge in Southeastern Europe – Implications for Poland”. This joint event was developed by the Centre for Eastern Studies and Ideas Lab – the President’s Expert Programme at the Chancellery of the President of the Republic of Poland. The chapters of the report reflect the main three subjects debated during the conference. In the first chapter Spasimir Domaradzki assesses the rule of law and its influence on the consolidation of democracy in the region. In the second chapter Marta Szpala focuses on economic development in Southeastern Europe. The third chapter is written by Adam Balcer and analyses the key instruments Russia is using to build influence in this region.

Maintenance of homeostasis is pivotal to all forms of life. In the case of plants, homeostasis is constantly threatened by the inability to escape environmental fluctuations, and therefore sensitive mechanisms must have evolved to allow rapid perception of environmental cues and concomitant modification of growth and developmental patterns for adaptation and survival. Re-establishment of homeostasis in response to environmental perturbations requires reprogramming of metabolism and gene expression to shunt energy sources from growth-related biosynthetic processes to defense, acclimation, and, ultimately, adaptation. Failure to mount an initial 'emergency' response may result in nutrient deprivation and irreversible senescence and cell death. Early signaling events largely determine the capacity of plants to orchestrate a successful adaptive response. Early events, on the other hand, are likely to be shared by different conditions through the generation of similar signals and before more specific responses are elaborated. Recent studies lend credence to this hypothesis, underpinning the importance of a shared energy signal in the transcriptional response to various types of stress. Energy deficiency is associated with most environmental perturbations due to their direct or indirect deleterious impact on photosynthesis and/or respiration. Several systems are known to have evolved for monitoring the available resources and triggering metabolic, growth, and developmental decisions accordingly. In doing so, energy-sensing systems regulate gene expression at multiple levels to allow flexibility in the diversity and the kinetics of the stress response.
