In this study we investigated predation rates on third instar larvae of Chrysomya putoria and C. megacephala by third instar larvae of C. albiceps in a two-choice situation. The highest predation rate occurred on C. putoria larvae and this result is compared to previous experiments, in which C. macellaria larvae were present. Our results suggest that, when C. macellaria is absent C. albiceps larvae attack more C. putoria than C. megacephala larvae. Prey choice decisions and its implications for introduced and native blowflies are discussed.

The Primary Care Steering Group met on the 23rd and 24th September 2003 to discuss the ongoing implementation of the Primary Care Strategy in the context of the Health Service Reform Programme and to make recommendations in this regard to the Secretary General of the Department of Health & Children. At the outset the steering group acknowledges that the reform programme provides significant opportunity for the successful implementation of the primary care strategy. The implementation phase of ten project Primary Care Teams (PCTs) has to-date provided valuable information in relation to current barriers to successful implementation which may now be addressed within the overall reform process. Key themes arising from the discussions formed the basis for the recommendations which are considered by the steering group to be critical elements in achieving successful reform.

This paper characterizes and evaluates the potential of three commercial CT iterative reconstruction methods (ASIR?, VEO? and iDose(4 ()?())) for dose reduction and image quality improvement. We measured CT number accuracy, standard deviation (SD), noise power spectrum (NPS) and modulation transfer function (MTF) metrics on Catphan phantom images while five human observers performed four-alternative forced-choice (4AFC) experiments to assess the detectability of low- and high-contrast objects embedded in two pediatric phantoms. Results show that 40% and 100% ASIR as well as iDose(4) levels 3 and 6 do not affect CT number and strongly decrease image noise with relative SD constant in a large range of dose. However, while ASIR produces a shift of the NPS curve apex, less change is observed with iDose(4) with respect to FBP methods. With second-generation iterative reconstruction VEO, physical metrics are even further improved: SD decreased to 70.4% at 0.5 mGy and spatial resolution improved to 37% (MTF(50%)). 4AFC experiments show that few improvements in detection task performance are obtained with ASIR and iDose(4), whereas VEO makes excellent detections possible even at an ultra-low-dose (0.3 mGy), leading to a potential dose reduction of a factor 3 to 7 (67%-86%). In spite of its longer reconstruction time and the fact that clinical studies are still required to complete these results, VEO clearly confirms the tremendous potential of iterative reconstructions for dose reduction in CT and appears to be an important tool for patient follow-up, especially for pediatric patients where cumulative lifetime dose still remains high.

Treatment of cancer using gene therapy is based on adding a property to the cell leading to its elimination. One possibility is the use of suicide genes that code for enzymes that transform a pro-drug into a cytotoxic product. The most extensively used is the herpes simplex virus thymidine kinase (TK) gene, followed by administration of the antiviral drug ganciclovir (GCV). The choice of the promoter to drive the transcription of a transgene is one of the determinants of a given transfer vector usefulness, as different promoters show different efficiencies depending on the target cell type. In the experiments presented here, we report the construction of a recombinant adenovirus carrying TK gene (Ad-TK) driven by three strong promoters (P CMV IE, SV40 and EN1) and its effectiveness in two cell types. Human HeLa and mouse CCR2 tumor cells were transduced with Ad-TK and efficiently killed after addition of GCV. We could detect two sizes of transcripts of TK gene, one derived from the close together P CMV IE/SV40 promoters and the other from the 1.5 Kb downstream EN1 promoter. The relative amounts of these transcripts were different in each cell type thus indicating a higher flexibility of this system.

RÉSUMÉ Le système rénine-angiotensine joue un rôle prépondérant dans la régulation de la pression sanguine et de la balance des sels ainsi que dans d'autres processus physiologiques et pathologiques. Lorsque la pression sanguine est trop basse, les cellules juxtaglomérulaires sécrètent la rénine, qui clivera l'angiotensinogène circulant (sécrété majoritairement par le foie) pour libérer l'angiotensine I, qui sera alors transformée en angiotensine II par l'enzyme de conversion de l'angiotensine. Ce système est régulé au niveau de la sécrétion de la rénine par le rein. La rénine est une enzyme de type protéase aspartique. Elle est produite sous la forme d'un précurseur inactif de haut poids moléculaire appelé prorénine, qui peut être transformé en rénine active. Si le rôle de la prorégion de la rénine n'est pas encore connu, plusieurs études ont montré qu'elle pourrait être un auto-inhibiteur. Des travaux menés sur d'autres enzymes protéolytique ont mis en évidence un rôle de chaperon de leurs prorégions. Dans la circulation, la prorénine est majoritaire (90%) et la rénine active ne représente que 10% de la rénine circulante. L'enzyme qui transforme, in vivo, la prorénine en rénine active n'est pas connue. De même, l'endroit précis du clivage n'est pas élucidé. Dans ce travail, nous avons généré plusieurs mutants de la prorénine et les avons exprimés dans deux types cellulaires : les CV1 (modèle constitutif) et AtT-20 (modèle régulé). Nous avons montré que la prorégion joue un rôle important aussi bien dans l'acquisition de l'activité enzymatique que dans la sécrétion de la rénine, mais fonctionne différemment d'un type cellulaire à l'autre. Nous avons montré pour la première fois que la prorégion interagit de façon intermoléculaire à l'intérieur de la cellule. Les expériences de complémentation montre que l'interaction favorable de la rénine avec la prorégion dépend de la taille de cette dernière : prorénine (383 acides aminés) > pro62 (62 acides aminés) > pro43 (43 acides aminés). Par ailleurs nos résultats montrent qu'une faible partie de la rénine est dirigée vers la voie de sécrétion régulée classique tandis que la majorité est dirigée vers les lysosomes. Ceci suggère qu'une internalisation de la rénine circulante via le récepteur mannose-6-phosphate est possible. Cette dernière concernerait essentiellement la prorénine (dont les taux circulants sont 10 fois plus élevés que la rénine active). La suite de ce travail porterait sur la confirmation de cette hypothèse et l'identification de son possible rôle physiologique. SUMMARY The renin-angiotensin system is critical for the control of blood pressure and salt balance and other physiological and pathological processes. When blood pressure is too low, renin is secreted by the juxtaglomerular cells. It will cleave the N-terminus of circulating angiotensinogen (mostly secreted by the liver) to angiotensin-1, which is then transformed in angiotensin-II by the angiotensin-converting-enzyme (ACE). This system is regulated at the level of renin release. Renin, an aspartyl protease, is produced from a larger precursor (called prorenin) which is matured into active renin. Although the role of the renin proregion remains unknown, it has been reported that it could act as an autoinhibitor. Works on other proteolytic enzymes showed that their prorégion can act as chaperones. prorenin is the major circulating form of renin, while active renin represents only 10%. The enzyme which transforms, in vivo, the prorenin into active renin is unknown and the exact cleavage site remains to be elucidated. In this study, we generated some prorenin mutants, which were expressed in CV1 cells (constitutive pathway model) or AtT-20 cells (regulated pathway model). We showed that the proregion plays a pivotal role in the enzymatic activity and secretion of renin in a different manner in the two cell types. For the first time, it has been demonstrated that the proregion acts in an intermolecular way into the cell. Complementation assays showed that interaction between renin and proregion depends on the size of the proregion: prorenin (383 amino acids) > pro62 (62 amino acids) > pro43 (43 amino acids). Furthermore, our results showed that only a small amount of the cellular renin pool is targeted to the "canonical" regulated pathway and that the remaining is targeted to the lysosomes. Those results suggest a possible internalizátion of the circulating renin through the mannose-6-phosphate receptor pathway. This would mostly concern the prorenin (whose levels are ten times higher than active renin). Further studies would confirm or infirm this hypothesis and elucidate a potential physiological role.

Thymus regression upon stressing stimuli, such as infectious diseases, is followed by organ reconstitution, paralleling its development in ontogeny. A narrow window of thymus development was here studied, encompassing the pro-T lymphoid precursor expansion during specification stages, by the use of epidermal growth factor plus insulin (INS) in murine fetal thymus organ cultures. Aiming to disclose signaling pathways related to these stages, cultured thymus lobes had their RNA extracted, for the search of transcripts differentially expressed using RNAse protection assays and reverse transcriptase-polymerase chain reactions. We found no difference that could explain INS-driven thymocyte growth, in the pattern of transcripts for death/proliferation mediators, or for a series of growth factor receptors and transcriptional regulators known as essential for thymus development. Thymocyte suspensions from cultured lobes, stained for phenotype analysis by fluorescence activated cell sorting, showed a decreased staining for Notch1 protein at cell surfaces upon INS addition. We analyzed the expression of Notch-related elements, and observed the recruitment of a specific set of transcripts simultaneous and compatible with INS-driven thymocyte growth, namely, transcripts for Notch3, for its ligand Jagged2, and for Deltex1, a mediator of a poorly characterized alternative pathway downstream of the Notch receptor.

Introduction: Apoptosis plays a central role in chronic hepatitis C virus (HCV) infection. Although the activation of cell death signals has been reported, HCV infection persists in most patients suggesting a pro-survival adaptation, eventually developing hepatocellular carcinoma. This study focused on the role of mitochondria in the activation of pro- and antiapoptotic response in cells expressing HCV proteins. Materials and Methods: Human Osteosarcoma U2-OS cells inducibly expressing the HCV polyprotein; huh7.5 hepatoma cells transfected with full length HCV genome. Results: Long term induction of viral proteins in U2-OS cells induced a cyclosporine A-sensitive cytochrome c partial release from mitochondria, revealed by immunofluorescence, western blot and spectral analysis. In HCV-transfected Huh7.5 cells, release of the apoptosis inducing factor (AIF) with no apparent nuclear translocation was also observed. HCV positive cells displayed an HIF-dependent enhanced glycolysis, charachterized by up-regulation of the mitochondria-bound Hexokinase II (HKII); preliminary data on signal transduction pathway revealed the iperphosphorylation of Glycogen synthase kinase 3b(GSK3b). Conclusion: HCV causes a cell stress activating an early apoptotic response, the entity of which likely depends on the cell type. Nevertheless a wide series of cell survival mechanisms are also triggered resulting in a metabolic adaptation possibly favouring carcinogenesis. Based on our results, we propose a pro-survival mechanism linking HCV infection to inhibition of GSK-3b, stabilization of HIF1a and up-regulation of HKII, the last events causing a glycolytic shift and protecting from apoptosis.

As many metalloproteinases (MMPs), macrophage elastase (MMP-12) is able to degrade extracellular matrix components such as elastin and is involved in tissue remodeling processes. Studies using animal models of acute and chronic pulmonary inflammatory diseases, such as pulmonary fibrosis and chronic obstrutive pulmonary disease (COPD), have given evidences that MMP-12 is an important mediator of the pathogenesis of these diseases. However, as very few data regarding the direct involvement of MMP-12 in inflammatory process in the airways were available, we have instilled a recombinant form of human MMP-12 (rhMMP-12) in mouse airways. Hence, we have demonstrated that this instillation induced a severe inflammatory cell recruitment characterized by an early accumulation of neutrophils correlated with an increase in proinflammatory cytokines and in gelatinases and then by a relatively stable recruitment of macrophages in the lungs over a period of ten days. Another recent study suggests that resident alveolar macrophages and recruited neutrophils are not involved in the delayed macrophage recruitment. However, epithelial cells could be one of the main targets of rhMMP-12 in our model. We have also reported that a corticoid, dexamethasone, phosphodiesterase 4 inhibitor, rolipram and a non-selective MMP inhibitor, marimastat could reverse some of these inflammatory events. These data indicate that our rhMMP-12 model could mimic some of the inflammatory features observed in COPD patients and could be used for the pharmacological evaluation of new anti-inflammatory treatment. In this review, data demonstrating the involvement of MMP-12 in the pathogenesis of pulmonary fibrosis and COPD as well as our data showing a pro-inflammatory role for MMP-12 in mouse airways will be summarized.
