The hybrid created from the crossbreeding of European and African bees, known as the Africanised bee, has provided numerous advantages for current beekeeping. However, this new species exhibits undesirable behaviours, such as colony defence instinct and a propensity to attack en masse, which can result in serious accidents. To date, there is no effective treatment for cases of Africanised bee envenomation. One promising technique for developing an efficient antivenom is the use of phage display technology, which enables the production of human antibodies, thus avoiding the complications of serum therapy, such as anaphylaxis and serum sickness. The aim of this study was to produce human monoclonal single-chain Fv (scFv) antibody fragments capable of inhibiting the toxic effects of Africanised bee venom. We conducted four rounds of selection of antibodies against the venom and three rounds of selection of antibodies against purified melittin. Three clones were selected and tested by enzyme-linked immunosorbent assay to verify their specificity for melittin and phospholipase A2. Two clones (C5 and C12) were specific for melittin, and one (A7) was specific for phospholipase A2. In a kinetic haemolytic assay, these clones were evaluated individually and in pairs. The A7-C12 combination had the best synergistic effect and was chosen to be used in the assays of myotoxicity inhibition and lethality. The A7-C12 combination inhibited the in vivo myotoxic effect of the venom and increased the survival of treated animals.

Using Fluorescence Recovery After Photobleaching, we investigate the Brownian motion of DNA rod-like fragments in two distinct anisotropic phases with a local nematic symmetry. The height of the measurement volume ensures the averaging of the anisotropy of the in-plane diffusive motion parallel or perpendicular to the local nematic director in aligned domains. Still, as shown in using a model specifically designed to handle such a situation and predicting a non-Gaussian shape for the bleached spot as fluorescence recovery proceeds, the two distinct diffusion coefficients of the DNA particles can be retrieved from data analysis. In the first system investigated (a ternary DNA-lipid lamellar complex), the magnitude and anisotropy of the diffusion coefficient of the DNA fragments confined by the lipid bilayers are obtained for the first time. In the second, binary DNA-solvent system, the magnitude of the diffusion coefficient is found to decrease markedly as DNA concentration is increased from isotropic to cholesteric phase. In addition, the diffusion coefficient anisotropy measured within cholesteric domains in the phase coexistence region increases with concentration, and eventually reaches a high value in the cholesteric phase.

The cationic lipid dioctadecyldimethylammonium bromide (DODAB) and the CpG oligonucleotide (CpG) have been separately used as potent immunoadjuvants driving Th1 responses. Here DODAB bilayer fragments (BF) and CpG (5 -TTGACGTTCG-3) assemblies have their physical properties and immunoadjuvant activity determined using ovalbumin (OVA) as a model antigen. At 0.1 mg/mL OVA, the dependence of DODAB BF/OVA size and zeta-potential on time and [DODAB] establishes 0.1 mMDODAB as suitable for obtaining stable and cationic DODAB BF/OVA assemblies. At 0.1 mMDODAB, 0.1 mg/mL OVA and 0.006 mMCpG, the zeta-potential is zero. At [CpG]>0.006 mM, good colloidal stability for the anionic assemblies is due to charge overcompensation. At 0.020 mM CpG, these DODAB BF/OVA/CpG assemblies are highly effective in vivo generating responses similar to those elicited by the stable and cationic DODAB BF/OVA. The anti-OVA DTH reaction and the secretion of IFN-gamma and IL-12 are 6, 42 and 9 times larger for the DODAB BF/OVA/CpG-immunized mice than the same responses by OVA-immunized mice, respectively. Leptospira, the causative agent of leptospirosis, interacts with several host molecules, including extracellular matrix components, coagulation cascade proteins, and human complement regulators. Here we demonstrate that acquisition of factor H (FH) on the Leptospira surface is crucial for bacterial survival in the serum and that these spirochetes, besides interacting with FH, FH related-1, and C4b binding protein (C4BP), also acquire FH like-1 from human serum. We also demonstrate that binding to these complement regulators is mediated by leptospiral immunoglobulin-like (Lig) proteins, previously shown to interact with fibronectin, laminin, collagen, elastin, tropoelastin, and fibrinogen. Factor H binds to Lig proteins via short consensus repeat domains 5 and 20. Competition assays suggest that FH and C4BP have distinct binding sites on Lig proteins. Moreover, FH and C4BP bound to immobilized Ligs display cofactor activity, mediating C3b and C4b degradation by factor I. In conclusion, Lig proteins are multifunctional molecules, contributing to leptospiral adhesion and immune evasion.

The affinity of the d-galactose-binding lectin from Artocarpus heterophyllus lectin, known as jacalin, with immonuglobulins (Igs) was determined by biofunctionalization of a piezoelectric transducer. This piezoelectric biofunctionalized transducer was used as a mass-sensitive analytical tool, allowing the real-time binding analysis of jacalin-human immunoglobulin A1 (IgA(1)) and jacalin-bovine IgG(1) interactions from which the apparent affinity constant was calculated. The strategy was centered in immobilizing jacalin on the gold electrode's surface of the piezoelectric crystal resonator using appropriate procedures based on self-assembling of 11-mercaptoundecanoic acid and 2-mercaptoethanol thiol's mixture, a particular immobilization strategy by which it was possible to avoid cross-interaction between the proteins over electrode's surface. The apparent affinity constants obtained between jacalin-human IgA(1) and jacalin-bovine IgG(1) differed by 1 order of magnitude [(8.0 +/- 0.9) x 10(5) vs (8.3 +/- 0.1) x 10(6) L mol(-1)]. On the other hand, the difference found between human IgA(1) and human IgA(2) interaction with jacalin, eight times higher for IgA(1), was attributed to the presence of O-linked glycans in the IgA(1) hinge region, which is absent in IgA(2). Specific interaction of jacalin with O-glycans, proved to be present in the human IgA(1) and hypothetically present in bovine IgG(1) structures, is discussed as responsible for the obtained affinity values.

Seventy-seven males of Hypsiboas prasinus from 2 Atlantic forest fragments in the municipalities of Botucatu and Jundiai, sao Paulo State, Brazil, were examined for endoparasites. The frogs were captured in summer (January until March) and winter (July/August) of 2008 and 2009. Thirty-three males (75%) from Botucatu were infected with Rhabdias cf. fuelleborni, cosmocerciid nematodes, and Cylindrotaenia americana. Twenty-five tree frogs (78.5%) from Jundiai were infected by Rhabdias cf. fuelleborni, Physaloptera sp., and cosmocerciid nematodes. Only cosmocerciid nematodes presented a statistically significance difference in prevalence (z = 4.345; P < 0.001) and mean abundance (t = 562.0; P < 0.001) between Botucatu and Jundiai during the winter. Also, the cosmocerciids exhibited higher mean abundance (t = 196.0; P = 0.034) in winter when compared with summer at the Jundiai site. Moreover, to our knowledge, this is the first report of C. americana in the Brazilian Hylidae. This study presents 4 new records of nematodes in H. prasinus.

Questions What are the main features of the seed rain in a fragmented Atlantic forest landscape? Can seed rain species attributes (life form, dispersal mode, successional status) relate to the spatial arrangement (size and number of fragments, edge density and presence of corridor) of forest fragments in the landscape? How does the rain forest landscape structure affect the seed rain? Location Atlantic rainforest, Sao Paulo State, Southeastern Brazil. Methods Seed rain samples were collected monthly throughout 1yr, counted, identified and classified according to species dispersal mode, successional status and life form. Seed rain composition was compared with woody species near the seed traps. Relationships between seed rain composition and landscape spatial arrangement (fragment area, presence of corridor, number of fragments in the surroundings, proximity of fragments, and edge density) were tested using canonical correspondence analysis (CCA). Results We collected 20142 seeds belonging to 115 taxa, most of them early successional and anemochorous trees. In general, the seed rain had a species composition distinct from that of the nearby forest tree community. Small isolated fragments contained more seeds, mainly of anemochorous, epiphytic and early-successional species; large fragments showed higher association with zoochorous and late-successional species compared to small fragments. The CCA significantly distinguished the species dispersal mode according to fragment size and isolation, anemochorous species being associated to small and isolated fragments, and zoochorous species to larger areas and fragment aggregation. Nevertheless, a gradient driven by proximity (PROX) and edge density (ED) segregated lianas (in the positive extremity), early successional and epiphyte species (in the negative end); large fragments were positively associated to PROX and ED. Conclusions The results highlight the importance of the size and spatial arrangement of forest patches to promote habitat connectivity and improve the flux of animal-dispersed seeds. Landscape structure controls seed fluxes and affects plant dispersal capacity, potentially influencing the composition and structure of forest fragments. The seed rain composition may be used to assess the effects of landscape spatial structure on plant assemblages, and provide relevant information for biodiversity conservation.

The interaction between the antimicrobial peptide gramicidin (Gr) and dipalmitoylphosphatidylcholine (DPPC)/dioctadecyldimethylammonium bromide (DODAB) 1:1 large unilamellar vesicles (LVs) or bilayer fragments (BFs) was evaluated by means of several techniques. The major methods were: 1) Gr intrinsic fluorescence and circular dichroism (CD) spectroscopy; 2) dynamic light scattering for sizing and zeta-potential analysis; 3) determination of the bilayer phase transition from extrinsic fluorescence of bilayer probes; 4) pictures of the dispersions for evaluation of coloidal stability over a range of time and NaCl concentration. For Gr in LVs, the Gr dimeric channel conformation is suggested from: 1) CD and intrinsic fluorescence spectra similar to those in trifluoroethanol (TFE); 2) KCl or glucose permeation through the LVs/Gr bilayer. For Gr in BFs, the intertwined dimeric, non-channel Gr conformation is evidenced by CD and intrinsic fluorescence spectra similar to those in ethanol. Both LVs and BFs shield Gr tryptophans against quenching by acrylamide but the Stern-Volmer quenching constant was slightly higher for Gr in BFs confirming that the peptide is more exposed to the water phase in BFs than in LVs. The DPPC/DODAB/Gr supramolecular assemblies may predict the behavior of other antimicrobial peptides in assemblies with lipids. (C) 2012 Elsevier B.V. All rights reserved.

Abstract: Introduction: Xenotransplantation is a potential solution for the high mortality of patients on the waiting list for multivisceral transplantation; nevertheless, hyperacute rejection (HAR) hampers this practice and motivates innovative research. In this report, we describe a model of multivisceral xenotransplantation in which we observed immunoglobulin G (IgG) involvement in HAR. Methods: We recovered en bloc multivisceral grafts (distal esophagus, stomach, small intestine, colon, liver, pancreas, and kidneys) from rabbits (n = 20) and implanted them in the swine (n = 15) or rabbits (n = 5, control). Three hours after graft reperfusion, we collected samples from all graft organs for histological study and to assess IgG fixation by immunofluorescence. Histopathologic findings were graded according to previously described methods. Results: No histopathological features of rejection were seen in the rabbit allografts. In the swine-to-rabbit grafts, features of HAR were moderate in the liver and severe in esophagus, stomach, intestines, spleen, pancreas, and kidney. Xenograft vessels were the central target of HAR. The main lesions included edema, hemorrhage, thrombosis, myosites, fibrinoid degeneration, and necrosis. IgG deposition was intense on cell membranes, mainly in the vascular endothelium. Conclusions: Rabbit-to-swine multivisceral xenotransplants undergo moderate HAR in the liver and severe HAR in the other organs. Moderate HAR in the liver suggests a degree of resistance to the humoral immune response in this organ. Strong IgG fixation in cell membranes, including vascular endothelium, confirms HAR characterized by a primary humoral immune response. This model allows appraisal of HAR in multiple organs and investigation of the livers relative resistance to this immune response.

Abstract Background Preconception allergen immunization prevents neonatal allergen sensitization in mice by a complex interaction between regulatory cells/factors and antibodies. The present study assessed the influence of maternal immunization with ovalbumin (OVA) on the immune response of 3 day-old and 3 week-old offspring immunized or non-immunized with OVA and evaluated the effect of IgG treatment during fetal development or neonatal period. Results Maternal immunization with OVA showed increased levels of FcγRIIb expression in splenic B cells of neonates, which were maintained for up to 3 weeks and not affected by additional postnatal OVA immunization. Maternal immunization also exerted a down-modulatory effect on both IL-4 and IFN-γ-secreting T cells and IL-4 and IL-12- secreting B cells. Furthermore, immunized neonates from immunized mothers showed a marked inhibition of antigen-specifc IgE Ab production and lowered Th2/Th1 cytokine levels, whereas displaying enhanced FcγRIIb expression on B cells. These offspring also showed reduced antigen-specific proliferative response and lowered B cell responsiveness. Moreover, in vitro evaluation revealed an impairment of B cell activation upon engagement of B cell antigen receptor by IgG from OVA-immunized mice. Finally, in vivo IgG transference during pregnancy or breastfeeding revealed that maternal Ab transference was able to increase regulatory cytokines, such as IL-10, in the prenatal stage; yet only the postnatal treatment prevented neonatal sensitization. None of the IgG treatments induced immunological changes in the offspring, as it was observed for those from OVA-immunized mothers. Conclusion Maternal immunization upregulates the inhibitory FcγRIIb expression on offspring B cells, avoiding skewed Th2 response and development of allergy. These findings contribute to the advancement of prophylactic strategies to prevent allergic diseases in early life.

The aim of this study was to compare the techniques of indirect immunofluorescence assay (IFA) and flow cytometry to clinical and laboratorial evaluation of patients before and after clinical cure and to evaluate the applicability of flow cytometry in post-therapeutic monitoring of patients with American tegumentary leishmaniasis (ATL). Sera from 14 patients before treatment (BT), 13 patients 1 year after treatment (AT), 10 patients 2 and 5 years AT were evaluated. The results from flow cytometry were expressed as levels of IgG reactivity, based on the percentage of positive fluorescent parasites (PPFP). The 1:256 sample dilution allowed us to differentiate individuals BT and AT. Comparative analysis of IFA and flow cytometry by ROC (receiver operating characteristic curve) showed, respectively, AUC (area under curve) = 0.8 (95% CI = 0.64–0.89) and AUC = 0.90 (95% CI = 0.75–0.95), demonstrating that the flow cytometry had equivalent accuracy. Our data demonstrated that 20% was the best cut-off point identified by the ROC curve for the flow cytometry assay. This test showed a sensitivity of 86% and specificity of 77% while the IFA had a sensitivity of 78% and specificity of 85%. The after-treatment screening, through comparative analysis of the technique performance indexes, 1, 2 and 5 years AT, showed an equal performance of the flow cytometry compared with the IFA. However, flow cytometry shows to be a better diagnostic alternative when applied to the study of ATL in the cure criterion. The information obtained in this work opens perspectives to monitor cure after treatment of ATL.

[EN]Parliamentary websites have become one of the most important windows for citizens and media to follow the activities of their legislatures and to hold parliaments to account. Therefore, most parliamentary institutions aim to provide new multimedia solutions capable of displaying video fragments on demand on plenary activities. This paper presents a multimedia system for parliamentary institutions to produce video fragments on demand through a website with linked information and public feedback that helps to explain the content shown in these fragments. A prototype implementation has been developed for the Canary Islands Parliament (Spain) and shows how traditional parliamentary streaming systems can be enhanced by the use of semantics and computer vision for video analytics...

Questa tesi si pone come obiettivo la riqualificazione architettonica del plesso scolastico del Comune di Galeata, cittadina della media valle del Bidente conosciuta sia per gli estesi ritrovamenti archeologici dell’antica città romana di Mevaniola e del cosiddetto “Palazzo di Teodorico”, sia per essere la città con la percentuale più alta di immigrati regolari a livello nazionale. Sono proprio queste peculiarità a guidare il progetto verso un intervento mirato non solo alla rigenerazione urbana dell’area ma anche civica e sociale attraverso la dotazione di nuovi servizi e spazi pubblici. Il progetto nasce dall’idea di dare un carattere unitario e riconoscibile ad un’area attualmente caratterizzata da singoli edifici che semplicemente si susseguono l’uno dopo l’altro in modo paratattico. La volontà è invece quella di dar vita ad un vero e proprio “palazzo” dell’istruzione, caratterizzato da spazi privati e spazi pubblici, spazi aperti e spazi chiusi in grado di dare una molteplicità di risposte e soluzioni alla complessità della società odierna galeatese. Proprio lo spazio pubblico è infatti il vero motore dell’intervento; quello spazio pubblico attualmente negato a Galeata, che si è vista mutilata nella parte centrale del suo centro storico sventrato alla fine dell’Ottocento per la costruzione della strada statale bidentina, diventa ora generatore di nuove relazioni con gli edifici dell’area e di nuove dinamiche tra il plesso scolastico e la città grazie alla realizzazione di corti aperte e terrazzate, piazze e strade pedonali che fungono sia da giardini sia da spazi espositivi o comunque spazi polifunzionali che possono accogliere eventi nei vari mesi dell’anno, entrando così a far parte di quella serie di attrezzature a servizio delle manifestazioni culturali in cui è sempre attiva la valle del Bidente e la comunità di Galeata. La scuola d’infanzia e di istruzione primaria viene così a configurarsi non solo come il primo luogo della crescita culturale del bambino, ma anche come primo luogo di integrazione sociale, perché il nuovo progetto genera all’interno e all’esterno degli edifici occasioni di incontro e confronto tra gli scolari grazie all’attenzione riservata agli spazi per le attività di interciclo e per le attività libere. Inoltre l’area viene dotata di un centro maternità/consultorio dove sono collocati servizi di assistenza per le donne in gravidanza e dove le stesse possono confrontare e condividere la propria esperienza con le altre future mamme. L’intero intervento è guidato a livello progettuale anche da criteri di ecoefficienza come educazione alla responsabilità ambientale.

La Rocca delle Caminate nella coscienza e nell’immaginario Ben volentieri mi sono occupato dell’ambizioso progetto di restauro e recupero funzionale della Rocca delle Caminate, perché se è vero che il complesso ha rivestito un’importanza rilevante sotto il profilo storico e politico di questa parte di Romagna, è pur vero che vive nei miei ricordi fin dall’epoca dell’infanzia, ed è venuto acquistando nel tempo un preciso valore nella mia coscienza e nel mio immaginario. Questa rocca, questa fortificazione, questa torre che si staglia all’orizzonte dominando con austerità le amene vallate, l’ho sempre veduta e ha sempre sollevato in me suggestioni e interrogativi che però non mi ero mai preoccupato di chiarire: ero, per così dire, rimasto fedele alla mia emotività e mi erano bastati i racconti di nonna che andava spesso rammentando di quando, durante il ventennio fascista, insieme alle sorelle percorreva a piedi i sentieri che dalla vicina Dogheria conducevano alle Caminate, in occasione di una tal festa o di una tal funzione religiosa. Le sue parole e i suoi racconti continuano a donarmi un po’ del profumo dell’epoca, e fa specie notare come ancora oggi – in un tempo che fa dello sfavillio delle luci sfoggio di opulenza e miraggio di benessere – vengano puntualmente traditi dall’ammirazione per un faro che all’epoca ruotava emanando luce verde, bianca e rossa: quella del tricolore italiano. La Rocca delle Caminate – dicevo – è sempre stata una veduta familiare e fin troppo consuetudinaria, nei cui riguardi ero quasi giunto a una sorta d’indolenza intellettuale. Eppure, alla vigilia di decidere l’oggetto di questa tesi, ho volto lo sguardo ancora verso la “torre che domina a meraviglia il circostante paese”, ma in questa circostanza trovandomi cambiato: non più solo emozionato dal racconto nella memoria degli anziani, bensì spinto a una sfida anche per onorare il loro attaccamento verso questa fortificazione. È un popolo, quello che ho trovato, che mi ha stupito alquanto per la capacità di ricordare. Voglio dire ricordo, non trasognata visione; sottolineo fulgida testimonianza, non di rado animata dalla fierezza di chi sente fortemente il radicamento a una terra e rivendica il diritto di poter un giorno rivedere la ‘propria’ Rocca come l’ha veduta un tempo, poterla finalmente visitare e toccare, al di là di ogni colore politico, al di là di ogni vicenda passata, bella o brutta che sia. Poterla, in concreto, vivere. Così, un giorno ho varcato il limite dell’ingresso della Rocca e ho iniziato a camminare lungo il viale che conduce al castello; mi sono immerso nel parco e ho goduto della frescura dei pini, dei frassini e dei cipressi che fanno da contorno alla fortificazione. Certamente, ho avvertito un po’ di soggezione giunto ai piedi dell’arce, e mi è sorto spontaneo pormi alcune domande: quanti fanti e cavalieri saranno giunti a questa sommità in armi? Quanti ne saranno morti travolti nella furia della battaglia? Quanti uomini avranno provveduto alla sicurezza di questi bastioni oggi privati per sempre delle originarie mura difensive? Quanti castellani avranno presieduto alla difesa del castro? Ma più che a ogni altra cosa, il mio pensiero è andato a un protagonista della storia recente, a Benito Mussolini, che fra la fine degli anni venti e l’inizio degli anni trenta aveva eletto questo luogo a dimora estiva. Camminando lungo i corridoi della residenza e visitando le ampie stanze, nel rimbombo dei miei passi è stato pressoché impossibile non pensare a quest’uomo che si riconosceva in un duce e che qui ha passato le sue giornate, qui ha indetto feste, qui ha tenuto incontri politici e ha preso importanti decisioni. Come dimenticare quel mezzobusto, quella testa, quella postura impettita? Come dimenticare quello sguardo sempre un po’ accigliato e quei proclami che coglievano il plauso delle masse prima ancora di essere compiutamente formulati? In questo clima, fra castellani, capitani, cavalieri e duces di ogni epoca, sono entrato in punta di piedi, procedendo ai rilevamenti e ai calcoli per la realizzazione di questo progetto. È vero, non è stato facile muoversi nel silenzio che avvolge questo posto magico senza romperne l’incanto, e se a volte, maldestramente, ho fatto più rumore del solito, me ne rammarico. Sono sicuro che questi illustri signori sapranno scusarmene.
