A hemorrhagic proteinase, jerdohagin, was purified from Trimeresurus jerdonii venom by gel filtration and ion-exchange chromatographies. It was a single chain polypeptide with an apparent molecular weight of 96 kDa as estimated by SDS-PAGE under the non-reducing and reducing conditions. Internal peptide sequencing indicated that it consisted of metalloproteinase, disintegrin-like and cysteine-rich domains and belonged to the class III snake venom metalloproteinases (class P-III SVMPs). Like other typical metalloproteinases, hemorrhagic activities of jerdohagin were completely inhibited by EDTA, but not by PMSF. Jerdohagin preferentially degraded a-chain of human fibrinogen. Interestingly, jerdohagin did not activate human prothrombin, whereas it cleaved human prothrombin and fragment F1 of activated human prothrombin. (C) 2004 Elsevier Ltd. All rights reserved.

Three 26 kDa proteins, named as TJ-CRVP, NA-CRVP1 and NA-CRVP2, were isolated from the venoms of Trimeresurus jerdonii and Naja atra, respectively. The N-terminal sequences of TJ-CRVP and NA-CRVPs were determined. These components were devoid of the enzymatic activities tested, such as phospholipase A(2), arginine esterase, proteolysis, L-amino acid oxidase, 5' nucleotidase, acetylcholinesterase. Furthermore, these three components did not have the following biological activities: coagulant and anticoagulant activities, lethal activity, myotoxicity, hemorrhagic activity, platelet aggregation and platelet aggregation-inhibiting activities. These proteins are named as cysteine-rich venom protein (CRVP) because their sequences showed high level of similarity with mammalian cysteine-rich secretory protein (CRISP) family. Recently, some CRISP-like proteins were also isolated from several different snake venoms, including Agkistrodon blomhoffi, Trimeresurus flavoviridis, Lanticauda semifascita and king cobra. We presumed that CRVP might be a common component in snake venoms. Of particular interest, phylogenetic analysis and sequence alignment showed that NA-CRVP1 and ophanin, both from elapid snakes, share higher similarity with CRVPs from Viperidae snakes. While carp themselves may destroy carp eggs by sucking from outside the happa, destruction in the absence of carp also occurs. During observations of the effect of electrical field on developing carp eggs it was seen that the egg membrane ruptured prematurely under certain potential differences. In natural waters and mud potential differences occur in the presence of high concentrations of certain salts compared with the internal concentration. Temperature and pH fluctuations following strong sun or heavy rain accelerates the diffusion of ions along side water through the egg membrane. The egg cells expand rapidly and the embryos were rapidly dropped.

Chunks of Labeo rohita, Cirrhinus mrigala and Catla catla wrapped in polythene film were stored at -8 to -10°C in the freezer cabinet of the refrigerator. It was found that L. rohita and C. mrigala were acceptable up to 33 days and C. catch up to 35 days. Total volatile base nitrogen, free fatty acids and degree of sponginess of the samples showed increasing trend during frozen storage.

Trichosanthin (TCS) was the first ribosome inactivating protein found to possess anti-HIV-1 activity. Phase I/II clinical trial of this compound had been done. Antigenicity and short plasma half-life were the major side effects preventing further clinical trial. Modification of TCS is therefore necessary to revive the interest to develop this compound as an anti-HIV agent. Three potential antigenic sites (Ser-7, Lys-173, and Gln-219) were identified by computer modeling. Through site-directed mutagenesis, these three antigenic amino acids were mutated to a cysteine residue resulting in 3 TCS mutants, namely S7C, K173C, and Q219C. These mutants were further coupled to polyethylene glycol with a molecular size of 20 kDa (PEG) via the cysteine residue. This produced another three TCS derivatives, namely PEG(20)k-S7C, PEG(20)k-K173C, and PEG(20)k-Q219C. PEGylation had been widely used recently to decrease immunogenicity by masking the antigenic sites and prolong plasma half-life by expanding the molecular size. The in vitro anti-HIV-1 activity of these mutants and derivatives was tested. Results showed that the anti-HIV-1 activity of S7C, K173C, and Q219C was decreased by about 1.5- to 5.5-fold with slightly lower cytotoxicity. On the other hand, PEGylation produced larger decrease (20- to 30-fold) in anti-HIV activity. Cytotoxicity was, however, weakened only slightly by about 3-fold. The in vitro study showed that the anti-HIV activity of PEGylated TCS was retained with reduced potency. The in vivo activity is expected to have only slightly changed due to other beneficial effects like prolonged half-life. (C) 2004 Elsevier Inc. All rights reserved.

A laboratory based 2x3 factorial experiment was conducted for 12 weeks to investigate the influences of dietary lipid and phosphorus (P) levels on retention and excretion of phosphorus and nitrogen (N) in fingerling red sea bream. Two levels of lipid (210 and 260 g/kg) and three levels of phosphorus (17, 14 and 12 g/kgˉ¹) in the dry diets were tested. Duplicate groups of 25 red sea bream (average weight 3.74±0.07 g) per 60L glass tank were fed experimental diets three times a day near to satiation level at 22 to 28°C water temperature. A reduction in dietary fish meal from 500 to 300 g/kg dry diet, corresponding to a supplementation in both dietary lipid and P resulted in significant increase in both P and N retention which resulted in the reduction of their excretion by red sea bream. The overall results of the present study demonstrated that both lipid and phosphorus supplementation are necessary for developing less-polluting feed which in turn, reduce fish meal level in the diet of fingerling red sea bream. Further studies in this regard with different size and age groups of red sea bream are warranted.

A study was undertaken to find out the impact of ten days fishing ban in the major spawning grounds of hilsa during October to December. The study revealed a positive impact of fishing ban during spawning season on reproduction success of hilsa. Proportionate distribution of male and female hilsa also showed a significant level of distribution in and around the spawning grounds where the ratio was 35:65. Percent of oozing hilsa during fishing ban period in the spawning grounds was observed 1.61%. About 5% spent hilsa was observed in the fish landing centers and was compared with the data of Global Environment Facility (GEF) and Bangladesh Fisheries Research Institute (BFRI) studies and was found about 2.80-3.57 times higher than that of the findings of 2002 and 2003. Estimated egg production value showed about 46,800 Kg of eggs could have been produced that indicating a positive impact of 10 days fishing ban in spawning season. Abundance of higher percentages of gravid hilsa was found which were not available in the same quantity and condition in the non-fishing ban period. During the present investigation, fairly higher amount of spent hilsa and juveniles were also observed in the spawning grounds. Along with the jatka fry, spawn and fries of other fishes were also found in higher quantity than the previous years and thus it is assumed that fishing ban also might have positive impact on the successful breeding of other fishes. Overall, the fishing ban was found effective for successful breeding of hilsa.

An investigation on growth, production and fishery of three Indian major carps: rohu, Labeo rohita, catla, Catla catla and mrigal, Cirrhinus mrigala and three exotic carps: silver carp, Hypophthalmichthys molitrix, grass carp, Ctenopharyngodon idella, and common carp, Cyprinus carpio was carried out in Nasti baor during February to April months. In catch per unit effort (CPUE) study the highest catch/day/person (3.13 kg) and catch/day/gear (40.65 kg) was recorded in the month of March for kochal fishing. In komar fishing catch/day/person (15.08 kg) and catch/day/gear (1206 kg) was also found higher in March. Komar fishing was done only in March and April and its CPUE was greater in both the months than that of kochal. The average recovery rate (combination of all six species) was 37.80 considering the stocking from July month of the previous year. The recovery rate of common carp (54.1) was the highest and lowest (13.90) in case of silver carp. When the recovery was calculated on the basis of one year data and stocking, it was 55.6%. Analysis of production model revealed that the present production (54,806 kg/year) is less than both theoretical production (model I- 85,285 kg/year and model II -75,952 kg/year) estimated. Therefore, it may be concluded that the fish production from Nasti baor could still be increased from the present level of production.

This project was done for identifying the mesopelagic fish of the Iranian waters of Oman Sea, during two year from 2008 to 2010. The specimens were collected using two trawler vessel from nine station. All the specimens were fixed in formalin then in 70% alcohol and carried to the laboratory. In total of 19 species belonged to 14 families of 6 orders identified including: Echinorhinidae, Stomidae, Phosichthyidae, Synodontidae, Paralepididae, Myctophidae, Acropomatidae, Priacanthidae, Pentacerotidae, Champsodontidae, Gempylidae, Trichiuridae, Nomeidae and Congridae. Of which 17 species were identified up to species level including: Echinorhinus brucus, Bathophilus indicus, Chauliodus sloani, Harpadon nehereus, Lestrolepis japonica, Benthosema pterotum, Diaphus garmani, Diaphus effulgens, Bolinichthys photothorax, Acropoma japonicum, Synagrops adeni, Cookeolus boops, Histiopterus typus, Champsodon sagittus, Neoepinnula orientalis, Trichiurus lepturus, Cubiceps baxteri. Vinciguerria was identified up to genus level because only one specimen caught during the survey and one species (Congridae) was identified up to family level because only 3 specimens of this fish in early stage of life were caught and their characters were not suitable for identify up to species level. The highest species belong to Myctophidae family of Myctophiformes order.

The Victoria and Kyoga lake basins form the major aquatic system of this study (Fig. I). The two lake basins share a common evolutionary history and have similar native fish faunas (Graham 1929, Worthington 1929). The two main lakes have also had similar impacts by introduction of Nile perch Lates niloticus and therefore these two lakes can be considered to be similar for ichiogeographical purposes. These lake basins have many satellite lakes isolated from one another and from the main lakes Victoria and Kyoga by swamps and other barriers. Some of these satellite lakes still possess stocks of endemic fish species which are almost extinct from the main water bodies. It was therefore considered that understanding of these lakes would contribute to the knowledge base required to solve some of the problems experienced in Lake Victoria and Kyoga especially the loss in trophic diversity arising. The study was carried out in these two main water bodies (Kyoga and Victoria) and on other satellite lakes e.g Wamala, Kachera, Mburo, Kayanja and Kayugi in the Victoria lake basin and lakes Nawampasa, Nyaguo, Agu, Gigate, Lemwa and Kawi in the Kyoga lake basin (Figs. 2, 3, 4, 5 & 6).
