Eugenitin, a chromone derivative and a metabolite of the endophyte Mycoleptodiscus indicus, at 5 mM activated a recombinant GH11 endo-xylanase by 40 %. The in silico prediction of ligand-binding sites on the three-dimensional structure of the endo-xylanase revealed that eugenitin interacts mainly by a hydrogen bond with a serine residue and a stacking interaction of the heterocyclic aromatic ring system with a tryptophan residue. Eugenitin improved the GH11 endo-xylanase activity on different substrates, modified the optimal pH and temperature activities and slightly affected the kinetic parameters of the enzyme.

Ruthenium compounds of the type trans-[Ru(NO)(NH3)(4)(L)] X-3, L = N-heterocyclic ligands, P(OEt)(3), SO32-, X BF4- or PF6-, or [Ru(NO)Hedta], were tested for antitumour activity in vitro against murine melanoma and human tumour cells. The ruthenium complexes induced DNA fragmentation and morphological alterations suggestive of necrotic tumour cell death. The calculated IC50 values were lower than 100 mu M. Complexes for which L = isn or imN were partially effective in vivo in a syngeneic model of murine melanoma B16F10, increasing animal survival. In addition, the same ruthenium complexes effectively inhibited angiogenesis of HUVEC cells in vitro. The results suggest that these nitrosyl complexes are a promising platform to be explored for the development of novel antitumour agents.

Twenty-three naphthoimidazoles and six naphthoxazoles were synthesised and evaluated against susceptible and rifampicin- and isoniazid-resistant strains of Mycobacterium tuberculosis. Among all the compounds evaluated, fourteen presented MIC values in the range of 0.78 to 6.25 mu g/mL against susceptible and resistant strains of M. tuberculosis, Five structures were solved by X-ray crystallographic analysis. Abstract Background N-acetyltransferase type 2 (Nat2) is a phase II drug- metabolizing enzyme that plays a key role in the bioactivation of aromatic and heterocyclic amines. Its relevance in drug metabolism and disease susceptibility remains a central theme for pharmacogenetic research, mainly because of its genetic variability among human populations. In fact, the evolutionary and ethnic-specific SNPs on the NAT2 gene remain a focus for the potential discoveries in personalized drug therapy and genetic markers of diseases. Despite the wide characterization of NAT2 SNPs frequency in established ethnic groups, little data are available for highly admixed populations. In this context, five common NAT2 SNPs (G191A, C481T, G590A, A803G and G857A) were investigated in a highly admixed population comprised of Afro-Brazilians, Whites, and Amerindians in northeastern Brazil. Thus, we sought to determine whether the distribution of NAT2 polymorphism is different among these three ethnic groups. Results Overall, there were no statistically significant differences in the distribution of NAT2 polymorphism when Afro-Brazilian and White groups were compared. Even the allele frequency of 191A, relatively common in African descendents, was not different between the Afro-Brazilian and White groups. However, allele and genotype frequencies of G590A were significantly higher in the Amerindian group than either in the Afro-Brazilian or White groups. Interestingly, a haplotype block between G590A and A803G was verified exclusively among Amerindians. Conclusions Our results indicate that ethnic admixture might contribute to a particular pattern of genetic diversity in the NAT2 gene and also offer new insights for the investigation of possible new NAT2 gene-environment effects in admixed populations.

Reactions initiated by collisions with low-energy secondary electrons has been found to be the prominent mechanism toward the radiation damage on living tissues through DNA strand breaks. Now it is widely accepted that during the interaction with these secondary species the selective breaking of chemical bonds is triggered by dissociative electron attachment (DEA), that is, the capture of the incident electron and the formation of temporary negative ion states [1,2,3]. One of the approaches largely used toward a deeper understanding of the radiation damage to DNA is through modeling of DEA with its basic constituents (nucleotide bases, sugar and other subunits). We have tried to simplify this approach and attempt to make it comprehensible at a more fundamental level by looking at even simple molecules. Studies involving organic systems such as carboxylic acids, alcohols and simple ¯ve-membered heterocyclic compounds are taken as starting points for these understanding. In the present study we investigate the role played by elastic scattering and electronic excitation of molecules on electron-driven chemical processes. Special attention is focused on the analysis of the in°uence of polarization and multichannel coupling e®ects on the magnitude of elastic and electronically inelastic cross-sections. Our aim is also to investigate the existence of resonances in the elastic and electronically inelastic channels as well as to characterize them with respect to its type (shape, core-excited or Feshbach), symmetry and position. The relevance of these issues is evaluated within the context of possible applications for the modeling of discharge environments and implications in the understanding of mutagenic rupture of DNA chains. The scattering calculations were carried out with the Schwinger multichannel method (SMC) [4] and its implementation with pseudopotentials (SMCPP) [5] at di®erent levels of approximation for impact energies ranging from 0.5 eV to 30 eV. A theoretical approach is used here to explain experimental results obtained from the electrosynthesis of polypyrrole-2-carboxylic acid (PPY-2-COOH) films in nonaqueous medium. An analysis of the Fukui function (reactivity index) indicates that the monomer (pyrrole-2-carboxylic acid, PY-2-COOH), and dimers and trimers are oxidized in the C4 or C5 positions of the heterocyclic ring of the PY-2-COOH structure. After calculating the heat of formation using semiempirical Austin Model 1 post-Hartree-Fock parameterization for dimer species, both C4 and C5 positions adjacent to the aromatic rings of PPY-2-COOH were considered the most susceptible ones to oxidative coupling reactions. The ZINDO-S/CI semiempirical method was used to simulate the electronic transitions typically seen in the UV-VIS-NIR range in monomer and oligomers with different conjugation lengths. The use of an electrochemical quartz crystal microbalance provides sufficient information to propose a polymerization mechanism of PY-2-COOH based on molecular modeling and experimental results.

Heterocyclic compounds represent almost two-thirds of all the known organic compounds: they are widely distributed in nature and play a key role in a huge number of biologically important molecules including some of the most significant for human beings. A powerful tool for the synthesis of such compounds is the hetero Diels-Alder reaction (HDA), that involve a [4+2] cycloaddition reaction between heterodienes and suitable dienophiles. Among heterodienes to be used in such six-membered heterocyclic construction strategy, 3-trialkylsilyloxy-2-aza-1,3-dienes (Fig 1) has been demonstrated particularly attractive. In this thesis work, HDA reactions between 2-azadienes and carbonylic and/or olefinic dienophiles, are described. Moreover, substitution of conventional heating by the corresponding dielectric heating as been explored in the frame of Microwave-Assisted-Organic-Synthesis (MAOS) which constitutes an up-to-grade research field of great interest both from an academic and industrial point of view. Reaction of the azadiene 1 (Fig 1) will be described using as dienophiles carbonyl compounds as aldehyde and ketones. The six-membered adducts thus obtained (Scheme 1) have been elaborated to biologically active compounds like 1,3-aminols which constitutes the scaffold for a wide range of drugs (Prozac®, Duloxetine, Venlafaxine) with large applications in the treatment of severe diseases of nervous central system (NCS). Scheme 1 The reaction provides the formation of three new stereogenic centres (C-2; C-5; C-6). The diastereoselective outcome of these reactions has been deeply investigated by the use of various combination of achiral and chiral azadienes and aliphatic, aromatic or heteroaromatic aldehydes. The same approach, basically, has been used in the synthesis of piperidin-2-one scaffold substituting the carbonyl dienophile with an electron poor olefin. Scheme 2 As a matter of fact, this scaffold is present in a very large number of natural substances and, more interesting, is a required scaffold for an huge variety of biologically active compounds. Activated olefins bearing one or two sulfone groups, were choose as dienophiles both for the intrinsic characteristic flexibility of the “sulfone group” which may be easily removed or elaborated to more complex decorations of the heterocyclic ring, and for the electron poor property of this dienophiles which makes the resulting HDA reaction of the type “normal electron demand”. Synthesis of natural compounds like racemic (±)-Anabasine (alkaloid of Tobacco’s leaves) and (R)- and (S)-Conhydrine (alkaloid of Conium Maculatum’s seeds and leaves) and its congeners, are described (Fig 2).

The aspartic protease BACE1 (β-amyloid precursor protein cleaving enzyme, β-secretase) is recognized as one of the most promising targets in the treatment of Alzheimer's disease (AD). The accumulation of β-amyloid peptide (Aβ) in the brain is a major factor in the pathogenesis of AD. Aβ is formed by initial cleavage of β-amyloid precursor protein (APP) by β-secretase, therefore BACE1 inhibition represents one of the therapeutic approaches to control progression of AD, by preventing the abnormal generation of Aβ. For this reason, in the last decade, many research efforts have focused at the identification of new BACE1 inhibitors as drug candidates. Generally, BACE1 inhibitors are grouped into two families: substrate-based inhibitors, designed as peptidomimetic inhibitors, and non-peptidomimetic ones. The research on non-peptidomimetic small molecules BACE1 inhibitors remains the most interesting approach, since these compounds hold an improved bioavailability after systemic administration, due to a good blood-brain barrier permeability in comparison to peptidomimetic inhibitors. Very recently, our research group discovered a new promising lead compound for the treatment of AD, named lipocrine, a hybrid derivative between lipoic acid and the AChE inhibitor (AChEI) tacrine, characterized by a tetrahydroacridinic moiety. Lipocrine is one of the first compounds able to inhibit the catalytic activity of AChE and AChE-induced amyloid-β aggregation and to protect against reactive oxygen species. Due to this interesting profile, lipocrine was also evaluated for BACE1 inhibitory activity, resulting in a potent lead compound for BACE1 inhibition. Starting from this interesting profile, a series of tetrahydroacridine analogues were synthesised varying the chain length between the two fragments. Moreover, following the approach of combining in a single molecule two different pharmacophores, we designed and synthesised different compounds bearing the moieties of known AChEIs (rivastigmine and caproctamine) coupled with lipoic acid, since it was shown that dithiolane group is an important structural feature of lipocrine for the optimal inhibition of BACE1. All the tetrahydroacridines, rivastigmine and caproctamine-based compounds, were evaluated for BACE1 inhibitory activity in a FRET (fluorescence resonance energy transfer) enzymatic assay (test A). With the aim to enhancing the biological activity of the lead compound, we applied the molecular simplification approach to design and synthesize novel heterocyclic compounds related to lipocrine, in which the tetrahydroacridine moiety was replaced by 4-amino-quinoline or 4-amino-quinazoline rings. All the synthesized compounds were also evaluated in a modified FRET enzymatic assay (test B), changing the fluorescent substrate for enzymatic BACE1 cleavage. This test method guided deep structure-activity relationships for BACE1 inhibition on the most promising quinazoline-based derivatives. By varying the substituent on the 2-position of the quinazoline ring and by replacing the lipoic acid residue in lateral chain with different moieties (i.e. trans-ferulic acid, a known antioxidant molecule), a series of quinazoline derivatives were obtained. In order to confirm inhibitory activity of the most active compounds, they were evaluated with a third FRET assay (test C) which, surprisingly, did not confirm the previous good activity profiles. An evaluation study of kinetic parameters of the three assays revealed that method C is endowed with the best specificity and enzymatic efficiency. Biological evaluation of the modified 2,4-diamino-quinazoline derivatives measured through the method C, allow to obtain a new lead compound bearing the trans-ferulic acid residue coupled to 2,4-diamino-quinazoline core endowed with a good BACE1 inhibitory activity (IC50 = 0.8 mM). We reported on the variability of the results in the three different FRET assays that are known to have some disadvantages in term of interference rates that are strongly dependent on compound properties. The observed results variability could be also ascribed to different enzyme origin, varied substrate and different fluorescent groups. The inhibitors should be tested on a parallel screening in order to have a more reliable data prior to be tested into cellular assay. With this aim, preliminary cellular BACE1 inhibition assay carried out on lipocrine confirmed a good cellular activity profile (EC50 = 3.7 mM) strengthening the idea to find a small molecule non-peptidomimetic compound as BACE1 inhibitor. In conclusion, the present study allowed to identify a new lead compound endowed with BACE1 inhibitory activity in submicromolar range. Further lead optimization to the obtained derivative is needed in order to obtain a more potent and a selective BACE1 inhibitor based on 2,4-diamino-quinazoline scaffold. A side project related to the synthesis of novel enzymatic inhibitors of BACE1 in order to explore the pseudopeptidic transition-state isosteres chemistry was carried out during research stage at Università de Montrèal (Canada) in Hanessian's group. The aim of this work has been the synthesis of the δ-aminocyclohexane carboxylic acid motif with stereochemically defined substitution to incorporating such a constrained core in potential BACE1 inhibitors. This fragment, endowed with reduced peptidic character, is not known in the context of peptidomimetic design. In particular, we envisioned an alternative route based on an organocatalytic asymmetric conjugate addition of nitroalkanes to cyclohexenone in presence of D-proline and trans-2,5-dimethylpiperazine. The enantioenriched obtained 3-(α-nitroalkyl)-cyclohexanones were further functionalized to give the corresponding δ-nitroalkyl cyclohexane carboxylic acids. These intermediates were elaborated to the target structures 3-(α-aminoalkyl)-1-cyclohexane carboxylic acids in a new readily accessible way.

ZusammenfassungDie selbstkondensierende Gruppenübertragungspolymerisation von 2-[(2-Methyl-1-triethylsiloxy-1-propenyl)oxy]ethyl-methacrylat (MTSHEMA) und die Copolymerisation mit Methylmethacrylat und tert-Butylmethacrylat wurde untersucht. Da MTSHEMA eine polymerisierbare Methacryloyl-Einheit und eine zur Initiierung einer Gruppenübertragungspolymerisation befähigte Silylketenacetal-Einheit besitzt, führt die Homopolymerisation zu hyperverzweigten und die Copolymerisation zu hochverzweigten Polymeren.Bei der Homopolymerisation von MTSHEMA konnten nur niedrige Molekulargewichte erreicht werden. Dies wird auf Nebenreaktionen der aktiven Kettenenden zurückgeführt, welche die Carbonylgruppen nucleophil angreifen und, mit der Doppelbindung Kern-Einheit reagieren. Die Copolymerisation mit Methylmethacrylat verlauft ohne Nebenreaktionen. Durch die Variation des molaren Verhältnisses von MTSHEMA zu den Comonomeren war es möglich, das Molekulargewicht, den Verzweigungsgrad und dadurch die Viskosität in Lösung zu kontrollieren. Die Bestimmung der Molekulargewichtsverteilung sämtlicher Polymere erfolgte durch Kopplung der Gelpermeationschromatographie mit einem Viskositätsdetektor und einem Vielwinkel Lichtstreu-Photometer. Die aus dem Vergleich der Viskositäten und Trägheitsradien ermittelten Schrumpfungspa-rameter lassen Schlüsse auf den Verzweigungsgrad zu.Nach den Ergebnissen der viskoelastischen Spektroskopie folgt das Verhalten der verzweigten Polymere in der Schmelze der Rouse-Theorie und deutet damit auf die Abwesenheit von Verschlaufungen hin.Durch die Copolymerisation mit tert-Butylmethacrylat und MTSHEMA konnte hochverzweigtes Poly(tert-butylmethacrylat) synthetisiert werden. Die Verseifung dieser Polymere ergab verzweigte Polymethacrylsäure.

Ein metallfreies Reaktionssystem mit Bis(triphenylphosphoranyliden)ammonium-Kationen als Gegenionen sowie das Additiv Lithium-2-methoxyethoxid wurden auf ihre Eignung zur anionischen Synthese von Blockcopolymeren mit (Meth)acrylatsegmenten bei moderaten Temperaturen im Strömungsrohr-Reaktor untersucht. Das metallfreie System ist zur lebenden Polymerisation von Methacrylaten in THF mit engen Molekulargewichtsverteilungen bei Reaktionszeiten < 1 s bis zu Temperaturen von 0 °C geeignet. In Gegenwart von Metallionen (Li+) findet eine Verlangsamung der Polymerisation unter Verlust der Reaktionskontrolle statt, Lithiumenolate verursachen nun breite, multimodale Molekulargewichtsverteilungen. Eine lebende Polymerisation von Acrylaten ist nicht möglich, massenspektroskopische Untersuchungen der Produkte weisen auf einen komplexen Reaktionsmechanismus mit Abbruch- und Übertragungsreaktionen hin. Die Synthese von Poly(styrol)-block-Poly(1,4-butadien)-block-Poly(methylmethacrylat)-Copolymeren in Toluol ist mit Lithium-2-methoxyethoxid als Additiv für die MMA-Polymerisation bei moderaten Mischtemperaturen (T < 0 °C) im Strömungsrohr-Reaktor möglich, die Effektivität des Wechselschritts von Polybutadien zu PMMA beträgt im Durchschnitt ca. 50 %. Untersuchungen verschiedener Reaktionsparameter, wie z.B. der Endfunktionalisierung des Polybutadiens mit 1,1-Diphenylethylen und der Temperatur während der Verkappung und der MMA-Polymerisation, geben keine eindeutigen Hinweise auf die Ursache dieses Phänomens. MALDI-TOF-Massenspektren des unreagierten Polybutadien Precursors zeigen die Anlagerung von 1-3 Molekülen Methylmethacrylat und keinen Abbruch durch Backbiting, was auf die Ausbildung stabiler Aggregate hindeutet.

Zusammenfassung:In der vorliegenden Arbeit konnte gezeigt werden, dass durch die Anwendung von Cyclodextrinen in der Polymerchemie ein neuer Weg gefunden wurde, Homo- und Copolymerisationen von hydrophoben, fluorophilen sowie ionischen Monomeren in wässrigem Medium durchzuführen, die bislang nur durch den Einsatz von Emulgatoren oder in organischen Lösemittelgemischen möglich waren. Standardmonomere wie z.B. Styrol konnten erfolgreich aus wässriger Phase in Gegenwart von Cyclodextrin polymerisiert werden; unter Anwendung des Zulauf-Verfahrens wurden stabile Latices mit monodisperser Teilchengrössenverteilung erhalten. Durch Verwendung von Cyclodextrinen als Löslichkeitsvermittler konnten fluorierte Methacrylate in wässriger Lösung in fast quantitativer Ausbeute homo- bzw. mit Styrol copolymerisiert werden, während bei Abwesenheit von Cyclodextrin keine bzw. nur geringe Umsätze erzielt wurden. Weiterhin ließen sich auch sehr unterschiedliche Monomerpaare wie 1H,1H,2H,2H-Perfluordecylacrylat mit Stearylmethacrylat in wässriger Phase unter Verwendung von Cyclodextrin copolymerisieren. Ein ebenfalls gegensätzliches Monomerenpaar, das sich als Cyclodextrin-Komplex in hohen Ausbeuten und zu hohen Molekulargewichten copolymerisieren ließ, war Natrium-4-(acrylamido)-phenyldiazosulfonat und Styrol, dessen Copolymerisation bislang nur in org. Lösemittelgemischen möglich war. In der Synthese leitfähiger Polymere konnte ein neuer, umweltfreundlicher Weg zur Herstellung von Polyheterocyclen durch oxidative Polymerisation der komplexierten Monomere in Wasser, wie z.B. 3,4-Ethylendioxythiophen oder Pyrrol, die ihre Anwendung zur Herstellung leitfähiger Polymere finden, erarbeitet werden.Zur Variation der Polymereigenschaften wurde auch erstmalig die neue Substanzklasse von 2H-Benzo[5,6][1,4]dioxino[2,3-c]pyrrol-Derivaten synthetisiert.

Die innerhalb dieser Arbeit mittels moderner Festkörper-NMR-Methoden untersuchte molekulare Dynamik in Poly(methacrylat)-Schmelzen und Polyphenylen-Dendrimeren ist durch eine bemerkenswerte Anisotropie gekennzeichnet.Die Anisotropie der molekularen Dynamik zeigt sich in geschmolzenen, ataktischen und isotaktischen Poly(ethylmethacrylaten) (PEMA) durch die Zeitskalenseparation der segmentellen alpha-Relaxation von einem etwa zwei Größenordnungen langsameren Relaxationsprozeß, welcher die Isotropisierung der Polymerhauptkette wiedergibt. Die Isotropisierungsdynamik der Polymerhauptkette wird - mit Ausnahme von PMMA - durch eine universelle, nicht-korrelationszeitenverteilte Relaxationsmode der Poly(methacrylate) quantifiziert, deren Temperaturabhängigkeit durch einen einheitlichen WLF-Parametersatz beschrieben werden kann. Geometrisch läßt sich die Isotropisierung der Hauptkette durch Sprungprozesse beliebiger Amplitude von Kettenstücken mit gestreckter all-trans-Konformation interpretieren. Die Kette zeigt eine außergewöhnliche konformative Stabilität. WAXS-Messungen deuten für PEMA und seine höheren Homologen die Existenz einer Schichtstruktur an, in der sich die steifen, polaren Hauptketten lokal in Monolagen anordnen, welche durch Bereiche zusammengelagerter Seitengruppen getrennt sind. Die Festkörper-NMR-Untersuchungen an Polyphenylen-Dendrimeren bringen zwei zentrale Aspekte in der wechselseitigen Beziehung von Struktur und Dynamik hervor. Zum einen ist die beobachtete molekulare Dynamik auf lokale Reorientierungen einzelner, terminaler Phenylringe um definierte Achsen beschränkt. Polyphenylen-Dendrimermoleküle sind unter diesen Bewegungen formstabil. Zum anderen können sowohl schnelle, als auch langsame Phenylreorientierungen nachgewiesen werden, wobei jeweils die intramolekulare Packungsdichte der Phenylringe das dynamische Verhalten der Polyphenylen-Dendrimere kontrolliert.

AbstractIn this thesis t-BOC and methacrylate functionalisedoligothiophenes with 3 – 5 thiophene units where synthesizedby Stille coupling. The material was structured by methodslike stamping and photolithography.The polymerizability of the methacrylates was proved by thepolymerization in solution with AIBN and subsequent analysisby gel permeation chromatography and differential scanningcalorimetry. The conductivities of the doped polymer filmswere in the range published for oligothiophenes.The optical and electrochemical properties of the compoundswere measured and compared to known compounds.The cleavage of the t-BOC groups was followed bythermogravimetry and infrared spectroscopy. The cleavagetemperature can be lowered by up to 100 °C by the use ofphoto acid generators. The cleavage is complete after 2hours annealing.The methacrylates were structured by microinjection mouldingin capillaries (MIMIC), stamping with soft silicone moulds,filling of patterned substrates and preperation of an opalreplica. By MIMIC line patterns with 5- 50 µm line widthswere obtained. The stamping succeeded in structures with 500nm line width only, on which liquid crystals wereorientated. This shows the possible application asorientation layers in LED’s with polarized emission. Withsilica opal templates the three-dimensional structuring ofthe oligomers succeeded.ZusammenfassungIn dieser Arbeit wurden neue funktionalisierteOligothiophene hergestellt und unter Anwendung verschiedenerVerfahren wie Stempeltechniken oder Photolithographiestrukturiert. Dazu wurden Oligothiophene mit drei bis fünfThiopheneinheiten durch Stille-Kupplung synthetisiert. Alsfunktionelle Gruppen wurden t-BOC-Ester undMethacrylsäureester eingeführt.Die Polymerisierbarkeit der Methacrylate wurde durch diePolymerisation mit AIBN in Lösung und anschließendeGelpermeationschromatographie und Differentialkalorimetrienachgewiesen. Die Leitfähigkeiten der dotierten Polymerfilmelagen im Bereich der für Oligothiophene bekannten Werte.Die optischen und elektrochemischen Eigenschaften derVerbindungen wurden untersucht und mit den Eigenschaftenbekannter Verbindungen verglichen. Die Abspaltung der t-BOC-Gruppen wurde thermogravimetrischund infrarotspektroskopisch verfolgt. Es wurde gezeigt, daßdie Abspaltungstemperatur durch den Zusatz einesPhotosäuregenerators um bis zu 100°C gesenkt wird und dieAbspaltung nach zweistündigem Tempern vollständig ist.Die methacrylatfunktionalisierten Verbindungen wurdenstrukturiert durch Micro Injection Moulding in Capillaries(MIMIC), Prägen mit weichen Silikonstempeln, Füllen vonstrukturierten Substraten und die Herstellung einerOpalreplika. Durch die Strukturierung mit MIMIC wurdenLinienstrukturen mit Linienbreiten von 5-50 µm erhalten.Durch Prägen wurden Strukturen mit Linienbreiten von nur 500nm erreicht, auf diesen gelang die Orientierung vonFlüssigkristallen. Dies zeigt die mögliche Anwendung alsOrientierungsschichten in Leuchtdioden mit polarisierterEmission. Durch die Verwendung von Siliziumoxidopalen alsTemplate gelang die dreidimensionale Strukturierung derOligomere.

The aim of this thesis was to investigate the synthesis of enantiomerically enriched heterocycles and dehydro-β-amino acid derivatives which can be used as scaffolds or intermediates of biologically active compounds, in particular as novel αvβ3 and α5β1 integrin ligands. The starting materials of all the compounds here synthesized are alkylideneacetoacetates. Alkylidene derivates are very usefull compounds, they are usually used as unsaturated electrophiles and they have the advantage of introducing different kind of functionality that may be further elaborated. In chapter 1, regio- and stereoselective allylic amination of pure carbonates is presented. The reaction proceeds via uncatalyzed or palladium-catalyzed conditions and affords enantiopure dehydro-β-amino esters that are useful precursor of biologically active compounds. Chapter 2 illustrates the synthesis of substituted isoxazolidines and isoxazolines via Michael addition followed by intramolecular hemiketalisation. The investigation on the effect of the Lewis acid catalysis on the regioselectivity of the addition it also reported. Isoxazolidines and isoxazolines are interesting heterocyclic compounds that may be regarded as unusual constrained -amino acids or as furanose mimetics. The synthesis of unusual cyclic amino acids precursors, that may be envisaged as proline analogues, as scaffolds for the design of bioactive peptidomimetics is presented in chapter 3. The synthesis of 2-substituted-3,4-dehydropyrrole derivatives starting from allylic carbonates via a two step allylic amination/ring closing metathesis (RCM) protocol is carried out. The reaction was optimized by testing different Grubbs’ catalysts and carbamate nitrogen protecting groups. Moreover, in view of a future application of these dehydro-β-amino acids as central core of peptidomimetics , the malonate chain was also used to protect nitrogen prior to RCM. Finally, chapter 4 presents the synthesis of two novel different classes of integrin antagonists, one derived from dehydro-β-amino acid prepared as described in chapter 1 and the other one has isoxazolidines synthesized in chapter 2 as rigid constrained core. Since that these compounds are promising RGD mimetics for αvβ3 and α5β1 integrins, they have been submitted to biological assay. and to interpret on a molecular basis their different affinities for the αvβ3 receptor, docking studies were performed using Glide program.

Ziel dieser Arbeit war es, ausgehend von auxiliargebundenen Piperidinderivaten, unterschiedliche chirale bi- und tricyclische Verbindungen darzustellen. Dazu wurde das 2,3,4,6-Tetra-O-pivaloyl--D-galactosylamin durch Kondensation mit Aldehyden in die entsprechenden Galactosylaldimine überführt, die in einer Lewissäure-katalysierten hochdiastereoselektiven Tandem-Mannich-Michael-Reaktionssequenz mit Danishefsky-Dien zu 2-substituierten Dehydropiperidinonen umgesetzt wurden. Die auf diese Weise zugänglichen chiralen Heterocyclen wurden diastereoselektiv in trans-konfigurierte 5-Bromverbindungen überführt. In einer Thiazolsynthese nach Hantzsch konnten die -Bromketone mit ambidenten Nukleophilen, wie Thiobenzamiden und unsymmetrischen Thioharnstoffderivaten, in niedrigen Ausbeuten zu bicyclischen Tetrahydro-thiazolo[5,4-c]pyridinen umgesetzt werden. Weitere bicyclische Heterocyclen mit einem Tetrahydro-thieno[2,3-c]pyridin-System konnten durch eine Gewald-Cyclisierung an 2-substituierten N-Galactosyl-piperidinonen erhalten werden. Durch Palladium-katalysierte Kreuzkupplungen an heterocyclischen Enoltriflaten, die ausgehend von den N-Galactosyl-dehydropiperidinonen synthetisiert wurden, gelang die Einführung von Aryl-, Alkinyl- und Alkenylsubstituenten in 4-Position des Piperidinringes. Zur Freisetzung der 2,4-disubstituierten Dehydropiperidinen wurde die N-glycosidische Bindung im sauren Milieu gespalten. Verbindungen mit einer exocyclischen Doppelbindung wurden einer Diels-Alder-Reaktion mit N-Phenylmaleinimid zum Aufbau von Isochinolinderivaten eingesetzt. Des Weiteren gelang die Synthese von 2-benzylsubstituierten N-Galactosyl-dehydropiperidinonen, wobei ortho-halogensubstituierte Phenylacetaldehyde eingesetzt wurden. Die in hohen Diastereomerenüberschüssen gebildeten Dehydropiperidinone wurden in die entsprechenden Enoltriflate überführt und einer Domino-Suzuki-Heck-Reaktion unterworfen. In dieser Kaskadenreaktion konnten tricyclische diastereomerenreine Benzomorphanderivate synthetisiert werden.
