Glass transition temperature of freeze-dried pineapple conditioned by adsorption at various water activities at 25 degreesC was determined by differential scanning calorimetry (DSC). High moisture content samples corresponding to water activities higher than 0.9, obtained by liquid water addition, were also analysed. The DSC traces showed a well-visible shift in baseline at the glass transition temperature (T(g)). Besides, no ice formation was observed until water activity was equal to 0.75. For water activities lower than 0.88, the glass transition curve showed that T(g) decreased with increasing moisture content and the experimental data could be well-correlated by the Gordon-Taylor equation. For higher water activities, this curve exhibited a discontinuity, with suddenly increasing glass transition temperatures approaching a constant value that corresponds to the T(g) of the maximally freeze-concentrated amorphous matrix. The unfreezable water content was determined through melting enthalpy dependence on the sample moisture content.

Observations of cosmic rays arrival directions made with the Pierre Auger Observatory have previously provided evidence of anisotropy at the 99% CL using the correlation of ultra high energy cosmic rays (UHECRs) with objects drawn from the Veron-Cetty Veron catalog. In this paper we report on the use of three catalog independent methods to search for anisotropy. The 2pt-L, 2pt+ and 3pt methods, each giving a different measure of self-clustering in arrival directions, were tested on mock cosmic ray data sets to study the impacts of sample size and magnetic smearing on their results, accounting for both angular and energy resolutions. If the sources of UHECRs follow the same large scale structure as ordinary galaxies in the local Universe and if UHECRs are deflected no more than a few degrees, a study of mock maps suggests that these three method can efficiently respond to the resulting anisotropy with a P-value = 1.0% or smaller with data sets as few as 100 events. using data taken from January 1, 2004 to July 31, 2010 we examined the 20, 30, ... , 110 highest energy events with a corresponding minimum energy threshold of about 49.3 EeV. The minimum P-values found were 13.5% using the 2pt-L method, 1.0% using the 2pt+ method and 1.1% using the 3pt method for the highest 100 energy events. In view of the multiple (correlated) scans performed on the data set, these catalog-independent methods do not yield strong evidence of anisotropy in the highest energy cosmic rays.

Atmospheric conditions at the site of a cosmic ray observatory must be known for reconstructing observed extensive air showers. The Global Data Assimilation System (GDAS) is a global atmospheric model predicated on meteorological measurements and numerical weather predictions. GDAS provides altitude-dependent profiles of the main state variables of the atmosphere like temperature, pressure, and humidity. The original data and their application to the air shower reconstruction of the Pierre Auger Observatory are described. By comparisons with radiosonde and weather station measurements obtained on-site in Malargue and averaged monthly models, the utility of the GDAS data is shown. (C) 2012 Elsevier B.V. The Pierre Auger Observatory is a facility built to detect air showers produced by cosmic rays above 10(17) eV. During clear nights with a low illuminated moon fraction, the UV fluorescence light produced by air showers is recorded by optical telescopes at the Observatory. To correct the observations for variations in atmospheric conditions, atmospheric monitoring is performed at regular intervals ranging from several minutes (for cloud identification) to several hours (for aerosol conditions) to several days (for vertical profiles of temperature, pressure, and humidity). In 2009, the monitoring program was upgraded to allow for additional targeted measurements of atmospheric conditions shortly after the detection of air showers of special interest, e. g., showers produced by very high-energy cosmic rays or showers with atypical longitudinal profiles. The former events are of particular importance for the determination of the energy scale of the Observatory, and the latter are characteristic of unusual air shower physics or exotic primary particle types. The purpose of targeted (or "rapid") monitoring is to improve the resolution of the atmospheric measurements for such events. In this paper, we report on the implementation of the rapid monitoring program and its current status. The rapid monitoring data have been analyzed and applied to the reconstruction of air showers of high interest, and indicate that the air fluorescence measurements affected by clouds and aerosols are effectively corrected using measurements from the regular atmospheric monitoring program. We find that the rapid monitoring program has potential for supporting dedicated physics analyses beyond the standard event reconstruction.

The surface detector array of the Pierre Auger Observatory can detect neutrinos with energy E-nu between 10(17) eV and 10(20) eV from point-like sources across the sky south of +55 degrees and north of -65 degrees declinations. A search has been performed for highly inclined extensive air showers produced by the interaction of neutrinos of all flavors in the atmosphere (downward-going neutrinos), and by the decay of tau leptons originating from tau neutrino interactions in Earth's crust (Earth-skimming neutrinos). No candidate neutrinos have been found in data up to 2010 May 31. This corresponds to an equivalent exposure of similar to 3.5 years of a full surface detector array for the Earth-skimming channel and similar to 2 years for the downward-going channel. An improved upper limit on the diffuse flux of tau neutrinos has been derived. Upper limits on the neutrino flux from point-like sources have been derived as a function of the source declination. Assuming a differential neutrino flux k(PS) . E-nu(-2). from a point-like source, 90% confidence level upper limits for k(PS) at the level of approximate to 5x10(-7) and 2.5x10(-6) GeV cm(-2) s(-1) have been obtained over a broad range of declinations from the searches for Earth-skimming and downward-going neutrinos, respectively.

A thorough search of the sky exposed at the Pierre Auger Cosmic Ray Observatory reveals no statistically significant excess of events in any small solid angle that would be indicative of a flux of neutral particles from a discrete source. The search covers from -90 degrees to +15 degrees in declination using four different energy ranges above 1 EeV (10(18) eV). The method used in this search is more sensitive to neutrons than to photons. The upper limit on a neutron flux is derived for a dense grid of directions for each of the four energy ranges. These results constrain scenarios for the production of ultrahigh energy cosmic rays in the Galaxy.

A thorough search for large-scale anisotropies in the distribution of arrival directions of cosmic rays detected above 10(18) eV at the Pierre Auger Observatory is presented. This search is performed as a function of both declination and right ascension in several energy ranges above 10(18) eV, and reported in terms of dipolar and quadrupolar coefficients. Within the systematic uncertainties, no significant deviation from isotropy is revealed. Assuming that any cosmic-ray anisotropy is dominated by dipole and quadrupole moments in this energy range, upper limits on their amplitudes are derived. These upper limits allow us to test the origin of cosmic rays above 10(18) eV from stationary Galactic sources densely distributed in the Galactic disk and predominantly emitting light particles in all directions.

We report a measurement of the proton-air cross section for particle production at the center-of-mass energy per nucleon of 57 TeV. This is derived from the distribution of the depths of shower maxima observed with the Pierre Auger Observatory: systematic uncertainties are studied in detail. Analyzing the tail of the distribution of the shower maxima, a proton-air cross section of [505 +/- 22(stat)(-36)(+28)(syst)] mb is found.

In 1936, the African-American intellectual W.E.B. Du Bois visited Nazi Germany for a period of five months. Two years later, the eleven-year-long American exile of the German philosopher Theodor W. Adorno began. From the latter’s perspective, the United States was the “home” of the Culture Industry. One intuitively assumes that these sojourns abroad must have amounted to “hell on earth” for both the civil rights activist W.E.B. Du Bois and the subtle intellectual Adorno. But was this really the case? Or did they perhaps arrive at totally different conclusions? This thesis deals with these questions and attempts to make sense of the experiences of both men. By way of a systematic and comparative analysis of published texts, hitherto unpublished documents and secondary literature, this dissertation first contextualizes Du Bois’s and Adorno’s transatlantic negotiations and then depicts them. The panoply of topics with which both men concerned themselves was diverse. In Du Bois’s case it encompassed Europe, science and technology, Wagner operas, the Olympics, industrial education, race relations, National Socialism and the German Africanist Diedrich Westermann. The opinion pieces which Du Bois wrote for the newspaper “Pittsburgh Courier” during his stay in Germany serve as a major source for this thesis. In his writings on America, Adorno concentrated on what he regarded as the universally victorious Enlightenment and the predominance of mass culture. This investigation also sheds light on the correspondences between the philosopher and Max Horkheimer, Thomas Mann, Walter Benjamin, Siegfried Kracauer and Oskar and Maria Wiesengrund. In these autobiographical texts, Adorno’s thoughts revolve around such diverse topics as the American landscape, his fears as German, Jew and Left-Hegelian as well as the loneliness of the refugee. This dissertation has to refute the intuitive assumption that Du Bois’s and Adorno’s experiences abroad were horrible events for them. Both men judged the foreign countries in which they were staying in an extremely differentiated and subtle manner. Du Bois, for example, was not racially discriminated against in Germany. He was also delighted by the country’s rich cultural offerings. Adorno, for his part, praised the U.S.’s humanity of everyday life and democratic spirit. In short: Although both men partly did have to deal with utterly negative experiences, the metaphor of “hell on earth” is simply untenable as an overall conclusion.

Im Rahmen dieser Arbeit wurde untersucht, welche Rolle die Marginalisierungspolitikrnin Sudan in Bezug auf den Staatsbildungs- und Demokratisierungsprozessrnspielt. Dabei wurde der Schwerpunkt auf die Region Darfur gelegt.rnZu Beginn der Arbeit werden die besonderen Verhältnisse im Sudan beleuchtet,rndie geprägt sind durch eine jahrzehntelange Kolonialpolitik, die auch nach der Unabhängigkeitrndes Sudans durch die herrschenden Eliten im eigenen Land gegenüberrnden peripheren Gebieten weitergeführt worden ist.rnEs wurde aufgezeigt, dass Marginalisierungspolitik unterschiedliche Ursachen hat.rnIm Sudan dient diese Politik dem Machterhalt bestimmter Gruppen und damit verbundenrnauch Vorteilen in allen Bereichen der Politik, Wirtschaft und Gesellschaft.rnDer Autor hat dargelegt, in welchen Formen Marginalisierung stattfindet. Beispielerndafür sind:rn• eine Sprachpolitik, die neben Arabisch keine andere Sprache zulässtrn• eine gezielte Politik der Vernachlässigung der peripheren Gebiete, die sichrnin einer fehlenden Infrastruktur und Bildung zeigt.rn• Erschwerter Zugang zu öffentlichen Ämtern, je nachdem aus welcher ethnischenrnGruppe jemand kommtrnDie Auswirkungen dieser Marginalisierungspolitik sind Verhinderung von Entwicklung,rnArmut, innerstaatliche Migration, Bürgerkriege und schließlich der Zerfall desrnStaates.rnAm Beispiel von der Region Darfur untermauert der Autor nochmals diese Vernachlässigungspolitikrnund begründet damit den Widerstand von Rebellengruppenrnseit 2003. Diese sind die Frage der Religion und Staatenbildungrnsowie die Frage, in welcher Form eine Demokratisierung Sudans stattfindenrnkann.rnEr stellt mögliche Ansätze zur Transformation des politischen Systems vor, um diernMarginalisierungspolitik zu beenden und das Land zu stabilisieren. Diese Transformationrnmuss in mehreren Phasen erfolgen. Als erstes muss das diktatorischernRegime abgelöst werden, um im Anschluss daran, die Demokratie zu institutionalisierenrnund schließlich zu konsolidieren.rnWenn dieser Weg nicht beschritten wird, sieht der Autor keine Zukunft für eine sudanesischernEinheit, in der alle Ethnien gleichberechtigt in allen Bereichen wie Politik,rnWirtschaft und Gesellschaft vertreten sein müssen. In diesem Fall wird esrnzwangsläufig zu einem Zerfall des Landes kommen.

The physicochemical properties of nanoparticles make them suitable for biomedical applications. Due to their ‘straight-forward’ synthesis, their known biocompatibility, their strong optical properties, their ability for targeted drug delivery and their uptake potential into cells gold nanoparticles are highly interesting for biomedical applications. In particular, the therapy of brain diseases (neurodegenerative diseases, ischemic stroke) is a challenge for contemporary medicine and gold nanoparticles are currently being studied in the hope of improving drug delivery to the brain.rnIn this thesis three major conclusions from the generated data are emphasized.rn1. After improvement of the isolation protocol and culture conditions, the formation of a monolayer of porcine brain endothelial cells on transwell filters lead to a reproducible and tight in vitro monoculture which exhibited in vivo blood brain barrier (BBB) characteristics. The transport of nanoparticles across the barrier was studied using this model.rn2. Although gold nanoparticles are known to be relatively bioinert, contaminants of the nanoparticle synthesis (i.e. CTAB or sodium citrate) increased the cytotoxicity of gold nanoparticles, as shown by various publications. The results presented in this thesis demonstrate that contaminants of the nanoparticle synthesis such as sodium citrate increased the cytotoxicity of the gold nanoparticles in endothelial cells but in a more dramatic manner in epithelial cells. Considering the increased uptake of these particles by epithelial cells compared to endothelial cells it was demonstrated that the observed decrease of cell viability appeared to be related to the amount of internalized gold nanoparticles in combination with the presence of the contaminant.rn3. Systematically synthesized gold nanoparticles of different sizes with a variety of surface modifications (different chemical groups and net charges) were investigated for their uptake behaviour and functional impairment of endothelial cells, one of the major cell types making up the BBB. The targeting of these different nanoparticles to endothelial cells from different parts of the body was investigated in a comparative study of human microvascular dermal and cerebral endothelial cells. In these experiments it was demonstrated that different properties of the nanoparticles resulted in a variety of uptake patterns into cells. Positively charged gold nanoparticles were internalized in high amounts, while PEGylated nanoparticles were not taken up by both cell types. Differences in the uptake behavior were also demonstrated for neutrally charged particles of different sizes, coated with hydroxypropylamine or glucosamine. Endothelial cells of the brain specifically internalized 35nm neutrally charged hydroxypropylamine-coated gold nanoparticles in larger amounts compared to dermal microvascular endothelial cells, indicating a "targeting" for brain endothelial cells. Co-localization studies with flotillin-1 and flotillin-2 showed that the gold nanoparticles were internalized by endocytotic pathways. Furthermore, these nanoparticles exhibited transcytosis across the endothelial cell barrier in an in vitro BBB model generated with primary porcine brain endothelial cells (1.). In conclusion, gold nanoparticles with different sizes and surface characteristics showed different uptake patterns in dermal and cerebral endothelial cells. In addition, gold nanoparticles with a specific size and defined surface modification were able to cross the blood-brain barrier in a porcine in vitro model and may thus be useful for controlled delivery of drugs to the brain.
